生物入侵在全世界广泛发生,目前已经受到全球关注。入侵生物对群落生物多样性和生态系统功能造成严重威胁,导致严重的环境问题和惨重的经济损失。薇甘菊(Mikania micrantha)、五爪金龙(Ipomoea cairica)和南美蟛蜞菊(Wedelia trilobata)是我国华南地区危害最严重的三种外来入侵种,其中以薇甘菊危害最严重,是世界十大有害杂草之一。从20世纪80年代发生以来,薇甘菊已在我国广东农林区域造成严重危害。 机械防治、化学防治和传统的生物防治等治理措施,未能有效治理外来入侵种的危害,直到使用本地种菟丝子(Cuscuta spp)的防治策略。与从原产地引进有害生物天敌的传统生物防治方法不同,本地种由于适应当地气候且与其他物种协同进化,因此,对生态环境潜在的负作用小。从2000年,被发现寄生和抑制薇甘菊之后,菟丝子被认为是防治薇甘菊的有效措施。 为了探讨菟丝子寄生对外来入侵种的治理,及对入侵群落的恢复,本研究在内伶仃岛的林地(入侵种群落被引入菟丝子寄生1-4年),以及东莞、深圳和海丰的干扰样地(入侵种群落被菟丝子自然寄生5年以上)开展野外调查。在每个样地分别设立外来种入侵亚群和菟丝子治理亚群,通过测定群落结构与组成、土壤性质与养分含量,以及外来种和菟丝子的生长与养分含量等参数之后,本研究得出以下主要结论。 (1) 虽然,被寄生的外来入侵种薇甘菊、五爪金龙和南美蟛蜞菊通过调节资源分配以抵御南方菟丝子(Cuscuta australis)的寄生影响,但是,菟丝子寄生导致外来入侵种生物量降低、繁殖能力下降、养分含量降低。虽然,很多寄生植物都是广谱寄生,能同时寄生多种寄主植物,但是,在本研究的被入侵的群落中,菟丝子主要寄生外来入侵植物。尤其是寄生于南美蟛蜞菊和薇甘菊的菟丝子,生长旺盛、繁殖能力强,表现出高度的适应性。因此,菟丝子对外来入侵种(南美蟛蜞菊和薇甘菊)有寄生偏好性,并对本地种的负面影响小。 (2) 通过吸收寄主的养分,田野菟丝子(Cuscuta campestris)有效地抑制了薇甘菊的危害。由于入侵种的凋落物养分含量高且分解效率高,而且,菟丝子能够促进其它凋落物的分解,并使难以被植物吸收的养分转化成易于被吸收利用的状态。因此,菟丝子与薇甘菊的寄生作用导致土壤养分含量的升高。在薇甘菊被抑制之后,本地种利用丰富的土壤养分资源,提高生长适应性,增强抵抗入侵的能力,甚至抵制薇甘菊的再生。 (3) 菟丝子的寄生作用改变了外来寄主与本地非寄主的竞争平衡,促进本地植物的生长与重建。在外来种被抑制之后,本地种的丰度和群落的物种多样性逐渐增加。本地种如:野葛(Pueraria lobata)和芦苇(Phragmites australis),取代了入侵群落中的入侵种,成了群落的优势种。而其它原先被薇甘菊抑制的本地草本、藤本和灌木,在引入菟丝子防治之后长势较好。群落稳定性与物种多样性密切相关,被治理群落本地种的增加有利于群落的演替与稳定。 (4) 被干扰的生态系统往往更容易被外来种入侵,而外来入侵种又常导致人工干扰生境的严重退化。在人工干扰样地的菟丝子对薇甘菊的抑制效果与在林地的效果一致,导致被寄生的薇甘菊生长衰退、养分竞争能力下降、入侵危害能力降低。而在薇甘菊被菟丝子治理之后,土壤养分资源增加,入侵群落的物种丰度和生物多样性提高。本 地种的重建与本地群落的恢复密切相关,利于本地被治理群落的稳定,促进被干扰植被的恢复。 菟丝子是一种治理外来入侵种危害的有效措施,适用于破碎的生境和被干扰的生态系统,尤其是在采用目前防治措施难以治理的情况下。本研究为本地种防治外来入侵种提供科学依据,且表明以入侵地的本地种治理外来入侵种有可能成为有效且可持续发展的生物防治策略。

The deep sea is Earth’s largest habitat but little is known about the nature of deep-sea parasitism. In contrast to a few characterized cases of bacterial and protistan parasites, the existence and biological significance of deep-sea parasitic fungi is yet to be understood. Here we report the discovery of a fungus-related parasitic microsporidium, Nematocenator marisprofundi n. gen. n. sp. that infects benthic nematodes at Pacific Ocean methane seeps on the Pacific Ocean floor. This infection is species-specific and has been temporally and spatially stable over two years of sampling, indicating an ecologically consistent host-parasite interaction. A high distribution of spores in the reproductive tracts of infected males and females and their absence from host nematodes’ intestines suggests a sexual transmission strategy in contrast to the fecal-oral transmission of most microsporidia. N. marisprofundi targets the host’s body wall muscles causing cell lysis, and in severe infection even muscle filament degradation. Phylogenetic analyses placed N. marisprofundi in a novel and basal clade not closely related to any described microsporidia clade, suggesting either that microsporidia-nematode parasitism occurred early in microsporidia evolution or that host specialization occurred late in an ancient deep-sea microsporidian lineage. Our findings reveal that methane seeps support complex ecosystems involving interkingdom interactions between bacteria, nematodes, and parasitic fungi and that microsporidia parasitism exists also in the deep sea biosphere.

The nematodes Trichinella spiralis and Trichinella pseudospiralis are both intracellular parasites of skeletal muscle cells and induce profound alterations in the host cell resulting in a re-alignment of muscle-specific gene expression. While T. spiralis induces the production of a collagen capsule surrounding the host-parasite complex, T. pseudospiralis exists in a non-encapsulated form and is also characterised by suppression of the host inflammatory response in the muscle. These observed differences between the two species are thought to be due to variation in the proteins excreted or secreted (ES proteins) by the muscle larva. In this study, we use a global proteomics approach to compare the ES protein profiles from both species and to identify individual T. pseudospiralis proteins that complement earlier studies with T. spiralis. Following two-dimensional gel electrophoresis, tandem mass spectrometry was used to identify the peptide spots. In many cases identification was aided by the determination of partial peptide sequence from selected mass ions. The T. pseudospiralis spots identified included the major secreted glycoproteins and the secreted 5'-nucleotidase. Furthermore, two major groups of T. spiralis-specific proteins and several T. pseudospiralis-specific proteins were identified. Our results demonstrate the value of proteomics as a tool for the identification of ES proteins that are differentially expressed between Trichinella species and as an aid to identifying key parasite proteins that are involved in the host-parasite interaction. The value of this approach will be further enhanced by data arising out the current T. spiralis genome sequencing project.

Toll-like receptors (TLRs) present in innate immune cells recognize pathogen molecular patterns and influence immunity to control the host-parasite interaction. The objective of this study was to characterize the involvement of TLR4 in the innate and adaptive immunity to Paracoccidioides brasiliensis, the most important primary fungal pathogen of Latin America. We compared the responses of C3H/HeJ mice, which are naturally defective in TLR4 signaling, with those of C3H/HePas mice, which express functional receptors, after in vitro and in vivo infection with P. brasiliensis. Unexpectedly, we verified that TLR4-defective macrophages infected in vitro with P. brasiliensis presented decreased fungal loads associated with impaired synthesis of nitric oxide, interleukin-12 (IL-12), and macrophage chemotactic protein 1 (MCP-1). After intratracheal infection with 1 million yeasts, TLR4-defective mice developed reduced fungal burdens and decreased levels of pulmonary nitric oxide, proinflammatory cytokines, and antibodies. TLR4-competent mice produced elevated levels of IL-12 and tumor necrosis factor alpha (TNF-alpha), besides cytokines of the Th17 pattern, indicating a proinflammatory role for TLR4 signaling. The more severe infection of TLR4-normal mice resulted in increased influx of activated macrophages and T cells to the lungs and progressive control of fungal burdens but impaired expansion of regulatory T cells (Treg cells). In contrast, TLR4-defective mice were not able to clear their diminished fungal burdens totally, a defect associated with deficient activation of T-cell immunity and enhanced development of Treg cells. These divergent patterns of immunity, however, resulted in equivalent mortality rates, indicating that control of elevated fungal growth mediated by vigorous inflammatory reactions is as deleterious to the hosts as low fungal loads inefficiently controlled by limited inflammatory reactions.

Human impacts through habitat destruction, introduction of invasive species and climate change are increasing the number of species threatened with extinction. Decreases in population size simultaneously lead to reductions in genetic diversity, ultimately reducing the ability of populations to adapt to a changing environment. In this way, loss of genetic polymorphism is linked with extinction risk. Recent advances in sequencing technologies mean that obtaining measures of genetic diversity at functionally important genes is within reach for conservation programs. A key region of the genome that should be targeted for population genetic studies is the Major Histocompatibility Complex (MHC). MHC genes, found in all jawed vertebrates, are the most polymorphic genes in vertebrate genomes. They play key roles in immune function via immune-recognition and -surveillance and host-parasite interaction. Therefore, measuring levels of polymorphism at these genes can provide indirect measures of the immunological fitness of populations. The MHC has also been linked with mate-choice and pregnancy outcomes and has application for improving mating success in captive breeding programs. The recent discovery that genetic diversity at MHC genes may protect against the spread of contagious cancers provides an added impetus for managing and protecting MHC diversity in wild populations. Here we review the field and focus on the successful applications of MHC-typing for conservation management. We emphasize the importance of using MHC markers when planning and executing wildlife rescue and conservation programs but stress that this should not be done to the detriment of genome-wide diversity.

O presente estudo consiste em uma caracterização preliminar da atividade proteolítica de frações de proteínas purificadas a partir de lisados de trofozoítos de cepa isolada e axenizada no Brasil. Frações obtidas por cromatografia líquida (FPLC) foram analisadas quanto ao perfil eletroforético em géis de poliacrilamida (SDS-PAGE) e a atividade proteolítica foi avaliada em géis contendo gelatina como substrato. A caracterização das enzimas foi realizada a partir da análise do efeito de inibidores sintéticos de cisteína-proteases (E-64, IAA), serina-proteases (PMSF), serina e cisteína-proteases (TPCK, TLCK, elastatinal), metalo-proteases (EDTA) e aspartil proteases (pepstatina) sobre a degradação do substrato. Entre 30 frações eluídas, bandas de proteínas foram observadas em oito delas, entretanto, atividade proteolítica foi detectada apenas nas frações 23, 24, 25 e 26. O perfil eletroforético das proteínas revelou poucas bandas distribuídas na faixa de 45 a 18 kDa. Os zimogramas revelaram zonas de proteólise na faixa de aproximadamente 62 a 35 kDa, entretanto destacaram-se as bandas de hidrólise de 62, 55, 53, 50, 46 e 40 kDa. Nos ensaios de inibição, a proteólise foi marcantemente inibida por E-64, TPCK, TLCK e elastatinal. Redução discreta da proteólise foi observada com IAA e PMSF, enquanto que EDTA e pepstatina não promoveram alteração dos perfis de hidrólise. Estas observações são relevantes, especialmente se considerarmos que para elucidar o envolvimento das proteases na relação parasita-hospedeiro, a purificação dessas moléculas é um requisito importante.

A comparative approach is potentially useful for understanding the role of mammal innate immunity role in stimulating adaptive immunity as well as the relationship between these two types of immune strategies. Considerable progress has been made in the elucidation of the co-ordinated events involved in plant perception of infection and their mobilisation of defence responses. Although lacking immunoglobulin molecules, circulating cells, and phagocytic processes, plants successfully use pre-formed physical and chemical innate defences, as well as inducible adaptive immune strategies. O Trypanosoma cruzi, agente etiológico da doença de Chagas, apresenta elevado grau de variabilidade genética intra-específica, com possíveis implicações na forma clínica da doença, como o desenvolvimento de cardiopatia, do megaesôfago e do megacólon de forma isolada ou em associação. Este tropismo tecidual envolvido na patogênese da doença não está totalmente esclarecido. Assim, nesta revisão são abordados alguns aspectos referentes à diversidade genética dos parasitas isolados, às formas clínicas da doença de Chagas, ao processo de infecção do parasita na célula hospedeira e resposta imune. Outros aspectos também são enfocados, como os fatores imunossupressivos liberados pelo parasita que atuam na regulação das respostas imunes, a inibição da apoptose da célula hospedeira, assim como da patogênese do megaesôfago chagásico que pode estar relacionada à interação hospedeiro- parasita e sua associação com risco aumentado para o desenvolvimento do carcinoma epidermóide do esôfago. Porém, apesar dos avanços no entendimento desta doença, ainda não é possível estabelecer o verdadeiro perfil da variabilidade genética do parasita com a forma clínica da doença de Chagas.

Some aspects of the widely observed over-dispersed pattern of the distribution of parasites within the host population are examined. It has been established in the parasitological literature that most hosts usually harbour few parasites, while only few hosts harbour a large proportion of the parasite population. Factors that may influence the pattern of distribution of parasites, the relation between the level of parasite aggregation and the prevalence of infection, and changes in this level of aggregation as a function of host age are analysed. Factors which determine the diversity of species in parasite communities are presented, and aspects of exploitative and interference competition among parasites and their relations with biological control procedures are also considered. Attention is also focused on the regulatory and destabilizing processes influencing the dynamic behaviour of host-parasite population interactions.

Giardia duodenalis isolates from asymptomatic or symptomatic patients and from animals present similarities and differences in the protein composition, antigenic profile, pattern of proteases and isoenzymes, as well as in nucleic acids analysis. In the present overview, these differences and similarities are reviewed with emphasis in the host-parasite interplay and possible mechanisms of virulence of the protozoon.

This study describes the occurrence and the seasonality of parasites of cultivated fish from a fee fishing farm located in Guariba, São Paulo State, Brazil (21°15'22'' S, 48°18'58'' W and 595 m of altitude), from August, 2001 to July, 2002. The presence of parasites was researched in pacu Piaractus mesopotamicus (Characidae), common carp Cyprinus carpio (Cyprinidae), nile-tilapia Oreochromis niloticus (Cichlidae), tambacu hybrid (male of P. mesopotamicus x female of tambaqui-Colossoma macropomum) and piraputanga Brycon hillari (Characidae). Results demonstrate that out of 100 fish examined, 15% were sponged for at least one of the following parasites: Trichodina sp.; monogenean helminths; copepodits of Lemaea cyprinacea; adults of L. cyprinacea; or Dolops carvalhoi. In decreasing order, the susceptibility degree of the hosts was C. carpio, P. mesopotamicus, B. hillari, tambacu hybrid and O. niloticus. In decreasing order the reported parasites were monogenean helminths, Dolops carvalhoi, Trichodina sp., adults of Lernaea cyprinacea and their young shapes.

The present study demonstrated the effects of the arthropod growth regulator, fluazuron (Acatak®), in the formation of the integument and digestive processes of Rhipicephalus sanguineus nymphs fed on rabbits treated with different doses of this chemical acaricide. For this, three different doses of fluazuron (20, 40, or 80 mg/kg) were applied pour on to the hosts (groups II, III, and IV), as well as distilled water to the control group. On the first day after treatment (24 h), the hosts were artificially infested with R. sanguineus nymphs. After full engorgement (7 days), the nymphs were removed, placed on labeled Petri dishes, and kept in biochemical oxygen demand incubator for 7 days. The engorged nymphs were then taken for morphological, histochemical, and histological analyses. The results showed the occurrence of cytological, morphohistological, and histochemical alterations in the integument and midgut of nymphs from all the different treated groups. These alterations occurred at cuticular level in the subdivisions of the cuticle, related to the size of the digestive cells, amount of accumulated blood elements, and digestive residues, as well as the presence of vacuoles in the cytoplasm of the digestive cells. Thus, this study demonstrated that fluazuron acts on the integument and midgut cells of R. sanguineus nymphs fed on treated rabbits and pointed out the possibility of the use of this chemical - which is more specific, less toxic, and less harmful to the environment and nontarget organisms - in the control of R. sanguineus, at least in the nymphal stage of its biological cycle. © 2012 Springer-Verlag.
