A pattern recognition protein (PRP), lipopolysaccharide and beta-1,3-glucan binding protein (LGBP) cDNA was cloned from the haemocyte of Chinese shrimp Fenneropenaeus chinensis by the techniques of homology cloning and RACE. Analysis of nucleotide sequence revealed that the full-length cDNA of 1,275 bp has an open reading frame of 1,098 bp encoding a protein of 366 amino acids including a 17 amino acid signal peptide. Sequence comparison of the deduced amino acid sequence of F. chinensis LGBP showed a high identity of 94%, 90%, 87%, 72% and 63% with Penaeus monodon BGBP, Litopenaeus stylirostris LGBP, Marsupenaeu japonicus BGBP, Homarus gammarus BGBP and Pacifastacus leniusculus LGBP, respectively. The calculated molecular mass of the mature protein is 39,857 Da with a deduced pI of 4.39. Two putative integrin binding motifs, RGD (Arg-Gly-Asp) and a potential recognition motif for beta-1,3-linkage of polysaccharides were observed in LGBP sequence. RT-PCR analysis showed that LGBP gene expresses in haemocyte and hepatopancreas only, but not in other tissues. Capillary electrophoresis RT-PCR method was used to quantify the variation of mRNA transcription level during artificial infection with heat-killed Vibrio anguillarum and Staphylococcus aureusin. A significant enhancement of LGBP transcription was appeared at 6 h post-injection in response to bacterial infection. These results have provided useful information to understand the function of LGBP in shrimp.

性比是反映动物种群的基本特征之一,因此成为种群生态学的主要研究内容。就种群的性比而言,在不同的物种、或同一物种在不同的时期和条件下都有极大的不同~[1-4]。性比的改变将影响动物的种群结构、家庭组成和交配关系,也影响动物两性的配偶竞争、繁殖投入、繁殖成功以及性二型的分化等~[4-13]。在繁殖过程中,动物可以采用改变性比的方法调节种群的密度和大小[2,14,15]。同时,动物的性比受多种因素的影响,不仅存在季节、年间以及地理变化的差异,而且受环境条件(如气温和降水量)的影响~[2~4]。捕食风险也是影响性比的主要因素。虽然许多动物出生时的性比是一样的,但是,由于捕食者在捕食猎物的过程中存在明显的选择性,使其性比发生了明显的变化~[16]。对于动物种群性比的研究已有诸多报道,其研究的动物包括海蛇尾(Ophiactis savignyi)~[5]、龙虾(Homarus gam- marus)~[6]、射毒蛙(Dendrobates purnilio)~[9]、斑马雀(Taeniopygyia guttata castanotis)~[8]、褐家鼠(Rattus norvegicus)~[17]等。高原鼠兔(Ochotona curzoniae)是青藏高原草地生态系统中的优势小哺乳类动物。有关高原鼠兔种群性比的研究,国内学者有过涉及~[18,19],但所作工作多是在高原鼠兔繁殖生态学研究中进行的静态描述,对种群性比的年龄变化,季节和年度变化及其与环境条件的关系等方面的研究尚缺详细的报道。本文通过对高原鼠兔种群性比的年间比较、不同胎次性比的分析和越冬对其性比影响的研究,旨在探讨高原鼠兔的性比变化在其种群调节中的作用。
The co-location of offshore windfarms and decapod fisheries in the UK: Constraints and opportunities

The offshore wind sector in the UK is expanding rapidly and is set to occupy significant areas of the coastal zone, making it necessary to explore the potential for co-location with other economic activities. The presence of turbine foundations introduces hard substrates into areas previously dominated by soft sediments, implying that artificial reef effects may occur, with potential benefits for fisheries. This review focuses on the possibilities for locating fisheries for two commercially important decapods, the brown crab Cancer pagurus and the European lobster Homarus gammarus, within offshore wind farms. Existing understanding of habitat use by C pagurus and H. gammarus suggests that turbine foundations have the potential to act as artificial reefs, although the responses of these species to noise and electromagnetic fields are poorly understood. Offshore wind farm monitoring programmes provide very limited information, but do suggest that adult C pagurus associate with turbine foundations, which may also serve as nursery areas. There was insufficient deployment and monitoring of rock armouring to draw conclusions about the association of H. gammarus with offshore wind farm foundations. The limited information currently available demonstrates the need for further research into the ecological and socioeconomic issues surrounding fishery co-location potential.

Ultrastructural investigations of eggs can be important in helping to understand embryonic development. There are few transmission electron microscope studies of marine arthropod eggs, however, as they have proved difficult to fix and infiltrate with resin. Here, we describe a modification of a standard method that allows the preparation of the quite different eggs of the marine copepod, Acartia tonsa and the lobster, Homarus gammarus, for transmission electron microscopy. By using double fixation and an extended resin infiltration time we obtained good preparations for electron microscopy. We anticipate that these modifications to the standard protocol will be widely applicable and useful for the study of the eggs and early developmental stages of many marine arthropod taxa. Les recherches sur l'ultrastructure des oeufs peuvent être importantes en aidant à comprendre le développement embryonnaire. Il existe cependant peu d'études en microscopie électronique à transmission sur les oeufs d'arthropodes marins, car il est difficile de les fixer et d'y infiltrer de la résine. Dans ce travail, nous décrivons une modification de la méthode standard, qui permet la préparation pour la microscopie électronique à transmission d'oeufs aussi différents que ceux du copépode marin Acartia tonsa et du homard Homarus gammarus. En utilisant une double fixation et un temps plus long d'infiltration de la résine, nous avons obtenu de bonnes préparations pour la microscopie électronique. Nous prévoyons que ces modifications du protocole standard seront largement applicables et utiles pour l'étude des oeufs et des premiers stades de développement de nombreux taxons d'arthropodes marins.

Lobsters are renowned the world over as tasty delicacies and are in great demand for Epicurian gourests. As a result these animals found in different parts of the world from countries like India they are mostly frozen and exported thus earning considerable foreign exchange for the country. Importance of this crustacean is thus well known and whatever harvested from nature are utilized to the maximum extent. The demand being much more than what is being produced the need for artificial culture becomes evident. In this context a complete knowledge of the lifecycle ,larval biology and their culture is very essential. Thus with the object of studying the life history of the commercially important lobsters fished from Indian waters the larval biology of panulirus homarus has been taken up as the project for the present study

Success achieved in seed production and culture of prawns induced researchers to explore the feasibiltiy of culture of other important crustaceans like lobsters and crabs. The CMFRI initiated a project to study the prospects of spiny lobster’ culture and I was associated with this project from the beginning. Compared to shrimp farming, culture of spiny lobsters is an entirely different proposition due to inherent problems in producing their seeds. The emphasis on .obster culture, therefore, shifted to rearing of juvenile lobsters, which form a major portion in commercial lobster landings in India. Commendable success was achieved in enhancing growth rate of four important speices of spiny lobsters by bilateral eyestalk ablation. But due to strong reservations expressed against the "blinding" of lobsters by people from all walks of life, this technique could not be tested commercially for its economic viability. As in prawns, energetics of reproduction and egg development have not yet been documented in spiny lobsters and so these aspects were included in this study. Though some limited informations on food conversion efficiency are availble in spiny lobster, a detailed study on the effect of size and sexual maturity on food conversion was wanting and this also was included in this investigation. The most dominant spiny lobster in south east and south west coast of India, Panulirus homarus (Linnaeus) was selected for this study. Uptake, accumulation and utilization of minerals and trace elements are important events during embryogenesis of aquatic invertebrates. The scope of this study was widened to include these aspects also in order to understand how specific requirements of minerals and trace elements are met in the developing eggs of these two species of crustaceans.

The management of exploited species requires the identification of demographically isolated populations that can be considered as independent management units (MUs), failuring in which can lead to over -fishing and depletion of less productive stocks. By characterizing the distribution of genetic variation, population sub structuring can be detected and the degree of connectivity among populations can be estimated. The genetic variation can be observed using identified molecular markers of both nuclear and mitochondrial origin. Hence, the present work was undertaken to study the genetic diversity and population/stock structure in P. homarus homarus and T. unimaculatus from different landing centres along the Indian coast using nuclear (RAPD) and mitochondrial DNA marker tools which will help towards developing management strategies for management and conservation of these declining resources.To make consistent conservation and fisheries management decisions, accurate species identifications are needed. It is also suggested that it is not always desirable to rely on a single sequence for taxonomic identification. Thus, the feasibility of using partial sequences of additional mitochondrial genes like 16SrRNA, 12SrRNA and nuclear 18SrRNA has also been explored in our study. Phylogenies provide a sound foundation for establishing taxonomy. The present work also attempts to reconstruct the phylogeny of eleven species of commercially important lobsters from the Indian EEZ using molecular markers

The developmental stages of the ovaries of the lobster Panulirus echinatus Smith, 1869 were characterized using macroscopic and microscopic features and the gonadosomatic relation (GSR). Based on monthly samples (November, 1999 to October, 2000), a total of 711 females were captured using gillnets. The dorsal region of the carapace was removed to evaluate the ovaries, which were dissected, weighted, preserved in Bouin`s solution, and submitted to histological procedures. The microscopic analysis of the ovaries was assessed by the presence of germinative cells in different developmental phases. When this analysis was combined with macroscopical observations (changes in color and volume of the gonads in the cephalothorax) and GSR, five developmental stages could be identified: immature (I), intermediate (II); pre-maturation (III); mature (IV) and post-spawning stage (V). Statistical analyses confirmed that GSR can be used as an indicator of developmental stage.

This paper characterizes the developmental stages of the testes and vasa deferentia of the Panulirus echinatus Smith, 1869 through comparisons between microscopic findings, macroscopic aspects, and gonadosomatic index (GSR). The lobsters were sampled monthly (November 1999 to October 2000) using seine nets and a total of 1716 males were obtained at Tamandare Bay. Each carapace was cut to allow evaluation of the reproductive organs; the testes and vasa deferentia were dissected, weighed, fixed in Bouin`s solution up to 12 hours and submitted for histological analysis to determine the presence and/or absence of spermatozoa. These measures, along with change in color, size, diameter, development of the spermatophores and the GSR allowed the caracterization of three development stages: immature, intermediate and ripe. In conclusion, the maturity of the testes precedes the maturity of the vasa deferentia. To evaluate if gonadosomatic relation was a good quantitative indicator of the maturity stage, t tests (alpha = 0,05) were used and verified significant difference in the averages of GSR. The statistics corroborated that GSR can be used as indicative of the developmental stages for P. echinatus.

We investigated modulation by ATP, Mg2+, Na+, K+ and NH4 (+) and inhibition by ouabain of (Na+,K+)-ATPase activity in microsomal homogenates of whole zoeae I and decapodid III (formerly zoea IX) and whole-body and gill homogenates of juvenile and adult Amazon River shrimps, . (Na+,K+)-ATPase-specific activity was increased twofold in decapodid III compared to zoea I, juveniles and adults, suggesting an important role in this ontogenetic stage. The apparent affinity for ATP ( (M) = 0.09 +/- A 0.01 mmol L-1) of the decapodid III (Na+,K+)-ATPase, about twofold greater than the other stages, further highlights this relevance. Modulation of (Na+,K+)-ATPase activity by K+ also revealed a threefold greater affinity for K+ ( (0.5) = 0.91 +/- A 0.04 mmol L-1) in decapodid III than in other stages; NH4 (+) had no modulatory effect. The affinity for Na+ ( (0.5) = 13.2 +/- A 0.6 mmol L-1) of zoea I (Na+,K+)-ATPase was fourfold less than other stages. Modulation by Na+, Mg2+ and NH4 (+) obeyed cooperative kinetics, while K+ modulation exhibited Michaelis-Menten behavior. Rates of maximal Mg2+ stimulation of ouabain-insensitive ATPase activity differed in each ontogenetic stage, suggesting that Mg2+-stimulated ATPases other than (Na+,K+)-ATPase are present. Ouabain inhibition suggests that, among the various ATPase activities present in the different stages, Na+-ATPase may be involved in the ontogeny of osmoregulation in larval The NH4 (+)-stimulated, ouabain-insensitive ATPase activity seen in zoea I and decapodid III may reflect a stage-specific means of ammonia excretion since functional gills are absent in the early larval stages.

In vielen Arthropoden wird Sauerstoff mittels des Kupferproteins Hämocyanin transportiert. In der vorliegenden Arbeit wurde das Hämocyanin einiger Vertreter der Arthropoden molekularbiologisch untersucht. Bei den Crustaceen (Homarus americanus und Palinurus elephas) konnten vier Untereinheiten isoliert werden, bei denen es sich um Hämocyanine des a-Typs handelt. Bei den Myriapoden (Chilopoda, Scutigera coleoptrata und Diplopoda, Spirostreptus spec.) konnten drei unterschiedliche Hämocyaninuntereinheiten gefunden werden. Erst seit einiger Zeit ist das Hämocyanin bei Myriapoden biochemisch charakterisiert und bei diesen Hämocyaninsequenzen handelt es sich um die ersten von Myriapoden. Bei den Onychophoren (Epiperipatus spec.) konnte ebenfalls erstmals ein Hämocyanin isoliert und sequenziert werden. Hierbei handelt es sich um den ersten Hinweis, dass Onychophoren über ein respiratorisches Protein verfügen. In einer phylogenetischen Analyse der Hämocyaninsequenzen konnte ein Stammbaum der Hämocyaninsuperfamilie erstellt werden. Innerhalb der Crustacea ordnen sich die verschiedenen Hämocyaninuntereinheiten in distinkten Ästen an. Die Hämocyanine der Myriapoda sind monophyletisch, wobei die Auftrennung in distinkte Untereinheiten bereits vor der Trennung der Chilopoden und Diplopoden erfolgte; die phylogenetische Stellung der Myriapoda kann anhand der Hämocyaninsequenzen nicht zuverlässig aufgelöst werden. Eine gemeinsame Anordnung mit den Hexapoda ('Tracheata'-Hypothese) kann jedoch mit hoher Sicherheit ausgeschlossen werden. Die Onychophora stehen im Arthropodenstammbaum an basaler Position und können somit als Proarthropoda angesprochen werden. Es konnte gezeigt werden, dass die Hämocyaninevolution der Arthropodenevolution entspricht.

Zusammenfassung rnrnIn dieser Arbeit wurden Untersuchungen an zwei verschiedenen multimeren Proteinkomplexen durchgeführt: Zum einen am Hämocyanin aus Homarus americanus mittels Röntgen-L-Kantenspektroskopie und zum anderen am α-Toxin aus Staphylococcus aureus, hinsichtlich der Interaktion an speziellen Raft-artigen Membranabschnitten, mittels AFM.rnFür das Hämocyanin aus Homarus americanus konnte ein neuer Aspekt bezüglich der Bindung von Sauerstoff aufgezeigt werden. Ein zuvor nicht in Betracht gezogener und diskutierter Einfluss von Wassermolekülen auf diesen Vorgang konnte mittels der Methode der Röntgen-L-Kantenspektroskopie dargestellt werden. Erstmals war es möglich die beiden verschiedenen Beladungszustände (Oxy-, Deoxy-Zustand) des Hämocyanin mittels dieser Methode in physiologisch ähnlicher Umgebung zu untersuchen. Vergleiche der erhaltenen L-Kanten-Spektren mit denen anorganischer Vergleichslösungen ließen auf eine Interaktion von Wassermolekülen mit den beiden Kupferatomen des aktiven Zentrums schließen. Dadurch wurde erstmals ein möglicher Einfluss des Wassers auf den Oxygenierungsprozess des Hämocyanins auf elektronischer Ebene aufgezeigt. Vergleichende Betrachtungen von Röntgenkristallstrukturen verschiedener Typ-3-Kupferproteine bestätigten, dass auch hier ein Einfluss von Wassermolekülen auf die aktiven Zentren möglich ist. Vorgeschlagen wird dabei, dass an Stelle der Überlappung der 3d-Orbitale des Kupfers mit den 2p-Orbitalen des Sauerstoffs, wie sie im sauerstoffbeladenen Zustand auftritt, im sauerstoffunbeladenen Zustand eine Wechselwirkung der 3d-Orbitale des Kupfers mit den LUMOS der Wassermoleküle möglich wird, und ein Elektronen- bzw. Ladungstransfer von den Kupfern auf die Wassermoleküle erfolgen kann. rnAFM-Untersuchungen hinsichtlich der Interaktion des α-Toxins aus Staphylococcus aureus mit oberflächenunterstützten Modellmembranen wiesen darauf hin, dass eine bevorzugte Anbindung und zumindest teilweise Integration der α-Toxine in Raft-artige Membranbereiche stattfindet. Für verschiedene ternäre Lipidsysteme konnten phasenseparierte Modellmembranen abgebildet und die unterschiedlichen Domänenformen zugeordnet werden. Der Anbindungsprozess der Toxine an diese oberflächenunterstützte Modellmembranen erfolgte dann wahrscheinlich vornehmlich an den speziellen Raft-artigen Domänen, wohingegen die Insertion der Poren vorrangig an den Grenzbereichen zwischen den Domänen auftrat. Mögliche Ursache dafür sind die räumlichen Besonderheiten dieser Grenzflächen. Membranen weisen an den Schnittstellen zwischen zwei Domänenformen eine erhöhte Unordnung auf, was sich u.a. in einer geringeren Packungsdichte der Phospholipide und dem erhöhten Freiheitsgrad ihrer Kopfgruppen bemerkbar macht. Außerdem kommt es auf Grund der Interaktion der beteiligten Membranbestandteile Sphingomyelin und Cholesterol untereinander zu einer speziellen Ausrichtung der Phosphocholin-Kopfgruppen und innerhalb der Raft-artigen Domänen zu einer erhöhten Packungsdichte der Phospholipide. Die in dieser Arbeit präsentierten Ergebnisse unterstützten demnach die in der Literatur postulierte Vermutung der bevorzugten Interaktion und Integration der Toxin-Moleküle mit Raft-artigen Membrandomänen. Die Insertion der Pore erfolgt aber wahrscheinlich bevorzugt an den Grenzbereichen zwischen den auftretenden Domänen.rn

Approximately 350 base pairs (bp) of the mitochondrial 16S rRNA gene were used to study the phylogenetic relationships among 5 genera of the clawed lobster family Nephropidae (infraorder Astacidea), including Homarus, Homarinus, Metanephrops, Nephrops, and Nephropsis. Maximum-parsimony analysis, using a hermit crab, Pagurus pollicaris (infraorder Anomura), as an outgroup. produced a tree topology in which Homarus and Nephrops formed a well-supported clade that excluded Homarinus. The same tree topology was obtained from both neighbor-joining and maximum-likelihood analyses, Some morphological characters that appear synapomorphic for Nephrops and Metanephrops may be due to convergence rather than symplesiomorphy. The current taxonomy, therefore, does not reflect the phylogeny of this group as suggested by the molecular data. More molecular data and studies using homologous morphological characters me needed to reach a better understanding of the phylogenetic history of clawed lobsters.

In the coming decades, artificial defence structures will increase in importance worldwide for the protection of coasts against the impacts of global warming. However, the ecological effects of such structures on the natural surroundings remain unclear. We investigated the impact of experimentally introduced tetrapod fields on the demersal fish community in a hard-bottom area in the southern North Sea. The results indicated a significant decrease in fish abundance in the surrounding area caused by migration effects towards the artificial structures. Diversity (HB) and evenness (E) values exhibited greater variation after the introduction of the tetrapods. Additionally, a distinct increase in young-of-the-year (YOY) fish was observed near the structures within the second year after introduction. We suggest that the availability of adequate refuges in combination with additional food resources provided by the artificial structures has a highly species-specific attraction effect. However, these findings also demonstrate that our knowledge regarding the impact of artificial structures on temperate fish communities is still too limited to truly understand the ecological processes that are initiated by the introduction of artificial structures. Long-term investigations and additional experimental in situ work worldwide will be indispensable for a full understanding of the mechanisms by which coastal defence structures interact with the coastal environment.
