Broiler chicks aged 12 h after hatching were allotted according to a block design in a 7 x 2 factorial schedule of 14 treatments and four replications of 50 chicks each one. The main experimental factors were fasting for 0, 6, 12, 18, 24, 30, and 36 h after chick placement and sex. Independent of sex, fasting had a negative linear effect on weight and productivity of broilers at market age (42 d) without affecting feed conversion or mortality index. Groups subjected to 18 and 36 h of fasting after placement, corresponding to 30 and 48 h posthatching fasting, had lower biometrical values for small intestine (length, weight, and size; villus height; and crypt depth) than chicks fed immediately after placement. According to the Pearson test, BW of birds at 21 and 42 d were significantly correlated to BW at 7 d (r = 0.77) and 21 d (r = 0.45), respectively. Males performed better than females but had higher mortality rates. Fasting did not influence serum concentrations of corticosterone or sexual steroid hormones. Nevertheless, early signs of sexual dimorphism arose from the high estradiol (E2) concentration on female serum. Heterophil:lymphocyte ratio was not different among treatments, indicating that early fasting did not seem to be a stress factor 21 or 42 d after fasting. The results suggested a maximum fasting of 24 h after hatching in order to preserve broiler productivity at market age.

A field trial was conducted designed in a completely randomized block in a 4 x 3 factorial arrangement to evaluate the application of nitrogen doses (N) (0, 40, 80 and 160 kg/ha) on the morphogenical characteristics and dry matter partition of three forage grasses (Panicum maximum cvs. Mombasa and Tanzania and Brachiaria sp. Hybrid Mulato). The leaf appearance (LAR, leaf/day) and stretching (LER; mm/day) rates, the number of green leaves per tiller (NLT) and the average weight of tillers (MTW; g) presented s positive linear response to the N dose while the phyllochron (Phil; day/leaves) showed a negative linear response. The highest LER, IAL and final leaf length (FLL; cm) occurred in the Mombaca and Tanzania grasses, while the highest LAR occurred in the Mulato grass. There was a negative quadratic effect of the N dose on the stem elongation rate (SER; mm/day) and LF. The Mombaca and Tanzania grasses presented the highest SER; however, in just two forages. The production of total dry matter (TDM; kg/ha), leaves (LDM; kg/ha) and stems (SDM; kg/ha) increased linearly and quadratically with the N dose, respectively, for the Mombaca and Tanzania grasses. There was a high positive correlation among DM, LDM and SDM and the Mombaca grass MTW. The dry matter production and morphogenic characteristics were influenced by the nitrogen fertilization as a result of the substantial increase in the flow of tissues stimulated by fertilization, proving the importance of N for forage biomass accumulation.

Com o objetivo de avaliar as características de carcaça e a alometria dos cortes comerciais e dos tecidos de cabritos F1 Boer × Saanen, 35 animais foram abatidos ao atingirem 5, 10, 15, 20 e 25 kg de peso vivo (PV). A dieta dos animais foi composta de leite de vaca nos primeiros 49 dias e ração à vontade do sétimo dia até o abate. Os cortes foram obtidos após o resfriamento da carcaça e a perna foi dissecada em músculo, osso e gordura. O PV teve efeito linear decrescente no rendimento de carcaça fria e na área de olho-de-lombo por kg de carcaça. Os pesos de perna, paleta e pescoço em relação à carcaça fria decresceram linearmente, mas houve efeito quadrático sobre o rendimento de costelas e lombo. O crescimento de paleta, pescoço e perna foi isométrico (b=1) ao do corpo, enquanto o das costelas e do lombo foi mais lento (b¹ 1). Os músculos da perna cresceram igualmente, a gordura mais lenta e os ossos mais rapidamente que a perna, enquanto o desenvolvimento da gordura subcutânea foi mais tardio que o da intermuscular. Para obtenção de carcaça de 8 a 11 kg com rendimento superior a 44%, boa proporção de músculo e gordura com menor perda durante o resfriamento, recomenda-se abater os animais com PV entre 20 e 25 kg, mas, se o objetivo for carcaça de menor peso, o abate dos animais deve ser feito ao final do aleitamento com aproximadamente 10 kg de peso corporal.

Background. Identifying changes in the oral health status of older populations, and their predictors and explanations, is necessary for public health planning. The authors assessed patterns of change in oral health-related quality of life in a large cohort of older adults in Brazil during a five-year period and evaluated associations between baseline characteristics and those changes. Methods. The sample consisted of 747 older people enrolled in a Brazilian cohort study called the Health, Well-Being and Aging (Saude, Bem-estar e Envelhecimento [SABE]) Study. Trained examiners measured participants' self-perceived oral health by using the General Oral Health Assessment Index (GOHAI). The authors calculated changes in the overall GOHAI score and in the scores for each of the GOHAI's three dimensions individually by subtracting the baseline score from the score at follow-up. A positive difference indicated improvement in oral health, a negative difference indicated a decline and a difference of zero indicated no change. Results. The authors found that 48.56 percent of the participants experienced a decline in oral health and 33.48 percent experienced an improvement. Participants with 16 or more missing teeth and eight or more years of education were more likely to have an improvement in total GOHAI score. Deterioration was more likely to occur among those with two or more diseases. Improvement and decline in GOHAI functional scores were related to the number of missing teeth. The authors found no significant model for the change in the psychosocial score, and Self-rated general health was the only variable related to both improvement and decline in pain or discomfort scores. Conclusions. The authors observed a bidirectional change in self-perceived oral health, with deterioration predominating. The strongest predictor of improvement in the total GOHAI score was the number of missing teeth, whereas the number of diseases was the strongest predictor of deterioration. Clinical Implications. Dental professionals and policymakers need to know the directions of change in older adults' oral health to establish treatment priorities and evaluate the impact of services directed at this population.

Abstract Introduction Mechanisms underlying inotropic failure in septic shock are incompletely understood. We previously identified the presence of exosomes in the plasma of septic shock patients. These exosomes are released mainly by platelets, produce superoxide, and induce apoptosis in vascular cells by a redox-dependent pathway. We hypothesized that circulating platelet-derived exosomes could contribute to inotropic dysfunction of sepsis. Methods We collected blood samples from 55 patients with septic shock and 12 healthy volunteers for exosome separation. Exosomes from septic patients and healthy individuals were investigated concerning their myocardial depressant effect in isolated heart and papillary muscle preparations. Results Exosomes from the plasma of septic patients significantly decreased positive and negative derivatives of left ventricular pressure in isolated rabbit hearts or developed tension and its first positive derivative in papillary muscles. Exosomes from healthy individuals decreased these variables non-significantly. In hearts from rabbits previously exposed to endotoxin, septic exosomes decreased positive and negative derivatives of ventricular pressure. This negative inotropic effect was fully reversible upon withdrawal of exosomes. Nitric oxide (NO) production from exosomes derived from septic shock patients was demonstrated by fluorescence. Also, there was an increase in myocardial nitrate content after exposure to septic exosomes. Conclusion Circulating platelet-derived exosomes from septic patients induced myocardial dysfunction in isolated heart and papillary muscle preparations, a phenomenon enhanced by previous in vivo exposure to lipopolysaccharide. The generation of NO by septic exosomes and the increased myocardial nitrate content after incubation with exosomes from septic patients suggest an NO-dependent mechanism that may contribute to myocardial dysfunction of sepsis.

Many potential diltiazem related L-VDCC blockers were developed using a multidisciplinary approach. This current study was to investigate and compare diltiazem with to the newly developed compounds by mouse Langendorff-perfused heart, Ca2+-transient and on recombinant L-VDCC. Twenty particular compounds were selected by the ligand-based virtual screening procedure (LBVS). From these compounds, five of them (5b, M2, M7, M8 and P1) showed a potent and selective inotropic activity on guinea-pig left atria driven 1 Hz. Further assays displayed an interesting negative inotropic effect of M2, M8, P1 and M7 on guinea pig isolated left papillary muscle driven at 1 Hz, a relevant vasorelaxant activity of 5b, M2, M7, M8 and P1 on K+-depolarized guinea-pig ileum longitudinal smooth muscle and a significant inhibition of contraction of 5b, M2, M8 and P1 on carbachol stimulated ileum longitudinal smooth muscle. Wild-type human heart and rabbit lung α1 subunits were expressed (combined with the regulatory α2δ and β3 subunits) in Xenopus Leavis oocytes using a two-electrode voltage clamp technique. Diltiazem is a benzothiazepine Ca2+ channel blocker used clinically for its antihypertensive and antiarrhythmic effects. Previous radioligand binding assays revealed a complex interaction with the benzothiazepine binding site for M2, M7 and M8. (Carosati E. et al. J. Med Chem. 2006, 49; 5206). In agreement with this findings, the relative order of increased rates of contraction and relaxation at lower concentrations s(≤10-6M) in unpaced hearts was M7>M2>M8>P1. Similar increases in Ca2+ transient were observed in cardiomyocytes. Diltiazem showed negative inotropic effects whereas 5b had no significant effect. Diltiazem blocks Ca2+current in a use-dependent manner and facilitates the channel by accelerating the inactivation and decelerating the recovery from inactivation. In contrast to diltiazem, the new analogs had no pronounced use-dependence. Application of 100 μM M8, M2 showed ~ 10% tonic block; in addition, M8, M2 and P1 shifted the steady state inactivation in hyperpolarized direction and the current inactivation time was significantly decreased compared with control (219.6 ± 11.5 ms, 226 ± 14.5 vs. 269 ± 12.9 vs. 199.28 ± 8.19 ms). Contrary to diltiazem, the recovery from the block by M8 and M2 was comparable to control. Only P1 showed a significantly decrease of the time for the recovery from inactivation. All of the compounds displayed the same sensitivity on the Ca2+ channel rabbit lung α1 except P1. Taken together, these findings suggest that M8, M2 and P1 might directly decrease the binding affinity or allow rapid dissociation from the benzothiazepine binding site.

Zusammenfassung: Thema dieser Arbeit ist der Einfluss von elektrischen Feldern auf die Viskosität nematischer Flüssigkristalle sowie isotroper Polymerlösungen.Für zwei thermotrope nematische Flüssigkristalle, 4-(trans-4'-n-Pentylcyclohexyl)-benzonitril (PCH-5) und 4-n-Pentyl-4'-cyanobiphenyl (5CB), wurde die Viskosität mit elektrorheologischen Messungen in Abhängigkeit von Feldstärke und Scherrate bestimmt. Alle Messwerte kommen bei Auftragung gegen E?/? auf einer Masterkurve zu liegen. Die Anpassung dieser Daten mit einer auf der Theorie von Ericksen und Leslie basierenden Gleichung liefert gleichzeitig die Scherviskositäten ? und ? sowie die Rotationsviskosität ? in guter Übereinstimmung mit Literaturwerten. Elektrorheologische Messungen stellen somit eine Methode zur Bestimmung dieser Viskositätskoeffizienten für Nematen dar, wenn diese eine ausreichend große positive dielektrische Anisotropie aufweisen. Bei den isotropen Polymerlösungen ließ sich der elektrorheologische Effekt des Systems Decalin/Polystyrol im Bereich der Entmischungstemperatur klären. Während im Ein-Phasen-Gebiet nur eine geringe Viskositätsänderung im E-Feld aufgrund elektrohydrodynamischer Strömungen zu beobachten ist, tritt unterhalb der Entmischungstemperatur ein deutlicher, aber nicht reproduzierbarer ER-Effekt auf. Dieser ist auf einen Einfluss des E-Felds auf die Zwei-Phasen-Morphologie zurückzuführen.Für das System Diethylbenzol/Isopentylcellulose wurde ein negativer elektrorheologischer Effekt auf die Dissoziation von assoziierten Polymermolekülen zurückgeführt. Die Dissoziation erfolgt aufgrund der Wirkung des elektrischen Feldes auf freie Ladungen.

Heusler- und Halb-Heusler-Phasen konnten als Verbindungsklasse identifiziert werden, neue Materialien für die Magnetoelektronik bereitzustellen. Auf Basis eines theoretischen Modells konnte das Auftreten eines hohen MR-Effekts auf bestimmte Valenzelektronenzahlen (VEK) präzisiert werden. Dazu muß ein dreiteiliger 'Fingerabdruck' in der Bandstruktur (Sattelpunkt, Spindichtewelle, lokales magnetisches Moment) vorliegen. Es existieren eine Vielzahl von halbmetallischen Ferromagneten in dieser Verbindungsklasse. Die variable Valenzelektronenkonzentrationen, die sich auch aus den hochsymmetrischen Strukturen ergeben, erlauben eine gute Dotierbarkeit der Phasen. Dadurch ist die Möglichkeit gegeben, die Phase exakt mit der geforderten VEK zu synthetisieren. Curietemperaturen > 500 K sind in Hinblick auf die technische Anwendbarkeit notwendig. PdMnTe hat nicht die Voraussetzungen für einen PMR-Effekt in der Bandstrukturrechnung, doch die Nähe zu einer halbmetallischen Zustandsdichte resultiert in einen negativen CMR-Effekt unterhalb des magnetischen Übergangs von MR0 = 18 % bei 4 K. Die Zusammenhänge von Probenpräparation zum magnetischen Sättigungsmoment konnten an Co2CrAl aufgedeckt werden. Die unter Anwendung des vorgestellten Modells synthetisierte Heusler-Phase Co2Cr0.6Fe0.4Al (VEK = 27.8) weisen den erwarteten MR-Effekt auf. Der gemessene PMR-Effekt ist größer als bei den GMR-Systemen (bei geringerer Feldempfindlichkeit) und anderen granularen Materialien wie CrO2 bei 295 K. Co2Cr0.6Fe0.4Al zeigt bei 295 K einen hohen negativen Magnetowiderstand von 30 % bei einem Sättigungsfeld von 0.2 Tesla. Durch die Beimischung von Oxiden und Polymeren sind Komposit-Materialien entwickelt worden, die MR0-Effekte von bis zu 88% mit einer verbesserten Feldempfindlichkeit von 0.1 Tesla bei Al2O3 und 0.05 Tesla bei den Oberflächenbeschichtungen zeigen.
