The concept of sequential injection chromatography (SIC) was exploited to automate the fluorimetric determination of amino acids after pre-column derivatization with ophthaldialdehyde (OPA) in presence of 2-mercaptoethanol (2MCE) using a reverse phase monolithic C(18) stationary phase. The method is low-priced and based on five steps of isocratic elutions. The first step employs the mixture methanol: tetrahydrofuran: 10 mmol L(-1) phosphate buffer (pH 7.2) at the volumetric ratio of 8:1:91; the other steps use methanol: 10 mmol L-1 phosphate buffer (pH 7.2) at volumetric ratios of 20:80, 35:65, SO:SO and 65:35. At a flow rate of 10 mu L s(-1) a 25 mm long-column was able to separate aspartic acid (Asp), glutamic acid (Glu), asparagine (Asn), serine (Ser), glutamine (Gln), glycine (Gly), threonine (Thr), citruline (Ctr), arginine (Arg), alanine (Ala), tyrosine (Tyr), phenylalanine (Phe), ornithine (Orn) and lysine (Lys) with resolution >1.2 as well as methionine (Met) and valine (Val) with resolution of 0.6. Under these conditions isoleucine (Ile) and leucine (Leu) co-eluted. The entire cycle of amino acids derivatization, chromatographic separation and column conditioning at the end of separation lasted 25 min. At a flow rate of 40 mu L s(-1) such time was reduced to 10 min at the cost of resolution worsening for the pairs Ctr/Arg and Orn/Lys. The detection limits varied from 0.092 mu mol L(-1) for Tyr to 0.51 mu mol L(-1) for Orn. The method was successfully applied to the determination of intracellular free amino acids in the green alga Tetraselmis gracilis during a period of seven days of cultivation. In a hospital environment, these bacteria can be spread by insects such as ants, which are characterized by high adaptability to the urban environment. Staphylococcus is a leading cause of hospital infection. In Europe, Latin America, USA and Canada, the group of coagulase negative staphylococci (CoNS) is the second leading cause of these infections, according to SENTRY (antimicrobial surveillance program- EUA). In this study, we investigated the potential of ants (Hymenoptera: Formicidae) as vehicle mechanics of Staphylococcus bacteria in a public hospital, in Natal-RN. The ants were collected, day and night, from June 2007 to may 2008, in the following sectors: hospitals, laundry, kitchen, blood bank. The ants were identified according to the identification key of Bolton, 1997. For the analysis of staphylococci, the ants were incubated in broth Tryptic Soy Broth (TSB) for 24 hours at 35 º C and then incubated on Mannitol Salt Agar. The typical colonies of staphylococci incubated for 24 hours at 35 ° C in Tryptic Soy Agar for the characterization tests (Gram stain, catalase, susceptibility to bacitracin and free coagulase). The identification of CoNS was performed through biochemical tests: susceptibility to novobiocin, growth under anaerobic conditions, presence of urease, the ornithine decarboxylation and acid production from the sugars mannose, maltose, trehalose, mannitol and xylose. The antimicrobial susceptibility examined by disk-diffusion technique. The technique of Polymerase Chain Reaction was used to confirm the presence of mecA gene and the ability to produce biofilm was verified by testing in vitro using polystyrene inert surface, in samples of resistant staphylococci. Among 440 ants, 85 (19.1%) were carrying coagulase-negative staphylococci (CoNS) of the species Staphylococcus saprophyticus (17), Staphylococcus epidermidis (15), Staphylococcus xylosus (13), Staphylococcus hominis hominis (10), Staphylococcus lugdunensis (10), Staphylococcus warneri (6), Staphylococcus cohnii urealyticum (5), Staphylococcus haemolyticus (3), Staphylococcus simulans (3), Staphylococcus cohnii cohnii (2), and Staphylococcus capitis (1). No Staphylococcus aureus was found. Among the isolates, 30.58% showed resistance to erythromycin. Two samples of CoNS (2.35%), obtained from the ant Tapinoma melanocephalum collected in the post-surgical female ward, S. Hominis hominis and S. lugdunensis harbored the mecA gene and were resistant to multiple antibiotics, and the specie S. hominis hominis even showed to be a biofilm producer. This study proves that ants act as carriers of multidrug-resistant coagulase-negative Staphylococci and biofilm producers and points to the risk of the spreading of pathogenic microorganisms by this insect in the hospital environment

Arginine was hypothesized to be a model compound in the present study on molecular forms of indispensable amino acid (IAA) dietary supplementation. Juvenile South American pacu (Piaractus mesopotamicus) were fed diets containing arginine in a protein base (casein-wheat gluten or casein-gelatin), or the casein-wheat gluten base supplemented with dipeptide or free arginine at two levels (5 and 10 g kg(-1)). Growth and protein efficiency ratios were significantly affected by diets, but not by arginine molecular form. Three free dispensable amino acids (DAA) and four IAA in plasma were affected by diet, but plasma arginine concentrations did not differ. Plasma urea concentrations, being very low in the pacu, and hepatic arginase activities, were not affected by diet (P = 0.10-0.11), but together with plasma ornithine, mirrored the growth data. Molecular form of arginine supplementation, free or dipeptide, significantly changed several free IAA (Phe, Leu, Ile, His) and urea, with a higher mean plasma concentration in dipeptide fed fish. The dietary treatments, or molecular form of the arginine supplementation, did not change proximate composition, except that calcium levels decreased with higher dietary arginine supplementation level. The present study indicates that dipeptides can provide IAA to pacu, and that arginine supplemented in this form is utilized as efficiently as in free form.

A S. Pullorum (SP) é muito semelhante à S. Gallinarum (SG), agentes da Pulorose e Tifo aviário, respectivamente, sendo que as duas enfermidades são responsáveis por perdas econômicas no setor avícola. SP e SG são de difícil diferenciação em procedimento laboratorial rotineiro, mas uma prova bioquímica muito utilizada na distinção das duas refere-se à capacidade de assimilar o aminoácido ornitina: SP descarboxila este aminoácido enquanto SG não. No entanto, o isolamento de cepas com comportamento bioquímico atípico, tem dificultado tal diferenciação. Um dos genes relacionados à assimilação do aminoácido ornitina, denomina-se gene speC, o qual está presente nos dois sorovares. Analisando 21 amostras de SP e 15 de SG com a utilização da PCR não foi possível realizar a diferenciação dos dois sorovares pois os fragmentos gerados eram idênticos. Posteriormente, com o uso da técnica de tratamento enzimático com a enzima de restrição Eco RI, foi possível observar que o padrão de bandas gerado em cada sorovar era diferente, mesmo quando amostras que apresentavam comportamento bioquímico atípico eram analisadas. Tal fato permitiu a padronização da técnica para ser utilizada na diferenciação entre os sorovares Pullorum e Gallinarum de maneira rápida e segura.

Although Salmonella Pullorum and Salmonella Gallinarum cause different diseases in poultry, they are very similar. Both are non-motile and present the same somatic antigenic structure. They are differentiated by biochemical tests. Certain atypical strains are very difficult to distinguish. They do not produce the expected results when dulcitol and ornithine descarxboxylase tests are performed. Therefore, additional tests could be helpful. Many studies have chose the part I of the gene that encodes flagellin (fliC) to differentiate serotypes. Most Salmonella strains have two structural genes (fliC and fliB) that encode flagellins. Non-motile strains generally present these structural genes, but are not able to build a functional flagellum. It was demonstrated that enzymatic restriction of the amplified fliC gene using Hinp1I enzyme can differentiate SG from SP. In the present study, this method was adopted to analyze 14 SP and 22 SG strains, including some strains with atypical results in biochemical tests assessing the utilization of dulcitol and ornithine. The results showed that all SG strains were broken by the enzyme, whereas the 14 SP strains were not.

Most studies on the antioxidants, lipoic acid (LA) and ascorbic acid (AA), focused on species that, unlike teleost fish, are not scurvy-prone, and are able to synthesize AA. The antioxidant properties of LA may make it useful in aquaculture nutrition, but several effects must first be investigated, and we address here plasma free amino acids (FAA). In mammals, LA and AA in high doses were claimed to alter plasma FAA profile; to our knowledge, however, no data are available in fish. We therefore studied the effects of dietary LA and AA on plasma FAA in the South American teleost fish pacu, which is being used increasingly in aquaculture. LA treatment decreased concentrations of 18 of 23 individual FAA; specifically, dispensable and total FAA were significantly affected. Ornithine was elevated (+26%) in LA-treated fish and significantly decreased ratios of plasma [Arg]/[Orn] and other individual [FAA]/[Orn] were observed. LA and AA both affected sulfur FAA concentrations. Plasma cystine levels were significantly increased in the LA-supplemented groups. AA had little effect on most amino acids, and no interaction with LA was detected. AA supplementation did, however, significantly lower taurine (-42%) and cystathionine (-31%) levels in plasma. No effect on the branched chain:aromatic amino acid ratios was observed. The data indicate that at the dietary level studied, LA and AA independently affect selected plasma FAA in pacu, and suggest that any use of LA in particular as a dietary supplement should take into account an altered plasma FAA profile.

Th1 cells, in cooperation with activated macrophages, are required to overcome Yersinia enterocolitica infection in mice. The pathway macrophages utilize to metabolize arginine can alter the outcome of inflammation in different ways. The objective of this study was to verify the pattern of macrophages activation in Y. enterocolitica infection of BALB/c (Yersinia-susceptible) and C57BL/6 (Yersinia-resistant) mice. Both strains of mice were infected with Y. enterocolitica O:8 WA 2707. Peritoneal macrophages and spleen cells were obtained on the 1st, 3rd and 5th day post-infection. The iNOS and the arginase activities were assayed in supernatants of macrophage cultures, by measuring their NO/citrulline and ornithine products, respectively. TGFβ-1 production was also assayed. The Th1 and Th2 responses were evaluated in supernatants of lymphocyte cultures, by IFN-γ and IL-4 production. Our results showed that in the early phase of Y. enterocolitica infection (1st and 3rd day), the macrophages from C57BL/6 mice produced higher levels of NO/citrulline and lower levels of ornithine than macrophages from BALB/c mice. The infection with Y. enterocolitica leads to an increase in the TGF-β1 and IL-4 production by BALB/c mice and to an increase in the IFN-γ levels produced by C57BL/6 mice. These results suggest that Y. enterocolitica infection leads to the modulation of M1 macrophages in C57Bl/6 mice, and M2 macrophages in BALB/c mice. The predominant macrophage population (M1 or M2) at the 1st and 3rd day of infection thus seems to be important in determining Y. enterocolitica susceptibility or resistance.

A Streptomyces was isolated from poultry plant wastewater, showed high keratinolytic activity when cultured on feather meal medium. Optimum keratinolytic activity was observed at 40°C and pH 8.0. The enzyme also showed to be stable between 40 and 60°C. The keratinolytic activity was not inhibited by EDTA, DMSO and Tween 80. On the other hand, CaCl2, ZnCl2, and BaCl2 slightly inhibited the keratinolytic activity. The Streptomyces isolated might be useful in leather, keratin waste treatment, animal feeding industry, and also cosmetic industry. © 2008 Academic Journals.

With the emergence of new genetic lines due to intense breeding improvement on swine production in recent years, there is the need to adapt more accurately diets for the current sows, which have higher nutritional demands. The use of functional amino acids aimsto optimize the sows production and among these amino acids arginine has excelled. Arginine is involved in several important metabolic pathways, for example, it serves as a substrate forsynthesis of protein, creatine, nitric oxide, polyamines, citrulline, agmatine, ornithine, proline, and glutamate. It also helps to stimulate the secretion of some hormones such as insulin, prolactin, and growth hormone.As arginine plays such important roles, its supplementation has been suggested in lactation feed once it may enhance the development of the mammary gland and milk nutritional profile, thus, providing a better piglet development.Thus, the objective was to evaluate the effect of lactation feed supplementation with L-Arginine on the productive performance of primiparoussows and their respective litter.One hundred forty sows from the same genetic lineage on a commercial farm, located in the city of Oliveira, MG were used in this study, in a completely randomized design with five treatments: control diet without amino acid supplementation and four diets with increasing levels of L-Arginine supplementation (containing 98.5% purity) - 0.5, 1.0, 1.5, and 2.0%. Each treatment hadtwenty-eight swine sows, and the experimental unit was the sowand its litter.It was used ‘on top’ amino acid supplementation.All data was submitted to variance analysis using the SAEG Software: version 9.1 (SAEG, 2005).The data relating to days of lactation were compared by Tukey test (5%). L-Arginine supplementation levels in lactation feed did not influence (P>0.05) average daily feed intake, body condition variables, and blood parameters of the sows (urea, creatinine, and non-esterified fatty acids) as well as it did not affect the dry matter, crude protein, and amino acid profile of milk and the litter performance. There was effect (P<0.05) of days of lactation on the percentage of crude protein and amino acids in milk, which reduced througout the days of lactation. The L-Arginine supplementation on the lactation diet at levels of 0.5, 1.0, 1.5, and 2.0% did not influence the sow and its respective litter performance.

Two experiments were carried out to evaluate the effect of adding glutamine, fish oil or yeast cellular wall to the diet of weaned piglets on the expression of the enzyme ornithine decarboxylase (ODC), the protein content and DNA in small intestine samples and on performance. In the first experiment, 24 weaned piglets were used to measure the performance at the phases prestarter, starter 1 and starter 2. Four diets were tested (T1 -basal diet (BD); T2 -BD + 1% of glutamine; T3 -BD + 0,2% of yeast cellular wall; T4 -BD + 5% of fish oil). At the second experiment 45 weaned piglets were used and distributed in a randomized block design, in factorial outline with four diets and three slaughter ages (on the day of weaning, on the seventh and fourteenth days postweaning). The tested diets did not alter the piglets' performance in none of the phases. There was reduction of the expression of the ODC enzyme, of the protein concentration and of the relationship protein/DNA to the seven days postweaning, with increase of the values on the 14th day, evidencing a state of hypotrophy of the mucous membrane, suggesting that the process of protein synthesis in the small intestine was diminished in the first week after weaning, but it presented recovery signs in the second week.
