As part of two research projects for analysing bycatch and discards, we quantified catch composition, catch rates, bycatch and discard,, in two important commercial bottom trawl fisheries (crustacean and fish trawls) off the Southern coast of Portugal (Algarve). Stratified sampling by onboard observers took place from February 1999 to March 2001 and data were collected from 165 tows during 52 fishing trips. Commercial tat-get species included crustaceans: blue and red shrimp (Aristeus antennatus), deep-water rose shrimp (Parapenaeus longirostris), Norway lobster (Nepharops norvegicus); and fishes: scabreams (Diplodus spl). and Pagellus Spp.), horse Mackerels (Trachurus spp.) and European hake (Merluccius merluccius). The trawl fisheries are characterised by considerable amounts of bycatch: 59.5% and 80.4% of the overall total catch for crustacean and fish trawlers respectively. A total of 255 species were identified, which belonged to 15 classes of organisms ( 137 vertebrates, 112 invertebrates and algae). Crustacean trawlers had higher bycatch biodiversity. Bony fish (45.6% and 37.8%) followed by crustaceans (14.6% and 11.5%) were the dominant bycatch components of both crustacean and fish trawlers respectively. The influence of a number of factors (e.g. depth, fishing gear, tow duration and season) on bycatch and discards is discussed.

A prototype for a Responsive Fisheries Management System (RFMS) was developed in the context of the European FP7 project EcoFishMan and tested on the Portuguese crustacean trawl fishery. Building on Results Based Management principles, RFMS involves the definition of specific and measurable objectives for a fishery by the relevant authorities but allows resource users the freedom to find ways to achieve the objectives and to provide adequate documentation. Taking into account the main goals of the new Common Fisheries Policy, such as sustainable utilization of the resources, end of discards and unwanted catches, a management plan for the Portuguese crustacean trawl fishery was developed in cooperation with the fishing industry, following the process and design laid out in the RFMS concept. The plan considers biological, social and economic goals and assigns a responsibility for increased data collection to the resource users. The performance of the plan with regard to selected indicators was evaluated through simulations. In this paper the process towards a RFMS is described and the lessons learnt from the interaction with stakeholders in the development of an alternative management plan are discussed. The crustacean trawl fishery off the Algarve coast (southern Portugal) takes place on the lower continental shelf and upper continental slope at depths between 150 and 600 m. This is a multi-species fishery targeting the shrimps Parapenaeus longirostris and Aristeus antennatus and the Norway lobster Nephrops norvegicus, with the latter two species the most important in the landings. The fishery is characterised by significant by-catch and discarding of a large number of species. As part of a study on the fate of trawl fishery discards, this component of the study focused on the quantification of the by-catch and discards of crustacean trawlers. Sampling took place on board seven commercial trawlers from June 1998 to October 1999. Data was collected from 48 tows in 22 fishing trips. The observers collected all of the catch that was discarded by the crew during the sorting operation and samples were taken to the laboratory for identification, weighing and measuring. The quantities of target species were recorded along with the presence of retained by-catch. Commercially valuable species that were retained included Plesiopenaeus edwardsianus, Aristeomorpha foliacea, Plesionika sp., and the fishes Lophius piscatorius and Merluccius merluccius. However, most of the species had no or little commercial value and were almost always discarded to the sea (90%). A total of 91 species were identified, 47 vertebrates and 44 invertebrates corresponding to 65 families. The Teleostei (78% and 68%) were the dominant group, both in number and weight. The species Micromesistius poutassou (34%), Gadiculus argenteus (10%) and Hoplostethus mediterraneus (8%) were the most important in weight. Ten species represented more than 82% of all discards in weight. Gadiculus argenteus (29%), Hoplostethus mediterraneus (21%) and Nezumia sclerorhynchus (10%) accounted for 60% of all discards in numbers. Data on the landed species composition is also presented.

The effects of an increase in cod end mesh size from 55 to 60 and 70 mm and a change of mesh configuration from 55 mm diamond to 55 turn square mesh on the size selectivity of four by-catch species (the red shrimp Aristeus antennatus, the European hake Merluccius merluccius, the horse mackerel Trachurus trachurus and the blue whiting Micromesistius poutassou) commonly captured in the crustacean fishery off the Portuguese south coast, were evaluated. Selectivity parameters for blue whiting, the most abundant species in the catches, were estimated taking into account between-haul variation, while for the remaining species, captured in much lower quantities, the selectivity estimates were based on pooled data by length class for all hauls within the same cod end. Length at 50% retention, L-50, was found to increase with mesh size and with the change in mesh configuration for all the studied species. For blue whiting trawling depth and cod end catch were found to play a role in between-haul variation by increasing L-50 as well. The results suggest that an increase in the current minimum mesh size of 55-70 mm would be advisable to be compatible with the minimum landing sizes (MLSs) of 29 mm carapace length and 27 cm total length for red shrimp and hake, respectively, while it would greatly reduce the amount of discards, particularly those for blue whiting, that accounted for approximately 50% of the total catch weight. Horse mackerel was the only species for which the use of a larger mesh size would result in a significant escapement of individuals above the MLS of 15 cm. (C) 2002 Elsevier Science B.V. By-catch and discards are a common problem to all fisheries. Recent studies of the Portuguese crustacean-trawl fishery show that discards average 70% of the total catch. This situation poses a threat not only to the management of target and commercial fish by-catch species stocks, but also in terms of the ecosystem impact of the discarded species whose survival is generally low. Previous attempts to address this problem by using oblique separator mesh panels associated with square-mesh windows, or square-mesh windows alone, provided encouraging results, but these are unlikely to have commercial applicability at present. Therefore, we tested the efficiency of a modified Nordmore grid in excluding some of the most captured non-commercial by-catch species, such as the blue whiting, Micromesistius poutassou, and the boarfish, Capros aper, evaluating simultaneously the losses of the crustacean target species (rose shrimp, Parapenaeus longirostris, Norway lobster, Nephrops norvegicus and red shrimp, Aristeus antennatus) and commercially valuable fish by-catch. A total of 41 valid hauls were carried out during 2001, and 15 in 2002 on board the R/V "Noruega". Results from both years varied considerably, with higher by-catch exclusion rates, in weight, registered in the second survey (73-74 and 48-63% for blue whiting and boarfish, respectively). The corresponding percentage reduction for target species was 8-15% for the Norway lobster, 4-9% for rose shrimp and 7-10% for red shrimp. The level of exclusion of the non-commercial by-catch is encouraging, although short-term loss of Norway lobster, where the escapees comprised larger individuals, raises some concern with regards to fishers' acceptance of mandatory use of grids. This study was conducted to determine the reduction in bycatch and the loss of target catch in trawl hauls for three commercial crustacean species (yellow squat lobster, Cervimunida johni, nylon shrimp, Heterocarpus reedi, and red squat lobster, Pleuroncodes monodon) in central Chile. Two experiments were carried out using the covered codend method; the first used different mesh sizes (56 and 70 mm, knot-centre to knot-centre) and shapes (diamond and square), and the second tested different reduction devices (escape panel and sorting grid). We analysed the escape proportions of the species caught and the size frequency distributions of the target species and Chilean hake (Merluccius gayi), the main commercial bycatch species. The results demonstrate that it is feasible to significantly reduce bycatch while keeping losses within acceptable ranges. In particular, the best option for yellow squat lobster was the combination of a 70-mm diamond mesh with an escape panel that reduced non-target and target catches by 48% and 7%, respectively. For nylon shrimp, the best option was the use of a 56-mm diamond mesh with a sorting grid that reduced non-target and target catches by 41% and 11%, respectively. Although the results for red squat lobster were not as promising, they will help to guide future research.

Trawling for crustaceans takes place on the south coast of Portugal at depths between 200 and 800 m. Large amounts of discards are released back to sea, reaching the bottom in the general area where trawling occurs. The objective of this work was to study the time taken for decomposition of the discards, to identify the most important scavenging species involved, and to evaluate the impact on the species targeted by the fishery. We conducted a series of 22 trials, using traps baited with samples of the discards. The time of immersion varied between 1 and 40 h. The amount of tissue removed from the discards was evaluated on a qualitative scale of 1–3, and quantitatively. The species caught in the traps were identified. Considerable consumption of the bait had occurred after 5 h of immersion. After 24 h of immersion only fish bones were left; past 40 h, consumption was complete. The conger eel (Conger conger) was the most common fish species present in the traps. Two species, the amphipod Scopelocheirus hopei and the isopod Natatolana (Cirolana) borealis, were identified as most important for the recycling of organic matter in the system. The stomach contents of a sample of species caught in trawls were analysed for the presence of small scavengers. Preliminary results show that S. hopei and N. (C.) borealis are part of the diet of some of the target species of this fishery, such as the Norway lobster, Nephrops norvegicus and the rose shrimp Parapenaeus longirostris.

The porpoise of this study was to implement research methodologies and assess the effectiveness and impact of management tools to promote best practices for the long term conservation of the endangered African wild dog (Lycaon pictus). Different methods were included in the project framework to investigate and expand the applicability of these methodologies to free-ranging African wild dogs in the southern African region: ethology, behavioural endocrinology and ecology field methodologies were tested and implemented. Additionally, research was performed to test the effectiveness and implication of a contraceptive implant (Suprenolin) as a management tool for the species of a subpopulation hosted in fenced areas. Attention was especially given to social structure and survival of treated packs. This research provides useful tools and advances the applicability of these methods for field studies, standardizing and improving research instruments in the field of conservation biology and behavioural endocrinology. Results reported here provide effective methodologies to expand the applicability of non-invasive endocrine assessment to previously prohibited fields, and validation of sampling methods for faecal hormone analysis. The final aim was to fill a knowledge gap on behaviours of the species and provide a common ground for future researchers to apply non-invasive methods to this species research and to test the effectiveness of the contraception on a managed metapopulation.

Intermittent fasting (IF) is an often-used intervention to decrease body mass. In male Sprague-Dawley rats, 24 hour cycles of IF result in light caloric restriction, reduced body mass gain, and significant decreases in the efficiency of energy conversion. Here, we study the metabolic effects of IF in order to uncover mechanisms involved in this lower energy conversion efficiency. After 3 weeks, IF animals displayed overeating during fed periods and lower body mass, accompanied by alterations in energy-related tissue mass. The lower efficiency of energy use was not due to uncoupling of muscle mitochondria. Enhanced lipid oxidation was observed during fasting days, whereas fed days were accompanied by higher metabolic rates. Furthermore, an increased expression of orexigenic neurotransmitters AGRP and NPY in the hypothalamus of IF animals was found, even on feeding days, which could explain the overeating pattern. Together, these effects provide a mechanistic explanation for the lower efficiency of energy conversion observed. Overall, we find that IF promotes changes in hypothalamic function that explain differences in body mass and caloric intake.

There is great interindividual variability in the response to GH therapy. Ascertaining genetic factors can improve the accuracy of growth response predictions. Suppressor of cytokine signaling (SOCS)-2 is an intracellular negative regulator of GH receptor (GHR) signaling. The objective of the study was to assess the influence of a SOCS2 polymorphism (rs3782415) and its interactive effect with GHR exon 3 and -202 A/C IGFBP3 (rs2854744) polymorphisms on adult height of patients treated with recombinant human GH (rhGH). Genotypes were correlated with adult height data of 65 Turner syndrome (TS) and 47 GH deficiency (GHD) patients treated with rhGH, by multiple linear regressions. Generalized multifactor dimensionality reduction was used to evaluate gene-gene interactions. Baseline clinical data were indistinguishable among patients with different genotypes. Adult height SD scores of patients with at least one SOCS2 single-nucleotide polymorphism rs3782415-C were 0.7 higher than those homozygous for the T allele (P < .001). SOCS2 (P = .003), GHR-exon 3 (P= .016) and -202 A/C IGFBP3 (P = .013) polymorphisms, together with clinical factors accounted for 58% of the variability in adult height and 82% of the total height SD score gain. Patients harboring any two negative genotypes in these three different loci (homozygosity for SOCS2 T allele; the GHR exon 3 full-length allele and/or the -202C-IGFBP3 allele) were more likely to achieve an adult height at the lower quartile (odds ratio of 13.3; 95% confidence interval of 3.2-54.2, P = .0001). The SOCS2 polymorphism (rs3782415) has an influence on the adult height of children with TS and GHD after long-term rhGH therapy. Polymorphisms located in GHR, IGFBP3, and SOCS2 loci have an influence on the growth outcomes of TS and GHD patients treated with rhGH. The use of these genetic markers could identify among rhGH-treated patients those who are genetically predisposed to have less favorable outcomes.

Most epidemiological studies concerning differentiated thyroid cancers (DTC) indicate an increasing incidence over the last two decades. This increase might be partially explained by the better access to health services worldwide, but clinicopathological analyses do not fully support this hypothesis, indicating that there are carcinogenetic factors behind this noticeable increasing incidence. Although we have undoubtedly understood the biology and molecular pathways underlying thyroid carcinogenesis in a better way, we have made very little progresses in identifying a risk profile for DTC, and our knowledge of risk factors is very similar to what we knew 30-40 years ago. In addition to ionizing radiation exposure, the most documented and established risk factor for DTC, we also investigated the role of other factors, including eating habits, tobacco smoking, living in a volcanic area, xenobiotics, and viruses, which could be involved in thyroid carcinogenesis, thus, contributing to the increase in DTC incidence rates observed.

The control of energy homeostasis relies on robust neuronal circuits that regulate food intake and energy expenditure. Although the physiology of these circuits is well understood, the molecular and cellular response of this program to chronic diseases is still largely unclear. Hypothalamic inflammation has emerged as a major driver of energy homeostasis dysfunction in both obesity and anorexia. Importantly, this inflammation disrupts the action of metabolic signals promoting anabolism or supporting catabolism. In this review, we address the evidence that favors hypothalamic inflammation as a factor that resets energy homeostasis in pathological states.

There is an increasing rate of papillary thyroid carcinomas that may never progress to cause symptoms or death. Predicting outcome and determining tumour aggressiveness could help diminish the number of patients submitted to aggressive treatments. We aimed to evaluate whether markers of the immune system response and of tumour-associated inflammation could predict outcome of differentiated thyroid cancer (DTC) patients. Retrospective cohort study. We studied 399 consecutive patients, including 325 papillary and 74 follicular thyroid carcinomas. Immune cell markers were evaluated using immunohistochemistry, including tumour-associated macrophages (CD68) and subsets of tumour-infiltrating lymphocytes (TIL), such as CD3, CD4, CD8, CD16, CD20, CD45RO, GRANZYME B, CD69 and CD25. We also investigated the expression of cyclooxygenase 2 (COX2) in tumour cells and the presence of concurrent lymphocytic infiltration characterizing chronic thyroiditis. Concurrent lymphocytic infiltration characterizing chronic thyroiditis was observed in 29% of the cases. Among all the immunological parameters evaluated, only the enrichment of CD8+ lymphocytes (P = 0·001) and expression of COX2 (P =0·01) were associated with recurrence. A multivariate model analysis identified CD8+ TIL/COX2 as independent risk factor for recurrence. A multivariate analysis using Cox's proportional-hazards model adjusted for the presence of concurrent chronic thyroiditis demonstrated that the presence of concurrent chronic thyroiditis had no effect on prognostic prediction mediated by CD8+ TIL and COX2. In conclusion, we suggest the use of a relatively simple pathology tool to help select cases that may benefit of a more aggressive approach sparing the majority of patients from unnecessary procedures.

The overall prevalence of infertility was estimated to be 3.5-16.7% in developing countries and 6.9-9.3% in developed countries. Furthermore, according to reports from some regions of sub-Saharan Africa, the prevalence rate is 30-40%. The consequences of infertility and how it affects the lives of women in poor-resource settings, particularly in developing countries, has become an important issue to be discussed in reproductive health. In some societies, the inability to fulfill the desire to have children makes life difficult for the infertile couple. In many regions, infertility is considered a tragedy that affects not only the infertile couple or woman, but the entire family. This is a position paper which encompasses a review of the needs of low-income infertile couples, mainly those living in developing countries, regarding access to infertility care, including ART and initiatives to provide ART at low or affordable cost. Information was gathered from the databases MEDLINE, CENTRAL, POPLINE, EMBASE, LILACS, and ICTRP with the key words: infertility, low income, assisted reproductive technologies, affordable cost, low cost. There are few initiatives geared toward implementing ART procedures at low cost or at least at affordable cost in low-income populations. Nevertheless, from recent studies, possibilities have emerged for new low-cost initiatives that can help millions of couples to achieve the desire of having a biological child. It is necessary for healthcare professionals and policymakers to take into account these new initiatives in order to implement ART in resource-constrained settings.

Glucocorticoid (GC) therapies may adversely cause insulin resistance (IR) that lead to a compensatory hyperinsulinemia due to insulin hypersecretion. The increased β-cell function is associated with increased insulin signaling that has the protein kinase B (AKT) substrate with 160 kDa (AS160) as an important downstream AKT effector. In muscle, both insulin and AMP-activated protein kinase (AMPK) signaling phosphorylate and inactivate AS160, which favors the glucose transporter (GLUT)-4 translocation to plasma membrane. Whether AS160 phosphorylation is modulated in islets from GC-treated subjects is unknown. For this, two animal models, Swiss mice and Wistar rats, were treated with dexamethasone (DEX) (1 mg/kg body weight) for 5 consecutive days. DEX treatment induced IR, hyperinsulinemia, and dyslipidemia in both species, but glucose intolerance and hyperglycemia only in rats. DEX treatment caused increased insulin secretion in response to glucose and augmented β-cell mass in both species that were associated with increased islet content and increased phosphorylation of the AS160 protein. Protein AKT phosphorylation, but not AMPK phosphorylation, was found significantly enhanced in islets from DEX-treated animals. We conclude that the augmented β-cell function developed in response to the GC-induced IR involves inhibition of the islet AS160 protein activity.
