Em geral, o efeito magnetocalórico (EMC) é caracterizado pela variação adiabática da temperatura (ΔTad) e a variação isotérmica da entropia (ΔST) sob variações do campo magnético. Devido as aplicações tecnológicas do EMC na refrigeração magnética, que não apresentam efeitos perigosos para o meio ambiente e tem o potencial para reduzir o consumo de energia, os estudos sobre o EMC tem crescido ao longo dos anos . Neste trabalho, estudamos as propriedades magnéticas e magnetocalóricos da série Gd (1-Y) Pr (Y) Ni2 com Y = 0; 0,25; 0,5; 0,75 e 1 A série dos compostos RNi2 compostos cristalizam na fase de Laves cúbico C15, o que torna o Campo Elétrico Cristalino cúbico um quadro adequado para descrever a anisotropia magnética sobre estes compostos . Além do modelo hamiltoniano inclui contribuições do efeito Zeeman e as interações de troca indireta entre Gd-Gd, Gd-Pr e íons Pr-Pr. Vale a pena notar que o GdNi2 apresenta um arranjo ferromagnético com temperatura de transição de cerca de 78 K e o composto PrNi2 é paramagnético. Os potenciais magnetocalóricos foram calculados e comparados com os dados experimentais. Além disso, investigamos a influência da direção do campo magnético sobre as quantidades magnéticas e no EMC investigada.

Choque séptico é caracterizado por desequilíbrio entre o transporte e o consumo de oxigênio, podendo acarretar hipóxia tecidual. A disfunção microcirculatória, característica cardinal da fisiopatologia do choque séptico, causa má distribuição de fluxo sanguíneo microvascular e, consequentemente, shunt de oxigênio, disóxia tissular e, teoricamente, diminuição no consumo de oxigênio (VO2) pela célula. No presente estudo, foi investigada a associação entre alterações microcirculatórias causadas pela sepse e o consumo de oxigênio em pacientes pediátricos. Dezessete crianças com choque séptico ressuscitadas foram estudadas em quatro momentos durante a internação na unidade de terapia intensiva (dentro de 24, 48 e 72 horas após a admissão ou diagnóstico de choque e após a resolução deste, antes da extubação traqueal). A microcirculação sublingual foi avaliada utilizando o método de imagem Sidestream dark field (SDF) e o VO2 foi calculado através da calorimetria indireta. Outras variáveis hemodinâmicas, como transporte de oxigênio, índice cardíaco, pressão arterial invasiva, lactato arterial e saturação venosa central, foram coletadas. Embora as variáveis hemodinâmicas tenham se mantido em níveis satisfatórios, graves alterações na microcirculação foram visualizadas, especialmente na densidade de vasos pequenos perfundidos (DVPP), na proporção de vasos pequenos perfundidos (PVPP) e no índice de fluxo microvascular (MFI). Foram encontradas assosciações significativas entre o VO2 e os parâmetros da microcirculação: dVO2 e dDVPP (β coefficient= 6,875; p<0,001), dVO2 e dPVPP (β coefficient=92,246; p<0,001) e dVO2 e dMFI (β coefficient=21,213; p<0,001). Não foram encontradas correlações entre as alterações microcirculatórias e as outras variáveis. Em conclusão, este estudo mostrou que pacientes pediátricos com choque séptico apresentaram grave disfunção microvascular e que o fluxo microcirculatório alterado estava associado ao VO2, podendo estar implicado na fisiopatologia da disóxia tecidual da sepse.

Atualmente grande parte dos resíduos descartados rotineiramente no Brasil é composta por material plástico como, por exemplo, o polietileno de alta densidade (PEAD), o qual é muito comum no lixo doméstico. Nesta Dissertação, embalagens de PEAD pós-consumo foram coletadas e moídas. Após processamento em extrusora, o material definido como PEAD reciclado (mistura de embalagens brancas, marfins e incolores) foi injetado e as propriedades mecânicas foram avaliadas e comparadas com uma amostra de PEAD comercial (HD7600U). A análise térmica através da calorimetria exploratória diferencial (DSC), em conjunto com a análise estatística dos resultados experimentais, também foi conduzida. Em relação à resistência à tração, a diferença entre as amostras de PEAD reciclado e PEAD comercial foram tão pouco expressivas que, dentro das condições experimentais adotadas, se pode afirmar que o material reciclado é equivalente ao comercial.Por sua vez, os modelos cinéticos aplicados na análise térmica revelaram quea amostra de PEAD comercial demonstra um processo de nucleação e crescimento dos cristais mais homogêneo e simples, embora a energia de ativação seja consideravelmente maior do que o das demais amostras de PEAD.

种子贮藏稳定性对于种质资源的长期保存具有重要意义,目前关于种子贮藏的最新理论为玻璃态理论,该理论认为种子的玻璃化有利于种子的长期贮藏。当种子处于玻璃态时,玻璃化物质的高度粘滞性降低了种子细胞内分子流动性,阻止了细胞质中分子的扩散,从而减少老化过程中细胞结构的损伤和化学组分的变化,延缓种子老化劣变反应速率,延长贮藏寿命。评价玻璃态的一个重要指标是玻璃化转变温度,当种子贮藏于玻璃化温度或以下10℃~30℃范围内时,种子具有最佳的贮藏稳定性。因此,检测种子的玻璃化转变温度对于种子的长期有效贮藏具有重要指导意义。 本研究将差示量热扫描技术(DSC)与电子顺磁共振波谱仪技术(EPR)应用于杜仲种子玻璃化转变温度方面的研究。在DSC方法中,选用4.4%~31.6%含水量范围的杜仲种胚分别进行了DSC图谱扫描。EPR方法选用3-羧基-2,2,5,5-四甲基吡咯烷-1-氧(3-carboxy-2,2,5,5-tetramethylpyrrolidine-1-oxyl,CP)和2,2,6,6-四甲基哌啶(4-hydroxy-2,2,6,6-tetramethyl-1-piperidinyloxy,TEMPO)作为探针标记杜仲种胚, 利用EPR技术测定不同含水量杜仲种胚的分子运动,通过对EPR图谱参数的分析计算,最终确定不同含水量杜仲种胚的玻璃化转变温度。 DSC实验结果显示,含水量为22.3%、28.0%、31.6%的杜仲种胚在0℃ 左右出现了一个水的熔融峰。该熔融峰的面积代表了自由水含量的多少,随着种胚含水量的降低该熔融峰面积减小。4.4%~31.6%含水量范围的杜仲种胚在-28℃左右还出现了一个熔融峰,推测此峰为杜仲种胚中某类物质熔融所形成的熔融峰。然而在此曲线上我们未观察到标志玻璃化转变的“台阶”出现。 CP-EPR实验的结果表明,利用EPR测定得到含水量为4.4%~11.6%的杜仲种胚在-110℃~20℃温度范围内,同一含水量的杜仲种胚随着温度的升高,分子运动速率加快;在同一温度条件下,高含水量的种胚比低含水量种胚的分子运动速率快。通过CP-EPR波谱两外缘峰最大距离(2Azz)的测定和数据统计分析,得到含水量为4.4%、5.7%、8.6%、10.3%、11.6%杜仲种胚的玻璃化转变温度分别约为44℃、25℃、4℃、-31℃、-43℃。可以把测定的杜仲种胚的这几个含水量的玻璃化转变温度与杜仲种子贮藏相结合,用于指导杜仲种子的贮藏。 TEMPO-EPR实验测定分析得到含水量为2.1%、3.4%、4.8%、8.3%、11.2% 的杜仲种胚的玻璃化转变温度分别为-21℃、-18℃、-24℃、-20℃、-27℃,玻璃化转变温度随含水量升高其变化的规律不明显,这与CP-EPR实验测得的结果有着较明显的差别。通过分析,认为对于脂质含量较高的杜仲种胚,随着含水量的降低,作为标记化合物的TEMPO随着脱水进入脂相,从而不能真实反映出不同含水量种胚的分子运动情况。与TEMPO标记相比,CP标记可能能够更真实地反映不同含水量杜仲种胚细胞质分子运动的情况,根据其分子运动情况得到的玻璃化转变温度更准确。

New chiral compounds 3R-methylcyclohexanone derivatives were synthesized. These compounds were revealed to exhibit the mesomorphic behavior within rather wide temperature ranges. Types of formed mesophases and phase transition temperatures were determined by polarizing microscopy, differential scanning calorimetry and small angle scattering of X-ray. Mesomorphic properties of the new chiral compounds were compared with those for the chiral 2-arylidene derivatives of 3R,6R-3-methyl-6-isopropylcyclohexanone (d-isomenthone) studied earlier. Distinctions between these two types of compounds in an ability to form mesophases and also in twisting properties as chiral dopants in induced cholesteric mesophases are considered.

Compact Fluorescent Lamps (CFL) incorporating electronic ballasts are widely used in lighting. In many cases the ability to dim the lamp is a requirement Dimming can be achieved by varying the voltage supplied to the inverter or by changing the switching frequency of the inverter. The effect of dimming by both approaches on the power losses in the inverter is studied in this work. The lamp and associated inverter has been modeled in PSPICE, using a behavioral model for the CFL. Predicted losses are in good agreement with experimental data obtained from calorimetry. The model was then used to determine the distribution of losses within the inverter, enabling a comparison of the effects of the two dimming methods to be made. © 2006 IEEE.

(1R,4R)-2-(4-Hydroxybenzylidene)- and (1R,4R)-2-(4′-hydroxybiphenyl- 4-yl)methylene-p-menthan-3-ones were synthesized by condensation of (-)-menthone with O-tetrahydropyran-2-yl derivatives of 4-hydroxybenzaldehyde and 4′-hydroxy-4-formylbiphenyl, respectively, in a DMSO - base medium followed by the removal of the protective group. The reactions of these hydroxy derivatives with 4-alkylbenzoic, 4-alkyloxybenzoic, trans-4-alkylcyclohexane-4- carboxylic, and 4′-alkylbiphenyl-4-carboxylic acids afforded three series of new chiral esters. Compounds containing the arylidene moiety with three benzene rings were found to exhibit liquid-crystalline properties. The characteristic features of these compounds are discussed based on the results of studies by polarizing microscopy, differential scanning calorimetry, and small-angle X-ray scattering. It was found that the mesomorphic compounds under study can form a smectic A mesophase, twist grain boundary mesophases (TGBA), and blue phases in a wide temperature range. Upon dissolution of certain of chiral compounds in 4′-cyano-4-pentylbiphenyl, a rather high twisting power and the thermal stabilizing effect on mesophases were observed.

The quasi-static and dynamic behaviour of Linear Low Density Polyethylene (LLDPE) and two LLDPE nanocomposites were studied. Nanocomposites consisting of LLDPE filled with 1% carbon black and 0.5% nanoclay fillers, by weight, were considered. Under quasi-static tensile loading, an improvement in the energy absorbing capability was achieved by adding 1% carbon black fillers. However, during quasi-static puncture and dynamic impact loading, the advantage provided by the fillers was lost. Thermal softening due to adiabatic heating under high strain rate deformation and difference s in the state of stress are considered as reasons for this reduction. © 2011 Published by Elsevier Ltd.

Hemoglobin (Hb) variability is a commonly used index of phylogenetic differentiation and molecular adaptation in fish. In the current study, the structural and functional characteristics of Hbs from two Sturgeon species of the Southern Caspian Sea Basin were investigated. After extraction and separation of hemoglobin from whole blood , the polyacrylamide gel electrophoresis (SDSPAGE), native-PAGE and isoelectric focusing (IEF) were used to confirm Hb variability in these fishes. Ion-exchange on CM-cellulose chromatography was used for purification of the dominant Hbs from these fishes. The accuracy of the methods was confirmed by IEF and SDS-PAGE. Spectral studies using fluorescence spectrophotometery, circular dichroism spectropolarimetry (CD) analysis and UV–vis spectrophotometery. Oxygen affinities of these Hbs were compared using Hb-oxygen dissociation curves. Also, the dominant Hbs from these blood fishes were utilized for further experiments. The behavior of Hbs during the denaturation process by n-dodecyl trimethylammonium bromide (DTAB) is investigated by UV–vis spectrophotometer and circular dichroism spectropolarimetry. The thermal denaturation properties of the Hbs wereinvestigated by differential scanning calorimetry (DSC) and Hbs aggregation performed chemically in the presence of dithiotreitol (DTT) by UV–vis spectrophotometer and chemometric study. The results demonstrate a significant relationship between stability of fish hemoglobins and the ability of fish for entering to deeper depths. The UV–Vis absorption spectra identified species of hemoglobin and showed the concentration of oxyHb and metHb decreases and deoxyHb increases upon interaction with DTAB. Besides the UV–vis spectrophotometry, the interaction of DTAB with hemoglobins has been studied using circular dichroism spectropolarimetry analysis. This experiment was utilized to measure the unfolding mechanism and compared alpha-helix secondary structure under different conditions for Hbs. The results reveal that the Acipenser stellatus Hb in comparison with Acipenser persicus Hb has more stability and more structural compactness. Besides, the results confirm the hypothesis that there is a meaningful relation between average habitat depth, partial oxygen pressure, oxygen affinity, structural compactness of Hb, and its stability.

We apply adjoint-based sensitivity analysis to a time-delayed thermo-acoustic system: a Rijke tube containing a hot wire. We calculate how the growth rate and frequency of small oscillations about a base state are affected either by a generic passive control element in the system (the structural sensitivity analysis) or by a generic change to its base state (the base-state sensitivity analysis). We illustrate the structural sensitivity by calculating the effect of a second hot wire with a small heat-release parameter. In a single calculation, this shows how the second hot wire changes the growth rate and frequency of the small oscillations, as a function of its position in the tube. We then examine the components of the structural sensitivity in order to determine the passive control mechanism that has the strongest influence on the growth rate. We find that a force applied to the acoustic momentum equation in the opposite direction to the instantaneous velocity is the most stabilizing feedback mechanism. We also find that its effect is maximized when it is placed at the downstream end of the tube. This feedback mechanism could be supplied, for example, by an adiabatic mesh. We illustrate the base-state sensitivity by calculating the effects of small variations in the damping factor, the heat-release time-delay coefficient, the heat-release parameter, and the hot-wire location. The successful application of sensitivity analysis to thermo-acoustics opens up new possibilities for the passive control of thermo-acoustic oscillations by providing gradient information that can be combined with constrained optimization algorithms in order to reduce linear growth rates. © Cambridge University Press 2013.

Nanobodies are single-domain fragments of camelid antibodies that are emerging as versatile tools in biotechnology. We describe here the interactions of a specific nanobody, NbSyn87, with the monomeric and fibrillar forms of α-synuclein (αSyn), a 140-residue protein whose aggregation is associated with Parkinson's disease. We have characterized these interactions using a range of biophysical techniques, including nuclear magnetic resonance and circular dichroism spectroscopy, isothermal titration calorimetry and quartz crystal microbalance measurements. In addition, we have compared the results with those that we have reported previously for a different nanobody, NbSyn2, also raised against monomeric αSyn. This comparison indicates that NbSyn87 and NbSyn2 bind with nanomolar affinity to distinctive epitopes within the C-terminal domain of soluble αSyn, comprising approximately amino acids 118-131 and 137-140, respectively. The calorimetric and quartz crystal microbalance data indicate that the epitopes of both nanobodies are still accessible when αSyn converts into its fibrillar structure. The apparent affinities and other thermodynamic parameters defining the binding between the nanobody and the fibrils, however, vary significantly with the length of time that the process of fibril formation has been allowed to progress and with the conditions under which formation occurs, indicating that the environment of the C-terminal domain of αSyn changes as fibril assembly takes place. These results demonstrate that nanobodies are able to target forms of potentially pathogenic aggregates that differ from each other in relatively minor details of their structure, such as those associated with fibril maturation.

A novel method of measuring cylinder gas temperature in an internal combustion engine cylinder is introduced. The physical basis for the technique is that the flow rate through an orifice is a function of the temperature of the gas flowing through the orifice. Using a pressure transducer in the cylinder, and another in a chamber connected to the cylinder via an orifice, it is shown how the cylinder temperature can be determined with useful sensitivity. In this paper the governing equations are derived, which show that the heat transfer characteristics of the chamber are critical to the performance of the system, and that isothermal or adiabatic conditions give the optimum performance. For a typical internal combustion engine, it is found that the pre-compression cylinder temperature is related to the chamber pressure late in the compression process with sensitivity of the order of 0.005 bar/K.
