Os autores determinaram o diâmetro médio dos granulomas de fígado de camundongos infectados com cercárias de duas cepas geográficas bem definidas do, Schistosoma mansoni (LE, Belo Horizonte e SJ, São Paulo). No total foram medidos 1.170 granulomas. Os granulomas com 60 dias eram de dimensões maiores do que os de 90 dias. A modulação da resposta imunopatológica foi significativamente mais eficiente na cepa LE e os granulomas, tanto aos 60 como aos 90 dias, da cepa SJ eram significativamente maiores. Os dados obtidos indicam uma maior patogenicidade da cepa SJ. Especula-se se o significado destes achados poderiam, em parte, explicar a ocorrência das variações regionais das formas anatomo-clínicas da esquistossomose.

In vivo flushing of the ear canals with 50 to 100 ml collected all larvae in 100% of infested cattle. All adult mites were collected from 22 ou 27 parasitized ears (81.5%). Both stages adult and larva were collected from 18 out 22 parasitized ears (82%). The technique is adequate as a measure of prevalence, incidence, density/intensity of infestation as well as in vivo diagnosis. No damage to the host ear has been noticed.

Raillietia flechtmanni sp. n. is described from the ear canal of its type host the domestic buffalo. The new species parasitizes cattle as well in Brazil.

The life cycle of ear mites of the genus Raillietia Trouessart consists of egg, larva, proto-and deutonymph and adult. The proto-and deutonymph are free living, non feeding instars. The teneral adult is the transfer stage. The minimum period required for completion of the life cycle is approximately eight days.

The compound Ro-15.5458/000, derivative in the class of 9-acridanone-hydrazones, was found to be effective against Schistosoma mansoni in mice, killing almost all the skin schistosomules (24 hr after infection), when administered at the dose of 100 mg/kg. In experiments carried out with Cebus monkeys, the drug was shown to be fully effective at 25 mg/kg, 7 days after infection. These data, associated with the good results obtained earlier at the post-postural phase of schistosomiasis, allow the inference that this promising compound may be important in the set of antischistosomal drugs, depending on further toxicological and clinical tests.

The concomitant immunity in the presence of repeated infections (with 15 cercariae) was studied in mice sacrificed on the 20th day after each infection. The comparison of the averages of immature worms, recovered from mice submitted to reinfection, with those of their respective controls (previously uninfected) showed a significantly lower worm recovery rate in the animals with previous infections (concomitant immunity). However, statiscally significant differences could not be detected among the various groups of animals, when the mice that accumulated worms in this mature stage were perfused. The theoretical projection based on the accumulation of young worms which developed to adult ones indicates a lower recovery rate of adult worms in the animals with concomitant immunity, but this projection was not corroborated by the experimental data. The visceral hemodynamic alterations that occurred in reinfections due to the pathogeny, favouring recirculation of the recent arriving worms to the other organs on the occasion of perfusion of the portal system. These results suggest that special care should be taken when one wants to investigate concomitant immunity in mice based on the distinction of the immature worms from challenge infection and the mature ones from primary infection.

To verify the prevalence of infection by human immunodeficiency virus (HIV) in alcoholics we studied 131 alcoholic patients (119 males and 12 females) with a mean age of 44.3 ± 10.8 years. Serum samples were collected from this group and analysed, by ELISA, for antibodies against HIV as well as for serological markers for hepatitis B virus (HBV) and hepatitis C virus (HCV). As we have previously described, we found a high prevalence of HBV (26.4%) and HCV (4.2%) markers as compared to the prevalence of these markers in samples of normal blood donors from Uberlândia's Hemocentro Regional, which are 4% and 0.4%, respectively. Of the 131 patients, four (3%) had antibodies against HIV, three (75%) of which were injecting drug users (IDU). In the HIV-negative group, only one patient was an IDU. The prevalence of HIV in our population, according to data from the city's Health Secretary, varies from 3.1% to 6.2%. We conclude that, at least for the moment, alcoholism per se, did not constitute an important risk factor for HIV infection. However, acquired immunodeficiency syndrome is a rather recent disease as compared to hepatitis B and C and, as the transmission routes are similar for HIV and hepatitis viruses, an increase in the incidence of HIV infection in alcoholics may be just a question of time.

Determination of seric levels of adenosine deaminase (ADA), an enzyme produced by monocytes/macrophages and lymphocytes, has been used in the diagnosis of human tuberculosis (TB). In the present study, ADA seric activity was evaluated comparatively to the comparative tuberculin test in the diagnosis of bovine tuberculosis. Two hundred fifty-six cattle were classified by origin and by the comparative tuberculin test as TB-positive animals (n = 52, from herds where the Mycobacterium bovis had previously been isolated), and TB-negative animals (n = 204, TB-free herds). The mean ADA seric value from the TB-positive group (4.45 ± 2.33 U/L) was significantly lower (p = 0.008) than that observed in sera from the TB-negative group (6.12 ± 4.47 U/L). When animals from a herd with clinical cases of enzootic bovine leukosis of TB-negative group were withdrawn from analysis, the mean ADA seric values of TB-negative group (5.12 ± 3.75 U/L) was not significantly different anymore from that of the TB-positive group (p = 0.28). There was no agreement in the diagnosis of bovine TB between comparative tuberculin test and determination of ADA seric values, using two different cutoff points, being 6.12 U/L and 15.0 U/L, (kappa = -0.086 and kappa = -0.082, respectively). In conclusion, the determination of ADA seric activity was not a good auxiliary test for bovine TB, because it was not able to distinguish between TB-positive and TB-negative animals.

A series of seven limonene β-amino alcohol derivatives has been regioselectively synthesised in moderate to good yields. Two of these compounds were found to be significantly effective against in vitro cultures of the Leishmania (Viannia) braziliensis promastigote form in the micromolar range. The activities found for 3b and 3f were about 100-fold more potent than the standard drug, Pentamidine, in the same test, while limonene did not display any activity. This is the first report of antileishmanial activity by limonene β-amino alcohol derivatives.

Trypanosoma cruzi infection induces progressive cardiac inflammation that leads to fibrosis and modifications in the heart architecture and functionality. Statins, such as 3-hydroxy-3-methylglutaryl coenzyme A (HMG CoA) reductase inhibitors, have been studied due to their pleiotropic roles in modulating the inflammatory response. Our goal was to evaluate the effects of simvastatin on the cardiac inflammatory process using a cardiotropic strain of T. cruzi in a murine model of Chagas cardiomyopathy. C57BL/6 mice were infected with 500 trypomastigotes of the Colombian strain of T. cruzi and treated with an oral dose of simvastatin (20 mg/Kg/day) for one month and inflammatory and morphometric parameters were subsequently evaluated in the serum and in the heart, respectively. Simvastatin reduced the total cholesterol and inflammatory mediators (interferon-gamma, tumour necrosis factor-alpha, CCL2 and CCL5) in the serum and in the heart tissue at 30 days post-infection. Additionally, a proportional reduction in heart weight and inflammatory infiltration was observed. Simvastatin also reduced epimastigote proliferation in a dose-dependent manner in vitro and was able to reduce blood trypomastigotes and heart amastigote nests during the acute phase of Chagas disease in vivo. Based on these data, we conclude that simvastatin exerts a modulatory effect on the inflammatory mediators that are elicited by the Colombian strain of T. cruzi and ameliorates the heart damage that is observed in a murine model of Chagas disease.

Analisar a associação entre excesso de peso e diferentes fatores de risco familiares em adolescentes da região oeste do estado de São Paulo. Estudo transversal com 1779 adolescentes de ambos os sexos, e idade compreendida entre 11 e 17 anos. Calculou-se o índice de massa corporal e os fatores de risco familiares foram analisados por meio de questionário. O excesso de peso foi associado com o sexo masculino (RC=1,55 [1,22-1,97]), estudar em escola particular (RC=2,14 [1,56-2,94]) e maior escolaridade materna (RC=0,52 [0,33-0,83]). Iniciativas de combate à obesidade devem ser instauradas em meio escolar e atingir toda a estrutura familiar, bem como levar em consideração particularidades decorrentes do sexo.

Verificar a relação entre capacidade cardiorrespiratória e gordura corporal em mulheres. Foram analisadas 229 mulheres com idade entre 50 e 84 anos. Foi realizada avaliação antropométrica e calculada a razão cintura/quadril, bem como o índice de conicidade. A capacidade cardiorrespiratória foi avaliada pelo teste de caminhada de seis minutos. Com o pedômetro foi possível avaliar: velocidade, comprimento da passada e total de passos. Os resultados foram distribuídos em quartil, e a comparação feita pela análise de variância com post-hoc. A relação entre teste de caminhada de seis minutos e a antropometria foi feita pela correlação de Pearson. Os resultados revelaram que mulheres com capacidade cardiorrespiratória no quartil4 apresentaram idade estatisticamente menor (p<0,05) que as do quartil1. As medidas antropométricas apresentaram valores menores (p<0,05), quando comparados os resultados dos quartis3 e 4 com os quartis1 e 2. Conclui-se que mulheres com maior tecido adiposo e acúmulo de gordura central apresentam menor capacidade cardiorrespiratória.
