Several tissue types of Lupinus albus L. were investigated as sources for the isolation of protoplasts. Cotyledons from in vitro seedlings were found to yield the highest number of protoplasts compared with leaves, hypocotyls and roots. A combination of the protoplast isolation enzymes, cellulase and Pectolyase Y23, was capable of releasing the highest number of protoplasts compared with a combination of cellulase and Macerase. Protoplast yield increased with increasing cotyledon age but was accompanied by a progressive decline in protoplast viability. The optimal combination of protoplast yield and viability occurred when the protoplasts were isolated from 14- to 18-day-old cotyledons. The ratio between the volume of enzyme solution and the tissue biomass did not affect the protoplast production significantly. This is the first report of the isolation of protoplasts from a lupin cotyledon and, following the procedure described in this paper, an average yield of 1.2 x 10(6) protoplasts per gram of fresh tissue was obtainable.

Six nutrient formulations were studied for their efficacy in inducing mitosis in white lupin seedling cotyledon protoplasts of which the formulations of Schafer-Menuhr & Sturmer (AS) and Kao (K8p) were found to be superior over the other four when supplemented with 6-benzylaminopurine and alpha-naphthaleneacetic acid (alpha-NAA). An unltrafiltration treatment of K8p increased mitotic frequency by 130% when compared with the untreated control. Medium enrichment with 0.2% bovine serum albumin (BSA) brought about a dramatic 1341% rise in protoplast division in comparison with BSA-free medium but only when the enrichment was carried out in Kao and Michayluk (KM8p) background containing 2, 4-dichlorophenoxyacetic acid, alpha-NAA and zeatin. A higher number of protocolonies (each proliferating from single protoplast following multiple divisions) were seen in 0.4% BSA. With this breakthrough in white lupin protoplast research, it is now possible to reproducibly obtain protocolonies that was hitherto not possible.

Plantas de duas formas botânicas de Catharanthus roseus, de flores lilases e de flores brancas foram cultivadas em soluções nutritivas deficientes em N, P, K, Ca, Mg, S e B, e em solução completa, a fim de se obter o quadro sintomatológico das deficiências, assim como os níveis analíticos de nutrientes nas folhas, caules, raízes e flores. Manifestaram-se sintomas de deficiência claros para todos os nutrientes estudados. Nas plantas de flores lilases, as concentrações de nutrientes na matéria seca de folhas de plantas normais e deficientes foram, respectivamente, para cada nutriente estudado: N(%): 3,53-1,20; P(%): 0,35-0,11; K(%): 2,45-0,76; Ca(%): 1,77-0,81; Mg(%): 0,55-0,46; S(%):0,21-0,12; B(ppm): 382-37. Nas plantas de flores brancas, estas concentrações foram: N(%): 3,78-0,92; P(%): 0,38-0,09; K(%): 2,60-0,86; Ca(%): 1,37-1,15; Mg(%): 0,56-0,44; S(%): 0,10-0,07; B(ppm):372-39.

O objetivo deste estudo foi avaliar as concentrações de mercúrio total em penas de Ardea albus coletadas em uma assembléia de aves localizada nas imediações da cidade de Belém, Pará, com vistas a investigar a possibilidade do uso desta espécie nos estudos de biomonitoramento deste metal. Para determinação de mercúrio total foi utilizada a espectrofotometria de absorção atômica com amalgamação. A concentração média de mercúrio total nas penas do corpo foi 2,2 ± 1,5 µg.g-1 e nas penas das asas foi 1.3 ± 0.9 µg/g-1. Não foi observada correlação entre a concentração de mercúrio total e o comprimento das penas do corpo e da asa. Foi observado teor de mercúrio total superior a 5 µg.g-1 em apenas uma amostra de pena do corpo.

The aim of the present study was to verify, through the response surface methodology, the optimum conditions for the combined use of aqueous extract of white lupine (Lupinus albus L.) and tropical pitanga juice (Eugenia unifl ora L.) in the development of a drink. Therefore, the independent variables were represented by the volume of aqueous extract of lupine (mL) and by the volume of pitanga juice (mL). The dependent variables (responses) were obtained through sensory test of acceptance (“appearance”, “aroma”, “taste” and “overall acceptability”). The combined optimization of the variables pointed the second formulation (50mL of aqueous extract of lupine and 30mL of pitanga juice) of the experimental project as the most adequate. In conclusion it was possible to get a drink with attractive sensory characteristics using aqueous extract of white lupine and tropical pitanga juice. The usage of response surface methodology made possible the determination of regions of maximum acceptance for each attribute evaluated in the formulation of a drink with aqueous extract of white lupine and pitanga juice, with a minimum of tests.
