The ingress of water into poly(2-hydroxyethyl methacrylate), PHEMA, loaded with either one of two model drugs, vitamin B-12 or aspirin, was studied at 37 degreesC using three-dimensional NMR imaging. PHEMA was loaded with 5 and 10 wt % of the drugs. From the imaging profiles, it was observed that incorporation of vitamin B-12 into PHEMA resulted in enhanced crack formation on sorption of water and the crack healing behind the diffusion front was slower than for PHEMA without added drug. This was accounted for by the anti-plasticization of PHEMA by vitamin B-12. Crack formation was inhibited in the P-HEMA-aspirin systems because of the plasticizing effect of the aspirin on the PHEMA matrix. All of the polymers were found to absorb water according to an underlying Fickian diffusion mechanism. For PHEMA loaded with 5 wt % of aspirin or vitamin B-12, the best values of the water diffusion coefficients were both found to be 1.3 +/- 0.1 x 10(-11) m(2) s(-1) at 37 degreesC, while the values for the polymer loaded with 10 wt % of the drugs were slightly higher, 1.5 +/- 0.1 x 10(-11) m(2) s(-1).

Purpose To study the protective effects and underlying molecular mechanisms of SAMC on carbon tetrachloride (CCl4)-induced acute hepatotoxicity in the mouse model. Methods Mice were intraperitoneally injected with CCl4 (50 μl/kg; single dose) to induce acute hepatotoxicity with or without a 2-h pre-treatment of SAMC intraperitoneal injection (200 mg/kg; single dose). After 8 h, the blood serum and liver samples of mice were collected and subjected to measurements of histological and molecular parameters of hepatotoxicity. Results SAMC reduced CCl4-triggered cellular necrosis and inflammation in the liver under histological analysis. Since co-treatment of SAMC and CCl4 enhanced the expressions of antioxidant enzymes, reduced the nitric oxide (NO)-dependent oxidative stress, and inhibited lipid peroxidation induced by CCl4. SAMC played an essential antioxidative role during CCl4-induced hepatotoxicity. Administration of SAMC also ameliorated hepatic inflammation induced by CCl4 via inhibiting the activity of NF-κB subunits p50 and p65, thus reducing the expressions of pro-inflammatory cytokines, mediators, and chemokines, as well as promoting pro-regenerative factors at both transcriptional and translational levels. Conclusions Our results indicate that SAMC mitigates cellular damage, oxidative stress, and inflammation in CCl4-induced acute hepatotoxicity mouse model through regulation of NF-κB. Garlic or garlic derivatives may therefore be a potential food supplement in the prevention of liver damage.

Although there is a paucity of scientific support for the benefits of warm-up, athletes commonly warm up prior to activity with the intention of improving performance and reducing the incidence of injuries. The purpose of this study was to examine the role of warm-up intensity on both range of motion (ROM) and anaerobic performance. Nine males (age = 21.7 +/- 1.6 years, height = 1.77 +/- 0.04 m, weight = 80.2 +/- 6.8 kg, and VO2max = 60.4 +/- 5.4 ml/kg/min) completed four trials. Each trial consisted of hip, knee, and ankle ROM evaluation using an electronic inclinometer and an anaerobic capacity test on the treadmill (time to fatigue at 13 km/hr and 20% grade). Subjects underwent no warm-up or a warm-up of 15 minutes running at 60, 70 or 80% VO2max followed by a series of lower limb stretches. Intensity of warm-up had little effect on ROM, since ankle dorsiflexion and hip extension significantly increased in all warm-up conditions, hip flexion significantly increased only after the 80% VO2max warm-up, and knee flexion did not change after any warm-up. Heart rate and body temperature were significantly increased (p < 0.05) prior to anaerobic performance for each of the warm-up conditions, but anaerobic performance improved significantly only after warm-up at 60% VO2max (10%) and 70% VO2max (13%). A 15-minute warm-up at an intensity of 60-70% VO2max is therefore recommended to improve ROM and enhance subsequent anaerobic performance.

Our understanding of the origin and fate of the IgE-switched B cell has been markedly improved by studies in mouse models. The immediate precursor of the IgE-switched B cell is either a relatively naive nonswitched B cell or a mature IgG-switched B cell. These 2 routes are referred to as the direct and indirect pathways, respectively. IgE responses derived from each pathway differ significantly, largely reflecting the difference in time spent in a germinal center and thus time for clonal expansion, somatic hypermutation, affinity maturation, and acquisition of a memory phenotype. The clinical and therapeutic implications for IgE responses in human subjects are still a matter of debate, largely because the immunization procedures used in the animal models are significantly different from classical atopic sensitization to allergens from pollen and mites. On the basis of the limited information available, it seems likely that these atopic IgE responses are characterized by a relatively low IgG/IgE ratio, low B-cell memory, and modest affinity maturation, which fits well with the direct switching pathway. It is still unresolved how the IgE response evolves to cover a wide epitope repertoire involving many epitopes per allergen, as well as many different allergens from a single allergen source. © 2013 American Academy of Allergy, Asthma & Immunology.

The HLA-B27 subtypes have a varied racial and ethnic prevalence throughout the world. However, the association of B27-subtypes with ankylosing spondylitis (AS) in the mainland China is unknown. To determine the association of B27-subtypes with AS in the Mainland Chinese Han population, a total of unrelated 153 patients with AS were enrolled in a large case-control association study, and 1545 unrelated, healthy, ethnically matched blood donors were included as controls. The genotyping of B27 and its subtypes was performed using the polymerase chain reaction with sequence specific primers (PCR-SSP). A total of 130 (84.97%) AS patients and 61 (3.95%) healthy controls were B27 positive. Three B27-subtypes, B*2704, B*2705 and B*2710, were further identified, of which both B*2704 and B*2705 were strongly AS associated. B*2710 was only detected in one AS patient and two other healthy controls. Considering only B27-positive cases and controls, a statistically different frequency of B27-subtypes was observed, with an over-representation of B*2704 (P = 0.018). B*2704 was clearly more strongly associated than B*2705 with AS [odds ratio (OR) = 2.4, P = 0.011]. Furthermore, a combined analysis including three previous studies of B27-subtype distributions in Chinese AS cases confirmed the stronger association of B*2704 with AS than B*2705 (OR = 2.5, P = 0.00094).

B. cereus is one of the most frequent occurring bacteria in foods . It produces several heat-labile enterotoxins and one stable non-protein toxin, cereulide (emetic), which may be pre-formed in food. Cereulide is a heat stable peptide whose structure and mechanism of action were in the past decade elucidated. Until this work, the detection of cereulide was done by biological assays. With my mentors, I developed the first quantitative chemical assay for cereulide. The assay is based on liquid chromatography (HPLC) combined with ion trap mass spectrometry and the calibration is done with valinomycin and purified cereulide. To detect and quantitate valinomycin and cereulide, their [NH4+] adducts, m/z 1128.9 and m/z 1171 respectively, were used. This was a breakthrough in the cereulide research and became a very powerful tool of investigation. This tool made it possible to prove for the first time that the toxin produced by B. cereus in heat-treated food caused human illness. Until this thesis work (Paper II), cereulide producing B. cereus strains were believed to represent a homogenous group of clonal strains. The cereulide producing strains investigated in those studies originated mostly from food poisoning incidents. We used strains of many origins and analyzed them using a polyphasic approach. We found that the cereulide producing B. cereus strains are genetically and biologically more diverse than assumed in earlier studies. The strains diverge in the adenylate kinase (adk) gene (two sequence types), in ribopatterns obtained with EcoRI and PvuII (three patterns), tyrosin decomposition, haemolysis and lecithine hydrolysis (two phenotypes). Our study was the first demonstration of diversity within the cereulide producing strains of B. cereus. To manage the risk for cereulide production in food, understanding is needed on factors that may upregulate cereulide production in a given food matrix and the environmental factors affecting it. As a contribution towards this direction, we adjusted the growth environment and measured the cereulide production by strains selected for diversity. The temperature range where cereulide is produced was narrower than that for growth for most of the producer strains. Most cereulide was by most strains produced at room temperature (20 - 23ºC). Exceptions to this were two faecal isolates which produced the same amount of cereulide from 23 ºC up until 39ºC. We also found that at 37º C the choice of growth media for cereulide production differed from that at the room temperature. The food composition and temperature may thus be a key for understanding cereulide production in foods as well as in the gut. We investigated the contents of [K+], [Na+] and amino acids of six growth media. Statistical evaluation indicated a significant positive correlation between the ratio [K+]:[Na+] and the production of cereulide, but only when the concentrations of glycine and [Na+] were constant. Of the amino acids only glycine correlated positively with high cereulide production. Glycine is used worldwide as food additive (E 640), flavor modifier, humectant, acidity regulator, and is permitted in the European Union countries, with no regulatory quantitative limitation, in most types of foods. B. subtilis group members are endospore-forming bacteria ubiquitous in the environment, similar to B. cereus in this respect. Bacillus species other than B. cereus have only sporadically been identified as causative agents of food-borne illnesses. We found (Paper IV) that food-borne isolates of B. subtilis and B. mojavensis produced amylosin. It is possible that amylosin was the agent responsible for the food-borne illness, since no other toxic substance was found in the strains. This is the first report on amylosin production by strains isolated from food. We found that the temperature requirement for amylosin production was higher for the B. subtilis strain F 2564/96, a mesophilic producer, than for B. mojavensis strains eela 2293 and B 31, psychrotolerant producers. We also found that an atmosphere with low oxygen did not prevent the production of amylosin. Ready-to-eat foods packaged in micro-aerophilic atmosphere and/or stored at temperatures above 10 °C, may thus pose a risk when toxigenic strains of B. subtilis or B. mojavensis are present.

We search for b to s\mu^+\mu^- transitions in B meson (B^+, B^0, or B^0_s) decays with 924pb^{-1} of p pbar collisions at sqrt(s)=1.96 TeV collected with the CDF II detector at the Fermilab Tevatron. We find excesses with significances of 4.5, 2.9, and 2.4 standard deviations in the B^+ to \mu^+\mu^-K^+, B^0 to \mu^+\mu^-K^*(892)^0, and B_s^0 to \mu^+\mu^-\phi decay modes, respectively. Using B to J/psi h (h = K^+, K^*(892)^0, phi) decays as normalization channels, we report branching fractions for the previously observed B^+ and B^0 decays, BR(B^+ to \mu^+\mu^-K^+)=(0.59\pm0.15\pm0.04) x 10^{-6}, and BR(B^0 to \mu^+\mu^-K^*(892)^0)=(0.81\pm0.30\pm0.10) x 10^{-6}, where the first uncertainty is statistical, and the second is systematic. These measurements are consistent with the world average results, and are competitive with the best available measurements. We set an upper limit on the relative branching fraction BR(B_s^0 to \mu^+\mu^-\phi)/BR(B_s^0 to J/\psi\phi)

We search for †’sμ+μ- transitions in B meson (B+, B0, or Bs0) decays with 924  pb-1 of ppÌ… collisions at √s=1.96  TeV collected with the CDF II detector at the Fermilab Tevatron. We find excesses with significances of 4.5, 2.9, and 2.4 standard deviations in the B+→μ+μ-K+, B0→μ+μ-K*(892)0, and Bs0→μ+μ-Ï• decay modes, respectively. Using †’J/ψh (h=K+, K*(892)0, Ï•) decays as normalization channels, we report branching fractions for the previously observed B+ and B0 decays, B(B+→μ+μ-K+)=(0.59±0.15±0.04)×10-6, and B(B0→μ+μ-K*(892)0)=(0.81±0.30±0.10)×10-6, where the first uncertainty is statistical, and the second is systematic. We set an upper limit on the relative branching fraction B(Bs0→μ+μ-Ï•)/B(Bs0→J/ψϕ)<2.6(2.3)×10-3 at the 95(90)% confidence level, which is the most stringent to date.

The lowest T = 2 states have been identified and studied in the nuclei 12C, 12B, 20F and and 28Al. The first two of these were produced in the reactions 14C(p,t)12C and 14C (p,3He)12B, at 50.5 and 63.4 MeV incident proton energy respectively, at the Oak Ridge National Laboratory. The T = 2 states in 20F and 28Al were observed in (3He,p) reactions at 12-MeV incident energy, with the Caltech Tandem accelerator.

The results for the four nuclei studied are summarized below:

(1) 12C: the lowest T = 2 state was located at an excitation energy of 27595 ± 20 keV, and has a width less than 35 keV.

(2) 12B: the lowest T = 2 state was found at an excitation energy of 12710 ± 20 keV. The width was determined to be less than 54 keV and the spin and parity were confirmed to be 0+. A second 12B state (or doublet) was observed at an excitation energy of 14860 ± 30 keV with a width (if the group corresponds to a single state) of 226 ± 30 keV.

(3) 20F: the lowest T = 2 state was observed at an excitation of 6513 ± 5 keV; the spin and parity were confirmed to be 0+. A second state, tentatively identified as T = 2 from the level spacing, was located at 8210 ± 6 keV.

(4) 28Al: the lowest T = 2 state was identified at an excitation of 5997 ± 6 keV; the spin and parity were confirmed to be 0+. A second state at an excitation energy of 7491 ± 11 keV is tentatively identified as T = 2, with a corresponding (tentative) spin and parity assignment Jπ = 2+.

The results of the present work and the other known masses of T = 2 states and nuclei for 8 ≤ A ≤ 28 are summarized, and massequation coefficients have been extracted for these multiplets. These coefficients were compared with those from T = 1 multiplets, and then used to predict the mass and stability of each of the unobserved members of the T = 2 multiplets.

O efeito magnetocalórico, i.e., o aquecimento e/ou resfriamento de um material magnético sob variação do campo magnético aplicado é a base da refrigeração magnética.O efeito magnetocalórico é caracterizado pela variação da entropia em um processo isotérmico (O efeito magnetocalórico, i.e., o aquecimento e/ou resfriamento de um material magnético sob variação do campo magnético aplicado é a base da refrigeração magnética. O efeito magnetocalórico é caracterizado pela variação da entropia em um processo isotérmico (ΔSiso) e pela variação da temperatura em um processo adia¡tico ΔTad.Apesar dos inúmeros trabalhos experimentais e teóricos publicados nessa área, muitos aspectos desse efeito ainda não são bem compreendidos.Nesse trabalho discutimos os efeitos da anisotropia sobre as propriedades magnetocalóricas de um sistema de momentos magnéticos localizados. Para essa finalidade, utilizamos um modelo de spins interagentes com um termo de anisotropia uniaxial do tipo DS2 z , onde D é um parâmetro. Nesse modelo, em que o eixo z é a direção de fácil magnetização, a magnitude do parâmetro de anisotropia e a direção do campo magnético aplicado têm um papel fundamental no comportamento das grandezas magnetocalóricas ΔSiso e ΔTad. Realizamos um estudo sistemático para um sistema com J = 1 aplicando o campo magnético em diferentes direções. Os resultados mostram que, quando o campo magnético é aplicado ao longo da direção z, as grandezas magnetocalóricas apresentam o comportamento normal (valores positivos de ΔTad e valores negativos de ΔSiso para ΔB > 0). Quando o campo magnético é aplicado em uma direção diferente do eixo z, as grandezas magnetocalóricas podem apresentar o comportamento inverso (valores negativos de ΔTad e valores positivos de ΔSiso para ΔB > 0) ou o comportamento anômalo (troca de sinal nas curvas de ΔTad e ΔSiso). Resultados equivalentes tam©m foram obtidos para um sistema com J = 7=2.

A esporotricose é uma micose subcutânea, crônica, causada por espécies termo-dimórficas do complexo Sporothrix schenckii. Esta micose apresenta diferentes manifestações clínicas sendo mais comum a forma linfocutânea. Casos graves causados por Sporothrix brasiliensis têm sido descritos recentemente, exigindo um tratamento prolongado com antifúngicos de alta toxicidade como a anfotericina B-desoxicolato ou suas versões menos tóxicas, mas de alto custo. Neste trabalho visamos testar in vitro e in vivo a eficácia de uma nova formulação intravenosa de anfotericina B poliagregada (P-AmB) e testar in vivo sua versão semi-sólida (AmB tópica), comparando-a com o itraconazol (ITC) e a anfotericina B-desoxicolato (D-AmB). Ensaios de susceptibilidade in vitro com S. brasiliensis mostraram que esta espécie é suscetível aos antifúngicos testados. Para os testes de eficácia in vivo foram estabelecidos um modelo de esporotricose disseminada e outro de esporotricose subcutânea, causados por S. brasiliensis. No modelo de esporotricose disseminada camundongos BALB/c foram inoculados intravenosamente com leveduras de S. brasiliensis e, 72 h pós-infecção, tratados sob diferentes regimes terapêuticos: i) uma monoterapia de ITC, D-AmB ou P-AmB; ii) uma combinação terapêutica entre D-AmB e ITC ou P-AmB e ITC; iii) um regime de pulso com D-AmB ou P-AmB. A sobrevivência (n= nove) e a carga fúngica em órgãos internos (n= três, no mínimo) foram avaliadas, sendo observado que o regime de pulso com D-AmB ou P-AmB foi o mais efetivo em prolongar a sobrevivência dos animais e reduzir a carga fúngica nos órgãos, seguido pela combinação terapêutica, porém o tratamento com D-AmB e ITC foi a combinação mais efetiva. A monoterapia com ITC e P-AmB e D-AmB foram menos eficazes, sendo corroborados pelas análises histopatológicas. Ensaios de toxicidade in vivo com as diferentes drogas revelaram que ITC e D-AmB induziram a uma toxicidade hepática e renal nos animais, respectivamente, mas P-AmB não induziu a nenhuma toxicidade. Nos ensaios de citoxicidade in vitro foi observado que ITC foi a menos citotóxica e hemolítica e a mais seletiva das drogas testadas, seguida por P-AmB, que foi menos citotóxica e mais seletiva que D-AmB. No modelo de esporotricose subcutânea camundongos da mesma linhagem foram inoculados por via subcutânea com conídios de S. schenckii e de S. brasiliensis (n=9/ grupo). Os animais infectados com S. brasiliensis apresentaram regressão das lesões primárias e disseminação. Usando o modelo de esporotricose subcutânea murina causada por S. brasiliensis testamos peliminarmente a formulação tópica de AmB poliagregada, que reduziu a extensão das lesões de animais infectados. Este é o primeiro trabalho a avaliar diferentes regimes de tratamento da esporotricose disseminada murina causada por S. brasiliensis utilizando ITC, D-AmB e uma nova formulação menos tóxica de anfotericina B poliagregada. O estudo revelou que o regime de pulso foi o mais eficaz para as formulações intravenosas de AmB. Nosso estudo tam©m estabeleceu pioneiramente um modelo de esporotricose subcutânea induzido por S. brasiliensis, que se revelou uma ferramenta útil para comparar a virulência das espécies do complexo S. schenckii e para testar a eficácia de antifúngicos contra essas novas espécies.
