A resposta do amendoim à calagem tem sido atribuída ao fornecimento de cálcio, mas também tem sido observada maior absorção de nitrogênio. Um experimento foi desenvolvido em casa de vegetação, em vasos de polietileno, utilizando-se um Latossolo Vermelho distrófico textura média, para estudar os efeitos da calagem e da aplicação de cobalto e molibdênio sobre a nodulação e a absorção de nitrogênio pelo amendoim, cultivar Tatu. O delineamento experimental empregado foi o de blocos ao acaso, em esquema fatorial 4x3, com quatro repetições. Os tratamentos do fatorial constituíram-se de quatro aplicações de nutrientes nas sementes (sem aplicação; 0,16 g kg-1 de Co; 0,58 g kg-1 de Mo e 0,16 + 0,58 g kg-1 de Co e Mo, respectivamente) e três doses de calcário (0, 4 e 6 t ha-1). A aplicação de cobalto nas sementes não exerceu efeito sobre a nodulação e a absorção de nitrogênio pelo amendoim. A calagem aumentou a absorção de nitrogênio pelo amendoim, provavelmente por causar melhoria na fixação simbiótica do N2, em decorrência da formação de maior número de nódulos. Apesar do molibdênio ter aumentado a matéria seca de nódulos na planta de amendoim, a sua aplicação não influenciou a absorção de nitrogênio, mostrando que a redução da toxicidade de manganês pela calagem foi mais importante do que o aumento da disponibilidade de molibdênio para a formação de nódulos e a fixação simbiótica do N2.

Soybean (Glycine max (L.) Merrill) is known to have a high ability as a potassium extractor, and different cultivars show different potassium requirements. An experiment was run to study the potassium nutrition of soybean as related to plant characteristics. Six soybean cultivars (FT-2, Bossier, IAC-11, IAC-17, IAC-18 and IAC-19) were grown in 6 kg pots filled with the topsoil of a Dark Red Latosol (sandy loam), either with and without K fertilization. The plants were harvested 70 days after emergence. Soybean response to potassium was not related to growth habit or group of maturation. There was a different response to K. The cultivars IAC-18 and FT-2 were less tolerant to K deficiency. Potassium deficiency in the leaves was not related to top dry matter production. With K fertilization soybean plants showed small root volume and higher ratio canopy/root. With high K in soil, all of the cultivars showed higher nodulation.

Insects manifest effective immune responses that include both cellular and humoral components. Morphological and quantitative aspects of cellular and Immoral cooperation during nodule formation in Chrysomya megacephala hemolymph against Saccharomyces cerevisae yeast cells were demonstrated for the first time. The analyses were performed in non-injected larvae (NIL), saline-injected larvae (SIL) and yeast-injected larvae (YIL). The hemolymph of injected groups was collected 0.5, 1, 2, 4, 12, 24, 36, or 48-h post-injection. Morphological aspects of YIL nodulation were investigated using transmission electron microscopy (TEM). Quantitative analyses consisted of total (THC) and differential hemocyte counts (DHC) in all the groups and total yeast count (TYC) in YIL, which were performed in an improved Neubauer chamber. Nodule formation was initiated at approximately 2-h post-injection. Twelve hours after the injection, TEM revealed the presence of an amorphous membrane, at the same time that circulating hemocyte number decreased significantly contrasting the increase of yeast number. Our results showed the ability of C megacephala hemolymph to perform humoral encapsulation when hemocyte population is insufficient to eliminate the microorganisms, warranting consideration in future investigations on the relative roles played by cellular and humoral elements of innate immunity of this calliphorid. The relationship between soil fungi and actinomycetes, with rhizobia that produce nodulation in Neonotonia wightii (perennial soybean) was studied in culture medium in order to obtain material to be used in mixed inoculations of this legume. A field experiment was designed to provide soil samples and isolate microorganisms belonging to these groups that are capable of interfering in the development of two selected rhizobium strains. Results show that the Bradyrhizobium strains used in the experiment, which are efficient in nodulation of Neonotonia wightii, are highly sensitive to substances produced in the culture medium by actinomycetes and fungi.

Several serological methods have been used for the characterization and identification of soybean bradyrhizobia. However, some problem were non-reactivity of certain strains and cross-reactivity among others. Since lipopolysaccharide (LPS) can often be used in strain identification, the objective was to investigate the antigenic properties and polyacrylamide gel electrophoretic pattern of 12 Brazilian strains of Bradyrhizobium japonicum that nodulate soybean and to compare them to standard strains. The close correlation between the LPS patterns obtained by SDS-PAGE and the serological analysis permitted us to assign the strains to nine groups different or the same as the standard strains.

It is well established that nitrate is a potent inhibitor of nodulation and nitrogen fixation in legumes. The objective of this study was to demonstrate the relative insensitivity of these processes to nitrate with Calopogonium mucunoides, a tropical South American perennial legume, native to the cerrado (savannah) region. It was found that nodule number was reduced by about half in the presence of high levels of nitrate (15 mM) but nodule growth (total nodule mass per plant) and nitrogen fixation (acetylene reduction activity and xylem sap ureide levels) were not affected. Other sources of N (ammonium and urea) were also without effect at these concentrations. At even higher concentrations (30 mM), nitrate did promote significant inhibition (ca. 50%) of acetylene reduction activity, but no significant reduction in xylem sap ureides was found. The extraordinary insensitivity of nodulation and N2 fixation of C. mucunoides to nitrate suggests that this species should be useful in studies aimed at elucidating the mechanisms of nitrate inhibition of these processes. © 2010 Springer Science+Business Media B.V.

The tree leguminous gliricídia (Gliricidia sepium), acácia (Acacia mangium), leucena (Leucaena leucocephala) and sombreiro (Clitoria fairchildiana) are indicated for agroforestry systems, reclamation of degraded lands, reforestation and other purposes in the wet tropic. Despite the importance of legumes the preamazon region it is lacking in information about the symbiotic capacity and diversity of indigenous rhizobia of these legumes. The aim of this work was to evaluate the phenotypic and genetic diversity of rhizobia species nodulating gliricidia sombreiro, leucena and acacia in the Maranhão pre-Amazon region and authenticate isolates of these species in siratro (Macroptilium atropurpureum). For this they were installed two experiments. Sampling was carried out on a alley cropping system, was sampled 20 plants of each species by collecting 10 nodules per plant. It was made isolation, cultural characterization, partial 16S rRNA gene sequencing and analysis of the symbiotic ability of bacterial strains with siratro. The authentication experiment was done in two steps for each legume (gliricidia, acácia, sombreiro and leucena), in the greenhouse and in a completely randomized design with three replications with sterile Hoagland nutrient solution as substrate. Gliricídia, Sombreiro, leucena and acacia are colonized by distinct groups of rhizobia. Gliricidia nodulate preferably with Rhizobium, sombreiro and acacia nodulate preferably with Bradyrhizobium and leucena has Mesorhizobium main symbiote. Endophytic strains of ten genera were found colonizing the gliricidia, sombrero, leucena and acacia nodules and a strain of Arthrobacter sp. had a positive nodulation with siratro. This is the first report on isolation of Methylobacterium sp. in gliricidia nodules and endophytic ability of Terriglobus sp. strains. Indigenous strains of pre-Amazon region of Bradyrhizobium, Mesorhizobium and Rhizobium genus nodulate with siratro, but are ineficiente or had low efficiency to promote their growth.
