Com o presente trabalho pretende-se discutir a importância do DNA na resolução de casos de investigação forense. O Homem, desde os tempos mais remotos tem revelado interesse na confirmação da identidade dos seus semelhantes, pelo desenvolvimento e prática de diversas técnicas de identificação. A partir de uma revisão da literatura, fez-se o estudo dos métodos de identificação humana, sendo este mais aprofundado no que diz respeito ao DNA. Frequentemente são encontrados corpos em que a única peça disponível para o processo de identificação é o dente. As peças dentárias são as estruturas mais resistentes e estáveis do corpo humano, mantendo as suas características e propriedades por longos períodos de tempo pós-morte. A análise do DNA contribui de forma muito importante para os processos de reconhecimento humano, principalmente em casos em que outros métodos de identificação falham devido a decomposição, fragmentação, incineração ou inexistência de dados comparativos ante-morte. Para a obtenção de um perfil genético é necessário executar as seguintes etapas: extração do DNA, sua quantificação e amplificação seguida de análise de determinadas regiões do genoma. Nos dias de hoje, muitos casos de identificação necessitam de uma abordagem multidisciplinar, isto porque em algumas situações existe falta de material padrão ou, por outro lado, pode haver mais do que uma evidência a ser examinada.

A implementação de sistemas de gestão da qualidade na área da educação e formação permite reforçar e consolidar as organizações que atuam num mercado cada vez mais competitivo, permitindo-lhes satisfazer, numa base permanente e sistemática, as expetativas dos clientes através do fornecimento de produtos de formação de melhor qualidade. Neste contexto, o objetivo deste estudo é explorar a temática dos sistemas de gestão da qualidade ao nível do setor de educação. Em específico pretende-se efetuar uma revisão de literatura sobre qualidade, formação e ensino à distância;analisar normas, projetos e iniciativas em matéria de ensino à distância e implementar um Sistema de Gestão da Formação, de acordo com a NP 4512, numa unidade de e-learning. A metodologia adotada foi investigação–ação e centrou-se no levantamento bibliográfico e na aplicação dos conceitos num contexto específico de um organização de ensino. Foi escolhida a unidade de e-learning do IPP (e-IPP) como contexto do estudo por ser uma unidade de ensino superior. Os principais resultados obtidos são: (1) maior conhecimento das normas projetos e iniciativas em matéria de ensino à distância a nível nacional e europeu; (2) análise detalhada da recente norma portuguesa NP 4512; (3) elaboração da documentação associada ao Sistema de Gestão da Formação (SGF) na unidade e-IPP, em específico, identificação e monitorização dos processos, descrição dos procedimentos obrigatórios e elaboração do manual do SGF. Como principal limitação deste estudo destaca-se a implementação parcial do sistema de gestão da formação na unidade e-IPP, devido à falta de tempo e à falta de maturidade da unidade e-IPP.

Among several process variability sources, valve friction and inadequate controller tuning are supposed to be two of the most prevalent. Friction quantification methods can be applied to the development of model-based compensators or to diagnose valves that need repair, whereas accurate process models can be used in controller retuning. This paper extends existing methods that jointly estimate the friction and process parameters, so that a nonlinear structure is adopted to represent the process model. The developed estimation algorithm is tested with three different data sources: a simulated first order plus dead time process, a hybrid setup (composed of a real valve and a simulated pH neutralization process) and from three industrial datasets corresponding to real control loops. The results demonstrate that the friction is accurately quantified, as well as ""good"" process models are estimated in several situations. Furthermore, when a nonlinear process model is considered, the proposed extension presents significant advantages: (i) greater accuracy for friction quantification and (ii) reasonable estimates of the nonlinear steady-state characteristics of the process. An important consideration in the development of mathematical models for dynamic simulation, is the identification of the appropriate mathematical structure. By building models with an efficient structure which is devoid of redundancy, it is possible to create simple, accurate and functional models. This leads not only to efficient simulation, but to a deeper understanding of the important dynamic relationships within the process. In this paper, a method is proposed for systematic model development for startup and shutdown simulation which is based on the identification of the essential process structure. The key tool in this analysis is the method of nonlinear perturbations for structural identification and model reduction. Starting from a detailed mathematical process description both singular and regular structural perturbations are detected. These techniques are then used to give insight into the system structure and where appropriate to eliminate superfluous model equations or reduce them to other forms. This process retains the ability to interpret the reduced order model in terms of the physico-chemical phenomena. Using this model reduction technique it is possible to attribute observable dynamics to particular unit operations within the process. This relationship then highlights the unit operations which must be accurately modelled in order to develop a robust plant model. The technique generates detailed insight into the dynamic structure of the models providing a basis for system re-design and dynamic analysis. This study focuses on identification and exploitation processes among Finnish design entrepreneurs (i.e. selfemployed industrial designers). More specifically, this study strives to find out what design entrepreneurs do when they create new ventures, how venture ideas are identified and how entrepreneurial processes are organized to identify and exploit such venture ideas in the given industrial context. Indeed, what does educated and creative individuals do when they decide to create new ventures, where do the venture ideas originally come from, and moreover, how are venture ideas identified and developed into viable business concepts that are introduced on the markets? From an academic perspective: there is a need to increase our understanding of the interaction between the identification and exploitation of emerging ventures, in this and other empirical contexts. Rather than assuming that venture ideas are constant in time, this study examines how emerging ideas are adjusted to enable exploitation in dynamic market settings. It builds on the insights from previous entrepreneurship process research. The interpretations from the theoretical discussion build on the assumption that the subprocesses of identification and exploitation interact, and moreover, they are closely entwined with each other (e.g. McKelvie & Wiklund, 2004, Davidsson, 2005). This explanation challenges the common assumption that entrepreneurs would first identify venture ideas and then exploit them (e.g. Shane, 2003). The assumption is that exploitation influences identification, just as identification influences exploitation. Based on interviews with design entrepreneurs and external actors (e.g. potential customers, suppliers and collaborators), it appears as identification and exploitation of venture ideas are carried out in close interaction between a number of actors, rather than alone by entrepreneurs. Due to their available resources, design entrepreneurs have a desire to focus on identification related activities and to find external actors that take care of exploitation related activities. The involvement of external actors may have a direct impact on decisionmaking and various activities along the processes of identification and exploitation, which is something that previous research does not particularly emphasize. For instance, Bhave (1994) suggests both operative and strategic feedback from the market, but does not explain how external parties are actually involved in the decisionmaking, and in carrying out various activities along the entrepreneurial process.

A class identification algorithms is introduced for Gaussian process(GP)models.The fundamental approach is to propose a new kernel function which leads to a covariance matrix with low rank,a property that is consequently exploited for computational efficiency for both model parameter estimation and model predictions.The objective of either maximizing the marginal likelihood or the Kullback–Leibler (K–L) divergence between the estimated output probability density function(pdf)and the true pdf has been used as respective cost functions.For each cost function,an efficient coordinate descent algorithm is proposed to estimate the kernel parameters using a one dimensional derivative free search, and noise variance using a fast gradient descent algorithm. Numerical examples are included to demonstrate the effectiveness of the new identification approaches.

Paracoccidioides brasiliensis causes paracoccidioidomycosis (PCM), a systemic mycosis presenting clinical manifestations ranging from mild to severe forms. A P. brasiliensis cDNA expression library was produced and screened with pooled sera from PCM patients adsorbed against antigens derived from in vitro-grown P. brasiliensis yeast cells. Sequencing DNA inserts from clones reactive with PCM patients sera indicated 35 open reading frames presenting homology to genes involved in metabolic pathways, transport, among other predicted functions. The complete cDNAs encoding aromatic-L-amino-acid decarboxylase (Pbddc), lumazine synthase (Pbls) and a homologue of the high affinity copper transporter (Pbctr3) were obtained. Recombinant proteins PbDDC and PbLS were obtained; a peptide was synthesized for PbCTR3. The proteins and the synthetic peptide were recognized by sera of patients with confirmed PCM and not by sera of healthy patients. Using the in vivo-induced antigen technology (IVIAT), we identified immunogenic proteins expressed at high levels during infection. Quantitative real time RTPCR demonstrated high transcript levels of Pbddc, Pbls and Pbctr3 in yeast cells infecting macrophages. Transcripts in yeast cells derived from spleen and liver of infected mice were also measured by qRT-PCR. Our results suggest a putative role for the immunogenic proteins in the infectious process of P. brasiliensis. (C) 2009 Elsevier Masson SAS. All rights reserved.

The purpose of the first part of the research activity was to develop an aerobic cometabolic process in packed bed reactors (PBR) to treat real groundwater contaminated by trichloroethylene (TCE) and 1,1,2,2-tetrachloroethane (TeCA). In an initial screening conducted in batch bioreactors, different groundwater samples from 5 wells of the contaminated site were fed with 5 growth substrates. The work led to the selection of butane as the best growth substrate, and to the development and characterization from the site’s indigenous biomass of a suspended-cell consortium capable to degrade TCE with a 90 % mineralization of the organic chlorine. A kinetic study conducted in batch and continuous flow PBRs and led to the identification of the best carrier. A kinetic study of butane and TCE biodegradation indicated that the attached-cell consortium is characterized by a lower TCE specific degredation rates and by a lower level of mutual butane-TCE inhibition. A 31 L bioreactor was designed and set up for upscaling the experiment. The second part of the research focused on the biodegradation of 4 polymers, with and with-out chemical pre-treatments: linear low density polyethylene (LLDPE), polyethylene (PP), polystyrene (PS) and polyvinyl chloride (PVC). Initially, the 4 polymers were subjected to different chemical pre-treatments: ozonation and UV/ozonation, in gaseous and aqueous phase. It was found that, for LLDPE and PP, the coupling UV and ozone in gas phase is the most effective way to oxidize the polymers and to generate carbonyl groups on the polymer surface. In further tests, the effect of chemical pretreatment on polyner biodegrability was studied. Gas-phase ozonated and virgin polymers were incubated aerobically with: (a) a pure strain, (b) a mixed culture of bacteria; and (c) a fungal culture, together with saccharose as a co-substrate.

Questa tesi ha come oggetto di studio i problemi riscontrati nelle trascrizioni di testi orali in lingua inglese prodotte da 39 studenti della laurea magistrale in Interpretazione. Nel Capitolo 1 viene presentato l’ascolto, che viene definito prima da un punto di vista storico, poi analizzato come processo composto da quattro fasi, come sostiene Michael Rost (2011). Il capitolo si conclude con l’ascolto nell’ambito dell’interpretazione. Nel Capitolo 2 viene analizzato in maniera contrastiva l’apprendimento dell’ascolto nella prima e nella seconda lingua. I primi due capitoli forniscono le basi per comprendere il caso di studio. Il Capitolo 3 concerne la metodologia dello studio. Vengono presentati il metodo di analisi delle trascrizioni, la categorizzazione dei problemi riscontrati e il processo di creazione del sistema di analisi utilizzato. Nel Capitolo 4 vengono presentati i dati ottenuti seguendo il metodo esposto nel Capitolo 3. Si presentano i problemi riscontrati, che vengono suddivisi in categorie in base a ciò che può averli causati. Il Capitolo 5 è dedicato alle conclusioni. Qui vengono suggerite possibili strategie mirate ad aiutare gli studenti di Interpretazione a migliorare le proprie capacità di ascolto in lingua inglese. Esta tesis quiere analizar los problemas encontrados en transcripciones de textos orales en inglés hechas por 39 estudiantes del máster en Interpretación. En el Capítulo 1 se presenta la escucha, que se define primero desde una perspectiva histórica, y luego como un proceso formado por cuatro fases, como argumenta Michael Rost (2011). El capítulo se cierra con la escucha en el ámbito de la interpretación. En el Capítulo 2 se analizan de forma contrastiva el aprendizaje de la escucha en la primera y segunda lengua. Los primeros dos capítulos constituyen la base para comprender el caso de estudio. El Capítulo 3 atañe a la metodología del estudio. Se presentan el método de análisis de las transcripciones, la categorización de los problemas encontrados y el proceso de creación del sistema de análisis que se ha empleado. En el Capítulo 4 se proporcionan los datos obtenidos gracias al método presentado en el Capítulo 3. Se presentan los problemas encontrados, que han sido divididos en categorías según qué puede haberlos ocasionado. El Capítulo 5 está dedicado a las conclusiones. Aquí se sugieren posibles estrategias cuyo objetivo es ayudar a los estudiantes de Interpretación a mejorar sus capacidades de escucha en inglés.

Initially this thesis examines the various mechanisms by which technology is acquired within anodizing plants. In so doing the history of the evolution of anodizing technology is recorded, with particular reference to the growth of major markets and to the contribution of the marketing efforts of the aluminium industry. The business economics of various types of anodizing plants are analyzed. Consideration is also given to the impact of developments in anodizing technology on production economics and market growth. The economic costs associated with work rejected for process defects are considered. Recent changes in the industry have created conditions whereby information technology has a potentially important role to play in retaining existing knowledge. One such contribution is exemplified by the expert system which has been developed for the identification of anodizing process defects. Instead of using a "rule-based" expert system, a commercial neural networks program has been adapted for the task. The advantages of neural networks over 'rule-based' systems is that they are better suited to production problems, since the actual conditions prevailing when the defect was produced are often not known with certainty. In using the expert system, the user first identifies the process stage at which the defect probably occurred and is then directed to a file enabling the actual defects to be identified. After making this identification, the user can consult a database which gives a more detailed description of the defect, advises on remedial action and provides a bibliography of papers relating to the defect. The database uses a proprietary hypertext program, which also provides rapid cross-referencing to similar types of defect. Additionally, a graphics file can be accessed which (where appropriate) will display a graphic of the defect on screen. A total of 117 defects are included, together with 221 literature references, supplemented by 48 cross-reference hyperlinks. The main text of the thesis contains 179 literature references. This study examined an integrated model of the antecedents and outcomes of organisational and overall justice using a sample of Indian Call Centre employees (n = 458). Results of structural equation modelling (SEM) revealed that the four organisational justice dimensions relate to overall justice. Further, work group identification mediated the influence of overall justice on counterproductive work behaviors, such as presenteeism and social loafing, while conscientiousness was a significant moderator between work group identification and presenteeism and social loafing. Theoretical and practical implications are discussed.

Growth of complexity and functional importance of integrated navigation systems (INS) leads to high losses at the equipment refusals. The paper is devoted to the INS diagnosis system development, allowing identifying the cause of malfunction. The proposed solutions permit taking into account any changes in sensors dynamic and accuracy characteristics by means of the appropriate error models coefficients. Under actual conditions of INS operation, the determination of current values of the sensor models and estimation filter parameters rely on identification procedures. The results of full-scale experiments are given, which corroborate the expediency of INS error models parametric identification in bench test process.

In this paper, we examine the injunction issued by the prominent politician, broadcaster and older people's advocate, Baroness Joan Bakewell, to engage in ‘death talk’. We see positive ethical potential in this injunction, insofar as it serves as a call to confront more directly the prospects of death and dying, thereby releasing creative energies with which to change our outlook on life and ageing more generally. However, when set against a culture that valorises choice, independence and control, the positive ethical potential of such injunctions is invariably thwarted. We illustrate this with reference to one of Bakewell's interventions in a debate on scientific innovation and population ageing. In examining the context of her intervention, we affirm her intuition about its positive ethical potential, but we also point to an ambivalence that accompanies the formulation of the injunction – one that ultimately blunts the force and significance of her intuition. We suggest that Gilleard and Higgs' idea of the third age/fourth age dialectic, combined with the psycho-analytic concepts of fantasy and mourning, allow us to express this intuition better. In particular, we argue that the expression ‘loss talk’ (rather than ‘death talk’) better captures the ethical negotiations that should ultimately underpin the transformation processes associated with ageing, and that our theoretical contextualisation of her remarks can help us see this more clearly. In this view, deteriorations in our physical and mental capacities are best understood as involving changes in how we see ourselves, i.e. in our identifications, and so what is at stake are losses of identity and the conditions under which we can engage in new processes of identification.
