The striking illusions produced by simultaneous brightness contrast generally are attributed to the center-surround receptive field organization of lower order neurons in the primary visual pathway. Here we show that the apparent brightness of test objects can be either increased or decreased in a predictable manner depending on how light and shadow are portrayed in the scene. This evidence suggests that perceptions of brightness are generated empirically by experience with luminance relationships, an idea whose implications we pursue in the accompanying paper.

In this second part of our study on the mechanism of perceived brightness, we explore the effects of manipulating three-dimensional geometry. The additional scenes portrayed here demonstrate that the same luminance profile can elicit different sensations of brightness as a function of how the objects in the scene are arranged in space. This further evidence confirms the implication of the scenes presented in the accompanying paper, namely that sensations of relative brightness—including standard demonstrations of simultaneous brightness contrast—cannot arise by computations of local contrast. The most plausible explanation of the full range of perceptual phenomena we have described is an empirical strategy that links the luminance profile in a visual stimulus with an association (the percept) that represents the profile’s most probable real-world source.

Somatic mosaicism has been observed previously in the lymphocyte population of patients with Fanconi anemia (FA). To identify the cellular origin of the genotypic reversion, we examined each lymphohematopoietic and stromal cell lineage in an FA patient with a 2815–2816ins19 mutation in FANCA and known lymphocyte somatic mosaicism. DNA extracted from individually plucked peripheral blood T cell colonies and marrow colony-forming unit granulocyte–macrophage and burst-forming unit erythroid cells revealed absence of the maternal FANCA exon 29 mutation in 74.0%, 80.3%, and 86.2% of colonies, respectively. These data, together with the absence of the FANCA exon 29 mutation in Epstein–Barr virus-transformed B cells and its presence in fibroblasts, indicate that genotypic reversion, most likely because of back mutation, originated in a lymphohematopoietic stem cell and not solely in a lymphocyte population. Contrary to a predicted increase in marrow cellularity resulting from reversion in a hematopoietic stem cell, pancytopenia was progressive. Additional evaluations revealed a partial deletion of 11q in 3 of 20 bone marrow metaphase cells. By using interphase fluorescence in situ hybridization with an MLL gene probe mapped to band 11q23 to identify colony-forming unit granulocyte–macrophage and burst-forming unit erythroid cells with the 11q deletion, the abnormal clone was exclusive to colonies with the FANCA exon 29 mutation. Thus, we demonstrate the spontaneous genotypic reversion in a lymphohematopoietic stem cell. The subsequent development of a clonal cytogenetic abnormality in nonrevertant cells suggests that ex vivo correction of hematopoietic stem cells by gene transfer may not be sufficient for providing life-long stable hematopoiesis in patients with FA.

Somatic mosaicism caused by in vivo reversion of inherited mutations has been described in several human genetic disorders. Back mutations resulting in restoration of wild-type sequences and second-site mutations leading to compensatory changes have been shown in mosaic individuals. In most cases, however, the precise genetic mechanisms underlying the reversion events have remained unclear, except for the few instances where crossing over or gene conversion have been demonstrated. Here, we report a patient affected with Wiskott–Aldrich syndrome (WAS) caused by a 6-bp insertion (ACGAGG) in the WAS protein gene, which abrogates protein expression. Somatic mosaicism was documented in this patient whose majority of T lymphocytes expressed nearly normal levels of WAS protein. These lymphocytes were found to lack the deleterious mutation and showed a selective growth advantage in vivo. Analysis of the sequence surrounding the mutation site showed that the 6-bp insertion followed a tandem repeat of the same six nucleotides. These findings strongly suggest that DNA polymerase slippage was the cause of the original germ-line insertion mutation in this family and that the same mechanism was responsible for its deletion in one of the propositus T cell progenitors, thus leading to reversion mosaicism.

Adaptive reversion of a lac- frameshift mutation in Escherichia coli appears to be due to DNA polymerase errors, implying that DNA is being synthesized although the cells are not dividing. Here we report that the production of adaptive lac+ revertants (i) is much higher when the mutational target is on the F' episome than when it is on the bacterial chromosome; (ii) is enhanced by functions required for conjugation; but (iii) does not require conjugation per se. These results suggest that, in static cells, DNA synthesis is initiated from the conjugal origin of transfer. Mutations may arise as polymerase errors during this synthesis or during synthesis stimulated by recombination among the multiple gene copies.

La hipótesis de la reconsolidación propone que una memoria consolidada entra nuevamente en un estado lábil seguido por un nuevo proceso de estabilización dependiente de síntesis de proteínas, definido como reconsolidación. Durante el proceso de desestabilización, la traza puede actualizarse incorporando nueva información ajustándose a nuevas exigencias ambientales. Esta visión le otorga a la memoria una propiedad flexible y dinámica, contrariamente a lo sugerido por la clásica teoría de la consolidación que definía el trazo de memoria como algo inflexible e inmutable. La reconsolidación de la memoria consta de dos fases: un proceso de labilización o desestabilización, seguido de una fase de re-estabilización. Evidencias recientes de nuestro laboratorio sugieren que la ocurrencia de un estado emocional negativo, como el generado por un evento traumático (estrés) reduce la vulnerabilidad de la traza durante la evocación, es decir la memoria bajo estas condiciones se vuelve resistente a la interferencia y disminuye las probabilidades de la ocurrencia de la reconsolidación. La abstinencia al etanol genera un estado emocional negativo de características similares al generado por estrés en el complejo basolateral de la Amígdala (BLA). El presente proyecto está dirigido a estudiar los mecanismos neurobiológicos implicados en la memoria resistente, en dos áreas cerebrales críticamente involucradas en la memoria de miedo contextual: BLA y el hipocampo dorsal (HD). Se verificará la reactividad a los agentes interferentes (Midazolam y Propranolol) sobre la reconsolidación tanto en animales estresados como en abstinentes. Además, estudiaremos la expresión de las subunidades de receptores NMDA y GABA-A en las áreas mencionadas. Además nos proponemos re-establecer la capacidad dinámica de la traza con herramientas farmacológicas como la infusión local de un agonista parcial de receptores NMDA. Bajo este esquema hipotético, este tratamiento permitirá re-establecer las propiedades biológicas que definen el proceso de labilización/reconsolidación, con la recuperación de la vulnerabilidad a los agentes interferentes de esta fase ante la presentación del recordatorio. En síntesis, proponemos que los cambios originados por la instauración de un estado emocional negativo modelan la dinámica de la memoria resultante al momento de la evocación. We present BVI photometry of 190 galaxies in the central 4 x 3 deg(2) region of the Fornax cluster observed with the Michigan Curtis Schmidt Telescope. Results from the Fornax Cluster Spectroscopic Survey (FCSS) and the Flair-II Fornax Surveys have been used to confirm the membership status of galaxies in the Fornax Cluster Catalogue (FCC). In our catalogue of 213 member galaxies, 92 (43 per cent) have confirmed radial velocities. In this paper, we investigate the surface brightness-magnitude relation for Fornax cluster galaxies. Particular attention is given to the sample of cluster dwarfs and the newly discovered ultracompact dwarf galaxies (UCDs) from the FCSS. We examine the reliability of the surface brightness-magnitude relation as a method for determining cluster membership and find that at surface brightnesses fainter than 22 mag arcsec(-2), it fails in its ability to distinguish between cluster members and barely resolved background galaxies. Cluster members exhibit a strong surface brightness-magnitude relation. Both elliptical (E) galaxies and dwarf elliptical (dE) galaxies increase in surface brightness as luminosity decreases. The UCDs lie off the locus of the relation. B-V and V-I colours are determined for a sample of 113 cluster galaxies and the colour-magnitude relation is explored for each morphological type. The UCDs lie off the locus of the colour-magnitude relation. Their mean V - I colours (similar to1.09) are similar to those of globular clusters associated with NGC 1399. The location of the UCDs on both surface brightness and colour-magnitude plots supports the 'galaxy threshing' model for infalling nucleated dwarf elliptical (dE, N) galaxies.

Using imaging from the Hubble Space Telescope, we derive surface brightness profiles for ultracompact dwarfs in the Fornax Cluster and for the nuclei of dwarf elliptical galaxies in the Virgo Cluster. Ultracompact dwarfs are more extended and have higher surface brightnesses than typical dwarf nuclei, while the luminosities, colors, and sizes of the nuclei are closer to those of Galactic globular clusters. This calls into question the production of ultracompact dwarfs via threshing, whereby the lower surface brightness envelope of a dwarf elliptical galaxy is removed by tidal processes, leaving behind a bare nucleus. Threshing may still be a viable model if the relatively bright Fornax ultracompact dwarfs considered here are descended from dwarf elliptical galaxies whose nuclei are at the upper end of their luminosity and size distributions.

This paper will show that short horizon stock returns for UK portfolios are more predictable than suggested by sample autocorrelation co-efficients. Four capitalisation based portfolios are constructed for the period 1976–1991. It is shown that the first order autocorrelation coefficient of monthly returns can explain no more than 10% of the variation in monthly portfolio returns. Monthly autocorrelation coefficients assume that each weekly return of the previous month contains the same amount of information. However, this will not be the case if short horizon returns contain predictable components which dissipate rapidly. In this case, the return of the most recent week would say a lot more about the future monthly portfolio return than other weeks. This suggests that when predicting future monthly portfolio returns more weight should be given to the most recent weeks of the previous month, because, the most recent weekly returns provide the most information about the subsequent months' performance. We construct a model which exploits the mean reverting characteristics of monthly portfolio returns. Using this model we forecast future monthly portfolio returns. When compared to forecasts that utilise the autocorrelation statistic the model which exploits the mean reverting characteristics of monthlyportfolio returns can forecast future returns better than the autocorrelation statistic, both in and out of sample.

Internal quantum efficiency (IQE) of a blue high-brightness InGaN/GaN light-emitting diode (LED) was evaluated from the external quantum efficiency measured as a function of current at various temperatures ranged between 13 and 440 K. Processing the data with a novel evaluation procedure based on the ABC-model, we have determined the temperature-dependent IQE of the LED structure and light extraction efficiency of the LED chip. Separate evaluation of these parameters is helpful for further optimization of the heterostructure and chip designs. The data obtained enable making a guess on the temperature dependence of the radiative and Auger recombination coefficients, which may be important for identification of dominant mechanisms responsible for the efficiency droop in III-nitride LEDs. Thermal degradation of the LED performance in terms of the emission efficiency is also considered.
