Lactate dehydrogenase (LDH) of the malaria parasite, Plasmodium vivax (Pv), serves as a drug target and immunodiagnostic marker. The LDH cDNA generated from total RNA of a clinical isolate of the parasite was cloned into pRSETA plasmid. Recombinant his-tagged PvLDH was over-expressed in E. coli Rosetta2DE3pLysS and purified using Ni2+-NTA resin giving a yield of 25-30 mg/litre bacterial culture. The recombinant protein was enzymatically active and its catalytic efficiency for pyruvate was 5.4 x 10(8) min(-1) M-1, 14.5 fold higher than a low yield preparation reported earlier to obtain PvLDH crystal structure. The enzyme activity was inhibited by gossypol and sodium oxamate. The recombinant PvLDH was reactive in lateral flow immunochromatographic assays detecting pan- and vivax-specific LDH. The soluble recombinant PvLDH purified using heterologous expression system can facilitate the generation of vivax LDH-specific monoclonals and the screening of chemical compound libraries for PvLDH inhibitors.

Glyoxalase I which is synonymously known as lactoylglutathione lyase is a critical enzyme in methylglyoxal (MG) detoxification. We assessed the STM3117 encoded lactoylglutathione lyase (Lgl) of Salmonella Typhimurium, which is known to function as a virulence factor, due in part to its ability to detoxify methylglyoxal. We found that STM3117 encoded Lgl isomerises the hemithioacetal adduct of MG and glutathione (GSH) into S-lactoylglutathione. Lgl was observed to be an outer membrane bound protein with maximum expression at the exponential growth phase. The deletion mutant of S. Typhimurium (lgl) exhibited a notable growth inhibition coupled with oxidative DNA damage and membrane disruptions, in accordance with the growth arrest phenomenon associated with typical glyoxalase I deletion. However, growth in glucose minimal medium did not result in any inhibition. Endogenous expression of recombinant Lgl in serovar Typhi led to an increased resistance and growth in presence of external MG. Being a metalloprotein, Lgl was found to get activated maximally by Co2+ ion followed by Ni2+, while Zn2+ did not activate the enzyme and this could be attributed to the geometry of the particular protein-metal complex attained in the catalytically active state. Our results offer an insight on the pivotal role of the virulence associated and horizontally acquired STM3117 gene in non-typhoidal serovars with direct correlation of its activity in lending survival advantage to Salmonella spp.

Nanocrystalline strontium hexaferrites SrFe12-2x (Ni2+-Zr4+)(x)O-19] nanoparticles were successfully synthesized by sal gel process. For densification the powders were sintered at 950 degrees C/4 h. The sintered samples were characterized by X-ray diffraction (XRD), surface area measurement, and field emission scanning electron microscope (FESEM). The lattice parameter a is almost constant but c increased with x upto 0.8 and then decreased. The frequency dependent complex permittivity (epsilon and epsilon `' and permeability (mu' and mu `') and magnetic properties such as saturation magnetization (M-s), coercive field (H-c) were studied. If is observed that saturation magnetization increased gradually from 57.82 emuig to 67.2 emufg as x increased from 0.2 to 0.4 and then decreased from 672 emufg to 31.63 ernufg for x=1.0. In present study, x=0.4 shows high value of M-s 67.2 emu/g. The real part of permittivity (epsilon') remains constant upto a frequency 1 GHz and increases further with an increase of frequency, a resonance and anti resonance peak was observed above 1 GHz for all the samples. In real part of permeability (mu') the relaxation frequency is observed above 1 GHz for all the samples and it is attributed to the domain wall motion. It is well known that the permeability for polycrystalline ferrites can be described as the superposition of two different magnetizing mechanisms: spin rotation and domain wall motion. These low coercive strontium hexaferrites are suitable for magnetic recording applications in hard disks, floppy disks, video tapes, etc. (C) 2015 Elsevier B.V. All rights reserved.

Ni2+ ion induced unusual conductivity reversal and an enhancement in the gas sensing properties of ferrites based gas sensors, is reported. The Co1-xNixFe2O4 (for x = 0, 0.5 and 1) nanoparticles were synthesized by wet chemical co-precipitation method and gas sensing properties were studied as a function of composition and temperature. The structural, morphological and microstructural characterization revealed crystallite size of in the range 10-20 nm with porous morphology consisting of nano-sized grains. The Energy Dispersive X-ray (EDX) mapping confirms homogeneous distribution of Co, Ni, Fe and O elements in the ferrites. The non-stoichiometry of the inverse spinel type ferrites and the relative concentration of Ni3+/Co3+ defects were studied using X-ray photoelectron spectroscopy. It is found that the addition of Ni2+ ions into cobalt ferrite shows preferred selectivity towards CO gas at high temperature (325 degrees C) and ethanol gas at low temperature (250 degrees C), unlike undoped cobalt ferrite or undoped nickel ferrite, which show similar response for both these gases. Moreover, an unusual conductivity reversal is observed, except cobalt ferrite due to the difference in reactivity of the gases as well as characteristic non-stoichiometry of ferrites. This behavior is highly gas ambient dependent and hence can be well-exploited for selective detection of gases. (C) 2015 Elsevier B.V. All rights reserved.

Introduction Nucleoside diphosphate kinase (NDK), conserved across bacteria to humans, synthesises NTP from NDP and ATP. The eukaryotic homologue, the NDPK, uses ATP to phosphorylate the tubulin-bound GDP to GTP for tubulin polymerisation. The bacterial cytokinetic protein FtsZ, which is the tubulin homologue, also uses GTP for polymerisation. Therefore, we examined whether NDK can interact with FtsZ to convert FtsZ-bound GDP and/or free GDP to GTP to trigger FtsZ polymerisation. Methods Recombinant and native NDK and FtsZ proteins of Mycobacterium smegmatis and Mycobacterium tuberculosis were used as the experimental samples. FtsZ polymersation was monitored using 90 degrees light scattering and FtsZ polymer pelleting assays. The gamma 32P-GTP synthesised by NDK from GDP and gamma 32P-ATP was detected using thin layer chromatography and quantitated using phosphorimager. The FtsZ bound P-32-GTP was quantitated using phosphorimager, after UV-crosslinking, followed by SDS-PAGE. The NDK-FtsZ interaction was determined using Ni2+-NTA-pulldown assay and co-immunoprecipitation of the recombinant and native proteins in vitro and ex vivo, respectively. Results NDK triggered instantaneous polymerisation of GDP-precharged recombinant FtsZ in the presence of ATP, similar to the polymerisation of recombinant FtsZ (not GDP-precharged) upon the direct addition of GTP. Similarly, NDK triggered polymerisation of recombinant FtsZ (not GDP-precharged) in the presence of free GDP and ATP as well. Mutant NDK, partially deficient in GTP synthesis from ATP and GDP, triggered low level of polymerisation of MsFtsZ, but not of MtFtsZ. As characteristic of NDK's NTP substrate non-specificity, it used CTP, TTP, and UTP also to convert GDP to GTP, to trigger FtsZ polymerisation. The NDK of one mycobacterial species could trigger the polymerisation of the FtsZ of another mycobacterial species. Both the recombinant and the native NDK and FtsZ showed interaction with each other in vitro and ex vivo, alluding to the possibility of direct phosphorylation of FtsZ-bound GDP by NDK. Conclusion Irrespective of the bacterial species, NDK interacts with FtsZ in vitro and ex vivo and, through the synthesis of GTP from FtsZ-bound GDP and/or free GDP, and ATP (CTP/TTP/UTP), triggers FtsZ polymerisation. The possible biological context of this novel activity of NDK is presented.

The double perovskite Y2NiMnO6 displays ferromagnetic transition at T-c approximate to 81 K. The ferromagnetic order at low temperature is confirmed by the saturation value of magnetization (Ms) and also validated by the refined ordered magnetic moment values extracted from neutron powder diffraction data at 10 K. This way, the dominant Mn4+ and Ni2+ cationic ordering is confirmed. The cation-ordered P2(1)/n nuclear structure is revealed by neutron powder diffraction studies at 300 and 10 K. Analysis of the frequency-dependent dielectric constant and equivalent circuit analysis of impedance data take into account the bulk contribution to the total dielectric constant. This reveals an anomaly which coincides with the ferromagnetic transition temperature (T-c). Pyrocurrent measurements register a current flow with onset near T-c and a peak at 57 K that shifts with temperature ramp rate. The extrinsic nature of the observed pyrocurrent is established by employing a special protocol measurement. It is realized that the origin is due to reorientation of electric dipoles created by the free charge carriers and not by spontaneous electric polarization at variance with recently reported magnetism-driven ferroelectricity in this material.

Here, we demonstrate a green and environment-friendly pyrolysis route for the synthesis of metal-rich sulphide embedded in an N-doped carbon (NC) framework in the absence of sulphide ions (S2-). The metal-chelate complex (tris(ethylenediamine) metal(II) sulfate) serves as a new and single source precursor for the synthesis of earth abundant and non-precious hybrid structures such as metal-rich sulphides Co9S8@NC and Ni3S2@ NC when M-II = Co2+ and Ni2+ and counter sulphate (SO42-) ions are the source of S. Both the hybrids show superior OER activity as compared to commercial RuO2.

This letter demonstrates an alternative method to form gallium silicate glass ceramics using high-energy electron irradiation. Compared with glass ceramics obtained from the conventional thermal treatment method, the distribution and crystal sizes of the precipitated Ga2O3 nanoparticles are the same. An advantage of this method is that the spatial distribution of the precipitated nanoparticles can be easily controlled. However, optically active dopants Ni2+ ions do not participate in the precipitation during electron irradiation. (c) 2007 American Institute of Physics.

Quando se trata de efluentes muito complexos e de amostras de água ou de sedimento de locais poluÃdos, é inviável ou até mesmo impossÃvel detectar a presença de todas as substâncias presentes. Os principais contaminantes associados à poluição das águas naturais, tem-se os metais pesados, uma classe de compostos de toxicidade elevada e que são bioacumulados nos seres vivos. Desses metais, o cobre e o nÃquel se destacam, tanto por seu amplo uso em processos industriais, o que acarreta sua presença em diversos tipos de despejos, quanto por sua toxicidade elevada. O objetivo deste trabalho foi determinar o potencial tóxico do sulfato de cobre e sulfato de nÃquel, de sua mistura e seus complexos com EDTA frente a Daphnia similis para utilização na avaliação do desempenho do processo de separação com membranas no tratamento de efluentes. Em um estudo de toxicidade, as interações do metal com o organismo-teste são influenciadas pela espécie testada, pela combinação dos metais ou pela composição do meio aquoso. O Ãon Cu2+ apresentou toxicidade superior ao Ni2+, sendo que na mistura desses dois Ãons, prevaleceu o resultado obtido para o Cu2+, decorrente de sua maior toxicidade. A complexação dos metais reduz significativamente o potencial tóxico dos metais

O objetivo deste trabalho foi investigar o desempenho de uma membrana comercial na remoção de um metal pesado (nÃquel) de efluentes sintéticos, por osmose inversa. Na primeira etapa foi realizada uma comparação com os resultados obtidos com soluções de alimentação contendo sais, como NaCl, NaNO3 e Ni(NO3)2.6 H2O, nas concentrações de 50, 100 e 200 ppm, e nas pressões de 10, 20 e 26 bar. Os resultados mostraram que a influência da concentração e da pressão aplicada ao sistema não afetaram as rejeições de forma significativa. Na segunda etapa, como os parâmetros não influÃram significativamente na rejeição dos sais, optou-se, pela aplicação de uma pressão de operação de 10 bar, para avaliar a eficiência de remoção de nÃquel. A membrana utilizada, constituÃda de poliamida, modelo HR98PP e fornecida pela DOW/Filmtec, apresentou uma boa permeabilidade hidráulica. Os resultados mostraram que para todas as concentrações testadas, as rejeições de nÃquel ultrapassaram 96%, comprovando a boa seletividade deste tipo de membrana na rejeição do referido metal, com fluxos de permeado variando entre 4,78 e 5,55 L/h.m2 , sob pressão de operação de 10 bar. Para estudar o efeito do tamanho iônico na rejeição da membrana, o nÃquel foi complexado pela adição de um agente quelante na solução de alimentação. O agente escolhido foi o Na2EDTA, devido à formação de um complexo estável com o nÃquel e por ser um agente não prejudicial à saúde humana. Os resultados com adição de EDTA indicaram um aumento na rejeição de nÃquel, atingindo o Ãndice máximo de 98,22 %, partindo-se de uma solução com 40,39 ppm de Ni2+, e confirmam que o processo de osmose inversa com a membrana HR98PP é altamente adequado para o tratamento de efluentes contendo nÃquel

Neste estudo, a sorção e recuperação de Ãons metálicos de resÃduos sólidos industriais provenientes de uma indústria de galvanoplastia situada no Rio de Janeiro (Brasil) foram investigadas através da utilização de duas resinas comerciais de troca iônica: Lewatit VPOC 1800 (fortemente ácida, tipo gel) e Lewatit VPOC 1960 (fortemente básica, tipo gel), produzidas pela Lanxess-Bayer Chemicals. As caracterÃsticas fÃsico-quimicas das resinas e do lodo galvânico foram determinadas. Os estudos de sorção das resinas foram conduzidos em batelada e em coluna. Baseado nesses estudos, os parâmetros de sorção e das curvas de ruptura foram determinados. Os estudos de equilÃbrio e cinética de sorção também foram realizados. O resÃduo de galvanoplastia era composto pelos metais: Cu2+, Fe3+, Al3+, Ni2+ e Cr3+. A capacidade de sorção qe das resinas Lewatit VPOC 1800 variou entre 0,1-1,9 mg g-1 para Cu2+, 0,01-0,6 mg g-1 para Fe3+ e 0,2-0,4 mg g-1 para Al3+. Enquanto que para a resina Lewatit VPOC 1960, os valores de qe variou entre 0,01-0,4 mg g-1 para Cu2+ e 0,01 0,2 mg g-1 para Fe3+ dependendo da concentração do metal e do tempo de contato. A capacidade de sorção para a resina Lewatit VPOC 1960 foi restrita para Ãons Cu2+ e Fe3+ os quais formam complexos aniônicos com Ãons Cl-. O modelo de Freundlich foi o mais adequado para descrever o equilÃbrio de troca iônica de ambas as resinas. Já em relação ao mecanismo de sorção, o modelo pseudo-segunda ordem tipo 1 foi o mais aplicável. O ponto de ruptura das resinas Lewatit VPOC 1800 e Lewatit VPOC 1960 em relação aos Ãons Cu2+ocorreu quando passou através da coluna, 1860 cm3 e 2220 cm3 de solução de resÃduo sólido respectivamente (20 g de resina, 100 mg L-1 de Ãons Cu2+, vazão de 60 cm3 min-1). Os Ãons metálicos Cu2+, Fe3+, Al3+, foram dessorvidos em alta proporção da resina Lewatit VPOC 1800 passando pela coluna solução aquosa de H2SO4 2,4 mol L-1. Já os metais Cu2+ e Fe3+ foram eluÃdos da resina Lewatit VPOC 1960 com solução aquosa de HCl 2,0 mol L-1. A recuperação seletiva de Cu2+ não foi alcançada porque Cu2+ e Fe3+ precipitam na mesma faixa de pH

Two new azo dyes of alpha-isoxazolylazo-beta-dilcetones and their Ni(II) and Cu(II) complexes with blue-violet light wavelength were synthesized using a coupling component, different diazo components and metal (II) ions (Ni2+ and Cu2+). Based on the elemental analysis, MS spectra and FT-IR spectral analyses, azo dyes were unequivocally shown to exist as hydrazoketo and azoenol forms which were respectively obtained from the solution forms and from the solid forms. The action of sodium methoxide (NaOMe) on azo dyes in solutions converts hydrazoketo form into azoenol form, so azo dyes are coordinated with metal (II) ions as co-ligands in the azoenol forms. The solubility of all the compounds in common organic solvents such as 2,2,3,3-tetrafluoro-1-propanol (TFP) or chloroform (CHCl3) and absorption properties of spin-coating thin films were measured. The difference of absorption maxima from the complexes to their ligands was discussed. In addition, the TG analysis of the complexes was also determined, and their thermal stability was evaluated. It is found that these new metal (II) complexes had potential application for high-density digital versatile disc-recordable (HD-DVD-R) system due to their good solubility in organic solvents, reasonable and controllable absorption spectra in blue-violet light region and high thermal stability. Thiazolyl heterocyclic azo dye and its metal (Ni2+, Co2+)-azo complexes were synthesized. Their structures were confirmed by elemental analysis, UV-VIS absorption spectra, FT-IR, H-1 NMR and MALDI-MS. The thermal properties of metal complexes were studied by DSC-TGA. The optical constants (complex refractive index N=n + ik) and thickness of the complex thin films on polished single-crystal silicon substrates were investigated on a scanning ellipsometer. Results indicate that thiazolyl metal-azo complexes possess good optical and thermal properties. They would be a promising recording medium candidate for NVD with the Super-resolution near field structure (Super-RENS) technology. (c) 2007 Elsevier B.V. All rights reserved.

The emission intensity of Ni2+ at 1200 nm in transparent ZnO-Al2O3-SiO2 glass ceramics containing ZnAl2O4 nanocrystals is improved approximately 8 times by Cr3+ codoping with 532 nm excitation. This enhanced emission could be attributed to an efficient energy transfer from Cr3+ to Ni2+, which is confirmed by time-resolved emission spectra. The energy transfer efficiency is estimated to be 57% and the energy transfer mechanism is also discussed. (C) 2008 Optical Society of America.

The near-infrared emission intensity of Ni2+ in Yb3+/Ni2+ codoped transparent MgO-Al2O3-Ga2O3-SiO2-TiO2 glass ceramics could be enhanced up to 4.4 times via energy transfer from Yb3+ to Ni2+ in nanocrystals. The best Yb2O3 concentration was about 1.00 mol%. For the Yb3+/Ni2+ codoped glass ceramic with 1.00 mol% Yb2O3, a broadband near-infrared emission centered at 1265 nm with full width at half maximum of about 300 nm and lifetime of about 220 mu s was observed.
