Conceptual database design is an unusually difficult and error-prone task for novice designers. This study examined how two training approaches---rule-based and pattern-based---might improve performance on database design tasks. A rule-based approach prescribes a sequence of rules for modeling conceptual constructs, and the action to be taken at various stages while developing a conceptual model. A pattern-based approach presents data modeling structures that occur frequently in practice, and prescribes guidelines on how to recognize and use these structures. This study describes the conceptual framework, experimental design, and results of a laboratory experiment that employed novice designers to compare the effectiveness of the two training approaches (between-subjects) at three levels of task complexity (within subjects). Results indicate an interaction effect between treatment and task complexity. The rule-based approach was significantly better in the low-complexity and the high-complexity cases; there was no statistical difference in the medium-complexity case. Designer performance fell significantly as complexity increased. Overall, though the rule-based approach was not significantly superior to the pattern-based approach in all instances, it out-performed the pattern-based approach at two out of three complexity levels. The primary contributions of the study are (1) the operationalization of the complexity construct to a degree not addressed in previous studies; (2) the development of a pattern-based instructional approach to database design; and (3) the finding that the effectiveness of a particular training approach may depend on the complexity of the task.

New designer drugs are constantly emerging onto the illicit drug market and it is often difficult to validate and maintaincomprehensive analytical methods for accurate detection of these compounds. Generally, toxicology laboratories utilize a screening method, such as immunoassay, for the presumptive identification of drugs of abuse. When a positive result occurs, confirmatory methods, such as gas chromatography (GC) or liquid chromatography (LC) coupled with mass spectrometry (MS), are required for more sensitive and specific analyses. In recent years, the need to study the activities of these compounds in screening assays as well as to develop confirmatory techniques to detect them in biological specimens has been recognized. Severe intoxications and fatalities have been encountered with emerging designer drugs, presenting analytical challenges for detection and identification of such novel compounds. The first major task of this research was to evaluate the performance of commercially available immunoassays to determine if designer drugs were cross-reactive. The second major task was to develop and validate a confirmatory method, using LC-MS, to identify and quantify these designer drugs in biological specimens.^ Cross-reactivity towards the cathinone derivatives was found to be minimal. Several other phenethylamines demonstrated cross-reactivity at low concentrations, but results were consistent with those published by the assay manufacturer or as reported in the literature. Current immunoassay-based screening methods may not be ideal for presumptively identifying most designer drugs, including the "bath salts." For this reason, an LC-MS based confirmatory method was developed for 32 compounds, including eight cathinone derivatives, with limits of quantification in the range of 1-10 ng/mL. The method was fully validated for selectivity, matrix effects, stability, recovery, precision, and accuracy. In order to compare the screening and confirmatory techniques, several human specimens were analyzed to demonstrate the importance of using a specific analytical method, such as LC-MS, to detect designer drugs in serum as immunoassays lack cross-reactivity with the novel compounds. Overall, minimal cross-reactivity was observed, highlighting the conclusion that these presumptive screens cannot detect many of the designer drugs and that a confirmatory technique, such as the LC-MS, is required for the comprehensive forensic toxicological analysis of designer drugs.^

Designing for users rather than with users is still a common practice in technology design and innovation as opposed to taking them on board in the process. Design for inclusion aims to define and understand end-users, their needs, context of use, and, by doing so, ensure that end-users are catered for and included, while the results are geared towards universality of use. We describe the central role of end-user and designer participation, immersion and perspective to build user-driven solutions. These approaches provided a critical understanding of the counterpart role. Designer(s) could understand what the user’s needs were, experience physical impairments, and see from other’s perspective the interaction with the environment. Users could understand challenges of designing for physical impairments, build a sense of ownership with technology and explore it from a creative perspective. The understanding of the peer’s role (user and designer), needs and perspective enhanced user participation and inclusion.

The most promising concept for low frequency (millihertz to hertz) gravitational wave observatories are laser interferometric detectors in space. It is usually assumed that the noise floor for such a detector is dominated by optical shot noise in the signal readout. For this to be true, a careful balance of mission parameters is crucial to keep all other parasitic disturbances below shot noise. We developed a web application that uses over 30 input parameters and considers many important technical noise sources and noise suppression techniques to derive a realistic position noise budget. It optimizes free parameters automatically and generates a detailed report on all individual noise contributions. Thus one can easily explore the entire parameter space and design a realistic gravitational wave observatory. In this document we describe the different parameters, present all underlying calculations, and compare the final observatory's sensitivity with astrophysical sources of gravitational waves. We use as an example parameters currently assumed to be likely applied to a space mission proposed to be launched in 2034 by the European Space Agency. The web application itself is publicly available on the Internet at http://spacegravity.org/designer. Future versions of the web application will incorporate the frequency dependence of different noise sources and include a more detailed model of the observatory's residual acceleration noise.

Le corps humain est composé de plusieurs milliards de cellules neuronales, lesquelles ont un impact primordial sur les systèmes vitaux. En effet, le système nerveux, étant lui-même un système vital du corps humain, effectue de nombreuses fonctions afin de voir au bon fonctionnement des autres systèmes du corps, tels que le système cardiovasculaire, le système digestif, le système musculaire et bien d’autres. Plusieurs maladies neurodégénératives telles que l’Alzheimer et la maladie de Creutzfeldt-Jakob, affectent les cellules nerveuses du corps et occasionnent la perte de différentes fonctions causant une diminution du bien-être et pouvant mener à la mort. La hausse du nombre d’individus atteint de maladies neurodégénératives observée au fils des ans nous montre l’importance de comprendre les phénomènes moléculaires qui se produisent lors des mécanismes de dégénérescence axonale dans les neurones. Lors de ma maîtrise dans le laboratoire du professeur Richard Blouin, j’ai étudié l’impact d’une augmentation intracellulaire de calcium sur la protéine dual leucine zipper kinase (DLK) en utilisant un modèle cellulaire humain, les SH-SY5Y. J’ai pu démontrer que l’entrée de calcium dans la cellule avait un impact sur la quantité de protéine DLK présente dans celles-ci. En effet, j’ai pu observer une diminution de la quantité de DLK dans les cellules suite à une entrée de calcium, un phénomène exclusivement calcium-dépendant. À l’aide d’inhibiteurs pharmacologiques, j’ai pu montrer l’implication, des calpaïnes, des protéases calcium-dépendantes reconnues pour leur rôle dans la dégénérescence axonale. Les essais in vitro montrent que DLK est une cible spécifique des calpaïnes.

Ma recherche porte sur l’adaptation auprès d’un groupe de jeunes adultes québécois d’une méthode de théâtre d’intervention réalisée et développée au Brésil. Il s’agit d’une recherche-action qui vise à valoriser le processus d’acquisition de savoirs qui, à son tour, pourra contribuer à l’émancipation des participants et promouvoir les valeurs d’humanité, de respect de la vie, de dignité et de partage de racines communes mises en valeur par l’art. Elle traite des conditions nécessaires à la pratique artistique réalisée par la communauté et appuyée par l’intégration des théories pédagogiques de Paulo Freire et d’Augusto Boal. Les résultats des expériences menées au Brésil avec le groupe de théâtre FACES (faire de l’art avec l’esthétique sociale) sont significatifs et leur impact a contribué au rayonnement de la communauté Nordeste de Amaralina de la ville de Salvador-Bahia. Est venu alors le désir de reprendre cette expérience et d’adapter la méthode de théâtre d’intervention brésilienne, mais cette fois-ci, pour une communauté de Québec. Quatre semaines ont été consacrées à la création d’un spectacle par les jeunes qui a été présenté aux amis et aux familles des participants. Durant cette expérience, les jeunes ou cochercheurs ont fait l’apprentissage du théâtre et de la création collective, et l’acquisition de connaissances personnelles et sociales. Les données provenant de cette expérience de théâtre d’intervention ont été non seulement recueillies dans un journal de bord tenu par l’animatrice chercheure tout au long du processus, mais aussi sur la base de deux entrevues individuelles effectuées au début et à la fin du projet. Les résultats ont été analysés et interprétés afin de répondre aux questions de recherche qui concernent autant l’adaptation de la méthode de théâtre d’intervention et l’acquisition de connaissances personnelles et collectives que l’émancipation de la chercheure et des cochercheurs.

La ricerca indaga il ruolo del designer nella transizione sostenibile e circolare all’uso di materiali polimerici. Nel contesto contemporaneo la plastica è utilizzata in quasi ogni settore merceologico ma la sua futura applicazione è messa in forte discussione a causa dei visibili impatti ambientali del suo uso irresponsabile. Un passaggio netto dalla totale dipendenza alla liberazione dei polimeri è difficile; è necessario un periodo di transizione che permetta di coesistere responsabilmente con i polimeri in attesa di trovare dei validi sostituti. L’obiettivo della ricerca è lavorare su questo periodo ponendo il designer e le sue competenze come soggetti chiave del movimento. La tesi di ricerca propone un approccio per calare le pratiche del Transition Design nella progettazione di sistemi-prodotto, nutrendosi degli attributi anticipatori dell’Advanced Design e puntando agli obiettivi del Circular Design, lavorando a partire dalle merci più critiche nel contesto contemporaneo: quelle in polimero fossile non riciclabile. Contributo della tesi è la figura del Transition Matter Designer, un progettista di transizioni dei materiali che prevede metamorfosi di sistemi-prodotto nel tempo grazie alle sue competenze a diverse scale del progetto: forma l’utente agli atteggiamenti circolari e sostenibili, caratterizza i materiali per individuarne nuovi usi, seleziona i processi produttivi adatti a prevenire scarti e ne anticipa i cicli di vita nei prodotti. I Knitted Fasteners sono il risultato della simulazione del lavoro del Transition Matter Designer nel tessile: un sistema di elementi di fissaggio, personalizzabili dallo stilista e integrati negli abiti a maglia, che permettono di eliminare l’uso di fashion fasteners in plastica e metallo, elementi che rendono difficile il riciclo dei capi. Dalla sperimentazione è emerso il modello concettuale della Transindustrial Production: un lavoro di collaborazione fra Transition Matter Designer e creativo per dare identità ai materiali polimerici circolari attraverso l’ibridazione fra artigianato e industria, tipico del Made in Italy.

Oggigiorno, quando una persona “ha un’idea” e vuole concretizzarla si rivolge agli incubatori: la diffusa metodologia Lean Startup è determinante nel far comprendere agli aspiranti founders la portata della propria “idea” abbattendo i rischi. Tuttavia, ci sono due problemi: accedere ai percorsi di incubazione non è scontato e, una volta concluso il percorso, si hanno diverse conoscenze per passare dalla validazione alla fase Seed, ma non sufficienti per un’efficace Execution. Partendo dalla personale esperienza del progetto Climby, si cercherà di dare un’overview sugli aspetti più importanti da considerare, evidenziando ciò che si potrebbe sottovalutare. Il fine è dunque supportare chi “ha un’idea” nell’intraprendere il percorso di incubazione in modo più consapevole e illustrare i limiti dei percorsi stessi, suggerendo approfondimenti di immediata applicazione ed utilità nelle diverse fasi citate, come integrazioni tratte dal Service Design. Pertanto, sarà posta particolare attenzione a ciò che è stato appreso in prima persona, considerando errori e aspetti positivi del percorso di un’“idea” nata nel 2019 e trasformatasi drasticamente durante l’incubazione, fino a ciò che accade dopo il Pitch Day. In tal modo, si intende anche esplicitare alcuni aspetti importanti, talvolta trascurati, ma rilevanti per le scelte da compiere prima ancora di iniziare un percorso. Il fine, quindi, è quello di produrre un contenuto complementare agli autorevoli scritti che trattano di Lean Startup, invece di proporne trattazione alternativa. Infine, conducendo queste riflessioni, si proporrà anche un confronto più specifico tra le metodologie e gli strumenti di Lean Startup e Service Design e - anticipando le conclusioni - suggerendo che sarebbe interessante approfondire l’integrazione di questi due ambiti, di cui emerge ancora una forte mancanza di reciproca conoscenza e che, invece, potrebbero rappresentare reciprocamente una risorsa se considerati senza visioni di parte.

Chlorophenylpiperazines (CPP) are psychotropic drugs used in nightclub parties and are frequently used in a state of sleep deprivation, a condition which can potentiate the effects of psychoactive drugs. This study aimed to investigate the effects of sleep deprivation and sleep rebound (RB) on anxiety-like measures in mCPP-treated mice using the open field test. We first optimized our procedure by performing dose-effect curves and examining different pretreatment times in naïve male Swiss mice. Subsequently, a separate cohort of mice underwent paradoxical sleep deprivation (PSD) for 24 or 48h. In the last experiment, immediately after the 24h-PSD period, mice received an injection of saline or mCPP, but their general activity was quantified in the open field only after the RB period (24 or 48h). The dose of 5mgmL(-1) of mCPP was the most effective at decreasing rearing behavior, with peak effects 15min after injection. PSD decreased locomotion and rearing behaviors, thereby inhibiting a further impairment induced by mCPP. Plasma concentrations of mCPP were significantly higher in PSD 48h animals compared to the non-PSD control group. Twenty-four hours of RB combined with mCPP administration produced a slight reduction in locomotion. Our results show that mCPP was able to significantly change the behavior of naïve, PSD, and RB mice. When combined with sleep deprivation, there was a higher availability of drug in plasma levels. Taken together, our results suggest that sleep loss can enhance the behavioral effects of the potent psychoactive drug, mCPP, even after a period of rebound sleep.
