Pouco se sabe a respeito da marcha de pacientes com doença de Parkinson (DP) em ambientes irregulares. Mais especificamente, não foram encontrados estudos que manipularam a informação visual na tarefa de ultrapassar obstáculo. Assim, o objetivo deste estudo foi analisar o papel da visão no comportamento locomotor durante a ultrapassagem de obstáculos de diferentes alturas em indivíduos com DP idiopática, manipulando experimentalmente a disponibilidade das informações visuais. Participaram 14 idosos com DP, classificados entre os estágios 1 e 2,5 da escala de Hoehn & Yahr. Os participantes foram convidados a percorrer andando uma passarela de 8 metros, em condições sem obstáculo (SO) e com obstáculo baixo (OB) ou alto (OA). As tentativas foram apresentadas combinando condições de obstáculo (SO, OB e OA) e de visão (IVD e IVE), em 3 tentativas por condição, totalizando 18 tentativas; realizadas em 2 blocos. A manipulação de informação visual foi realizada através do uso de óculos de lentes de cristal líquido e a análise cinemática foi realizada tridimensionalmente. As variáveis dependentes selecionadas incluíram os parâmetros espaciais e temporais das fases de aproximação e ultrapassagem do obstáculo. Para o tratamento dos dados, foram empregadas duas MANOVAs. Os resultados revelaram que tanto a manipulação visual como a manipulação do terreno influenciaram no comportamento locomotor dos pacientes. Na fase de aproximação, a presença do obstáculo provocou modificações nos parâmetros locomotores, refletindo em menores valores médios de comprimento e velocidade da passada nas condições OB e OA do que na condição SO. Em relação à informação visual nesta mesma fase, a anulação do fluxo óptico provocou diminuição do comprimento e da velocidade da passada, da cadência e da porcentagem da fase de suporte simples e aumento da duração... Monócitos e macrófagos são células da imunidade inata que desempenham importante papel na defesa contra infecções fúngicas, através do reconhecimento dos fungos, ativação e desenvolvimento de atividade fungicida. A função das células fagocitárias depende do seu estado de ativação, induzido principalmente pelo ambiente de citocinas, presentes durante a interação com o microrganismo. O presente projeto teve por objetivo avaliar o efeito da opsonização de células leveduriformes de Paracoccidioides brasiliensis com soro humano sobre a produção de peróxido de hidrogênio (H2O2) e atividade fungicida de monócitos humanos, estimulados ou não com Interferon-gama (IFN-γ), contra P. brasiliensis. Monócitos de sangue periférico, obtidos de indivíduos saudáveis, foram cultivados na ausência ou presença de IFN-γ por 24h a 37oC. A seguir, essas células foram desafiadas com a amostra Pb18 de P. brasiliensis por 60 min para determinação da produção de H2O2 e por 4h para a avaliação da atividade fungicida nas proporções fungomonócito de 1:50. A suspensão fúngica foi previamente incubada por 30 min a 37oC na ausência de soro (SS) ou na presença de pool de soro humano normal, inativado (SI) por aquecimento a 56oC ou soro humano fresco (SF) não submetido à inativação do sistema complemento, nas concentrações de 5%, 10%, 20% e 30% em meio de cultura RPMI. A atividade fungicida de monócitos contra a amostra Pb18 foi avaliada por plaqueamento dessas co-culturas em meio de cultivo BHIágar e determinação da recuperação de fungos viáveis após 14 dias de incubação a 36oC. A produção de H2O2 por monócitos foi determinada através da técnica de redução do vermelho de fenol. Currently the use of pesticides in agriculture is widespread due to their high effectiveness in combating pests, weeds and diseases leading to better productivity and economical performance in agricultural area. The use of pesticides affects the whole world and their use is often performed in an improper and indiscriminate way and for long periods. Several studies have been carried out in order to verify the presence of pesticides in nature, with worrying results. The presence of higher levels of pesticides and their degradation products in soil and surface and groundwater have indicated increasing contamination. Among the most widely used pesticides, herbicides are present and among these trifluralin has occupied an important place due to its widespread use; it is an herbicide originated from benzene derivatives belonging to dinitroanilines family; it is classified as belonging to group C, being possibly carcinogenic for humans, present a high persistence in soil as a result of its low mobility and therefore may affect local edaphic fauna. Diplopods belong to a group of invertebrates considered important in the soil dynamics; due to their close contact with it, these animals can be used as bioindicators of substrates toxicity. This study aimed to expose diplopod specimens of the species R. padbergi to different concentrations of trifluralin and therefore it was mounted five bioassays containing soil from the site where animals were collected (control group) and the same soil mixed to different concentrations of trifluralin herbicide (concentration recommended for agriculture use, that is, the dose recommended by the producer 0.0534g/m2, double, haft and quarter of this dose), animals were exposed for 7 and 90 days. Nos idosos, o andar tem sido associado com a preservação das funções cognitivas. Entretanto, pacientes com Demência de Alzheimer (DA), apresentam modulações durante o andar, decorrente das disfunções no lobo frontal, principalmente no córtex motor e declínio nas funções executivas. Contudo, a marcha de idosos com DA pode ser mais vulnerável na presença de uma tarefa dupla, pois esses idosos apresentam déficits nas ações que exigem planejamento, atenção e ajustes motores. Desse modo, o objetivo deste estudo foi analisar no paradigma da tarefa dupla os valores médios das variáveis espaciais e temporais do andar de pacientes com DA. A amostra do estudo foi composta por 19 idosos com DA, sendo 14 mulheres e 5 homens. Os pacientes com DA realizaram 5 tentativas das seguintes tarefas: andar livre sem tarefa dupla e andar livre com tarefa dupla. A tarefa dupla realizada pelos pacientes foi uma contagem regressiva iniciando do número vinte. As variáveis espaço-temporais foram coletadas através de um sistema optoeletrônico de análise do movimento. As variáveis analisadas foram comprimento da passada, largura do passo, duração da fase de suporte simples e duplo suporte, duração da passada e velocidade da passada. Para verificar as diferenças entre as condições experimentais no andar livre foi utilizado o teste t de Student pareado (p<0,05) com fator para tarefa. O teste t pareado revelou efeito da tarefa no comprimento da passada, largura do passo, suporte simples, duplo suporte, duração da passada e velocidade da passada. Os pacientes reduziram o comprimento e a velocidade da passada e aumentaram a largura do passo e as durações do suporte simples, duplo suporte e da passada na condição marcha livre com contagem em comparação com a marcha livre sem contagem. Aedes (Stegomyia) aegypti is a species of mosquitoes known to be the vector of diseases such as dengue and yellow fever, and a better understanding of aspects of their biology can help in the establishment of control strategies for the same. Several previous studies showed that temperature significantly affects the development of immature stages of insects. In general, higher temperatures (up to a threshold) accelerate the development of insects, and lower temperature retards the same. This rule also applies to mosquitoes, including Ae. aegypti. But not still know the effects of daily variation of temperature on the developmental stages of mosquitoes. And this detail is very important, since in natural breeding or artificial, The mosquitoes usually face temperature variations over a single day, which should interfere with its development until the emergence of the adult forms. For this reason, the objective of this study is to analyze the effect of alternating temperatures on the development of Ae. aegypti. To conduct the study, adults were collected active in the neighborhood Bela Vista Campus of UNESP - Rio Claro, SP, using a sweep net or using ovitraps for immatures, and the active search for breeding. Individuals collected were kept under experimental conditions in the laboratory. The adult samples were identified to species level, were considered for the experiments, only samples of Ae. aegypti. The insects were housed in plastic cages, suitable for creating flies. These were fed with sugar solution and blood meal on alternate days. Photodynamic Therapy (PDT) is a therapeutic method which employs a photosensitizer and light to cause cellular death. The chemical compounds have low or none toxicity for hosts cells. Under the incidence of light, in an appropriated wavelength, these chemical compounds produce reactive oxygen which affects the biomolecules of the target-cells. The specific illumination of the affected area increases the selectivity of the therapy, since the photodynamic process occurs only in the irradiated area. Pythiosis, for instance, is a life-threatening emerging disease caused by a fungus-like organism called Pythium insidiosum. The disease occurs in man and other animals, being mostly observed in horses. Human pythiosis may present as ophthalmic, cutaneous-subcutaneous and systemic forms of lesions. Due to the fact that P. insidiosum is not a true fungus, it is refractory to most antifungal drugs and the treatment of the disease is difficult. Extensive surgery procedures, such as limb amputation, are the treatment of choice, however relapses may occur frequently. Although not totally effective, the use of immunotherapy associated to surgery have shown some results. Considering that pythiosis is an emerging disease few explored in its etiological and therapeutic aspects, which are limited and few effective, it is of great importance to encourage the development of researches for new strategies of treatment. In this sense, it was evaluated the effect of PDT on in vitro growth of the pathogen employing two chemical compounds as photosensitizer, porphyrin and chlorine, at different concentrations in combination with several energetic dosages. Females of the painted turtle, Trachemys scripta, have an annual reproductive cycle. Their nest periods in Brazil starts around September and they lay about 7 - 10 eggs in nests dug in the soil. The eggs hatch in December. It is possible that the nest cover cause problems for the gas diffusion, also, the rain can wet or compact the soil covering the nest, or even soaked the whole area. Those problems may change the nests intern atmosphere decreasing O2 partial pressure (hypoxia) and increasing CO2 partial pressure (hypercarbia). The main objective of this study was to determine if and how hypercarbia (3% and 6%CO2), could influence the eggs and early posthatching development of the Trachemys scripta. We used three experimental groups of eggs incubated in: normal atmosphere (Control group CG, n = 14), hypercarbia with 3% of CO2 (G3%, n = 16) and with 6% of CO2 (G6%, n = 15). The profile of the mass increment during the egg development were not different among the groups, GC = 9.89±0.81g; G3% = 10.11±0.60g and G6% = 10.58±0.36g. In addition, the mass of the post-hatching turtles were the not different. There were not differences between the duration of the incubation period among the experimental groups, GC = 60.12±0.97days, G3% = 59.00±0.98days and G6% = 60.80±1.14days. The mortality rates were not affected by the hypercarbia, GC = 0.43 (43%), G3% = 0.27 (27%) and G6% = 0.37 (37%). Furthermore, the mass increment and the metabolic rate were the same among the early development of the turtles until 3 month after hatching. We conclude that hypercarbia (up to 6% of CO2) does not cause any alteration on the eggs or post-hatching normal development. The aim of this study was to analyze the influence of prior exercise on different intensity distribution strategies (pacing). The study included five male individuals, apparently healthy, aged between 18 and 25 years, and without regular practice of physical activities. The subjects were tested on different days following the protocols on a cycle ergometer: 1) a progressive ramp test, 2) three constant load tests in the intensities of 85%, 90% and 95% IVO2max to exhaustion, and 3) six tests with and without holding a prior exercise intensity of 70%  with different strategies for Even-intensity pace, ES (401 ± 70 W), which consists of an exercise at a pace and / or constant intensity from beginning to end; where intensity is initially increased to 10% less than the ES progressively increase to reach 10% above the intensity of ES, and; Fast-start; FS, where there is a reversal in the way of distributing intensity, ie the initial intensity is 10% higher than the value of ES decreased progressively to 10% below that ES. All these tests were performed in random order. The tlim with previous exercise was significantly shorter than without previous exercise at FS condition (p < 0.05). The VO2final obtained at ES condition was similar with (3243 ± 599 ml.min-1) and without (3252 ± 384 ml.min-1) previous aerobic exercise (p > 0.05). However, the VO2final obtained at FS condition was higher with (3291 ± 218 ml.min-1) than without (3097 ± 207 ml.min-1) previous aerobic exercise (p < 0.05). The heart rate was higher at ES condition with than without previous aerobic exercise (p < 0.05). There was no significant difference in this variable for FS condition with and without previous aerobic exercise (p > 0.05). It can be concluded that the results the previous exercise (70%) achieved lasting 6 minutes followed by 6 minutes of recovery appears to influence / commit ting the conditions during the pacing for this population

A quimioterapia antitumoral convencional, baseada na aplicação da dose máxima tolerável de drogas antineoplásicas, está comumente associada com mielossupressão e causa diminuição da viabilidade e número de células dendríticas (DCs), principais células apresentadoras de antígenos (APCs) do sistema imune. Uma alternativa para limitar os efeitos tóxicos desses agentes é a quimioterapia metronômica, baseada na administração das drogas em doses baixas e frequentes que, entre outros aspectos, melhora a eficácia antitumoral pela inibição da angiogênese no sítio do tumor. Estudos prévios do grupo indicam que doses ultrabaixas de determinados agentes antineoplásicos modulam positivamente o estado de ativação e as funções das células dendríticas, sugerindo que a associação entre vacinas de DCs e quimioterapia metronômica possibilitaria a estimulação da resposta imune, ao mesmo tempo em que as células tumorais sofrem o efeito dos agentes antineoplásicos em doses ultrabaixas. Assim, hipotetizamos que o uso de 5-Fluorouracil e Leucovorina, drogas de escolha para o tratamento do câncer colorretal, em dose efetiva mínima ou dose ultrabaixa não tóxica, poderia modular positivamente as DCs humanas e potencializar a imunogenicidade de células tumorais, além de torná-las mais sensíveis à ação de linfócitos T citotóxicos. Desse modo, o presente estudo tem o objetivo de avaliar o efeito in vitro da dose efetiva mínima e dose não tóxica de 5-Fluorouracil e Leucovorina sobre as funções de células dendríticas humanas e sobre a imunogenicidade de células de câncer colorretal humano da linhagem HT-29. Adequate testosterone levels are necessary for the development, growth and maintenance of the male reproductive system. Testosterone deficiency is common in men with diabetes in whom it may contribute to impaired performance, with consequent reduction of the activity of the androgen regulated organs, such as the prostate. However, little attention has been given to the plasma dihydrotestosterone (DHT) level, the most potent androgen, nor to the expression of the androgen receptor (AR), insulin-like growth factor type I (IGF-1) and receptor (IGF-1R) in target tissues. Here, we investigated the effect of type I diabetes mellitus on DHT plasma levels and on prostate AR, IGF-1 and IGF-1R expression during rat pubertal growth. Diabetes was induced in prepubertal male rats through administration of streptozotocin (STZ; 40 mg/kg). Diabetic, diabetic treated with insulin, and age-matched control animals were killed by overdoses of pentobarbital. The ventral prostatic lobe (VP) was dissected, weighed and processed for immunohistochemistry for AR, IGF-1 e IGF-1R; plasma T and DHT levels were also determined. Hyperglycemia at puberty reduced VP weight gain to about 50% and plasma T level to about 80% of the control levels. In contrast there were no changes in plasma DHT levels. Insulin replacement restored the VP weight gain, but not the plasma T levels, which remained 90% below the ones of controls. Immunohistochemistry showed that AR, IGF-1 and IGF-1R expression in the prostate epithelial cells did not change with hyperglycemia or insulin replacement. Thus, the AR expression in the prostate epithelial cells appears to be regulated by DHT, and to a minor extent it also controls glandular growth

O objetivo do presente estudo foi verificar o efeito da adição de um prebiótico nas características de crescimento, resistência às condições do trato gastrintestinal e remoção de colesterol in vitro, das cepas de Enterococcus faecium CRL183 e Lactobacillus helveticus 416. A fonte de prebiótico foi composta por Inulina GR – Orafti, sendo testadas as seguintes concentrações: 1,0%, 2,0%, 3,0%; 4,0%; 5,0% e 6,0% p/v). Para a avaliação da resistência às condições do trato gastrintestinal, os microrganismos foram colocados em contato com um fluido gástrico simulado por 2 horas (9g/L de NaCl, 3g/L de pepsina, pH 1,8) e com um fluido intestinal simulado por 3 horas (0,9g/L de pancreatina, 12,5g/L de bicarbonato de sódio e 6,0 g/L de Oxgall). Após o período de contato com os fluidos, foram realizadas diluições seriais e plaqueamento em meios específicos. No estudo de redução de colesterol, meios de cultura contendo inulina e colesterol (1%) foram inoculados com as cepas probióticas e incubados em anaerobiose a 37°C/24h. A capacidade de redução do colesterol foi determinada pela diferença entre a concentração de colesterol adicionada ao meio de cultura e a de colesterol residual. Os resultados foram submetidos à análise de variância e teste de médias de Tukey (p≤0,05). A adição de inulina não resultou em alteração no ciclo logarítmico de crescimento dos microrganismos. Porém, quando os dados foram submetidos à análise estatística, constatou-se que a adição de 1% a 4% de inulina inibiu o crescimento de Lactobacillus helveticus 416, sendo que para as concentrações de 5% a 6% estas alterações não foram perceptíveis. A cepa de Enterococcus faecium CRL 183 apresentou crescimento superior na presença de 2% e 4% de inulina. A bothropstoxina-I (BthTX-I) é uma fosfolipase A2 (PLA2) Lys49 miotóxica isolada do veneno da Bothrops jararacussu. Embora seja desprovida de atividade neurotóxica in vivo, esta toxina bloqueia a transmissão neuromuscular in vitro. A relação entre as atividades miotóxica e paralisante da BthTX-I ainda não está esclarecida. A crotapotina corresponde à subunidade não-enzimática da crotoxina, principal fração tóxica do veneno da Crotalus durissus terrificus. Isoladamente a crotapotina é atóxica, porém atua como carreadora da PLA2 Asp49 da crotoxina, potencializando sua ação neurotóxica. Esta proteína também é capaz de se complexar com outras PLA2s (Asp49 ou Lys49) de venenos ofídicos, alterando suas toxicidades. Neste trabalho avaliamos a influência da crotapotina sobre o bloqueio neuromuscular e a atividade miotóxica da BthTX-I in vitro. Preparações do nervo frênico-músculo diafragma de camundongos machos foram montadas em cubas para o registro das contrações musculares evocadas direta e indiretamente. Cortes transversais do músculo foram submetidos à coloração por hematoxilina e eosina para a avaliação do padrão morfológico. A BthTX-I (1 μM) isoladamente, ou pré-incubada com crotapotina (2 M) à 35 ºC por 30 minutos, foram adicionadas às preparações. A análise dos dados foi realizada por testes não paramétricos (p<0.05). A BthTX-I induziu bloqueio irreversível e tempo-dependente das contrações musculares diretas e indiretas. O tempo para o bloqueio de 50% das contrações indiretas (18,98 ± 1,94 min, n=4) foi significativamente menor que o das diretas (45,97 ± 5,61 min, n=5). A pré-incubação com a crotapotina não alterou de forma significativa o bloqueio das contrações diretas ou indiretas induzidos pela BthTX-I. Isoladamente, a crotapotina não afetou as contrações... In the present study it was investigated, in plant and population scale, the relationship between the amount of resource and body size, species diversity and abundance and biomass of bruchids and their parasitoids. Possible effects of resource quality (soil quality and concentration of tannins in seeds) in response to the variables mentioned above were also analyzed. Mimosa bimucronata fruits were collected in two areas during the occurrence of bruchids during the years 2009 and 2010. Only the species of bruchid Acanthoscelides schrankiae was found infesting the fruits of M. bimucronata. The parasitoids belong to families Braconidae, Encyrtidae, Eulophidae, Eupelmidae, Eurytomidae and Pteromalidae. Most local-level analysis showed no significant results for both years of collection and study areas. There was only significant result for the relationship between the biomass of parasitoids and the concentration of tannins considering the year 2009 (negative trend). The soil from Lageado showed higher percentages of silt and clay. With respect to chemical analysis, pH, cation exchange capacity, and concentrations of organic matter, potassium, calcium and magnesium were all significantly higher in soil from Lageado. Therefore, it was found that the plants from Lageado are growing in more fertile soils. However, it was found that the amount of fruits and seeds was significantly higher in Rubião. Comparisons of the abundance of bruchids and parasitoids diversity among the areas that showed the highest values were observed in Rubião. In this study it was found that the abundance of bruchids and parasitoids, as well as the diversity of parasitoids was greatest in the area that had higher amounts of fruits and seeds, suggesting a significant relationship between the amount of resources and the abundance and diversity. Beside aging process comes the incidence of dementia and, among them Alzheimer's disease (AD) accounts for approximately 60% of cases. This disease is characterized as a neuropathology with unknown etiology that causes cognitive deficits and behavioral disorders. Caring for patients with AD can cause an overload, both physical and psychological, which can cause high levels of stress on the primary caregiver. It is necessary that the caregiver also receives attention and develop activities that promote health benefits, while providing moments of distraction from the task of caring. Nonpharmacological interventions may be favorable for improving health with consequent decreased on the levels of stress. The objective of this study was to conduct a systematic review of scientific papers that aimed to verify the effect of nonpharmacological interventions on stress levels in caregivers of patients with AD. To contamplate this goal was accomplished a systematic search in the following databases: Biological Abstracts, PsycINFO, PubMed/Medline, Web of Science, LILACS and SciELO. The following keywords and Boolean operators was used: “caregivers” OR “family” and “nonpharmacological interventions” OR “support groups” OR “therapies” AND “Alzheimer's disease” OR “Alzheimer's dementia” OR “Alzheimer” OR “elderly” AND “stress”. There were found 3studies that met inclusion criteria adopted for the present work, and none showed significant results for the variable stress. It is not possible to affirm, according to the studies, that nonpharmacologial interventions programs for caregivers of patients with AD are effective to influence and to control the stress. The study of gastrointestinal tract (GIT) functions is necessary due to the increasing number of pathologies associated with it. Directly influencing the quality of life, the gastrointestinal tract provides a number of parameters that, when analyzed, allow us to describe its dysfunctions. Thus, many techniques can be combined to obtain these properties related to the GIT. However, these techniques are often invasive, require surgery, catheter insertion, or to build a temporal model of these functions, require the sacrifice of animals in a series of data collection. The technique used in this study has the advantage of having a low operating cost, being free of ionizing radiation, non-invasive and is known as biosusceptometry AC (BAC), used to evaluate the properties of the GI tract by monitoring the position and concentration of materials magnetically marked. The sensor consists of two pairs of coils, one reference and one for detection. A fixed base line separates the sensing and reference coils, and also functions as support for the instrumentation. It is also important to note that the detection coils are arranged in a first order (subtraction) gradiometric way. The objective of this study was to analyze the effects of gastrectomy in gastric emptying and gastrointestinal transit time of solid food in rats using a BAC system associated with magnetic markers. To realize this study was constructed a dedicated BAC sensor, built to analyze these GIT properties. Data acquisition was obtained by aligning the magnetic sensor with the stomach and colon of the animal at pre-determined intervals.
