It is very known that due to inflammatory processes the obesity leads to resistance to leptin, it reduces phosphorylation via JAK-2/STAT-3, which generates lower STAT-3 activity in the cell nucleus, and it leads to decrease the number of transcription of anorexigenic neurons (POMC/CART) and allowing transcription of orexigenic (NPY/AgRP). PURPOSE: The present study aimed to evaluate the effects of moderate aerobic training on food intake of obese mice through analysis of activity of hypothalamic proteins JAK-2/STAT-3. METHODS: It were used 30 Swiss mice (30 days old) divided into 3 groups: Control Group (C): sedentary animals fed with balanced diet ; Obese (OB) sedentary animals fed with high-fat diet throughout the experiment and Trained Obese (TOB) : animals fed with high fat diet throughout the experiment , kept sedentary during the first half of the experiment (8 weeks) and submitted to physical training protocol during the second half of the experiment (8 weeks). The exercise program consisted of treadmill running 1h, 5 days/week during 8 weeks at a speed equivalent to 60 % of maximum potency determined at the beginning of training period. To assess the leptin resistance, after rats were deprived of food for 6h with free access to water, they received i.p injection with leptin (2.0µl, 10-6M), after this, the chow was returned and food intake was determined by measuring the quantity and Kcal consumed at the end of 2h. The hypothalami was removed for determination of JAK-2 and STAt-3 activity. RESULTS: Our results showed that moderate physical exercise was effective in improving the JAK/STAT signaling pathway in the hypothalamus of obese animals. This has made these obese animals had reduced food intake and consequently lower body mass gain. CONCLUSION: It can be concluded that physical exercise, for restoring leptin signaling in the hypothalamus, controls the synthesis of neurons responsible for appetite and thus is an important tool in the treatment of obesity.

The fishery products are sources of protein of high biological value and an important component in a balanced diet. However, they are highly subject to deterioration, which represents a serious risk to public health. Thus, rigorous methods are needed for quality assurance of fish in all stages of the production chain. Traditionally, the Brazilian population does not have the habit of eating fish, although the country presents a great potential for aquaculture, which is underexploited. The consumption of fish is limited by socio-economic factors. Currently, the search for food security is a global trend. Thus, it is necessary to change the profile of the consumer, by offering a competitive and safe food. In this paper, we address issues related to quality, hygiene and health of fish

This study compares the impact of obesogenic environment (OE) in six different periods of development on sperm parameters and the testicular structure of adult rats and their correlations with sex steroid and metabolic scenario. Wistar rats were exposed to OE during gestation (O1), during gestation/lactation (O2), from weaning to adulthood (O3), from lactation to adulthood (O4), from gestation to sexual maturity (O5), and after sexual maturation (O6). OE was induced by a 20% fat diet, and control groups were fed a balanced diet (4% fat). Serum leptin levels and adiposity index indicate that all groups were obese, except for O1. Three progressive levels of impaired metabolic status were observed: O1 presented insulin resistance, O2 were insulin resistant and obese, and groups O3, O4, and O5 were insulin resistant, obese, and diabetic. These three levels of metabolic damage were proportional to the increase of leptin and decreased circulating testosterone. The impairment in the daily sperm production (DSP) paralleled these three levels of metabolic and hormonal damage being marginal in O1, increasing in O2, and being higher in groups O3, O4, O5, and O6. None of the OE periods affected the sperm transit time in the epididymis, and the lower sperm reserves were caused mainly by impaired DSP. In conclusion, OE during sexual maturation markedly reduces the DSP at adulthood in the rat. A severe reduction in the DSP also occurs in OE exposure during gestation/lactation but not in gestation, indicating that breast-feeding is a critical period for spermatogenic impairment under obesogenic conditions.

Recent studies have shown a positive association of cancer and obesity, but the morphological and molecular mechanisms involved in this relationship are still unknown. This study analysed the impact of long-term obesity on rat prostate, focusing on stromal changes. Male adult Wistar rats were treated with high-fat diet to induce obesity, while the control group received a balanced diet. After 30 weeks of feeding, the ventral prostate was analysed by immunohistochemistry for cell proliferation, smooth muscle α-actin, vimentin, chondroitin sulphate and metalloproteinases (MMP-2 and 9). The content of androgen receptor (AR), oestrogen receptors (ERs) and vascular endothelial growth factor (VEGF) was measured by Western blotting, and activity of catalase and Glutathione-S-Transferase (GST) were quantified by enzymatic assay. Long-term obesity decreased testosterone plasma levels by 70% and resulted in stromal prostate hyperplasia, as evidenced by increased collagen fibres. Such stromal hyperplasia was associated with increased number of blood vessels and raised VEGF content, and increased expression of chondroitin sulphate, vimentin, α-actin and MMP-9. In spite of the high cell density in prostate, the proliferative activity was lower in the prostates of obese rats, indicating that hyperplasia was established during the early phases in this obesity model. AR levels increased significantly, whereas the ERα decreased in this group. Moreover, the levels of catalase and GST were changed considerably. These findings indicate that long-term obesity, besides disturbing the antioxidant control, causes intense stromal remodelling and release of factors that create an environment that can promote proliferative disorders in the gland, culminating with diffuse hyperplasia.

Free radicals are produced during aerobic cellular metabolism and have key roles as regulatory mediators in signaling processes. Oxidative stress reflects an imbalance between production of reactive oxygen species and an adequate antioxidant defense. This adverse condition may lead to cellular and tissue damage of components, and is involved in different physiopathological states, including aging, exercise, inflammatory, cardiovascular and neurodegenerative diseases, and cancer. In particular, the relationship between exercise and oxidative stress is extremely complex, depending on the mode, intensity, and duration of exercise. Regular moderate training appears beneficial for oxidative stress and health. Conversely, acute exercise leads to increased oxidative stress, although this same stimulus is necessary to allow an up-regulation in endogenous antioxidant defenses (hormesis). Supporting endogenous defenses with additional oral antioxidant supplementation may represent a suitable noninvasive tool for preventing or reducing oxidative stress during training. However, excess of exogenous antioxidants may have detrimental effects on health and performance. Whole foods, rather than capsules, contain antioxidants in natural ratios and proportions, which may act in synergy to optimize the antioxidant effect. Thus, an adequate intake of vitamins and minerals through a varied and balanced diet remains the best approach to maintain an optimal antioxidant status. Antioxidant supplementation may be warranted in particular conditions, when athletes are exposed to high oxidative stress or fail to meet dietary antioxidant requirements. Aim of this review is to discuss the evidence on the relationship between exercise and oxidative stress, and the potential effects of dietary strategies in athletes. The differences between diet and exogenous supplementation as well as available tools to estimate effectiveness of antioxidant intake are also reported. Finally, we advocate the need to adopt an individualized diet for each athlete performing a specific sport or in a specific period of training, clinically supervised with inclusion of blood analysis and physiological tests, in a comprehensive nutritional assessment. The knowledge of how animals deposited chemical components as water, protein, fat and ash in the carcass is importance for the formulation of a balanced diet, allowing maximum performance with a low environmental impact. So, the study was carried out to evaluate the influence of different tilapia strains (Chitralada, Commercial, Red and Universidade Federal de Lavras [UFLA]) on the deposition of bodily chemical components in the carcass. The bodily components analyzed were water, protein, fat and ash. For the determination of the bodily chemical deposition curves by age, the exponential, Brody, logistic, Gompertz and von Bertalanffy models were adjusted. The Commercial and UFLA strains deposited water at a faster speed (P<0.05) compared with the remaining strains. As for protein, the Red strain had a lower estimated maturity weight (49.37 g), and was more precocious (202 days) with regard to maximum deposition in comparison to the other strains (Chitralada, UFLA and Commercial) in which there was an estimated maturity weight of 231.5 g and maximum depositionfor 337 days. There were no differences (P>0.05) for the logistic model parameter between Red, UFLA and Commercial strains for fat, which presented a maximum fat deposition (0.23 g) at 310 days of age. Regarding ash deposition, the Commercial strain presented a higher maximum deposition (0.10 g) at 337 days, occurring later than the other strains that presented maximum deposition (0.033g) at 254 days of age. Thus, it was concluded that the genetic strains evaluated differ in chemical deposition curves of water, protein, fat and ash.
