Na América do Sul, alguns canídeos silvestres são considerados reservatórios naturais da Leishmania chagasi. A resposta imunológica desses animais à Leishmania é pouco conhecida, havendo a necessidade de métodos diagnósticos adequados para esse fim. No presente estudo, é descrita a padronização do ensaio imunoenzimático indireto (ELISA) para o diagnóstico sorológico de leishmaniose visceral em canídeos silvestres brasileiros. Foram estudadas amostras de soro e plasma de 12 canídeos cativos: sete lobos-guará (Chrysocyon brachyurus), três raposinhas (Lycalopex vetulus) e dois cachorros-do-mato (Cerdocyon thous). As amostras de um C. brachyurus e uma L. vetulus, cativos em área endêmica para LV, que apresentavam doença clínica e positividade em testes de Imunofluorescência Indireta e Reação em Cadeia de Polimerase, foram utilizadas como controles positivos. Foram comparados os conjugados anti-IgG de cão e proteína A, ambos ligados a peroxidase, cujos testes detectaram quatro (04/12) e três (03/12) C. brachyurus soropositivos para anticorpos anti-Leishmania sp., respectivamente. As médias das densidades ópticas (DOs) das amostras negativas foram nitidamente mais baixas do que as médias das DOs dos positivos tanto no ELISA com anti-IgG de cão (4,8 vezes) como com proteína A (15,5 vezes). Os soros de três C. brachyurus positivos no ELISA indireto foram avaliados por Western blotting e identificaram 22 bandas, sendo imunodominantes as de peso molecular de 19, 22, 24, 45 e 66 kDa. Os testes ELISA com a proteína A e o conjugado anti-IgG de cão apresentaram respectivamente concordância excelente (Kappa = 1; p<0,001) e moderada (Kappa = 0,8; p<0,0015), com o Western blotting. Ambos foram, portanto, considerados adequados a avaliações de triagem de animais cuja resposta humoral de anticorpos indica contato com o parasito, úteis para subsidiar estudos para adequação de metodologias específicas para os canídeos silvestres.

A lectin present in the marine red alga Pterocladiella capillacea was purified and characterised by extraction of soluble proteins (crude extract) in 20 mM Tris-HCl buffer, pH 7.5. Among the analysed erythrocytes (human blood group A, B and O and the animals ox, goat, chicken and rabbit) the lectin agglutinated specifically rabbit erythrocytes. The hemagglutinating activity assay showed that the lectin was not dependent on divalent cations and was shown to be inhibited by the glycoproteins avidin and mucin. The purification procedure was conduced by precipitation of the crude extract with 80% saturation ammonium sulfate (F0/80) followed by affinity chromatography on guar-gum column. The lectin of P. capillacea was purified 14.5 fold and had a recovery of 27.4% of the original total specific activity present in the crude extract. The absence of carbohydrate suggested that the lectin is not a glycoprotein. The molecular mass of P. capillacea lectin, determined by gel filtration, was 5.8 kDa. SDS-PAGE in the presence of ß-mercaptoethanol gave one band, indicating that the native lectin is a monomeric protein. The activation energy of denaturation process (D G') was calculated to be 106.87 kJ . mol-1 at 70 ºC.

A lectina da alga marinha vermelha Vidalia obtusiloba foi purificada através da combinação de precipitação com sulfato de amônio, cromatografia de troca iônica em DEAE-celulose e cromatografia de afinidade em goma de guar reticulada. A lectina preferencialmente aglutinou eritrócitos do grupo humano O, nativos e tratados com bromelaina. A atividade hemaglutinante revelou que a lectina era dependente de cátions divalentes (Ca++ ou Mn++) e inibida por N-acetil-galactosamina, D-galactosamina, alfa-lactose and D-galactose e pela glicoproteína mucina de estômago de porco. A massa molecular da lectina, estimada por filtração em gel, foi de 78,9 kDa, enquanto por SDS-PAGE, em presença de beta-mercaptoetanol, a lectina exibiu duas subunidades protéicas diferentes com Mr de 59,6 e 15,2 kDa, sugerindo que a lectina é uma proteína dimérica. Focalização isoelétrica revelou a presença de uma proteína ácida simples, com um ponto isoelétrico entre 4 e 5. A lectina purificada exibiu um conteúdo de carboidrato de 43,2% e uma predominância dos aminoácidos Asp/Asn, Glu/Gln e Leu. A energia de ativação (deltaG') para a desnaturação da lectina foi calculada como sendo 25,4 kcalm.mol-1 a 90 ºC. Ensaios imunoquímicos usando um anti-soro de coelho produzido contra a lectina purificada de V. obtusiloba mostraram que foi possível detectar a presença da lectina em diferentes etapas do processo de purificação. Western blotting de géis de SDS-PAGE mostraram imunocoramento apenas da maior das subunidades da lectina.
Viscosity of gums in vitro and their ability to reduce postprandial hyperglycemia in normal subjects

Experiments were carried out in vitro with three viscous polysaccharides (guar gum, pectin, and carboxymethylcellulose (CMC)) of similar initial viscosity submitted to conditions that mimic events occurring in the stomach and duodenum, and their viscosity in these situations was compared to their actions on postprandial hyperglycemia in normal human subjects. Guar gum showed greater viscosity than the other gums during acidification and/or alkalinization and also showed larger effects on plasma glucose levels (35% reduction in maximum rise in plasma glucose) and on the total area under the curve of plasma glucose (control: 20,314 ± 1007 mg dl-1 180 min-1 vs guar gum: 18,277 ± 699 mg dl-1 180 min-1, P<0.01). Pectin, which showed a marked reduction in viscosity at 37oC and after events mimicking those that occur in the stomach and duodenum, did not have a significant effect on postprandial hyperglycemia. The performance of viscosity and the glycemia response to CMC were at an intermediate level between guar gum and pectin. In conclusion, these data suggest that temperature, the process of acidification, alkalinization and exposure to intestinal ions induce different viscosity changes in gums having similar initial viscosity, establishing a direct relationship between a minor decrease of gum viscosity in vitro and a reduction of postprandial hyperglycemia

In recent years, there have been studies that show a correlation between the hyperactivity of children and use of artificial food additives, including colorants. This has, in part, led to preference of natural products over products with artificial additives. Consumers have also become more aware of health issues. Natural food colorants have many bioactive functions, mainly vitamin A activity of carotenoids and antioxidativity, and therefore they could be more easily accepted by the consumers. However, natural colorant compounds are usually unstable, which restricts their usage. Microencapsulation could be one way to enhance the stability of natural colorant compounds and thus enable better usage for them as food colorants. Microencapsulation is a term used for processes in which the active material is totally enveloped in a coating or capsule, and thus it is separated and protected from the surrounding environment. In addition to protection by the capsule, microencapsulation can also be used to modify solubility and other properties of the encapsulated material, for example, to incorporate fat-soluble compounds into aqueous matrices. The aim of this thesis work was to study the stability of two natural pigments, lutein (carotenoid) and betanin (betalain), and to determine possible ways to enhance their stability with different microencapsulation techniques. Another aim was the extraction of pigments without the use of organic solvents and the development of previously used extraction methods. Stability of pigments in microencapsulated pigment preparations and model foods containing these were studied by measuring the pigment content after storage in different conditions. Preliminary studies on the bioavailability of microencapsulated pigments and sensory evaluation for consumer acceptance of model foods containing microencapsulated pigments were also carried out. Enzyme-assisted oil extraction was used to extract lutein from marigold (Tagetes erecta) flower without organic solvents, and the yield was comparable to solvent extraction of lutein from the same flowers. The effects of temperature, extraction time, and beet:water ratio on extraction efficiency of betanin from red beet (Beta vulgaris) were studied and the optimal conditions for maximum yield and maximum betanin concentration were determined. In both cases, extraction at 40 °C was better than extraction at 80 °C and the extraction for five minutes was as efficient as 15 or 30 minutes. For maximum betanin yield, the beet:water ratio of 1:2 was better, with possibly repeated extraction, but for maximum betanin concentration, a ratio of 1:1 was better. Lutein was incorporated into oil-in-water (o/w) emulsions with a polar oil fraction from oat (Avena sativa) as an emulsifier and mixtures of guar gum and xanthan gum or locust bean gum and xanthan gum as stabilizers to retard creaming. The stability of lutein in these emulsions was quite good, with 77 to 91 percent of lutein being left after storage in the dark at 20 to 22°C for 10 weeks whereas in spray dried emulsions the retention of lutein was 67 to 75 percent. The retention of lutein in oil was also good at 85 percent. Betanin was incorporated into the inner w1 water phase of a water1-in-oil-inwater2 (w1/o/w2) double emulsion with primary w1/o emulsion droplet size of 0.34 μm and secondary w1/o/w2 emulsion droplet size of 5.5 μm and encapsulation efficiency of betanin of 89 percent. In vitro intestinal lipid digestion was performed on the double emulsion, and during the first two hours, coalescence of the inner water phase droplets was observed, and the sizes of the double emulsion droplets increased quickly because of aggregation. This period also corresponded to gradual release of betanin, with a final release of 35 percent. The double emulsion structure was retained throughout the three-hour experiment. Betanin was also spray dried and incorporated into model juices with different pH and dry matter content. Model juices were stored in the dark at -20, 4, 20–24 or 60 °C (accelerated test) for several months. Betanin degraded quite rapidly in all of the samples and higher temperature and a lower pH accelerated degradation. Stability of betanin was much better in the spray dried powder, with practically no degradation during six months of storage in the dark at 20 to 24 °C and good stability also for six months in the dark at 60 °C with 60 percent retention. Consumer acceptance of model juices colored with spray dried betanin was compared with similar model juices colored with anthocyanins or beet extract. Consumers preferred beet extract and anthocyanin colored model juices over juices colored with spray dried betanin. However, spray dried betanin did not impart any off-odors or off-flavors into the model juices contrary to the beet extract. In conclusion, this thesis describes novel solvent-free extraction and encapsulation processes for lutein and betanin from plant sources. Lutein showed good stability in oil and in o/w emulsions, but slightly inferior in spray dried emulsions. In vitro intestinal lipid digestion showed a good stability of w1/o/w2 double emulsion and quite high retention of betanin during digestion. Consumer acceptance of model juices colored with spray dried betanin was not as good as model juices colored with anthocyanins, but addition of betanin to real berry juice could produce better results with mixture of added betanin and natural berry anthocyanins could produce a more acceptable color. Overall, further studies are needed to obtain natural colorants with good stability for the use in food products.

A utilização de gomas de sementes tem trazido muitos benefícios à indústria de alimentos, melhorando as propriedades dos produtos, principalmente pelo seu poder espessante, estabilizante e geleificante. Gomas obtidas a partir do endosperma de sementes de leguminosas são compostas por manose e galactose (galactomanano). A partir da trituração e posterior peneiramento das sementes de faveiro (Dimorphandra mollis), espécie brasileira que ocorre com alta freqüência nos cerrados e caatinga, foi obtido um extrato rico em galactomanano (83,2%) com proporção manose:galactose 2,7. Esses parâmetros são comparáveis aos obtidos para os galactomananos de uso consagrado em formulações de alimentos, tais como a goma guar com 78-82% de rendimento e razão manose galactose igual a 2 e a goma locusta com 88% de rendimento e razão manose:galactose igual a 4. Ensaios biológicos demonstraram que a goma do faveiro é considerada praticamente atóxica. Esses resultados sugerem que a goma de Dimorphandra mollis apresenta potencial para ser utilizada em formulações de alimentos. Além disso, como é espécie já utilizada para a extração de rutina e também espécie de larga ocorrência em ecossistemas de grande importância tais como o Cerrado e a Caatinga, sua exploração poderia ser feita de forma ecologicamente sustentável.

O presente trabalho tem como objetivo determinar a influência das interações das gomas xantana e guar com o amido de milho de alto teor de amilose na textura de gel durante o armazenamento. Foram utilizadas amostras de amido milho Hylon VII® (71% de amilose, National Starch, goma guar (Higum 55I®, Rhodia) e goma xantana (Rhodigel 200®, Rhodia). Foram utilizadas diferentes concentrações das gomas guar e xantana, que variaram de 0 a 1%, de acordo com o delineamento experimental central composto rotacional. Em cada tratamento foram utilizadas 50g de amido com alto teor de amilose (Hylon VII), adicionadas das gomas. Estas amostras foram diluídas em água destilada e submetidas à agitação mecânica até completa dissolução. As soluções foram aquecidas até 95°C por 5 minutos para formação dos géis, os quais foram acondicionados em recipientes plásticos de 50mL e mantidos em temperatura de 5-10°C até 120h. Nos tempos T1 (24h), T2 (48h), T3 (72h), T4 (96h) e T5 (120h) de armazenamento foram feitas medidas da força máxima do gel de amilose em texturômetro (Stable Micro-System, Modelo TAX-T2). No período inicial, de 24 horas, a goma guar não apresentou influência positiva na redução da força do gel, sendo que a aplicação de goma xantana entre 0,7 e 1,0% apresentou os menores valores de força do gel de amilose. Após 120h de armazenamento, a força do gel de amilose diminuiu com a adição de 0,5-1,0% de goma xantana e 0-0,15% de goma guar.

O café é uma das bebidas mais consumidas no mundo e seus polissacarídeos têm sido investigados devido a sua grande importância comercial. Os principais polissacarídeos de café são os galactomananos e arabinogalactanos, considerados como fibras alimentares não digeridas no sistema digestivo humano. O presente trabalho teve por objetivo quantificar e determinar a composição de monossacarídeos e de polissacarídeos na bebida do café. As preparações foram realizadas em coador de papel tradicional, em cafeteira elétrica e em máquina expresso, utilizando café Arábica (Coffea arabica). Determinações quantitativas e qualitativas dos açúcares foram realizadas, respectivamente, por análises colorimétricas e cromatografia de troca aniônica (HPAEC). O rendimento de sólidos solúveis foi maior em café expresso, sendo os valores cerca de 3,6 vezes superiores àqueles de café de cafeteira elétrica com filtro de papel. Preparações de café expresso foram as que resultaram em maiores teores de açúcares totais não redutores por volume de bebida. Entretanto, os valores de açúcares totais foram significativamente maiores na preparação em cafeteira elétrica, quando expressos pelo total de sólidos solúveis extraídos, indicando que o tempo de extração é provavelmente mais eficiente do que a temperatura. A composição em monossacarídeos foi similar em todas as amostras e independente do modo de preparação do café. Todos apresentaram arabinose (14-15%), galactose (29-30%), glucose (6%) e manose 48-49%). Não se sabe se os polissacarídeos de café exercem as mesmas funções que a goma guar como fibra, mas, se isto ocorrer, esta é possivelmente uma das mais importantes fibras alimentares que ingerimos, levando em consideração a popularidade do café.

O efeito de diferentes combinações de gomas sobre os parâmetros de textura instrumental de queijo tipo petit-suisse probiótico foi avaliado. Queijos petit-suisse foram produzidos utilizando-se massa-base de queijo quark preparada com o starter Streptococcus thermophilus e os probióticos Lactobacillus acidophilus e Bifidobacterium longum. Três formulações de queijo petit-suisse foram preparadas, a partir de massa-base de queijo quark adicionada de 0,75% do produto final da mistura de gomas - xantana (X), carragena (C), guar (G), pectina (P): F1 = 2,5X:2,5C:5G; F2 = 2X:3C:5P; F3 = 5C:5G. Os parâmetros avaliados após 1, 7, 14 e 21 dias de armazenamento do produto a 4±1ºC incluíram a contagem de probióticos, a textura instrumental, o pH e a umidade. As populações de probióticos mantiveram-se acima de 6,40 log UFC/g para L. acidophilus e de 7,30 log UFC/g para B. longum. Houve diferença significativa entre as formulações (p<0,05) para todos os parâmetros de textura, exceto a firmeza de F1. O pH e a umidade apresentaram comportamento semelhante para as três formulações. Dentre as formulações estudadas, F1 foi considerada a melhor, por sua firmeza mais estável durante o período de armazenamento.

In this Master’s Thesis work the rheological properties of different polysaccharide gels have been studied. The results of this study are used as a starting point for further investigations of potential applications. In order to understand rheological behavior of studied materials, the commercial hydrocolloids such as sodium carboxymethyl cellulose, xanthan gum and guar gum were used as reference and comparison material for rheological studies. As a part the rheological research the development and implementation of proper measurement methods for studied materials were carried out. In the literature review, short introductions of studied materials and application areas of rheological modifiers are summarized. In addition, basic rheological concepts and key fundamentals are explained. In the experimental part the focus was on the rheological characterization of aqueous suspensions of studied materials. Especially, gel strength and solution stability were investigated. The rheological measurements included both rotational and oscillatory measurements in different conditions, where several chemical and physical properties were measured with Anton Paar MCR302 dynamic rotational rheometer. Studied polysaccharide gels can be clearly defined to be shear thinning and thixotropic materials. They have strong gel forming properties even at low concentrations, which explains the superior thickening behavior for some of the samples. Along with rheological characterization of selected materials the factors behind different phenomena were investigated. To reveal value and potential use of polysaccharide gels the influence of various factors such as concentration, temperature and ionic strength were determined. The measurements showed a clear difference between studied materials under investigated external parameters.

Presently different audio watermarking methods are available; most of them inclined towards copyright protection and copy protection. This is the key motive for the notion to develop a speaker verification scheme that guar- antees non-repudiation services and the thesis is its outcome. The research presented in this thesis scrutinizes the field of audio water- marking and the outcome is a speaker verification scheme that is proficient in addressing issues allied to non-repudiation to a great extent. This work aimed in developing novel audio watermarking schemes utilizing the fun- damental ideas of Fast-Fourier Transform (FFT) or Fast Walsh-Hadamard Transform (FWHT). The Mel-Frequency Cepstral Coefficients (MFCC) the best parametric representation of the acoustic signals along with few other key acoustic characteristics is employed in crafting of new schemes. The au- dio watermark created is entirely dependent to the acoustic features, hence named as FeatureMark and is crucial in this work. In any watermarking scheme, the quality of the extracted watermark de- pends exclusively on the pre-processing action and in this work framing and windowing techniques are involved. The theme non-repudiation provides immense significance in the audio watermarking schemes proposed in this work. Modification of the signal spectrum is achieved in a variety of ways by selecting appropriate FFT/FWHT coefficients and the watermarking schemes were evaluated for imperceptibility, robustness and capacity char- acteristics. The proposed schemes are unequivocally effective in terms of maintaining the sound quality, retrieving the embedded FeatureMark and in terms of the capacity to hold the mark bits. Robust nature of these marking schemes is achieved with the help of syn- chronization codes such as Barker Code with FFT based FeatureMarking scheme and Walsh Code with FWHT based FeatureMarking scheme. An- other important feature associated with this scheme is the employment of an encryption scheme towards the preparation of its FeatureMark that scrambles the signal features that helps to keep the signal features unreve- laed. A comparative study with the existing watermarking schemes and the ex- periments to evaluate imperceptibility, robustness and capacity tests guar- antee that the proposed schemes can be baselined as efficient audio water- marking schemes. The four new digital audio watermarking algorithms in terms of their performance are remarkable thereby opening more opportu- nities for further research.

The rheological, emulsification and certain physicochemical properties of purified exopolysaccharides (EPS) of Bifidobacterium longum subsp. infantis CCUG 52486 and Bifidobacterium infantis NCIMB 702205 were studied and compared with those of guar gum and xanthan gum. The two strains were grown in skim milk supplemented with 1.5% (w/v) casein hydrolysate at 37 ◦C for 24 h; they both produced heteropolysaccharides with different molecular mass and composition. The carbohydrate content of both polymers was more than 92% and no protein was detected. The EPS of B. longum subsp. infantis CCUG 52486 showed highly branched entangled porous structure under scanning electron microscopy. Higher intrinsic viscosity was observed for the EPS of B. longum subsp. infantis CCUG 52486 compared to the EPS of B. infantis NCIMB 702205 and guar gum. Both polymers showed pseudoplastic non-Newtonian fluid behaviour in an aqueous solution. The EPS of B. infantis NCIMB 702205 and B. longum subsp. infantis CCUG 52486 produced more stable emulsions with orange oil, sunflower seed oil, coconut oil and xylene compared to guar and xanthan gum. The EPS of B. longum subsp. infantis CCUG 52486 is the most promising one for applications in the food industry, as it had higher intrinsic viscosity, higher apparent viscosity in aqueous solution, porous dense entangled structure and good emulsification activity.

A utilização de inoculantes em culturas de leguminosas é uma prática conhecida e empregada há longo tempo e o substrato mais utilizado até o momento tem sido a turfa. Diversos tipos de formulações inoculantes que existem no mercado visam oferecer uma alternativa ao emprego da turfa, porém muitas apresentam baixa capacidade para manter a sobrevivência e eficiência dos rizóbios. Este trabalho avaliou a utilização de catorze misturas diferentes de polímeros naturais e/ou sintéticos como suportes para inoculantes, visando a produção de inoculantes comerciais para soja, e a capacidade de manter a sobrevivência e preservar as características de infectividade e de efetividade das estirpes SEMIA 587 de Bradyrhizobium elkanii e SEMIA 5079 de Bradyrhizobium japonicum. Avaliou-se a sobrevivência dos rizóbios nas formulações armazenadas, a capacidade de aderência das formulações em sementes, a sobrevivência dos rizóbios em sementes a 40 ºC e a eficiência dos inoculantes em plantas de soja, cultivadas em vasos com solo, submetidas a estresse hídrico e térmico. Observou-se que as misturas contendo goma xantana, jataí e guar, tanto nas formulações em gel como líquidas, podem ser usadas como veículo para inoculantes proporcionando maior proteção aos rizóbios contra as condições de dessecação e temperatura. Todos os inoculantes mantiveram a sobrevivência da população de rizóbios durante um ano de armazenamento. Os rizóbios das formulações G5, L1 e L7, que continham goma arábica, foram afetados pelo estresse hídrico e pelas temperaturas elevadas, reduzindo o número de nódulos formados. Dentre as formulações líquidas, as que continham xantana e glicerol (L2) e com adição de polivinilpirrolidona (L5) e a formulação que continha xantana, carboximetilcelulose e polivinilpirrolidona (L6), foram as mais promissoras para a formulação de inoculantes para soja.
