Nedbrytning av blågrönalgtoxiner med hälsobefrämjande mjölksyrebakterier Blomningar av cyanobakterier (blågrönalger) har blivit ett världsomfattande fenomen i eutrofierade vattenmiljöer. Cyanobakterier producerar toxiner, både levergifter och nervgifter, vilka utgör en hälsorisk för människan. Exponeringsrutter omfattar både dricksvatten och förorenade matvaror. Rening av dricksvatten från dessa toxiner är således av hög prioritet. Konventionella vatttenreningsprocesser är inte alltid tillräckligt effektiva mot cyanotoxiner. Därför behövs utveckling av nya effektiva biologiska metoder för vattenrening, vilka kunde komplettera de redan existerande metoderna. FM Sonja Nybom har i sin doktorsavhandling undersökt eliminering av cyanotoxiner från dricksvatten med hjälp av probioter. Probiotiska bakterier, såsom mjölksyrebakterier och bifidobakterier, finns i den naturliga tarmfloran och har även visats ha gynnsamma effekter för människans hälsa. I avhandlingen visades flera olika stammar av probiotiska mjölksyrebakterier och bifidobakterier effektivt eliminera cyanotoxiner, såsom levergiftiga microcystiner, från vatten. Elimineringen undersöktes under olika omständigheter och visades vara beroende av bland annat vattentemperatur, pH, celldensitet och närvaro av kolkälla (glukos) för bakterierna. Metaboliskt aktiva, levande bakterier krävdes för effektiv toxineliminering. En kombination av flera probiotstammar resulterade i effektivare nedbrytning av toxiner i jämförelse med enskilda bakteriestammar. Även reaktionstiden var av betydelse för effektiviteten; efter ett dygns inkubering åstadkoms nästan total nedbrytning. Sammanfattningsvis tyder resultaten på att metoder utnyttjande dessa hälsobefrämjande probiotiska bakterier kunde utvecklas till att användas vid rening av dricksvatten från cyanotoxiner samt användas som en personlig skyddsmekanism mot cyanotoxiner i mag-tarmkanalen.

Adrenoceptors (ARs), G-protein coupled receptors (GPCRs) at the plasma membrane, respond to endogenous catecholamines noradrenaline and adrenaline. These receptors mediate several important physiological functions being especially important in the cardiovascular system and in the regulation of smooth muscle contraction. Impairments in the function of these receptors can thus lead to severe diseases and disorders such as to cardiovascular diseases and benign prostatic hyperplasia. The Eastern green mamba (Dendroaspis angusticeps) venom has been shown to contain toxins that can antagonize the functions of GPCRs. The most well-known are muscarinic toxins (MTs) targeting muscarinic acetylcholine receptors (mAChRs) with high affinity and selectivity. However, some reports have indicated that these toxins might also act on the α1- and α2-ARs which can be divided into various subtypes; the α1-ARs to α1A-, α1B- and α1D-ARs and α2-ARs to α2A-, α2B- and α2C-ARs. In this thesis, the interaction of four common MTs (MT1, MT3, MT7 and MTα) with the adrenoceptors was characterized. It was also evaluated whether these toxins could be anchored to the plasma membrane via glycosylphosphatidylinositol (GPI) tail. Results of this thesis reveal that muscarinic toxins are targeting several α-adrenoceptor subtypes in addition to their previously identified target receptors, mAChRs. MTα was found to interact with high affinity and selectivity with the α2B-AR whereas MT7 confirmed its selectivity for the M1 mAChR. Unlike MTα and MT7, MT1 and MT3 have a broad range of target receptors among the α-ARs. All the MTs characterized were found to behave as non-competitive antagonists of receptor action. The interaction between MTα and the α2B-AR was studied more closely and it was observed that the second extracellular loop of the receptor functions as a structural entity enabling toxin binding. The binding of MTα to the α2B-AR appears to be rather complex and probably involves dimerized receptor. Anchoring MTs to the plasma membrane did not interfere with their pharmacological profile; all the GPI-anchored toxins created retained their ability to block their target receptors. This thesis shows that muscarinic toxins are able to target several subtypes of α-ARs and mAChRs. These toxins offer thus a possibility to create new subtype specific ligands for the α-AR subtypes. Membrane anchored MTs on the other hand could be used to block α-AR and mAChR actions in disease conditions such as in hypertension and in gastrointestinal and urinary bladder disorders in a cell-specific manner and to study the physiological functions of ARs and mAChRs in vivo in model organisms.

The possibility of producing neutralizing antibodies against the lethal effects of scorpion toxins was evaluated in the mouse model by immunization with an immunogen devoid of toxicity. A toxic fraction (5 mg) from the venom of the scorpion Tityus serrulatus was entrapped in sphingomyelin-cholesterol liposomes. The liposomes were treated for 1 h at 37oC with a 1% (w/w) trypsin solution in 0.2 M sodium carbonate buffer, pH 8.3. This treatment led to a strong reduction in venom toxicity. Immunization was performed as follows: mice were injected sc with 20 µg of the liposome-entrapped toxic fraction on days 1 and 21 and a final injection (20 µg) was administered ip on day 36. After injection of the immunogen, all mice developed an IgG response which was shown to be specific for the toxic antigen. The antibodies were measured 10 days after the end of the immunization protocol. In an in vitro neutralization assay we observed that pre-incubation of a lethal dose of the toxic fraction with immune serum strongly reduced its toxicity. In vivo protection assays showed that mice with anti-toxin antibodies could resist the challenge with the toxic fraction, which killed, 30 min after injection, all non-immune control mice

Toxic cyanobacteria are common in Portuguese freshwaters and the most common toxins are microcystins. The occurrence of microcystin-LR (MCYST-LR) has been reported since 1990 and a significant number of water reservoirs that are used for drinking water attain high levels of this toxin. Aquatic animals that live in eutrophic freshwater ecosystems may be killed by microcystins but in many cases the toxicity is sublethal and so the animals can survive long enough to accumulate the toxins and transfer them along the food chain. Among these, edible mollusks, fish and crayfish are especially important because they are harvested and sold for human consumption. Mussels that live in estuarine waters and rivers where toxic blooms occur may accumulate toxins without many significant acute toxic effects. In this study data are presented in order to understand the dynamics of the accumulation and depuration of MCYST-LR in mussels. The toxin is readily accumulated and persists in the shellfish for several days after contact. In the crayfish the toxin is accumulated mainly in the gut but is also cleared very slowly. In carps, although the levels of the toxins found in naturally caught specimens were not very high, some toxin was found in the muscle and not only in the viscera. This raises the problem of the toxin accumulation by fish and possible transfer through the food chain. The data gathered from these experiments and from naturally caught specimens are analyzed in terms of risk for human consumption. The occurrence of microcystins in tap water and the incidence of toxic cyanobacteria in fresh water beaches in Portugal are reported. The Portuguese National Monitoring Program of cyanobacteria is mentioned and its implications are discussed.

Loxoscelism, the term used to describe lesions and clinical manifestations induced by brown spider's venom (Loxosceles genus), has attracted much attention over the last years. Brown spider bites have been reported to cause a local and acute inflammatory reaction that may evolve to dermonecrosis (a hallmark of envenomation) and hemorrhage at the bite site, besides systemic manifestations such as thrombocytopenia, disseminated intravascular coagulation, hemolysis, and renal failure. The molecular mechanisms by which Loxosceles venoms induce injury are currently under investigation. In this review, we focused on the latest reports describing the biological and physiopathological aspects of loxoscelism, with reference mainly to the proteases recently described as metalloproteases and serine proteases, as well as on the proteolytic effects triggered by L. intermedia venom upon extracellular matrix constituents such as fibronectin, fibrinogen, entactin and heparan sulfate proteoglycan, besides the disruptive activity of the venom on Engelbreth-Holm-Swarm basement membranes. Degradation of these extracellular matrix molecules and the observed disruption of basement membranes could be related to deleterious activities of the venom such as loss of vessel and glomerular integrity and spreading of the venom toxins to underlying tissues.

Chronic kidney disease is characterized by a progressive reduction of glomerular filtration rate and/or the appearance of proteinuria, and subsequently the progressive retention of organic waste compounds called uremic toxins (UT). Over the last decades, a large number of such compounds have been identified and their effects on organs and tissues, especially the cardiovascular system, has been demonstrated. In this review, we present the current classification of UT, as proposed by the EUTox Group, and the effects of some of the probably most important UTs, such as phosphate, FGF-23, PTH, AGEs, indoxyl sulfate and para-cresyl sulfate. We provide an overview on therapeutic approaches aimed to increase their extracorporeal removal via convective and/or adsorptive strategies and to lower their intestinal production/ absorption via dietetic and pharmacological interventions. The recognition that multiple toxins contribute to the uremia supports the need for new therapeutic targets, with a potentially positive impact on CKD progression and survival.

O objetivo deste estudo foi avaliar a qualidade fisiológica das sementes de milho-doce em função do teor de água na colheita e da temperatura de secagem em espiga. O experimento foi instalado na área experimental da FCA/Unesp, Botucatu-SP. Utilizou-se a cultivar BR 400 (bt) 'Super doce'. O delineamento experimental empregado foi o de blocos ao acaso com seis repetições, constituindo os tratamentos as épocas de colheitas. As colheitas das espigas foram iniciadas após a maturidade fisiológica; após despalhadas e divididas em duas porções, as espigas foram submetidas a secagem em estufas com circulação forçada nas temperaturas de 30 e 40ºC. Foi utilizada uma testemunha com sementes secadas no campo com 10,1% de teor de água. Foram determinados os teores de água das sementes, inicial e após a secagem, de todas as colheitas. Após a secagem, as espigas foram debulhadas manualmente, as sementes acondicionadas em saco de papel e armazenadas em condições ambientais de laboratório. As avaliações da qualidade fisiológica das sementes (emergência de plântulas no campo, índice de velocidade de emergência, matéria seca de plântulas, germinação, vigor-primeira contagem do teste de germinação, envelhecimento acelerado, teste de frio, condutividade elétrica e teores de Ca, Mg, K e Na lixiviados na solução do teste de condutividade elétrica) foram realizadas antes e após seis meses de armazenamento. As sementes de milho-doce cultivar BR 400 (bt), com teor de água igual ou menor do que 35%, podem ser submetidas à secagem em espiga a temperatura de 30 ou 40ºC, sem perdas significativas em sua qualidade fisiológica.

The monoconjugates of phenolic acids (i.e. coumaric acid) with polyamines such as spermidine and spermine are strikingly similar to some toxins from spiders and predatory wasps. Many plants contain phenolic acid polyamine conjugates and there is some reliable information supporting their roles as plant defense chemicals. Eleven monoacylated compounds of diamines, triamines, tetraamines and oxa-polyamine amines were prepared in three to seven steps: 22, 23, 24, 25, 26, 27, 28, 29, 30, 31 and 32. The synthesis proceeds through stepwise construction of the polyamine backbone (as in 62 and 72), followed by protection and deprotection steps of the amino functions. Desymmetrization of readily available and prepared symmetrical polyamines is a key step in the synthesis. The protecting groups employed were tert-butoxycarbonyl (BOC) and trifluoroacetyl (TFA) group which were removed under different conditions: acid and base respectively. Deprotection and refunctionalization of the polyamine reagent demonstrated the versatility of these systems for N-acylation.

In der vorliegenden Arbeit ging es um die Erarbeitung, Anwendung und Beurteilung von quantitativen Analysenverfahren / Methoden für ein Monitoring von durch Bt-Mais verursachbaren Umwelteffekten im Boden. Die Ausgangsthese besagte, dass sich transgene Maisstreu beim mikrobiellen Abbau anders verhält als konventionelle. Bezugnehmend auf die These wurden zwei Freilandversuche (Freilandmikrokosmenmethode nach Raubuch 1997 über 2 Jahre, Quantifizierung des Maisstreuabbaus mit Hilfe kleiner Bodensäulen über 1 Jahr) und zwei Inkubationsversuche im Labor (INK bei drei verschiedenen Temperaturen über 49 Tage und INK mit verschiedenen landwirtschaftlich genutzten Böden über 49 Tage mit jeweils kontinuierlicher Respirationsratenermittlung nach Isermeyer 1952) sowie Inhaltsstoffbestimmungen der Maisstreu durchgeführt. Für alle Untersuchungen wurde Streu der vier Maissorten Novelis (transgen, Monsanto 810), Nobilis (Isolinie von Novelis), Valmont (transgen, Bt 176, Fa. Syngenta) und Prelude (Isolinie von Valmont) eingesetzt. Nach Beendigung der Laborversuche sowie des Freilandversuches nach der Freilandmikrokosmenmethode wurden mikrobielle Messgrößen wie Adenylategehalt, Ergosterolgehalt, Cmik- und Nmik-Gehalt am Boden-Streu-Gemisch bestimmt. Der Einsatz der Isotopentechnik (Bestimmung von 13C/12C an gemahlenem Boden-Streu-Gemisch bzw. gefriergetrocknetem K2SO4 als Extrakt aus dem Boden-Streu-Gemisch) ermöglichte eine genaue Quantifizierung der abgebauten Maisstreu und brachte dadurch Aufschluss über das Abbauverhalten verschiedener Maissorten. Bezüglich der Ermittlung der mikrobiellen Messgrößen ergab sich für die transgene Sorte Novelis* stets eine durchschnittlich geringere pilzliche Biomasse. Langfristig ergaben sich bei der Kohlenstoff- und Stickstoffdynamik keine Trends hinsichtlich transgener bzw. konventioneller Maisstreu. Sowohl im Freilandversuch nach der Mikrokosmenmethode als auch in den Inkubationsversuchen trat das Phänomen der kurzzeitigen Respirationsratenerhöhung der Mikroorganismen nach Zugabe der transgenen Maissorten auf, welches nicht bei Zugabe der konventionellen Maisstreu auszumachen war. ______________________________

This paper describes some of the results of a detailed farm-level survey of 32 small-scale cotton farmers in the Makhathini Flats region of South Africa. The aim was to assess and measure some of the impacts (especially in terms of savings in pesticide and labour as well as benefits to human health) attributable to the use of insect-tolerant Bt cotton. The study reveals a direct cost benefit for Bt growers of SAR416 ($51) per hectare per season due to a reduction in the number of insecticide applications. Cost savings emerged in the form of lower requirements for pesticide, but also important were reduced requirements for water and labour. The reduction in the number of sprays was particularly beneficial to women who do some spraying and children who collect water and assist in spraying. The increasing adoption rate of Bt cotton appears to have a health benefit measured in terms of reported rates of accidental insecticide poisoning. These appear to be declining as the uptake of Bt cotton increases. However, the understanding of refugia and their management by local farmers are deficient and need improving. Finally, Bt cotton growers emerge as more resilient in absorbing price fluctuations.

The study reported presents the findings relating to commercial growing of genetically-modified Bt cotton in South Africa by a large sample of smallholder farmers over three seasons (1998/99, 1999/2000, 2000/01) following adoption. The analysis presents constructs and compares groupwise differences for key variables in Bt v. non-Bt technology and uses regressions to further analyse the production and profit impacts of Bt adoption. Analysis of the distribution of benefits between farmers due to the technology is also presented. In parallel with these socio-economic measures, the toxic loads being presented to the environment following the introduction of Bt cotton are monitored in terms of insecticide active ingredient (ai) and the Biocide Index. The latter adjusts ai to allow for differing persistence and toxicity of insecticides. Results show substantial and significant financial benefits to smallholder cotton growers of adopting Bt cotton over three seasons in terms of increased yields, lower insecticide spray costs and higher gross margins. This includes one particularly wet, poor growing season. In addition, those with the smaller holdings appeared to benefit proportionately more from the technology (in terms of higher gross margins) than those with larger holdings. Analysis using the Gini-coefficient suggests that the Bt technology has helped to reduce inequality amongst smallholder cotton growers in Makhathini compared to what may have been the position if they had grown conventional cotton. However, while Bt growers applied lower amounts of insecticide and had lower Biocide Indices (per ha) than growers of non-Bt cotton, some of this advantage was due to a reduction in non-bollworm insecticide. Indeed, the Biocide Index for all farmers in the population actually increased with the introduction of Bt cotton. The results indicate the complexity of such studies on the socio-economic and environmental impacts of GM varieties in the developing world.
