The objective of this work was to evaluate the total endogenous N losses, and protein and energy net requirements for maintenance in growing lambs. Thirty-four castrated lambs, 17 F1 Ideal X Ile de France wool and 17 Santa Inas hair lambs, averaging 20 +/- 0.14 kg BW, were used in the experiment. Five animals from each genotype were slaughtered at the beginning of the experiment and taken as controls. Diets (D) were composed of concentrate mix (C) and Cynodon sp. c.v. Tifton 85 hay (R), combined in three different ratios: D1 = 60C:40R; D2 = 40C:60R and D3 = 20C:80R. Animals of each group of three lambs, that showed BW of 20 kg at the beginning of the dietary regimen, were slaughtered when one of them reached 35 kg, what always happened to be the one fed with D1. Total endogenous N losses estimated for wool lambs were 250 mg N/kg BW0.75. For hair lambs, total endogenous N losses reached 324 mg N/kg BW0.75 . Hair lambs showed higher (P < 0.01) (29.9%) net requirements of protein for maintenance than wool lambs. In contrast, net energy (NE) requirement for maintenance was similar (P > 0.05) for both genotypes (74.27 kcal/kg BW0.75 per day), the average of the antilog of the two intercept values obtained from the estimated regression equations of heat production for zero metabolizable energy (ME) consumption. Further studies should be done to check if this trend is also true for metabolizable energy and protein in animals exhibiting BW gains in tropical region. (C) 2003 Elsevier B.V. B.V. The present study aimed at determining the influence of condensed tannins present in the Brazilian legume species Mimosa hostilis, Mimosa caesalpinifolia and Bauhinia cheilantha on ruminal degradability, microbial colonization and enzymatic activity. Polyethylene glycol (PEG) was used to reduce the astringency and concentration of soluble condensed tannins. Four ruminally-cannulated Saanen goats (60 +/- 8 kg BW) were fed, in two experimental periods, with a hay diet based on the studied legumes treated or non-treated with PEG. Voluntary intake, microbial colonization, DM, CP, NDF, and ruminal degradability of PEG treated and non-treated forage leaves, as well as pH, ammonia and 1,4 P-endoglucanase activity of the rumen content were evaluated. Astringency and soluble tannin concentration of the studied legumes were reduced by approximately 70% and 50%, respectively, with PEG treatment. Average DM intake was higher for the treated diet (16.76 g DM/kg BW/day against 13.06 g DM/kg BW/day). Percentile values for degradation parameters and for potential and effective degradabilities of DM, CP and NDF were also affected by the tannins, but at different intensities. Electron microscopic observations of ruminally-incubated legume leaves showed a more effective microbial colonization of PEG-treated leaves for all legume species. A decrease in pH and an increase in ammonia concentration and in endoglucanase activity in the ruminal content was also observed for PEG-treated diets at all sampling periods. Condensed tannins of the studied legume species have influenced the adhesion conditions, colonization and enzymatic activity of the microbial ecosystem, and consequently the ruminal degradation of the different dietary fractions. For this reason, the reduction in condensed tannin would be of great importance to improve the nutrition of ruminant feeding of these species. (c) 2005 Elsevier B.V. All rights reserved.

This work was conducted to evaluate food intake and digestive efficiency of temperate wool and tropic semi-arid hair lambs, according to different concentrate: forage ratios in diet. Twenty-four lambs, averaging 90 +/- 1.8 days old and a mean body weight of 20 +/- 0.69 kg, 12 of them wool lambs, F, from Ideal x Ile de France crossing, and 12 others pure Santa Ines hair lambs, were distributed into a four replication 3 x 2 factorial arrangement consisting of three diets and two genotypes. Experimental diets consisted of: D1 = 60% concentrate mix (C) and 40% Cynodon sp. cv. Tifton-85 hay (F), D2 = 40% C and 60% F, and D3 = 20% C and 80% F. D1 was formulated for a daily gain of 300g per animal. Increasing forage levels in diets resulted in linear reductions (P < 0.01) in DM, OM, CP, TCH and metabolizable energy (ME) intake, and a linear increase (P < 0.01) in NDF ingestion. Tropic semi-arid hair lambs had higher DM, OM, NDF, CP, and TCH intake than temperate wool lambs. Although there were no genotype effects in OM and GE coefficient of digestibility, hair lambs showed more efficient (P < 0.05) digestibility of DM, CP, NDF and TCH. Increases in forage levels of diets corresponded to a negative linear effect (P < 0.01) in the apparent digestibility of DM, OM, CP, TCH and GE, while apparent digestibility of NDF increased linearly (P < 0.01). Total endogenous nitrogen (fecal plus urinary N) for F(1) Ideal x Ilede France wool and Santa Ines hair lambs were, respectively, 182 and 312 mg/kg(0.75) per day. Thus, Santa Ines tropic semi-arid hair lambs showed to be more responsive than F(1) Ideal x Ile de France temperate wool lambs to low quality fibrous diets. (C) 2004 Elsevier B.V. A total of 116 goats originating from three heterozygous bucks for rob (5/15) were cytogenetically studied at Botucatu, Brazil. In the mating 59,XY,t(5/15) x 60,XX, the offspring karyotype proportion did not differ significantly from the expected ratio of 1 normal:1 heterozygous. In the mating 59,XY,t(5/15) x 59,XX,t(5/15) the observed proportion was 1 normal:2 heterozygous:1 homozygous, and in the mating 59,XY,t(5/15) X 58,XX,tt(5/15) the observed proportion was 1 heterozygous:1 homozygous. No animals had unbalanced karyotypes and no differences in the weight at birth of normal, heterozygous and homozygous kids were observed (P > 0.10).

Testicular biopsy has been a complementary technique for clinical and research purposes to evaluate reproductive function in males. However, hemorrhage, inflammation, degeneration, and adhesion are factors that might limit the use of this procedure. In order to minimize these potential problems, fibrin glue derived from snake venom, a tissue adhesive with sealing, hemostatic, and healing properties, was used in conjunction with bilateral testicular biopsy with the Tru-Cut needle and was compared with a more conventional technique that uses nylon suture. Thirty mature rams were randomly assigned to three groups of 10 animals each, as follows: nonsurgical control group (no scrotal surgery, or biopsy); biopsy + glue group (fibrin glue on puncture sites and skin incisions) and biopsy + suture group (compression with swab on puncture sites and suturing of skin incision). The surgeries of the rams in the biopsy groups were performed on the same day, which was designated Day 0 for all three groups. Data of scrotal circumference, number of spermatozoa per ejaculum, percentage of morphologically abnormal spermatozoa, spermatozoa motility, and serum testosterone concentrations from Days -7, 20, 40, 60, 80 and 100 were evaluated. There were no significant differences between groups within days for any of the parameters evaluated. In conclusion, the testicular biopsy procedure using the Tru-Cut needle in conjunction with conventional nylon suture or the more novel fibrin glue in rams did not affect any of the parameters of testicular function evaluated in this study and was shown to be relatively simple, safe and efficient. An outbreak of botulism was detected in goats in the semiarid region of Brazil. In a flock of 460 goats, 38 does were affected and 37 died. Kids and younger goats were not affected. The main clinical signs were flaccid tetraparesis leading to tetraplegia that was often accompanied by twisted neck, tongue paralysis, and muscle tremors. At the time of the visit, 4 out of 11 affected goats were recumbent. Ambulatory goats had uncoordinated and swaying gaits with hypometria and weakness, mainly of the hind limbs. Two recumbent and four ambulatory goats showed twisted neck. Two recumbent goats were euthanized and necropsied. Non-significant gross and histologic lesions were observed. Samples of the liver, gut and rumen content were collected from the two goats and examined for botulinum toxins using the mouse serum neutralization test. The three samples from one goat were positive for type C toxin. Marked osteophagia was observed when the goats had access to bones in the pasture, and the farmer mentioned that osteophagia was common among goats of the flock. A sample of the plant Hybantus ipecaconha, the most abundant forage available for the goats, contained 2800 mg/kg of Ca and 450 mg/kg of P. One soil sample contained 58.12 mg/kg of Ca and 2.02 mg/kg of P. It was concluded that in this outbreak, botulism was associated with osteophagia probably due to phosphorus deficiency. The present work evaluated the pathological effects in both dams and their fetuses of cyanide ingestion during pregnancy using goat as animal model. From the Day 24 of pregnancy, three pregnant goats were dosed by gavage with 3.0 mg KCN/kg bw/day, and two others received only tap water. All dams were euthanized and necropsied at Day 120 of pregnancy. The fetuses were examined carefully for gross abnormalities. Determinations of cyanide and thiocyanate were performed in maternal and fetal blood and in amniotic fluid. Samples of several organs were collected for histopathologic evaluation. No clinical changes were seen in any animal throughout the pregnancy. No malformations or dead fetuses were observed: however, placenta from one treated goat presented increased cotyledon surface area occupied by vascular lumina. Histological lesions in KCN-treated dams consisted of vacuolation of hepatocytes and thyroid follicular cells, increased number of vacuoles on thyroid follicular colloid. and spongiosis of cerebral, internal capsule, and cerebellar peduncles tracts. Fetuses from these mothers showed vacuolation of hepatocytes and thyroid follicular cells, and spheroids in the cerebellar white matter. Levels of cyanide and thiocyanate were higher in maternal than fetal blood, which suggests that these substances were largely but not freely transferred from mothers to fetuses. Two hundred and five animals were cytogenetically studied. They were related to a male goat called 'Harald', imported from Switzerland, who was a t 5 15 chromosomic translocation carrier. 29.27% of the analyzed animals were heterozygous and 4.88% were homozygous. All carrying animals were found in Botucatu, Limeira and São Manuel city in Brazil. Descendants of this buck were sold in several regions of Brazil, and may have spread this aberration. © 1992.

The aim of the present work was to evaluate plasma membrane integrity, motility, vigor and morphology of fresh and frozen goat spermatozoa with or without seminal plasma. Semen samples were diluted in Tris solution, before and after thawing, with a combination of carboxifluorescein diacetate and propidium iodide. The results showed differences (P < 0.01) for motility and minor defects in the presence or absence of seminal plasma, for both fresh and frozen samples. Periods of collection had a significant effect on motility, probably due to changes in the photoperiod. Plasma membrane integrity was significantly reduced by the freezing process, whether seminal plasma was present or absent. In conclusion, removal of seminal plasma decreased motility and vigor rates in frozen samples. The photoperiod probably decreased the testosterone level, contributing negatively to the high percentage of sperm abnormalities, mainly damaged membranes. The use of fluorescent probes allowed a better estimation of the percentage of functional cells, instead of only estimating the percentage of motile cells or morphology defects. © 2001 Elsevier Science B.V. Three grazing management systems were compared to examine pasture decontamination of gastrointestinal nematode parasites (GIN) of sheep (Ovies aries) and cattle (Bos taurus). They consisted of sheep and cattle grazing paddocks alternately for 32, 96 or 192 days over 2 years. Pastureland (8.43 ha) was subdivided into six areas of eight paddocks each to produce an eight-paddock rotational grazing system. Every paddock was grazed for 4 days and then rested for 28 days. Sixty-six Ile de France ewes and 12 steers were randomly divided into three groups (22 sheep and four cattle per group). Each grazing system included a cattle area and a sheep area. Sheep and cattle interchanged areas every 32 days in system 1 (Group 1), every 96 days in system 2 (Group 2) and every 192 days in system 3 (Group 3). Fecal examination and larvae counting on pasture were performed every 32 days. During summer, winter and spring 2005, tracer lambs free of nematode infection were introduced into each sheep group and later sacrificed for quantification and identification of GIN species. All cattle were sacrificed for the same purpose. The main parasites found in tracer lambs were Haemonchus contortus and Trichostrongylus colubriformis, and in cattle, Haemonchus similis, Cooperia punctata and Oesophagostomum radiatum. Pasture contamination by sheep-infective GIN larvae was considerably reduced after 96 or 192 days of cattle grazing. Cross-infections between sheep and cattle GIN were not significant, which suggested that integrated grazing using such animals could be used for pasture decontamination. However, as effective anthelmintics were not available, decontamination was not sufficient for proper prophylaxis of GIN infections in Ile de France sheep, which are quite susceptible to such parasites. © 2007 Elsevier B.V. It is believed the temporary meiosis arrest with roscovitine or cycloheximide may improve the in vitro developmental competence of oocytes in different animal species. However, little is known about the effects of these inhibitors on ultrastructure of ovines cumulus-oocyte complexes (COCs). The aim of this study was to evaluate the progression of cytoplasmic maturation and the ultrastructural changes in sheep COCs exposed to roscovitine or cycloheximide, at acceptable concentrations. COCs were in vitro cultured for 24. h in maturation medium (control group) containing 100 μM roscovitine or 1 μg/mL cycloheximide (treatment groups). After this time, some COCs were cultured for further 22. h in inhibitor-free medium. The ultrastructure organization of COCs was evaluated by transmission electron microscopy before (immature group) and after in vitro culture for 24 and 46. h. As expected, signs of immaturity and maturity were observed in immature and control groups, respectively. In treatment with roscovitine, there were cumulus cells degeneration, swelling of mitochondrias, few cortical granules and many vesicles with electron-dense material. However, in cycloheximide treatment there were not signs of degeneration or cellular senescence. Metabolic units and mitochondrial pleomorphism were found in all experimental groups. These evidences demonstrate that roscovitine promoted irreversible ultrastructural changes while cycloheximide did not affect the cytoplasmic maturation. However, the implications on embryo development are still unclear. © 2012 Elsevier B.V.

Hydrometra is considered a very important pathological condition, because it represents one of the main causes of temporary infertility in dairy goats. The objective was (i) to evaluate a protocol for the treatment of hydrometra associated (n=2) or not (n=17) with follicular ovarian cyst in 19 dairy goats and (ii) to assess its reproductive efficiency after treatment. For this purpose, 10. mg of dinoprost (PGF) divided in two equal doses were administered to all animals intravulvosubmucosally on Days 0 and 10. In addition, 500. IU hCG were administered on Day 7. Ultrasound exams were performed in all females from Days 0 to 3, 7 and 10 to 13 of treatment, in order to evaluate uterus drainage after each treatment. Goats were monitored for estrus after both treatments and mated after the second dose of PGF. Blood samples were collected from 11 goats to determine plasma progesterone concentrations before, during and after treatment. Of the 19 goats treated, 16 lost weight after the first dose, probably due to uterine discharge. Complete drainage of uterine fluid was observed in 11/19 (57.9%) and 17/19 (89.5%) after the first and second doses, respectively. Afterwards, we diagnosed 2 more goats with follicular cysts, for a total of 21.1% (4/19) of animals exhibiting hydrometra and ovarian cyst concomitantly. In one doe the diagnosis was on Day 2 and in the other on Day 11. All does showed progesterone concentrations superior to 1. ng/mL at Day 0, with an average of 10.6 ± 1.4. ng/mL. Out of the 10 goats mated, only two became pregnant after treatment, corresponding to 10.5% of the total (2/19). Although prostaglandin was effective to drain the uterine fluid and led to the onset of estrus, it did not improve the pregnancy rate. The use of hCG in female goats was not effective in luteinizing the cysts. It can be concluded that hydrometra alone or associated with ovarian follicular cyst may adversely affect goat reproductive performance. © 2012 Elsevier B.V.
