Doppler ultrasonography is a new technology that has been study by researchers to improve the physiologic and pathologic knowledge about reproduction. This technology is based on Doppler-shifts frequencies or ultrasonic, these frequencies can be increase or decrease according to the movements of the red cells in the vessel. Color Doppler and power Doppler are the two possibilities to use the Doppler ultrasonography. Color Doppler is based in more the one color that show the direction of the blood f low and power Doppler is based in one color that change according of the flow intensity. Doppler ultrasonography can be demonstrated with the spectral mode to verify blood flow in large vessels, because of this, it is not use in equine reproduction. Studies in equine reproduction have been doing to verify uterus blood flow in cyclic mares and to observe the vascular perfusion in mares with cists, uterine vascular perfusion post breeding and verify the affects of drugs to decrease the uterus fluid in mares with problems in uterus perfusion. The ovarian irrigation during the estrus cycle was analyze with the measurement of the principal hormones during the estrus cycle in mares, the integrity of the corpus luteus, the irrigation of the future dominant follicle and the consequences in the ovarian irrigation after luteolyse induction also were study. Nevertheless, more than the knowledge that existed about Doppler ultrasonography, new studies have been doing to improve the forms to use Doppler ultrasonography in equine reproduction

Methods of semen cryopreservation allow changes in spermatic cells, such as damage in plasma and acrossomal membrane and modifications in mitochondrial function due to a disorder in the lipidic bilayer. For effective oocyte fertilization, spermatozoa require functional competent membranes, and intact organelles, acrosome and DNA. However, most laboratory methods used to evaluate semen quality are not highly correlated with fertilizing capacity. The discovery of a variety of fluorochromes and compounds conjugated to fluorescent probes has enabled an accurate assessment of the viability, integrity and function of spermatozoa. Among the most used probes that label the various compartments of the sperm cell there are the membrane impermeable fluorescent dyes to test the membrane integrity, as well as acylated dyes that pass the intact membrane. For the acrossomal integrity the most commonly used method is lectins labeled by a fluorescent probe. The acrosome reaction and spermatic capacitation is detected by the evaluation of membrane architecture and disorder of lipids in plasma membrane. Mitochondrial function can be determined using markers for their aerobic activity. The DNA status of spermatozoa has been determined using the metachromatic properties of Acridine Orange, and the DNA fragmentation can also be assessed by TUNEL assay. Finally, DNA condensation is analyzed using a single cell DNA gel electrophoresis assay that indicates DNA compactation. This monograph aims to compile the various tests used to detect damaged spermatozoa under cryopreservation methods, searching for improve the predictive value of semen analysis with the intention of a successful conception

Equine neonatal isoerythrolysis is a neonatal foals’ illness. Results from the incompatibility of blood type between the foal and the mare and mediated by maternal antibody absorbed by the colostrum against foal’s red blood cells. Characterized by a type ll hypersensitivity reaction, where the exhibition of the organism to a strange antigen, that it takes the sensitization of the lymphocytes B that after the removal of the antigens by the reticule-endothelial system the production of immunoglobulin is decreased, with the formation of cellular immunological will cause the occurrence of the illness in foal of sensitized mares. The most important clinical signs are severe anemia and jaundice, and this illness should be differentiated of other as: hemolysis induced by bacterial toxins, diseases of the hepatobiliary system, disseminated intravascular coagulation and incompatibility in blood transfusions. Like the sensitization happens during the previous incompatible foal’s birth, most cases occur in foals of multiparous mares. However during the first pregnancy the mare can generate a foal with neonatal 7 isoerythrolysis if she have developed placental anomaly in the beginning of the pregnancy which blood cells in her circulation

Os hormônios esteróides anabólico-androgênicos (EAA) compreendem a testosterona e seus derivados. Atletas e não atletas interessados em aumentar o desempenho esportivo, a força, a aparência física e a massa muscular têm utilizado esteróides anabólico-androgênicos sintéticos (EAAs) sem indicação médica. O uso de EAAs pode causar câncer de próstata, doença coronariana e esterilidade. No presente trabalho, avaliou-se os efeitos de altas doses de decanoato de nandrolona, do exercício físico resistido e da interação desses fatores sobre os lobos prostáticos de ratos machos adultos. Vinte ratos foram divididos aleatoriamente em quatro grupos experimentais e tratados e pesados por oito semanas: Esteróide + Exercício (EE), Sedentário + Esteróide (ES), Veículo + Exercício (VE) e Veículo + Sedentário (VS). Eles receberam injeção i.m de decanoato de nandrolona (5mg/Kg) e veículo (propilenoglicol) (0,2 mL/Kg). O peso absoluto corpóreo e do lobo ventral da próstata foi aferido. Os lobos prostáticos foram coletados, dissecados e processados, empregando-se técnicas microscópicas (histológicas, histoquímicas e imunohistoquímicas) e morfométricas. Os dados foram analisados por ANOVA bifatorial e Tukey (p<0,05). Alterações estatisticamente significativas nas massas corpórea, do lobo ventral, no índice de proliferação celular, na população de células basais e na análise dos volumes relativos dos componentes prostáticos (epitélio, estroma e lúmen) foram observadas. O decanoato de nandrolona determinou o aumento da proliferação celular, sugerindo o desenvolvimento da hiperplasia hormonal. De modo geral, as respostas não foram homogêneas para os parâmetros analisados, visto que os tecidos apresentam níveis de expressão de receptores de andrógenos, afinidade de ligação e vias metabólicas de conversão... The success in implementing an embryo transfer program derives from many factors. The embryo recovery rate is one of the most important factors, and allows the transfer to the recipient mare. This rate comes from a group of elements such as donor´s characteristics, collection date, reproductive management, semen quality, technician skill. Mainly because of the great importance of this direct interference on the results of an embryo recovery program, this study aimed to review the factors involved in embryo recovery rates in donor mares during an embryo transfer, suggesting some ways of improving these results

O diagnóstico genético pré-implantacional de embriões equinos já é uma realidade nos Estados Unidos e Argentina. Embora o Brasil seja líder mundial de transferência de embriões equinos, essa tecnlogia ainda está em fase de desenvolvimento, com poucos grupos atuando nessa área no país. O objetivo do presente projeto foi desenvolver a metodologia de micromanipulação e identificação do sexo de embriões equinos a campo. Dessa forma, selecionamos sequências específicas para padronização laboratorial dos ensaios de PCR com amostras de DNA da espécie equina. Os primers que apresentaram maior sensibilidade foram selecionados para a realização da PCR em biópsias embrionárias. Além das análises de biologia molecular, foi desenvolvida a técnica de micromanipulação de embriões equinos. Foram realizadas 105 biópsias de embriões pela técnica de microaspiração, permitindo a identificação do sexo pela técnica de WGA (Whole Genome Amplification) e posteriormente PCR seguido de eletroforese. Os embriões sexados foram transferidos para receptoras sincronizadas, permitindo a avaliação da taxa de concepção dos embriões submetidos à biópsia. Além disso, um embrião foi biopsiado, vitrificado e sexado. Após a identificação do sexo (fêmea) o embrião foi desvitrificado e transferido em receptora criteriosamente selecionada, resultando em prenhez

As observações levadas a cabo em um galinheiro experimental mostraram que Psychodopygus intermedius tem a capacidade de nele abrigar-se. Para se chegar a este resultado, a metodologia utilizada consistiu na coleta total diurna e exame do estado de alimentação e digestão sangüínea dos Ps. intermedius como parâmetro da sua maior ou menor permanência no ecótopo estudado. Além disso, observou-se paralelamente, os tempos para o repasto sangüíneo, digestão completa, oviposição, sobrevivência e cópula sob a influência direta dos fatores físicos naturais. A importância epidemiológica dos resultados reside em novas elucidações experimentais sobre a viabilidade da transmissão da leshmaniose tegumentar ocorrer em ambiente domiciliar.

Species of the Byrsonima genus are widely distributed around the neotropical zone, being frequently used in folk medicine to treat gastrointestinal, respiratory and skin diseases. This article briefly reviews the ethnopharmacology, pharmacology and phytochemistry of the Byrsonima genus. Eighty three compounds isolated from different species are reported, most of them being flavonoids or triperpenes. The pharmacological studies carried out with the extracts from these plants emphasize on the antimicrobial activity, however other activities have also been investigated leading to promising results. The data presented in this work strongly supports the view that plants of Byrsonima genus have potential therapeutic action.

This work describes the biflavonoids found in species of Clusiaceae, particularly the genera Garcinia and Calophyllum, emphasizing the importance of these metabolites as chemical markers of this family, their contribution to the pharmacological potential of these species, besides the promising potential of these compounds in the search for new drugs.

The present study examines the chemical composition and their effects on free radicals, inflammation, angiogenesis, coagulation, VEGF effects and cellular proliferation of a polysaccharides from alga Sargassum vulgare. The sulfated polysaccharide was extracted from brown seaweed by proteolysis with enzymes maxataze. The presence of proteins and sugars were observed in crude polysaccharides. Fractionation of this crude extract was made with growing concentration of acetone (0.3-1.5 v) and produced four groups of polysaccharides. Anionic polysaccharides from brown seaweed Sargassum vulgare, SV1and PSV1 were fractionated (SV1) and purified (PSV1), and displayed with high total sugars and sulfate content and very low level of protein. This fucan SV1 contains low levels of protein and high carbohydrate and sulfate content. This polysaccharides prolonged activated partial thromboplastin time (aPTT) at 50 μg (>240 s). SV1 was found to have no effect on prothrombin time (PT), corresponding to the extrinsic pathway of coagulation. SV1 exhibits high antithrombotic action in vivo, with a concentration ten times higher than heparin. Polysaccharides from S. vulgare promoted direct inhibition enzymatic activity of thrombin and stimulated enzymatic activity of FXa. SV1 showed optimal inhibitory activity of thrombin (50.2±0.28%) at a concentration of 25 μg/mL. Its antioxidant action on scavenging radicals by DPPH was (22%), indicating the polymer has no cytotoxic action (hemolytic) on ABO and Rh blood types in different erythrocyte groups and displays strong anti-inflammatory action on all concentrations tested in the carrageenan-induced paw edema model, demonstrated by reduced edema and cellular infiltration. Angiogenesis is a dynamic process of proliferation and differentiation. It requires endothelial proliferation, migration, and tube formation. In this context, endothelial cells are a preferred target for several studies and therapies. The antiangiogenic efficacy of polysaccharides was examined in vivo in the chick chorioallantoic membrane (CAM) model by using fertilized eggs. Decreases in the density of the capillaries were assessed and scored. The results showed that SV1 and PSV1 have an inhibitory effect on angiogenesis. These results were also confirmed by inhibition tubulogenesis in rabbit aorta endothelial cell (RAEC) in matrigel. These compounds were assessed in Apoptosis assay (Annexin V - FITC / PI) and cell viability by MTT assay of RAEC. These polysaccharides do not affect the viability and do not have apoptotic or necrotic action. RAEC cell when incubated with SV1 and PSV1showed inhibition of VEGF secretion, observed when compounds were incubated at 25, 50 and 100 μg/μL. The VEGF secretion with the RAEC cell line for 24 h, was more effective for PSV1 at 50 μg/μL(71.4%) than SV1 100 μg/μL (75.9%). SV1 and PSV1 had an antiproliferative action (47%) against tumor cell line HeLa. Our results indicate that these sulfated polysaccharides have antiangiogenic and antitumoral actions

Reproduction and feeding aspects of Neoplecostomus microps (Loricariidae, Neoplecostominae) in the Ribeirao Grande system, eastern Serra da Mantiqueira (São Paulo State). The Ribeirao Grande system is located in the slope of the Serra da Mantiqueira and is surrounded by Atlantic forest. Eight sites were surveyed in the Ribeirao Grande system, during the winter (July 2001), spring (October 2001), summer (February 2002) and fall (April 2002). Samples were made with an electrofishing device. Gonad maturation, levels of stomach fullness and fat content were analyzed and their distributions reported in an annual cycle. Neoplecostomus microps has a spawning in the spring through to summer. The size at sexual maturity is about 5.9 cm total length for males and 6.9 cm for females. The high values in October and February by the gonadosomatic index and the relative condition factor coincided with the spawning period. In the diet of N. microps were found Diptera larvae (Simulium, Chironomus), Plecoptera nymphs and Coleoptera aquatic Larvae (Psephenus). The increased feeding from summer to fall provided fat accumulation. During subsequent seasons, fish may utilize visceral fat reserves for maintenance and reproduction. The reproductive pattern and feeding are interpreted as being an adaptation with respect to temporal and spatial variation and food availability.
