Purpose: Reverse iontophoresis (RI) is one of the potential techniques used to monitor the concentration of various analytes in body fluids non -invasively. Transdermal extraction of potassium is investigated using RI. In the present work, the effect of potassium on stratum corneum (SC) during RI, feasibility of RI for continuous monitoring of potassium, and use of potassium as internal standard in RI, are investigated. Methods: Tape stripping experiment is carried out to find potassium concentration in SC. RI is carried out continuously for 180 min without passive diffusion and after passive diffusion for 60 min. Skin impedance measurements are done at 20 Hz and 20 kHz. Results: Potassium is found to be in the range 300-650 nmol/cm(2) on SC by tape stripping experiment. Correlation coefficient between blood potassium and extracted potassium through RI after passive diffusion (R-2 = 0.5870) is more than without passive diffusion (R-2 = 0.5117). The skin impedance measurement shows that RI has more effect on SC than superficial layer of SC during RI. Conclusion: The present investigations conclude that it is possible to monitor potassium continuously through RI and using potassium as internal standard in RI.

An immunosensor based on imaging ellipsometry and its potential applications was demonstrated in this paper. It has been proven a fast, reliable, and convenient method to quantify the thickness distribution of protein layers or detect protein concentration in solution. Combined with a protein chip, the immunosensor was able to detect multiple analytes simultaneously without any labeling. Preliminary results demonstrated how this immunosensor could be used to monitor several independent biospecific binding processes in real-time and in situ conditions.

Cardiovascular diseases are nowadays the first cause of mortality worldwide, causing around the 30% of global deaths each year. The risk of suffering from cardiovascular illnesses is strongly related to some factors such as hypertension, high cholesterol levels, diabetes, obesity The combination of these different risk factors is known as metabolic syndrome and it is considered a pandemic due to the high prevalence worldwide. The pathology of the disorders implies a combined cardiovascular therapy with drugs which have different targets and mechanisms of action, to regulate each factor separately. The simultaneous analysis of these drugs turns interesting but it is a complex task since the determination of multiple substances with different physicochemical properties and physiological behavior is always a challenge for the analytical chemist. The complexity of the biological matrices and the difference in the expected concentrations of some analytes require the development of extremely sensitive and selective determination methods. The aim of this work is to fill the gap existing in this field of the drug analysis, developing analytical methods capable of quantifying the different drugs prescribed in combined cardiovascular therapy simultaneously. Liquid chromatography andem mass spectrometry (LCMS/MS) has been the technique of choice throughout the main part of this work, due to the high sensitivity and selectivity requirements.

This report summarizes the results of a characterization of chemical contaminants in the sediments in southwest Puerto Rico. The report is part of a project to integrate various analytical specialties to assess linkages between chemical contaminants and the condition of coral reefs. In this phase of the project, over 120 chemical contaminants were analyzed in sediments collected, including a number of organic (e.g., hydrocarbons), inorganic (e.g., metals), and biological (bacterial) compounds/analytes. The report also provides a preliminary analysis of the association between sediment contaminants and coral species richness. Overall, the levels of chemical contaminants in the study area between Guanica Bay and the town of La Parguera were fairly low. At most of the sites sampled, particularly adjacent to the town of La Parguera, concentrations of organic and inorganic contaminants were below the median values from NOAA’s National Status and Trends Program, which monitors the Nation’s coastal and estuarine waters for chemical contaminants. Elevated levels of a number of contaminant classes were seen at the two sites sampled within Guanica Bay. An initial analysis of modeled PAH (hydrocarbon) data and coral species richness (reef building species) indicated a strong negative correlation between the presence of PAHs in the sediments and coral species richness. Additional work is needed to assess possible reasons for this observed pattern. (PDF contains 126 pages).

Executive Summary: Information found in this report covers the years 1986 through 2005. Mussel Watch began monitoring a suite of trace metals and organic contaminants such as DDT, PCBs and PAHs. Through time additional chemicals were added, and today approximately 140 analytes are monitored. The Mussel Watch Program is the longest running estuarine and coastal pollutant monitoring effort conducted in the United States that is national in scope each year. Hundreds of scientific journal articles and technical reports based on Mussel Watch data have been written; however, this report is the first that presents local, regional and national findings across all years in a Quick Reference format, suitable for use by policy makers, scientists, resource managers and the general public. Pollution often starts at the local scale where high concentrations point to a specific source of contamination, yet some contaminants such as PCBs are atmospherically transported across regional and national scales, resulting in contamination far from their origin. Findings presented here showed few national trends for trace metals and decreasing trends for most organic contaminants; however, a wide variety of trends, both increasing and decreasing, emerge at regional and local levels. For most organic contaminants, trends have resulted from state and federal regulation. The highest concentrations for both metal and organic contaminants are found near urban and industrial areas. In addition to monitoring throughout the nation’s coastal shores and Great Lakes, Mussel Watch samples are stored in a specimen bank so that trends can be determined retrospectively for new and emerging contaminants of concern. For example, there is heightened awareness of a group of flame retardants that are finding their way into the marine environment. These compounds, known as polybrominated diphenyl ethers (PBDEs), are now being studied using historic samples from the specimen bank and current samples to determine their spatial distribution. We will continue to use this kind of investigation to assess new contaminant threats. This thesis investigates the design and implementation of a label-free optical biosensing system utilizing a robust on-chip integrated platform. The goal has been to transition optical micro-resonator based label-free biosensing from a laborious and delicate laboratory demonstration to a tool for the analytical life scientist. This has been pursued along four avenues: (1) the design and fabrication of high-$Q$ integrated planar microdisk optical resonators in silicon nitride on silica, (2) the demonstration of a high speed optoelectronic swept frequency laser source, (3) the development and integration of a microfluidic analyte delivery system, and (4) the introduction of a novel differential measurement technique for the reduction of environmental noise.

The optical part of this system combines the results of two major recent developments in the field of optical and laser physics: the high-$Q$ optical resonator and the phase-locked electronically controlled swept-frequency semiconductor laser. The laser operates at a wavelength relevant for aqueous sensing, and replaces expensive and fragile mechanically-tuned laser sources whose frequency sweeps have limited speed, accuracy and reliability. The high-$Q$ optical resonator is part of a monolithic unit with an integrated optical waveguide, and is fabricated using standard semiconductor lithography methods. Monolithic integration makes the system significantly more robust and flexible compared to current, fragile embodiments that rely on the precarious coupling of fragile optical fibers to resonators. The silicon nitride on silica material system allows for future manifestations at shorter wavelengths. The sensor also includes an integrated microfluidic flow cell for precise and low volume delivery of analytes to the resonator surface. We demonstrate the refractive index sensing action of the system as well as the specific and nonspecific adsorption of proteins onto the resonator surface with high sensitivity. Measurement challenges due to environmental noise that hamper system performance are discussed and a differential sensing measurement is proposed, implemented, and demonstrated resulting in the restoration of a high performance sensing measurement.

The instrument developed in this work represents an adaptable and cost-effective platform capable of various sensitive, label-free measurements relevant to the study of biophysics, biomolecular interactions, cell signaling, and a wide range of other life science fields. Further development is necessary for it to be capable of binding assays, or thermodynamic and kinetics measurements; however, this work has laid the foundation for the demonstration of these applications.

Desenvolvimento de metodologia para a determinação de aflatoxinas em amostras de amendoim usando espectrofluorimetria e análise dos fatores paralelos (PARAFAC). 2012. 92 f. Dissertação (Mestrado em Engenharia Química) - Instituto de Química, Universidade do Estado do Rio de Janeiro , Rio de Janeiro, 2012. Neste trabalho de pesquisa são descritos dois estudos de caso que se baseiam na determinação de aflatoxinas B1, B2, G1 e G2 em amostras de amendoim, utilizando a técnica de espectroscopia de fluorescência molecular. O primeiro estudo tem o objetivo de avaliar a metodologia empregada para a quantificação de aflatoxinas totais em amendoins, utilizando o método clássico de validação fazendo-se o uso da calibração univariada. Os principais parâmetros de desempenho foram avaliados visando certificar a possibilidade de implementação desta metodologia em laboratórios. O segundo estudo está focado na separação e quantificação destas aflatoxinas com a aplicação combinada da espectrofluorimetria e de um método quimiométrico de segunda ordem (PARAFAC) utilizando a calibração multivariada. Esta técnica pode ser empregada como uma alternativa viável para a determinação de aflatoxinas B1, B2, G1 e G2 isoladamente, tradicionalmente é feito por cromatografia líquida de alta eficiência com detector de fluorescência. Porém, como estes analitos apresentam uma larga faixa de sobreposição espectral e as aflatoxinas (B1 e G1) possuem intensidade de sinal de fluorescência bem abaixo das demais, a separação e quantificação das quatro aflatoxinas foi inviável. O estudo foi retomado com a utilização das aflatoxinas B2 e G2 e os resultados alcançados foram satisfatórios. O método utilizado para a quantificação de aflatoxinas totais apresentou bons resultados, mostrando-se como uma importante ferramenta para a determinação destes analitos. Alem disso, comtempla perfeitamente o que é requerido pela legislação brasileira para a análise de aflatoxinas B1, B2, G1 e G2, que tem como exigência em laudos finais de análise a declaração do somatório, em g/kg, destas aflatoxinas, ou seja, sem a necessidade de quantifica-las separadamente

The FOB-3, anew type fiber optic biosensor, is designed to rapidly detect a variety of biological agents or analytes with better stability, sensitivity and specificity. In order to detect Y. Pestis, a sandwich immunoassay was developed by using the purified antibody against antigen FI immobilized on polystyrene probes as the capture antibody and the monoclonal antibody-Cy5 conjugate as the detector. After a series of optimization for the stability, sensitivity and specificity of the FOB-3, 50-1000 ng/ml of antigen FI and 6 x 10(1)-6 x 10(7) CFU/ml Y. pestis could be detected constantly in about 20 min, and Y pestis could be detected specifically from Y. pseudotuberculosis, Y. enterocolitica, B. anthracis and E. coli. Then, 39 blind samples, including 27 tissues of mice infected with Y pestis and 12 tissues of healthy mice as negative control, were detected with the FOB-3. 92.6% infected tissues were identified from the tissues of healthy mice and the tissues containing more than 100 CFU/ml bacteria could be detected by the biosensor. The results demonstrated the feasibility of the FOB-3 as an effective method to detect Y. pestis rapidly and directly from the infected animal specimens with the advantage of portability, simple-operation as well as high sensitivity and specificity. Segundo O`Kane et al, em 2008, foi demonstrado que 88,9% dos erros laboratoriais são realizados na fase pré-analítica. No laboratório de análises clínicas, o sistema de controle da qualidade pode ser definido como toda a ação sistemática necessária para dar confiança e segurança em todos os exames e prevenir a ocorrência de erros. O tempo de armazenamento pode variar de dias a meses ou mesmo anos, influenciando na definição da temperatura de estocagem. O armazenamento de longo prazo pode resultar na criopreservação inadequada para determinados analitos e pode desnaturar as lipoproteínas. Objetivos: Este trabalho teve como objetivo avaliar a fase pré-analítica, controle da qualidade interno e também avaliar o efeito do congelamento (- 80C) quanto ao tempo de armazenamento do soro e plasma de sangue colhido com ácido etilenodiamino tetra-acético (EDTA). As dosagens foram realizadas no Laboratório de Lípides LabLip, e estocadas em - 80 C por três anos, as mesmas foram redosadas no Serviço de Patologia Clínica da Policlínica Piquet Carneiro da UERJ com metodologias iguais e realizada a comparabilidade dos resultados. Foram analisados o perfil lipídico (CT, HDLc e TG) e PCR-US de 103 amostras, 73 no soro e 30 no plasma em dois laboratórios altamente qualificados. Discussão: Após redosagem foram encontrados nas dosagens de HDLc e CT resultados diminuídos respectivamente (correlação coeficiente no soro 0,48 e 0,62) teste t pareado no soro (CT p 0,0012 e HDLc p 0,0001). Conclusões: Os dados obtidos nas avaliações dos resultados de diferentes laboratórios e tempo de estocagem revelaram que as amostras quando armazenadas por um longo período após redosagem no soro, apresentaram diferenças em certos analitos, tais como CT e HDLc, no qual obteve-se resultados significativamente diminuídos, diferentemente no plasma, que após três anos de estocagem a -80C foram redosados e aplicados no teste t pareado, os analitos CT e PCR-US mantiveram a estabilidade.

O objetivo deste estudo foi investigar na saliva de crianças pré-púberes obesas, em comparação com crianças eutróficas, a presença de possíveis diferenças na expressão de mediadores proteicos relacionados à metainflamação através de um estudo observacional transversal. Foram selecionadas 105 crianças pré-púberes com idade entre 5 e 9 anos, sem quaisquer outros comprometimentos sistêmicos ou bucais sendo realizada a mensuração do comprimento da circunferência da cintura (CC), além do peso e estatura para cálculo do IMC e seu escore Z (zscore IMC), de forma a compor 3 grupos: EU - eutrofia (-2SD≤ zscore IMC≤1SD), SP - sobrepeso (1SD < zscore IMC < 2SD) e OB obesidade (zscore IMC ≥ 2SD). Após o exame médico e odontológico foi feita a coleta de sangue e de saliva total não estimulada, de forma protocolada. Amostras séricas e salivares foram analisadas, individualmente, para a dosagem de IL1β, IL6, IL8. IL10, IL12p70, TNFα, MCP1, leptina, grelina e insulina, por Multiplex e adiponectina total e de alto peso molecular (adipoHMW), por ELISA. Além disso, as amostras séricas foram utilizadas para o delineamento hemodinâmico dos pacientes. As amostras salivares de EU e OB foram também analisadas em pools por espectrometria de massa (MS) e a presença de algumas proteínas foi validada por imunoblotting, para a comparação do perfil salivar proteico. As concentrações dos analitos foram comparadas tanto entre os grupos (teste de Kruswall-Wallis), como em relação ao zscore IMC e ao CC (Correlação de Spearman ou Pearson). Dos 12 analitos, somente a adipoHMW não foi detectada em nenhuma das amostras salivares. Na comparação OBxEU houve aumento na concentração total de proteínas salivares, da insulina e leptina sérica e salivar e do MCP1, além de diminuição da adiponectina sérica total e de adipoHMW e uma menor relação adiponectina/leptina (A/L) no grupo OB. As principais correlações obtidas com o zscore IMC foram com as concentrações séricas e salivares de insulina e leptina (positivas) e com a relação A/L (negativa), observando-se também a correlação negativa com a adiponectina total e adipoHMW séricas. Na comparação entre as concentrações séricas e salivares, foi possível detectar correlação positiva entre as dosagens de insulina e leptina, assim como com os valores da relação A/L. Na análise proteômica por MS foram identificadas 670 proteínas, sendo 163 delas com expressão diferenciada na saliva de OB em relação a EU. Dentre estas, encontram-se alteradas proteínas relacionadas à resposta inflamatória humoral, em especial com a via alternativa do sistema complemento e do metabolismo redox. Foram selecionadas três destas proteínas para validação por imunoblotting, que confirmou o aumento do Fator H e a diminuição do Fator B e da tioredoxina na saliva de OB em relação a EU. Considerando os resultados, podemos verificar que embora com menores concentrações absolutas dos mediadores, a saliva mostrou praticamente as mesmas associações observadas no sangue, em especial para a insulina, leptina e relação A/L, sendo possível admitir que a análise salivar venha a ser um bom método diagnóstico não invasivo para a obesidade infantil.

This paper presents a novel platform for the formation of cost-effective PCB-integrated optical waveguide sensors. The sensor design relies on the use of multimode polymer waveguides that can be formed directly on standard PCBs and commercially-available chemical dyes, enabling the integration of all essential sensor components (electronic, photonic, chemical) on low-cost substrates. Moreover, it enables the detection of multiple analytes from a single device by employing waveguide arrays functionalised with different chemical dyes. The devices can be manufactured with conventional methods of the PCB industry, such as solder-reflow processes and pick-and-place assembly techniques. As a proof of principle, a PCB-integrated ammonia gas sensor is fabricated on a FR4 substrate. The sensor operation relies on the change of the optical transmission characteristics of chemically functionalised optical waveguides in the presence of ammonia molecules. The fabrication and assembly of the sensor unit, as well as fundamental simulation and characterisation studies, are presented. The device achieves a sensitivity of approximately 30 ppm and a linear response up to 600 ppm at room temperature. Finally, the potential to detect multiple analytes from a single device is demonstrated using principal-component analysis. © 1983-2012 IEEE.

A rapid and sensitive method was developed and validated for the determination of MCYST (microcystin)-RR, -LR, and [Dha(7)] MCYST-LR in rat plasma by liquid chromatography-tandem mass spectrometry. The analytes were extracted from rat plasma by protein precipitation, followed by solid-phase extraction. Liquid chromatography with electrospray ionization mass spectrometry, operating in selected reaction monitoring (SRM) mode, was used to quantify MCYST-RR, -LR, and [Dha(7)] MCYST-LR in rat plasma. The recoveries for each analyte in rat plasma ranged from 70.8 to 88.7%. The calibration curve was linear within the range from 0.005 to 1.25 mu g mL(-1). The limit of detection were 1.4, 1.0, 0.6 ng mL(-1) for MCYST-RR, -LR, and [Dha(7)] MCYST-LR. The overall precision was determined on three different days. The values for within- and between-day precision in rat plasma were within 15%. This method was applied to the identification and quantification of microcystins in rat plasma with acute exposure of microcystins via intravenous injection.

A sensitive and selective liquid chromatography-tandem mass spectrometry method was developed and validated for the simultaneous quantitative determination of microcystin-LR (MC-LR) and its glutathione conjugate (MC-LR-GSH) in fish tissues. The analytes were extracted from fish liver and kidney using 0.01 M EDTA-Na-2-5% acetic acid, followed by a solid-phase extraction (SPE) on Oasis HLB and silica cartridges. High-performance liquid chromatography (HPLC) with electrospray ionization mass spectrometry, operating in selected reaction monitoring (SRM) mode, was used to quantify MC-LR and its glutathione conjugate in fish liver and kidney. Recoveries of analytes were assessed at three concentrations (0.2, 1.0, and 5 mu g g(-1) dry weight [DW]) and ranged from 91 to 103% for MC-LR, and from 65.0 to 75.7% for MC-LR-GSH. The assay was linear within the range from 0.02 to 5.0 mu g g(-1) DW, with a limit of quantification (LOQ) of 0.02 mu g g(-1) DW. The limit of detection (LOD) of the method was 0.007 mu g g(-1) DW in both fish liver and kidney. The overall precision was determined on three different days. The values for within- and between-day precision in liver and kidney were within 15%. This method was applied to the identification and quantification of MC-LR and its glutathione conjugate in liver and kidney of fish with acute exposure of MC-LR. (c) 2007 Elsevier B.V. This paper describes a novel method that applies pressure-assisted field-amplified sample injection with reverse migrating micelles (PA-FASI-RMM) for the online concentration of neutral analytes in MEKC with a low-pH BGE. After injection of a plug of water into the separation capillary, negative voltage and positive pressure were simultaneously applied to initialize PA-FASI-RMM injection. The hydrodynamic flow generated by the positive pressure compensated the reverse EOF in the water plug and allowed the water plug to remain in the capillary during FASI with reverse migrating micelles (FASI-RMM) to obtain a much longer injection time than usual, which improved stacking efficiency greatly. Equations describing this injection mode were introduced and were supported by experimental results. For a 450-s online PA-FASI-RMM injection, three orders of magnitude sample enhancement in terms of peak area could be observed for the steroids and an achievement of detection limits was between 1 and 10 ng/mL.

A preliminary survey was carried out in April 2003 to estimate the levels of nonylphenol (NP), bisphenol-A (BPA) and residual DDTs in Lake Donghu. After then, the sediments of some areas of the lake were moved out for clearing the lake by the local government. Therefore, the variance of NP and BPA after the clearance of sediment in the surface water was determined from December 2003 to May 2004. Sediments, surface water and fish were collected from four sub-lake areas and the analytes were qualified by GC/MS in SIM mode after concentration onto an Oasis solid phase extraction cartridge. NP and BPA values ranged between 5.46-119.10 and 0.9913.42 mg/kg dw, respectively, in sediments, 75.2-179.6 and 15.1-62.5 mu g/L in surface water. Meanwhile, the bioaccumulation factors (BCFs) of NP and BPA were calculated in fish livers. Plasma vitellogenin (VTG), a sensitive biomarker showing oestrogenicity was detected in the captured male Wuchang bream (Megalobrama amblycephala Yih) and common carp (Cyprinus carpio). After sediment clearance, the concentrations of NP and BPA decreased to 0.65-25.04 and 0.04-21.32 mu g/L. The results indicate the presence of high-dose xenoestrogenic NP and BPA, probably major contributors which associate with VTG induction in Lake Donghu male fish.
