A resistência aos antibióticos em bactérias Gram-negativas pode ser aumentada pela extrusão de antibióticos através de sistemas de efluxo. Em Escherichia coli, o principal sistema de efluxo é o AcrAB-TolC o qual tem como principal fonte energética a força proto-motriz. Este trabalho pretendeu estudar alguns aspectos essenciais da bioenergética na actividade de efluxo de E. coli usando três estirpes bem caracterizadas genotipica e fenotipicamente. Foi utilizado um método fluorimétrico semi-automático no qual a fluorescência do fluorocromo brometo de etídeo, substrato de bombas de efluxo foi seguida, permitindo a medição em tempo real da actividade de efluxo e acumulação de fluorocromo (inibição do efluxo). A utilização de brometo de etídeo é particularmente vantajosa pois emite baixa fluorescência no exterior da célula bacteriana tornando-se extremamente fluorescente no seu interior. Este método é uma nova aplicação do termociclador em tempo real RotorGeneTM 3000 que permite o cálculo da cinética de transporte reflectindo o balanço entre acumulação de substrato por difusão passiva através da membrana e a sua extrusão/efluxo, proporcionando uma detecção rápida e económica de inibidores de efluxo. Os resultados obtidos mostram, para todas as estirpes, que a GLU e o pH afectam a acumulação e o efluxo do brometo de etídeo. De todos os inibidores de vias biossintéticas testados, o ortovanadato de sódio, foi o que demonstrou maior actividade inibitória, a qual é revertida na presença de GLU. Em conclusão, este estudo mostra que a actividade de efluxo de E. coli depende não só da fosforilação oxidativa por via da força proto-motriz mas também da energia proveniente da hidrólise de ATP pelas ATPases. O ortovanadato de sódio tem potencial para ser um novo inibidor de bombas de efluxo de largo espectro. A tecnologia utilizada neste trabalho demonstrou ser apropriada para a caracterização bioenergética da actividade de bombas de efluxo e permite a selecção de novos inibidores de bombas de efluxo em bactérias.

O presente trabalho, desenvolvido nas instalações da FISIPE, teve como objectivo a caracterização de fibra oxidada (FOX),actualmente em desenvolvimento, com vista ao controlo de processo e de qualidade, e centrou-se em quatro parâmetros: i. Densidade; ii. Índice de Aromaticidade; iii. Índice de Oxigénio Limitante; iv. Grupos funcionais. No estudo da densidade realizou-se: a. Consolidação do método actualmente usado na empresa, observando-se uma redução da incerteza para metade (±0,005 gcm-3). b. Análise de diferentes amostras de FOX que apresentaram valores entre 1,348-1,387 gcm-3, que atendendo à incerteza do método, se encontram dentro dos valores de referência (1,35-1,39 gcm-3). c. Previsão de densidades ao longo da etapa de estabilização oxidativa. Confirmou-se ser apenas possível pela expressão apresentada numa das fontes consultada. Para o Índice de Aromaticidade (AI) efectuou-se: a. Estudo de quatro métodos de integração de curvas de DSC para determinação do calor de combustão que permite determinar o AI. Verificou-se que o mais indicado seria o uso da temperatura de on-set como limite inferior à linha de base de integração. b. Análise de diferentes amostras de FOX, observando-se valores de AI entre os 66% e 81%. Na determinação do Índice de Oxigénio Limitante, parâmetro exclusivo de qualidade, analisaram-se amostras de cabo pela adaptação da norma ASTM D 2863-06a. Obtiveram-se valores acima dos 40%O2 para a fibra oxidada. Para a verificação dos grupos funcionais realizou-se: a. Análise de FTIR, confirmando-se o desaparecimento de grupos nitrilo em função do tempo de oxidação, e a formação de bandas conjugadas e sobrepostas, indicando a presença de estruturas ciclizadas e aromatizadas. b. Proposta de um mecanismo reaccional para a reacção de oxidação de uma determinada fibra precursora. Com o intuito de minimizar o tempo de caracterização das FOX, procurou-se ainda encontrar uma correlação entre a densidade e o Índice de Aromaticidade, tanto ao longo da etapa de oxidação, como da fibra oxidada final. Conclui-se que não é possível estabelecer uma relação matemática para as amostras analisadas.

En aquest treball es caracteriza per primera vegada la capacitat de coordinació metàl·lica d'una metal·lotineïna (MT) de planta i es proposa un model de plegament per a les MTs de planta en general. Els resultat mostren que aquestes proteïnes poden tenir un paper molt important en la regulació de l'estat redox de les cèl·lules, probablement a través de la coordinació a Cu. Les MTs de planta són proteïnes molt desconegudes. Es postula que participen en l'homeòstasi del Cu i en la protecció contra l'estrès oxidatiu, però es desconeix la capacitat de coordinació metàl·lica i el plegament. En aquest treball s'han estudiat una metal·lotioneïna d'alzina surera, QsMT, aïllada d'una llibreria de cDNA de fel·lema. Els objectius concrets han estat: (1) estudiar l'expressió de QsMT i la resposta a l'estrès oxidatiu; (2) determinar la capacitat de coordinació metàl·lica i la funcionalitat in vivo; (3) fer una aproximació al plegament de les MTs de planta. L'expressió del gen s'ha estudiat mitjançant hibridació in situ en plàntules i en embrions d'alzina surera. QsMT s'expressa majoritàriament en cèl·lules amb fort estrès oxidatiu, associat a la síntesi de polifenols (suberització i lignificació) i a la senescència. També s'expressa en cèl·lules meristemàtiques, cèl·lules en divisió molt activa on la funció de les MTs podria estar relacionada amb el manteniment de l'estat redox. L'aplicació d'estrès oxidatiu exogen (H2O2 i paraquat) incrementa fortament l'expressió de QsMT en teixits amb expressió constitutiva, confirmant la regulació de l'expressió del gen per estrès oxidatiu. Per l'estudi de les propietats de coordinació metàl·lica es va expressar QsMT en cèl·lules d'E. coli en medi de cultiu suplementat amb Cu, Zn o Cd. Es van aïllar els agregats metàl·lics corresponents i es van analitzar mitjançant tècniques espectroscòpiques i espectromètriques (ICP-OES, ESI-MS i CD). Els resultats mostren que QsMT coordina de forma estable Cu (8 ions metàl·lics/molècula), Zn (4 ions de Zn/molècula) i Cd (6 ions de Cd/molècula), i adopta una estructura especialment quiral en coordinació a Cu. L'elevada capacitat quelant de la proteïna i la quiralitat de l'estructura indiquen que QsMT possiblement té preferència metàl·lica pel Cu i per tant una funció relacionada amb aquest metall in vivo. Estudis de complementació en llevat demostren que QsMT coordina Cu de forma funcional in vivo. En coordinació a Cd QsMT presenta una peculiaritat no observada fins ara en altres MTs: la participació d'ions sulfur en la formació de l'agregat metàl·lic incrementant la capacitat de coordinació metàl·lica (6 ions metàl·lics divalents de Cd enlloc de 4 ions de Zn). A més QsMT coordina Cd de forma funcional en llevat, i per tant la seva funció també podria estar relacionada amb la destoxicació de Cd en la planta. QsMT s'ha utilitzat com a model per fer una aproximació al plegament de les MTs de planta. Amb aquest objectiu vam dissenyar tres pèptids mutants derivats de QsMT: N25 corresponent a la zona rica en cisteïna en posició amino-terminal, C18 corresponent a la zona rica en cisteïna en posició carboxil-terminal, i N25-C18 corresponent a les dues zones riques en cisteïna enllaçades per 4 glicines substituint la zona central de 39 aminoàcids. Es van expressar i estudiar aquests pèptids per les mateixes tècniques utilitzades en l'estudi de QsMT. Els resultats indiquen que QsMT es plega formant un sol agregat metàl·lic per la interacció de les dues zones riques en cisteïna. En aquest model la zona central d'enllaç, típica de les MTs de planta, no participa en la coordinació metàl·lica però és imprescindible per a la funció de la proteïna. El paper de la zona central podria variar en funció del metall que coordina, participant en el plegament i estructura de la proteïna quan coordina Zn i Cd i en la seva regulació i estabilització quan coordina Cu.

Los fenómenos oxidativos y en particular la oxidación lipídica son uno de los principales responsables de la pérdida de calidad en la carne y en los productos cárnicos. Como consecuencia de estos procesos se generan compuestos que pueden afectar el flavor, color y textura de la carne disminuyendo la aceptabilidad por parte del consumidor y reduciendo su valor nutritivo. Por otro lado, el estrés oxidativo está relacionado con la etiología de diversas enfermedades comunes en nuestra sociedad. Las carnes de pollo y de pavo son particularmente sensibles a los procesos oxidativos debido a su elevada proporción de ácidos grasos poliinsaturados en comparación con otros tipos de carne. La suplementación de antioxidantes en la dieta de determinados animales es una de las estrategias más eficaces para proteger la carne de la oxidación. Otro aspecto que afecta a la calidad y seguridad de la carne es la presencia de residuos en los tejidos animales destinados al consumo humano, una parte de los cuáles puede proceder de la administración de antibióticos. En este trabajo se estudió la eficacia de tres compuestos antioxidantes, alfa-tocoferol, beta-caroteno y licopeno, adicionados en distintas concentraciones y combinaciones a la dieta de pollos y pavos. Para ello se determinó la estabilidad oxidativa de los tejidos musculares de pechuga y muslo mediante el análisis de los valores de TBARS, de las actividades de los enzimas antioxidantes GSHPx, CAT y SOD y desde un punto de vista sensorial. Asimismo, se analizaron las concentraciones de vitamina E presentes en ambos músculos. Por otro lado, se investigó la presencia de residuos del antibiótico enrofloxacina y de su metabolito en los tejidos muscular y hepático de ambas especies después de la administración del fármaco con o sin periodo de retirada. Finalmente, y dada la aparente relación existente entre el metabolismo de determinados antibióticos y los fenómenos oxidativos, se valoró la posible interacción entre el fármaco y la vitamina E suplementada a la dieta. La vitamina E, a dosis de 100 ppm y 200 ppm en pollos y pavos respectivamente, se comportó como un antioxidante eficaz disminuyendo la rancidez de la carne tanto en pechuga como en muslo. La dosis de vitamina E necesaria para conseguir un incremento significativo de la estabilidad oxidativa de la carne varió en función de la especie y de las características bioquímicas del tejido analizado. El beta-caroteno, suplementado en la dieta de pollos y pavos conjuntamente con la vitamina E, no sólo no manifestó propiedades antioxidantes sino que enmascaró la efectividad de la vitamina E. El licopeno, de cuya utilización en nutrición animal no existían estudios publicados anteriormente, no mostró eficacia antioxidante en la carne de pollo a una dosis de 10 ppm. Respecto al análisis de residuos de antibiótico se observó que tras el periodo de retirada del fármaco los niveles residuales de enrofloxacina y su metabolito disminuyeron notablemente. Debe tenerse en cuenta que se apreciaron diferencias en función de la especie y del tejido considerados, estando los residuos en algunos casos por encima de los límites máximos permitidos. Por otro lado, se observó una relación entre la enrofloxacina y la vitamina E suplementada en la dieta que, parecía depender tanto de la dosis de antioxidante como del metabolismo del fármaco. Esta interacción afectó tanto a los niveles de vitamina E como a la presencia de residuos de enrofloxacina en el tejido muscular, resaltando la importancia de no subestimar posibles interacciones entre distintos compuestos presentes en la dieta animal.

The [2+2+2] cycloaddition reaction involves the formation of three carbon-carbon bonds in one single step using alkynes, alkenes, nitriles, carbonyls and other unsaturated reagents as reactants. This is one of the most elegant methods for the construction of polycyclic aromatic compounds and heteroaromatic, which have important academic and industrial uses. The thesis is divided into ten chapters including six related publications. The first study based on the Wilkinson’s catalyst, RhCl(PPh3)3, compares the reaction mechanism of the [2+2+2] cycloaddition process of acetylene with the cycloaddition obtained for the model of the complex, RhCl(PH3)3. In an attempt to reduce computational costs in DFT studies, this research project aimed to substitute PPh3 ligands for PH3, despite the electronic and steric effects produced by PPh3 ligands being significantly different to those created by PH3 ones. In this first study, detailed theoretical calculations were performed to determine the reaction mechanism of the two complexes. Despite some differences being detected, it was found that modelling PPh3 by PH3 in the catalyst helps to reduce the computational cost significantly while at the same time providing qualitatively acceptable results. Taking into account the results obtained in this earlier study, the model of the Wilkinson’s catalyst, RhCl(PH3)3, was applied to study different [2+2+2] cycloaddition reactions with unsaturated systems conducted in the laboratory. Our research group found that in the case of totally closed systems, specifically 15- and 25-membered azamacrocycles can afford benzenic compounds, except in the case of 20-membered azamacrocycle (20-MAA) which was inactive with the Wilkinson’s catalyst. In this study, theoretical calculations allowed to determine the origin of the different reactivity of the 20-MAA, where it was found that the activation barrier of the oxidative addition of two alkynes is higher than those obtained for the 15- and 25-membered macrocycles. This barrier was attributed primarily to the interaction energy, which corresponds to the energy that is released when the two deformed reagents interact in the transition state. The main factor that helped to provide an explanation to the different reactivity observed was that the 20-MAA had a more stable and delocalized HOMO orbital in the oxidative addition step. Moreover, we observed that the formation of a strained ten-membered ring during the cycloaddition of 20-MAA presents significant steric hindrance. Furthermore, in Chapter 5, an electrochemical study is presented in collaboration with Prof. Anny Jutand from Paris. This work allowed studying the main steps of the catalytic cycle of the [2+2+2] cycloaddition reaction between diynes with a monoalkyne. First kinetic data were obtained of the [2+2+2] cycloaddition process catalyzed by the Wilkinson’s catalyst, where it was observed that the rate-determining step of the reaction can change depending on the structure of the starting reagents. In the case of the [2+2+2] cycloaddition reaction involving two alkynes and one alkene in the same molecule (enediynes), it is well known that the oxidative coupling may occur between two alkynes giving the corresponding metallacyclopentadiene, or between one alkyne and the alkene affording the metallacyclopentene complex. Wilkinson’s model was used in DFT calculations to analyze the different factors that may influence in the reaction mechanism. Here it was observed that the cyclic enediynes always prefer the oxidative coupling between two alkynes moieties, while the acyclic cases have different preferences depending on the linker and the substituents used in the alkynes. Moreover, the Wilkinson’s model was used to explain the experimental results achieved in Chapter 7 where the [2+2+2] cycloaddition reaction of enediynes is studied varying the position of the double bond in the starting reagent. It was observed that enediynes type yne-ene-yne preferred the standard [2+2+2] cycloaddition reaction, while enediynes type yne-yne-ene suffered β-hydride elimination followed a reductive elimination of Wilkinson’s catalyst giving cyclohexadiene compounds, which are isomers from those that would be obtained through standard [2+2+2] cycloaddition reactions. Finally, the last chapter of this thesis is based on the use of DFT calculations to determine the reaction mechanism when the macrocycles are treated with transition metals that are inactive to the [2+2+2] cycloaddition reaction, but which are thermally active leading to new polycyclic compounds. Thus, a domino process was described combining an ene reaction and a Diels-Alder cycloaddition.

O estresse oxidativo tem um papel importante no desenvolvimento da falência orgânica múltipla e choque séptico. O presente estudo foi realizado para determinar diferentes parâmetros de dano oxidativo a biomoléculas, produção de superóxido mitocondrial, atividades da superóxido dismutase e catalase e suas relações com a mortalidade por sepse em um modelo animal. Além disso, nós avaliamos os efeitos da combinação de antioxidantes (N-acetilcisteína e deferoxamina) em um modelo murino de sepse polimicrobiana induzida por ligação cecal e punção (CLP). Ratos Wistar machos (250-300 g) foram randomicamente divididos em quatro grupos: sham-operated, CLP ressuscitado com solução salina (50 ml/Kg imediatamente e 12h após CLP), CLP ressuscitado com solução salina e antibioticoterapia (solução salina 50 ml/ Kg imediatamente e 12h após CLP + ceftriaxone 30 mg/Kg e clindamicina 25 mg/Kg de 6/6h), e CLP com N-acetilcisteína (20 mg/kg subcutâneo, 3h, 6h, 12h, 18h e 24h após CLP) + deferoxamina (20mg/kg, subcutâneo, 3h e 24h após CLP). A peroxidação lipídica, a carbonilação protéica e a atividade da superóxido dismutase se apresentaram significativamente aumentadas nos animais não-sobreviventes podendo predizer a mortalidade. Nós demonstramos que existe uma modulação diferente da superóxido dismutase e da catalase nos animais não-sobreviventes Existe um grande aumento da atividade da superóxido dismutase sem um aumento proporcional da atividade da catalase nos não-sobreviventes. Os ratos tratados com antioxidantes apresentaram uma redução significante da atividade da mieloperoxidase e da peroxidação lipídica em todos os órgãos estudados. A produção de superóxido mitocondrial apresentou uma redução significante nos animais tratados com antioxidantes. Além disto, o tratamento com antioxidantes melhorou o equilíbrio entre a atividade da catalase e superóxido dismutase. A sobrevivências nos ratos sépticos não tratados foi de 10%. A sobrevivência aumentou para 40% com a ressuscitação volêmica e antibióticos. Ratos tratados apenas con N-acetilcisteína e desferoxamina apresentaram uma sobrevivência de 47%, similar aos ratos que receberam suporte básico. Este estudo demonstrou pela primeira vez que a atividade da superóxido dismutase deve ser um marcador precoce de mortalidade em sepse. Nossos resultados auxiliam o entendimento de aspectos importantes da resposta oxidativa na sepse; um aumento na atividade da superóxido dismutase sem um aumento proporcional na atividade da catalase. Além disto, nossos resultados demonstraram que o tratamento combinado com N-acetilcisteína e desferoxamina reduzem as conseqüências da sepse, reduzindo o estresse oxidativo, limitando a ativação de neutrófilos e a disfunçào mitocondrial, levando a uma melhor sobrevida.

Biodiesel is a fuel obtained from vegetable oils, such as soy, castorbean, among others. The monoester of fatty acid of these oils have chains with mono, di and tri double connections. The presence of these insaturations are susceptible to oxidization. Antioxidants are substances able to prevent oxidization from oils, fats, fat foods, as well as esters of Alquila( biodiesel). The objective of this work is to summarize a new antioxidant from the Cashew Nut Shell Liquid (CNSL) using the electrolysis technique. A current of 2 amperes was used in a single cell of only one group and two eletrodos of stainless steel 304 in a solution of methanol, together with the eletrolits: acetic acid, sodium chloride and sodium hydroxide, for two hours of agitation. The electrolysis products are characterized by the techniques of cromatography in a thin layer, spectroscopy of infrared and gravimetric analysis. The material was submitted to tests of oxidative stability made by the techniques of spectropy of impendancy and Rancimat (EN 14112). The analyses of characterization suggest that the polimerization of the electrolytic material ocurred. The application results of these materials as antioxidants of soy biodiesel showed that the order of the oxidative stability was obtained by both techniques used

There are strong interests in the evaluation of the biological effects of natural and synthetic products. Blood constituents labeled with technetium-99m (99mTc) are used in nuclear medicine. The aim of this work was to study the effects of Clove (Caryophyllus aromaticas L.) and OZE (preparation used in the Health Sciences) on the labeling blood constituents with 99mTc and on the morphologic red blood cells (RBC) and the action of an extract of tomato (TO) on the labeling of blood constituents Blood samples were incubated with clove or OZE or TO, stannous chloride and 99mTc. Plasma (P), blood cells (BC), insoluble fractions (IF) of plasma and blood cells were separated. The radioactivity was counted and percentage of radioactivity (%ATI) to each blood fraction was calculated. The shape and morphometric parameter (perimeter/area ratio) were evaluated in the studies with clove and OZE. Clove extract and OZE altered significantly (p<0.05) the %ATI of blood constituents and the shape of red blood cells. However, clove extract not altered the red blood cells perimeter/area ratio. The tomato extract used at the highest concentrations reduced significantly (p<0.05) the %ATI in IF-P, although this extract did not modify the radiolabeling on BC, neither the radioactivity fixation on IFBC. The results indicate that these chemical compounds would have oxidative/chelating actions

The lubricants found in the market are of mineral or synthetic origin and harm to humans and the environment, mainly due to their improper discard. Therefore industries are seeking to develop products that cause less environmental impact, so to decrease mainly, operator aggression the Cutting Fluids became an emulsion of oil / water or water / oil. However, the emulsion was not considered the most suitable solution for environmental question, therefore the search for biodegradable lubricants and which no are toxic continues and so vegetable oils are seen, again, as a basis for the production of lubricants. The biggest problem with these oils is their oxidative instability that is intensified when working at high temperatures. The process transesterification decreases the oxidation, however changes some physical and chemical properties. Therefore soybean oil after the transesterification process was subjected to tests of density, dynamic viscosity, kinematic viscosity which is calculated from two parameters mentioned, flash point and acidity. Besides the physico-chemical test the soybean oil was subjected to a dynamic test in a tribometer adapted from a table vise, whose induced wear was the adhesive and ultimately was used as cutting fluid in a process of turning in two different materials, steel 1045 and cast iron. This latter test presented results below the mineral cutting fluid which it was compared in all tests, already in other experiments the result was satisfactory and other experiments not, so that chemical additives can be added to the oil analyzed to try equate all parameters and so formulate a biolubrificante not toxic to apply in machining processes of metalworking industry

The biodiesel use has become important due to its renewable character and to reduce environmental impacts during the fuel burning. Theses benefit will be valid if the fuel shows good performance, chemistry stability and compatibility with engines. Biodiesel is a good fuel to diesel engines due to its lubricity. Then, the aimed of this study was to verify the physicalchemistry properties of biodiesel and their correlations with possible elastomers damage after biodiesel be used as fuel in an injection system. The methodology was divided in three steps: biodiesels synthesis by transesterification of three vegetable oil (soybean, palm and sunflower) and their physical-chemistry characterization (viscosity, oxidative stability, flash point, acidity, humidity and density); pressurized test of compatibility between elastomers (NBR and VITON) and biodiesel, and the last one, analyze of biodiesels lubricity by tribological test ball-plan( HFRR). Also, the effect of mixture of biodiesel and diesel in different concentrations was evaluated. The results showed that VITON showed better compatibility with all biodiesel blends in relation to NBR, however when VITON had contact with sunflower biodiesel and its blends the swelling degree suffer higher influences due to biodiesel humidity. For others biodiesels and theirs blends, this elastomer kept its mechanical properties constant. The better tribological performance was observed for blends with high biodiesel concentration, lower friction coefficient was obtained when palm biodiesel was used. Natural gas, although basically composed by light hydrocarbons, also presents in its composition gaseous contaminants such as CO2 (carbon dioxide) and H2S (hydrogen sulfide). Hydrogen sulfide, which commonly occurs in oil and gas exploration and production activities, besides being among the gases that are responsible by the acid rain and greenhouse effect, can also cause serious harm to health, leading even to death, and damages to oil and natural gas pipelines. Therefore, the removal of hydrogen sulfide will significantly reduce operational costs and will result in oil with best quality to be sent to refinery, thereby resulting in economical, environmental, and social benefits. These factors highlight the need for the development and improvement of hydrogen sulfide sequestrating agents to be used in the oil industry. Nowadays there are several procedures for hydrogen sulfide removal from natural gas used by the petroleum industry. However, they produce derivatives of amines that are harmful to the distillation towers, form insoluble precipitates that cause pipe clogging and produce wastes of high environmental impact. Therefore, the obtaining of a stable system, in inorganic or organic reaction media, that is able to remove hydrogen sulfide without forming by-products that affect the quality and costs of natural gas processing, transport and distribution is of great importance. In this context, the evaluation of the kinetics of H2S removal is a valuable procedure for the treatment of natural gas and disposal of the byproducts generated by the process. This evaluation was made in an absorption column packed with Raschig ring, where natural gas with H2S passes through a stagnant solution, being the contaminant absorbed by it. The content of H2S in natural gas in column output was monitored by an H2S analyzer. The comparison between the obtained curves and the study of the involved reactions have not only allowed to determine the efficiency and mass transfer controlling step of the involved processes but also make possible to effect a more detailed kinetic study and evaluate the commercial potential of each reagent

The constant search for biodegradable materials for applications in several fields shows that carnauba wax can be a viable alternative in the manufacturing of biolubricants. Carnauba wax is the unique among the natural waxes to have a combination of properties of great importance. In previous studies it was verified the presence of metals in wax composition that can harm the oxidative stability of lubricants. Considering these factors, it was decided to develop a research to evaluate iron removal from carnauba wax, using microemulsion systems (Me) and perform the optimization of parameters, such as: extraction pH, temperature, extraction time, among others. Iron concentration was determined by atomic absorption and, to perform this analysis, sample digestion in microwave oven was used, showing that this process was very efficient. It was performed some analysis in order to characterize the wax sample, such as: attenuated total reflectance infrared spectroscopy (ATR-IR), thermogravimetry (TG), differential scanning calorimetry (DSC), energy dispersive X-ray fluorescence (EDXRF), scanning electron microscopy (SEM) and melting point (FP). The microemulsion systems were composed by: coconut oil as surfactant, n-butanol as cosurfactant, kerosene and/or heptanes as oil phase, distilled water as water phase. The pH chosen for this study was 4.5 and the metal extraction was performed in finite experiments. To evaluate Me extraction it was performed a factorial design for systems with heptane and kerosene as oil phase, also investigating the influence of temperature time and wax/Me ratio, that showed an statistically significant answer for iron extraction at 95% confidence level. The best result was obtained at 60°C, 10 hours contact time and 1: 10 wax/Me ratio, in both systems with kerosene and heptanes as oil phase. The wet oxidation of organic compounds with CO2 and H2O has been demonstrated to be an efficient technique for effluent treatment. This work focuses on the synthesis, characterization and catalytic performance of Fe-MnO2/CeO2, K-MnO2/CeO2/ palygorskite and Fe/ palygorskite toward the wet oxidative degradation of phenol. The experiments were conducted in a sludge bed reactor with controlled temperature, pressure and stirring speed and sampling of the liquid phase. Experiments were performed on the following operating conditions: temperature 130 ° C, pressure 20.4 atm, catalyst mass concentration of 5 g / L initial concentration of phenol and 0.5 g / L. The catalytic tests were performed in a slurry agitated reactor provided with temperature, pressure and agitation control and reactor liquid sampling. The influences of iron loaded on the support (0.3; 7 and 10%, m/m) and the initial pH of the reactant medium (3.1; 6.8; 8.7) were studied. The iron dispersion on the palygorskite, the phase purity and the elemental composition of the catalyst were evaluated by X-Ray Difraction (XRD), Scanning Electron Microscopy (SEM) and X-Ray Flourescence (XRF). The use of palygorskite as support to increase the surface area was confirmed by the B.E.T. surface results. The phenol degradation curves showed that the Fe3+ over palygorskite when compared with the other materials tested has the best performance toward the (Total Organic carbonic) TOC conversion. The decrease in alkalinity of the reaction medium also favors the conversion of TOC. The maximum conversion obtained from the TOC with the catalyst 3% Fe / palygorskite was around 95% for a reaction time of 60 minutes, while reducing the formation of acids, especially acetic acid. With products obtained from wet oxidation of phenol, hydroquinone, p-benzoquinone, catechol and oxalic acid, identified and quantified by High Performance Liquid Chromatography was possible to propose a reaction mechanism of the process where the phenol is transformed into the homogeneous and heterogeneous phase in the other by applying a kinetic model, Langmuir-Hinshelwood type, with evaluation of kinetic constants of different reactions involved.

The chemical nature of the hidrocarbons found in gasoline directly affects the formation of oxidation complexes known as gums. Such complexes are slightly soluble in gasoline and their formation is delayed with the use of inhibitors, known as antioxidants. In this study, a synthetic compound (βnaphtol) and a natural subproduct (hydrogenated cardanol, derived from cashewnut shell liquid CNSL) have been used in order to generate novel antioxidant substances. These compounds were submitted to chemical reactions including alkylation, nitration and reduction, with the purpose of forming the following derivatives: 6(Nethyl,Nethylamino)βnaphtol (AO1); 6(Nethyl,Ndiethylamino)βnaphtol (AO2); aminoβnaphtol (AO3); 2(Nethyl,Nethylamino)pentadecylphenol (AOC1), 2(Nethyl,Ndiethylamino)pentadecylphenol (AOC2) and aminopentadecylphenol (AOC3). The derivatives were subjected to accelerated oxidative stability assays (Potential Gum and Induction Period) and to storage assays (Washed Gum and ASTM Color) during six months, with naphtha provided by the petroleum refinery RPBC (Refinaria Presidente Bernardes de Cubatão, in Brazil). The results for the derivatives were compared to those for commercial additives [DBPC (2,6ditbutyl4methylpcresol) and PDA (N,N disecbutylpphenylenediamine)], which were also added to the naphtha produced at RPBC at the moment of sampling. From all tested antioxidants, the novel antioxidant AOC1 (derived from hydrogenated cardanol) yielded a better global performance. During the period of time in which the naphtha was stored, an examination of this material was carried out in parallel, using the mass spectrometry technique. This study allowed to monitor the formation of a triolefinic compound, as well as the observation of subsequent formation and rupture of the olefinic constituents. As an eventual result from these experimental investigations, a reaction route leading to gum formation has been suggested

Este estudo teve por objetivo analisar as alterações histológicas, histoquímicas e morfométricas das fibras do músculo sóleo de ratos submetidos a um programa de natação, associado ou não à administração do esteróide anabólico decanoato de nandrolona. Foram utilizados 22 ratos Wistar machos, 12 dos quais receberam injeção intramuscular do esteróide (5mg/kg) e 10, óleo mineral (5mg/kg), duas vezes por semana. Os animais foram submetidos a 42 sessões de natação por nove semanas (de segunda a sexta-feira), com aumento progressivo de carga por meio do tempo de natação. Após o sacrifício, o músculo sóleo esquerdo foi retirado, imerso em n-hexana e acondicionado em nitrogênio líquido. Cortes do terço médio desse músculo foram feitos em micrótomo criostato (-20ºC) e corados pela técnica HE e pelo método histoquímico NADH-TR. Os animais submetidos a treinamento físico e a esteróide (TA) ou óleo mineral (TO) apresentaram fibras musculares com maior diâmetro, quando comparados com os animais-controle (NTA e NTO). Não houve diferença significativa entre as medidas das médias dos diâmetros das fibras dos grupos NTA e NTO e entre TA e TO. Nos grupos TA e NTA notou-se acentuado processo de fagocitose, arredondamento e hialinização das fibras musculares. Já nos grupos TA, TO e NTA observou-se perda da atividade enzimática oxidativa. Os resultados sugerem que a natação produz hipertrofia muscular de forma semelhante, tanto no grupo que recebeu esteróide como no que recebeu óleo mineral. No entanto, o grupo que recebeu esteróide apresentou sinais claros de maior degeneração muscular.
