Ethnopharmacological relevance: In Brazil, a phytotherapeutic preparation produced from a standardized tincture of Cinchona calisaya Weddel such that each mL of product contains 400 mu g of quinine, known in Portuguese as Agua Inglesa (R) (English water), is indicated by the manufacturer as a tonic, appetite stimulant, and digestive. However, this preparation has long been used in folk medicine as a female fertility stimulant. Despite its widespread use in folk medicine to stimulate female fertility, no study has been undertaken to assess the potential teratogenic and genotoxic effects of this phytotherapeutic preparation. The aim of the present study was to investigate possible toxic reproductive effects in mice caused by exposure to Agua Inglesa (R), either before mating or during the pre- and post-embryo implantation periods. The genotoxic potential was evaluated using the micronucleus assay.Material, Methods, and Results: Virgin female mice, with at least one estrous cycle evidenced by vaginal cytology, were divided into five groups of 15 individuals each (Group I - control, Group II - treated with ethanol solution at 16%, Groups III, IV and V treated with phytotherapeutic preparation at 1.5 mL/kg/day, 3.0 mL/kg/day and 4.5 mL/kg/day, respectively). After the first 28 days of treatment, females were caged individually with adult fertile males. Pregnant females continued to receive treatment for seven days (preimplantation period). Body weight was recorded weekly during treatment. Signs of toxicity (weight loss, food intake, piloerection, apathy, prostration, diarrhea, seizures, behavioral changes, and locomotion) were also observed. The females were sacrificed on the 15th day of pregnancy, uterine horns were evaluated for implantation, and the placental index was recorded. In the micronucleus test, 2000 polychromatic erythrocytes (PCE) per animal, obtained from bone marrow, were scored. Results The results showed that exposure of the females during the pre- and post-implantation periods did not significantly alter the reproductive capacity (p < 0.05); however, in higher dose (three times human dose)reduction of fetal weight was observed. There was no difference between the control and phytotherapeutic preparation (p > 0.05) in terms of the average number of micronucleated polychromatic erythrocytes. Conclusions: Although folk medicine suggests that the Agua Inglesa (R) preparation is useful as a female fertility stimulant, no such effect was confirmed in mice. Palicourea rigida H.B.K. (Rubiaceae), a medicinal species commonly known as douradinha, has wide distribution across ecosystems in Central and South America. This species exhibits seed dormancy delaying germination until optimal conditions for seedling growth and development are in place. While dormancy ensures species survival, it also presents a technical problem for developing P. rigida’s plant production program. Thus, the objective of this study was to investigate if secondary metabolites present in seeds influence the seed dormancy of P. rigida. Mature fruits were harvested from the native habitat, in the savanna region of the State of Minas Gerais during February 2009, 2010 and 2011. The content of phenolic compounds in the seed of P. rigida was measured, and the allelopathic effects were assessed using the germination of lettuces as model to detect phytotoxicity. The P. rigida seeds geminated at rates varying between 7% and 31% with a Seed Germination Index (SGI) of 0.09. Data suggest that the phenolic compounds present in the seeds may be responsible for seed dormancy.

The therapeutic use of medicinal plants has contributed since antiquity in a beneficial way for health. However, many species lacks of scientific evidence which provide basis for their use in therapeutic practice. In this context is the Genipa americana L. species (Rubiaceae), popularly known as jenipapo and used to treat syfilis, ulcer and hemorrhagic disturbs. It's also used against bruising, as tonic and as aphrodisiac. Due this species lacks toxicological studies, the aim of this study was to evaluate the toxicity in vivo (acute and sub-chronic toxicity) and in vitro (cytotoxicity) of the hydroethanolic extract from G. americana fruits. The hydroethanolic extract of G. americana fruits was prepared by maceration. A preliminary phytochemical analysis was performed to assess the presence of secondary metabolites in the extract. The cytotoxicity study of the extract (0.1, 1.0, 10, 100 and 1000 mg / 100 ul) were performed against normal cells (3T3) and tumor (786-0, HepG2 and B16), analyzed by the MTT assay. To evaluate the acute (single dose of 2000 mg / Kg) and subchronic (100, 500 and 1000 mg / kg for 30 days) toxicity Swiss mice of both sexes were used. At the end of the experiment, blood samples and organs were collected for analysis. Data between groups were compared by t test or ANOVA with Dunnett's post-test with 5% significance level. The phytochemical study of the extracts mainly indicated the presence of iridoids. Results for cytotoxicity tests showed up to 70% inhibition of B16 cell line at a dose of 1000 mg / 100 ul, and up to 29% inhibition of 786-0 at a dose of 10 ug / 100 ul. The extract did not cause death in 3T3 and HepG2 cells. During the in vivo assays, there were no animal deaths. Analysis of blood samples revealed that the animals submitted to the evaluation of acute toxicity had changes in AST and ALT, and that the animals evaluated for subchronic toxicity showed changes in the relative wet weight of the kidney and plasma urea concentration. No differences were observed between groups on histopathological evaluation of the collected organs. Despite the changes found in the in vivo toxicity tests, using the criteria described by the OECD Guidelines, it is suggested that the hydroethanolic extract of the fruits of the G. americana is classified as low toxicity. The cytotoxicity of the extract suggests that they have potential against melanoma cell lines (B16).

The strong selection pressure exerted by intensive use of glyphosate in cultivated areas has selected populations of the Rubiaceae weed species Borreria latifolia (Aubl.) K.Shum. (broadleaf buttonweed), Galianthe chodatiana (Standl.) E.L. Cabral (galiante) and Richardia brasiliensis Gomes (Brazilian pusley) with differential sensitivity to this herbicide in the South region of Brazil. The control of these weeds with herbicides is troublesome and signals the need to incorporate management practices of ruderal flora and crops, more sustainable and that results in more efficient control for long-term. Therefore, it is very important to expand the information about their biology and management. This study aimed: (a) select efficient methods to overcome dormancy of B. latifolia and G. chodatiana and determine how they influence the kinetics of seeds germination; (b) analyze the effects of temperature, irradiance, pH, aluminum and salinity on seed germination and initial growth of the B. latifolia, G. chodatiana e R. brasiliensis seedlings; (c) evaluate tolerance to glyphosate levels in biotypes of B. latifolia, G. chodatiana e R. brasiliensis through dose-response curves and compare two methods to evaluate herbicidal control; (d) and evaluated the effectiveness of alternative herbicides in pre-emergence and in early and late post-emergence of the three species. The treatment with KNO3 2%/3h + gibberellic acid 400 ppm resulted in higher percentage of G. chodatiana seed germination. This treatment and also the dry heat (60°C/30 min) + KNO3 2%/3h were more effective in overcoming dormancy of B. latifolia. G. chodatiana and R. brasiliensis tolerate lower temperatures during the germination process, while B. latifolia tolerate higher temperatures. B. latifolia and R. brasiliensis are positive photoblastic while G. chodatiana is indifferent to the photoperiod. B. latifolia shows higher germination and early development in pH 3, while G. chodatiana and R. brasiliensis prefer pH range between 5 and 7. B. latifolia and G. chodatiana were more tolerant to the aluminum during the germination process than R. brasiliensis. Low salt levels were sufficient to reduce the seed germination of the three species. Some biotypes of B. latifolia and R. brasiliensis showed medium-high glyphosate tolerance, not being controlled by higher doses than recommended. The G. chodatiana specie was not controlled with the highest dose used, showing a high glyphosate tolerance. The sulfentrazone, s-metolachlor and saflufenacil herbicides sprayed in pre-emergence showed high efficacy both on B. latifolia and R. brasiliensis, while chlorimuron-ethyl and diclosulan were effective only on R. brasiliensis. In early post-emergence the fomesafen, lactofem and flumioxazin herbicides efficiently controlled plants of all species, while bentazon showed high efficacy only on B. latifolia. Noteworthy the susceptibility of the G. chodatiana specie for applications in early post-emergence, because the control effectiveness and the number of effective herbicides are reduced with increasing the plant age. Many treatments with tank mix or sequencial applications with glyphosate, were effective in controlling B. latifolia and R. brasiliensis plants in advanced stage of development.
