Temporal and spatial acoustic intensity (SATA) of therapeutic ultrasound (US) equipment should be monitored periodically. In order to evaluate the conditions of US equipment in use in the city of Piracicaba-Sao Paulo, Brazil, 31 machines - representing all Brazilian manufacturers - were analysed under continuous and pulsed conditions at a frequency of 1 MHz. Data about temporal and spatial acoustic intensity were collected and the use of equipment was surveyed. Intensities of 0.1, 0.2, 0.5, 0.8, 1.0, 1.5, 2.0, 2.5 and 3.0 Wcm -2, indicated on the equipment panel were analysed using a previously calibrated digital radiation pressure scale, model UPM-DT-1 (Ohmic Instruments Co). The acoustic intensity (I) results were expressed as superior and inferior quartile ranges for transducers with metal surfaces of 9 cm 2 and an effective radiation area (ERA) Of 4 cm 2. The results under continuous conditions were: I 0.1 = -20.0% and -96%. I 0.2 = -3.1% and -83.7%. I 0.5 = -35.0% and -86.5%. I 0.8 = -37.5% and -71.0%. I 2.5 = -49.0% and -69.5%. I 3.0 = -58.1% and -77.6%. For pulsed conditions, intensities were: I 0.1 = -40.0% and -86.2%. I 1.0 = -50.0% and -86.5%. I 1.5 = -62.5% and -82.5%. I 2.0 = -62.5% and -81.6%. I 2.5 = -64.7% and -88.8%. I 3.0 = -87.1% and -94.8%. In reply to the questionnaire drawn up to check the conditions of use of equipment, all users reported the use of hydrosoluble gel as a coupling medium and none had carried out previous calibrations. Most users used intensities in the range of 0.4. to 1.0 Wcm -2 and used machines for 300 to 400 minutes per week. The majority of machines had been bought during the previous seven years and weekly use ranged from less than 100 minutes to 700 minutes (11 hours 40 minutes). Findings confirm previous observations of discrepancy between the intensity indicated on the equipment panel and that emitted by the transducer and highlight the necessity for periodic evaluations of US equipment.

Grundlage für die hier gezeigte Arbeit stellt die Eigenschaft von amphiphilen Blockcopolymeren dar immer den Block mit der niedrigsten Grenzflächenenergie zum angrenzenden Medium an die Oberfläche zu bringen. Durch einen Austausch des Mediums an der Grenzfläche zum Blockcopolymer kann eine Reorientierung erzwungen werden, wenn die Grenzflächenenergie des anderen Blocks nun die niedrigere Grenzflächenenergie besitzt. Dieses Verhalten von dünnen amphiphilen Blockcopolymerfilmen wurde zur Strukturierung von Oberflächen ausgenutzt und in nachfolgenden Synthesen weiter verstärkt. Um dies zu erreichen wurde das zur Strukturierung erforderliche Poly(4-Octylstyrol)block(4-hydroxystyrol) durch kontrollierte radikalische Polymerisationsmethode mit dem Tempo Unimer (2,2,6,6-Tetramethyl-1-1(1-phenyl-ethoxy)-piperidin) synthetisiert. Für die geplanten Reorientierungen und Modifizierungen von Oberflächen wurden dünne Filme durch Schleuderbeschichtung auf verschiedenen Substraten (Siliziumwafern, Glassubstraten und Goldoberflächen) hergestellt. Das Verhalten der Oberflächen von diesen Filmen wurde durch Kontaktwinkelmessungen untersucht. Auf diese Weise konnte gezeigt werden, dass die Oberfläche von Polymerfilmen nach der Präparation aus dem hydrophoben Block des Polymers gebildet wird. Durch Kontakt des Polymerfilms mit Wasser kann dieser zur Reorientierung gebracht werden, so dass der hydrophile Block des Polymers an der Oberfläche erscheint. Dieses Verhalten wurde zur Strukturierung mit softlithographischen Techniken genutzt. Dazu wurden hydrophil/hydrophob strukturierte Oberflächen durch Aufsetzen von hydrophoben PDMS-Stempeln, die Teile der Oberfläche selektiv abdeckten, und Einbringen von Wasser in die dabei entstehenden Kapillaren hergestellt. Dies ermöglichte es die Oberfläche selektiv im Größenbereich von 500nm bis zu 50µm zu strukturieren und an den reaktiven Bereichen Materialien, wie z.B. Kupfer, Titandioxid, Polyelektrolyte, photonische Kristalle und angegraftete Polymere, mit verschiedenen Methoden selektiv auf die Oberfläche aufzubringen. Um den Reorganisationsprozess der Oberfläche genauer zu studieren, wurde ein für diese Aufgabe besser geeignetes Polymer (Poly(Styrol)-block-poly(essigsäure-2-(2-(4-vinyl-phenoxy)-ethoxy)ethylester)) synthetisiert. Aus diesem Blockcopolymer wurden wieder dünne Filme durch Spincoaten hergestellt. Die Reorientierung dieses Polymers in 70°C warmen Wasser konnte durch Kontaktwinkelmessungen und NEXAFS Spektroskopie nachgewiesen werden. Mit Hilfe der NEXAFS Spektroskopie konnte festgestellt werden, dass die Geschwindigkeit der Reorientierung durch eine exponentielle Funktion beschrieben werden kann. Eine Auswertung der Geschwindigkeitskonstante für die Reorientierung einer hydrophilen zu einer hydrophoben Oberfläche des Polymers bei 60°C führt zu =75min. Aufgrund des exponentiellen Charakters der Reorientierung macht es den Anschein, dass die Reorientierung bei verschiedenen Reorientierungstemperaturen bis zu einem gewissen Grad erfolgt und dann stoppt. Eine weitere Reorientierung scheint erst wieder bei einer Temperaturerhöhung zu beginnen. Aus AFM Messungen ist ein Beginnen der Reorientierung durch Bildung kleiner Löcher in der Polymeroberfläche zu erkennen, die sich zu runden Erhöhungen und Vertiefungen vergrößern, um letztendlich in ein spinodales Entmischungsmuster über zu gehen. Dieses heilt dann im Laufe der Zeit langsam durch Verschwinden der hydrophilen Bereiche langsam aus. Der Beginn des zuvor beschriebenen Reorientierungsprozesses einer hydrophilen Oberfläche in eine hydrophobe konnte sowohl in den AFM, als auch in den NEXAFS-Messungen zu ca. 50°C bestimmt werden.

Biologische Membranen sind Fettmolekül-Doppelschichten, die sich wie zweidimensionale Flüssigkeiten verhalten. Die Energie einer solchen fluiden Oberfläche kann häufig mit Hilfe eines Hamiltonians beschrieben werden, der invariant unter Reparametrisierungen der Oberfläche ist und nur von ihrer Geometrie abhängt. Beiträge innerer Freiheitsgrade und der Umgebung können in den Formalismus mit einbezogen werden. Dieser Ansatz wird in der vorliegenden Arbeit dazu verwendet, die Mechanik fluider Membranen und ähnlicher Oberflächen zu untersuchen. Spannungen und Drehmomente in der Oberfläche lassen sich durch kovariante Tensoren ausdrücken. Diese können dann z. B. dazu verwendet werden, die Gleichgewichtsposition der Kontaktlinie zu bestimmen, an der sich zwei aneinander haftende Oberflächen voneinander trennen. Mit Ausnahme von Kapillarphänomenen ist die Oberflächenenergie nicht nur abhängig von Translationen der Kontaktlinie, sondern auch von Änderungen in der Steigung oder sogar Krümmung. Die sich ergebenden Randbedingungen entsprechen den Gleichgewichtsbedingungen an Kräfte und Drehmomente, falls sich die Kontaktlinie frei bewegen kann. Wenn eine der Oberflächen starr ist, muss die Variation lokal dieser Fläche folgen. Spannungen und Drehmomente tragen dann zu einer einzigen Gleichgewichtsbedingung bei; ihre Beiträge können nicht mehr einzeln identifiziert werden. Um quantitative Aussagen über das Verhalten einer fluiden Oberfläche zu machen, müssen ihre elastischen Eigenschaften bekannt sein. Der "Nanotrommel"-Versuchsaufbau ermöglicht es, Membraneigenschaften lokal zu untersuchen: Er besteht aus einer porenüberspannenden Membran, die während des Experiments durch die Spitze eines Rasterkraftmikroskops in die Pore gedrückt wird. Der lineare Verlauf der resultierenden Kraft-Abstands-Kurven kann mit Hilfe der in dieser Arbeit entwickelten Theorie reproduziert werden, wenn der Einfluss von Adhäsion zwischen Spitze und Membran vernachlässigt wird. Bezieht man diesen Effekt in die Rechnungen mit ein, ändert sich das Resultat erheblich: Kraft-Abstands-Kurven sind nicht länger linear, Hysterese und nichtverschwindende Trennkräfte treten auf. Die Voraussagen der Rechnungen könnten in zukünftigen Experimenten dazu verwendet werden, Parameter wie die Biegesteifigkeit der Membran mit einer Auflösung im Nanometerbereich zu bestimmen. Wenn die Materialeigenschaften bekannt sind, können Probleme der Membranmechanik genauer betrachtet werden. Oberflächenvermittelte Wechselwirkungen sind in diesem Zusammenhang ein interessantes Beispiel. Mit Hilfe des oben erwähnten Spannungstensors können analytische Ausdrücke für die krümmungsvermittelte Kraft zwischen zwei Teilchen, die z. B. Proteine repräsentieren, hergeleitet werden. Zusätzlich wird das Gleichgewicht der Kräfte und Drehmomente genutzt, um mehrere Bedingungen an die Geometrie der Membran abzuleiten. Für den Fall zweier unendlich langer Zylinder auf der Membran werden diese Bedingungen zusammen mit Profilberechnungen kombiniert, um quantitative Aussagen über die Wechselwirkung zu treffen. Theorie und Experiment stoßen an ihre Grenzen, wenn es darum geht, die Relevanz von krümmungsvermittelten Wechselwirkungen in der biologischen Zelle korrekt zu beurteilen. In einem solchen Fall bieten Computersimulationen einen alternativen Ansatz: Die hier präsentierten Simulationen sagen voraus, dass Proteine zusammenfinden und Membranbläschen (Vesikel) bilden können, sobald jedes der Proteine eine Mindestkrümmung in der Membran induziert. Der Radius der Vesikel hängt dabei stark von der lokal aufgeprägten Krümmung ab. Das Resultat der Simulationen wird in dieser Arbeit durch ein approximatives theoretisches Modell qualitativ bestätigt.

The aim of my thesis is to parallelize the Weighting Histogram Analysis Method (WHAM), which is a popular algorithm used to calculate the Free Energy of a molucular system in Molecular Dynamics simulations. WHAM works in post processing in cooperation with another algorithm called Umbrella Sampling. Umbrella Sampling has the purpose to add a biasing in the potential energy of the system in order to force the system to sample a specific region in the configurational space. Several N independent simulations are performed in order to sample all the region of interest. Subsequently, the WHAM algorithm is used to estimate the original system energy starting from the N atomic trajectories. The parallelization of WHAM has been performed through CUDA, a language that allows to work in GPUs of NVIDIA graphic cards, which have a parallel achitecture. The parallel implementation may sensibly speed up the WHAM execution compared to previous serial CPU imlementations. However, the WHAM CPU code presents some temporal criticalities to very high numbers of interactions. The algorithm has been written in C++ and executed in UNIX systems provided with NVIDIA graphic cards. The results were satisfying obtaining an increase of performances when the model was executed on graphics cards with compute capability greater. Nonetheless, the GPUs used to test the algorithm is quite old and not designated for scientific calculations. It is likely that a further performance increase will be obtained if the algorithm would be executed in clusters of GPU at high level of computational efficiency. The thesis is organized in the following way: I will first describe the mathematical formulation of Umbrella Sampling and WHAM algorithm with their apllications in the study of ionic channels and in Molecular Docking (Chapter 1); then, I will present the CUDA architectures used to implement the model (Chapter 2); and finally, the results obtained on model systems will be presented (Chapter 3).

Byrd Glacier has one of the largest ice catchment areas in Antarctica, delivers more ice to the Ross Ice Shelf than any other ice stream, and is the fastest of these ice streams. A force balance, combined with a mass balance, demonstrates that stream flow in Byrd Glacier is transitional from sheet flow in East Antarctica to shelf flow in the Ross Ice Shelf. The longitudinal pulling stress, calculated along an ice flowband from the force balance, is linked to variations of ice thickness, to the ratio of the basal water pressure to the ice overburden pressure where Byrd Glacier is grounded, and is reduced by an ice-shelf buttressing stress where Byrd Glacier is floating. Longitudinal tension peaks at pressure-ratio maxima in grounded ice and close to minima in the ratio of the pulling stress to the buttressing stress in floating ice. The longitudinal spacing of these tension peaks is rather uniform and, for grounded ice, the peaks occur at maxima in surface slope that have no clear relation to the bed slope. This implies that the maxima in surface slope constitute a "wave train" that is related to regular variations in ice-bed coupling, not primarily to bed topography. It is unclear whether these surface "waves" are "standing waves" or are migrating either upslope or downslope, possibly causing the grounding line to either retreat or advance. Deciding which is the case will require obtaining bed topography in the map plane, a new map of surface topography, and more sophisticated modeling that includes ice flow linked to subglacial hydrology in the map plane.

When a gas is introduced at high velocity through a nozzle into a packed bed, it creates a raceway in the packed bed. It has been found that the raceway size is larger when it is formed by decreasing the gas velocity from its highest value than when it is formed by increasing the gas velocity. This phenomenon is known as raceway hysteresis. A hypothesis has been oroposed to explain the hysteresis phenomenon based on a force-balance approach which includes frictional, bed-weight, and pressure forces. According to this hypothesis, the frictional force acts in different directions when the raceway is expanding and contracting. In this article, the entire packed bed has been divided into radial and Cartesian co-ordinate systems, and the forces acting on the raceway have been solved analytically for a simplified one-dimensional case. Based on the force-balance approach, a general equation has been obtained to predict the diameter of the raceway for increasing And decreasing velocities. A reasonable agreement has been found between the theoretical predictions and experimental observations. The model has also been compared with published experimental and plant data. The hysteresis mechanism in the packed beds can be described reasonably by taking into consideration the direction of frictional forces for the increasing- and decreasin-velocity cases. The effects of the particleshape factor and void fraction on the raceway hysteresis are examined.

Spouted beds have been used in industry for operations such as drying, catalytic reactions, and granulation. Conventional cylindrical spouted beds suffer from the disadvantage of scaleup. Two-dimensional beds have been proposed by other authors as a solution for this problem. Minimum spouting velocity has been studied for such two-dimensional beds. A force balance model has been developed to predict the minimum spouting velocity and the maximum pressure drop. Effect of porosity on minimum spouting velocity and maximum pressure drop has been studied using the model. The predictions are in good agreement with the experiments as well as with the experimental results of other investigators.

Lift, pitching moment, and thrust/drag on a supersonic combustion ramjet were measured in the T4 free-piston shock tunnel using a three-component stress-wave force balance. The scramjet model was 0.567 m long and weighed approximately 6 kg. Combustion occurred at a nozzle-supply enthalpy of 3.3 MJ/kg and nozzle-supply pressure of 32 MPa at Mach 6.6 for equivalence ratios up to 1.4. The force coefficients varied approximately linearly with equivalence ratio. The location of the center of pressure changed by 10% of the chord of the model over the range of equivalence ratios tested. Lift and pitching-moment coefficients remained constant when the nozzle-supply enthalpy was increased to 4.9 MJ/kg at an equivalence ratio of 0.8, but the thrust coefficient decreased rapidly. When the nozzle-supply pressure was reduced at a nozzle-supply enthalpy of 3.3 MJ/kg and an equivalence ratio of 0.8, the combustion-generated increment of lift and thrust was maintained at 26 MPa, but disappeared at 16 MPa. Measured lift and thrust forces agreed well with calculations made using a simplified force prediction model, but the measured pitching moment substantially exceeded predictions. Choking occurred at nozzle-supply enthalpies of less than 3.0 MJ/kg with an equivalence ratio of 0.8. The tests failed to yield a positive thrust because of the skin-friction drag that accounted for up to 50% of the fuel-off drag.

The coupling of sandy beach aquifers with the swash zone in the vicinity of the water table exit point is investigated through simultaneous measurements of the instantaneous shoreline (swash front) location, pore pressures and the water table exit point. The field observations reveal new insights into swash-aquifer coupling not previously gleaned from measurements of pore pressure only. In particular, for the case where the exit point is seaward of the observation point, the pore pressure response is correlated with the distance between the exit point and the shoreline in that when the distance is large the rate of pressure drop is fast and when the distance is small the rate decreases. The observations expose limitations in a simple model describing exit point dynamics which is based only on the force balance on a particle of water at the sand surface and neglects subsurface pressures. A new modified form of the model is shown to significantly improve the model-data comparison through a parameterization of the effects of capillarity into the aquifer storage coefficient. The model enables sufficiently accurate predictions of the exit point to determine when the swash uprush propagates over a saturated or a partially saturated sand surface, potentially an important factor in the morphological evolution of the beach face. Observations of the shoreward propagation of the swash-induced pore pressure waves ahead of the runup limit shows that the magnitude of the pressure fluctuation decays exponentially and that there is a linear increase in time lags, behavior similar to that of tidally induced water table waves. The location of the exit point and the intermittency of wave runup events is also shown to be significant in terms of the shore-normal energy distribution. Seaward of the mean exit point location, peak energies are small because of the saturated sand surface within the seepage face acting as a "rigid lid'' and limiting pressure fluctuations. Landward of the mean exit point the peak energies grow before decreasing landward of the maximum shoreline position.

The drag on a nacelle model was investigated experimentally and computationally to provide guidance and insight into the capabilities of RANS-based CFD. The research goal was to determine whether industry constrained CFD could participate in the aerodynamic design of nacelle bodies. Grid refinement level, turbulence model and near wall treatment settings, to predict drag to the highest accuracy, were key deliverables. Cold flow low-speed wind tunnel experiments were conducted at a Reynolds number of 6∙〖10〗^5, 293 K and a Mach number of 0.1. Total drag force was measured by a six-component force balance. Detailed wake analysis, using a seven-hole pressure probe traverse, allowed for drag decomposition via the far-field method. Drag decomposition was performed through a range of angles of attack between 0o and 45o. Both methods agreed on total drag within their respective uncertainties. Reversed flow at the measurement plane and saturation of the load cell caused discrepancies at high angles of attack. A parallel CFD study was conducted using commercial software, ICEM 15.0 and FLUENT 15.0. Simulating a similar nacelle geometry operating under inlet boundary conditions obtained through wind tunnel characterization allowed for direct comparisons with experiment. It was determined that the Realizable k-ϵ was best suited for drag prediction of this geometry. This model predicted the axial momentum loss and secondary flow in the wake, as well as the integrated surface forces, within experimental error up to 20o angle of attack. SST k-ω required additional surface grid resolution on the nacelle suction side, resulting in 15% more elements, due to separation point prediction sensitivity. It was further recommended to apply enhanced wall treatment to more accurately capture the viscous drag and separated flow structures. Overall, total drag was predicted within 5% at 0o angle of attack and 10% at 20o, each within experimental uncertainty. What is more, the form and induced drag predicted by CFD and measured by the wake traverse shared good agreement. Which indicated CFD captured the key flow features accurately despite simplification of the nacelle interior geometry.

A new overground body-weight support system called ZeroG has been developed that allows patients with severe gait impairments to practice gait and balance activities in a safe, controlled manner. The unloading system is capable of providing up to 300 lb of static support and 150 lb of dynamic (or constant force) support using a custom-series elastic actuator. The unloading system is mounted to a driven trolley, which rides along an overhead rail. We evaluated the performance of ZeroG's unloading system, as well as the trolley tracking system, using benchtop and human-subject testing. Average root-mean-square and peak errors in unloading were 2.2 and 7.2 percent, respectively, over the range of forces tested while trolley tracking errors were less than 3 degrees, indicating the system was able to maintain its position above the subject. We believe training with ZeroG will allow patients to practice activities that are critical to achieving functional independence at home and in the community.
