The microfilamentous green alga Uronema curvatum is widely distributed along the western and eastern coasts of the north Atlantic Ocean where it typically grows on crustose red algae and on haptera of kelps in subtidal habitats. The placement of this marine species in a genus of freshwater Chlorophyceae had been questioned. Molecular phylogenetic analysis of nuclear-encoded small and large subunit rDNA sequences reveal that U. curvatum is closely related to the ulvophycean order Cladophorales, with which it shares a number of morphological features, including a siphonocladous level of organization and zoidangial development. The divergent phylogenetic position of U. curvatum, sister to the rest of the Cladophorales, along with a combination of distinctive morphological features, such as the absence of pyrenoids, the diminutive size of the unbranched filaments and the discoid holdfast, warrants the recognition of a separate genus, Okellya, within a new family of Cladophorales, Okellyaceae. The epiphytic Urospora microscopica from Norway, which has been allied with U. curvatum, is revealed as a member of the cladophoralean genus Chaetomorpha and is herein transferred to that genus as C. norvegica nom. nov.

The overall biotic pressure on a newly introduced species may be less than that experienced within its native range, facilitating invasion. The brown alga Sargassum muticum (Yendo) Fensholt is a conspicuous and successful invasive species originally from Japan and China. We compared S. muticum and native macroalgae with respect to the biotic pressures of mesoherbivore grazing and ectocarpoid fouling. In Strangford Lough, Northern Ireland, S. muticum thalli were as heavily overgrown with seasonal blooms of epiphytic algae as native macroalgal species were. The herbivorous amphipod Dexamine spinosa was much more abundant on S. muticum than on any native macroalga. When cultured with this amphipod, S. muticum lost more tissue than three native macroalgae, Saccharina latissima (Linnaeus) Lane et al., Halidrys siliquosa (Linnaeus) Lyngbye and Fucus serratus Linnaeus. Sargassum muticum cultured with both ectocarpoid fouling and amphipods showed a severe impact, consistent with our previous findings of large declines in the density of S. muticum observed in the field during the peak of fouling. Despite being a recent introduction into the macroalgal community in Strangford Lough, S. muticum appears to be under biotic pressure at least equal to that on native species, suggesting that release from grazing and epiphytism does not contribute to the invasiveness of this species in Strangford Lough.

Esta tese debruça-se sobre a biodiversidade de líquenes epífitos de pinhais dunares portugueses e sobre uso de líquenes como biomonitores de poluição atmosférica nesse habitat. A Mata Nacional das Dunas de Quiaios (Figueira da Foz) foi o ponto de partida dos estudos de biodiversidade efetuados nesta tese, mas alguns deles estenderam-se à maior parte da costa portuguesa. Como resultado, encontrou-se uma espécie nova para a ciência, Lecanora sorediomarginata Rodrigues, Terrón & Elix, epifítica sobre Pinus pinaster Aiton e P. pinea L, que se encontra distribuída na maior parte da costa. Esta espécie caracteriza-se morfologicamente por um talo crustáceo, de cor esbranquiçada a acinzentada ou esverdeada e que desenvolve sorálios a partir de pequenas verrugas marginais. Quimicamente caracteriza-se pela presença dos ácidos 3,5-dicloro-2'-O-metilnorestenospórico [maior], 3,5-dicloro-2 -O-metilanziaico [menor], 3,5-dicloro-2 -O-metilnordivaricático [menor], 5-cloro-2'-Ometilanziaico [traço] e úsnico [traço]; atranorina [menor] e cloroatranorina [menor]. É quimicamente semelhante a L. lividocinerea Bagl., com a qual apresenta afinidades filogenéticas com base na análise da sequência ITS do rDNA, e a L. sulphurella Hepp. Adicionalmente, espécies Chrysothrix flavovirens Tønsberg e Ochrolechia arborea (Kreyer) Almb, também se encontraram epifíticas sobre P. pinaster e P. pinea em vários pinhais ao longo da costa, representando novos registos para a flora liquénica portuguesa, bem como a espécie Lepraria elobata Tønsberg encontrada epifítica sobre P. pinaster apenas nas Dunas de Quiaios. Além disso, as espécies Hypotrachyna lividescens (Kurok.) Hale e H. pseudosinuosa (Asahina) Hale encontraram-se epifíticas sobre P. pinaster e outros forófitos nas Dunas de Quiaios, constituindo novos registos para a flora liquénica da Península Ibérica. Estes resultados põe em evidência a importância dos pinhais dunares como habitat para líquenes epífitos. Num estudo conduzido entre janeiro e julho de 2008 num pinhal dunar (Mata do Urso, Figueira da Foz), em cuja bordadura existe uma fábrica de celulose de papel, usaram-se transplantes de líquenes da espécie Flavoparmelia caperata (L.) Hale para avaliar a acumulação de trinta e três elementos putativamente emitidos por fábricas de papel e pasta de papel. A cinética da fluorescência da clorofila a foi estudada nos líquenes transplantados, através da análise dos parâmetros Fv/Fm, F0, Fm, qP, NPQ, PSII, e Exc, de forma a avaliar os efeitos decorrentes da acumulação de elementos na vitalidade dos líquenes. Pretendeu-se avaliar se a acumulação de elementos e a cinética da fluorescência da clorofila a variavam significativamente com o local e o tempo de exposição, tendo em consideração os resultados obtidos de transplantes colocados num local de referência (Dunas de Quiaios) durante o mesmo período de tempo. (Continua no verso) resumo A maior parte dos elementos — Al, B, Ba, Ca, Co, Cr, Cu, Fe, Hg, Li, Mg, Mn, Mo, Na, Ni, P, S, Sb, Sc, Sr, Ti e V — ocorreu em concentrações significativamente mais elevadas nos transplantes expostos a 500 m da fábrica. Cerca de metade dos elementos estudados — B, Ba, Cr, Fe, Hg, Mg, Mn, Mo, Na, P, Pb, S, Sb e V — encontraram-se em concentrações significativamente mais elevadas nos transplantes expostos durante 180 dias. O solo foi identificado como uma fonte parcial da maior parte dos elementos. Os parâmetros Fv/Fm, Fm, PSII e Exc variaram significativamente com o local e/ou com o tempo de exposição. Observou-se um decréscimo significativo nos parâmetros Fv/Fm e Fm nos transplantes expostos a 500 e 1000 da fábrica, e também naqueles expostos durante 135 e 180 dias. Observou-se também um decréscimo significativo nos parâmetros PSII e Exc expostos durante 180 dias. Estes parâmetros correlacionaram-se de forma negativa e significativa com a acumulação de elementos: Fv/Fm: B, Ba, Co, Fe, Hg, Mg, Mn, Mo, N, P, S, Sb e Zn; Fm: Ba, Co, Hg, Mn, Mo, N, P, S, Sb e Zn; PSII: N e P; Exc: Mn, N, P e S. Estudos acerca da diversidade liquénica efetuados nos mesmos locais onde os transplantes foram colocados no local impactado, revelaram um menor valor de diversidade liquénica a 500 m da fábrica, que foi também o único local onde se encontraram espécies nitrófilas, o que se poderá dever à deposição de amónia e/ou poeiras. À semelhança de outros estudos, este trabalho confirma que os líquenes podem ser usados com sucesso em estudos de biomonitorização, mesmo em locais florestados. Além disso, traz também informações adicionais sobre como a acumulação de elementos pode influenciar a cinética da fluorescência da clorofila a em líquenes.

Fire blight is an economically important disease of apples and pears that is caused by the bacterium Erwinia amylovora. Control of the disease depends on limiting primaly blosson1 infection in the spring, and rapidly removing infected tissue. The possibility of using phages to control E.amylovora populations has been suggested, but previous studies have. failed to show high treatment efficacies. This work describes the development of a phage-based biopesticide that controls E. amylovora populations under field conditions, and significantly reduces the incidence of fire blight. This work reports the first use ofPantoea agglomerans, a non-pathogenic relative ofE. amylovora, as a carrier for E. amylovora.phages. Its role is to support a replicating population of these phages on blossom surfaces during the period when the flowers are most susceptible to infection. Seven phages and one carrier isolate were selected for field trials from existing collections of 56 E. amylovora phages and 249 epiphytic orchard bacteria. Selection of the . /' phages and carrier was based on characteristics relevant to the production and field perfonnance of a biopesticide: host range, genetic diversity, growth under the conditions of large-scale production, and the ability to prevent E. amylovora from infecting pear blossoms. In planta assays showed that both the phages and the carrier make significant contributions to reducirig the development of fire blight symptoms in pear blossoms. Field-scale phage production and purification methods were developed based on the growth characteristics of the phages and bacteria in liquid culture, and on the survival of phages in various liquid media. Six of twelve phage-carrier biopesticide treatments caused statistically signiflcant reductions in disease incidence during orchard trials. Multiplex real-time PCR was used to simultaneously monitor the phage, carrier, and pathogen populations over the course of selected treatments. In all cases. the observed population dynamics of the biocontrol agents and the pathogen were consistent with the success or failure of each treatment to control disease incidence. In treatments exhibiting a significantly reduced incidel1ce of fire blight, the average blossom population ofE.amylovora had been reduced to pre-experiment epiphytic levels. In successful treatments the phages grew on the P. agglomerans carrier for 2 to 3 d after treatment application. The phages then grew preferentially on the pathogen, once it was introduced into this blossom ecosystem. The efficacy of the successful phage-based treatnlents was statistically similar to that of streptomycin, which is the most effective bactericide currently available for fire blight prevention. The in planta behaviour ofE. amylovora was compared to that ofErwinia pyrifoliae, a closely related species that causes fire blight-like synlptoms on pears in southeast Asia. Duplex real-time PCR was used to monitor the population dynamics of both species on single blossonls. E. amylovora exhibited a greater competitive fitness on Bartlett pear blossoms than E. pyrifoliae. The genome ofErwinia phage Ea21-4 was sequenced and annotated. Most of the 8-4.7 kB genome is substantially different from previously described sequences, though some regions are notably similar to Salmonella phage Felix 01 . Putative functions were assigned to approximately 30% of the predicted open reading frames based on amino acid sequence comparisons and N-terminal sequencing of structural proteins.

Forty-four bacteriophage isolates of Erwinia amy/ovora, the causal agent of fire blight, were collected from sites in and around the Niagara Region of Southern Ontario in the summer of 1998. Phages were isolated only from sites where fire blight was present. Thirty-seven of these phages were isolated from the soil surrounding infected trees, with the remainder isolated from aerial plant tissue samples. A mixture of six E. amy/ovora bacterial host strains was used to enrich field samples in order to avoid the selection bias of a single-host system. Molecular characterization of the phages with a combination of peR and restriction endonuclease digestions showed that six distinct phage types were isolated. Ten phage isolates related to the previously characterized E. amy/ovora phage PEa1 were isolated, with some divergence of molecular markers between phages isolated from different sites. The host ranges of the phages revealed that certain types were unable to efficiently lyse some E. amy/ovora strains, and that some types were able to lyse the epiphytic bacterium Pantoea agg/omerans. Biological control of E. amy/ovora by the bacteriophages was assessed in a bioassay using discs of immature pear fruit. Twenty-three phage isolates were able to significantly suppress the incidence of bacterial exudate on the pear disc surface. Quantification of the bacterial population remaining on the disc surface indicated that population reductions of up to 97% were obtainable by phage treatment, but that elimination of bacteria from the surface was not possible with this model system.

In natural systems phytoplankton interact with planktonic (free living) and attached epiphytic bacteria both synergistically and antagonistically. The specificity of the association with micro algae and bacteria differs in terms of adhesion mechanisms and metabolic cooperation. Present research was carried out to study the effect of bacterial isolates namely Bacillus sp. and Pseudomonas sp. from algal culture systems on the growth of micro algae such as Chaetoceros calcitrans and Nannochloropsis oculata. C. calcitrans (F= 15.34; P<0.05) and N. oculata (F=12.52; P<0.05) showed significantly higher growth, in treatments with Bacillus sp. and Pseudomonas sp when compared to control.

Im ersten Teil dieser Dissertation stand die Analyse der Motilitätsentwicklung bei Vertretern der Gattung Methylobacterium im Vordergrund. Diese zu den pink pigmentierten fakultativ methylotrophen Mikroorganismen (PPFMs) gehörenden Prokaryoten sind in der Umwelt weit verbreitet. Besonders häufig besiedeln die Mikroben pflanzliche Oberflächen und können als so genannte Phytosymbionten in einer wechselseitigen Beziehung zu pflanzlichen Organismen stehen. In aquatischer Umgebung können Methylobakterien Flagellen aufweisen. Hierbei handelt es sich um spezielle Fortbewegungsorganellen, die den Mikroben eine aktive Beweglichkeit ermöglichen. Die Ausbildung polarer Einzelflagellen bei Methylobacterium-Zellen in planktonischer Lebensweise konnte unter Anwendung verschiedener mikroskopischer Techniken dokumentiert werden. Quantitative Beweglichkeitsstudien zeigten einen charakteristischen Entwicklungsverlauf, korreliert mit den Wachstumsphasen der Bakterienkulturen und machten deutlich, dass die Motilitätsrate durch Umweltfaktoren, wie z. B. die Nährstoffversorgung, beeinflusst werden kann. Es konnte gezeigt werden, dass die Pflanzen-assoziierten PPFMs in der Lage sind, zwischen einer sessilen und planktonischen Lebensweise zu wechseln und dass sowohl die zelluläre Beweglichkeit als auch die Biofilm-Bildung der Prokaryoten ein reversibles, reaktivierbares Verhalten darstellt. Weiterhin konnte belegt werden, dass die Motilität der epiphytischen Mikroben bezüglich der Besiedelung von Pflanzen, z. B. bei der Ausbreitung auf Keimblatt-Oberflächen von Sonnenblumen (Helianthus annuus), keine zentrale Rolle spielt und eine endophytische Lebensweise unwahrscheinlich ist. Ziel der Arbeit war weiterhin die Charakterisierung und Identifizierung eines aus der Phyllosphäre der Echten Feige (Ficus carica, Standort Griechenland) isolierten Bakterien-Stammes (Mtb. sp. Fc1). Die fakultativ methylotrophe Stoffwechseleigenschaft, sowie die auffällige rötliche Pigmentierung belegen, dass es sich um einen Vertreter der PPFMs handelt. Die Analyse morphologischer, physiologischer und biochemischer Eigenschaften bestätigte in Übereinstimmung mit molekularphylogenetischen Untersuchungen zur Klassifizierung und taxonomischen Einordnung, dass es sich um Pflanzen-assoziierte Mikroben der Gattung Methylobacterium handelt. Analysen der 16S rDNA sowie partieller Sequenzen der für Methylobakterien etablierten Marker-Gene mxaF und gyrB verdeutlichten die phylogenetische Stellung und die evolutionären Beziehungen des Ficus-Isolates. Obwohl enge Verwandtschaftsverhältnisse zu anderen Methylobacterium-Arten ermittelt werden konnten, war eine Identifizierung als valide beschriebene Spezies nicht möglich. Die Resultate legen den Schluss nahe, dass es sich um eine neue, unbeschriebene Spezies der epiphytisch lebenden Methylobakterien handelt.

El foc bacterià és una malaltia que afecta a plantes de la família de la rosàcies, causada pel bacteri Erwinia amylovora. El seu rang d'hostes inclou arbres fruiters, com la perera, la pomera o el codonyer, i plantes ornamentals de gran interès comercial i econòmic. Actualment, la malaltia s'ha dispersat i es troba àmpliament distribuïda en totes les zones de clima temperat del món. A Espanya, on la malaltia no és endèmica, el foc bacterià es va detectar per primer cop al 1995 al nord del país (Euskadi) i posteriorment, han aparegut varis focus en altres localitzacions, que han estat convenientment eradicats. El control del foc bacterià, és molt poc efectiu en plantes afectades per la malaltia, de manera que es basa en mesures encaminades a evitar la dispersió del patogen, i la introducció de la malaltia en regions no endèmiques. En aquest treball, la termoteràpia ha estat avaluada com a mètode d'eradicació d'E. amylovora de material vegetal de propagació asimptomàtic. S'ha demostrat que la termoteràpia és un mètode viable d'eradicar E. amylovora de material de propagació. Gairebé totes les espècies i varietats de rosàcies mantingudes en condicions d'humitat sobrevivien 7 hores a 45 ºC i més de 3 hores a 50 ºC, mentre que més d'1 hora d'exposició a 50 ºC amb calor seca produïa danys en el material vegetal i reduïa la brotació. Tractaments de 60 min a 45 ºC o 30 min a 50 ºC van ser suficients per reduir la població epífita d'E. amylovora a nivells no detectables (5 x 102 ufc g-1 p.f.) en branques de perera. Els derivats dels fosfonats i el benzotiadiazol són efectius en el control del foc bacterià en perera i pomera, tant en condicions de laboratori, com d'hivernacle i camp. Els inductors de defensa de les plantes redueixen els nivells de malaltia fins al 40-60%. Els intervals de temps mínims per aconseguir el millor control de la malaltia van ser 5 dies pel fosetil-Al, i 7 dies per l'etefon i el benzotiadiazol, i les dosis òptimes pel fosetil-Al i el benzotiadiazol van ser 3.72 g HPO32- L-1 i 150 mg i.a. L-1, respectivament. Es millora l'eficàcia del fosetil-Al i del benzotiadiazol en el control del foc bacterià, quan es combinen amb els antibiòtics a la meitat de la dosi d'aquests últims. Tot i que l'estratègia de barrejar productes és més pràctica i fàcil de dur a terme a camp, que l'estratègia de combinar productes, el millor nivell de control de la malaltia s'aconsegueix amb l'estratègia de combinar productes. Es va analitzar a nivell histològic i ultrastructural l'efecte del benzotiadiazol i dels fosfonats en la interacció Erwinia amylovora-perera. Ni el benzotiadiazol, ni el fosetil-Al, ni l'etefon van induir canvis estructurals en els teixits de perera 7 dies després de la seva aplicació. No obstant, després de la inoculació d'E. amylovora es va observar en plantes tractades amb fosetil-Al i etefon una desorganització estructural cel·lular, mentre que en les plantes tractades amb benzotiadiazol aquestes alteracions tissulars van ser retardades. S'han avaluat dos models (Maryblyt, Cougarblight) en un camp a Espanya afectat per la malaltia, per determinar la precisió de les prediccions. Es van utilitzar dos models per elaborar el mapa de risc, el BRS-Powell combinat i el BIS95 modificat. Els resultats van mostrar dos zones amb elevat i baix risc de la malaltia. Maryblyt i Cougarblight són dos models de fàcil ús, tot i que la seva implementació en programes de maneig de la malaltia requereix que siguin avaluats i validats per un període de temps més llarg i en àrees on la malaltia hi estigui present.

To further our understanding of powdery mildew biology during infection, we undertook a systematic shotgun proteomics analysis of the obligate biotroph Blumeria graminis f. sp. hordei at different stages of development in the host. Moreover we used a proteogenomics approach to feed information into the annotation of the newly sequenced genome. We analyzed and compared the proteomes from three stages of development representing different functions during the plant-dependent vegetative life cycle of this fungus. We identified 441 proteins in ungerminated spores, 775 proteins in epiphytic sporulating hyphae, and 47 proteins from haustoria inside barley leaf epidermal cells and used the data to aid annotation of the B. graminis f. sp. hordei genome. We also compared the differences in the protein complement of these key stages. Although confirming some of the previously reported findings and models derived from the analysis of transcriptome dynamics, our results also suggest that the intracellular haustoria are subject to stress possibly as a result of the plant defense strategy, including the production of reactive oxygen species. In addition, a number of small haustorial proteins with a predicted N-terminal signal peptide for secretion were identified in infected tissues: these represent candidate effector proteins that may play a role in controlling host metabolism and immunity. We conducted the first molecular phylogenetic study of Ficus section Malvanthera (Moraceae; subgenus Urostigma) based on 32 Malvanthera accessions and seven outgroups representing other sections of Ficus subgenus Urostigma. We used DNA sequences from the nuclear ribosomal internal and external transcribed spacers (ITS and ETS), and the glyceraldehyde-3-phosphate dehydrogenase (G3pdh) region. Phylogenetic analysis using maximum parsimony, maximum likelihood and Bayesian methods recovered a monophyletic section Malvanthera to the exclusion of the rubber fig, Ficus elastica. The results of the phylogenetic analyses do not conform to any previously proposed taxonomic subdivision of the section and characters used for previous classification are homoplasious. Geographic distribution, however, is highly conserved and Melanesian Malvanthera are monophyletic. A new subdivision of section Malvanthera reflecting phylogenetic relationships is presented. Section Malvanthera likely diversified during a period of isolation in Australia and subsequently colonized New Guinea. Two Australian series are consistent with a pattern of dispersal out of rainforest habitat into drier habitats accompanied by a reduction in plant height during the transition from hemi-epiphytic trees to lithophytic trees and shrubs. In contradiction with a previous study of Pleistodontes phylogeny suggesting multiple changes in pollination behaviour, reconstruction of changes in pollination behaviour on Malvanthera, suggests only one or a few gains of active pollination within the section. The possibility that parents of one sex may preferentially invest in offspring of a certain sex raises profound evolutionary questions about the relative worth of sons and daughters to their mothers and fathers. Post-fledging brood division-in which cacti parent feeds a different subset of offspring-has been well documented in birds. However, a lack of empirical evidence that this may be based oil offspring sex, combined with the theoretical difficulty of explaining such an interaction, has led researchers to consider a gender bias in post-fledging brood division highly unlikely. Here we show that in the toc-toc, Foudia sechellarum, postfledging brood division is extreme and determined by sex; where brood composition allows, male parents exclusively provision male fledglings, whereas female parents provision female fledglings. This is the first study to provide unambiguous evidence, based on molecular sexing, that sex-biased post-fledging brood division can occur in birds. Male and female parents provisioned at the same rate and neither offspring nor parent survival appeared to be affected by the sex of the parent or offspring, respectively. The current hypotheses predicting advantages for brood division and preferential care for one specific type of offspring are discussed in the light of our results.

While only about 1-200 species are used intensively in commercial floriculture (e.g. carnations, chrysanthemums, gerbera, narcissus, orchids, tulips, lilies, roses, pansies and violas, saintpaulias, etc.) and 4-500 as house plants, several thousand species of herbs, shrubs and trees are traded commercially by nurseries and garden centres as ornamentals or amenity species. Most of these have been introduced from the wild with little selection or breeding. In Europe alone, 12 000 species are found in cultivation in general garden collections (i.e. excluding specialist collections and botanic gardens). In addition, specialist collections (often very large) of many other species and/or cultivars of groups such as orchids, bromeliads, cacti and succulents, primulas, rhododendrons, conifers and cycads are maintained in several centres such as botanic gardens and specialist nurseries, as are 'national collections' of cultivated species and cultivars in some countries. Specialist growers, both professional and amateur, also maintain collections of plants for cultivation, including, increasingly, native plants. The trade in ornamental and amenity horticulture cannot be fully estimated but runs into many billions of dollars annually and there is considerable potential for further development and the introduction of many new species into the trade. Despite this, most of the collections are ad hoc and no co-ordinated efforts have been made to ensure that adequate germplasm samples of these species are maintained for conservation purposes and few of them are represented at all adequately in seed banks. Few countries have paid much attention to germplasm needs of ornamentals and the Ornamental Plant Germplasm Center in conjunction with the USDA National Plant Germplasm System at The Ohio State University is an exception. Generally there is a serious gap in national and international germplasm strategies, which have tended to focus primarily on food plants and some forage and industrial crops. Adequate arrangements need to be put in place to ensure the long- and medium-term conservation of representative samples of the genetic diversity of ornamental species. The problems of achieving this will be discussed. In addition, a policy for the conservation of old cultivars or 'heritage' varieties of ornamentals needs to be formulated. The considerable potential for introduction of new ornamental species needs to be assessed. Consideration needs to be given to setting up a co-ordinating structure with overall responsibility for the conservation of germplasm of ornamental and amenity plants.

To further our understanding of powdery mildew biology during infection, we undertook a systematic shotgun proteomics analysis of the obligate biotroph Blumeria graminis f. sp. hordei at different stages of development in the host. Moreover we used a proteogenomics approach to feed information into the annotation of the newly sequenced genome. We analyzed and compared the proteomes from three stages of development representing different functions during the plant-dependent vegetative life cycle of this fungus. We identified 441 proteins in ungerminated spores, 775 proteins in epiphytic sporulating hyphae, and 47 proteins from haustoria inside barley leaf epidermal cells and used the data to aid annotation of the B. graminis f. sp. hordei genome. We also compared the differences in the protein complement of these key stages. Although confirming some of the previously reported findings and models derived from the analysis of transcriptome dynamics, our results also suggest that the intracellular haustoria are subject to stress possibly as a result of the plant defense strategy, including the production of reactive oxygen species. In addition, a number of small haustorial proteins with a predicted N-terminal signal peptide for secretion were identified in infected tissues: these represent candidate effector proteins that may play a role in controlling host metabolism and immunity. DNA barcodes could be a useful tool for plant conservation. Of particular importance is the ability to identify unknown plant material, such as from customs seizures of illegally collected specimens. Mexican cacti are an example of a threatened group, under pressure because of wild collection for the xeriscaping trade and private collectors. Mexican cacti also provide a taxonomically and geographically coherent group with which to test DNA barcodes. Here, we sample the matK barcode for 528 species of Cactaceae including approximately 75% of Mexican species and test the utility of the matK region for species-level identification. We find that the matK DNA barcode can be used to identify uniquely 77% of species sampled, and 79-87% of species of particular conservation importance. However, this is far below the desired rate of 95% and there are significant issues for PCR amplification because of the variability of primer sites. Additionally, we test the nuclear ITS regions for the cactus subfamily Opuntioideae and for the genus Ariocarpus (subfamily Cactoideae). We observed higher rates of variation for ITS (86% unique for Opuntioideae sampled) but a much lower PCR success, encountering significant intra-individual polymorphism in Ariocarpus precluding the use of this marker in this taxon. We conclude that the matK region should provide useful information as a DNA barcode for Cactaceae if the problems with primers can be addressed, but matK alone is not sufficiently variable to achieve species-level identification. Additional complementary regions should be investigated as ITS is shown to be unsuitable

More than 30 epiphytic lichens, collected in Agadir (Morroco) and along a 150-km transect from the Atlantic Ocean eastward, were analyzed for their metal content and lead isotopic composition. This dataset was used to evaluate atmospheric metal contamination and the impact of the city on the surrounding area. The concentrations of Cu, Pb, and Zn (average ± 1 SD) were 20.9 ± 15.2 μg g−1, 13.8 ± 9.0 μg g−1, and 56.6 ± 26.6 μg g−1, respectively, with the highest values observed in lichens collected within the urban area. The 206Pb/207Pb and 208Pb/207Pb ratios in the lichens varied from 1.146 to 1.186 and from 2.423 to 2.460, respectively. Alkyllead-gasoline sold in Morocco by the major petrol companies gave isotopic ratios of 206Pb/207Pb = 1.076–1.081 and 208Pb/207Pb = 2.348–2.360. These new, homogeneous values for gasoline-derived lead improve and update the scarce isotopic database of potential lead sources in Morocco, and may be of great value to future environmental surveys on the presence of lead in natural reservoirs, where it persists over time (e.g., soils and sediments). The interest of normalizing metal concentrations in lichens to concentrations of a lithogenic element is demonstrated by the consistency of the results thus obtained with lead isotopic ratios. Leaded gasoline contributed less than 50% of the total amount of lead accumulated in lichens, even in areas subject to high vehicular traffic. This strongly suggests that the recent banishment of leaded gasoline in Morocco will not trigger a drastic improvement in air quality, at least in Agadir.
