The sequential extraction procedure of Zinc and lead performed in a Brazilian soil showed that it presents high pollution potential once over 90% of total lead is present in fractions where the metals can be easily mobilized. The fraction contents are as follow: F1 = 174 and 15 mg kg-1; F2 = 3155 and 9.7 mg kg -1; F3 = 99 and 1.6 mg kg -1; Residual fraction = 38 and 5.5 mg kg -1 for lead and zinc, respectively. The comparison with non contaminated soil only Pb 2+ concentration is above its intervention reference concentration, 900 mg kg -1.

The following research project deals with the development of new analytical procedures aimed at the identification and quantification of the element zinc in different fractions of vegetable drugs used for weight loss, using the high-resolution continuum source atomic absorption spectrometry in flame. The determination of the levels of zinc was made in an AnalytikJena ContrAA 300 spectrometer in the principal line of Zn at 213.857 nm and wavelength integrated absorbance equal to 3 pixels. Calibration curves were obtained with linear dynamic range from 0.10 to 1.00 mg L-1, correlation coefficient equal to 0.9991 and limit of detection equal to 0.020 mg L-1. The total Zn content in the samples obtained after microwave-assisted acid digestion system were: 28.06 mg kg-1 (Artichoke), 31.49 mg kg-1 (Chamomile), 77.16 mg kg-1 (Gorse), 105.38 mg kg-1 (Horsetail), 32.37 mg kg-1 (Fennel) and 5.00 mg kg-1 (Senna). The extraction in aqueous environment assisted by microwave radiation produced extracts containing: 5.40 mg kg-1 (Artichoke), 10.65 mg kg-1 (Chamomile), 14.83 mg kg-1 (Gorse), 18.90 mg kg-1 (Horsetail), 7.80 mg kg-1 (Fennel), 3.24 mg kg-1 (Senna), corresponding to 19%, 34%, 19%, 18%, 24% and 64% of their total content of Zn, respectively. The amounts of Zn in the extracted samples from some plants indicate that this metal was strongly bounded to the plant's matrix. The sequential extraction using solvents and mixtures with different polarities (n-hexane, ethyl acetate, propanol, methanol, ethanol-water 1:1 (v/v), water) aimed the separation of different Zn containing species and water was considered the most efficient extractor. The Zn contents extracted were: 14.82 mg kg-1 (Artichoke), 16.82 mg kg-1 (Chamomile), 34.09 mg kg-1 (Gorse), 4.75 mg kg-1 (Senna), corresponding to 53% in the first two samples, 44% and 95% for the last two, respectively. The effects of domestic cooking on proteins, organic compounds and Fe distribution in beans (Phaseolus vulgaris L.) were investigated. Sequential extraction with different extractant solutions (mixture of methanol and chloroform 1:2 v/v, water, 0.5 mol L-1 NaCl, 70% v/v ethanol and 0.5 mol L-1 NaOH) were used for extracting lipids, albumins, globulins, prolamins and glutelins, respectively. Iron determination by graphite furnace atomic absorption spectrometry (GF AAS), proteins by Bradford method and organic compounds by matrix-assisted laser desorption ionization time-of-flight mass spectrometry (MALDI-TOF MS) were carried out in this work. High concentration of albumins, globulins and glutelins were found in raw beans, while in the cooked beans, albumins and glutelins are main proteins types. The MALDI-TOF MS spectra of raw and cooked beans revealed that the domestic cooking altered the molecular weight of the organic compounds, since that in the cooked beans were found compounds between 2 and 3.5 kDa, which were not presented in the raw beans. Besides this, in cooked beans were also observed the presence of four compounds of high molecular weight (12-16 kDa), being that in the raw grains there is only one (ca. 15.2 kDa). In raw grains is possible to observe that Fe is mainly associated to albumins, globulins and glutelins. For cooked grains, Fe is associated to albumins and globulins.

This paper describes a rapid method for arsenic (As) speciation by LC-ICP-MS in several types of food samples. Prior to analysis, samples were milled and the As species extracted from biological tissues by sonication in only 2 min with a solution containing MeOH (10%, v/v) plus HNO3 (2%, v/v). As species were separated by LC using an anion exchange column. Method detection limits for AsB, As3+,DMA, MMA and As5+ were 1.3, 0.9, 0.6, 0.7 and 0.8 ng g(-1), respectively. Method accuracy and precision were traceable to Certified Reference Materials SRM1577 bovine liver from the National Institute of Standards and Technology, CE278 mussel tissue from the Institute of Reference Materials and Measurements and DOLT-3 dogfish liver tissue and DORM-3 fish protein from the National Research Council of Canada. Finally, the method was applied to speciate As in food samples (egg, fish muscle, beef and chicken) purchased in Brazilian markets.

Throughout the world, pressures on water resources are increasing, mainly as a result of human activity. Because of their accessibility, groundwater and surface water are the most used reservoirs. The evaluation of the water quality requires the identification of the interconnections among the water reservoirs, natural landscape features, human activities and aquatic health. This study focuses on the estimation of the water pollution linked to two different environmental issues: salt water intrusion and acid mine drainage related to the exploitation of natural resources. Effects of salt water intrusion occurring in the shallow aquifer north of Ravenna (Italy) was analysed through the study of ion- exchange occurring in the area and its variance throughout the year, applying a depth-specific sampling method. In the study area were identified ion exchange, calcite and dolomite precipitation, and gypsum dissolution and sulphate reduction as the main processes controlling the groundwater composition. High concentrations of arsenic detected only at specific depth indicate its connexion with the organic matter. Acid mine drainage effects related to the tin extraction in the Bolivian Altiplano was studied, on water and sediment matrix. Water contamination results strictly dependent on the seasonal variation, on pH and redox conditions. During the dry season the strong evaporation and scarce water flow lead to low pH values, high concentrations of heavy metals in surface waters and precipitation of secondary minerals along the river, which could be released in oxidizing conditions as demonstrated through the sequential extraction analysis. The increase of the water flow during the wet season lead to an increase of pH values and a decrease in heavy metal concentrations, due to dilution effect and, as e.g. for the iron, to precipitation.

Die Verwendung von Metallen zur Entwicklung der heutigen fortschrittlichen technologischenrnGesellschaft lässt auf eine lange Geschichte zurück blicken. Im Zuge des letzten Jahrhundertsrnwurde realisiert, dass die chemischen und radioaktiven Eigenschaften von Metallen einernernsthafte Bedrohung für die Menschheit darstellen können. In der modernen Geochemie ist esrnallgemein akzeptiert, dass die spezifischen physikochemische Formen entscheidender sind, alsrndas Verhalten der gesamten Konzentration der Spurenmetalle in der Umwelt. Die Definition derrnArtbildung kann grob als die Identifizierung und Quantifizierung der verschiedenen Formen oderrnPhasen für ein Element zugeordnet werden. Die chemische Extraktion ist eine gemeinsamernSpeziierungstechnik bei der die Fraktionierung des Gesamtmetallgehaltes zur Analyse der Quellernanthropogener Metallkontamination und zur Vorhersage der Bioverfügbarkeit von verschiedenenrnMetallformen dient. Die Philosophie der partiellen und sequenziellen Extraktionsmethodernbesteht darin, dass insbesondere das Extraktionsmittel phasenspezifisch unter chemischemrnAngriff unterschiedlicher Mischungsformen steht. Die Speziation von Metall ist wichtig bei derrnBestimmung der Toxizität, Mobilität, Bioverfügbarkeit des Metalls und damit ihr Schicksal inrnder Umwelt und biologischem System. Die Artenbildungsanalyse kann für das Verständnis derrnAuswirkung auf die menschliche Gesundheit und bei ökologischen Risiken durch diernQuantifizierung von Metallspezies bei einem Untersuchungs-standort angewendet werden undrnanschließend können Sanierungsstrategien für den Standort umgesetzt werden. Mit Hilfe derrnSpezifizierung wurden Arsen und Kupfer in landwirtschaftlichem Kalkdünger und Thallium inrnkontaminierten Böden untersucht und in den folgenden Abschnitten im Einzelnen dargestellt.

Nd and Pb isotopic compositions extracted from bulk deep sea sediments have been shown to be robust proxies for deep water circulation as well as weathering provenance and intensity over geologically young time scales. In this study we evaluated ten deep sea samples from Ocean Drilling Program (ODP) site 1090 ranging in age from mid Eocene to early-Miocene to test whether Pb isotopic compositions extracted from geologically older sediments record reliable seawater isotopic ratios and to evaluate the source of the extracted Pb. The sequential extraction protocol used in this study is similar to protocols reported for previous studies and produces acetic acid, hydroxylamine hydrochloride (HH) and residue fractions. Each extracted fraction was analyzed for Pb isotopes, rare earth elements (REEs), and a suite of major elements. Similar 206Pb/204Pb, 207Pb/204Pb, and 208Pb/204Pb ratios are recorded from the acetic acid and HH fractions for ~70-80% of the samples, suggesting that either the acetic acid dissolves Fe-Mn oxides or multiple phases are recording the same seawater isotopic value. Several indirect tests, such as Al mass balance, comparison of Sr isotopes in HH extracts to contemporaneous seawater Sr isotopes, and comparison of Nd isotopic compositions in HH extracts to published fossil fish teeth values, provide evidence that Pb isotopic compositions measured in our bulk HH extracts record bottom water values. The relationship between Pb, Mn and Ca concentrations in HH fractions indicates that Fe-Mn oxides and a Mn-bearing carbonate are the dominant phases contributing seawater Pb. Comparison of REE patterns derived from the HH fraction and total digestions of Fe-Mn nodule standards reveals that the trivalent REEs exhibit patterns consistent with the parent archive, but Ce can be fractionated during extraction. Ratios of REEs also produce unique fields for each fraction and can be used to test the purity of the seawater signal of the extraction protocol. Finally, an initial evaluation of Pb isotopic compositions in fossil fish indicates that this archive is not suitable for bottom water Pb isotope studies.
