Contexte L’américanisation des habitudes de vie, notamment de l’alimentation, semble être en grande partie responsable de l’augmentation incessante de l’incidence élevée des maladies cardiovasculaires. La période postprandiale, où l’homéostasie vasculaire et métabolique est mise à l’épreuve, est d’une importance considérable dans le développement ou la prévention de l’athérosclérose et représente maintenant la majeure partie du temps d’éveil. La compréhension de l’influence de la composition d’un repas sur son impact postprandial est donc essentielle dans notre lutte dans la prévention de ces maladies. Objectifs L’objectif principal de ce projet de recherche était d’étudier les réponses endothéliale et métabolique à un repas de type méditerranéen mixte (MMM), puisqu’elles sont inconnues. Ce projet avait aussi pour objectifs d’évaluer l’impact microvasculaire d’un tel repas et de caractériser la composition postprandiale des acides gras plasmatiques. À titre comparatif, ces éléments ont aussi été étudiés suite à un repas riche en gras saturés (HSFAM). Méthodes Vingt-huit (28) hommes sains, exempts de facteurs de risque de maladies cardiovasculaires ont reçu de façon randomisée les deux repas à l’étude. Le MMM, composé de saumon frais et de légumes cuits dans l'huile d'olive, contenait 7.87g de SFA et 2.29g d’acides gras polyinsaturés oméga-3, tandis que le HSFAM, composé d'un sandwich déjeuner avec œuf, saucisse et fromage, contenait 14.78g de SFA. Les mesures de la fonction endothéliale mesurée par échographie brachiale (FMD), de la fonction microvasculaire mesurée par spectroscopie proche de l’infrarouge (NIRS) et de la composition des acides gras plasmatique ont été effectuées à jeun et en période postprandiale. Résultats Deux sous-groupes de répondeurs aux repas à l’étude se sont dégagés de ces travaux. Un premier sous-groupe de sujets ayant une triglycéridémie à jeun élevée, mais normale (hnTG) a démontré des altérations endothéliales seulement suivant le repas HSFAM. Un second sous-groupe de sujets ayant une triglycéridémie plus faible et normale (lnTG) n’a quant à lui pas subi d’altérations endothéliales suivant les deux repas à l’étude. Les sujets hnTG ont aussi démontré une charge triglycéridémique postprandiale (iAUC) plus importante et qui était de surcroît enrichie en acide stéarique suivant la HSFAM et en acide gras polyinsaturés oméga-3 suivant le MMM. L’évaluation par NIRS de la fonction microvasculaire nous révèle un ralentissement de la réoxygénation post-ischémique qui suggère une altération postprandiale du recrutement capillaire chez les sujets hnTG seulement. De telles altérations, qui semblent être plus importantes suivant le HSFAM, pourraient être en partie responsables de l’impact endothélial de ce repas. Conclusions Cet essai clinique démontre donc de façon novatrice qu’un repas MMM n’a pas d’effet endothélial délétère et que cette neutralité est indépendante de la triglycéridémie à jeun. De plus, une triglycéridémie à jeun élevée mais normale semble indiquer des dysfonctions endothéliales et métaboliques à des épreuves nutritionnelles tel un repas HSFAM. La consommation de repas méditerranéens par des individus sains à la triglycéridémie marginale serait bénéfique, peut-être même davantage que pour des individus de triglycéridémie plus faible, dans la prévention de l’athérogénèse et des maladies cardiovasculaires.

Functional near-infrared spectroscopy (fNIRS) acquired with electroencephalography (EEG) is a relatively new non-invasive neuroimaging technique with potential for long term monitoring of the epileptic brain. Simultaneous EEG-fNIRS recording allows the spatio-temporal reconstruction of the hemodynamic response in terms of the concentration changes in oxy-hemoglobin (HbO) and deoxy-hemoglobin (HbR) associated with recorded epileptic events such as interictal epileptic discharges (IEDs) or seizures. While most previous studies investigating fNIRS in epilepsy had limitations due to restricted spatial coverage and small sample sizes, this work includes a sufficiently large number of channels to provide an extensive bilateral coverage of the surface of the brain for a sample size of 40 patients with focal epilepsies. Topographic maps of significant activations due to each IED type were generated in four different views (dorsal, frontal, left and right) and were compared with the epileptic focus previously identified by an epileptologist. After excluding 5 patients due to the absence of IEDs and 6 more with mesial temporal foci too deep for fNIRS, we report that significant HbR (respectively HbO) concentration changes corresponding to IEDs were observed in 62% (resp. 38%) of patients with neocortical epilepsies. This HbR/HbO response was most significant in the epileptic focus region among all the activations in 28%/21% of patients.

A real-time analysis of renewable energy sources, such as arable crops, is of great importance with regard to an optimised process management, since aspects of ecology and biodiversity are considered in crop production in order to provide a sustainable energy supply by biomass. This study was undertaken to explore the potential of spectroscopic measurement procedures for the prediction of potassium (K), chloride (Cl), and phosphate (P), of dry matter (DM) yield, metabolisable energy (ME), ash and crude fibre contents (ash, CF), crude lipid (EE), nitrate free extracts (NfE) as well as of crude protein (CP) and nitrogen (N), respectively in pretreated samples and undisturbed crops. Three experiments were conducted, one in a laboratory using near infrared reflectance spectroscopy (NIRS) and two field spectroscopic experiments. Laboratory NIRS measurements were conducted to evaluate to what extent a prediction of quality parameters is possible examining press cakes characterised by a wide heterogeneity of their parent material. 210 samples were analysed subsequent to a mechanical dehydration using a screw press. Press cakes serve as solid fuel for thermal conversion. Field spectroscopic measurements were carried out with regard to further technical development using different field grown crops. A one year lasting experiment over a binary mixture of grass and red clover examined the impact of different degrees of sky cover on prediction accuracies of distinct plant parameters. Furthermore, an artificial light source was used in order to evaluate to what extent such a light source is able to minimise cloud effects on prediction accuracies. A three years lasting experiment with maize was conducted in order to evaluate the potential of off-nadir measurements inside a canopy to predict different quality parameters in total biomass and DM yield using one sensor for a potential on-the-go application. This approach implements a measurement of the plants in 50 cm segments, since a sensor adjusted sideways is not able to record the entire plant height. Calibration results obtained by nadir top-of-canopy reflectance measurements were compared to calibration results obtained by off-nadir measurements. Results of all experiments approve the applicability of spectroscopic measurements for the prediction of distinct biophysical and biochemical parameters in the laboratory and under field conditions, respectively. The estimation of parameters could be conducted to a great extent with high accuracy. An enhanced basis of calibration for the laboratory study and the first field experiment (grass/clover-mixture) yields in improved robustness of calibration models and allows for an extended application of spectroscopic measurement techniques, even under varying conditions. Furthermore, off-nadir measurements inside a canopy yield in higher prediction accuracies, particularly for crops characterised by distinct height increment as observed for maize.

In dieser Arbeit wurden zum einen die Auswirkungen mineralischer und organischer Düngung und zum anderen der Einfluss reduzierter Bodenbearbeitung (Kreiselegge) im Vergleich zum Einsatz des Wendepfluges auf die Nährstoffspeicherung der mikrobiellen Biomasse untersucht. Dabei wurde deutlich, dass sowohl organische Düngung als auch die reduzierte Bodenbearbeitung zu einer Erhöhung des organischen Kohlenstoffs (Corg), des Stickstoff-Gesamtgehalts (N) und des mikrobiellen Biomasse Kohlenstoffs (Cmik) und Stickstoffs (Nmik) führten. Der Einsatz organischer Dünger wies darüber hinaus höhere Gehalte an Schwefel (S) auf, während der Einsatz der Kreiselegge auch den mikrobiellen Biomasse Phosphor (Pmik) gegenüber der Pflugvariante erhöhte. Des Weiteren zeigten die Untersuchungen des pilzlichen Biomarkers Ergosterol einen signifikant höheren Gehalt mit mineralischer Düngung plus Strohrückführung und beim Einsatz des Wendepfluges im Vergleich zu organischer Düngung und reduzierter Bodenbearbeitung. Dies führte sowohl bei mineralischer Düngung als auch bei dem Einsatz des Wendepfluges zu einem signifikant höheren Ergosterol/Cmik-Quotienten, der bei hohen Werten auf einen höheren Anteil saprotropher Pilze an der gesamten mikrobiellen Gemeinschaft schließen lässt. Im Zusammenhang mit der Möglichkeit saprotropher Pilze, ihre Schwefelinkorporation um 130% zu steigern, zeigten sich höhere Gehalte des mikrobiellen Schwefels unter mineralischer Düngung und des Einsatzes des Wendepflugs, bei denen der Anteil saprotropher Pilze erhöht war. Dies führte zu einem geringeren C/Smik-Quotienten, der somit als Maß für eine Veränderung der mikrobiellen Gemeinschaft herangezogen werden kann. Des Weiteren ließ sich feststellen, dass trotz eines erhöhten Pilzanteils an der mikrobiellen Gemeinschaft unter Pflugbearbeitung und mineralischer Düngung die Substratnutzungseffizienz, die durch Pilze gewöhnlich erhöht wird, hier nicht gesteigert wurde. Somit lässt sich festhalten, dass sowohl organische Düngung als auch der Einsatz der Kreiselegge mit reduzierter Bodenbearbeitung zu einer Erhöhung Bodenfruchtbarkeit erhaltender Messgrößen, wie Corg, N, Cmik und Nmik führten. Das Bodenmaterial beider Langzeitversuche wurde nach den Laboruntersuchungen mit flüssigem Stickstoff schockgefroren und anschließend gefriergetrocknet, um mittels Nahinfrarot Spektroskopie (NIRS) und unterschiedlichen Berechnungsverfahren die Bodeneigenschaften vorherzusagen. Die Messgrößen des pH-Werts, Corg, N, P, S, K, Mg, Mn, Fe, Al wurden mit NIRS in einer exzellenten Vorhersagegüte bestimmt, während die Vorhersage von Cmik, Nmik und dem Ergosterol/Cmik-Quotienten nur zu einer annähernd quantitativen Bestimmung führte. Die unterschiedlichen Berechnungen mittels Kreuzvalidierung und separater Kalibrierung/Validierung zeigten zwar, dass die Kreuzvalidierung zu einer höheren Anzahl erfolgreich vorhergesagter Parameter führte, diese jedoch nicht zur Erstellung einer Kalibrationsdatenbank herangezogen werden kann, da hier nur eine geschlossene Population betrachtet wird. Die unterschiedlichen Selektionsverfahren zeigten nur einen geringen Einfluss auf die Vorhersagegenauigkeit. Während die Aufteilung des Datensatzes in einen kleineren, homogeneren Datensatz der einzelnen Standorte zu keiner besseren Vorhersagegüte im Vergleich zum Gesamtdatensatz führte. Daher bleibt festzuhalten, dass NIRS vor allem für die Vorhersage der Grundmessgrößen wie Corg, den Gesamtgehalten und Cmik und Nmik Einsatz finden kann, während für die erfolgreiche Vorhersage für weitere mikrobielle Biomasse-Parameter noch Forschungsbedarf besteht, wie z.B. zur Probenvorbereitung.

Samples of whole crop wheat (WCW, n = 134) and whole crop barley (WCB, n = 16) were collected from commercial farms in the UK over a 2-year period (2003/2004 and 2004/2005). Near infrared reflectance spectroscopy (NIRS) was compared with laboratory and in vitro digestibility measures to predict digestible organic matter in the dry matter (DOMD) and metabolisable energy (ME) contents measured in vivo using sheep. Spectral models using the mean spectra of two scans were compared with those using individual spectra (duplicate spectra). Overall NIRS accurately predicted the concentration of chemical components in whole crop cereals apart from crude protein. ammonia-nitrogen, water-soluble carbohydrates, fermentation acids and solubility values. In addition. the spectral models had higher prediction power for in vivo DOMD and ME than chemical components or in vitro digestion methods. Overall there Was a benefit from the use of duplicate spectra rather than mean spectra and this was especially so for predicting in vivo DOMD and ME where the sample population size was smaller. The spectral models derived deal equally well with WCW and WCB and Would he of considerable practical value allowing rapid determination of nutritive value of these forages before their use in diets of productive animals. Bacterial resistance to antibiotics poses a serious health threat. Since research into new antibiotics is not progressing at the same rate as the development of bacterial resistance, widespread calls for alternatives to antibiotics have been made. Phage therapy is an ideal alternative candidate to be investigated. However the success of phage therapy may be hampered by a lack of investment support from large pharmaceutical companies, due to their narrow spectrum of activity in antibiotics, very large costs associated with clinical trials of the variety of phages needed, and regulatory requirements remaining unclear. Intellectual property is difficult to secure for therapeutic phage products for a variety of reasons, and patenting procedures vary widely between the US and the EU. Consequently, companies are more likely to invest in phage products for decontamination or veterinary use, rather than clinical use in humans. Some still raise questions as to the safety of phage therapy overall, suggesting the possibility of cytotoxicity and immunogenicity, depending on the phage preparation and route. On the other hand, with patients dying because of infections untreatable with conventional antibiotics, the question arises as to whether it is ethical not to pursue phage therapy more diligently. A paradigm shift about how phage therapy is perceived is required, as well as more rigorous proof of efficacy in the form of clinical trials of existing medicinal phage products. Phage therapy potential may be fulfilled in the meantime by allowing individual preparations to be used on a named-patient basis, with extensive monitoring and multidisciplinary team input. The National Health Service and academia have a role in carrying out clinical phage research, which would be beneficial to public health, but not necessarily financially rewarding.

Este trabalho foi desenvolvido com o intuito de avaliar nutricionalmente variedades agrícolas regionais madeirenses e açorianas de Ipomoea batatas L. (batata-doce). No total 18 amostras foram analisadas, entre as quais 11 são variedades provindas da Madeira e outras 7 provindas dos Açores. O maior objectivo deste estudo foi comparar a composição nutricional e mineral de um conjunto de variedades de batata-doce, valorizando assim os recursos agrícolas da Madeira e Açores. A caracterização nutricional e mineral dos rizomas, produzidos nestas regiões, proporcionou informação relevante para a valorização destes recursos. Sendo um rizoma tropical consumido habitualmente pela população e disponível todo o ano, torna essencial a sua avaliação. A análise laboratorial dos rizomas permitiu determinar um conjunto de características nutricionais. De um modo geral, os valores nutricionais médios detectados em rizomas de batata-doce destas duas regiões são de 93,64 g/100g de MS para o resíduo seco, 3,01 g/100g de MS para cinzas, 3,19 g/100g de MS para fibra (ADF), 1,21 g/100g de MS para gordura bruta, 3,20 g/100g de MS para proteína, 14,00 g/100g de MS para açúcares solúveis e 59,72 g/100g de MS para o amido. A análise mineral incluiu a determinação de fósforo, potássio, cálcio, magnésio, ferro, cobre, zinco, manganês e boro. Na variedade “De cenoura” (ISOP 1028), foi quantificado o nível de pro-vitamina A pela técnica de HPLC, que foi verificado ser 12,48 mg/100g em MS de β-caroteno. Os dados obtidos na análise nutricional foram usados para construir uma base de dados no NIRS (Near Infra-Red Spectroscopy) de modo a construir um modelo de previsão que possa predizer o valor nutritivo de um grande número de amostras em tempo reduzido, com um mínimo de impacto ambiental. Posteriormente, os dados obtidos através das análises nutricionais e minerais foram introduzidos no software estatístico SPSS 19.0 para determinar correlações e similaridades.

O presente trabalho teve como objectivo proceder à avaliação da qualidade nutricional de 20 variedades regionais de Phaseolus vulgaris L. e à análise comparativa dos parâmetros bioquímicos (nutricionais e anti-nutricionais) obtidos recorrendo às técnicas analíticas convencionais por química molhada e de NIRS (Near Infra-Red Spectroscopy). Uma tipificação das variedades regionais de feijão foi realizada recorrendo a sete parâmetros ou caracteres (traits) nutricionais compreendidos em humidade, proteína bruta, lípidos totais, açúcares solúveis, amido, cinzas e minerais. A faseolamina foi incluída na tipificação do feijão como parâmetro anti-nutricional enquanto inibidor de enzimas digestivas. A variedade que apresentou uma melhor qualidade nutricional foi o feijão vermelho (ISOP 00724), enquanto que o feijão Filipe (ISOP 00478) apresentou uma maior actividade inibitória da PPA (amilase do pâncreas suíno), contribuindo de igual forma como uma característica de qualidade deste feijão. A aplicação de técnicas de quimiometria na quantificação dos vários parâmetros de qualidade nutricional, através da técnica de NIRS, permitiu o desenvolvimento dos modelos PLS globais após a colecção dos valores de referência e obtenção dos respectivos espectros de cada ISOP em análise. A análise comparativa dos parâmetros nutricionais, recorrendo às técnicas analíticas convencionais por química molhada e de NIRS, permitiu relacionar os parâmetros cinzas e proteína bruta como os principais critérios nutricionais para distinção das variedades regionais quanto à sua qualidade, ao diferirem significativamente relativamente aos parâmetros restantes. A partir destas duas técnicas, conclui-se que a espectroscopia NIR associada a técnicas de análise multivariada consegue quantificar os parâmetros em estudo, permitindo distinguir amostras das variedades regionais, quanto às suas características nutricionais, exigindo uma preparação reduzida da amostra, com consequente custo de análise muito reduzido. Este trabalho representou o início de uma base de fenotipagem a partir de caracteres nutricionais, estabelecendo-se um perfil das variedades regionais de feijão e avaliação da importância dos caracteres na sua distinção e tipagem.

Near-infrared Raman spectroscopy (NIRS) is a particularly promising technique that is being used in recent years for many biomedical applications. Optical spectroscopy has gained increasing prominence as a tool for quantitative analysis of biological samples, clinical diagnostic, concentration measurements of blood metabolites and therapeutic drugs, and analysis of the chemical composition of human tissues. Toxoplasmosis is an important zoonosis in public health, and domestic cats are the most important transmitters of the disease. This disease can be detected by several serological tests, which usually have a high cost and require a long time. The goal of this work was to investigate a new method to diagnosis Toxoplasma gondii infections using NIRS. In order to confirm antibody detection, 24 cat blood scrum samples were analyzed by the Raman spectra, from which 23 presented positive serology to toxoplasmosis and one was a reference negative serum. Characteristic Raman peaks allowed differentiation between negative and positive sera, confirming the possibility of antibody detection by Raman spectroscopy. These results give the first evidence that this technique can be useful to quantify antibodies in cat sera.

The aim of this study was to evaluate the potential of near-infrared reflectance spectroscopy (NIRS) as a rapid and non-destructive method to determine the soluble solid content (SSC), pH and titratable acidity of intact plums. Samples of plum with a total solids content ranging from 5.7 to 15%, pH from 2.72 to 3.84 and titratable acidity from 0.88 a 3.6% were collected from supermarkets in Natal-Brazil, and NIR spectra were acquired in the 714 2500 nm range. A comparison of several multivariate calibration techniques with respect to several pre-processing data and variable selection algorithms, such as interval Partial Least Squares (iPLS), genetic algorithm (GA), successive projections algorithm (SPA) and ordered predictors selection (OPS), was performed. Validation models for SSC, pH and titratable acidity had a coefficient of correlation (R) of 0.95 0.90 and 0.80, as well as a root mean square error of prediction (RMSEP) of 0.45ºBrix, 0.07 and 0.40%, respectively. From these results, it can be concluded that NIR spectroscopy can be used as a non-destructive alternative for measuring the SSC, pH and titratable acidity in plums

Aiming to consumer s safety the presence of pathogenic contaminants in foods must be monitored because they are responsible for foodborne outbreaks that depending on the level of contamination can ultimately cause the death of those who consume them. In industry is necessary that this identification be fast and profitable. This study shows the utility and application of near-infrared (NIR) transflectance spectroscopy as an alternative method for the identification and classification of Escherichia coli and Salmonella Enteritidis in commercial fruit pulp (pineapple). Principal Component Analysis (PCA), Independent Modeling of Class Analogy (SIMCA) and Discriminant Analysis Partial Least Squares (PLS-DA) were used in the analysis. It was not possible to obtain total separation between samples using PCA and SIMCA. The PLS-DA showed good performance in prediction capacity reaching 87.5% for E. coli and 88.3% for S. Enteritides, respectively. The best models were obtained for the PLS-DA with second derivative spectra treated with a sensitivity and specificity of 0.87 and 0.83, respectively. These results suggest that the NIR spectroscopy and PLS-DA can be used to discriminate and detect bacteria in the fruit pulp

Objetivou-se com o estudo determinar o valor nutritivo da dieta selecionada por vacas de cria, numa área característica da sub-região da Nhecolândia, Pantanal, durante dois anos hidrológicos (outubro de 1997 a setembro de 1999). Mensalmente foram colhidas manualmente amostras das partes das principais forrageiras selecionadas por bovinos, identificadas por meio de observação direta, para análise de proteína bruta (PB), fibra em detergente neutro (FDN), fibra em detergente ácido (FDA) e lignina (LIG). A digestibilidade da matéria orgânica (DMO) da dieta foi avaliada em amostras fecais com o uso da espectrofotometria de reflectância no infravermelho proximal (NIRS). Houve efeito interativo de ano e época para todos os constituintes químicos analisados na dieta. O valor nutritivo da dieta selecionada por bovinos diminuiu no período de abril a junho, em ambos os anos estudados. As vacas apresentaram uma melhor condição corporal de setembro a março do ano hidrológico 1998/99, no qual a dieta apresentou valores médios de 60,4% de digestibilidade, 10% de PB; 40,20% de FDA; 71% de FDN e 6,0% de lignina, considerada adequada para obtenção de um desempenho reprodutivo satisfatório. Portanto, o valor nutritivo da dieta selecionada por bovinos na sub-região da Nhecolândia depende especialmente da intensidade e distribuição das chuvas, responsáveis pela maior ou menor presença de água nas unidades mais baixas, que possuem forrageiras de melhor qualidade.

The objective of this study was to evaluate the potential of near infrared spectroscopy (NIRS) associated with multivariate statistics to distinguish coal produced from wood of planted and native forests. Timber forest species from the C errado (Cedrela sp., Aspidosperma sp., Jacaranda sp. and unknown species) and Eucalyptus clones from forestry companies (Vallourec and Cenibra) were carbonized in the final temperatures of 300, 500 and 700°C. In each heat treatment were carbonized 15 specimens of each vegetal material totaling 270 samples (3 treatments x 15 reps x 6 materials) produced in 18 carbonization (3 treatments x 6 materials). The acquisition of the spectra of coals in the near infrared using a spectrometer was performed. Principal Component Analysis (PCA) and Partial Least Squares Regression (PLS-R) were carried out in the spectra. NIR Spectroscopy associated with PCA was not able to differentiate charcoals produced from native and planted woods when utilizing all carbonized samples at different temperatures in the same analysis; The PCA of all charcoals was able to distinguish the samples depending on temperature in which they were carbonized. However, the separation of native and planted charcoal was possible when the samples were analyzed separately by final temperature. The prediction of native or planted classes by PLS-R presented better performance for samples carbonized at 300°C followed by those at 500°C, 700°C and for all together.
