In order to assay the triatomine infestation and domiciliation in the rural area of Jaguaruana district, state of Ceará, Brazil, we studied, from November 2000 to April 2002, 4 localities comprising 158 domiciles as a whole, with an average of 4 inhabitants/house, who are dwelling in there for more than 7 years. Most houses have tile-covered roofs and the walls built with plaster-covered bricks (57%), followed by bricks without plaster (33%), and mud walls (7.5%). A total of 3082 triatomines were captured from different locations, according to the following capture plan: (a) intradomiciles: 238 Triatoma brasiliensis, 6 T. pseudomaculata, 9 Rhodnius nasutus, and 2 Panstrongylus lutzi; (b) peridomiciles (annexes): 2069 T. brasiliensis, 223 T. pseudomaculata, 121 R. nasutus, and 1 P. lutzi; (c) wild, in carnauba palms (Copernicia prunifera): 413 R. nasutus. From the captured triatomines, 1773 (57.5%) were examined. The natural index of Trypanosoma cruzi infection ranged from 10.8% to 30.2% (average of 17%), depending on the species and the location from where the triatomines were captured.

Rhodnius barretti , a new triatomine species, is described based on adult specimens collected in rainforest environments within the Napo ecoregion of western Amazonia (Colombia and Ecuador). R. barretti resembles Rhodnius robustus s.l. , but mitochondrial cytochrome b gene sequences reveal that it is a strongly divergent member of the “robustus lineage”, i.e., basal to the clade encompassing Rhodnius nasutus , Rhodnius neglectus , Rhodnius prolixus and five members of the R. robustus species complex. Morphometric analyses also reveal consistent divergence from R. robustus s.l. , including head and, as previously shown, wing shape and the length ratios of some anatomical structures. R. barretti occurs, often at high densities, in Attalea butyracea and Oenocarpus bataua palms. It is strikingly aggressive and adults may invade houses flying from peridomestic palms. R. barretti must therefore be regarded as a potential Trypanosoma cruzi vector in the Napo ecoregion, where Chagas disease is endemic.

Rhodnius prolixus, a blood-sucking triatomine with domiciliary anthropophilic habits, is the main vector of Chagas disease. The current paradigm of Trypanosoma cruzi transmission in Columbia includes a sylvatic and domiciliary cycle co-existing with domestic and sylvatic populations of reservoirs. The aim of this study is to evaluate the population densities and relative abundance of triatomines and mammals that may be involved in the sylvatic cycle of Chagas disease to clarify the epidemiological scenario in an endemic area in the province of Casanare. Insect vectors on Attalea butyracea palms were captured using both manual searches and bait traps. The capture of mammals was performed using Sherman and Tomahawk traps. We report an infestation index of 88.5% in 148 palms and an index of T. cruzi natural infection of 60.2% in 269 dissected insects and 11.9% in 160 captured mammals. High population densities of triatomines were observed in the sylvatic environment and there was a high relative abundance of reservoirs in the area, suggesting a stable enzootic cycle. We found no evidence of insect domiciliation. Taken together, these observations suggest that eco-epidemiological factors shape the transmission dynamics of T. cruzi, creating diverse scenarios of disease transmission.

Chagas disease, caused by Trypanosoma cruzi infection, is a zoonosis of humans, wild and domestic mammals, including dogs. In Panama, the main T. cruzi vector is Rhodnius pallescens, a triatomine bug whose main natural habitat is the royal palm, Attalea butyracea. In this paper, we present results from three T. cruzi serological tests (immunochromatographic dipstick, indirect immunofluorescence and ELISA) performed in 51 dogs from 24 houses in Trinidad de Las Minas, western Panama. We found that nine dogs were seropositive (17.6% prevalence). Dogs were 1.6 times more likely to become T. cruziseropositive with each year of age and 11.6 times if royal palms where present in the peridomiciliary area of the dog’s household or its two nearest neighbours. Mouse-baited-adhesive traps were employed to evaluate 12 peridomestic royal palms. All palms were found infested with R. pallescens with an average of 25.50 triatomines captured per palm. Of 35 adult bugs analysed, 88.6% showed protozoa flagellates in their intestinal contents. In addition, dogs were five times more likely to be infected by the presence of an additional domestic animal species in the dog’s peridomiciliary environment. Our results suggest that interventions focused on royal palms might reduce the exposure to T. cruzi infection.

Six individuals of the palm A. phalerata, in Poconé floodplains of Mato Grosso, were sprayed with a synthetic pyrethroid (0.25% concentration) in order to study the biomass, diversity, and richness of the canopy arthropods. A total of 17,188 (238.7±80.6 ind./m²) arthropods belonging to 22 Orders, was collected in a 72 m² funnel area. Two hours after spraying, 58.9% of the total number fell into the funnels, 37.6% was obtained by shaking the trees, and finally, 3.5% after cutting and washing all the palm leaves. The Coleoptera (27.4%), Hymenoptera-Formicidae (19.0%), Collembola (13.6%), Psocoptera (10.7%), Diptera (9.0%) and Araneae (6.4%) were the predominant. The total biomass was 15.1 g dry weight (0.4mg/m²; 0.13+0.04/tree). A total of 4,715 beetles representing 48 families and 326 morphospecies were obtained. Tenebrionidae (22.9%), Curculionidae (22.0%), Carabidae (10.9%) and Staphylinidae (7.9%) were the most abundant, while Curculionidae (44 spp.), Staphylinidae (40 spp.) and Chrysomelidae (34 spp.) presented the largest number of morphospecies. Herbivores (37.5%) were the dominant in the trophic guilds of adult Coleoptera, followed by predators (35.4%), fungivores (14.6%), and saprophages (12.5%). Although most arthropod Orders were represented in all the palms sampled, analysis of variance showed no significant differences in their composition, however there was a significant difference in their frequency of occurrence.

The African Republic of Cape Verde consists of nine inhabited and several uninhabited volcanic islands set out in the Atlantic Ocean, about 500 km off the most westerly point of the African mainland and 1500 km south of the Canary Islands @g. 2). Most are rugged and mountainous; three (Sal, Maio, and Boavista) are flat, desert islands with sand beaches. Precipitation is meagre and very erratic; indeed Cape Verde can be seen as an island extension of the arid Sahel zone. Three species of the genus Phoenix are recorded from the Cape Verde Islands, P. akzctyli&a L., P. canariensis Chabaud and P. atlantica A. Chev. While the former two species have almost certainly been introduced by man, the latter is said to be endemic to the islands. Perhaps because the Cape Verdes are a particularly isolated set of islands or because palms are notoriously awkward to collect, little is known about the taxonomy, origins and natural history of this species. Phoenix atlantica was described by the French botanist Auguste Chevalier (1935a) following field exploration in the Cape Verdes in 1934 (Chevalier 1934: 1153). Chevalier provided limited diagnostic characters, defining the species as a clustering palm with 2-6 trunks, 5-15 m in height with dark green leaves 2-3 m in length. He considered it to be most similar in form to P. &ctyZzjkra and P. canariensis, possessing characters of both (Chevalier 1935a). Chevalier’s description indicates that Phoenix atlantica can be distinguished easily from P. canariensis by its clustering growth form (P. canariensis always has a single, stout trunk) and its shorter, straighter leaves. However, the differences between P. atlantica and P. dactylzjkra appear much more subtle. For example, while P. dacfylifera is usually observed as single-stemmed, when left undisturbed for a number of years it becomes clustering like the Cape Verde Phoenix, so this character on its own is unreliable. Phlebotomine specimens were captured in domiciliary and forest environments in 47 municipalities between 1982 and 2005 with the aid of CDC light traps. A total of 91 species were found, of which four belonged to genus Brumptomyia and 87 to genus Lutzomyia, distributed among the following subgenera: Evandromyia (6), Lutzomyia (5), Micropygomyia (2), Nyssomyia (9), Pintomyia (2), Pressatia (3), Psathyromyia (6), Psychodopygus (14), Sciopemyia (4), Trichophoromyia (2), Viannamyia (2); species groups: Aragaoi (2), Baityi (1), Dreisbachi (1), Migonei (12), Oswaldoi (8), Pilosa (1), Saulensis (2), Verrucarum (4) and ungrouped (1). Species diversity was greatest in areas where there was dense evergreen seasonal forest (52 species), ombrophilous forest (31) and meridional cerrados (23) and lowest in areas with mixed forest (forest with babassu palms, cerrado and caatinga). The greatest similarity index was observed for restinga and open evergreen seasonal forest (J=0.48). Dense evergreen seasonal forest had greatest similarity with ombrophilous forest (J=0.38). The phlebotomine fauna was species rich and unevenly distributed in Maranhão, reflecting the phytogeographical complexity of the state, which is a result of the great variety of ecosystems and climate zones.

First report of parasitism on pupae of Opsiphanes invirae amplificatus Stichel (Lepidoptera, Nymphalidae) by Conura (Conura) maculata (Fabricius) (Hymenoptera, Chalcididae) in Rio Grande do Sul, Brazil. This scientific note records the parasitism on pupae of Opsiphanes invirae amplificatus Stichel (1904) by Conura (Conura) maculata (Fabricius). This is a report of this interaction in the subtropical region of South America and the first report of the occurrence of this parasitoid in the state of Rio Grande do Sul, Brazil.

Adults of Cyclocephala distincta are flower visitors of Neotropical palms (Arecaceae) and commonly found in the Atlantic Forest of Pernambuco, Brazil. Males and females were collected in the wild and subjected to captive rearing and breeding. The egg hatching rate, the life cycle, longevity of immatures and adults, and oviposition parameters in captivity were analyzed. The average duration of the life cycle of C. distinctawas 108.2 days (n = 45). The egg stage lasted on average 10.9 days, and the egg-hatching rate was 73.9%. The immature stage lasted on average 93.4 days. The larvae stage exhibited negative phototaxis, and the size of their head capsules increased at a constant rate of 1.6 between instars, following Dyar's rule. The average duration of the first instar was 24.8 days (n = 88), whereas the second and third instars lasted for 17.2 (n = 76) and 40.4 (n = 74) days respectively, and survival rates were 21.6%, 86.4% and 97.4%. The pre-pupal stage was recorded, and pupal chambers were built before pupation. The average number of eggs laid per female was 15.5, the total reproductive period lasted for 3.3 days, and the total fertility was 81.2%. Adults that emerged in captivity exhibited an average longevity of 18.9 days. Adult C. distincta exhibited thanatosis behavior upon manipulation, a strategy observed for the first time in Cyclocephala.

RESUME: Etude de l'activation et de l'inactivation pH-dépendantes des canaux ASICs (Acid-Sensing Ion Channels) Benoîte BARGETON, Département de Pharmacologie et de Toxicologie, Université de Lausanne, rue du Bugnon 27, CH-1005 Lausanne, Suisse Les canaux sodiques ASICs (Acid-Sensing Ion Channels) participent à la signalisation neuronale dans les systèmes nerveux périphérique et central. Ces canaux non voltage dépendants sont impliqués dans l'apprentissage, l'expression de la peur, la neurodégénération consécutive à une attaque cérébrale et la douleur. Les bases moléculaires sous-tendant leur activité ne sont pas encore totalement comprises. Ces canaux sont activés par une acidification du milieu extracellulaire et régulés, entre autres, par des ions tels que le Ca2+, le Zn2+ et le CI". La cristallisation de ASIC inactivé a été publiée. Le canal est un trimére de sous-unités identiques ou homologues. Chaque sous-unité a été décrite en analogie à un avant bras, un poignet et une main constituée d'un pouce, d'un doigt, d'une articulation, une boule β et une paume. Nous avons appliqué une approche bioinformatique systématique pour identifier les pH senseurs putatifs de ASICIa. Le rôle des pH senseurs putatifs a été testé par mutagénèse dirigée et des modifications chimiques combinées à une analyse fonctionnelle afin de comprendre comment les variations de ρ H ouvrent ces canaux. Les pH senseurs sont des acides aspartiques et glutamiques éparpillés sur la boucle extracellulaire suggérant que les changements de pH contrôlent l'activation et l'inactivation de ASIC en (dé)protonant ces résidus en divers endroits de la protéine. Par exemple lors de l'activation, la protonation des résidus à l'interface entre le pouce, la boule β et le doigt d'une même sous-unité induit un mouvement du pouce vers la bouie β et le doigt. De même lors de l'inactivation du canal les paumes des trois sous-unités formant une cavité se rapprochent. D'après notre approche bioinformatique, aucune histidine n'est impliquée dans la détection des variations de pH extracellulaire c'est-à-dire qu'aucune histidine ne serait un pH-senseur. Deux histidines de ASIC2a lient le Zn2+ et modifient l'affinité apparente du canal pour les protons. Une seule des deux est conservée parmi tous les ASICs, hASICIa H163. Elle forme un réseau de liaison hydrogène avec ses voisins conservés. L'étude détaillée de ce domaine, Pinterzone, montre son importance dans l'expression fonctionnelle des canaux. La perturbation de ce réseau par l'introduction d'un résidu hydrophobe (cystéine) par mutagénèse dirigée diminue l'expression du canal à la membrane plasmique. La modification des cystéines introduites par des réactifs spécifiques aux groupements sulfhydryle inhibe les canaux mutés en diminuant leur probabilité d'ouverture. Ces travaux décrivent les effets de l'acidification du milieu extracellulaire sur les canaux ASICs. These non-voltage-gated channels are involved in learning, the expression of fear, neurodegeneration after ischemia and pain sensation. The molecular bases underlying their activity are not yet fully understood. ASICs are activated by extracellular acidification and regulated, eg by ions such as Ca2+, the Zn2+ and CI". The crystallization of inactivated ASIC has been published. The channel is a trimer of identical or homologous subunits. Each subunit has been described in analogy to a forearm, wrist and hand consisting of a thumb, a finger, a knuckle, a β-ball and a palm. We applied a systematic computational approach to identify putative pH sensor(s) of ASICIa. The role of putative pH sensors has been tested by site-directed mutagenesis and chemical modification combined with functional analysis in order to understand how changes in pH open these channels. The pH sensors are aspartic and glutamic acids distributed throughout the extracellular loop, suggesting that changes in pH control activation and inactivation of ASIC by protonation / deprotonation of many residues in different parts of the protein. During activation the protonation of various residues at the interface between the finger, the thumb and the β-ball induces the movement of the thumb toward the finger and the β-ball. During inactivation of the channel the palms of the three subunits forming a cavity approach each other. No histidine has been shown to be involved in extracellular pH changes detection, i.e. no histidine is a pH- sensor. Two histidines of ASIC2 bind Zn2+ and alter the apparent affinity of channel for protons. Only one of the two His is conserved among all ASICs, hASICIa H163. This residue is part of a network of hydrogen bonding with its conserved neighbors. The detailed study of this area, the interzone, shows its importance in the functional expression of ASICs. Disturbance of this network by the introduction of hydrophobic residues decreases the cell surface channel expression. Chemical modification of the introduced cysteines by thiol reactive compounds inhibits the mutated channels by a reduction of their open probability. These studies describe the effects of extracellular acidification on ASICs. Un dysfonctionnement de certains types de synapses est à l'origine de beaucoup de troubles nerveux, tels que certaine forme d'épilepsie et de l'attention. Les récepteurs des neurotransmetteurs sont de très bonnes cibles thérapeutiques dans de nombreuses neuropathologies. Les canaux ASICs sont impliqués dans la neurodégénération consécutive à une attaque cérébrale et les bloquer pourraient permettre aux patients d'avoir moins de séquelles. Les canaux ASICs sont des détecteurs de l'acidité qui apparaît lors de situations pathologiques comme l'ischémie et l'inflammation. Ces canaux sont également impliqués dans des douleurs. Cibler spécifiquement ces canaux permettrait d'avoir de nouveaux outils thérapeutiques car à l'heure actuelle l'inhibiteur de choix, l'amiloride, bloque beaucoup d'autres canaux empêchant son utilisation pour bloquer les ASICs. C'est pourquoi il faut connaître et comprendre les bases moléculaires du fonctionnement de ces récepteurs. Les ASICs formés de trois sous-unités détectent les variations de l'acidité puis s'ouvrent transitoirement pour laisser entrer des ions chargés positivement dans la cellule ce qui active la signalisation neuronale. Afin de comprendre les bases moléculaires de l'activité des ASICs nous avons déterminé les sites de liaison des protons (pH-senseurs), ligands naturels des ASICs et décrit une zone importante pour l'expression fonctionnelle de ces canaux. Grâce à une validation systématique de résultats obtenus en collaboration avec l'Institut Suisse de Bioinformatique, nous avons décrit les pH-senseurs de ASICIa. Ces résultats, combinés à ceux d'autres groupes de recherche, nous ont permis de mieux comprendre comment les ASICs sont ouverts par une acidification du milieu extracellulaire. Une seconde étude souligne le rôle structural crucial d'une région conservée parmi tous les canaux ASICs : y toucher c'est diminuer l'activité de la protéine. Ce domaine permet l'harmonisation des changements dus à l'acidification du milieu extracellulaire au sein d'une même sous-unité c'est-à-dire qu'elle participe à l'induction de l'inactivation due à l'activation du canal Cette étude décrit donc quelle région de la protéine atteindre pour la bloquer efficacement en faisant une cible thérapeutique de choix.

There is no definite theory yet for the mechanism by which the pattern of epidermal ridges on fingers, palms and soles forming friction ridge skin (FRS) patterns is created. For a long time growth forces in the embryonal epidermis have been believed to be involved in FRS formation. More recent evidence suggests that Merkel cells play an important part in this process as well. Here we suggest a model for the formation of FRS patterns that links Merkel cells to the epidermal stress distribution. The Merkel cells are modeled as agents in an agent based model that move anisotropically where the anisotropy is created by the epidermal stress tensor. As a result ridge patterns are created with pattern defects as they occur in real FRS patterns. As a consequence we suggest why the topology of FRS patterns is indeed unique as the arrangement of pattern defects is sensitive to the initial configuration of Merkel cells.

ABSTRACT This work aimed to evaluate the consequences of the monthly extraction of immature leaves in survival, leaf production and reproductive performance ofCopernicia prunifera H. E. Moore palm, popularly known as carnaúba. One hundred sixty reproductive adult palms were monitored for 17 months in four extractive communities located at the coast of the state of Piauí. As a result, it was observed that leaves, flowers, fruits and seeds production were reduced in the palm submitted to 50% or 75% monthly extraction. Higher levels of extraction were followed by smaller levels in seed germination. No deaths were observed even in the group subjected to 75% monthly leaves exploration. In order not to produce damage to palm trees development it is suggested that leaf extraction rate should not exceed 25% monthly as well as pausing of extractive activity preferentially during fruit maturation.

A resistência de plantas jovens de açaí (Euterpe oleracea Mart.) à deficiência de água foi determinada em plantas de quatro meses de idade cultivadas em sacos plásticos perfurados contendo 7 kg de terra. O experimento foi conduzido em casa de vegetação e a irrigação foi suspensa até as plantas atingirem fotossíntese zero, quando, então, foram reidratadas. Durante este período foram feitas medidas diárias da fotossíntese líquida, condutância estomática e transpiração. Pequenas reduções na condutância estomática, transpiração e fotossíntese foram observadas a partir do décimo dia e se acentuaram com o tempo. A taxa de fotossíntese reduziu-se à metade da fotossíntese máxima quando o potencial de água atingiu -1,9 MPa. O estresse hídrico atingiu nível suficientemente severo para induzir uma fotossíntese nula após 61 dias sem irrigação, com um potencial de água médio foliar de -2,5 MPa. A recuperação da fotossíntese, condutância estomática e transpiração iniciaram-se um dia após a irrigação, atingindo valores semelhantes ao do controle após 14 dias de reidratação. A análise das curvas de pressão e volume mostrou que a deficiência de água diminuiu o potencial osmótico, resultando em um ajustamento osmótico de 0,29 MPa na turgescência zero e 0,36 MPa na turgescência máxima. Isto indica que essa espécie possui mecanismos de adaptação a um estresse hídrico moderado.
