This paper examines the daily morphological responses of Sununga Beach, an embayed beach located on the south-eastern Brazilian coast, to storms in the South Atlantic Ocean. The main mechanisms and timing of beach erosion and accretion, the relationship between wave height and direction, and beach volume changes are considered, to establish a qualitative model for short-term embayed beach morphological changes. The methodology consisted of daily topographic surveys during the month of May in 2001, 2002, and 2003, using an RTK-GPS (real-time kinematics global positioning system). Weather and wave model results were used to correlate hydrodynamics and beach morphology. The results indicate that the morphodynamics of Sununga Beach are characterized by a process of beach rotation, which occurred more or less clearly during all three surveys. Unlike what has been commonly described in the literature for longer time intervals and alternations of fair and stormy weather, the beach rotation processes on Sununga Beach occurred under conditions of moderate-to-high wave energy change (wave heights greater than 2 m). An integrated evaluation of the behaviour of the meteorological aspects, together with beach morphology, enabled us to recognize that extra-tropical cyclones were the most important agent in remobilizing the beach planform, whether in beach rotation or in cross-shore erosion. Objective: The aim of this study was to compare the correspondence between gap formation and apical microleakage in root canals filled with epoxy resin-based (AH Plus) combined or not with resinous primer or with a dimethacrylate-based root canal sealer (Epiphany). Material and Methods: Thirty-nine lower single-rooted human premolars were filled by the lateral condensation technique (LC) and immersed in a 50-wt% aqueous silver nitrate solution at 37 degrees C (24 h). After longitudinal sectioning, epoxy resin replicas were made from the tooth specimens. Both the replicas and the specimens were prepared for scanning electron microscopy (SEM). The gaps were observed in the replicas. Apical microleakage was detected in the specimens by SEM/energy dispersive spectroscopy (SEM/EDS). The data were analyzed statistically using an Ordinal Logistic Regression model and Analysis of Correspondence (alpha=0.05). Results: Epiphany presented more regions containing gaps between dentin and sealer (p<0.05). There was correspondence between the presence of gaps and microleakage (p<0.05). Microleakage was similar among the root-filling materials (p>0.05). Conclusions: The resinous primer did not improve the sealing ability of AH Plus sealer and the presence of gaps had an effect on apical microleakage for all materials.

Objective: To compare two methods of respiratory inductive plethysmography (RIP) calibration in three different positions. Methods: We evaluated 28 healthy subjects (18 women and 10 men), with a mean age of 25.4 +/- 3.9 years. For all of the subjects, isovolume maneuver calibration (ISOCAL) and qualitative diagnostic calibration (QDC) were used in the orthostatic, sitting, and supine positions. In order to evaluate the concordance between the two calibration methods, we used ANOVA and Bland-Altman plots. Results: The values of the constant of proportionality (X) were significantly different between ISOCAL and QDC in the three positions evaluated: 1.6 +/- 0.5 vs. 2.0 +/- 1.2, in the supine position, 2.5 +/- 0.8 vs. 0.6 +/- 0.3 in the sitting position, and 2.0 +/- 0.8 vs. 0.6 +/- 0.3 in the orthostatic position (p < 0.05 for all). Conclusions: Our results suggest that QDC is an inaccurate method for the calibration of RIP. The K values obtained with ISOCAL reveal that RIP should be calibrated for each position evaluated.

Background: Medical education can affect medical students' physical and mental health as well as their quality of life. The aim of this study was to assess medical students' perceptions of their quality of life and its relationship with medical education. Methods: First-to sixth-year students from six Brazilian medical schools were interviewed using focus groups to explore what medical student's lives are like, factors related to increases and decreases of their quality of life during medical school, and how they deal with the difficulties in their training. Results: Students reported a variety of difficulties and crises during medical school. Factors that were reported to decrease their quality of life included competition, unprepared teachers, excessive activities, and medical school schedules that demanded exclusive dedication. Contact with pain, death and suffering and harsh social realities influence their quality of life, as well as frustrations with the program and insecurity regarding their professional future. The scarcity of time for studying, leisure activities, relationships, and rest was considered the main factor of influence. Among factors that increase quality of life are good teachers, classes with good didactic approaches, active learning methodologies, contact with patients, and efficient time management. Students also reported that meaningful relationships with family members, friends, or teachers increase their quality of life. Conclusion: Quality of teachers, curricula, healthy lifestyles related to eating habits, sleep, and physical activity modify medical students' quality of life. Lack of time due to medical school obligations was a major impact factor. Students affirm their quality of life is influenced by their medical school experiences, but they also reframe their difficulties, herein represented by their poor quality of life, understood as necessary and inherent to the process of becoming doctors.

The aim of this study was to analyze the rat temporomandibular joint (TMJ) synovial membrane at different ages using light, scanning, and transmission electron microscopy. Under light microscopic analysis, the TMJ structures were observed such as condyle, capsule, disk, the synovial membrane collagen type, and cells distribution. In the scanning electron microscopy, the synovial membrane surface exhibited a smooth aspect in young animals and there was an increase with ageing in the number of folds. The transmission electron microscopic analysis showed more synoviocytes in the synovial layer in the young group and still a great number of vesicles and cisterns dilation of rough endoplasmic reticulum in the aged group. In the three groups, a dense layer of collagen fibers in the synovial layer and cytoplasmic extensions were clearly seen. It was possible to conclude that synovial membrane structures in aged group showed alterations contributing to the decrease in joint lubrication and in the sliding between disk and joint surfaces. These characteristic will reflect in biomechanics of chewing, and may cause the TMJ disorders, currently observed in clinical processes. Microsc. Res. Tech. (c) 2012 Wiley Periodicals, Inc.

Semi-qualitative probabilistic networks (SQPNs) merge two important graphical model formalisms: Bayesian networks and qualitative probabilistic networks. They provade a very Complexity of inferences in polytree-shaped semi-qualitative probabilistic networks and qualitative probabilistic networks. They provide a very general modeling framework by allowing the combination of numeric and qualitative assessments over a discrete domain, and can be compactly encoded by exploiting the same factorization of joint probability distributions that are behind the bayesian networks. This paper explores the computational complexity of semi-qualitative probabilistic networks, and takes the polytree-shaped networks as its main target. We show that the inference problem is coNP-Complete for binary polytrees with multiple observed nodes. We also show that interferences can be performed in time linear in the number of nodes if there is a single observed node. Because our proof is construtive, we obtain an efficient linear time algorithm for SQPNs under such assumptions. To the best of our knowledge, this is the first exact polynominal-time algorithm for SQPn. Together these results provide a clear picture of the inferential complexity in polytree-shaped SQPNs.

[EN]Qualitative and quantitative research approaches are often considered as incompatible, and when they are brought together in a study, the analyses often stay within the realm of the same research field. The study at hand aims at combining the two methods from the perspectives of different disciplines and tries to determine to which degree a corpus-based analysis might support the traditional content-focused approach to qualitative data and render additional results.

Ziel der Arbeit ist die Entwicklung eines allgemeingültigen, im mittleren Maßstabsbereich durchgeführten, GIS-Bewertungsverfahrens zur Ermittlung der Grundwasserneubildungsrate unter Berücksichtigung einer langzeitlichen Landnutzungsänderung sowie die Darstellung eines Szenarios zur Grundwassergefährdungsabschätzung für ausgewählte rüstungsspezifi-sche organische Verbindungen. Das Untersuchungsgebiet befindet sich in einem während der beiden Weltkriege stark militär- und rüstungstechnisch genutzten Raum. Die Beurtei-lungsszenarien der Grundwassergefährdung erfolgen rein qualitativ.Die Abhängigkeit der Grundwasserneubildungsrate von der Landnutzungsänderung ist deut-lich zu erkennen. Mit zunehmender Flächenversiegelung, einher gehend mit veränderten Sickerwasserraten und des oberflächennahen Abflusses, reduziert sich die Grundwasser-neubildungsrate deutlich. Die Abschätzung der Grundwassergefährdung für die sprengstofftypischen Verbindungen erfolgt im ersten Schritt emissionsbezogen für die ungesättigte Zone unter Berücksichtigung der Standort- und Bodeneigenschaften und der physikalisch-chemischen Stoffeigenschaften. Im zweiten Schritt erfolgt eine immissionsbezogene Betrachtung der potenziellen Belastun-gen von Trinkwasserbrunnen.Trotz starker Generalisierungseffekte bietet dieser stoffspezifische Bewertungsansatz in Verbindung mit physikalischen Methoden für organische Verbindungen einen guten ersten Überblick über eine potenzielle Gefährdungen des Grundwassers. Die Datenbestände der entwickelten GIS-Anwendung sind leicht erweiterbar, so dass eine Abschätzung der potenzi-ellen Gefährdung auch für andere organische Stoffgruppen sowie auch eine Übertragung auf andere Untersuchungsgebiete möglich ist.

Iodine is an essential microelement for human health because it is a constituent of the thyroid hormones that regulate growth and development of the organism. Iodine Deficiency Disorders (IDDs) are believed to be one of the commonest preventable human health problems in the world today, according to the World Health Organization: that diseases include endemic goiter, cretinism and fetal abnormalities, among others, and they are caused by lack of iodine in the diet, that is the main source of iodine. Since iodine intake from food is not enough respect to human needs, this can be remedied through dietary diversification, mineral supplementation, food fortification, or increasing the concentration and/or bioavailability of mineral elements in the edible portions of crops through agricultural intervention or genetic selection (biofortification). The introduction of iodized salt is a strategy widely used and accepted to eradicate iodine deficiency, because it is an inexpensive source of stable iodine. Since the intake of salt, though iodized, must still be limited according to the risk of cardiovascular disease, so the increase of iodine content in plants for the production of functional foods is representing a field of study of particular interest and a potential market. In Italy potatoes enriched with iodine are produced by a patented procedure of agronomic biofortification for the fresh market since several years, furthermore they are recently accepted and recommended by Italian Thyroid Association, as an alternative source of iodine. Researches performed during the PhD course intended to characterize this innovative vegetables products, focusing the attention on different aspects, such as chemistry, agriculture, and quality of fresh and fried potatoes. For this purpose, lipid fraction of raw material was firstly investigated, in order to assess whether the presence of iodine in plant metabolism can affect fatty acid or sterol biosynthesis, according to the hypothesis that iodine can be bounded to polyunsaturated fatty acids of cell membranes, protecting them from peroxydation; phytosterols of plant sterol are also studied because their importance in reducing serum cholesterol, especially in potato plant sterols are also involved in synthesis of glycoalkaloid, a family of steroidal toxic secondary metabolites present in plants of the Solanaceae family. To achieve this goal chromatographic analytical techniques were employed to identify and quantify fatty acids and sterols profile of common and iodine enriched row potatoes. Another aim of the project was to evaluate the effects of frying on the quality of iodine-enriched and common potatoes. Since iodine-enriched potatoes are nowadays produced only for the fresh market, preliminary trials of cultivation under controlled environment were carried out to verify if potato varieties suitable for processing were able to absorb and accumulate iodine in the tuber. In a successive phase, these varieties were grown in the field, to evaluate their potential productivity and quality at harvest and after storage. The best potato variety to be destined for processing purposes, was finally subjected to repeated frying cycles; the effects of lipid oxidation on the composition and quality of both potatoes and frying oil bath were evaluated by chromatographic and spectrophotometric analytical techniques. Special attention were paid on volatile compounds of fried potatoes.

The present dissertation collects the results of three different research trials which have the common aim to understand the effects of swine welfare (both at farm level and during transport) on the main fresh and dry-cured meat characteristics. The first trial was carried out in order to compare the effects of illumination regimes differing in light duration or light intensity on meat and ham quality of Italian heavy pigs. The results of this trial support the conclusion that, within a moderate range of light intensity and given an appropriate dark period for animal rest, an increase of light duration or intensity above the minimum mandatory levels has no negative impact on carcass composition, meat or long-cured hams quality. The second trial was designed with the aim to investigate the effects of water restriction on growth traits, animal welfare and meat and ham quality of liquid-fed heavy pigs. Overall, the parameters analyzed as concerns growth rate, behavioural traits, blood, as well as carcass, fresh meat and cured hams quality were not affected by the absence of fresh drinking water. However, since liquid feeding did not suppress drinker use or drinker manipulation in the experimental groups, water restriction does not appear to be an applicable method to obtain a reduction of water waste. The third trial, which was carried out in Canada, tested the effectiveness of water sprinkling market-weight pigs (115±10Kg BW) before and after transport in reducing the heat stress experienced under commercial transport conditions. Our results show that the water sprinkling protocol proposed may reduce heat stress during transport and improve pork quality, particularly in specific trailer compartments. This body of research supports the general conclusion that swine welfare could be improved in different scenarios through simple and cost-effective means, without negatively affecting the quality of the main animal-derived products.

This thesis focused on the polymer’s influence on the interaction of polymeric NPs with epithelial cells. Furthermore, the measurement of single submicron nanoparticles in a commercially available flow cytometer was established, to provide a new method in the toolbox for nanoparticle-cell studies. This gave way to develop a routine for the absolute quantification of intracellular NPs via flow cytometry. rnThe cellular uptake of poly(methyl methacrylate) (PMMA), polystyrene (PS) and poly(L-lactide) (PLLA) nanoparticles was investigated via flow cytometry. PLLA-NPs were internalized the most efficiently. But upon co-incubation of PS and PLLA particles with cells, the two particles mutually influenced their uptake, slightly shifting the relative uptake efficiencies. This phenomenon should be based on specific properties of the different polymer materials. The findings indicated a competition (which is strongly influenced by properties of the respective polymeric material) for the uptake into the cells, allegedly due to competition for specific coatings with serum components that enhances the NPs’ cellular uptake. The fluorescence of single 150 nm particles was determined with a benchtop cytometer, breaching the machine’s detection limit but yielding precise NP fluorescence standardization factors. Up to now, these standardization factors are mostly determined by spectroscopic analysis of the particles’ dye content. Finally a flow cytometric routine for absolute particle counting in cells was devised. This quantitation revealed a low uptake efficiency for un-functionalized PMMA NPs of less than 150 NPs (approx. 0,001 % of added) per cell.rn

Il presente lavoro di tesi ha avuto lo scopo di valutare l’effetto dell’insorgenza delle anomalie “white-striping” e “wooden breast” (sia separatamente che contestualmente nell’ambito dello stesso filetto) su composizione chimica, profilo in acidi grassi e suscettibilità all’ossidazione lipidica e proteica. Le modificazioni riscontrate nella composizione chimica (maggiori livelli di collagene e lipidi a discapito del tenore in proteine) determinano una ridotta qualità nutrizionale con possibili effetti negativi sulla percezione del consumatore, oltre che sulla capacità di ritenzione idrica come riscontrato in precedenti studi. Per quanto riguarda il profilo in acidi grassi, l’effetto delle anomalie è stato più limitato rispetto a quanto ipotizzato. Infatti, nonostante l’aumento dei lipidi totali, le carni di petto anomale non presentano modificazioni nel contenuto di acidi grassi monoinsaturi e polinsaturi totali. Le uniche differenze hanno riguardato il totale degli acidi grassi saturi e il contenuto di acidi grassi essenziali (EPA, DPA, DHA) che sono risultati significativamente inferiori. Le ridotte quantità di EPA e DPA nei petti anomali possono essere correlate alla minore attività degli enzimi Δ5 e Δ6 desaturasi, probabilmente in ragione della complessiva riduzione nell’espressione dei geni che codificano tali enzimi. Da sottolineare infine che le carni anomale hanno presentato livelli inferiori di emepigmenti. Per quanto attiene alle caratteristiche tecnologiche, è stata osservato un maggiore livello di ossidazione delle proteine in relazione alla presenza dell’anomalia wooden breast. Al contrario, l’ossidazione dei lipidi non ha mostrato differenze sostanziali fra i campioni anomali e quelli normali. Nel complesso, pertanto questo lavoro di tesi ha ulteriormente evidenziato i rischi connessi all’utilizzo delle carni anomale, e soprattutto di quelle affette da wooden breast, nella preparazione di prodotti trasformati in relazione al peggioramento delle rese di lavorazione dovuto alla minore concentrazione e qualità delle proteine. La componente lipidica sembra invece essere meno compromessa dalla presenza di tali anomalie.

Lo scopo di questo elaborato di tesi è stato quello di condurre uno studio preliminare volto ad indagare le principali caratteristiche qualitative delle carni bovine in funzione dell’origine e dell’età degli animali. Nel corso della sperimentazione sono stati analizzati 24 campioni di carne bovina ottenuti da animali di origine francese ed italiana di età compresa fra 15 e 23 mesi, acquistati presso la grande distribuzione o acquisiti direttamente dalle aziende produttrici. Da ciascun campione sono state ricavate delle porzioni di 6 mm di spessore impiegate, a loro volta, per la preparazione di sotto-campioni da sottoporre a determinazione di pH, colore, Expressible Moisture (EM %) e sforzo di taglio. Nell’ambito di ciascun parametro, l’insieme dei dati è stato analizzato mediante analisi statistica di tipo descrittivo. Successivamente, calcolata la matrice delle correlazioni fra i parametri oggetto di studio i dati sono stati elaborati mediante analisi multivariata con il metodo delle componenti principali (Principal Component Analysis, PCA) allo scopo di verificare se fosse possibile discriminare la qualità della carne in funzione dell’origine e dell’età degli animali. Quanto emerso evidenzia come non sia possibile discriminare la qualità dei campioni di carne bovina, sia acquistati al dettaglio presso la grande distribuzione che acquisiti direttamente dalle aziende produttrici, sulla base delle informazioni riportate in etichetta circa l’età e l’origine degli animali. Ciò può trovare spiegazione nella molteplicità di fattori intrinseci (specie, razza o tipo genetico, genere, età e peso degli animali alla macellazione) ed estrinseci (fasi pre- e post-macellazione) in grado di svolgere un ruolo rilevante nel determinare la qualità della carne. Pertanto, future ricerche dovranno essere intraprese per individuare quali parametri possano essere considerati più idonei a valorizzare la qualità tecnologica e sensoriale delle carni bovine.

Background A key aim of England's National Strategy for Sexual Health is to extend high-quality sexual health services in primary care. Objectives To explore the expectations and experiences of men and women who initially presented at their general practice with a suspected sexually transmitted infection in order to identify areas where change could improve service delivery. Methods Semi-structured interviews were carried out in six general practices and two genitourinary medicine (GUM) clinics in Brent primary care trust (London) and Bristol (southwest England). Patients within general practice, and GUM patients who had initially attended general practice were eligible to participate. Interview transcripts were analysed using thematic analysis. Results 49 patients (29 women, 20 men) were interviewed. Patients approaching their GP practice typically expected written referral or in-house care, but this expectation was often not met. Absence of formal referral, lack of information and perceived avoidance of sexual health matters by practitioners were commonly cited as reasons for disappointment. However, a dedicated service within general practice met expectations well. Conclusion Purchasers and providers of all general practice services should ensure that any patient consulting in primary care with a suspected sexually transmitted infection can either receive appropriate care there, or a formal and supported referral to a specialised GUM clinic or primary care service.
