White mice were used to study the infectivity of the eggs of Lagochilascaris minor Leiper, 1909 after incubation in liquid media, with or without preservative substances. Potassium bichromate (K2Cr2O7) at 1% restrict hatching, while 1% formalin gave a greater larval yield. Incubation of eggs in distilled water, in Roux or Falcon flasks gave a good yield, whether the eggs were obtained from human feces or from experimentally infected cats. Treatment of eggs with Sodium hypochlorite (NaOCl) at 5.25% for 2 min prior to inoculation, produced a notable increment of the larval yield in the infections.

At least eighteen species of triatominae have been found in the Brazilian Amazon, nine of them naturally infected with Trypanosoma cruzi or "cruzi-like" trypanosomes and associated with numerous wild reservoirs. Despite the small number of human cases of Chagas' disease described to date in the Brazilian Amazon the risk that the disease will become endemic in this area is increasing for the following reasons: a) uncontrolled deforestation and colonization altering the ecological balance between reservoir hosts and wild vectors; b) the adaptation of reservoir hosts of T.cruzi and wild vectors to peripheral and intradomiciliary areas, as the sole feeding alternative; c) migration of infected human population from endemic areas, accompanied by domestic reservoir hosts (dogs and cats) or accidentally carrying in their baggage vectors already adapted to the domestic habitat. In short, risks that Chagas' disease will become endemic to the Amazon appear to be linked to the transposition of the wild cycle to the domestic cycle in that area or to transfer of the domestic cycle from endemic areas to the Amazon.

A set of radiation measurements were carried out in several public and private institutions. These were selected with basis on the people affluence and passage to these sites. These measurements were registration formed either indoor, outdoor or underground and were compiled in three Case Studies. Radiation doses measurements were also made, surface and underground locations, and compiled in other two Case Studies. There were sampled, at the same time, humidity, temperature, atmospheric pressure and relevant construction materials at sampling locations. They were collected and registration formed to analyse if there is any relation or contribution for the measured value in each specific place. Geostatistical models were used to elaborate maps of the results both for radiation values and for doses. Preliminary relations were established among the measured parameters.

Num mundo onde cerca de 1% da população necessita de utilizar uma cadeira de rodas para a sua deslocação, a acessibilidade a este tipo de veículo no exterior ainda encontra-se num estado bastante precário. Falta de rampas nos passeios, buracos, caminhos em declive e determinados obstáculos como carros estacionados por cima do passeio, são alguns de muitos exemplos dos desafios que uma pessoa em cadeira de rodas enfrenta quando tenta deslocar-se no exterior, o que faz com que a autonomia e a segurança na condução de uma cadeira de rodas seja bastante reduzida, sendo por muitas vezes necessário recorrer de forma constante à ajuda de terceiros, que, por consequência, reduz a sua, já limitada, liberdade de locomoção. É com base nestes problemas que o projeto RevoChair surgiu. Este projeto consiste no desenvolvimento de um sistema para cadeira de rodas elétrica que permita ao utilizador movimentar-se no exterior automaticamente, sempre que possível, sem ter de se preocupar com os obstáculos a sua volta. Esta tese incide-se concretamente sobre três dos problemas do desenvolvimento deste projeto, a definição das componentes necessárias para a implementação do sistema, a forma como o utilizador interage com o sistema e a forma como os componentes comunicam entre si, tendo como foco a unidade de interação e a unidade de controlo responsável por gerir a comunicação entre os diversos componentes do sistema.

Plague caused by Yersinia pestis, has persisted in Brazil in several natural foci spread throughout rural areas in the States of Ceara, Paraiba, Pernambuco, Piaui, Rio Grande do Norte, Alagoas, Bahia, Minas Gerais and Rio de Janeiro. Nationwide surveillance of plague in Brazil based on serological testing started in 1983. We now present an update report of the examinations carried out in our laboratory from 1983 to 1992. The passive hemagglutination test for antibodies against fraction 1A antigen of Y. pestis and the passive hemagglutination inhibition control were employed for testing a total of 220,769 sera. Samples analyzed included 2,856 sera from clinically diagnosed plague cases or suspects, 49,848 sera from rodents of 24 species and 2 species of small wild carnivores (marsupials), 122,890 sera from dogs, and 45,175 sera from cats. Specific antibodies were found in 92 (3.22%) human sera; 143 (0.29%) sera from rodents of 8 species and from the two species of marsupials, 1,105 (0.90%) sera from dogs and 290 (0.64%) sera from cats. The presence of significant levels of specific anti-F1A antibodies among rodents and wild or domestic carnivores (dogs and cats) indicates that all the Brazilian plague foci remain active in spite of the absence of human cases in some of them.

Visceral larva migrans (VLM) is a clinical syndrome caused by infection of man by Toxocara spp, the common roundworm of dogs and cats. Tissue migration of larval stages causes illness specially in children. Because larvae are difficult to detect in tissues, diagnosis is mostly based on serology. After the introduction of the enzyme-linked immunosorbent assay (ELISA) using the larval excretory-secretory antigen of T. canis (TES), the diagnosis specificity was greatly improved although cross-reactivity with other helminths are still being reported. In Brazil, diagnosis is routinely made after absorption of serum samples with Ascaris suum antigens, a nematode antigenicaly related with Ascaris lumbricoides which is a common intestinal nematode of children. In order to identify T. canis antigens that cross react to A. suum antigens we analyzed TES antigen by SDS-PAGE and Western blotting techniques. When we used serum samples from patients suspected of VLM and positive result by ELISA as well as a reference serum sample numerous bands were seen (molecular weight of 210-200 kDa, 116-97 kDa, 55-50 kDa and 35-29 kDa). Among these there is at least one band with molecular weight around 55-66 kDa that seem to be responsible for the cross-reactivity between T. canis e A. suum once it disappears when previous absorption of serum samples with A. suum antigens is performed

Actualmente, os sistemas de localização são uma área em forte expansão sendo que para espaços exteriores existe uma grande variedade de sistemas de localização enquanto que para espaços interiores as soluções são mais escassas. Este trabalho apresenta o estudo e implementação de um sistema de localização indoor baseado no protocolo ZigBee, utilizando a informação da intensidade de sinal recebido (RSSI - Received Signal Strength Indication). Para a realização deste projecto foi necessário iniciar uma pesquisa mais pormenorizada do protocolo ZigBee. O dispositivo móvel a ser localizado é o módulo XBee Serie 2 que se baseia no mesmo protocolo. Posto isto, foi necessário efectuar um estudo sobre sistemas de localização existentes e analisar as técnicas de localização utilizadas para ambientes interiores. Desta forma utiliza-se neste projecto uma técnica que consiste na análise de fingerprinting, onde é criado um mapa com os valores RSSI para diferentes coordenadas do espaço físico. As intensidades de sinal recebido são relativas a dispositivos XBee instalados em pontos fixos de referência. Para calcular a localização do dispositivo móvel é utilizado o algoritmo K-NN (K- Nearest Neighbors) que permite estimar a posição aproximada do dispositivo móvel. Por último é descrito todo o desenvolvimento do projecto assim como a apresentação e discussão de resultados.

Toxocariasis is caused by infection of man by Toxocara canis and Toxocara cati larvae, the common roundworm of dogs and cats. Because larvae are difficult to detect in tissues, diagnosis is mostly based on serology. Non specific reactions are observed mainly due to cross-reactivity with Ascaris sp antigens. This investigation aimed at developing and evaluating an indirect antibody competition ELISA (IACE) employing a specific rabbit IgG anti-Toxocara canis excretory-secretory antigens as the competition antibody, in order to improve indirect ELISA specificity performed for toxocariasis diagnosis. For that, the rabbit IgG was previously absorbed by Ascaris suum adult antigens. Sensitivity and specificity of IACE were first evaluated in 28 serum samples of mice experimentally infected with T. canis embryonated eggs. Adopting cut-off value established in this population before infection, sensitivity and specificity were 100% after 20 days post-inoculation. For human population IACE was evaluated using sera from 440 patients with clinical signs of toxocariasis and the cut-off value was established with 60 serum samples from apparently healthy individuals. Using as reference test the indirect ELISA performed by Adolfo Lutz Institute, sensitivity was 60.2%, specificity was 98% and concordance was 77.3%. Repeatability of IACE was evaluated by the inter-reactions variation coefficient (2.4%).

A serological survey of Toxoplasma gondii infection in population groups in Fortaleza, Brazil, was conducted, in order to identify seroprevalence rates according to age and risk factors associated with acquired infection. A Toxoplasma IgG-antibody enzyme immunoassay (Sanofi Pasteur Diagnostics, Marnes la Coquette, France) was employed to assess the immunity. Public day-care centers and schools were randomly selected, while three large antenatal clinics and maternity units were choosen by its importance. Population groups and results of 997 blood tests were: 227 children (mean age 3.8 years), 22.8% seropositives; 584 students (mean 11.4 years), 58.4%, and pregnant and postpartum women (mean 24 years), 71.5% seropositives (p < 0.001). Of 256 participants reporting close contact with cats, 59.8% were seropositive, in contrast with 51% seropositives without contact (relative risk 1.17; 95% confidence interval 1.04-1.33; p = 0.01). Having three or more siblings at home was related to higher seroprevalence in children and students (relative risk 1.39; 95% confidence interval 1.21-1.60; p < 0.01). In conclusion, toxoplasmosis seroprevalence showed a rapid increase during the first ten years of life, in association with close contact with cats and larger households, probably related to inappropriate hygiene and child-care practices.

No âmbito do desenvolvimento da dissertação do Mestrado de Engenharia Eletrotécnica – Sistemas Elétricos de Energia, surgiu a oportunidade de participar no desenvolvimento de um projeto integrante numa subestação de energia. O presente caso de estudo tem em vista a resolução de condicionantes na conceção desta subestação, tais como cumprir requisitos estéticos projetando todo o tipo de equipamentos dentro de edifícios, a ventilação das salas de transformação assim como insonorização de todo o ruído produzido de forma a cumprir os limites legais e não perturbar a vizinhança. A presente subestação de energia está situada numa zona urbana da cidade de Leiria, localidade de Parceiros, dando origem ao nome Subestação de Energia de Parceiros. Esta subestação pertence ao cliente Energias de Portugal, conhecida como EDP, e visa o melhoramento da distribuição do serviço eléctrico. As tradicionais subestações de energia, com aparência bastante desenquadrada dos meios urbanos, representam um entrave ao nível da aproximação destes centros de produção energética às zonas urbanas, comprometendo a melhoria da rede elétrica. Desta forma, foi implementada a tecnologia Gas-Insolated Switchgear e permite o enquadramento destes centros energéticos em zonas urbanas, constituindo mais um edifício urbano na zona onde se insere. Esta substitui os convencionais barramentos existentes nos Parques Exteriores de Aparelhagem das subestações e apresenta-se com dimensões muito reduzidas quando comparadas com as estruturas instaladas nestes parques. Dado que esta tecnologia é desenvolvida no sentido de permitir a construção destes centros energéticos em zonas urbanas, podem ser alojadas dentro de edifícios produzindo assim vantagens ao nível estético, não perturbando a paisagem. Dado que os principais equipamentos de funcionamento na subestação de Parceiros, nomeadamente o Transformador de Potência, se encontram instalados num edifício completamente fechado, foram construídas duas salas de ventilação, na parte superior deste edifício, cada uma adjacente a uma sala de transformação. O transformador instalado possui elevadas dimensões, pesando 53000 kg e contendo 11000 kg de óleo que em estado normal de funcionamento circula por quase todo o interior da máquina a elevadas temperaturas, provocando um aquecimento elevado no interior do edifício o que condiciona o bom funcionamento do transformador. Para ultrapassar esta condicionante foi realizado um estudo de um sistema de ventilação capaz de avaliar e controlar os valores térmicos existentes e proceder à circulação de ar, que será movimentado ou bloqueado, recorrendo a um sistema autónomo, mantendo a temperatura ideal nas salas de transformação. Este autómato é o cérebro de toda a cadeia lógica que, mediante as leituras efetuadas irá dar ordens de atuação aos diversos equipamentos. Apesar dos TP estarem protegidos dentro do edifício, estes produzem um maior ruído. A necessidade da existência do referido sistema de ventilação das divisões de funcionamento destas máquinas, implica a utilização de aparelhos que, apesar da sua evolução tecnológica os torna cada vez menos ruidosos, mas geram sempre alguma perturbação, o que pode representar um problema no cumprimento do regulamento geral do ruído.

An epizootic outbreak of rabies occurred in 1995 in Ribeirão Preto, SP, with 58 cases of animal rabies (54 dogs, 3 cats and 1 bat) confirmed by the Pasteur Institute of São Paulo, and one human death. The need to provide care to a large number of people for the application of equine rabies immune globulin (ERIG) prevented the execution of the skin sensitivity test (SST) and often also the execution of desensitization, procedures routinely used up to that time at the Emergency Unit of the University Hospital of the Faculty of Medicine of Ribeirão Preto, University of São Paulo (EU-UHFMRP-USP), a reference hospital for the application of heterologous sera. In view of our positive experience of several years with the abolition of SST and of the use of premedication before the application of antivenom sera, we used a similar schedule for ERIG application. Of the 1489 victims of animal bites, 1054 (71%) received ERIG; no patient was submitted to SST and all received intravenously anti-histamines (anti-H1 + anti-H2) and corticosteroids before the procedure. The patients were kept under observation for 60 to 180 minutes and no adverse reaction was observed. On the basis of these results, since December 1995 ERIG application has been decentralized in Ribeirão Preto and has become the responsibility of the Emergency Unit of the University Hospital and the Central Basic Health Unit, where the same routine is used. Since then, 4216 patients have received ERIG (1818 at the Basic Health Unit and 2398 at the EU-UHFMRP), with no problems. The ideal would be the routine use of human rabies immune globulin (HRIG) in public health programs, but this is problematic, because of their high cost. However, while this does not occur, the use of SST is no longer justified at the time of application of ERIG, in view of the clinical evidence of low predictive value and low sensitivity of SST involving the application of heterologous sera. It is very important to point out that a negative SST result may lead the health team to a feeling of false safety that no adverse reaction will occur, but this is not true for the anaphylactoid reactions. The decision to use premedication, which is based on knowledge about anaphylaxis and on the pharmacology of the medication used, is left to the judgment of health professionals, who should always be prepared for eventual untoward events.

The visceral larva migrans (VLM) is a syndrome observed in human infection with helminth larval eggs such as the Toxocara spp. that usually infects dogs and cats. Among the risk factors involved in the occurrence of VLM, particularly important is the size of these animal populations. Sorocaba is a city with a dog population twice as large as that recommended by the World Health Organization. This fact has led to a survey of the presence of Toxocara spp. eggs in public square soils of this city. Thirty squares were selected, fifteen located in the outskirts of the city and fifteen downtown. Soil samples were collected from five distinct sites in the same area. The material was homogenized and drained and 100 g was mixed with a saturated solution of magnesium sulfate and 5% potassium iodine. The floating material was analyzed under the light microscope. Toxocara spp. eggs were found in 16 squares, nine of which were located in the outskirts of the city and seven downtown. It was concluded that Sorocaba squares present a high rate of contamination with Toxocara spp. eggs. The squares in the outskirts of the city presented a higher occurrence of these eggs in comparison with those downtown, although the difference was not statistically significant (p > 0.05).

Animal and human rabies samples isolated between 1989 and 2000 were typified by means of a monoclonal antibody panel against the viral nucleoprotein. The panel had been previously established to study the molecular epidemiology of rabies virus in the Americas. Samples were isolated in the Diagnostic Laboratory of the Pasteur Institute and in other rabies diagnostic centers in Brazil. In addition to the fixed virus samples CVS-31/96-IP, preserved in mouse brain, and PV-BHK/97, preserved in cell culture, a total of 330 rabies virus samples were isolated from dogs, cats, cattle, horses, bats, sheep, goat, swine, foxes, marmosets, coati and humans. Six antigenic variants that were compatible with the pre-established monoclonal antibodies panel were defined: numbers 2 (dog), 3 (Desmodus rotundus), 4 (Tadarida brasiliensis), 5 (vampire bat from Venezuela), 6 (Lasiurus cinereus) and Lab (reacted to all used antibodies). Six unknown profiles, not compatible with the panel, were also found. Samples isolated from insectivore bats showed the greatest variability and the most commonly isolated variant was variant-3 (Desmodus rotundus). These findings may be related to the existence of multiple independent transmission cycles, involving different bat species.

Cat-Scratch Disease (CSD) is a benign lymphadenitis that may progress to severe or recurrent forms, and it is occasionally associated with morbidity. Between January of 1998 and March of 1999, forty-three suspected CSD patients were assessed in the Hospital Cayetano Heredia and the Instituto de Salud del Niño, in Lima, Peru. Twelve patients had a confirmed diagnosis, 8 of whom were women, and the mean age was 10 years old. The majority (53%) of the cases were encountered in the summer. All patients reported having had contact with cats. Fever, malaise, lymphadenopathy and skin lesions were the most frequent clinical features. Twelve patients had indirect immunofluorescence antibody test titers of between 1/50 and 1/800 for Bartonella henselae and Bartonella clarridgeiae. Two lymph node biopsies were histologically compatible with CSD. No positive blood cultures could be obtained. This is the first Peruvian prospective study able to identify B. henselae and B. clarridgeiae in pediatric patients.
