Lysinibacillus sphaericus (Lsp) é uma bactéria entomopatógena que produz a toxina Bináriav(Bin) com atividade larvicida para culicídeos. A sua ação em Culex quinquefasciatus depende da ligação da toxina Bin à α-glicosidase (Aglu) Cqm1, que atua como receptor no epitélio intestinal de larvas. Na colônia R2362, foram caracterizados dois alelos de resistência ao Lsp: cqm1REC e cqm1REC-2, cujas mutações impedem a expressão da Aglu Cqm1. O objetivo deste trabalho foi avaliar a atividade catalítica da Cqm1 e comparar a atividade α-glicosidase e o desenvolvimento pré-imaginal de larvas de indivíduos susceptíveis (S) e resistentes (R) para cada alelo. Para isto, foram avaliados os seguintes parâmetros: atividade catalítica da Cqm1 recombinante; padrão de transcrição de outras Aglus parálogas à Cqm1; atividade de Aglus nativas em larvas; sobrevivência de indivíduos frente a diferentes dietas. A Aglu Cqm1 mostrou atividade enzimática ótima à 37o C, pH 7,5-8,0 e utilizando o substrato sintético pNαG. A atividade α-glicosidase total em larvas S e R foi similar, apesar da ausência de expressão da Cqm1 nas larvas R. A investigação in silico revelou 18 proteínas parálogas à Cqm1 e, dentre 11 investigadas, nove são expressas em larvas S e R. A análise quantitativa de três parálogas demonstrou que duas tem um padrão de transcrição mais elevado em larvas resistentes, sugerindo a existência de um mecanismo de compensação de expressão de α-glicosidases. O desenvolvimento pré-imaginal de larvas S foi decrescente nas seguintes dietas: ração de gatos, ração de peixes, leite desnatado, extrato de levedura e sacarose. De uma forma global, a taxa de sobrevivência de larvas R foi inferior à S em todas as dietas testadas. Os dados obtidos mostram que as mutações ligadas aos alelos cqm1REC e cqm1REC-2 não parecem impactar a atividade Aglu nas larvas e que o custo biológico observado poderia estar relacionado a outros genes e vias metabólicas.

Os insetos podem atuar como pragas agrícolas e vetores de patógenos causadores de doenças ao homem e outros animais. Investigações a respeito do sistema imunológico de Ae. aegypti e Cx. quinquefasciatus poderão contribuir para o desenvolvimento de métodos de controle das doenças veiculadas por estes insetos, principalmente a dengue, enfermidade causadora de sério problema de saúde pública no mundo. Apesar de Ae. aegypti ser a única espécie vetora confirmada na transmissão do vírus Dengue no Brasil, considera-se também importante um melhor entendimento dos mecanismos imunológicos de Cx. quinquefasciatus tido como refratário ao vírus. Neste estudo foram utilizadas linhagens de Ae. aegypti e Cx. quinquefasciatus mantidas no Insetário do Departamento de Entomologia do CPqAM/FIOCRUZ. Três grupos experimentais de fêmeas com 10 dias de idade foram formados para cada espécie. Grupo I, composto por fêmeas alimentadas com solução sacarose (10 por cento); grupo II, fêmeas alimentadas com sangue limpo e grupo III, fêmeas alimentadas com sangue infectado com o sorotipo DENV-1. De cada grupo foram obtidos hemolinfa, glândula salivar, intestino médio e corpo gorduroso para avaliação da expressão dos antimicrobianos defensina e transferrina. Essa avaliação foi realizada através de PCR em Tempo Real utilizando o kit QuantiFast SYBR Green - One-Step qRT-PCR. A avaliação da hemodinâmica foi realizada utilizando 10 microlitros de hemolinfa de cada grupo, através da contagem das células em câmara de Neubauer. Nossos resultados demonstraram que o Cx. quinquefasciatus tem um maior aumento da expressão de defensina e um maior número total de hemócitos quando infectados com DENV-1 em relação ao Ae. aegypti e a transferrina teve sua expressão alterada somente no Ae. aegypti. Em ambas as espécies estudadas, apenas a alimentação sanguínea não interfere na produção de hemócitos ou quanto na indução de defensina e transferrina. Esses dados sugerem que fêmeas de Cx. quinquefasciatus parecem apresentar uma resposta imune celular e humoral mais intensa do que Ae. aegypti quando infectados com DENV-1

Parity rate, gonotrophic cycle length, and density of a Culex quinquefasciatus female population was estimated at the Parque Ecologico do Tiete (PET), Sao Paulo, Brazil. Adult Cx. quinquefasciatus females were collected from vegetation along the edges of a polluted drainage canal with the use of a battery-powered backpack aspirator from September to November 2005 and from February to April 2006. We examined 255 Cx. quinquefasciatus ovaries to establish the parity rate of 0.22 and determined the gonotrophic cycle length under laboratory conditions to be 3 and 4 days. From these data, we calculated the Cx. quinquefasciatus survival rate to be 0.60 and 0.68 per day. Density of the Cx. quinquefasciatus female (5.71 females per m(2)) was estimated based on a population size of 28,810 individuals divided by the sampled area of 5,040 m(2). Results of all experiments indicate medium survivorship and high density of the Cx. quinquefasciatus female population. This species is epidemiologically relevant in the PET area and should be a target of the vector control program of Sao Paulo municipality.

The adult female Culex quinquefasciatus midgut comprises a narrow anterior and a dilated posterior region, with epithelia composed of a monolayer of adjacent epithelial cells joined at the apical portion by septate junctions. Densely packed apical microvilli and an intricate basal labyrinth characterise each cell pole. Our morphological studies suggest that, during blood digestion, the anterior midgut region also participates in an initial absorptive stage which is probably related to the intake of water, salts and other small molecules. This activity peaked by 6 h after bloodmeal feeding (ABF) and ended approximately 18 h ABF, when the peritrophic membrane was already formed. After this time, absorption only occurred in the posterior region, with morphologic and biochemical evidence of high synthetic activity related to the secretion of proteases. Chymotrypsin, elastase, aminopeptidase, and trypsin reached their maximum activity at around 36 h ABF. Digestion products were apparently absorbed and transported to the basal labyrinth, from where they should be released to the hemolymph. At 72 h ABF, proteolysis had already ended and protein levels had returned to those observed before blood meal. The epithelium of the posterior region, however, did not return to its initial morphology, appearing quite disorganised. The salivary glands of Culex quinquefasciatus female mosquitoes are paired organs composed of two lateral lobes with proximal and distal secretory portions, and a medial lobe. All portions comprise a simple epithelium that surrounds a salivary duct. In the apical portion of the medial lobe, non-secretory cells strongly resemble cells involved in ion and water transport. The general architecture of the secretory portions is similar between lobes. The appearance of the secretory material and the morphological aspect of the apical cell membrane are the most distinctive features among the three secretory portions. Cells in the lateral proximal lobe display thin membrane projections extending into a translucent and finely filamentous secretory product. At the lateral distal portion, the apical cell membrane forms an intricate meshwork that encloses a dark secretory product. Medial lobe secretory cells also contain secretory cavities surrounded by intracytoplasmic vesicles, all containing a very dark and uniform product. Scattered cells holding numerous vacuoles, some of them containing a small and electron-dense granule eccentrically located and resembling those of the diffuse endocrine system, are frequently observed in the periphery of all secretory portions. Immunofluorescence assays revealed that the distal portion of the lateral lobes contains apyrase, an enzyme putatively responsible for platelet aggregation inhibition, diffusely distributed in the cell cytoplasm. © 2003 Elsevier Ltd. Culex quinquefasciatus is a vector of human pathogens, including filarial nematodes and several viruses. Although its epidemiological relevance is known to vary across geographical regions, an understanding of its population genetic structure is still incipient. In light of this, we evaluated the genetic diversity of Cx. quinquefasciatus and Cx. pipiens x Cx. quinquefasciatus hybrids collected from nine localities in Brazil and one site in Argentina. We used mitochondrial genes cox1 and nd4, along with the coxA and wsp genes of the maternally-inherited Wolbachia endosymbiont. The nd4 fragment was invariant between samples, whilst cox1 exhibited four haplotypes that separated two types of Cx. quinquefasciatus, one clustered in southern Brazil. Low sequence diversity was generally observed, being discussed. Both Brazilian and Argentinian mosquitoes were infected with a single Wolbachia strain. As reported in previous studies with these populations, cox1 and nd4 diversity is not congruent with the population structure revealed by nuclear markers or alar morphology. Future Cx. quinquefasciatus research should, if possible, evaluate mtDNA diversity in light of other markers.

Insect storage proteins accumulate at high levels during larval development of holometabolous insects. During metamorphosis they are degraded, supplying energy and amino acids for the completion of adult development. The genome of Culex quinquefasciatus contains eleven storage protein-coding genes. Their transcripts are more abundant in larvae than in pupae and in adults. In fact, only four of these genes are transcribed in adults, two of which in blood-fed adult females but not in adult males. Transcripts corresponding to all Cx. quinquefasciatus storage proteins were detected by RT-PCR, while mass spectrometric analysis of larval and pupal proteins identified all storage proteins with the exception of one encoded by Cq LSP1.8. Our results indicate that the identified Cx. quinquefasciatus storage protein-coding genes are candidates for identifying regulatory sequences for the development of molecular tools for vector control

Cry proteins produced by Bacillus thuringiensis are selective biodegradable insecticides used increasingly in bacterial insecticides and transgenic plants as alternatives to synthetic chemical insecticides. However, the potential for development of resistance and cross-resistance in target insect populations to Cry proteins used alone or in combination threatens the more widespread use of this novel pest control technology. Here we show that high levels of resistance to CryIV proteins in larvae of the mosquito, Culex quinquefasciatus, can be suppressed or reduced markedly by combining these proteins with sublethal quantities of CytA, a cytolytic endotoxin of B. thuringiensis. Resistance at the LC95 level of 127-fold for a combination of three CryIV toxins (CryIVA, B, and D), resulting from 60 generations of continuous selection, was completely suppressed by combining sporulated powders of CytA in a 1:3 ratio with sporulated powders of a CryIVA, CryIVB, and CryIVD strain. Combining the CytA strain with a CryIVA and CryIVB strain also completely suppressed mosquito resistance of 217-fold to the latter toxins at the LC95 level, whereas combination of CytA with CryIVD reduced resistance in a CryIVD-selected mosquito strain from greater than 1,000-fold to less than 8-fold. The CytA/CryIV model provides a potential molecular genetic strategy for engineering resistance management for Cry proteins directly into bacterial insecticides and transgenic plants.

Background: West Nile virus (WNV) infection, is an arbovirus infection with high morbidity and mortality, the vector responsible for both human and animal transmission is Culex pipens complex. Objective: To determine the species distribution and seasonal abundance of Culex pipens and Culex quinquefasciatus mosquitoes in Abeokuta, Nigeria. Methods: Mosquitoes belonging to the Culex pipens complex were captured in three different locations located within Abeokuta Metropolis between March 2012 and January 2013. Individual species were identified using morphometric methods. Amplification of the Ace2 gene by PCR confirmed morphormetric identification of the mosquitoes. Results: A total of 751 mosquitoes were captured. Culex quinquefaciatus recorded the highest distribution of vectors with 56.6% and Culex pipens 43.4% (P > 0.05). Idi aba community recorded the highest distribution of mosquito vectors with 42.9% (n=322) and Culex quinqueaciatus was more abundantly distributed with 183 mosquitoes. Aro community recorded 32% (n=240) of captured mosquitoes with Culex quinquefaciatus having a higher level of abundance and lastly Kemta with a distribution of 25.1% (n=189). Conclusion: Results from this study show that potential vectors of WNV abound within Abeokuta, putting residents at high risk of West Nile infection. We advocate for introduction of routine testing of WNV in Abeokuta and Nigeria. Keywords:

En el presente trabajo, se describen los niveles de expresión de siete transcritos de Culex quinquefasciatus que codifican para citocromos P450, estos medidos por qPCR en larvas expuestas a dos concentraciones del insecticida permetrina (0.51 y 0.71 mg) comparadas con un grupo control sin exponer, las cuales fueron colectadas en distintas localidades del noreste de México. Debido a que los genes analizados no se encontraban reportados, fue necesario diseñar oligonucleótidos utilizando secuencias trazas producto de la secuenciación del genoma del mosquito antes mencionado. Con los oligonucleótidos diseñados y utilizando DNA complementario, sintetizado a partir de RNA total de las larvas fueron amplificados por PCR los mensajeros, se procedió a clonar cada uno de estos genes en nueve poblaciones analizadas, posteriormente fueron secuenciados y éstas fueron comparadas para deducir los porcentajes de similitud. Se diseñaron sondas taqman en segmentos conservados, y utilizando el DNA complementario previamente sintetizado fueron deducidos los niveles de expresión de cada uno de los genes analizados bajo las condiciones citadas anteriormente para las poblaciones. Con el trabajo realizado, reportamos las secuencias nucleótidicos y aminoacídicas de cada uno de los genes analizados de cada una de las poblaciones estudiadas y sus niveles de expresión. Concluimos que los genes analizados son una herramienta potente para ahondar en el estatus de resistencia que han desarrollado las poblaciones silvestres de Culex quinquefasciatus en localidades del noreste de México.

Purpose: To evaluate the larvicidal, pupicidal and insecticidal activities of Cosmos bipinnatus , Foeniculum vulgare and Tagetes minuta against Culex quinquefasciatus mosquitoes. Methods: The leaves of the plants were extracted with distilled water, ethanol (95 %), and hexane and the extracts screened for their phytochemical profile. While larvicidal and pupicidal activities were assayed at concentrations ranging from 0.1 - 10 mg/mL, insecticidal property was tested at varying amounts (0.25 - 2 g) of the plant sample. The respective larval mortality was thereafter evaluated using Probit analysis. Results: Saponins, terpenoids, flavonoids and steroids were detected in the plant extracts. The ethanol extracts of F. vulgare, T. minuta and C. bipinnatus exhibited larvicidal activity half-maximal lethal concentration (LC50) of 0.10, 1.17 and 1.18 mg/mL, followed by hexane extracts with LC50 value of 1.03, 1.01 and 1.27 mg/mL, respectively, against the larvae of C. quinquefasciatus mosquito. Hexane extracts displayed pupicidal activity with LC50 of 1.07, 1.12 and 1.16 mg/mL against F. vulgare, T. minuta and C. bipinnatus, respectively, while the ethanol extracts of T. minuta, C. bipinnatus and F. vulgare displayed pupicidal activity at LC50 of 1.11, 1.14 and 1.31 mg/mL respectively, against pupa of C. quinquefasciatus mosquito. The aqueous extracts had no (p > 0.05) lethal effects on both larvae and pupa of C. quinquefasciatus at all evaluated concentrations. F. vulgare had the highest (p < 0.05) half-maximal knock-down effect (KD50 = 7.52 min-1), followed by T. minuta (KD50 = 8.64 min-1) on adult C. quinquefasciatus mosquitoes after 6 h of exposure. F. vulgare and T. minuta killed all evaluated mosquito adults within 12 h with LD99 = 0.25 g/air, while the leaves of C. bipinnatus had no (p > 0.05) knock-down or lethal effects on the adult mosquito. Conclusion: C. bipinnatus, F. vulgare and T. minuta possess larvicidal and pupicidal properties against C. quinquefasciatus, whereas only F. vulgare and T. minuta displayed insecticidal properties. Consequent upon these findings, all the plants can be considered naturally potent larvicidal and pupicidal agents against C. quinquefasciatus.

OBJETIVO: Observar a potencialidade sinantrópica de Anopheles bellator e de An. cruzii em vilarejo rodeado por ambiente natural. MÉTODOS: As observações foram levadas a efeito na localidade de Pedrinhas, no Sudeste do Estado de São Paulo, Brasil, de outubro de 1996 a janeiro de 2000. A sinantropia, ou domiciliação, foi estimada pelo índice de Nuorteva (s) acrescido do fator intradomiciliar de Mihályi (d) e adaptado para culicídeos hematófagos. RESULTADOS: Utilizando procedimento baseado na atração pela isca humana, as duas espécies de Kerteszia mostraram freqüência conjunta positiva correspondente a s=35,7. Isso parece indicar a existência de certo grau de sinantropia por parte desses mosquitos. Assim também, a população de Cx. sacchettae, correspondente à razão de sinantropia sr=12,8, obtida pelo cálculo da relação entre os dados conseguidos na armadilha de Shannon. Quanto ao método de procura ativa pela aspiração ambiental, os resultados obtidos consistiram em valores negativos de s. Assim, corresponderam a -43,1 para An. bellator, -48,2 para An. cruzii e -3,0 para Cx. sacchettae. Tais cifras foram relacionadas ao valor positivo de s=+93,8 apresentado por Cx. quinquefasciatus e considerado como equivalente a +100,00. CONCLUSÕES: O observado em relação a domicílios isolados rurais repetiu-se em conjunto de casas formando vila, também de caráter rural, e circundada por ambiente primitivo. Ou seja, esses mosquitos adultos tendem a procurar o ambiente antrópico na busca de repasto sangüíneo. Uma vez tendo-o conseguido, voltam para o ambiente de onde saíram. Todavia, revelou-se alguma tendência adaptativa revelada pelo encontro de formas imaturas de An. bellator em recipientes artificiais. Quanto a Cx. sacchettae, a população local parece mostrar tendência para adaptação ao meio antrópico.

IntroductionThe aim of this work was to identify possible lymphatic filariasis foci in the western Brazilian Amazonian that could be established from the reports of Rachou in the 1950s. The study was conducted in three cities of the western Brazilian Amazon region - Porto Velho and Guajará-Mirim (State of Rondônia) and Humaitá (State of Amazonas).MethodsFor human infection evaluation thick blood smear stained with Giemsa was used to analyze samples collected from 10pm to 1am. Polymerase chain reaction (PCR) was used to examine mosquito vectors for the presence of Wuchereria bancrofti DNA. Humans were randomly sampled from night schools students and from inhabitants in neighborhoods lacking sanitation. Mosquitoes were collected from residences only.ResultsA total 2,709 night students enrolled in the Program for Education of Young Adults (EJA), and 935 people registered in the residences near the schools were examined, being 641 from Porto Velho, 214 from Guajará-Mirim and 80 from Humaitá. No individual examined was positive for the presence of microfilariae in the blood stream. A total of 7,860 female Culex quinquefasciatus specimens examined were negative by PCR.ConclusionsThis survey including human and mosquito examinations indicates that the western Amazon region of Brazil is not a focus of Bancroftian filariasis infection or transmission. Therefore, there is no need to be included in the Brazilian lymphatic filariasis control program.
