Summary : The chemokines CCL19 and CCL21 and their common receptor CCR7 attract antigenpresenting dendritic cells (DCs) and naive T cells into the T zone of secondary lymphoid organs (SLO) and are therefore critically involved in homeostatic T cell recirculation and the initiation of adaptive immune responses. In addition. CCR7 ligands were proposed to mediate T cell exit from neonatal thymus, allowing colonization of T zones in SLOB. The relative contribution of CCL19 and CCL21 to these processes has remained unclear, as they were studied in mouse models lacking either CCR7 or both ligands. The aim of my thesis was to characterize Cc119-' mice and thereby investigate the relative roles of the two CCR7 ligands in development, homeostasis and immune response. The first study addressed the role of CCR7 ligands in DC biology. We found that CCL19 is dispensable for DC migration to lymph nodes and their localization to T zones. Furthermore, a CCL19-deficient environment did not lead to a defect in DC maturation or T cell priming. Therefore, CCL21 is sufficient to mediate CCR7-dependent processes during the initiation of adaptive immune responses. In the second study we investigated how the two CCR7 ligands affect CCR7 expression and function on naive T cells. We found that in SLOB CCR7 is constantly occupied with CCL19 and CCL21, eventually leading to its internalization. The reduced level of free CCR7 on these cells led to diminished ligand sensitivity and consequently impaired chemotactic responses. This effect was reversible by passage through aCCR7 ligand-free environment like the blood circulation. We propose that the different states of ligand sensitivity in SLOB and blood are important to allow for proper T cell recirculation. In the third study the role of CCL19 in neonatal thymus and spleen was analyzed. While neonatal Cc119-!- mice had no defect in thymic egress, we observed reduced T cell accumulation in the spleen but not lymph nodes. We identified reticular stromal cells in the developing white pulp (WP) as the major CCL 19 source. The development of these WP stromal cells as well as their CCL19 expression were dependent on LTalß2+ B cells. In conclusion, we have found that CCL21 can mostly compensate for lack of CCL19 in homeostasis and immunity. In contrast, during development. CCL19 has anon-redundant function for T cell colonization of the spleen.

Interactions between major histocompatibility complex (MHC) molecules expressed on stromal cells and antigen-specific receptors on T cells shape the repertoire of mature T lymphocytes emerging from the thymus. Some thymocytes with appropriate receptors are stimulated to undergo differentiation to the fully mature state (positive selection), whereas others with strongly autoreactive receptors are triggered to undergo programmed cell death before completing this differentiation process (negative selection). The quantitative impact of negative selection on the potentially available repertoire is currently unknown. To address this issue, we have constructed radiation bone marrow chimeras in which MHC molecules are present on radioresistant thymic epithelial cells (to allow positive selection) but absent from radiosensitive hematopoietic elements responsible for negative selection. In such chimeras, the number of mature thymocytes was increased by twofold as compared with appropriate control chimeras This increase in steady-state numbers of mature thymocytes was not related to proliferation, increased retention, or recirculation and was accompanied by a similar two- to threefold increase in the de novo rate of generation of mature cells. Taken together, our data indicate that half to two-thirds of the thymocytes able to undergo positive selection die before full maturation due to negative selection.

The chemokine receptor CCR7 is critical for the recirculation of naive T cells. It is required for T cell entry into secondary lymphoid organs (SLO) and for T cell motility and retention within these organs. How CCR7 activity is regulated during these processes in vivo is poorly understood. Here we show strong modulation of CCR7 surface expression and occupancy by the two CCR7 ligands, both in vitro and in vivo. In contrast to blood, T cells in SLO had most surface CCR7 occupied with CCL19, presumably leading to continuous signaling and cell motility. Both ligands triggered CCR7 internalization in vivo as shown in Ccl19(-/-) and plt/plt mice. Importantly, CCR7 occupancy and down-regulation led to strongly impaired chemotactic responses, an effect reversible by CCR7 resensitization. Therefore, during their recirculation, T cells cycle between states of free CCR7 with high ligand sensitivity in blood and occupied CCR7 associated with continual signaling and reduced ligand sensitivity within SLO. We propose that these two states of CCR7 are important to allow the various functions CCR7 plays in T cell recirculation.

The murine gut epithelium contains a large population of thymus-derived intraepithelial lymphocytes (IELs), including both conventional CD4(+) and CD8alphabeta(+) T cells (expressing T-cell receptor alphabeta [TCRalphabeta]) and unconventional CD8alphaalpha(+) T cells (expressing either TCRalphabeta or TCRgammadelta). Whereas conventional IELs are widely accepted to arise from recirculation of activated CD4(+) and CD8alphabeta(+) T cells from the secondary lymphoid organs to the gut, the origin and developmental pathway of unconventional CD8alphaalpha IELs remain controversial. We show here that CD4-Cre-mediated inactivation of c-Myc, a broadly expressed transcription factor with a wide range of biologic activities, selectively impairs the development of CD8alphaalpha TCRalphabeta IELs. In the absence of c-Myc, CD4(-) CD8(-) TCRalphabeta(+) thymic precursors of CD8alphaalpha TCRalphabeta IELs are present but fail to develop on adoptive transfer in immunoincompetent hosts. Residual c-Myc-deficient CD8alphaalpha TCRalphabeta IEL display reduced proliferation and increased apoptosis, which correlate with significantly decreased expression of interleukin-15 receptor subunits and lower levels of the antiapoptotic protein Bcl-2. Transgenic overexpression of human BCL-2 resulted in a pronounced rescue of CD8alphaalpha TCRalphabeta IEL in c-Myc-deficient mice. Taken together, our data support a model in which c-Myc controls the development of CD8alphaalpha TCRalphabeta IELs from thymic precursors by regulating interleukin-15 receptor expression and consequently Bcl-2-dependent survival.
Proyecto de ejecución de zona lúdica de verano formada por piscinas, jacuzzi y edificio de servicios

Este trabajo final de carrera se basa en la redacción de un proyecto de ejecución de una zona lúdica situada en el interior del recinto de un camping, la cual estará formada por una piscina de grandes dimensiones formada por tres vasos de tipo desbordante, una piscina de tipo infantil, un jacuzzi y finalmente un edificio de servicios complementarios (bar, aseos y enfermería). La piscina grande estará formada por tres vasos donde el agua va deslizándose de uno a otro hasta ser recogida por un depósito de compensación desde donde se realizará la filtración y tratamiento antes de ser devuelta al piscina de nuevo. La instalación está también preparada para ser utilizado cada vaso como una piscina individual, recogiendo el agua por los rebosaderos laterales y conduciéndola al mismo vaso de compensación. El tratamiento del agua se realizará de forma física a través de la filtración mediante filtros de arena y de forma química analizando la cantidad de cloro residual y el nivel de PH y posteriormente añadiendo de forma automática las cantidades de cloro y corrector de PH adecuados mediante bombas dosificadoras de precisión. La piscina infantil es de pequeñas dimensiones y esta preparada para ser utilizada por niños de muy corta edad. Se diferencia de la piscina grande en el sistema de recirculación de agua (en este casos e utilizarán skimmers) y en el tratamiento posterior, ya que se utilizará el sistema de hidrólisis salina para su desinfección en lugar de añadir directamente cloro. Este sistema de tratamiento del agua no es tan agresivo como el tradicional, siendo cada vez mas utilizado en piscinas de mediano y pequeño tamaño. El jacuzzi es de grandes dimensiones, disponiendo de su propio sistema de tratamiento de agua, utilizando el sistema de hidrólisis salina como en el caso de la piscina infantil. El edificio de servicios dispone de zonas de bar, enfermería y aseos. Se ha descrito desde la estructura a las instalaciones, justificando las conclusiones a través de los cálculos correspondientes, bien de forma manual, bien utilizando diverso software especializado.

SUMMARY IL-1R and TLRs are key players in innate immunity and inflammation. Tollip was identified as a component of IL-1RI, TLR2 and TLR4 signaling complexes that activate NF-κB and MAP kinase pathways. Tollip was previously shown as a negative regulator of NF-κB and MAP Kinase activation. We have characterized the role of Tollip in IL-R/TLRs induced signaling by the analysis of the Tollip deficient mice. We showed that NF-κB and MAPK (p38, JNK, or ERK1/2) signaling appeared normal in Tollip deficient cells following stimulation with IL-1β, lipopolysaccharide (LPS), and other TLR ligands. Also IL-1β and TLRs ligands induced activation of immune cells was indistinguishable from wild-type cells. Strikingly, in Tollip deficient mice the production of the inflammatory cytokines, IL-6 or TNF-α was significantly reduced relative to control mice after treatment with physiological doses of IL-1β or LPS, whereas no difference was observed at high doses of stimulation with LPS or in LPS induced septic shock. Therefore, Tollip could be critical for regulation of optimal responses to IL-1β and LPS, in addition to its role as negative regulator of the signaling. We also studied the role of Tollip as an endocytic adaptor for IL-1R endocytosis. We could show that Il-1R is ubiquitinated after IL-1β stimulation, and that Tollip's CUE domain binds IL-1RI in an ubiquitin-dependent manner. We followed IL-1R internalization and Tollip localization by confocal microscopy. Consistent with a role for Tollip in sorting of ubiquitinated IL-1RI, a significant amount of Tollip was also localized at the late endosomal compartment. We could show that Tollip is required for efficient lysosomal targeting of ubiquitinated IL-1R1, In the absence of Tollip or in Tollip deficient cells reconstituted with a Tollip mutant (defective in ubiquitin binding) IL-1RI accumulates in enlarged late endosomes. In addition, Tollip was shown to interact with, another endocytic adapter, Toml, and both interact with IL-1RI. In conclusion, we showed that Tollip is required for IL-1β and LPS signaling for cytokine production. In addition we showed and that Tollip has a role as an endocytic adapter, necessary for efficient trafficking and lysosomal degradation of IL-1RI. Resumé Le récepteur à l'interleukine-1 (IL-1R) et les récepteurs "Toll-like" (TLRs) sont des acteurs cruciaux de la réponse immunitaire innée et de l'inflammation. La proteine Tollip a été identifiée comme étant un élément des complexes de signalisation, induits par les récepteurs IL-1RI, TLR-2 et TLR-4, qui mènent à l'activation de la voie des MAP kinases et de NF-κB. Dans de précédentes études, il a été montré que Tollip pouvait inhiber ces deux voies de signalisation. Nous avons voulu caractériser plus précisément le rôle de Tollip dans l'activation des voies de signalisation mitées par IL-1R/TLRs en utilisant une lignée murine déficiente pour la protéine Tollip. Ainsi, en absence de Tollip, les cascades d'activation de NF-κB et MAPK (p38, JNK, or ERK1/2) ne semblent pas affectées après stimulation avec IL-1β, lipopolysaccharide (LPS) ou d' autres ligands des TLR. La réponse des cellules du système immunitaire induite par la stimulation avec IL-1β et les ligands des TLR est également comparable entre les souris sauvages et les souris deficientes pour Tollip. Par contre, dans cette lignée murine, la production de cytokines proinflammatoires IL-6 et TNFα induite par la stimulation à dose physiologique de IL-1β or LPS, est réduite. Cependant, lors de stimulation à plus hautes doses de LPS ou pendant un choc septique induit par de LPS, cette réduction n'est pas observée. Ces résultats montrent que Tollip pourrait avoir un rôle déterminant dans l'activation optimale en réponse à l' IL-1β et au LPS qui s'ajoute à sa fonction inhibitrice des mêmes voies de signalisation. Nous avons aussi étudié le rôle de Tollip comme molécule adaptatatrice du mécanisme endocytique d'internalisation de l' IL-1RI. Ainsi, l' IL-1R est ubiquitiné après stimulation par l' IL-1β , permettant à Tollip de se lier au récepteur. Cette interaction est réalisée entre le domaine CUE de Tollip et l'IL-1R via l'ubiquitine. L'internalisation et la localisation intracellulaire de l'IL-1RI et de Tollip ont été observés par microscopie confocale. En accord avec le rôle de Tollip dans le triage et la recirculation des IL-1R ubiquitiné, une quantité importante de Tollip été détectée dans l' endosome tardif. Nous avons pu démontrer que Tollip était nécessaire pour diriger efficacement ubiquitiné vers les lysosomes. Dans des cellules déficientes pour Tollip, ou reconstituées avec un mutant de Tollip (MF/AA) incapable de lier l'ubiquitine, IL-1RI s'accumule dans des vesicules anormales de l'endosome tardif. Dans ce travail, nous avons pu confirmer et préciser la fonction de la protéine Tollip dans l' activation de la production de cytokines induites par l' IL-1p and le LPS lors de l'inflammation et découvrir son rôle d'adaptateur dans l' internalisation et l'endocytose de l' IL-1RI.

The fiber recovery process is an essential part of the modern paper mill. It creates the basisfor mill's internal recirculation of the most important raw materials ¿ water and fiber. It is normally also a start point for further treatment of wastewater and if it works efficiently, it offers excellent basis to minimize effluents. This dissertation offers two different approaches to the subject. Firstly a novel save-all disc filter feeding system is developed and presented. This so-called precoat method is tested both in the laboratory and full-scale conditions. In laboratory scale it beats the traditional one clearly, when low freeness pulps are used as a sweetener stock. The full-scale application needs still some development work before it can be implemented to the paper mills. Secondly, the operationenvironment of save-all disc filter is studied mostly in laboratory conditions.The focus of this study is in cases, where low-freeness pulps are used as a sweetener stock of save-all filter. The effects of CSF-value, pressure drop, suspension consistency and retention chemicals to the quantity and quality of the filtrate was studied. Also the filtration resistance of the low freeness pulps was one studied.

Työssä selvitettiin perälaatikon muuttujien kuten huuliraon, huulisuihkun osumakohdan rintapöydälle, ohivirtauksen ja sivuvirtausten määrän sekä suihkusuhteen vaikutus formaatio-, neliömassa- ja kuituorientaatioprofiileihin. Analysoitiin huulisuureiden muutosten vaikutus rainanmuodostuksessa. Formaatiotasoa seurattiin koneella olevan vianilmaisinjärjestelmän mittauksen avulla ja kuituorientaatiota paperin poikkiratanäytteistä tehdyistä laboratoriomittauksista. Vianilmaisinjärjestelmän formaatiomittaus todettiin toimivaksi tarvittavan versiopäivityksen jälkeen. Selvitettiin huulisuureiden säädettävyyden ikkuna ja mahdolliset kehityskohteet. Kehitettäessä optimiprofiilien ajomallia raskaammille neliömassoille havaitsimme investointitarpeet kuten jäykempi huulilista perälaatikolle, määrämittaukset sivuvirtauksille ja tarvittavat parannukset kosteusprofiilin säätöön.

Työssä käsitellään hule- ja suotovesien hallintajärjestelmän kehittämistä Ämmässuon nykyisellä kaatopaikalla. Hallintajärjestelmää kehitettäessä on huomioitu ympäristölliset tekijät kuten topografia, sadanta, pinta- ja pohjavesiolosuhteet, maa- ja kallioperä sekä kaatopaikkaa ympäröivä alue. Vallitseva lainsäädäntö ja kaatopaikkaa koskevat luvat on selvitetty ennen hallintajärjestelmän kehittämisen suunnittelua. Kaatopaikkojen hule- ja suotovesien hallintajärjestelmän tärkeimpinä osina ovat olleet hule- ja suotovesien määrät ja niiden muutokset esim. kaatopaikan täyttösuunnitelman tai viimeistelysuunnitelman muutoksen vaikutuksesta. Määräarvioiden jälkeen on selvitetty vesien keräily, tasaus ja johtamistavat. Näitä osa-alueita suunniteltaessa on määritetty olemassa olevat rakenteet ja tarvittava lisärakennustarve. Myös mahdolliset poikkeustilanteet, kuten heikentynyt huleveden laatu, on huomioitu johtamisjärjestelyitä suunniteltaessa. Tämän lisäksi hallintajärjestelmään on liitetty suotoveden erillis- ja esikäsittelytarpeet sekä mahdollinen kierrättäminen takaisin jätetäyttöön paremman biohajoavuuden aikaan saamiseksi. Vesien keräilyn, virtauksen tasaamisen ja johtamisen lisäksi hallintajärjestelmään kuuluvat mittaavat ja ohjaavat automaatiojärjestelmät sekä tiedonkeräily- ja käsittelyjärjestelmät. Varsinainen mittaus- ja automaatiosuunnittelu on jätetty tämän työn ulkopuolelle. Työhön on lisäksi liitetty aikataulu- ja kustannustarkastelu. Näin on mahdollista varautua järjestelmän rakenteellisten osien toteuttamiseen sekä niistä aiheutuviin kustannuksiin

La transfusion de concentrés plaquettaires, dès 1958, a permis d'augmenter l'espérance et la qualité de vie des patients dans le domaine de l'onco-hématologie, notamment en réduisant les risques hémorragiques induits par les chimiothérapies intensives. Après le traumatisme de l'affaire du sang contaminé dans les années 1980-1990, la médecine transfusionnelle a adopté des exigences de sécurité et de qualité très strictes et régulées dans plusieurs pays par les instances politiques. Cependant même les mesures de qualité les plus strictes n'ont permis d'atteindre le risque « zéro », notamment en ce qui concerne les contaminations bactériennes. De plus, la prise de conscience de l'existence de nouveaux pathogènes (West Nile Virus, Chikungunya, Prions) a stimulé le développement de 3 techniques d'inactivation des pathogènes pouvant s'appliquer au plasma et aux concentrés plaquettaires : la technique INTERCEPT utilisant l'amotosalen/UVA, la technique MIRASOL avec la riboflavine/UV et la technique THERAFLEX avec les UVC. La Suisse a fait office de pionnière en étant le premier pays au monde à adopter de manière généralisée l'inactivation des pathogènes par la technique INTERCEPT pour les concentrés plaquettaires dès 2011 après sa validation par Swissmedic en 2009 et son implémentation réussie dans plusieurs centres de transfusion pilotes. Coïncidence? Le décès tragique d'un patient pédiatrique en 2009 suite à une contamination bactérienne par une transfusion de concentré plaquettaire a précédé cette décision. Les cliniciens ont besoin de disposer de concentrés plaquettaires sûrs d'un point de vue microbiologique mais également sur le plan hémostatique, d'où la nécessité de disposer de preuves solides sur l'efficacité thérapeutique de ces nouveaux produits. Ceci a fait l'objet de la revue publiée dans Blood Reviews « The clinical and biological impact of new pathogen inactivation technologies on platelets concentrates » dont l'originalité est de couvrir l'étude de l'effet des processus d'inactivation des pathogènes sur la fonction plaquettaire sous toutes ses facettes allant de la protéomique aux études d'hémovigilance. Ce travail montre l'efficacité de ces méthodes et leur sécurité et souligne que l'observation de taux de recirculation moindre peut être compensée par une augmentation du statut d'activation des plaquettes. Le deuxième article publié comme co-auteur dans le journal Blood Transfusion « In vitro evaluation of pathogen-inactivated buffy coat-derived platelet concentrates during storage: The psoralen-based photochemical treatment step-by-step » se pose la question de la modification des propriétés fonctionnelles des plaquettes dans une étude à deux bras (par comparaison entre plaquettes traitées et non traitées). En plus de deux tests utilisés en pratique clinique (agrégation plaquettaire et cytométrie de flux) un test expérimental d'adhésion plaquettaire au fibrinogène en condition statique a été développé en collaboration avec le Prof Angelillo-Scherrer dans le cadre du laboratoire de recherche et développement du SRTS-VD. Les résultats obtenus démontrent la conservation du métabolisme plaquettaire et des changements mineurs dans la capacité d'agrégation mais une augmentation de la capacité d'adhésion au fibrinogène des plaquettes traitées probablement via une augmentation de la conversion de l'intégrine ailb(B3 dans sa forme activée. Les techniques d'inactivation des pathogènes appliqués aux concentrés plaquettaires représentent un important progrès dans le domaine de la médecine transfusionnelle. Leur impact au niveau moléculaire reste cependant encore en partie inconnu et fait l'objet d'études. Le défi actuel consiste à réussir à les adopter aux concentrés érythrocytaires, ce qui révolutionnerait la médecine transfusionnelle.

The extravasation of leukocytes from the blood stream into the tissues is a prerequisite for adequate immune surveillance and immune reaction. The leukocyte movement from the bloodstream into the tissues is mediated by molecular bonds. The bonds are formed between adhesion molecules on endothelial cells and their counterparts expressed on leukocytes. Vascular adhesion protein-1 (VAP-1) is an endothelial adhesion molecule mediating leukocyte interactions with endothelium. It is also an enzyme having semicarbazide sensitive amine oxidase (SSAO) activity. The SSAOactivity catalyses deamination of primary amines into corresponding aldehyde and during the enzymatic reaction hydrogen peroxide and ammonia are produced. The aim of this study was to investigate the relationship between the adhesive and enzymatic activities of VAP-1. The role of VAP-1 in leukocyte traffic was studied in vivo under normal and pathological conditions in VAP-1 deficient mice. The results from in vitro flow-based assays indicated that VAP-1 uses both SSAOactivity and its adhesive epitope to bind leukocytes, and both are perquisites for VAP-1 mediated adhesion. Furthermore, in vivo results demonstrated that leukocyte trafficking was impaired in vivo by deleting VAP-1 or inhibiting SSAO-activity. There was impairment in lymphocyte recirculation as well as leukocyte accumulation into the inflamed area. Moreover, the VAP-1 deficient mice did not show generalized defects in antimicrobial responses, whereas significant reduction in tumor progression and neovascularization was observed. These results indicate that VAP-1 could be used as a target in anti-adhesive therapies either by blocking its adhesive epitope with antibodies or by inhibiting its SSAO-activity using inhibitors. Moreover, targeting of VAP-1 may provide a new way of inhibiting neovascularization in tumors.

The amount of water available is usually restricted, which leads to a situation where a complete understanding of the process, including water circulations and the influence of water components, is essential. The main aim of this thesis was to clarify the possibilities for the efficient use of residual peroxide by means of water circulation rearrangements. Rearranging water circulations and the reduction of water usage may cause new problems, such as metal induced peroxide decomposition that needs to be addressed. This thesis introduces theoretical methods of water circulations to combine two variables; effective utilization of residual peroxide and avoiding manganese in the alkaline peroxide bleaching stage. Results are mainly based on laboratory and mill site experiments concerning the utilization of residual peroxide. A simulation model (BALAS) was used to evaluate the manganese contents and residual peroxide doses. It was shown that with optimum recirculation of residual peroxide the brightness can be improved or chemical costs can be decreased. From the scientific perspective, it was also very important to discover that recycled peroxide was more effective pre-bleaching agent compared to fresh peroxide. This can be due to the organic acids i.e. per acetic acid in wash press filtrate that have been formed in alkaline bleaching stage. Even short retention time was adequate and the activation of residual peroxide using sodium hydroxide was not necessary. There are several possibilities for using residual peroxide in practice regarding bleaching. A typical modern mechanical pulping process line consist of defibering, screening, a disc filter, a bleach press, high consistency (HC) peroxide bleaching and a wash press. Furthermore there usually is not a particular medium consistency (MC) pre-bleaching stage that includes additional thickening equipment. The most advisable way to utilize residual peroxide in this kind of process is to recycle the wash press filtrate to the dilution of disc filter pulp (low MC pre-bleaching stage). An arrangement such as this would be beneficial in terms of the reduced convection of manganese to the alkaline bleaching stage. Manganese originates from wood material and will be removed to the water phase already in the early stages of the process. Recycling residual peroxide prior to the disc filter is not recommended because of low consistencies. Regarding water circulations, the novel point of view is that, it would be beneficial to divide water circulations into two sections and the critical location for the division is the disc filter. Both of these two sections have their own priority. Section one before the disc filter: manganese removal. Section two after the disc filter: brightening of pulp. This division can be carried out if the disc filter pulp is diluted only by wash press filtrate before the MC storage tower. The situation is even better if there is an additional press after the disc filter, which will improve the consistency of the pulp. This has a significant effect on the peroxide concentration in the MC pre-bleaching stage. In terms of manganese content, it is essential to avoid the use of disc filter filtrate in the bleach press and wash press showers. An additional cut-off press would also be beneficial for manganese removal. As a combination of higher initial brightness and lower manganese content, the typical brightness increase varies between approximately 0.5 and 1% ISO units after the alkaline peroxide bleaching stage. This improvement does not seem to be remarkable, but as it is generally known, the final brightness unit is the most expensive and difficult to achieve. The estimation of cost savings is not unambiguous. For example in GW/TMP mill case 0.6% ISO units higher final brightness gave 10% savings in the costs of bleaching chemicals. With an hypothetical 200 000 ton annual production, this means that the mill could save in the costs of bleaching chemicals more than 400 000 euros per year. In general, it can be said that there were no differences between the behavior of different types of processes (GW, PGW, TMP and BCTMP). The enhancement of recycling gave a similar response in all cases. However, we have to remember that the utilization of residual peroxide in older mills depends a great deal on the process equipment, the amount of water available and existing pipeline connections. In summary, it can be said that processes are individual and the same solutions cannot be applied to all cases.

Destruction of Padron® (dye and picloram) was evaluated using a photoreactor and a solar reactor. Photolysis was observed using only a germicide lamp (GL). Black light (BL) and H2O2 (172 mmol L-1) promoted a conversion of 49% and 6% of dye and picloram, respectively. Photocatalytic processes were more efficient using TiO2/GL (96%-dye; 60%-picloram) than TiO2/BL (44%-dye; 40%-picloram). Photolysis using sunlight was not observed during PadronÒ recirculation in the reactor constructed with four borosilicate tubes. Meanwhile, adding H2O2 resulted in 12% conversion of dissolved organic compounds. Finally, the most efficient mineralization (60%) was obtained using the Fenton reaction ( H2O2-176 mmol L-1; FeSO4x6H2O-90 mmol L-1) and sunlight.

The main aim of this study was to inspect the influence of the ultrafiltration implementation on the washing and on bleaching efficiency. Four cases corresponding to four washing stages were observed: two with hardwood pulp and two with softwood pulp; each case had a reference and a trial experiment. The experiments with hardwood pulp were arranged in a manner to explore predominantly the possibility of bleaching performance improvement by applying for washing instead of untreated filtrate (reference case) the same treated one (trial case). Despite that the ultrafiltration reduced the COD of the wash filtrates allowing the decreasing of COD carry-over to the bleaching stage it didn’t affect the bleaching performance. Another set was used in the experiments with softwood pulp. It implied the ultrafiltration and recirculation of the filtrate to the same washing stage with the purpose to reduce the volumes and pollution of the bleaching effluents. In one case the negative result was obtained which was expressed by worse parameters of the pulp after bleaching. Another case showed the opportunity to replace hot water with the filtrate and reduce the fresh water consumption.

Nitrous oxide emissions from an activated sludge plant which serves a research institute in Rio de Janeiro city were estimated from six unit processes (grit tank, sand trap, aeration tank, secondary settling tank, sludge recirculation line and aerobic digester sludge tank) and also from the plant effluent. Total estimated annual flux was 3.2 x 10(4) g N2O yr-1 of which about 90% was from the aeration tank. Emission factors estimated from population served, wastewater flow and nitrogen load (conversion ratio) were 13 g N2O person-1 yr-1, 9.0 x 10-5 g N2O Lwastewater-1 and 0.14%.
