As indústrias têm buscado constantemente reduzir gastos operacionais, visando o aumento do lucro e da competitividade. Para alcançar essa meta, são necessários, dentre outros fatores, o projeto e a implantação de novas ferramentas que permitam o acesso às informações relevantes do processo de forma precisa, eficiente e barata. Os sensores virtuais têm sido aplicados cada vez mais nas indústrias. Por ser flexível, ele pode ser adaptado a qualquer tipo de medição, promovendo uma redução de custos operacionais sem comprometer, e em alguns casos até melhorar, a qualidade da informação gerada. Como estão totalmente baseados em software, não estão sujeitos a danos físicos como os sensores reais, além de permitirem uma melhor adaptação a ambientes hostis e de difícil acesso. A razão do sucesso destes tipos de sensores é a utilização de técnicas de inteligência computacional, as quais têm sido usadas na modelagem de vários processos não lineares altamente complexos. Este trabalho tem como objetivo estimar a qualidade da alumina fluoretada proveniente de uma Planta de Tratamento de Gases (PTG), a qual é resultado da adsorção de gases poluentes em alumina virgem, via sensor virtual. O modelo que emula o comportamento de um sensor de qualidade de alumina foi criado através da técnica de inteligência computacional conhecida como Rede Neural Artificial. As motivações deste trabalho consistem em: realizar simulações virtuais, sem comprometer o funcionamento da PTG; tomar decisões mais precisas e não baseada somente na experiência do operador; diagnosticar potenciais problemas, antes que esses interfiram na qualidade da alumina fluoretada; manter o funcionamento do forno de redução de alumínio dentro da normalidade, pois a produção de alumina de baixa qualidade afeta a reação de quebra da molécula que contém este metal. Os benefícios que este projeto trará consistem em: aumentar a eficiência da PTG, produzindo alumina fluoretada de alta qualidade e emitindo menos gases poluentes na atmosfera, além de aumentar o tempo de vida útil do forno de redução.

This work brings to its content proposed deployment of a system of quality management in paving asphalt based on the elaboration of a basic training to meet those entering this area of civil engineering at a technical manual for the implementation of major services running on paving of roads. The training was designed to serve both those entering the field of asphalt paving, synthesizing the complex processes involved in paving services and providing an overview of the steps in implementation thereof, as well as entrants must meet the enterprise user of this system, providing them an idea of the method used by the company designing the paving of roads. The technical manual presented in turn, has 8 (eight) of the main services components of a work of paving, which are: Mobilization and Maintenance of construction site, Regularization and subfloor preparation, execution or sub-base BGS base (graded gravel plain), Execution of sub-base or base BGTC (graded gravel treated with cement), Implementation of subbase or base Macadam hydraulic Priming bituminous waterproofing, asphalt binder Priming and finally the execution flexible pavement - HMA (hot milled asphalt concrete). These services are presented in the form of IT-Education work, this seven items are presented that guide the performance of services, thus providing overall guidelines to perform the services described in it. The development of IT's was based on experiences in day-to-day paving near the theoretical precepts presented in the literature, so the conditions specified in these documents aim to always guide the implementation of services in general, giving the process a versatility major operation, as this work takes into account the dynamics of such services, as well as the variety of their execution according to local conditions and specified in the project. It is known that the process of capitalist restructuring resulted in socio-spatial transformations in various geographical scales. Space is continually diversifying and redefining their roles, driven mostly by external agents. This search analyzes how and intensities are consolidating these new refunctionalisation the District of Gardenia city of Rancharia-SP.The study brings some concepts in the definition of small town, which are fundamental to the definition of the area that is the object of our study. Some features are explained in the District, such as location, history of economic activities preterit in a record that cover the 1950s to the present day. From this record ,it will be exposed and analyzed the services (private and collective goods) offered to the population in order to highlight their importance for better social equity. The information for your preparation of this work were supported mostly on reports and records historical and contextualized with specific bibliography. In developing the search is portrayed as the district Gardenia was modifying its economic activities, physical space and how it is currently being structured with tourist activity. There is also intended to show that even though the district with urban core urban functions simple in content space is consolidating complex processes in its various dimensions

The vast majority of known proteins have not yet been experimentally characterized and little is known about their function. The design and implementation of computational tools can provide insight into the function of proteins based on their sequence, their structure, their evolutionary history and their association with other proteins. Knowledge of the three-dimensional (3D) structure of a protein can lead to a deep understanding of its mode of action and interaction, but currently the structures of <1% of sequences have been experimentally solved. For this reason, it became urgent to develop new methods that are able to computationally extract relevant information from protein sequence and structure. The starting point of my work has been the study of the properties of contacts between protein residues, since they constrain protein folding and characterize different protein structures. Prediction of residue contacts in proteins is an interesting problem whose solution may be useful in protein folding recognition and de novo design. The prediction of these contacts requires the study of the protein inter-residue distances related to the specific type of amino acid pair that are encoded in the so-called contact map. An interesting new way of analyzing those structures came out when network studies were introduced, with pivotal papers demonstrating that protein contact networks also exhibit small-world behavior. In order to highlight constraints for the prediction of protein contact maps and for applications in the field of protein structure prediction and/or reconstruction from experimentally determined contact maps, I studied to which extent the characteristic path length and clustering coefficient of the protein contacts network are values that reveal characteristic features of protein contact maps. Provided that residue contacts are known for a protein sequence, the major features of its 3D structure could be deduced by combining this knowledge with correctly predicted motifs of secondary structure. In the second part of my work I focused on a particular protein structural motif, the coiled-coil, known to mediate a variety of fundamental biological interactions. Coiled-coils are found in a variety of structural forms and in a wide range of proteins including, for example, small units such as leucine zippers that drive the dimerization of many transcription factors or more complex structures such as the family of viral proteins responsible for virus-host membrane fusion. The coiled-coil structural motif is estimated to account for 5-10% of the protein sequences in the various genomes. Given their biological importance, in my work I introduced a Hidden Markov Model (HMM) that exploits the evolutionary information derived from multiple sequence alignments, to predict coiled-coil regions and to discriminate coiled-coil sequences. The results indicate that the new HMM outperforms all the existing programs and can be adopted for the coiled-coil prediction and for large-scale genome annotation. Genome annotation is a key issue in modern computational biology, being the starting point towards the understanding of the complex processes involved in biological networks. The rapid growth in the number of protein sequences and structures available poses new fundamental problems that still deserve an interpretation. Nevertheless, these data are at the basis of the design of new strategies for tackling problems such as the prediction of protein structure and function. Experimental determination of the functions of all these proteins would be a hugely time-consuming and costly task and, in most instances, has not been carried out. As an example, currently, approximately only 20% of annotated proteins in the Homo sapiens genome have been experimentally characterized. A commonly adopted procedure for annotating protein sequences relies on the "inheritance through homology" based on the notion that similar sequences share similar functions and structures. This procedure consists in the assignment of sequences to a specific group of functionally related sequences which had been grouped through clustering techniques. The clustering procedure is based on suitable similarity rules, since predicting protein structure and function from sequence largely depends on the value of sequence identity. However, additional levels of complexity are due to multi-domain proteins, to proteins that share common domains but that do not necessarily share the same function, to the finding that different combinations of shared domains can lead to different biological roles. In the last part of this study I developed and validate a system that contributes to sequence annotation by taking advantage of a validated transfer through inheritance procedure of the molecular functions and of the structural templates. After a cross-genome comparison with the BLAST program, clusters were built on the basis of two stringent constraints on sequence identity and coverage of the alignment. The adopted measure explicity answers to the problem of multi-domain proteins annotation and allows a fine grain division of the whole set of proteomes used, that ensures cluster homogeneity in terms of sequence length. A high level of coverage of structure templates on the length of protein sequences within clusters ensures that multi-domain proteins when present can be templates for sequences of similar length. This annotation procedure includes the possibility of reliably transferring statistically validated functions and structures to sequences considering information available in the present data bases of molecular functions and structures.

Der N-methyl-D-aspartat-Rezeptor (NMDA), als Vertreter ionotroper Glutamat-Rezeptoren, ist essentiell für physiologische Lern- und Gedächtnisvorgänge und eine krankhafte Überaktivierung wird als potentielle Ursache für eine Reihe von akuten und chronischen neurodegenerativen Erkrankungen angesehen. Hierbei sind für die akuten Erkrankungen vor allem der Schlaganfall und für die chronischen Erkrankungen Morbus Parkinson sowie die Alzheimer´sche Demenz zu nennen. Durch seine einzigartige spannungsabhängige Mg2+-Blockade und der Notwendigkeit der gleichzeitigen Anwesenheit der endogenen Liganden Glutamat und Glycin zur Rezeptoraktivierung, stellt dieser Rezeptorkomplex daher ein sehr interessantes molekulares Target dar. NMDA-Rezeptor-Antagonisten der Glycin-Bindungsstelle und der verschiedenen allosterischen Bindungsstellen könnten als Neuroprotektiva bei den verschiedenen Krankheiten eine symptomatische Verbesserung bewirken und zur Therapie eingesetzt werden. Eine visuelle Darstellung des Rezeptors im Rahmen von Vorsorgeuntersuchungen ist jedoch derzeit nicht möglich. Zur Visualisierung dieser Prozesse mittels der Positronen-Emissions-Tomographie (PET) wurden basierend auf einer Hydantoin-substituierten Indol-2-carbonsäure als Leitstruktur, im Rahmen dieser Arbeit Fluorethoxy- und Methoxy-substituierte Derivate dargestellt und in pharmazeutischen und radiopharmazeutischen Studien evaluiert. Dazu wurde die Affinität und Spezifität zum Rezeptor in einem [3H]MDL-105,519 Rezeptorbindungsassay und die Lipophilie als Parameter für die Hirngängigkeit ermittelt. Anhand dieser Resultate wurden geeignete Markierungsvorläufer synthetisiert, welche eine phenolische Hydroxylfunktion besitzen und eine radioaktive Markierung mit den sekundären Markierungsvorläufern 2-[18F]Fluorethyltosylat ([18F]FETos) und [11C]Methyliodid ([11C]CH3I) ermöglichen. Unter Verwendung von 4,6-Dichlor-3-((3-(4-hydroxyphenyl)-2,4-dioxoimidazolidin-1-yl)methyl)-indol-2-carbonsäure wurde in einer Einstufenreaktion mit [18F]FETos die Zielverbindung 4,6-Dichlor-3-((3-(4-(2-[18F]fluorethoxy)phenyl)-2,4-dioxoimidazolidin-1-yl)methyl)-indol-2-carbonsäure in radiochemischen Ausbeuten von 6 % erhalten. Daher wurde eine alternative Markierung des Ethylester-geschützten Derivates 4,6-Dichlor-3-((3-(4-hydroxyphenyl)-2,4-dioxoimidazolidin-1-yl)methyl)-indol-2-carbonsäureethylester in einer Zweistufensynthese mit [18F]FETos und [11C]CH3I untersucht. Unter Verwendung dieser Strategie wurden unter optimierten Bedingungen 4,6-Dichlor-3-((3-4-(2-[18F]fluorethoxy)phenyl)-2,4-dioxoimidazolidin-1-yl)methyl)-indol-2-carbonsäureethylester und 4,6-Dichlor-3-((3-(4-[11C]methoxy-phenyl)-2,4-dioxoimidazolidin-1-yl)-methyl)-indol-2-carbonsäureethylester in radiochemischen Ausbeuten von 27 – 38 % erhalten. Die anschließende Entfernung der Schutzgruppe führte unter Bildung von Neben- und Zersetzungsreaktionen zu 4,6-Dichlor-3-((3-(4-(2-[18F]fluorethoxy)-phenyl)-2,4-dioxoimidazolidin-1-yl)methyl)-indol-2-carbonsäure und 4,6-Dichlor-3-((3-(4-[11C]methoxyphenyl)-2,4-dioxoimidazolidin-1-yl)methyl)-indol-2-carbonsäure in radiochemischen Gesamtausbeuten von 5 – 7 %. Die Überprüfung des biochemischen Konzepts in vivo durch µ-PET-Studien und durch autoradiographische Experimente an Rattenhirnschnitten, deuten auf eine niedrige in vivo-Aktivität hin, welche sich auf eine nicht ausreichende Passage der Blut-Hirn-Schranke zurückführen lässt.

Many industries and academic institutions share the vision that an appropriate use of information originated from the environment may add value to services in multiple domains and may help humans in dealing with the growing information overload which often seems to jeopardize our life. It is also clear that information sharing and mutual understanding between software agents may impact complex processes where many actors (humans and machines) are involved, leading to relevant socioeconomic benefits. Starting from these two input, architectural and technological solutions to enable “environment-related cooperative digital services” are here explored. The proposed analysis starts from the consideration that our environment is physical space and here diversity is a major value. On the other side diversity is detrimental to common technological solutions, and it is an obstacle to mutual understanding. An appropriate environment abstraction and a shared information model are needed to provide the required levels of interoperability in our heterogeneous habitat. This thesis reviews several approaches to support environment related applications and intends to demonstrate that smart-space-based, ontology-driven, information-sharing platforms may become a flexible and powerful solution to support interoperable services in virtually any domain and even in cross-domain scenarios. It also shows that semantic technologies can be fruitfully applied not only to represent application domain knowledge. For example semantic modeling of Human-Computer Interaction may support interaction interoperability and transformation of interaction primitives into actions, and the thesis shows how smart-space-based platforms driven by an interaction ontology may enable natural ad flexible ways of accessing resources and services, e.g, with gestures. An ontology for computational flow execution has also been built to represent abstract computation, with the goal of exploring new ways of scheduling computation flows with smart-space-based semantic platforms.

This thesis was driven by the ambition to create suitable model systems that mimic complex processes in nature, like intramolecular transitions, such as unfolding and refolding of proteins, or intermolecular interactions between different cell compo-nents. Novel biophysical approaches were adopted by employing atomic force mi-croscopy (AFM) as the main measurement technique due to its broad diversity. Thus, high-resolution imaging, adhesion measurements, and single-molecule force distance experiments were performed on the verge of the instrumental capabilities. As first objective, the interaction between plasma membrane and cytoskeleton, me-diated by the linker protein ezrin, was pursued. Therefore, the adsorption process and the lateral organization of ezrin on PIP2 containing solid-supported membranes were characterized and quantified as a fundament for the establishment of a biomimetic model system. As second component of the model system, actin filaments were coated on functionalized colloidal probes attached on cantilevers, serving as sensor elements. The zealous endeavor of creating this complex biomimetic system was rewarded by successful investigation of the activation process of ezrin. As a result, it can be stated that ezrin is activated by solely binding to PIP2 without any further stimulating agents. Additional cofactors may stabilize and prolong the active conformation but are not essentially required for triggering ezrin’s transformation into an active conformation. In the second project, single-molecule force distance experiments were performed on bis-loop tetra-urea calix[4]arene-catenanes with different loading rates (increase in force per second). These macromolecules were specifically designed to investigate the rupture and rejoining mechanism of hydrogen bonds under external load. The entangled loops of capsule-like molecules locked the unbound state of intramolecular hydrogen bonds mechanically, rendering a rebinding observable on the experimental time scale. In conjunction with Molecular Dynamics simulations, a three-well potential of the bond rupture process was established and all kinetically relevant parameters of the experiments were determined by means of Monte Carlo simulations and stochastic modeling. In summary, it can be stated that atomic force microscopy is an invaluable tool to scrutinize relevant processes in nature, such as investigating activation mechanisms in proteins, as shown by analysis of the interaction between F-actin and ezrin, as well as exploring fundamental properties of single hydrogen bonds that are of paramount interest for the complete understanding of complex supramolecular structures.

Aim: Previous studies revealed that diversification events in the western clade of the alpine Primula sect. Auricula were concentrated in the Quaternary cold periods. This implies that allopatric speciation in isolated glacial refugia was the most common mode of speciation. In the first part of the present dissertation, this hypothesis is further investigated by locating refugial areas of two sister species, Primula marginata & P. latifolia during the last glacial maximum, 21,000 years ago. In the second part, the glacial and postglacial history of P. hirsuta and P. daonensis is investigated. Location: European Alps. Methods: Glacial refugia were located using species distribution models, which are projected to last glacial maximum climate. These refugia are validated with geographic distribution patterns of intra-specific genetic diversity, rarity and variation. Results 1) Speciation: Glacial refugia of the sister taxa Primula marginata and P. latifolia were largely separated, only a small overlapping zone at the southern margin of the former glacier in the Maritime Alps exists. This overlapping zone is too small to indicate sympatric speciation. The largely separated glacial distribution of both species rather confirms our hypothesis of allopatric speciation in isolated glacial refugia. Results 2) Glacial and postglacial history: Surprizingly, the modelled potential refugia of three out of four Primula species are situated within the former ice-shield, except for P. marginata. This indicates that peripheral and central nunataks played an important role for the glacial survival in P. latifolia, P. hirsuta and P. daonensis, while peripheral refugia outside the maximum extend of the glacier were crucial in P. marginata. In P. hirsuta and P. latifolia SDMs allowed to exclude several hypothetical refugial areas that overlap with today’s distribution as potential refugia for the species. In P. marginata, hypothetical refugial areas at the periphery of the former ice-shield that overlap with today’s distribution were confirmed by the models. The results from the SDMs are confirmed by population genetic patterns in three out of four species. P. daonensis represents an exception, where population genetic data contradict the SDMs. Main conclusions: Species distribution models provide species specific scenarios of glacial distribution and postglacial re-colonization, which can be validated using population genetic analyses. This combined approach is useful and helps to understand the complex processes that have lead to the genetic and floristic patterns of biodiversity that is found today in the Alps.

Aseptic loosening of metal implants is mainly attributed to the formation of metal degradation products. These include particulate debris and corrosion products, such as metal ions (anodic half-reaction) and ROS (cathodic half-reaction). While numerous clinical studies describe various adverse effects of metal degradation products, detailed knowledge of metal-induced cellular reactions, which might be important for possible therapeutic intervention, is not comprehensive. Since endothelial cells are involved in inflammation and angiogenesis, two processes which are critical for wound healing and integration of metal implants, the effects of different metal alloys and their degradation products on these cells were investigated. Endothelial cells on Ti6Al4V alloy showed signs of oxidative stress, which was similar to the response of endothelial cells to cathodic partial reaction of corrosion induced directly on Ti6Al4V surfaces. Furthermore, oxidative stress on Ti6Al4V alloy reduced the pro-inflammatory stimulation of endothelial cells by TNF-α and LPS. Oxidative stress and other stress-related responses were observed in endothelial cells in contact with Co28Cr6Mo alloy. Importantly, these features could be reduced by coating Co28Cr6Mo with a TiO2 layer, thus favouring the use of such surface modification in the development of medical devices for orthopaedic surgery. The reaction of endothelial cells to Co28Cr6Mo alloy was partially similar to the effects exerted by Co2+, which is known to be released from metal implants. Co2+ also induced ROS formation and DNA damage in endothelial cells. This correlated with p53 and p21 up-regulation, indicating the possibility of cell cycle arrest. Since CoCl2 is used as an hypoxia-mimicking agent, HIF-1α-dependence of cellular responses to Co2+ was studied in comparison to anoxia-induced effects. Although important HIF-1α-dependent genes were identified, a more detailed analysis of microarray data will be required to provide additional information about the mechanisms of Co2+ action. All these reactions of endothelial cells to metal degradation products might play their role in the complex processes taking place in the body following metal device implantation. In the worst case this can lead to aseptic loosening of the implant and requirement for revision surgery. Knowledge of molecular mechanisms of metal-induced responses will hopefully provide the possibility to interfere with undesirable processes at the implant/tissue interface, thus extending the life-time of the implant and the overall success of metal implant applications.
