The goal of the RAP-WAM AND-parallel Prolog abstract architecture is to provide inference speeds significantly beyond those of sequential systems, while supporting Prolog semantics and preserving sequential performance and storage efficiency. This paper presents simulation results supporting these claims with special emphasis on memory performance on a two-level sharedmemory multiprocessor organization. Several solutions to the cache coherency problem are analyzed. It is shown that RAP-WAM offers good locality and storage efficiency and that it can effectively take advantage of broadcast caches. It is argued that speeds in excess of 2 ML IPS on real applications exhibiting medium parallelism can be attained with current technology.

The signaling pathway initiated by factor Xa on vascular endothelial cells was investigated. Factor Xa stimulated a 5- to 10-fold increased release of nitric oxide (NO) in a dose-dependent reaction (0.1–2.5 μg/ml) unaffected by the thrombin inhibitor hirudin but abolished by active site inhibitors, tick anticoagulant peptide, or Glu-Gly-Arg-chloromethyl ketone. In contrast, the homologous clotting protease factor IXa or another endothelial cell ligand, fibrinogen, was ineffective. A factor Xa inter-epidermal growth factor synthetic peptide L83FTRKL88(G) blocking ligand binding to effector cell protease receptor-1 inhibited NO release by factor Xa in a dose-dependent manner, whereas a control scrambled peptide KFTGRLL was ineffective. Catalytically active factor Xa induced hypotension in rats and vasorelaxation in the isolated rat mesentery, which was blocked by the NO synthase inhibitor l-NG-nitroarginine methyl ester (l-NAME) but not by d-NAME. Factor Xa/NO signaling also produced a dose-dependent endothelial cell release of interleukin 6 (range 0.55–3.1 ng/ml) in a reaction inhibited by l-NAME and by the inter-epidermal growth factor peptide Leu83–Leu88 but unaffected by hirudin. Maximal induction of interleukin 6 mRNA required a brief, 30-min stimulation with factor Xa, unaffected by subsequent addition of tissue factor pathway inhibitor. These data suggest that factor Xa-induced NO release modulates endothelial cell-dependent vasorelaxation and cytokine gene expression. This pathway requiring factor Xa binding to effector cell protease receptor-1 and a secondary step of ligand-dependent proteolysis may preserve an anti-thrombotic phenotype of endothelium but also trigger acute phase responses during activation of coagulation in vivo.

Passive and active immunization against outer surface protein A (OspA) has been successful in protecting laboratory animals against subsequent infection with Borrelia burgdorferi. Antibodies (Abs) to OspA convey full protection, but only when they are present at the time of infection. Abs inactivate spirochetes within the tick and block their transmission to mammals, but do not affect established infection because of the loss of OspA in the vertebrate host. Our initial finding that the presence of high serum titers of anti-OspC Abs (5 to 10 μg/ml) correlates with spontaneous resolution of disease and infection in experimentally challenged immunocompetent mice suggested that therapeutic vaccination with OspC may be feasible. We now show that polyclonal and monospecific mouse immune sera to recombinant OspC, but not to OspA, of B. burgdorferi resolve chronic arthritis and carditis and clear disseminated spirochetes in experimentally infected C.B.-17 severe combined immunodeficient mice in a dose-dependent manner. This was verified by macroscopical and microscopical examination of affected tissues and recultivation of spirochetes from ear biopsies. Complete resolution of disease and infection was achieved, independent of whether OspC-specific immune sera (10 μg OspC-specific Abs) were repeatedly given (4× in 3- to 4-day intervals) before the onset (day 10 postinfection) or at the time of fully established arthritis and carditis (days 19 or 60 postinfection). The results indicate that in mice spirochetes constitutively express OspC and are readily susceptible to protective OspC-specific Abs throughout the infection. Thus, an OspC-based vaccine appears to be a candidate for therapy of Lyme disease.

The phylogeny of 123 complete envelope gene sequences was reconstructed in order to understand the evolution of tick- and mosquito-borne flaviviruses. An analysis of phylogenetic tree structure reveals a continual and asymmetric branching process in the tick-borne flaviviruses, compared with an explosive radiation in the last 200 years in viruses transmitted by mosquitoes. The distinction between these two viral groups probably reflects differences in modes of dispersal, propagation, and changes in the size of host populations. The most serious implication of this work is that growing human populations are being exposed to an expanding range of increasingly diverse viral strains.

Human hookworm infection is a major cause of gastrointestinal blood loss and iron deficiency anemia, affecting up to one billion people in the developing world. These soil-transmitted helminths cause blood loss during attachment to the intestinal mucosa by lacerating capillaries and ingesting extravasated blood. We have isolated the major anticoagulant used by adult worms to facilitate feeding and exacerbate intestinal blood loss. This 8.7-kDa peptide, named the Ancylostoma caninum anticoagulant peptide (AcAP), was purified by using a combination of ion-exchange chromatography, gel-filtration chromatography, and reverse-phase HPLC. N-terminal sequencing of AcAP reveals no homology to any previously identified anticoagulant or protease inhibitor. Single-stage chromogenic assays reveal that AcAP is a highly potent and specific inhibitor of human coagulation, with an intrinsic K*i for the inhibition of free factor Xa of 323.5 pM. In plasma-based clotting time assays, AcAP was more effective at prolonging the prothrombin time than both recombinant hirudin and tick anticoagulant peptide. These data suggest that AcAP, a specific inhibitor of factor Xa, is one of the most potent naturally occurring anticoagulants described to date.

In colonial species, it is often assumed that locations in the center of the colony are of highest quality and provide highest breeding success. We tested this prediction, known as the "central-periphery model," in a King Penguin colony in the subantarctic Crozet Archipelago. Breeding activity and survival of 150 penguins, fitted with transponder tags, were monitored over an entire breeding season. Among these 150 birds, 50 bred on the slope at the upper periphery of the colony, where the rates of predation and parasitism by ticks were high. Fifty birds bred in the center of the colony, where rates of predation and tick parasitism were low, and 50 bred at the lower end of the colony, where the rate of tick parasitism was low but predation and flooding were important risks. We predicted that the center of the colony should provide the safest breeding place and consequently be characterized by the highest breeding success and be used by the highest-quality individuals. Yet we found that penguins breeding in the center of the colony had the same breeding success as those at both peripheral locations. In addition, penguins breeding on the upper slope had a higher survival rate than penguins breeding at the center or bottom of the slope and were likely of higher quality. Our study does not support the central-periphery model and emphasizes the complexity behind the relationships among breeding site, breeding success, and individual quality.

A major trough ('Belgica Trough') eroded by a palaeo-ice stream crosses the continental shelf of the southern Bellingshausen Sea (West Antarctica) and is associated with a trough mouth fan ('Belgica TMF') on the adjacent continental slope. Previous marine geophysical and geological studies investigated the bathymetry and geomorphology of Belgica Trough and Belgica TMF, erosional and depositional processes associated with bedform formation, and the temporal and spatial changes in clay mineral provenance of subglacial and glaciomarine sediments. Here, we present multi-proxy data from sediment cores recovered from the shelf and uppermost slope in the southern Bellingshausen Sea and reconstruct the ice-sheet history since the last glacial maximum (LGM) in this poorly studied area of West Antarctica. We combined new data (physical properties, sedimentary structures, geochemical and grain-size data) with published data (shear strength, clay mineral assemblages) to refine a previous facies classification for the sediments. The multi-proxy approach allowed us to distinguish four main facies types and to assign them to the following depositional settings: 1) subglacial, 2) proximal grounding-line, 3) distal sub-ice shelf/subsea ice, and 4) seasonal open-marine. In the seasonal open-marine facies we found evidence for episodic current-induced winnowing of near-seabed sediments on the middle to outer shelf and at the uppermost slope during the late Holocene. In addition, we obtained data on excess 210Pb activity at three core sites and 44 AMS 14C dates from the acid-insoluble fraction of organic matter (AIO) and calcareous (micro-)fossils, respectively, at 12 sites. These chronological data enabled us to reconstruct, for the first time, the timing of the last advance and retreat of the West Antarctic Ice Sheet (WAIS) and the Antarctic Peninsula Ice Sheet (APIS) in the southern Bellingshausen Sea. We used the down-core variability in sediment provenance inferred from clay mineral changes to identify the most reliable AIO 14C ages for ice-sheet retreat. The palaeo-ice stream advanced through Belgica Trough after ~36.0 corrected 14C ka before present (B.P.). It retreated from the outer shelf at ~25.5 ka B.P., the middle shelf at ~19.8 ka B.P., the inner shelf in Eltanin Bay at ~12.3 ka B.P., and the inner shelf in Ronne Entrance at ~6.3 ka B.P.. The retreat of the WAIS and APIS occurred slowly and stepwise, and may still be in progress. This dynamical ice-sheet behaviour has to be taken into account for the interpretation of recent and the prediction of future mass-balance changes in the study area. The glacial history of the southern Bellingshausen Sea is unique when compared to other regions in West Antarctica, but some open questions regarding its chronology need to be addressed by future work.
