O âmbito deste trabalho reside no contributo da Farmacovigilância para um Uso Racional do Medicamento, focando o relevante papel do farmacêutico nestes dois campos. A Farmacovigilância tem uma actuação a nível mundial, com um objectivo bem claro, o uso seguro do medicamento. Tem-se verificado uma evolução ao longo dos séculos, tendo sido a ocorrência de tragédias o impulso para a implementação de Sistemas de Farmacovigilância, a tragédia que ocorreu com a administração de talidomida a mulheres grávidas foi um dos grandes marcos para a criação do Sistema Internacional de Farmacovigilância. A Reacção Adversa a Medicamento(s) (RAM) é uma consequência do risco subjacente à utilização de fármacos, estando por isso a Notificação Espontânea (NE) de RAMs na base da Farmacovigilância. Todos os que contactam com tratamentos terapêuticos devem notificar, falamos portanto de profissionais de saúde, da indústria e dos utentes. Não menosprezando qualquer efeito adverso que decorra da utilização de um medicamento, deve-se notificar todas as RAMs estando elas já reconhecidas ou não. O Uso Racional do Medicamento pressupõe a intervenção do médico, do farmacêutico e do utente, havendo o objectivo bem claro através de várias medidas implementadas e do apoio dos Estudos de Utilização de Medicamentos (EUM) de uma prescrição, uma dispensa e um uso correctos. O farmacêutico é parte activa da Farmacovigilância e do Uso Racional do Medicamento. O farmacêutico tem contacto não só com a prescrição como com a dispensa e com o evoluir do tratamento terapêutico, sendo portanto um monitorizador. Devido à proximidade do dia-a-dia entre farmacêutico e doente cabe a este profissional de saúde educar para um uso correcto e seguro do medicamento, transmitindo sempre um elevado nível de confiança e fiabilidade.

Many aspects of the conditions required to maximize the ewe's response to ram introduction in the late anoestrous season remain unclear. The aim of this research was to determine whether grazing space allowances could influence the efficacy of the ram effect. In August 1995, at Reading (latitude 51degrees27'N), following a 3-month isolation period from rams, two groups of nulliparous Mule ewes, aged 15 months, were introduced to four rains in a low (12 ewes/ha; treatment L, n = 124) or in a high stocking rate (84 ewes/ha; treatment H, n = 126). From the beginning of August until the end of August oestrous behaviour was recorded by daily checks of mating marks on ewes. Rams were removed and in October all ewes were scanned (day 50) for pregnancy. No significant differences were found in the parameters investigated. Eighty-two percent of the L and 75.4% of the H ewes exhibited oestrus, with a pronounced peak on day 23 following ram introduction and a compact concentration in the 21-25-day period. The oestrous synchronisation rate in this 5-day period was 69.4 and 68.3%, respectively for L and H. The mean interval from ram introduction to oestrus was 23.17+/-2.4 days in L and 23.0+/-2.2 days in the H group. Conception rates were 84.3 and 87.4% for L and H groups, respectively. These results suggest that the response of anoestrous ewes to the introduction of rams was not affected by grazing space allowances and that yearling Mule ewes respond well to the ram effect in the late anoestrus season. It is accepted that an important source of variation in the response of anoestrous ewes, to the introduction of rams, is the intensity of male stimulation. The aim of this study was to investigate strategies capable of increasing the impact and transmission of the ram stimuli. In Experiment 1, two groups of seven ewes (Bluefaced Leicester male x Swaledale female) were individually penned with one ram and for the next 6 h the rams either remained in the pen or were replaced hourly. Blood samples revealed no difference in the pattern of plasma LH secretion. In Experiment 2, three groups of 16 ewes were either introduced to one ram, individually (H) or in groups of 8 (L), or remained isolated. Ram introduction increased the plasma LH pulsatility (P < 0.001). H ewes displayed more (nine versus six) male-induced LH pulses (pulses occurring within the first 45 min) and more pulses per 8 h intervals than the L group of ewes (1.9 +/- 0.3 versus 1.3 +/- 0.3), but these differences were not significant. It was concluded that (i) frequent replacement of rams within a few hours following ram introduction to ewes does not further improve the response of ewes, especially if the ram:ewe ratio is high; (ii) the characterization of the plasma LH secretion parameters during a period of 6-8 h does not seem to be an effective method to detect small differences in the intensity of stimulation received by the ewes when exposed to rams; (iii) North Country Mule ewes (Bluefaced Leicester male x Swaledale female) in the UK respond to the presence of rams in spring (late oestrous/early anoestrous season) with an elevation in plasma LH secretion. The formulation and implementation of LEAF-2, the Land Ecosystem–Atmosphere Feedback model, which comprises the representation of land–surface processes in the Regional Atmospheric Modeling System (RAMS), is described. LEAF-2 is a prognostic model for the temperature and water content of soil, snow cover, vegetation, and canopy air, and includes turbulent and radiative exchanges between these components and with the atmosphere. Subdivision of a RAMS surface grid cell into multiple areas of distinct land-use types is allowed, with each subgrid area, or patch, containing its own LEAF-2 model, and each patch interacts with the overlying atmospheric column with a weight proportional to its fractional area in the grid cell. A description is also given of TOPMODEL, a land hydrology model that represents surface and subsurface downslope lateral transport of groundwater. Details of the incorporation of a modified form of TOPMODEL into LEAF-2 are presented. Sensitivity tests of the coupled system are presented that demonstrate the potential importance of the patch representation and of lateral water transport in idealized model simulations. Independent studies that have applied LEAF-2 and verified its performance against observational data are cited. Linkage of RAMS and TOPMODEL through LEAF-2 creates a modeling system that can be used to explore the coupled atmosphere–biophysical–hydrologic response to altered climate forcing at local watershed and regional basin scales.

Background Dermatosparaxis (Ehlers–Danlos syndrome in humans) is characterized by extreme fragility of the skin. It is due to the lack of mature collagen caused by a failure in the enzymatic processing of procollagen I. We investigated the condition in a commercial sheep flock. Hypothesis/Objectives Mutations in the ADAM metallopeptidase with thrombospondin type 1 motif, 2 (ADAMTS2) locus, are involved in the development of dermatosparaxis in humans, cattle and the dorper sheep breed; consequently, this locus was investigated in the flock. Animals A single affected lamb, its dam, the dam of a second affected lamb and the rams in the flock were studied. Methods DNA was purified from blood, PCR primers were used to detect parts of the ADAMS2 gene and nucleotide sequencing was performed using Sanger's procedure. Skin samples were examined using standard histology procedures. Results A missense mutation was identified in the catalytic domain of ADAMTS2. The mutation is predicted to cause the substitution in the mature ADAMTS2 of a valine molecule by a methionine molecule (V15M) affecting the catalytic domain of the enzyme. Both the ‘sorting intolerant from tolerant’ (SIFT) and the PolyPhen-2 methodologies predicted a damaging effect for the mutation. Three-dimensional modelling suggested that this mutation may alter the stability of the protein folding or distort the structure, causing the protein to malfunction. Conclusions and clinical importance Detection of the mutation responsible for the pathology allowed us to remove the heterozygote ram, thus preventing additional cases in the flock.

The scavenging processes of chemical species have been previously studied with numerical modeling, in order to understand the gas and particulate matter intra-reservoir transferences. In this study, the atmospheric (RAMS) and scavenging (B.V.2) models were used, in order to simulate sulfate concentrations in rainwater using scavenging processes as well as the local atmospheric conditions obtained within the LBA Project in the State of Rondonia, during a dry-to-wet transition season. Two case studies were conducted. The RAMS atmospheric simulation of these events presented satisfactory results, showing the detailed microphysical processes of clouds in the Amazonian region. On the other hand, with cloud entrainments, observed values have been overestimated. Modeled sulfate rainwater concentration, using exponential decay and cloud heights of 16 km and no entrainments, presented the best results, reaching 97% of the observed value. O Regional Atmospheric Modeling System (RAMS), versão 3b, foi utilizado para simular o escoamento na Região Metropolitana de Porto Alegre (RMPA) para explicar a influência de circulações clássicas e não clássicas devido as proximidades do oceano e da Lagoa dos Patos, e da ilha de calor formada pelas cidades. O RAMS foi utilizado em um domínio tridimensional com duas grades sendo utilizadas, a grade mais grossa com resolução horizontal de 8 Km, e a grade mais fina com resolução horizontal de 2 Km. A inicialização do modelo é homogênea e parte do repouso, em uma situação de verão. Foram realizados 3 experimentos; o primeiro deles com apenas a grade mais grossa e com a parametrização da vegetação sendo grama curta em todo o domínio; o segundo utilizando as duas grades, sendo que a área coberta pela grade mais fina é parametrizada como deserto, nas duas grades; o terceiro diferencia-se do segundo pela adição de uma forçante térmica no primeiro nível da área coberta pela grade mais fina durante o período da noite (após o pôr do sol). Os resultados mostram que circulação de brisa lacustre/terrestre é responsável por uma forte tendência meridional no escoamento resultante sobre a RMPA, só sendo atenuada pela componente zonal da circulação de brisa marítima à partir do final da tarde. A confluência dessas duas circulações provoca uma tendência a elevação da camada de mistura. A presença da ilha de calor aumenta o movimento convectivo, com maior transporte vertical de calor sensível, e também retardou o avanço da frente de brisa marítima, embora durante a noite a sua influência não tenha sido muito significante.

O objetivo geral deste trabalho é desenvolver estudos relacionados à dispersão de poluentes, considerando a queima de carvão para geração de energia nas Usinas Termoelétricas de Charqueadas e São Jerônimo. O período de estudo foi do dia 17 a 23 de junho de 2003. Neste período houve a passagem de um sistema frontal. Sendo possível avaliar a dispersão dos poluentes em condições pré-frontal, frontal e pós-frontal. Para simular o comportamento dos poluentes neste período, foi utilizada uma subrotina de dispersão acoplada ao modelo RAMS (Regional Atmospheric Modeling System). Os resultados mostraram que nos dias classificados como pré-frontal e pós-frontal as concentrações do material particulado, dióxido de enxofre e do óxido de nitrogênio, atingiram seus valores máximos, pelo fato da umidade relativa do ar estar bastante baixa em torno de 60%, pressão atmosférica da ordem de 1021 hPa e a intensidade dos ventos fraca. No dia classificado como frontal, as concentrações estavam praticamente nulas, devido à passagem do sistema frontal que causou a queda na pressão atmosférica, aumento da umidade relativa e também pelo fato da ocorrência de precipitação atmosférica neste dia. As comparações dos resultados simulados, com os dados observados na estação de qualidade do ar mostraram-se satisfatórios. Com exceção do dia 22 de junho, que apresentou uma diferença da ordem de 90%.

O objetivo central deste trabalho consiste, a partir de um caso de ciclogênese ocorrido na costa leste do América do Sul, caracterizar as condições sinóticas favoráveis ao desenvolvimento de ciclones marítimos que possam gerar ressacas. Uma ciclogênese desenvolveu-se a leste da Argentina dia 03 de maio de 2001. Neste mesmo dia sobre o continente, existia um centro de alta pressão, que em conjunto com este ciclone marítimo formava um pista de vento de quadrante sul, paralelo ao continente, deixando o mar revolto, aumentando nível no extremo sul do Brasil, como registrado pela tábua de maré da cidade de Rio Grande, no estado do Rio Grande de Sul. No dia 04 de maio, à leste do estado de São Paulo, outro ciclone começa a se formar, intensificando a pista de vento pré existente. A metodologia utilizada afim de atingir os objetivos propostos, foi a da modelagem numérica. Utilizou-se modelo Regional Atmospheric Modeling System (RAMS). Foram utilizados como entrada dados de outros dois modelos, modelo Global e Modelo ETA. Para ambos os experimentos usando o RAMS, observou-se que o ciclone do dia 04 teve sua gênese em torno de 25ºS e 45ºW, se deslocando para sudeste. Na análise de mesoescala, foi observado que durante o período estudado os ventos sempre tiveram uma componente de sul e que os mais intensos ventos ocorreram em 28ºS e 48ºW, pois nesta região haviam maiores valores de gradiente de pressão, fluxo de calor sensível e fluxo de calor latente, já que é também nesta região onde se verifica a maior temperatura da superfície do mar.

The nutritive value of elephant grass silages, with increasing levels of dehydrated cashew stalk (DCS) was evaluated. Five addition levels were studied (0: 4: 8: 12 and 16%), using 20 rams in a completely randomized design with five treatments (addition levels) and four replicates (animals). Intake and digestibility of dry matter (DM), organic matter (OM). crude protein (CP), neutral detergent fiber (NDF), acid detergent fiber (ADF), ether extract (EE), total carbohydrates (TC) and non-fibrous carbohydrates (NFC) were determined, as well as total digestible nutrients (TDN) and silages nitrogen balance (NB) DCS did not affect intakes of DM, OM, NDF, ADF, TC and TDN, neither digestibility of OM, CB, NDF ADF and TC silages. Otherwise. the addition of DCS increased intakes of CP EE and NFC and digestibility of EE, NFC, as well as TON values and nitrogen balance. Inclusion of DCS, showed linear decreasing digestibility of ADF It is concluded that addition of DCS to silages of elephant grass up to 16% on a natural basis can be used as a forage source for ruminants.

The aim of this study was to investigate the histopathological changes in reproductive system (testicles, epididymis, seminal vesicles, and prostate) of small male ruminants after Toxoplasma gondii infection. Eight sheep were inoculated with T. gondii: group I, four sheep (2.0 x 10(5) P-strain oocysts); group II, four sheep (1.0 x 10(6) RH-strain tachyzoites); and group III, two uninfected sheep maintained as control. Infection with T. gondii was confirmed by seroconversion (indirect fluorescent antibody test-IgG) in all the infected animals beginning on post-inoculation day (PID) 7. on PID 70, all the animals were euthanized and tissue samples (testicles, epididymis, seminal vesicles, and prostate) were collected and processed for histological analysis. The main changes detected were a focal mononuclear interstitial inflammatory infiltrate in the prostate and seminal vesicles; diffuse testicular degeneration associated with calcification foci and a multifocal mononuclear interstitial inflammatory infiltrate; and a mononuclear interstitial infiltrate and focal necrotic areas of the muscle fibers surrounding the seminal vesicles. The histopathological findings of this work, along with the detection of T. gondii in the examined parenchyma tissues (immunohistochemistry) and the results obtained by other authors examining different tissues, suggest that histological changes diagnosed in the reproductive system of rams infected with T. gondii are strongly suggestive of toxoplasmatic infection.

The aim of this study was to analyze the anatomotopographic location of the mandibular foramen in the right and left ramus, and to verify the influence of the amount of dental alveoli on the foramen position. Thirty-five adult dry human mandibles of Araraquara Dental School, UNESP - São Paulo State University were assessed, with or without dental alveoli. Measurements were obtained, using a ruler and a digital caliper. The following distances were measured: Fl - distance between the lowest point of the mandibular incisure and the mandibular foramen (F point); FB - distance between the mandibular base and F point; FP - distance between the posterior margin of the ramus and F point; FA - distance between the anterior margin of the ramus and F point; FT - distance between the apex of the retromolar trigone and F point. The Mann-Whitney test was used to compare each measurement according to hemi-arch, and the Kruskal-Wallis test was used to analyze the influence of the presence of alveoli on the measures. For multiple comparison, Dunn's method was used. There was no statistically significant difference in the location of the mandibular foramen when compared to the right and left hemi-arches. The amount of dental alveoli influenced, significantly, only on FA and FP distances. Thus, it was concluded that the right and left mandibular ramus showed symmetry in the location of the mandibular foramen, and the amount of alveoli influenced on the distances of the anterior and posterior margins of the mandibular rams, in relation to the mandibular foramen.
