El objetivo general de la presente investigación está orientado a analizar en profundidad la perspectiva filosófico-pragmatista que desarrollara Clarence Irving Lewis en torno a la teoría del conocimiento, señalando las vinculaciones e implicancias de la misma para su teoría de la valoración y para la ética temas que consideramos centrales en las agendas de investigación actuales en epistemología. Todo ello con el propósito de contribuir a una mejor comprensión de su importancia e incidencia en el marco de la corriente pragmatista, de determinar el carácter original y novedoso de sus aportes al campo de la epistemología, y de evaluar su potencial para pensar las prácticas cognitivas y científicas actuales. Cabe señalar que C. I. Lewis forma parte de la tradición del pragmatismo clásico, un movimiento filosófico surgido en EEUU a finales del s. XIX en torno a la obra de pensadores como C. S. Peirce, W. James, J. Dewey y G. H. Mead. Si bien esta línea de pensamiento ha sido criticada y en muchos aspectos malinterpretada, desde hace ya unas décadas ha resurgido un neto interés en dicho pensamiento desde perspectivas interpretativas de innegable valor para el campo de la epistemología, de la vida social y de la ética; más allá de las coincidencias y divergencias que sin duda sigue suscitando. En lo que respecta especialmente a Lewis, ha sido sin duda uno de los epistemólogos más importantes de la filosofía norteamericana durante el siglo XX. Su largo paso por Harvard, como estudiante y como profesor, le significó una importante formación, una vinculación con grandes pensadores de la filosofía local y continental, y una indudable influencia en las generaciones que le sucedieron. Sin embargo, su filosofía no ha recibido la atención que merece en su propio contexto y, mucho menos, en la filosofía de habla hispana. Ha sido, además, objeto de diferentes interpretaciones y asociaciones, muchas de las cuales lo desvincularon de la corriente del pragmatismo clásico opacando sus más lúcidos aportes teóricos. Frente a este estado de la cuestión y teniendo en cuenta, por otra parte, el resurgimiento que ha tenido el pensamiento pragmatista en las últimas décadas y el renovado interés que se registra en torno a sus perspectivas interpretativas para el terreno de la epistemología, la filosofía política y la ética, es que nos proponemos los siguientes objetivos específicos de trabajo. En primer lugar, recobrar el pensamiento lewisiano analizando en profundidad su novedosa posición, denominada "pragmatismo conceptualista", sistematizando sus conceptos específicos y sus tesis fundamentales. En segundo lugar, dar cuenta de su filiación pragmatista para así habilitar lo que, a nuestro juicio y tal como pretendemos mostrar aquí, constituye una mejor y más cabal comprensión de su propuesta integral; o, dicho en sentido inverso, abordar desde una perspectiva global su propuesta filosófica en torno al conocimiento de modo de poner en evidencia su innegable veta pragmatista. En tercer lugar, dar a conocer para el caso de la filosofía hispanoparlante lo que consideramos una de las teorías gnoseológicas más complejas, potentes y acabadas del siglo XX (sobre todo teniendo en cuenta que es casi desconocida en dicho contexto y que no ha sido traducida al español), de manera tal de habilitar su incorporación a las reflexiones actuales vinculadas al resurgimiento que tiene desde hace unas décadas el pragmatismo. En cuarto lugar, dar cuenta de algunos problemas o puntos de tensión que presenta su teoría y revisar algunas de las principales críticas que ha recibido. Finalmente y acorde a todo lo anterior, evaluar la pertinencia, los aportes y la incidencia de la perspectiva lewisiana en el campo del conocimiento para así contribuir a las discusiones más actuales en epistemología y filosofía de las ciencias, así como también a las reflexiones éticas y axiológicas íntimamente vinculadas a ellas. En función de los objetivos anteriormente señalados, proponemos una metodología consistente en abordar analíticamente la concepción filosófica de Lewis a lo largo de diferentes escritos, centrándonos básicamente en su teoría del conocimiento; situar contextualmente su pensamiento tratando de reconstruir su trayecto filosófico a través de distintos tópicos e intereses; mostrar las influencias que recibió y las que supo ejercer a partir de diferentes intercambios con profesores, colegas y alumnos; y abordar algunos de los problemas gnoseológicos más importantes a los que Lewis pretendió dar respuesta, señalando el tono específico de los debates en los que se enmarcaban y analizando puntualmente algunas de las críticas de mayor relevancia que se le han formulado y que, a nuestro juicio, han incidido en la recepción de sus propuestas. Todo ello, en orden a dar cuenta crítica y argumentalmente de las siguientes tesis: que (1) Lewis es un pragmatista, a pesar de las importantes interpretaciones en contrario realizadas por destacados filósofos contemporáneos; que (2) su propuesta constituye un genuino sistema filosófico de ideas sólidamente articuladas sobre el trasfondo de su perspectiva pragmatista; que (3) si asumimos las dos tesis anteriores muchas de las dificultades interpretativas que por momentos pone de manifiesto la teoría lewisiana se resuelven reforzando consistentemente dichas premisas; lo cual nos permite pensar que (4) en muchos casos Lewis ha sido mal comprendido precisamente porque se lo ha desvinculado del pragmatismo o porque se han desarticulado algunas partes de su teoría, de su sistema como un todo; y finalmente, y en consonancia con los objetivos principales del trabajo, la tesis según la cual (5) la interpretación en clave pragmatista de Lewis ofrece una perspectiva integral de su teoría que resuelve ciertas aporías al mismo tiempo que permite visualizar un conjunto de aportes que resultan significativos para las discusiones epistemológicas actuales; todos éstos resultados que, a su vez, nos estimulan a seguir en nuestra dirección interpretativa. Por último, ofrecemos algunas conclusiones finales que muestran resumidamente los argumentos que nos permiten respaldar las afirmaciones anteriormente señaladas, que recapitulan en líneas generales los puntos más significativos desarrollados a lo largo del trabajo, y que retoman de manera orgánica las conclusiones parciales obtenidas. Asimismo, ofrecemos un conjunto de hipótesis que podrían orientar una futura investigación que de continuidad al trabajo aquí presentado y que se fundan en la convicción de que la propuesta de Lewis tiene todavía mucho que aportar y que su incorporación a las reflexiones actuales puede resultar muy enriquecedora

Purpose – This paper adds to growing research of psychiatric intensive care units (PICU) by recounting descriptions of psychiatric intensive care settings and discusses the perceptions held by nurses of the organisational interfaces, arrangements and provisions of care in these settings. Design/methodology/approach – Data gathered from focus groups held with nurses from two PICUs was used to establish terminology, defining attributes, related concepts, antecedents, values, processes and concepts related to current practices. A literature search was conducted to permit a review of the conceptual arrangements and contemporary understanding of intensive care for people experiencing acute psychiatric illness based on the perspectives held by the nurses from the focus groups. Findings – Dissonance between service needs and the needs and management of individual patients overshadow strategies to implement comprehensive recovery-oriented approaches. Three factors are reported in this paper that influence standards and procedural practice in PICU; organisational structures; physical structures; and subtype nomenclature. Practical implications – Acute inpatient care is an important part of a comprehensive approach to mental health services. Commonly intensive acute care is delivered in specialised wards or units co-located with acute mental health inpatient units mostly known as PICU. Evidence of the most effective treatment and approaches in intensive care settings that support comprehensive recovery for improved outcomes is nascent. Originality/value – Current descriptions from nurses substantiate wide variations in the provisions, design and classifications of psychiatric intensive care. Idiosyncratic and localised conceptions of psychiatric intensive care are not adequately entailing effective treatment and methods in support of recovery principles for improved and comprehensive outcomes. The authors suggest that more concrete descriptions, guidelines, training and policies for provision of intensive psychiatric health care encompassing the perspective of nursing professionals, would reinforce conceptual construction and thus optimum treatments within a comprehensive, recovery-oriented approach to mental health services.

Introduction: The delivery of health care in the 21st century will look like no other in the past. The fast paced technological advances that are being made will need to transition from the information age into clinical practice. The phenomenon of e-Health is the over-arching form of information technology and telehealth is one arm of that phenomenon. The uptake of telehealth both in Australia and overseas, has changed the face of health service delivery to many rural and remote communities for the better, removing what is known as the tyranny of distance. Many studies have evaluated the satisfaction and cost-benefit analysis of telehealth across the organisational aspects as well as the various adaptations of clinical pathways and this is the predominant focus of most studies published to date. However, whilst comments have been made by many researchers about the need to improve and attend to the communication and relationship building aspects of telehealth no studies have examined this further. The aim of this study was to identify the patient and clinician experiences, concerns, behaviours and perceptions of the telehealth interaction and develop a training tool to assist these clinicians to improve their interaction skills. Methods: A mixed methods design combining quantitative (survey analysis and data coding) and qualitative (interview analysis) approaches was adopted. This study utilised four phases to firstly qualitatively explore the needs of clients (patients) and clinicians within a telehealth consultation then designed, developed, piloted and quantitatively and qualitatively evaluated the telehealth communication training program. Qualitative data was collected and analysed during Phase 1 of this study to describe and define the missing 'communication and rapport building' aspects within telehealth. This data was then utilised to develop a self-paced communication training program that enhanced clinicians existing skills, which comprised of Phase 2 of this study to develop the interactive program. Phase 3 included evaluating the training program with 26 clinicians and results were recorded pre and post training, whilst phase 4 was the pilot for future recommendations of this training program using a patient group within a Queensland Health setting at two rural hospitals. Results: Comparisons of pre and post training data on 1) Effective communication styles, 2) Involvement in communication training package, 3) satisfaction pre and post training, and 4) health outcomes pre and post training indicated that there were differences between pre and post training in relation to effective communication style, increased satisfaction and no difference in health outcomes between pre and post training for this patient group. The post training results revealed over half of the participants (N= 17, 65%) were more responsive to non-verbal cues and were better able to reflect and respond to looks of anxiousness and confusion from a 'patient' within a telehealth consultation. It was also found that during post training evaluations, clinicians had enhanced their therapeutic communication with greater detail to their own body postures, eye contact and presentation. There was greater time spent looking at the 'patient' with an increase of 35 second intervals of direct eye contact and less time spent looking down at paperwork which decreased by 20 seconds. Overall 73% of the clinicians were satisfied with the training program and 61% strongly agreed that they recognised areas of their communication that needed improving during a telehealth consultation. For the patient group there was significant difference post training in rapport with a mean score from 42 (SD = 28, n = 27) to 48 (SD = 5.9, n = 24). For communication comfort of the patient group there was a significant difference between the pre and post training scores t(10) = 27.9, p = .002, which meant that overall the patients felt less inhibited whilst talking to the clinicians and more understood. Conclusion: The aim of this study was to explore the characteristics of good patient-clinician communication and unmet training needs for telehealth consultations. The study developed a training program that was specific for telehealth consultations and not dependent on a 'trainer' to deliver the content. In light of the existing literature this is a first of its kind and a valuable contribution to the research on this topic. It was found that the training program was effective in improving the clinician's communication style and increased the satisfaction of patient's within an e-health environment. This study has identified some historical myths that telehealth cannot be part of empathic patient centred care due to its technology tag.

Chlamydial infections are wide spread in koalas across their range and a solution to this debilitating disease has been sought for over a decade. Antibiotics are the currently accepted therapeutic measure, but are not an effective treatment due to the asymptomatic nature of some infections and a low efficacy rate. Thus, a vaccine would be an ideal way to address this infectious disease threat in the wild. Previous vaccine trials have used a three-dose regimen; however this is very difficult to apply in the field as it would require multiple capture events, which are stressful and invasive processes for the koala. In addition, it requires skilled koala handlers and a significant monetary investment. To overcome these challenges, in this study we utilized a polyphosphazine based poly I:C and a host defense peptide adjuvant combined with recombinant chlamydial major outer membrane protein (rMOMP) antigen to induce long lasting (54 weeks) cellular and humoral immunity in female koalas with a novel single immunizing dose. Immunized koalas produced a strong IgG response in plasma, as well as at mucosal sites. Moreover, they showed high levels of C. pecorum specific neutralizing antibodies in the plasma as well as vaginal and conjunctival secretions. Lastly, Chlamydia-specific lymphocyte proliferation responses were produced against both whole chlamydial elementary bodies and rMOMP protein, over the 12-month period. The results of this study suggest that a single dose rMOMP vaccine incorporating a poly I:C, host defense peptide and polyphosphazine adjuvant is able to stimulate both arms of the immune system in koalas, thereby providing an alternative to antibiotic treatment and/or a three-dose vaccine regime.

Earlier, we showed that, for the D form (n = 8 and h = 3.03 A, where n is number of nucleotide units per turn and h is height per nucleotide unit) of poly[d(A-T)], both right- and left-handed double helical models are stereochemically satisfactory and give good agreement with the observed fiber diffraction data. It was also noted that the conformations of the right- and left-handed D-DNA models are very similar to those of the right- and left-handed B-DNA models. This observation was consistent with the D leads to B transition in the solid phase. As a continuation of our earlier studies, we have carried out similar experiments with poly[d(I-C)]. We could obtain a crystalline D-form pattern (n = 8, h = 3.13 A) of the fiber at 75% relative humidity (r.h.); the hydrated (r.h. approximately equal to 95%) form of the same fiber gave the classical B-form pattern (n = 10, h = 3.40 A). In the present report, we show that both right- and left-handed double-helical models are consistent with the fiber diffraction data of poly[d(I-C)] in the D-form. Theoretical energy calculations also suggest that the right- and left-handed B- and D-DNA models are almost equally stable. Hence, we conclude that the right- and left-handed double-helical models of poly[d(I-C)] in a given form (B or D) are equally likely and that the fiber diffraction data do not permit discrimination.

It is currently believed that an unsubstituted axial hydroxyl at the specificity-determining C-4 locus of galactose is indispensable for recognition by galactose/N-acetylgalactosamine-specific lectins. Titration calorimetry demonstrates that 4-methoxygalactose retains binding allegiance to the Moraceae lectin jacalin and the Leguminosae lectin, winged bean (basic) agglutinin (WBA I). The binding reactions were driven by dominant favorable enthalpic contributions and exhibited significant enthalpy-entropy compensation. Proton NMR titration of C-methoxygalactose with jacalin and WBA I resulted in broadening of the sugar resonances without any change in chemical shift. The alpha-and beta-anomers of 4-methoxygalactose were found to be in slow exchange with free and lectin-bound states. Both the anomers experience magnetically equivalent environments at the respective binding sites. The binding constants derived from the dependence of NMR line widths on 4-methoxygalactose concentration agreed well with those obtained from titration calorimetry. The results unequivocally demonstrate that the loci corresponding to the axially oriented C-4 hydroxyl group of galactose within the primary binding site of these lectins exhibit plasticity. These analyses suggest, for the first time, the existence of C-H ... O-type hydrogen-bond(s) in protein-carbohydrate interactions in general and between the C-4 locus of galactose derivative and the lectins jacalin and WBA I in particular.

Acetohydroxyacid synthase (AHAS) is an enzyme involved in the biosynthesis of the branched chain amino acids viz, valine, leucine and isoleucine. The activity of this enzyme is regulated through feedback inhibition by the end products of the pathway. Here we report the backbone and side-chain assignments of ilvN, the 22 kDa dimeric regulatory subunit of E. coli AHAS isoenzyme I, in the valine bound form. Detailed analysis of the structure of ilvN and its interactions with the catalytic subunit of E. coli AHAS I will help in understanding the mechanism of activation and regulation of the branched chain amino acid biosynthesis.

Physical clustering of genes has been shown in plants; however, little is known about gene clusters that have different functions, particularly those expressed in the tomato fruit. A class I 17.6 small heat shock protein (Sl17.6 shsp) gene was cloned and used as a probe to screen a tomato (Solanum lycopersicum) genomic library. An 8.3-kb genomic fragment was isolated and its DNA sequence determined. Analysis of the genomic fragment identified intronless open reading frames of three class I shsp genes (Sl17.6, Sl20.0, and Sl20.1), the Sl17.6 gene flanked by Sl20.1 and Sl20.0, with complete 5' and 3' UTRs. Upstream of the Sl20.0 shsp, and within the shsp gene cluster, resides a box C/D snoRNA cluster made of SlsnoR12.1 and SlU24a. Characteristic C and D, and C' and D', boxes are conserved in SlsnoR12.1 and SlU24a while the upstream flanking region of SlsnoR12.1 carries TATA box 1, homol-E and homol-D box-like cis sequences, TM6 promoter, and an uncharacterized tomato EST. Molecular phylogenetic analysis revealed that this particular arrangement of shsps is conserved in tomato genome but is distinct from other species. The intronless genomic sequence is decorated with cis elements previously shown to be responsive to cues from plant hormones, dehydration, cold, heat, and MYC/MYB and WRKY71 transcription factors. Chromosomal mapping localized the tomato genomic sequence on the short arm of chromosome 6 in the introgression line (IL) 6-3. Quantitative polymerase chain reaction analysis of gene cluster members revealed differential expression during ripening of tomato fruit, and relatively different abundances in other plant parts.

Pluripotent stem cells are being actively studied as a cell source for regenerating damaged liver. For long-term survival of engrafting cells in the body, not only do the cells have to execute liver-specific function but also withstand the physical strains and invading pathogens. The cellular innate immune system orchestrated by the interferon (IFN) pathway provides the first line of defense against pathogens. The objective of this study is to assess the innate immune function as well as to systematically profile the IFN-induced genes during hepatic differentiation of pluripotent stem cells. To address this objective, we derived endodermal cells (day 5 post-differentiation), hepatoblast (day 15) and hepatocyte-like cells (day 21) from human embryonic stem cells (hESCs). Day 5, 15 and 21 cells were stimulated with IFN-alpha and subjected to IFN pathway analysis. Transcriptome analysis was carried out by RNA sequencing. The results showed that the IFN-alpha treatment activated STAT-JAK pathway in differentiating cells. Transcriptome analysis indicated stage specific expression of classical and non-classical IFN-stimulated genes (ISGs). Subsequent validation confirmed the expression of novel ISGs including RASGRP3, CLMP and TRANK1 by differentiated hepatic cells upon IFN treatment. Hepatitis C virus replication in hESC-derived hepatic cells induced the expression of ISGs - LAMP3, ETV7, RASGRP3, and TRANK1. The hESC-derived hepatic cells contain intact innate system and can recognize invading pathogens. Besides assessing the tissue-specific functions for cell therapy applications, it may also be important to test the innate immune function of engrafting cells to ensure adequate defense against infections and improve graft survival. Cronista do Reino, escreveu essa obra por ordem real, que foi publicada em 1628, utilizando documentação oficial e informações que as relações da época lhe proporcionavam. ‘Restauracion de la ciudad del Salvador, i Baia de Todos-Sanctos, en la Provincia del Brasil’ reflete a opinião oficial espanhola sobre a restauração da Bahia e mostra a repercussão que o acontecimento teve na metrópole espanhola que, na época, dominava o Brasil. Rodrigues considera essa uma edição ‘rara’ e Brunet afirma ser ‘muito rara’. Há uma edição brasileira, difícil de encontrar, publicada em Salvador pela Tipografia de Epifanio Pedroza (1847)
