Myxozoans, belonging to the recently described Class Malacosporea, parasitise freshwater bryozoans during at least part of their life cycle, but no complete malacosporean life cycle is known to date. One of the 2 described malacosporeans is Tetracapsuloides bryosalmonae, the causative agent of salmonid proliferative kidney disease. The other is Buddenbrockia plumatellae, so far only found in freshwater bryozoans. Our investigations evaluated malacosporean life cycles, focusing on transmission from fish to bryozoan and from bryozoan to bryozoan. We exposed bryozoans to possible infection from: stages of T bryosalmonae in fish kidney and released in fish urine; spores of T bryosalmonae that had developed in bryozoan hosts; and spores and sac stages of B. plumatellae that had developed in bryozoans. Infections were never observed by microscopic examination of post-exposure, cultured bryozoans and none were detected by PCR after culture. Our consistent negative results are compelling: trials incorporated a broad range of parasite stages and potential hosts, and failure of transmission across trials cannot be ascribed to low spore concentrations or immature infective stages. The absence of evidence for bryozoan to bryozoan transmissions for both malacosporeans strongly indicates that such transmission is precluded in malacosporean life cycles. Overall, our results imply that there may be another malacosporean host which remains unidentified, although transmission from fish to bryozoans requires further investigation. However, the highly clonal life history of freshwater bryozoans is likely to allow both long-term persistence and spread of infection within bryozoan populations, precluding the requirement for regular transmission from an alternate host.

Fluvial redeposition of stone artifacts is a major complicating factor in the interpretation of Lower Palaeolithic open-air archaeological sites. However, the microscopic examination of lithic surfaces may provide valuable background information on the transport history of artifacts, particularly in low energy settings. Replica flint artifacts were therefore abraded in an annular flume and examined with a scanning electron microscope. Results showed that abrasion time, sediment size, and artifact transport mode were very sensitive predictors of microscopic surface abrasion, ridge width, and edge damage (p < 0.000). These results suggest that patterns of micro-abrasion of stone artifacts may enhance understanding of archaeological assemblage formation in fluvial contexts

Objectives: The objective of this study was to evaluate the accuracy and reproducibility of three complete denture biofilm indices (Prosthesis Hygiene Index; Jeganathan et al. Index; Budtz-J circle divide rgensen Index) by means of a computerised comparison method. Background: Clinical studies into denture hygiene have employed a large number of biofilm indices among their outcome variables. However, the knowledge about the validity of these indices is still scarce. Materials and methods: Sixty-two complete denture wearers were selected. The internal surfaces of the upper complete dentures were stained (5% erythrosine) and photographed. The slides were projected on paper, and the biofilm indices were applied over the photos by means of a scoring method. For the computerised method, the areas (total and biofilm-covered) were measured by dedicated software (Image Tool). In addition, to compare the results of the computerised method and Prosthetic Hygiene Index, a new scoring scale (including four and five graded) was introduced. For the Jeganathan et al. and Budtz-J circle divide rgensen indices, the original scales were used. Values for each index were compared with the computerised method by the Friedman test. Their reproducibility was measured by means of weighed kappa. Significance for both tests was set at 0.05. Results: The indices tested provided similar mean measures but they tended to overestimate biofilm coverage when compared with the computerised method (p < 0.001). Agreement between the Prosthesis Hygiene Index and the computerised method was not significant, regardless of the scale used. Jeghanathan et al. Index showed weak agreement, and consistent results were found for Budtz-Jorgensen Index (kappa = 0.19 and 0.39 respectively). Conclusion: Assessment of accuracy for the biofilm indices showed instrument bias that was similar among the tested methods. Weak inter-instrument reproducibility was found for the indices, except for the Budtz-J circle divide rgensen Index. This should be the method of choice for clinical studies when more sophisticated approaches are not possible.

The study was undertaken to evaluate changes in the activity of adenosine deaminase (ADA) in brains of rats infected by Trypanosoma evansi. Each rat was intraperitoneally infected with 10(6) trypomastigotes either suspended in fresh (group A; n = 13) and cryopreserved blood (group B; n = 13). Thirteen animals were used as control (group C). ADA activity was estimated in the cerebellum, cerebral cortex, striatum and hippocampus. No differences (P > 0.05) in ADA activity were observed in the cerebellum between infected and non-infected animals. Significant (P < 0.05) reductions in ADA activity occurred in cerebral cortex in acutely (day 4 post-infection; PI) and chronically (day 20 PI) infected rats. ADA activity was significantly (P < 0.05) decreased in the hippocampus in acutely infected rats, but significantly (P < 0.05) increased in the chronically infected rats. Significant (P < 0.05) reductions in ADA activity occurred in the striatum of chronically infected rats. Parasites could be found in peripheral blood and brain tissue through microscopic examination and PCR assay, respectively, in acutely and chronically infected rats. The reduction of ADA activity in the brain was associated with high levels of parasitemia and anemia in acute infections. Alterations in ADA activity of the brain in T. evansi-infected rats may have implications for pathogenesis of the disease. (C) 2010 Elsevier Inc. All rights reserved.

XIXA doença do refluxo gastroesofágico (DRGE) comumente afeta o esôfago e provavelmente é a condição mais prevalente no segmento alto do trato gastrointestinal, acometendo entre 5 e 45% da população ocidental. A terapêutica atual para essa doença além de medidas gerais e dietéticas, inclui tratamento farmacológico e/ou cirurgia. Ambos podem ser eficazes, mas apresentam elevado custo financeiro. O tratamento com fármacos pode apresentar baixa aderência medicamentosa e a cirurgia tem baixa, mas não desprezível, morbidade e mortalidade. Idealmente o tratamento da DRGE deveria ser eficaz, com baixo risco e com baixo custo. Objetivos: 1. Desenvolver um modelo experimental em suínos para o estudo do Refluxo Gastroesofágico através da Pressão e do Volume de Vazão Gástricos; 2. Avaliar a eficácia do implante endoscópico de PMMA ao nível do Esfíncter Esofágico Inferior (EEI) para aumentar a Pressão de Vazão Gástrica, o Volume de Vazão Gástrico e a Pressão Basal do EEI; 3. Descrever as reações histológicas associadas ao implante de PMMA. Material e Métodos: Suínos da raça Large White, do sexo feminino com 8 semanas de vida foram estudados no experimento. Foi realizada manometria do esfíncter esofágico inferior com registro da pressão basal com cateter de perfusão com água e técnica de retirada lenta. Calculou-se, também, a extensão do EEI. Após, gastrostomia era realizada com colocação intragástrica da extremidade distal de uma sonda de Foley com três vias e um cateter de pHmetria esofágica introduzido via oral com o sensor distal posicionado 5 cm acima do bordo superior de esfíncter esofágico inferior. Iniciava-se, XXentão, a infusão contínua no estômago de uma solução de HCl a 0,02N com medida e registro simultâneos da Pressão e do Volume de Vazão Gástricos e do pH esofágico. Definiu-se a Pressão de Vazão Gástrica (PVG) e o Volume de Vazão Gástrico quando houve brusca e sustentada acidificação do esôfago distal (pH<3). Após, introduzia-se um tubo metálico (Tubo Introdutor ou TI) via oral e, em seguida, o endoscópio, seguindo ambos até o esôfago distal. Cateter de nylon com agulha calibre 16 era introduzido pelo tubo introdutor e o PMMA implantado na área correspondente ao EEI com uma pistola dosadora volumétrica que permitia a injeção de volumes prédeterminados em 3 ou 4 pontos da submucosa (total por ponto = 0,73 ml de PMMA). As medidas de PVG, VVG e pressão basal do EEI foram repetidas após 28 dias, sacrificando-se os animais e removendo-se esôfago médio e distal, junção esofagogástrica, fundo e corpo gástricos para estudo histológico. Previamente ao experimento, três projetos pilotos foram desenvolvidos. O primeiro designado como “Projeto Piloto: Refluxo Gastroesofágico por pHmetria de 24H” (RGE 24H) buscou a via de acesso transnasal para registro de pHmetria por 24 horas. No segundo, designado “Projeto Piloto: Esofagostomia”, para confirmação de refluxo gastroesofágico espontâneo em suínos da raça Large White, utilizou-se anestesia com associação de tiletamina 125 mg e zolazepam 125 mg em combinação com um sedativo, a xilazina a 2% . A pHmetria foi mantida por 24 horas. Os dados descritos por Kadirkamanathan et al. não foram reproduzidos no nosso laboratório. Optou-se então pela realização de um terceiro projeto, “Projeto Piloto: Gastrostomia” para induzir refluxo gastroesofágico através de um modelo experimental e testar a reprodutibilidade da Pressão de Vazão Gástrica. Obteve-se sucesso. Resultados: Trinta e sete animais foram estudados em 60 intervenções no Laboratório de motilidade experimental. O projeto piloto “RGE:24h” foi abandonado após perda de 5 animais pela anestesia com halotano e, solucionada esta questão, a sistemática infecção do tecido celular subcutâneo nasal, dificultando a manutenção do cateter de phmetria em outros 5 animais. No “Projeto Piloto: Esofagostomia”, em cinco animais estudados (cada animal em duas ocasiões diferentes) não se reproduziram os achados de refluxo espontâneo. No terceiro projeto “Projeto Piloto: Gastrostomia” quatro animais foram estudados em dois momentos diferentes obtendo-se sucesso e reprodutibilidade XXIdos dados. Criado o modelo experimental, quatorze suínos foram submetidos ao experimento com implante endoscópico de PMMA com os seguintes resultados: A média dos pesos com 8 semanas de idade foi 14,98 ± 2,43 e 28 dias após o implante, 20,26 ± 3,68. O ganho ponderal foi considerado normal para a espécie. A Pressão de Vazão Gástrica média no dia 1 foi 8,08 mmHg e no dia 28, 10,69mmHg (Teste t de Student: t = 2,72 gl = 13 p = 0,017). Os Volumes de Vazão Gástricos médios foram: 392,86 ml para o dia 1 e 996,71 ml no dia 28 (teste t de Student: t = 11,66 gl = 13 p< 0,001). A Pressão Basal do EEI e o comprimento do esfíncter, não apresentaram diferenças estatisticamente significativas. PMMA foi identificado como depósitos de grandes vacúolos no tecido intersticial ao exame histológico da junção esofagogástrica associado a histiócitos, plasmócitos e presença de células gigantes tipo Langhans indicando reação tecidual de corpo estranho em todos os animais. Fibrose e macrófagos com vacúolos intracelulares estiveram presentes em menor freqüência. Conclusões: 1. O modelo experimental, em suínos, desenvolvido viabilizou o estudo do Refluxo Gastroesofágico através da Pressão de Vazão Gástrica e Volume de Vazão Gástrico; 2. O implante de PMMA, no presente estudo, aumentou a Pressão de Vazão e o Volume de Vazão Gástricos, mas não a Pressão Basal do EEI, nem tampouco aumentou o seu comprimento; 3. Depósito de PMMA implantado e evidência de processo inflamatório crônico e reação tecidual de corpo estranho foi encontrada no local do implante de PMMA. Macrófagos com vacúolos intracelulares e fibrose foram encontrados com menos freqüência.

American visceral leishmaniasis is a zoonosis caused by Leishmania infantum and transmitted by the bite of the sand flies Lutzomia longipalpis.The main domestic reservoir is the dog, while foxes and opposums are the known wild reservoirs. However, identification of natural infections with L. infantum in rodents appears for need of investigating the participation of these rodents how source of infection of the parasite. In the present work the Leishmania infantum infection was investigated in rodents captured in Rio Grande do Norte, aiming at to offer subsidies to the understanding of the epidemic chains of LVA in the State. Thirteen Galea spixii were distributed in four groups, being G1 the group control with four animals and the others, G2, G3 and G4, with three animals each. Those animals were intraperitoneally inoculated with 107 promastigotas of L. infantum and accompanied for, respectively, 30, 90 and 180 days. Weekly the animals were monitored as for the corporal weight and rectal temperature. At the end of each stipulated period the animals were killed. Blood were used for determination of the parameters biochemical and haematological, PCR, ELISA, microscopic examination and cultivation in NNN medium. Liver, spleen and lymph node were used in Giemsa-stained impression and cultivation in NNN medium. Liver and spleen fragments were still used in PCR and histopathological, respectively. At the same time 79 rodents of the species Rattus rattus, Bolomys lasiurus, Oligoryzomys nigripis, Oryzomys subflavus and Trichomys apereoides were captured in the Municipal districts of Brejinho, Campo Grande, Coronel Ezequiel, Passa e Fica and Vázea for identification of natural infection with L. infantum. Evidence of infection was checked by direct examination of Giemsa-stained impression of liver, spleen and blood and culture of these tissues in NNN medium. Antibodies were researched by ELISA. They were not found differences among the weigh corporal final, rectal temperature and biochemical and haematological parameters of the Galea spixii controls and infected. The rectal temperature of the animals varied from 36OC to 40OC. For the first time values of the haematocrit (33,6% to 42,8%), hemoglobin (10,2 to 14,5g/dl), erythrocyts number (4,67x106 to 6,90x106/mm3), total leukocytes (0,9x103 to 9,2x103/mm3), platelets (49x103 to 509x103/mm3) total proteins (1,56 to 6,06 g/dl), albumin (1,34 to 3,05 g/dl) and globulins (0,20 to 3,01 g/dl) of the Galea spixii were determined. The lymphocytes were the most abundant leucocytes. Infection for L. infantum was diagnosed in two animals euthanasied 180 days after the infection. In one of the animals was also identified antibodies anti-Leishmania. The parasite was not found in none of the five other species of rodents captured. Galea spixii are resistant to the infection for L. infantum and they are not good models for the study for visceral leishmaniose, although they can act as infection sources. More studies are necessary to determine the paper of the rodents in the epidemic chain of transmission of the visceral leishmaniose in the State of Rio Grande do Norte

Three methods of sexual identification were evaluated in Nile tilapia (Oreochromis niloticus) at ages of 30, 60 and 90 days that were submitted to sexual reversion under administration of androgen hormone 17 alpha-methyltestosterone added to crumble diets with different granules sizes (0.25, 0.35 and 0.50 mm) in dosage of 60mg/kg. The techniques of sexual identification employed were: a) Macroscopic examination of urogenital papilla; b) Microscopic examination of gonads through stain with carmine-acetate and c) Microscopic examination of gonads through histological routine. In the presence of three methods of sexual identification evaluated, histology of gonads has shown the most confident diagnosis. The grain diameters of rations did not interfere in morphological characteristics of gonads, neither in secondary sexual characters of fish.

A Erliquiose é uma doença zoonótica causada por bactérias gram-negativas e intracelulares obrigatórias. A Anaplasmose Granulocítica Equina - AGE (anteriormente denominada Erliquiose Granulocítica Equina, EGE) é uma enfermidade sazonal, normalmente auto-limitante em equinos. No Brasil, existem poucos relatos deste agente erliquial, bem como de seus vetores naturais. Atualmente, veterinários têm levantado a suspeita de casos de AGE em equinos com sinais clínicos sugestivos de erliquiose e não responsivos ao tratamento para a piroplasmose equina. O objetivo do presente estudo foi identificar equinos expostos a A. phagocytophilum por meio de técnicas sorológicas e moleculares. Vinte amostras de sangue e soro de equinos da região Centro-oeste do Brasil foram avaliados por meio do exame microscópico de capa leucocitária, ensaio imunoenzimático indireto (ELISA), reação de imunofluorescência indireta (RIFI) e reação em cadeia da polimerase (nested PCR). Adicionalmente, o diagnóstico sorológico de Theileria equi pela RIFI e ELISA foram realizados, assim como o diagnóstico molecular pelo nPCR. Treze (65%) amostras de soro foram positivas para A. phagocytophilum pelo teste de ELISA, entretanto nenhum equino foi positivo pelo exame microscópico da capa leucocitária ou nPCR. Anticorpos IgG anti-T. equi foram detectados em 18 (90%) e 17 (85%) equinos pela RIFI e ELISA, respectivamente e o agente foi detectado em 9 (45%) animais pelo nPCR. Estes dados sugerem importante informação para o entendimento da ocorrência da AGE e piroplasmose equina no Centro-oeste do Brasil.

OBJETIVO: Comparar uma malha comercial de poliéster com o pericárdio bovino preservado em glicerina na reconstituição de defeitos da parede abdominal. MÉTODOS: Foram utilizados 30 ratos, divididos em dois grupos eqüitativos. Efetuou-se uma excisão retangular de 2,5 x 2 cm, incluindo toda a musculatura abdominal e peritônio. No grupo I a parede abdominal foi reparada com malha de poliéster e no grupo II com pericárdio bovino conservado em glicerina. Os animais foram sacrificados aos 15, 60 e 90 dias de pós-operatório, sendo o local cirúrgico avaliado macroscópica e histologicamente. RESULTADOS: Os animais do grupo I apresentaram aderências mais severas e em maior número quando comparados aos do grupo II; porém, sem comprometimento funcional. A análise histológica revelou incorporação dos tecidos aos implantes, com maior resposta fibroblástica nos animais do grupo I. CONCLUSÃO: A malha de poliéster oferece maior resistência estrutural e resposta fibroblástica mais intensa; contudo, promove grande quantidade de aderências às vísceras abdominais, quando comparada ao pericárdio bovino.

Objectives: To evaluate the possible association between microorganisms present in the cervical secretions and amniotic fluid of pregnant women with preterm premature rupture of membranes (PPROM), and histologic chorioamnionitis. Methods: Thirty-seven pregnant women with PPROM and 21 healthy pregnant women were studied. Secretions from the cervical canal and amniotic fluid were collected to isolate microorganisms present in the genital tract. Cervical smears were Gram stained and evaluated microscopically. At delivery, chorioamniotic membranes were collected for histopathologic analysis. Results: Microscopic examination of the cervical secretion smears obtained from the PPROM group showed a low rate of Lactobacillus species, large numbers of leukocytes, and a wide diversity of microorganisms compared with the control group. The PPROM group presented an 80% rate of chorioamnionitis. Staphylococcus aureus isolation in cervical secretion was associated with intense inflammatory infiltrate in the membranes and might play a role in the pathogenesis of PPROM. Conclusions: the low colonization of cervical flora by Lactobacillus species associated with an intense leukocyte infiltrate detected in Gram-stained cervical smears can be considered a rapid method of detecting chorioamnionitis in pregnant women with PPROM. Background: Salivary mucocele is an accumulation of saliva in a single or multiloculated cavity lined by connective tissue that is contiguous to a salivary gland-duct complex and is the most common condition affecting the salivary glands in dogs. Occasionally, different types of metaplastic lesions, such as squamous and osseous metaplasia - which are rare lesions in animals - can be observed in association with salivary mucocele.Case presentation: A right facial enlargement was suddenly observed in a 4-year-old non-spayed female Shih-Tzu dog. The lesion presented itself as a soft and fluctuant mass located in the right side of the face near to the neck. Histologically, the mass consisted of a cavitary formation without an epithelial lining. Additionally, microscopic examination revealed the presence of osteoid-producing cells which gave rise to areas of bone formation, probably induced by irritation due to the presence sialoliths. Such cells and bone formations were also present in the cavity wall, consequently leading us to classify the condition as a salivary mucocele with osseous metaplasia.Conclusions: In the present case, the pathogenesis was probably associated with the presence of sialoliths, which can behave as etiological agents for the metaplastic lesion. The occurrence of osteoid metaplasia is a rare peculiar condition in the canine salivar y gland, and due to the rarity and lack of information about this specific disease, no clinical data can yet be associated with the development of salivary mucocele with osseous metaplasia in dogs.

Background: Spontaneous gastrointestinal neoplasms in non-human primates are commonly seen in aged individuals. Due to genetic similarities between human and non-human primates, scientists have shown increasing interest in terms of comparative oncology studies.Case presentation: The present study is related to a case of an intestinal leiomyoma in a black crested macaque (Macaca nigra), kept on captivity by Mateca a Zoo, Pereira City, Colombia. The animal had abdominal distension, anorexia, vomiting, diarrhea and behavioral changes. Clinical examination showed an increased volume in the upper right abdominal quadrant caused by a neoplastic mass. The patient died during the surgical procedure. Necropsy revealed several small nodules in the peritoneum with adhesion to different portions of the small and large intestines, liver, stomach and diaphragm. Tissue samples were collected, routinely processed and stained by H&E. Microscopic examination revealed a mesenchymal tumor limited to tunica muscularis, resembling normal smooth muscle cells. Neoplastic cells were positive for alpha-smooth muscle actin and vimentin, and negative for cytokeratin AE1/AE3 by immunohistochemistry. Those morphological and immunohistochemical findings allowed to diagnose the intestinal leiomyoma referred above.Conclusion: Neoplastic diseases in primates have multifaceted causes. Their manifestations are understudied, leading to a greater difficulty in detection and measurement of the real impact provides by this disease.

OBJETIVO: Avaliar as lesões proliferativas que se desenvolvem na mucosa gástrica de ratos Wistar após modelo específico de refluxo duodeno-gástrico. MÉTODOS: Foram utilizados 75 ratos adultos machos divididos em três grupos experimentais: o grupo I (controle) submetido a gastrotomia na parede posterior do estômago glandular (25 animais); o grupo II (RDG), foi submetido a gastrojejunoanastomose látero-lateral na parede posterior do estômago glandular (25 animais) e o grupo III (RDG-P) submetido a gastrojejunoanastomose látero-lateral na parede posterior do estômago glandular, com secção e fechamento da alça (25 animais). Os animais foram observados durante 36 semanas, após o que foram realizados estudos macroscópicos e microscópicos da anastomose gastrojejunal, da região pré-pilórica e região escamosa do estômago. RESULTADOS: Os animais do Grupo I não apresentaram nenhum tipo de lesão. No grupo II observou-se 40% de lesões do tipo hiperplasia adenomatosa na anastomose e 12% de hiperplasia escamosa. No grupo III obteve-se 40% de hiperplasia adenomatosa na mucosa pré-pilórica, 72 % de hiperplasia adenomatosa na mucosa da anastomose, 20% de hiperplasia escamosa e 12 % de adenocarcinoma. CONCLUSÕES: O refluxo duodeno-gástrico induz a alta freqüência de lesões proliferativas na mucosa adjacente à anastomose gastrojejunal ou na mucosa pré-pilórica e o adenocarcinoma é um evento raro neste modelo experimental.
