La caracterització funcional de dos gens en la peridermis, la ω hidroxilasa d'àcids grassos CYP86A33 -candidata per la funcionalització del carboni ω-terminal dels monòmers alifàtics de la suberina- i la ketoacyl-CoA sintasa StKCS6 -candidata per elongar àcids grassos o derivats llargs de suberina i ceres- es realitza per silenciament per RNA d'interferència en patata. La deficiència de CYP86A33 comporta una gran reducció dels monòmers principals de la suberina, l'àcid gras ω-hidroxilat i l'α,ω-diàcid C18:1, juntament amb una reducció total de la quantitat de suberina del 60%. Aquesta deficiència altera l'estructura lamel·lar típica de la suberina, així com també la funció barrera de la peridermis. La deficiència en StKCS6 comporta que els monòmers de la suberina de 28 carbonis o més llargs es redueixin i que els de 26 carbonis o més curts s'incrementin. Aquesta deficiència suggereix que la llargada dels compostos alifàtics pot contribuir a les propietats impermeabilitzants de la peridermis.

The integration of processes at different scales is a key problem in the modelling of cell populations. Owing to increased computational resources and the accumulation of data at the cellular and subcellular scales, the use of discrete, cell-level models, which are typically solved using numerical simulations, has become prominent. One of the merits of this approach is that important biological factors, such as cell heterogeneity and noise, can be easily incorporated. However, it can be difficult to efficiently draw generalizations from the simulation results, as, often, many simulation runs are required to investigate model behaviour in typically large parameter spaces. In some cases, discrete cell-level models can be coarse-grained, yielding continuum models whose analysis can lead to the development of insight into the underlying simulations. In this paper we apply such an approach to the case of a discrete model of cell dynamics in the intestinal crypt. An analysis of the resulting continuum model demonstrates that there is a limited region of parameter space within which steady-state (and hence biologically realistic) solutions exist. Continuum model predictions show good agreement with corresponding results from the underlying simulations and experimental data taken from murine intestinal crypts.

Im Rahmen der vorliegenden Dissertation wurden Untersuchungen zur Expression und Funktion der respiratorischen Proteine Neuroglobin (Ngb) und Cytoglobin (Cygb) in Vertebraten durchgeführt. Beide Globine wurden erst kürzlich entdeckt, und ihre Funktionen konnten trotz vorliegender Daten zur Struktur und biochemischen Eigenschaften dieser Proteine bisher nicht eindeutig geklärt werden. Im ersten Abschnitt der vorliegenden Arbeit wurde die zelluläre und subzelluläre Lokalisation von Neuroglobin und Cytoglobin in murinen Gewebeschnitten untersucht. Die Expression von Ngb in neuronalen und endokrinen Geweben hängt offensichtlich mit den hohen metabolischen Aktivitäten dieser Organe zusammen. Insbesondere im Gehirn konnten regionale Unterschiede in der Ngb-Expression beobachtet werden. Dabei korrelierte eine besonders starke Neuroglobin-Expression mit Gehirnbereichen, die bekanntermaßen die höchsten Grundaktivitäten aufweisen. In Anbetracht dessen liegt die Funktion des Neuroglobins möglicherweise im basalen O2-Metabolismus dieser Gewebe, wobei Ngb als O2-Lieferant und kurzfristiger O2-Speicher den vergleichsweise hohen Sauerstoffbedarf vor Ort sicherstellen könnte. Weitere Funktionen in der Entgiftung von ROS bzw. RNS oder die kürzlich publizierte mögliche Rolle des Ngb bei der Verhinderung der Mitochondrien-vermittelten Apoptose durch eine Reduktion des freigesetzten Cytochrom c wären darüber hinaus denkbar. Die Cygb-Expression im Gehirn beschränkte sich auf relativ wenige Neurone in verschiedenen Gehirnbereichen und zeigte dort vorwiegend eine Co-Lokalisation mit der neuronalen NO-Synthase. Dieser Befund legt eine Funktion des Cytoglobins im NO-Metabolismus nahe. Quantitative RT-PCR-Experimente zur mRNA-Expression von Ngb und Cygb in alternden Säugern am Bsp. der Hamsterspezies Phodopus sungorus zeigten keine signifikanten Änderungen der mRNA-Mengen beider Globine in alten im Vergleich zu jungen Tieren. Dies widerspricht publizierten Daten, in denen bei der Maus anhand von Western Blot-Analysen eine Abnahme der Neuroglobin-Menge im Alter gezeigt wurde. Möglicherweise handelt es sich hierbei um speziesspezifische Differenzen. Die im Rahmen dieser Arbeit durchgeführte vergleichende Sequenzanalyse der humanen und murinen NGB/Ngb-Genregion liefert zum einen Hinweise auf die mögliche Regulation der Ngb-Expression und zum anderen eine wichtige Grundlage für die funktionellen Analysen dieses Gens. Es konnte ein minimaler Promotorbereich definiert werden, der zusammen mit einigen konservierten regulatorischen Elementen als Basis für experimentelle Untersuchungen der Promotoraktivität in Abhängigkeit von äußeren Einflüssen dienen wird. Bioinformatische Analysen führten zur Identifizierung des sog. „neuron restrictive silencer element“ (NRSE) im Ngb-Promotor, welches vermutlich für die vorwiegend neuronale Expression des Proteins verantwortlich ist. Die kontrovers diskutierte O2-abhängige Regulation der Ngb-Expression konnte hingegen anhand der durchgeführten komparativen Sequenzanalysen nicht bestätigt werden. Es wurden keine zwischen Mensch und Maus konservierten Bindestellen für den Transkriptionsfaktor HIF-1 identifiziert, der die Expression zahlreicher hypoxieregulierter Gene, z.B. Epo und VEGF, vermittelt. Zusammen mit den in vivo-Daten spricht dies eher gegen eine Regulation der Ngb-Expression bei verminderter Verfügbarkeit von Sauerstoff. Die Komplexität der Funktionen von Ngb und Cygb im O2-Stoffwechsel der Vertebraten macht den Einsatz muriner Modellsysteme unerlässlich, die eine sukzessive Aufklärung der Funktionen beider Proteine erlauben. Die vorliegende Arbeit liefert auch dazu einen wichtigen Beitrag. Die hergestellten „gene-targeting“-Vektorkonstrukte liefern in Verbindung mit den etablierten Nachweisverfahren zur Genotypisierung von embryonalen Stammzellen die Grundlage zur erfolgreichen Generierung von Ngb-knock out sowie Ngb- und Cygb-überexprimierenden transgenen Tieren. Diese werden für die endgültige Entschlüsselung funktionell relevanter Fragestellungen von enormer Bedeutung sein.

The ATP-binding cassette transporter A1 (ABCA1) mediates the transport of cholesterol, phospholipids, and other lipophilic molecules across cellular membranes. Recent data provide evidence that ABCA1 plays an important role in placental function but the exact cellular sites of ABCA1 action in the placenta remain controversial. To clarify this issue, we analyzed the cellular and subcellular localization of ABCA1 with immunocytochemistry, immunofluorescence and subsequent confocal or immunofluorescence microscopy in different types of isolated primary placenta cells: cytotrophoblast cells, amnion epithelial cells, villous macrophages (Hofbauer cells), and mesenchymal cells isolated from chorionic membrane and placental villi. After 12 h of cultivation, primary cytotrophoblast cells showed intensive membrane and cytoplasmic staining for ABCA1. After 24 h, with progressive syncytium formation, ABCA1 staining intensity was markedly reduced and ABCA1 was dispersed in the cytoplasm of the forming syncytial layer. In amnion epithelial cells, placental macrophages and mesenchymal cells, ABCA1 was predominantly localized at the cell membrane and cytoplasmic compartments partially corresponding to the endoplasmic reticulum. In these cell types, the ABCA1 staining intensity was not dependent on the cultivation time. In conclusion, ABCA1 shows marked expression levels in diverse placental cell types. The multitopic localization of ABCA1 in diverse human placental cells not all directly involved in materno-fetal exchange suggests that this protein may not only participate in transplacental lipid transport but could have additional regulatory functions.

Morphogenesis is the process by which the 3-dimensional structure of the developing embryo takes shape. We are studying xlcaax-1, a gene whose product can be used as a molecular marker for several morphogenetic events. We report here the cellular and subcellular localization of the xlcaax-1 protein during development of Xenopus laevis. Whole mount immunocytochemistry and immunoperoxidase staining of tissue sections showed that during development the xlcaax-1 protein accumulation was coincident with the differentiation of the epidermis, pronephros and mesonephros. In the pronephros and mesonephros the xlcaax-1 protein was localized to the basolateral membrane of differentiated tubule epithelial cells. Thus, the xlcaax-1 protein served as a marker for tubule formation and polarization during Xenopus kidney development. Xlcaax-1 may also be used as a marker for the functional differentiation of the epidermis and the epidermally derived portions of the lens and some cranial nerves. The xlcaax-1 protein was most abundant in kidney and immunogold EM analysis showed that the xlcaax-1 protein was highly enriched in the basal infoldings of the basolateral membrane of the epithelial cells in adult kidney distal tubules. The xlcaax-1 protein was also localized in other ion transporting epithelia. The localization pattern and preliminary functional assays of xlcaax-1 suggest that the protein may function in association with an ion transport channel or pump.^ Cell migration and cell-cell interactions play important roles in numerous processes during morphogenesis. One of these is the formation of the pronephric (wolffian) duct (PD), which connects the pronephros to the cloaca. It is currently accepted that in most amphibians the pronephric duct is formed by active migration of the pronephric duct rudiment (PDR) cells along a pre-determined pathway. However, there is evidence that in Xenopus, the PD may be formed entirely by in situ segregation of cells out of the lateral mesoderm. In this study, we showed, using PDR ablation and X. laevis - X. borealis chimeras, that PD elongation in Xenopus required both active cell migration and an induced recruitment of cells from the posterior lateral plate mesoderm. We also showed that PDR cell migration was limited to only a few stages during development and that this temporal control is due, at least in part, to changes in the competence of the PD pathway to support cell migration. In addition, our data suggested that an alkaline phosphatase (APase) adhesion gradient may be involved in determining this competence. ^

Three plant sulfate transporter cDNAs have been isolated by complementation of a yeast mutant with a cDNA library derived from the tropical forage legume Stylosanthes hamata. Two of these cDNAs, shst1 and shst2, encode high-affinity H+/sulfate cotransporters that mediate the uptake of sulfate by plant roots from low concentrations of sulfate in the soil solution. The third, shst3, represents a different subtype encoding a lower affinity H+/sulfate cotransporter, which may be involved in the internal transport of sulfate between cellular or subcellular compartments within the plant. The steady-state level of mRNA corresponding to both subtypes is subject to regulation by signals that ultimately respond to the external sulfate supply. These cDNAs represent the identification of plant members of a family of related sulfate transporter proteins whose sequences exhibit significant amino acid conservation in filamentous fungi, yeast, plants, and mammals.

The effect of high power ultrasound waves on physical, biochemical, and microbial properties of meat have been the subject of a great deal of interest in recent years. The present review details the basic principles underlying the effects of ultrasound on the properties of food systems, followed by discussion of specific effects of high power ultrasound on meat products, including muscle, cellular, and subcellular components. In addition, the specific effects of high power ultrasound on the following parameters are discussed: enzyme activities and efficiencies, muscle proteolysis, quality criteria such as tenderness; extraction of protein, gelation, and restructuring of meat products and germicidal properties against meat micro-organisms.

Cancer remains one of the world’s most devastating diseases, with more than 10 million new cases every year. However, traditional treatments have proven insufficient for successful medical management of cancer due to the chemotherapeutics’ difficulty in achieving therapeutic concentrations at the target site, non-specific cytotoxicity to normal tissues, and limited systemic circulation lifetime. Although, a concerted effort has been placed in developing and successfully employing nanoparticle(NP)-based drug delivery vehicles successfully mitigate the physiochemical and pharmacological limitations of chemotherapeutics, work towards controlling the subcellular fate of the carrier, and ultimately its payload, has been limited. Because efficient therapeutic action requires drug delivery to specific organelles, the subcellular barrier remains critical obstacle to maximize the full potential of NP-based delivery vehicles. The aim of my dissertation work is to better understand how NP-delivery vehicles’ structural, chemical, and physical properties affect the internalization method and subcellular localization of the nanocarrier. In this work we explored how side-chain and backbone modifications affect the conjugated polymer nanoparticle (CPN) toxicity and subcellular localization. We discovered how subtle chemical modifications had profound consequences on the polymer’s accumulation inside the cell and cellular retention. We also examined how complexation of CPN with polysaccharides affects uptake efficiency and subcellular localization. This work also presents how changes to CPN backbone biodegradability can significantly affect the subcellular localization of the material. A series of triphenyl phosphonium-containing CPNs were synthesized and the effect of backbone modifications have on the cellular toxicity and intracellular fate of the material. A mitochondrial-specific polymer exhibiting time-dependent release is reported. Finally, we present a novel polymerization technique which allows for the controlled incorporation of electron-accepting benzothiadiazole units onto the polymer chain. This facilitates tuning CPN emission towards red emission. The work presented here, specifically, the effect that side-chain and structure, polysaccharide formulation and CPN degradability have on material’s uptake behavior, can help maximize the full potential of NP-based delivery vehicles for improved chemotherapeutic drug delivery.

Algunes nanotècniques recents permeten la manipulació de biomolècules i cèl·lules en escala nanomètrica amb la mesura simultània de la força aplicada amb resolució de piconewtons. Aquestes escales de desplaçament i força, i la possibilitat de treballar en medi líquid, fan que siguin eines molt útils per a l'estudi de les propietats mecàniques de molècules i cèl·lules individuals en condicions fisiològiques. Entre les tècniques més utilitzades es troben el microscopi de força atòmica, les trampes de làser i les microesferes magnètiques. En aquest treball es descriuen els principis de funcionament d'aquestes tècniques en aplicacions biològiques i, en particular, en l'estudi de la mecànica molecular i cel·lular.

In this study, we have compared the effector functions and fate of a number of human CTL clones in vitro or ex vivo following contact with variant peptides presented either on the cell surface or in a soluble multimeric format. In the presence of CD8 coreceptor binding, there is a good correlation between TCR signaling, killing of the targets, and Fast-mediated CTL apoptosis. Blocking CD8 binding using (alpha3 domain mutants of MHC class I results in much reduced signaling and reduced killing of the targets. Surprisingly, however, Fast expression is induced to a similar degree on these CTLs, and apoptosis of CTL is unaffected. The ability to divorce these events may allow the deletion of antigen-specific and pathological CTL populations without the deleterious effects induced by full CTL activation.

Foram realizadas observações sobre a duração do período embrionário de Biomphalaria glabrata e Biomphalaria tenagophila, durante um ano, em condições de laboratório. As médias, expressas em dias, encontradas para Biomphalaria glabrata e Biomphalaria tenagophila foram respectivamente de 7,6 e 8,0 dias. Essas diferenças não foram significativas ao nível de 5%. O desenvolvimento mais rápido dos embriões até a eclosão verificou-se na estação quente, em ambas as espécies, quando a temperatura média da água foi de aproximadamente 28°C. Após a análise da matriz de correlação verificou-se que a influência da temperatura sobre o tempo de desenvolvimento embrionário foi pouco intensa: Biomphalaria glabrata apresentou maior correlação com a temperatura mínima do ambiente (36,2%) e com a temperatura da água (36,0%) enquanto Biomphalaria tenagophila apresentou maior correlação com a temperatura mínima do ambiente (27,0%).

En Charambirá, localidad del município de Istmina (Chocó) en la costa Pacífica de Colombia, donde existe un problema de malaria endémica, se estudió la fluctuación de la población larval y las características de los criaderos de Anopheles (Kerteszia) neivai, especie considerada como posible vectora de malaria en esa región del país. La vegetación circundante fue dividida en tres estratos de acuerdo al grado de cobertura foliar. Se demarcaron cuadrantes de 100 m² en cada estrato para el muestreo de plantas epífitas de la família Bromeliaceae, en las cuales se acumula agua que sirve como criadero para esta especie de anofelino. Se tomaron datos de temperatura, pH y volumen del agua contenida en cada bromelia. El mayor número de larvas se detectó en el estrato 1 (manglar) a una altitud inferior a 4 m, pero no se encontró evidencia significativa de estratificación vertical de la población larval de A. neivai hasta los 8 m. Se evidenció una correlación lineal positiva entre el número de larvas y el volumen de agua contenida en cada bromelia; por otra parte se observó también una correlación directa entre la precipitación mensual acumulada y la fluctuación poblacional de esta especie. Los índices larvales mas altos se observaron entre los meses de marzo a abril y de julio a agosto. La mortalidad larval fue alta en el primer estadío (43,5%) y solo un 23,7% sobrevivió hasta el cuarto.

Devido aos danos dos ataques de Urbanus acawoios (Williams, 1926) (Lepidoptera: Hesperiidae) em palheteiras (Clitoria racemosa) (Leguminosae) em Manaus, AM e a total ausência de qualquer informação a respeito desta espécie, o presente trabalho foi desenvolvido visando a obtenção de conhecimentos que facilitem a identificação de métodos seguros e eficientes para o seu controle. Os estudos biológicos revelaram que este inseto é multivoltino, tendo duração média de 41,89 ± 0,98 dias por geração. As condições climáticas na cidade de Manaus, a disponibilidade de alimentos, e possivelmente a ausência de inimigos naturais eficientes, parecem ser os fatores principais responsáveis pela ocorrência de erupções populacionais desta espécie. Enquanto o estágio larval consiste de 5 fases (ínstares) e dura em média 18,46 ± 1,28 dias, o da pupa necessita 9,96 ± 1,05, e o adulto 13,47 ± 0,27 dias, sob condições de 30°C e 85% UR. Estudos morfológicos foram incluídos.

A fase larval do predador Nusalala uruguaya (Navás, 1923) alimentada com pulgão-preto-dos-citros Toxoptera citricidus (Kirk), pulgão-da-couve Brevicoryne brassicae (L.) e pulgão-do-picao Dactynotus sp. foi estudada, em condições de laboratório à 25 ± 2°C, UR de 70±10% e fotofase de 14 horas. A espécie de presa consumida afetou a fase larval do predador, influenciando a duração e a viabilidade de cada instar, a largura da cápsula cefálica, o peso e o tamanho de suas larvas. Dactynotus sp. foi a espécie mais consumida e que propiciou o melhor desenvolvimento larval, sendo que T. citricidus acarretou 100% de mortalidade no primeiro instar. As larvas de N. uruguaya somente se alimentaram de pre-ovos e larvas da sua própria espécie.
