Apesar da sua localização perto de Belém, Pará, a Ilha de Cotijuba tem sido assolada por surtos de malária durante os últimos anos, principalmente nos meses de abril a maio. A ilha faz parte de um arquipélago situado às margens da baía do Marajó, a 29 km de Belém, constituindo uma parte insular dessa cidade. Cotijuba possui uma área de cerca de 60 km² e uma costa com 20 km de praias, que corresponde a 66% da área total da ilha. Por causa das repetidas epidemias de malária, nosso interesse voltou-se para conhecer as espécies de anofelinos vetoras. Nos anos de 2002 a 2004 foram realizadas coletas periódicas de mosquitos adultos utilizando-se o método de captura manual com atrativo humano no peridomicílio. Foram realizadas seis coletas bimensais com duração de 6 horas com o registro de apenas duas espécies em atividade atacando o homem, Anopheles (Nyssorhynchus) aquasalis e Anopheles (Anopheles) intermedius. Verificou-se que as condições climáticas, o comportamento dos residentes e a falta de recursos para o efetivo lcontrole dos vetores da doença, favorecem a persistência de malária na Ilha de Cotijuba.

Por causa da epidemia, tomou-se necessário conhecer as espécies de anofelinos vetores de malária na ilha. Nos anos de 2002 a 2004 foram realizadas coletas periódicas de larvas e adultos de mosquitos e os seus criatórios foram localizados e caracterizados. Os imaturos foram coletados com auxílio de conchas e em bandejas plásticas. Para os adultos utilizou-se o método de captura manual com isca humana. Duas coletas de adultos na floresta foram realizadas, uma no período seco e outra no período chuvoso, com duração de 24 horas ininterruptas. Seis coletas bimensais no peridomicílio foram feitas, com duração de 6 horas. Tanto na floresta como no peridomicilio foram registradas apenas duas espécies em atividade atacando o homem, Anopheles (Nyssorhynchus) aquasalis e Anopheles (Anopheles) intermedius. A. aquasalis foi mais freqüente no peridomicílio, enquanto A. intermedius teve maior freqüência na floresta. Verificou-se que o maior número de casos de malária na ilha ocorre dois meses depois do início das chuvas, no mês de maio. Na i, como um todo, existem quatro lagos que são os possíveis criatórios de anofelinos da ilha e o lago da Gabriela é o principal criatório da ilha de Cotijuba, sendo responslha de Cotijubaável por 42% dos casos de malária na ilha no ano de 2003. As condições climáticas, o comportamento dos residentes e a falta de recursos para o efetivo controle dos vetores da doença, entre outros fatores, favorecem a persistência de malária na Ilha de Cotijuba.

Across the Americas and the Caribbean, nearly 561,000 slide-confirmed malaria infections were reported officially in 2008. The nine Amazonian countries accounted for 89% of these infections; Brazil and Peru alone contributed 56% and 7% of them, respectively. Local populations of the relatively neglected parasite Plasmodium vivax, which currently accounts for 77% of the regional malaria burden, are extremely diverse genetically and geographically structured. At a time when malaria elimination is placed on the public health agenda of several endemic countries, it remains unclear why malaria proved so difficult to control in areas of relatively low levels of transmission such as the Amazon Basin. We hypothesize that asymptomatic parasite carriage and massive environmental changes that affect vector abundance and behavior are major contributors to malaria transmission in epidemiologically diverse areas across the Amazon Basin. Here we review available data supporting this hypothesis and discuss their implications for current and future malaria intervention policies in the region. Given that locally generated scientific evidence is urgently required to support malaria control interventions in Amazonia, we briefly describe the aims of our current field-oriented malaria research in rural villages and gold-mining enclaves in Peru and a recently opened agricultural settlement in Brazil. Results obtained with catches performed at several sites of the Ribeira Valley, SP (Brazil) are reported. Collections with Shannon type traps showed a concentration of An.albitarsis s.l. in the Experimental Farm of Pariquera-Açu County, associated with the development of irrigated rice cultivation. Two species of the complex, named A and B, were recognized. Species B predominated in the samples. Indoor and outdoor crepuscular human bait captures were performed a the domiciliary environment. The two species showed a generally low William's media valve of 0.2, the highest value obtained was 1.6. No differences were found between indoor and outdoor behaviours between the two species of the complex.

INTRODUCTION: Isoenzymatic analyses were performed involving species of the Nyssorhynchus and Anopheles subgenera in order to estimate the intra and interspecies genetic variability. METHODS: Mosquitoes were caught at different localities in the Amazon region. The collection and rearing of mosquitoes in the laboratory followed specific protocols. For the genetic variability analyses, the technique of horizontal electrophoresis on starch and starch-agarose gel with appropriate buffer systems was used. The alloenzyme variation was estimated using the Biosys-1 software. RESULTS: Out of the 13 loci, eight were polymorphic. Anopheles nuneztovari presented the largest number of alleles per locus, while the smallest number was detected in Anopheles marajoara from Macapá. The largest number of polymorphic loci was found for Anopheles marajoara from Maruanum and the smallest for Anopheles benarrochi (Guayará Mirim). Anopheles darlingi (Macapá) presented the greatest heterozygosity (Ho = 0.167 ± 0.071), while the lowest heterozygosity (Ho = 0.045 ± 0.019) was observed in Anopheles intermedius (Pacoval) of the subgenus Anopheles. Wright's F coefficient revealed considerable genetic structuring between the populations of Anopheles darlingi (Fst = 0.110) and between the populations of Anopheles marajoara (Fst = 0.082). CONCLUSIONS: Considering all the species studied, the genetic distance ranged from 0.008 to 1.114. The greatest distance was between Anopheles mattogrossensis and Anopheles oswaldoi, while the smallest was between the Anopheles benarrochi populations.

Anopheles halophylus n.sp. is described from Central Brazil. It is distinguished from An. triannulatus (Neiva and Pinto) by morphological and morphometric characters, especially in the male genitalia, larva and egg. Illustrations of the male and female genitalia, egg, larva and pupa, and discussion of the status of names associated with An. triannulatus are provided.

The most important vectors of human Plasmodium in the neotropics belong to the subgenus Nyssorhynchus. These species are generally sympatric in terms of their geographical distributions. Some are difficult to identify based solely on examination of adult females using the available morphological keys, in these cases examination of immature stages and male genitalia is required to make correct determinations. However, in epidemiological studies it is necessary to identify the species of adult females which are found near humans, i.e. in studies of malaria transmission or evaluation of control measures. The purpose of the present study was to evaluate the discrimination of adult females of different species of Nyssorhynchus isolated mainly from Southern Colombia (department of Putumayo), using morphometric analysis. Adult females were obtained after rearing larvae collected in natural breeding places and from the progeny of females collected on humans. The morphological characteristics of the immature stages allowed the identification of four species of the subgroup Oswaldoi from Southern Colombia: Anopheles rangeli Gabaldon, Cova Garcia & Lopez, An. oswaldoi (Peryassu), An. benarrochi Gabaldon, Cova Garcia & Lopez and An. triannulatus (Neiva & Pinto). The species An. nuneztovari (Gabaldon) from the Northwest of Colombia was included for comparison. Morphometric analysis allowed differentiation of the females of all species to a confidence level approaching 90% using principal components analysis of 10 wing and leg variables, followed by canonical variate analysis of the first four principal components. We conclude that morphometrics may represent a useful taxonomic tool for this group and that its use should be further studied.

An isoenzymatic comparative analysis of the variability and genetics differentiation among Anopheles species was done in populations of An. (Nys.) intermedius and An. (Ano.) mattogrossensis of the Anopheles subgenus, and of An. darlingi, An. albitarsis and An. triannulatus of the Nyssorhynchus subgenus, with the aim of detecting differences between both subgenera and of estimating the degree of genetic intere specific divergence. Samples from Macapá, State of Amapá and Janauari Lake, near Manaus, State of Amazonas, were analyzed for eight isoenzymatic loci. Analysis revealed differences in the average number of alleles per locus (1.6-2.3) and heterozygosity (0.060-0.284). However, the proportion of polymorphic loci was the same for An. (Nys.) darlingi, An. (Nys.) triannulatus and An. (Ano.) mattogrossensis (50%), but differed for An. (Nys.) albitarsis (62.5%) and An. (Ano.) intermedius (25%). Only the IDH1 (P > 0.5) locus in all species studied was in Hardy-Weinberg equilibrium. The fixation index demonstrated elevated genetic structuring among species, based on values of Fst = 0.644 and genetic distance (0.344-0.989). Genetic difference was higher between An. (Nys.) triannulatus and An. (Ano.) intermedius (0.989) and smaller between An. (Nys.) albitarsis sensu lato and An. (Nys.) darlingi (0.344). The data show interspecific genetic divergence which differs from the phylogenetic hypothesis based on morphological characters.

Anopheles darlingi Root is the major vector of human malaria in the Neotropics and has been considered to be the sole malaria vector in French Guiana. The presence of other potential vectors suggests that malaria may be transmitted by other species under certain conditions. From 2006-2011, all anopheline specimens collected from 11 localities were assayed to determine if the Plasmodium circumsporozoite protein was present. In addition to An. darlingi, we found Anopheles oswaldoi, Anopheles intermedius and Anopheles nuneztovari specimens that were infected with Plasmodium sp. Further investigations on the behaviour and ecology of An. oswaldoi, An. intermedius and An. nuneztovari are necessary to determine their role in malaria transmission in French Guiana.

Every year, autochthonous cases of Plasmodium vivax malaria occur in low-endemicity areas of Vale do Ribeira in the south-eastern part of the Atlantic Forest, state of São Paulo, where Anopheles cruzii and Anopheles bellator are considered the primary vectors. However, other species in the subgenus Nyssorhynchus of Anopheles (e.g., Anopheles marajoara) are abundant and may participate in the dynamics of malarial transmission in that region. The objectives of the present study were to assess the spatial distribution of An. cruzii, An. bellator and An. marajoara and to associate the presence of these species with malaria cases in the municipalities of the Vale do Ribeira. Potential habitat suitability modelling was applied to determine both the spatial distribution of An. cruzii, An. bellator and An. marajoara and to establish the density of each species. Poisson regression was utilized to associate malaria cases with estimated vector densities. As a result, An. cruzii was correlated with the forested slopes of the Serra do Mar, An. bellator with the coastal plain and An. marajoara with the deforested areas. Moreover, both An. marajoara and An. cruzii were positively associated with malaria cases. Considering that An. marajoara was demonstrated to be a primary vector of human Plasmodium in the rural areas of the state of Amapá, more attention should be given to the species in the deforested areas of the Atlantic Forest, where it might be a secondary vector.

Registram-se o encontro de larvas e uma pupa de anofelinos em recipientes artificiais desativados. Foram encontradas larvas de Anopheles argyritarsis em uma caixa d'água abandonada, na localidade de Pedregulho, distrito de Ponta Negra, e uma pupa de Anopheles aquasalis em um recipiente experimental, a céu aberto, no distrito de Itaipuaçu, onde ocorreram três casos de malária em 1997. Ambos os encontros ocorreram no município de Maricá, RJ, Brasil.

A riqueza, abundância relativa, flutuação sazonal, preferência por ambiente (peri e intradomicílio) e horários de hematofagismo de espécies de Anopheles foram estudados no município de Pinheiro, Maranhão. As fêmeas foram capturadas mensalmente sobre iscas humanas de maio/1998 a abril/1999, das 18 às 6 horas. Foram coletados 1.321 espécimens pertencentes a 10 espécies de Anopheles, todas do subgênero Nyssorhynchus: A. argyritarsis com 62%, A. darlingi (21,7%), A. albitarsis (4,5%), A. galvaoi (4%), A. triannulatus (3,1%) e A. evansae (2,8%). As espécies A. nuneztovari, A. braziliensis, A. rondoni e A. strodei representaram juntas 1,9%. Os anofelinos ocorreram o ano todo, não havendo diferença significativa no número de exemplares capturados entre as estações chuvosa (51,7%) e seca (48,3%). A freqüência dos anofelinos no peridomicílio foi significativamente maior (p < 0,01) (82%) do que no intradomicílio (18%), preferindo sugar no crepúsculo vespertino e nas primeiras horas da noite.

Anopheles galvaoi, a member of the subgenus Nyssorhynchus, is redescribed based on morphological characters of the adults male and female, fourth-instar larva and pupa. Female, male genitalia, larval and pupal stages are illustrated. Data about medical importance, bionomics, and distribution are given based on literature records. Adult female of An. galvaoi can be easily misidentified as An. benarrochi Gabaldón and An. aquasalis Curry. A few characters are indicated for identifying female and immatures of An. galvaoi. Phylogenetic relationships among An. galvaoi and six other species of the Oswaldoi Subgroup are estimated using COII mtDNA and ITS2 rDNA gene sequences. Lectotype of An. galvaoi, an adult female from Rio Branco, State of Acre, is invalidated.

Anopheles (Anopheles) intermedius and Anopheles (Ano.) mattogrossensis are Brazilian anopheline species belonging to the scarcely studied Anopheles subgenus. Few studies have been done on the genetic differentiation of these species. Both species have been found infected by Plasmodium and are sympatric with other anopheline species from the Nyssorhynchus subgenus. Eighteen enzymatic loci were analyzed in larval specimens of An. intermedius and An. mattogrossensis aiming to estimate the variability and genetic differentiation between these species. An. mattogrossensis population showed higher genetic variability (P = 44.4 and Ho = 0.081 ± 0.031) than that of An. intermedius (P = 33.3 and Ho = 0.048 ± 0.021). Most analyzed loci showed genotypic frequencies according to Hardy-Weinberg equilibrium, except for LAP1 and LAP2 in An. intermedius, and EST1 and PGM loci in An. mattogrossensis. The genetic distance between these species (D = 0.683) was consistent with the inter-specific values reported for Anopheles subgenus. We verified that the polymorphism and heterozygosity percentile values found in both species and compared to those in the literature, showed no relation between the level of isozyme variability and geographical distribution. The low variability found in these two species is probably more related to the niche they occupy than to their geographic distribution.

In the present study, we used morphological characters to estimate phylogenetic relationships among members of the subgenus Anopheles Meigen. Phylogenetic analyses were carried out for 36 species of Anopheles (Anopheles). An. (Stethomyia) kompi Edwards, An. (Lophopodomyia) gilesi (Peryassú), Bironella hollandi Taylor, An. (Nyssorhynchus) oswaldoi (Peryassú) and An. (Cellia) maculatus Theobald were employed as outgroups. One hundred one characters of the external morphology of the adult male, adult female, fourth-instar larva, and pupa were scored and analyzed under the parsimony criterion in PAUP. Phylogenetic relationships among the series and several species informal groups of Anopheles (Anopheles) were hypothesized. The results suggest that Anopheles (Anopheles) is monophyletic. Additionally, most species groups included in the analysis were demonstrated to be monophyletic.
