Background: Imbalance in bacterial species composition of the gut microbiota is one of the factors associated with the cause or complication of the symptoms of Crohn's disease (CD). This disequilibrium consists in the reduction of biodiversity, decrease of genus such as Bifidobacterium and elevation of species such as Escherichia coli. Human microbiota varies among subjects of a same population irrespective of their health condition and among individuals living in distinct geographic locations. In animal models, sex related differences could also be observed in gut bacterial species composition under some pathological conditions. Experiments conducted with mice have demonstrated that the manifestation of type 1 diabetes (T1D) could be under the influence of the animal sex and its serum level of testosterone, which in turn could be modulated by a particular gut microbiota. Considering the existence of similar features between T1D and CD, such as strong genetic component and malfunctioning of the immune system, we investigated whether differences could be observed in the gut microbiota dysbiosis of male and female CD patients. Methods: Fifty and 5 gut mucosal biopsies from 25 adult CD patients (11 males and 14 females) and 43 specimens of an equivalent clinical material from 22 control subjects (11 males and 11 females) were screened for bacterial biodiversity by analyzing sequences of 16SrDNA V6 region. A number of 2-3 samples each from distinct gut segments (from ileum to rectum) were taken from each subject. The 16SrDNA sequences were obtained by sequencing PCR amplicons of the corresponding gene in the Ion torrent PGM sequencer. Identification and classification of the bacterial groups followed the Ribosomal Database Project (RDP) website pipeline. The relationships of the bacterial taxa with each of the study parameters was performed by compiling the data in a MS Excel and the level of statistical significance determined by the Chi-square test. Results: A total of 3203 16SrDNA sequences were detected in the 98 biopsies samples, the majority of which matching Proteobacteria, Firmicutes, Bacterioidetes, and Actinobacteria. The percentage of DNA sequences for each of these phyla found in Male control subjects/Male CD patients was 40.5/33, 32.7/32.4, 20.8/24.5, and 4.4/4,4 for Proteobacteria, Firmicutes, Bacterioidetes, and Actinobacteria, respectively. In Female comparisons, these values were 35.6/42, 39.2/26.3, 19.8/23.3, 5.2/7. Both Male and Female CD patients presented higher numbers of sequences of Actinobacteria and Bacterioidetes than those of control subjects of the same gender. Case-control differences for Firmicutes could be observed only in female comparisons and, for Proteobacteria, although case-control differences were observed in both genders, the nature of difference was distinct, since while in CD female patients a higher number of sequences matching this phylum was detected, in males a reduced number was observed, in comparison with controls. The species responsible for the Proteobacteria variation in both gender was Escherichia coli. Conclusions: The data presented above suggest that any analysis of dysbiosis in CD must take in account the patient's gender, an observation particularly relevant for Escherichia coli, whose association with CD has been most intensively investigated and for which the present study shows a reverse quantitative variation regarding the patients' gender.

Background: The intestinal microbiome (IM) has extensively been studied in the search for a link of bacteria with the cause of Crohn`s disease (CD). The association might result from the action of a specific pathogen and/or an eventual imbalance in bacterial species composition of the gut. The innumerous virulence associated markers and strategies described for adherent and invasive Escherichia coli (AIEC) have made them putative candidate pathogens for CD. IM of CD patients shows dysbiosis, manifested by the proliferation of bacterial groups such as Enterobacteriaceae and reduction of others such as Lactobacillus and Bifidobacterium. The augmented bacterial population comprising of commensal and/or pathogenic organisms super stimulates the immune system, triggering the inflammatory reactions responsible for the clinical manifestations of the disease. Considering the role played by IM in CD and the multiple variables influencing its species composition, resulting in differences among populations, the objective of this study was to determine the bacterial biodiversity in the mucosa associated microbiome of CD patients from a population not previously subject to this analysis, living in the middle west region of Sao Paulo state. Methods: A total of 4 CD patients and 5 controls subjects attending the Botucatu Medical School of the Sao Paulo State University (UNESP) for routine colonoscopy and who signed an informed consent were included in the study. A number of 2 biopsies, one from the ileum and other from any part of the terminal colon, were taken from each subject and immediately frozen at -70[degrees]C until DNA purification. The bacterial biodiversity was assessed by next generation (ion torrent) sequencing of PCR amplicons of the ribosomal DNA 16S V6 region (16S V6 rDNA). The bacterial identification was performed at the genus level, by alignment of the generated DNA sequences with those available at the ribosomal database project (RDP) website. Results: The overall DNA sequence output was based on an average number of 526,427 reads per run, matching 50 bacterial genus 16SrDNA sequences available at the RDB website, and 22 non matching sequences. Over 95% of the sequences corresponded to taxa belonging to the major phyla: Firmicutes, Bacterioidetes, Proteobacteria and Actinobacteria. Irrespective of the intestinal site analyzed, no case-control differences could be observed in the prevalence of Actinobacteria and Firmicutes. The prevalence of Proteobacteria was higher (40%) in the biopsies of control subjects as compared to that of DC patients (16%). For Bacterioidetes, the higher prevalence was observed among DC patients (33% as opposed to 14,5% in controls). The significance for all comparisons considered a p value < 0,05 in a Chi2 test. No mucosal site specific differences could be observed in IM comparisons of CD and control subjects. Conclusions: The rise in the number of Bacterioidetes observed here among CD patients seems to be in agreement with most of studies published thus far. Yet, the reduction in the number of Proteobacteria along with an apparently unaltered population of Actinobacteria and Firmicutes, which include the so called "beneficial" organisms Bifidobacterium and Lactobacillus were rather surprising. These data suggest that the analyses on the role of IM in CD should consider the multiple variables that may influence its species composition.

Background: The number of Escherichia coli in the gut of Crohn's disease (CD) patients is higher than that of normal subjects, but the virulence potential of these bacteria is not fully known. Previous studies have shown that these E. coli are closely related to extraintestinal pathogenic categories (ExPEC), are able to invade epithelial cells, and usually do not produce exotoxins. We report here the detection, in a CD patient, of an E. coli which belongs to a classical enteropathogenic (EPEC) serotype and displays virulence markers of enteroinvasive (EIEC), enteroaggregative (EAEC) and enterohemorrhagic (EHEC) pathotypes. Methods: The E. coli strain was isolated, in 2009, by classical bacteriological procedures from a 56 year old woman who underwent ileo-terminal resection 1 year before, due to intestinal obstruction. The bacterial characterization was carried out by in vitro adhesion and invasion assays to cultured epithelial cells and macrophages and screening by PCR to identify virulence genetic markers of diarrheogenic E. coli (DEC) and to detect one of the gene combinations which define the phylogroups of the E. coli reference (EcoR) collection. The strain was also tested for the ability to produce biofilm and shiga cytotoxins and had its whole genome sequenced by Ion Torrent Sequencing Technology. Results: The studied strain, which was detected both in ileum biopsies and the stools of the patient, displayed the aggregative adherence (AA) phenotype to Hep-2 cells and an ability to enter Caco-2 cells 3x as high as that of EIEC reference strain and 89% of that of the prototype AIEC LF82 strain. Although it could invade cultured macrophages, the strain was unable to replicate inside these cells. PCR screening revealed the presence of eae, aggR and stx1. Tests with bacterial culture supernatants in Vero cells demonstrating cytotoxicity suggested the production of Stx1. In addition, the strain revealed to be a strong biofilm producer, belonged to the B2 EcoR phylogroup, to the O126:H27 serogroup and to the multilocus sequencing type (MLST) ST3057. The 2 later features were deduced from the whole genome sequence of the strain. Conclusions: The characterization of this E. coli isolate from a CD patient revealed a combination of virulence markers of distinct DEC pathotypes, namely eae and stx1 of EHEC, AA, aggR and biofilm formation of EAEC, and invasiveness of EIEC. These features along with its serotype and phylogroup identity seem to suggest a potential to be involved in CD, an observation which should be tested with additional studies.

Under conservation tillage systems is critical to maintaining plant residues from previous crops on the soil surface. A technique called Hormesis aims to increase the amount of straw and also cause a delay in straw decomposition. Therefore, this study aimed to evaluate the agronomic characteristics of corn under no-tillage system subjected to Hormesis. The experiment was carried out at UNESP campus in the city of Botucatu, SP . The experimental was design in completely randomized blocks. The treatments based on the technique of Hormesis were : control (no application) , sub Gliphosate low dosage (12.5 g.ai.ha - 1 ) , sub Gliphosate average dosage (25 g. ai.ha - 1 ) , sub Gliphosate high dose (50 g. ai.ha -1 ), 2,4-D under low dose (100 g. ai.ha -1 ), 2,4-D sub average dosage (200 g. ai.ha -1 ) 2,4- D under high dosage (300 g. ai.ha -1 ) sub Verdict low dose (0.625 g. ai.ha -1 ) sub Verdict average dosage (1.25 g. ai.ha -1 ) , sub Verdict high dosage (2.5 g. ai.ha - 1 ) . In addition, the following characteristics were evaluated: plant height, first ear growth, stem diameter , ear length , number of rows per cob, cob diameter , , percentage of grains on the cob , mass of plant dry matter, and yield. The results showed that all the treatments showed no statistical difference, the maize was not affected with sub doses of herbicides applications under the field and weather conditions in which the experiment was conducted.

Soil bulk density is an attribute often used to characterize soil physical structure, being an indicator of soil compaction. The objective of this study was to compare the values ​​of bulk density measured by the paraffin sealed clod and volumetric ring methods in conventional, no-tillage and minimum tillage systems on a Dystroferric Red Nitosol, clayey, in Botucatu, SP. The experiment design was a 3x2 factorial arrangement in randomized block with four replications. The density values ​​obtained by the paraffin sealed clod method were statistically higher than those obtained by the volumetric ring method. There was no difference between the managements when comparing the values ​​of soil density obtained by the paraffin sealed clod method. The soil under conventional management showed lower density when comparing the values ​​of soil density obtained by the volumetric ring. The volumetric ring method was more sensitive to show differences between the management systems than the paraffin sealed clod method.

This study aimed to evaluate the behavior of the deposition and mechanical damage in seeds using a continuous flow metering system under different slope and speed. Part of the study was conducted at Agricultural Research Foundation Agricultural - (FAPA), where seeds that are deposited by a metering system were collected, and the quality analysis verifying the percentage of mechanical damage were conducted at the Faculty of Agricultural Sciences, UNESP, city of Botucatu– SP. The mechanism deposition was subjected to three different speed conditions (4,7, and 10 km.h-1) and three differents working slopes, ( 3%, 8%, and 16%). The results were submitted to Tukey test (p ≤ 0.05), and an analysis of variance with F test at 5% significance level was performed. The results showed an interaction between the factor slope and speed of work, increasing the metering mechanism speed, results in a reduction of the seed deposition at a 3% slope but a working speed of 10 km h-1did not reduce the rate of seed deposition until the slope reaches 16%. Both the slope factor and the working speed caused at least 3.9 and 4.2% more damage to the seeds, respectively.

As emas são aves ratitas nativas do continente sul americano, são consideradas aves primitivas do ponto de vista filogenético que constituem um grupo altamente especializado. Este estudo buscou caracterizar macro e microscopicamente o fígado e pâncreas de emas. O material foi coletado no Centro de Multiplicação de Animais Silvestres (CEMAS), na cidade de Mossoró-RN, Brasil, (Registro IBAMA n° 14.78912). Utilizaram-se 20 animais jovens com idade entre dois e seis meses independente do sexo. Em emas, o fígado se relacionava cranialmente com o ápice do coração, dorsalmente com os pulmões, esôfago e o proventrículo gástrico, caudalmente, com o ventrículo gástrico, o baço, o duodeno e parte do jejuno. Apresentava coloração vermelha escura e possuía apenas dois lobos, sendo o direito ligeiramente menor que o esquerdo. Histologicamente, era revestido por uma cápsula de tecido conjuntivo delgada e cada lóbulo hepático pôde ser identificado pela presença evidente de veias centrais, com muitos sinusoides comunicando-se com elas. O pâncreas, ventralmente, apresentava-se como uma fita fina, formado por um lobo dorsal e um lobo ventral. Longitudinalmente o pâncreas em emas localiza-se no mesentério dorsal desde o fígado até a flexura cranial do duodeno, mantendo-se preso às alças duodenais por ligamentos. Histologicamente, era composto por uma cápsula delgada de tecido conjuntivo denso, com discretos lóbulos separados por tecido conjuntivo capsular, compostos por estruturas tubuloalveolares e ductos. O fígado e pâncreas de emas apresentam padrão morfológico similar ao descrito para aves domésticas.

Torque teno sus virus (TTSuV) is emergent in swine herds. Recent studies have shown an increased frequency of TTSuV2 in Porcine circovirus type 2 (PCV2)-associated diseases (PCVAD), which are endemic in many swine-producing countries, including Brazil. Coinfection with several other viral and bacterial agents results in an increased incidence of more severe PCVAD. Given the limited information on TTSuV and PCV2 coinfection, especially in Brazilian swine herds, this study made a preliminary estimation of the occurrence of coinfection in swine herds by testing samples from different categories. Between 2008 and 2009, 111 samples of feces and 23 serum samples from 5 swine herds were tested for PCV2 and TTSuVs and the results analyzed for associations between these agents. No significant differences in coinfection frequency were observed for PCV2 1 + TTSuV1 or for PCV2 1 TTSuV2 between nursery piglets (P = 0.730), growing pigs (P = 0.331), or sows (P = 0.472). However, a significant difference was observed for PCV2 1 TTSuV1 1 TTSuV2 between nursery piglets and growing pigs (P = 0.004; Fisher's exact test). Phylogenetic studies agreed with the grouping of TTSuV1 and TTSuV2 into 2 different clades, with no distinct pattern of clustering of these isolates with the animal categories.

Fluoxetine (FLX) is commonly used to treat anxiety and depressive disorders in pregnant women. Since FLX crosses the placenta and is excreted in milk, maternal treatment with this antidepressant may expose the fetus and neonate to increased levels of serotonin (5-HT). Long-term behavioral abnormalities have been reported in rodents exposed to higher levels of 5-HT during neurodevelopment. In this study we evaluated if maternal exposure to FLX during pregnancy and lactation would result in behavioral and/or stress response disruption in adolescent and adult rats. Our results indicate that exposure to FLX influenced restraint stress-induced Fos expression in the amygdala in a gender and age-specific manner. In male animals, a decreased expression was observed in the basolateral amygdala at adolescence and adulthood; whereas at adulthood, a decrease was also observed in the medial amygdala. A lack of FLX exposure effect was observed in females and also in the paraventricular nucleus of both genders. Regarding the behavioral evaluation, FLX exposure did not induce anhedonia in the sucrose preference test but decreased the latency to feed of both male and female adolescent rats evaluated in the novelty-suppressed feeding test. In conclusion, FLX exposure during pregnancy and lactation decreases acute amygdalar stress response to a psychological stressor in males (adolescents and adults) as well as influences the behavior of adolescents (males and females) in a model that evaluates anxiety and/or depressive-like behavior. Even though FLX seems to be a developmental neurotoxicant, the translation of these findings to human safe assessment remains to be determined since it is recognized that not treating a pregnant or lactating woman may also impact negatively the development of the descendants.

Com objectivo de melhorar a qualidade da carne de origem caprina e ovina proveniente de animais adultos de raças Serrana e Churra Galega Bragançana (CGB), de forma a atribuir-lhe um valor comercial acrescentado, foram elaborados diferentes tipos de patês com incorporação de diferentes níveis de gordura de porco. O estudo foi feito no laboratório de Tecnologias de Qualidade da Carcaça e da Carne da Escola Superior Agrária, do Instituto Politécnico de Bragança. Os patês foram submetidos a uma avaliação da qualidade físico-química no período de um ano como forma de estudar a vida útil dos mesmos, quando conservados à temperatura de 2 a 4º C. As determinações físico-químicas foram repetidas quatro vezes em cada período de estudo, nomeadamente 1, 6 e 12 meses de conservação. Diferenças entre as médias foram consideradas estatisticamente significantes, no nível de 5%, quando p ≤ 0,05. O tratamento estatístico dos dados foi realizado utilizando o programa Statistix 10. A transformação de carne de animais adultos em subprodutos originou patês de alto valor nutricional, com vida útil superior a um ano. Com a avaliação do pH em patês de ambas espécies, foi possível observar maior queda de pH em patês de origem caprina, podendo-se afirmar que os mesmos têm baixa estabilidade quando comparados com os de origem ovina. A incorporação de gordura de porco, proveniente de abdómen, influenciou positivamente na qualidade físico-química do produto final. Patês com incorporação de gordura de porco apresentaram maior teor de ácidos gordos saturados e poli-insaturados. Em relação às duas espécies, as amostras de patês de origem ovina sem incorporação de gordura de porco apresentaram maior teor de gordura saturada e menor teor de gordura mono-insaturada.

A presente dissertação destina-se a analisar o processo de emergência da identidade sul-americana, cuja primeira manifestação remonta à década de 1990, acentuando-se após a I Reunião de Presidentes da América do Sul (2000). A imersão histórica de longa duração do primeiro capítulo, que nos remete à segunda década do século XIX, oferece-nos um instrumental analítico complementar ao arcabouço teórico utilizado, de modo a apresentar a identidade latino-americana principal concorrente da sul-americana como elemento criado a partir de estímulos vinculados a uma conjuntura específica. Desconstruídas supostas naturalidades identitárias, o segundo capítulo concentra-se na análise das principais iniciativas da diplomacia brasileira para a edificação dos diversos modelos de regionalismo no âmbito sul-americano, destacando-se a reunião proposta por Fernando Henrique Cardoso. Ato contínuo, o terceiro capítulo busca verificar as principais ações da gestão Lula da Silva para a região, tendo em vista que esta última foi alçada à condição de prioridade da política externa durante seu governo. Nesse sentido, levantamos os argumentos que embasam e estruturam a identidade sul-americana na virada do século XX para o XXI, trazendo à tona divergências paradigmáticas atinentes às agremiações partidárias que polarizam o cenário político nacional e, por conseguinte, apresentam convicções distintas no que tange ao aprofundamento dessa nova vertente identitária.
