Malária é uma das mais incidentes doenças infecciosas do mundo. Na Amazônia existem muitos casos de malária causados principalmente por duas espécies de protozoários, o Plasmodium vivax e o Plasmodium falciparum, sendo este último responsável pela maioria dos casos de malária grave, que geralmente levam a morte devido ao acometimento de múltiplos órgãos, como o cérebro. Um dos mediadores químicos amplamente estudados nessa patogênese é o Óxido Nítrico (NO), o qual apresenta papel controverso. Atualmente duas hipóteses principais são apontadas como potencializadoras na patogênese. Uma, que a MC é causa da superprodução de NO, produzido pela Óxido Nítrico Sintase Neuronal (nNOS), após um quadro de hipóxia. Outra, diz que a MC é a causa da resposta exacerbada do sistema imunológico com produção de NO pela Óxido Nítrico Sintase Induzida (iNOS), presente nos macrófagos quando ativados pro determinantes antigênicos. Devido grande relevância da doença e dificuldade em enteder a patologia, modelos experimentais têm sido estabelecidos com a finalidade de esclarecer vias potenciais da evolução para MC, dentre eles o modelo de malária aviária causada pelo Plasmodium gallinaceum. Pouco se sabe sobre o seu papel do NO em modelos de malária aviária, principalmente devido inexistência de marcadores específicos para avaliar expressão das enzimas de síntese. Diante disso é importante estabelecer protocolos de purificação da iNOS de galinhas para a produção de um possível marcador. Para tanto se faz necessário investigar o papel do NO durante a malária aviária, em modelo experimental in vivo e in vitro, com linhagens de macrófagos de galinha HD11. Animais infectados com P. gallinaceum tratados com aminoguanidina (AG), um inibidor da produção de NO, tiveram maior sobrevida, além de menores níveis de nitrito no plasma e em macrófagos derivados de monócitos do sangue periférico, sugerindo a inibição da iNOS. Nos experimentos in vitro, células HD11 tratadas com LPS mostraram produção aumentada de NO, inferindo aumento na expressão e atividade da iNOS. Na separação proteica, observamos padrões diferentes que podem ser associados a uma elevada expressão da iNOS nos macrófagos ativados com LPS. Esse estudo proporcionará o melhor entendimento do modelo de malária aviária em galinhas, incluindo a cerebral, e envolvimento do sistema nitrérgico em galinhas infectadas com P. gallinaceum.

Tumor-induced osteomalacia (TIO) is a paraneoplastic bone mineral disturbance related to fibroblast growth factor 23 (FGF23) overproduction by the tumor, usually from mesenchymal origin. Such condition leads to high phosphate renal wasting and, consequently, to cumbersome symptoms as weakness, bone pain, and fractures. Case report. We report a case of an advanced castration-refractory prostate cancer patient, which developed severe hypophosphatemia with elevated phosphate excretion fraction. TIO was suspected, and increased levels of FGF23 reinforced such diagnosis. The patient died 4 months after being diagnosed with TIO. This case suggests that TIO has a dismal prognosis in prostate cancer patients. The clinical oncology community must be aware about such disturbance that can be present in those patients with weakness, bone pain, and hypophosphatemia.

It is well established that the development of insulin resistance shows a temporal sequence in different organs and tissues. Moreover, considering that the main aspect of insulin resistance in liver is a process of glucose overproduction from gluconeogenesis, we investigated if this metabolic change also shows temporal sequence. For this purpose, a well-established experimental model of insulin resistance induced by high-fat diet (HFD) was used. The mice received HFD (HFD group) or standard diet (COG group) for 1, 7, 14 or 56?days. The HFD group showed increased (P?<?0.05 versus COG) epididymal, retroperitoneal and inguinal fat weight from days 1 to 56. In agreement with these results, the HFD group also showed higher body weight (P?<?0.05 versus COG) from days 7 to 56. Moreover, the changes induced by HFD on liver gluconeogenesis were progressive because the increment (P?<?0.05 versus COG) in glucose production from l-lactate, glycerol, l-alanine and l-glutamine occurred 7, 14, 56 and 56 days after the introduction of the HFD schedule, respectively. Furthermore, glycaemia and cholesterolemia increased (P?<?0.05 versus COG) 14?days after starting the HFD schedule. Taken together, the results suggest that the intensification of liver gluconeogenesis induced by an HFD is not a synchronous all-or-nothing process but is specific for each gluconeogenic substrate and is integrated in a temporal manner with the progressive augmentation of fasting glycaemia. Background: Dietary creatine has been largely used as an ergogenic aid to improve strength and athletic performance, especially in short-term and high energy-demanding anaerobic exercise. Recent findings have also suggested a possible antioxidant role for creatine in muscle tissues during exercise. Here we evaluate the effects of a 1-week regimen of 20 g/day creatine supplementation on the plasma antioxidant capacity, free and heme iron content, and uric acid and lipid peroxidation levels of young subjects (23.1 +/- 5.8 years old) immediately before and 5 and 60 min after the exhaustive Wingate test. Results: Maximum anaerobic power was improved by acute creatine supplementation (10.5 %), but it was accompanied by a 2.4-fold increase in pro-oxidant free iron ions in the plasma. However, potential iron-driven oxidative insult was adequately counterbalanced by proportional increases in antioxidant ferric-reducing activity in plasma (FRAP), leading to unaltered lipid peroxidation levels. Interestingly, the FRAP index, found to be highly dependent on uric acid levels in the placebo group, also had an additional contribution from other circulating metabolites in creatine-fed subjects. Conclusions: Our data suggest that acute creatine supplementation improved the anaerobic performance of athletes and limited short-term oxidative insults, since creatine-induced iron overload was efficiently circumvented by acquired FRAP capacity attributed to: overproduction of uric acid in energy-depleted muscles (as an end-product of purine metabolism and a powerful iron chelating agent) and inherent antioxidant activity of creatine.

Anti-silencing factor 1 (ASF1) is a histone chaperone that contributes to the histone deposition during nucleosome assembly in newly replicated DNA. It is involved in chromatin disassembly, transcription activation and in the cellular response to DNA damage. In Leishmania major the ASF1 gene (LmASF1) is located in chromosome 20 and codes for a protein showing 67% of identity with the Trypanosoma brucei TbASF1a. Compared to orthologous proteins, LmASF1 conserves the main residues relevant for its various biological functions. To study ASF1 in Leishmania we generated a mutant overexpressing LmASF1 in L. major. We observed that the excess of LmASF1 impaired promastigotes growth rates and had no impact on cell cycle progress. Differently from yeast, ASF1 overproduction in Leishmania did not affect expression levels of genes located on telomeres, but led to an upregulation of proteins involved in chromatin remodelling and physiological stress, such as heat shock proteins, oxidoreductase activity and proteolysis. In addition, we observed that LmASF1 mutant is more susceptible to the DNA damaging agent, methyl methane sulphonate, than the control line. Therefore, our study suggests that ASF1 from Leishmania pertains to the chromatin remodelling machinery of the parasite and acts on its response to DNA damage.

Sigma (σ) receptors are well established as a non-opioid, non-phencyclidine, and haloperidol-sensitive receptor family with its own binding profile and a characteristic distribution in the central nervous system (CNS) as well as in endocrine, immune, and some peripheral tissues. Two σ receptors subtypes, termed σ1 and σ2, have been pharmacologically characterized, but, to date, only the σ1 has also been cloned. Activation of σ1 receptors alter several neurotransmitter systems and dopamine (DA) neurotrasmission has been often shown to constitute an important target of σ receptors in different experimental models; however the exact role of σ1 receptor in dopaminergic neurotransmission remains unclear. The DA transporter (DAT) modulates the spatial and temporal aspects of dopaminergic synaptic transmission and interprer the primary mechanism by wich dopaminergic neurons terminate the signal transmission. For this reason present studies have been focused in understanding whether, in cell models, the human subtype of σ1 (hσ1) receptor is able to directly modulate the human DA transporter (hDAT). In the first part of this thesis, HEK-293 and SH-SY5Y cells were permanently transfected with the hσ1 receptor. Subsequently, they were transfected with another plasmid for transiently expressing the hDAT. The hDAT activity was estimated using the described [3H]DA uptake assay and the effects of σ ligands were evaluated by measuring the uptaken [3H]DA after treating the cells with known σ agonists and antagonists. Results illustrated in this thesis demonstrate that activation of overexpressed hσ1 receptors by (+)-pentazocine, the σ1 agonist prototype, determines an increase of 40% of the extracellular [3H]DA uptake, in comparison to non-treated controls and the σ1 antagonists BD-1047 and NE-100 prevent the positive effect of (+)-pentazocine on DA reuptake DA is likely to be considered a neurotoxic molecule. In fact, when levels of intracellular DA abnormally invrease, vescicles can’t sequester the DA which is metabolized by MAO (A and B) and COMT with consequent overproduction of oxygen reactive species and toxic catabolites. Stress induced by these molecules leads cells to death. Thus, for the second part of this thesis, experiments have been performed in order to investigate functional alterations caused by the (+)-pentazocine-mediated increase of DA uptake; particularly it has been investigated if the increase of intracellular [DA] could affect cells viability. Results obtained from this study demonstrate that (+)-pentazocine alone increases DA cell toxicity in a concentration-dependent manner only in cells co-expressing hσ1 and hDAT and σ1 antagonists are able to revert the (+)-pentazocine-induced increase of cell susceptibility to DA toxicity. In the last part of this thesis, the functional cross-talking between hσ1 receptor and hDAT has been further investigated using confocal microscopy. From the acquired data it could be suggested that, following exposure to (+)-pentazocine, the hσ1 receptors massively translocate towards the plasma membrane and colocalize with the hDATs. However, any physical interaction between the two proteins remains to be proved. In conclusion, the presented study shows for the first time that, in cell models, hσ1 receptors directly modulate the hDAT activity. Facilitation of DA uptake induced by (+)-pentazocine is reflected on the increased cell susceptibility to DA toxicity; these effects are prevented by σ1 selective antagonists. Since numerous compounds, including several drugs of abuse, bind to σ1 receptors and activating them could facilitate the damage of dopaminergic neurons, the reported protective effect showed by σ1 antagonists would represent the pharmacological basis to test these compounds in experimental models of dopaminergic neurodegenerative diseases (i.e. Parkinson’s Disease).

This PhD thesis is aimed at studying the possible pathways and the mechanisms that can trigger oxylipins biosynthesis, and particularly that of short chain aldehydes and alcohols, in Lactobacillus helveticus, also in the presence of oxidative stress, using a totally labelled linoleic acid as precursor. In plants and fungi these molecules, involved in defence mechanisms against pathogens and in communication systems, derive from the oxidation of cellular unsaturated fatty acids (UFAs) and their accumulation is associated with stress exposure. Since some oxylipins are produced also by lactobacilli, it is possible to hypothesize that a metabolic pathway from UFAs to oxylipins, similar to what happens in plants and fungi, is present also in lactic acid bacteria. The results obtained pointed out that some volatile molecules are the result of UFAs catabolism, since they appear only when cells are incubated in their presence. Labelled linoleic acid is integrated in the membrane and subsequently transformed into aldehydes and alcohols, whose extent and carbon atoms number depend on stress exposure. The enzymes responsible for this metabolic pathway in plants and fungi (e.g. lipoxygenase, dioxygenase) seem to be absent in Lactobacillus helveticus and in other lactobacilli. Proteomic analyses show the over expression of many proteins, including thioredoxin reductase (part of the bacterial oxidative defence system), mainly in cells grown with linoleic acid without oxidative stress exposure, confirming that linoleic acid itself induces oxidative stress. 6 general oxidoreductases (class including dioxygenases and peroxidase) were found and therefore a deeper investigation on them could be productive in elucidating all steps involved in oxylipins biosynthesis in bacteria. Due to the multiple role of oxylipins (flavouring agents, antimicrobial compounds and interspecific signalling molecules) the identification of genes involved and regulating factors should have an important biotechnological impact, also allowing the overproduction of selected bioactive molecules.

TGF-beta ist ein Schlüsselmolekül zellvermittelter Immuntoleranz. So spielt es neben seiner pleiotropen Rolle in Immunzellen auch bei der Tumorentwicklung eine große Rolle. Das TGF-beta hat bei der Tumorentwicklung eine duale Rolle. So dient es in frühen Phasen als Tumorsuppressor, währenddessen es in späten Phasen der Entwicklung als Tumorpromotor wirkt. Eine strikte Regulation des TGF-beta Signalweges ist daher für ein funktionierendes Immunsystem von essentieller Bedeutung. Die Ubiquitin Ligase Smurf2 ist dabei ein wichtiger negativ Regulator des TGF-beta Signalweges.In der vorliegenden Arbeit konnte eine neue Spleißform des Smurf2 (dE2Smurf2) aus murinen CD4+ T-Zellen isoliert werden, deren Funktion in vitro und in vivo in T-Lymphozyten untersucht worden ist. Für diese Spleißform konnte zudem eine humane Relevanz nachgewiesen werden. Mit Hilfe von Überexpressionen in Cos7 Zellen konnte eine veränderte Lokalisation der Smurf2 Spleißformen (WT und dE2) festgestellt werden. Dabei konnten lysosomale und endosomale Kompartimente bei der Kolokalisation mit dem dE2Smurf2 Konstrukt beobachtet werden. Das Spleißen des Exons2 führte dabei zu Änderungen der Topologie der N-terminalen C2-Domäne, wodurch sich eine veränderte Lokalisation in der Zelle beschreiben ließ. Mit der veränderten intrazellulären Verteilung erfuhr auch die Funktion der dE2Smurf2 Ubiquitin Ligase eine Änderung. So konnte überraschenderweise eine positive Signalinduktion des TGF-beta Signalweges beobachtet werden, was im Gegensatz zum beschriebenen WTSmurf2 stand. Durch eine Überexpression des dE2Smurf2 Proteins in T-Lymphozyten wurde der TGF-beta Signalweg in CD4+ und CD8+ Zellen positiv reguliert, dabei wurde der TGFbetaRII vermehrt exprimiert und gleichzeitig fand eine verstärkte Phosphorylierung der Transkriptionsfaktoren Smad2 und Smad3 nach TGF-beta Stimulation statt. Die transgenen T-Lymphozyten waren somit sensitiver gegenüber TGF-beta. Dies führte zur Hypothese, die durch Western Blot Analyse bestätigt werden konnte, daß das dE2Smurf2 nach Überexpression seine WT-Form bindet und dadurch degradiert. Die Degradation der Ubiquitin Ligase war dabei Smad7 abhängig. Zur Analyse des Einflusses der Ubiquitin Ligase dE2Smurf2 auf die Differenzierung von CD4+ T-Zellen, sowie ihre Rolle bei der T-Zell Proliferation, konnte gezeigt werden, daß durch die höhere Sensitivität gegenüber TGF-beta naive T-Zellen unter Einfluß von TGF-beta und IL6 vermehrt in TH17 Zellen differenzierten. Zudem konnte gezeigt werden, daß die Proliferationsrate transgener naiver CD4+ T-Zellen bei geringen Mengen von TGF-beta starkt vermindert war. Weiterhin konnte gezeigt werden, daß bei einer Differenzierung der naiven CD4+ T-Zellen in TH1 Zellen, diese signifkant weniger das proinflammatorische Zytokin INFγ produzierten.So zeigten in vivo Versuche, daß die transgenen Tiere in der Entwicklung von Kolorektalen Karzinomen protektiert waren. Sowohl im kolitisassiziierten Tumor Modell als auch bei der spontanen Entwicklung von Tumoren im APCmin Modell. Dies konnte zum einen auf eine deutlich verminderte Entzündung (geringere Produktion an Zytokinen durch verminderte Proliferation) des Darms und zum anderen durch eine stärkere Produktion an zytotoxischen Genen, wie Perforin, INFγ und Granzym B erklärt werden. Interessanterweise konnte jedoch im Transfer Kolitis Modell eher eine proinflammatorische Wirkung des dE2Smurf2 Proteins nachgewiesen werden. So wiesen die immundefizienten Mäuse, in denen die transgenen T-Zellen injiziert wurden, eine signifikant stärkere Kolitis auf als die Kontrollen. Dies konnte mit einer Überproduktion an IL17 sezernierenden T-Zellen erklärt werden. Klonierungsexperimente führten zudem zur Identifikation einer bisher nicht beschriebenen nicht kodierenden RNA. Diese zeigte in Kombination mit dem dE2Smurf2 Protein in einer Reportergen Analyse eine Hyperaktivierung des Smad3 Promotors. Diese Daten liefern zum einen ein genaueres Modell über die Regulation des TGF-beta Signalweges sowie wichtige Erkenntnisse zur Pathophysiologie chronisch entzündlicher Darmerkrankung und daraus resultierende Tumorerkrankungen. So entwickelt sich das dE2Smurf2, Teil des TGF-beta Signalweges, als attraktives Zielprotein für die Modulation von chronisch entzündlichen Darmerkrankungen und (kolitisassoziierte) Kolonkarzinomen.

Le alterazioni della funzionalità mitocondriale detengono un ruolo cruciale nella patogenesi della malattia di Alzheimer (AD), sostenendo il processo neurodegenerativo attraverso meccanismi quali la riduzione della disponibilità energetica e la iperproduzione di ROS. Alle numerose ipotesi di patogenesi dell’AD, si è recentemente affiancata la cosiddetta ipotesi vascolare. Nei soggetti AD è stata riscontrata una significativa riduzione della disponibilità di ossigeno a livello neuronale (ipossia neuronale). Da numerosi studi è poi emerso che l’ipossia gioca un ruolo fondamentale nello sviluppo dell’AD contribuendo a più vie patogenetiche contemporaneamente. Tuttavia, non sono stati ancora chiariti tutti i meccanismi attraverso cui l’ipossia esplica la sua azione di danno. Lo scopo di questo studio è stato quello di contribuire a chiarire il ruolo patologico dell’ipossia nell’AD, analizzando principalmente le alterazioni della funzionalità mitocondriale indotte dalla riduzione della disponibilità di ossigeno. Nella prima fase dello studio cellule PC12 sono state coltivate in presenza di β-amiloide e ipossia. In questo modello abbiamo osservato un potenziamento dei fenomeni di deplezione dell’ATP e di generazione delle ROS indotti dalla Aβ quando anche l’ipossia era presente come fonte di danno cellulare, ipotizzando per i due fattori un effetto congiunto di tipo additivo. Nella seconda fase abbiamo esposto all’ipossia fibroblasti prelevati da pazienti AD portatori di mutazioni a carico dei geni APP e PSEN. La presenza di mutazioni predisponenti ad un fenotipo AD era in grado di determinare un danno bioenergetico e ossidativo. Le alterazioni bioenergetiche riscontrate in normossia risultavano ulteriormente potenziate quando i fibroblasti erano coltivati in ipossia, mentre lo stato di stress ossidativo veniva evidenziato solo in condizioni ipossiche. Sulla base dei risultati finora conseguiti si può ipotizzare che uno dei meccanismi attraverso cui l’ipossia esplica la sua azione di danno nella AD, possa essere dovuto alla capacità di potenziare ulteriormente le alterazioni della funzionalità mitocondriale.

During sepsis, activation of phagocytes leads to the overproduction of proinflammatory cytokines, causing systemic inflammation. Despite substantial information regarding the underlying molecular mechanisms that lead to sepsis, several elements in the pathway remain to be elucidated. We found that the enzyme sphingosine kinase 1 (SphK1) is up-regulated in stimulated human phagocytes and in peritoneal phagocytes of patients with severe sepsis. Blockade of SphK1 inhibited phagocyte production of endotoxin-induced proinflammatory cytokines. We observed protection against sepsis in mice treated with a specific SphK1 inhibitor that was enhanced by treatment with a broad-spectrum antibiotic. These results demonstrated a critical role for SphK1 in endotoxin signaling and sepsis-induced inflammatory responses and suggest that inhibition of SphK1 is a potential therapy for septic shock.
