A desnutrição proteica (DP) pode ocasionar alterações na matriz extracelular (MEC) de diferentes órgãos e tecidos, inclusive o hematopoético, com comprometimento funcional. Estudos do nosso laboratório demonstraram, em modelo murino de DP, aumento da expressão proteica de fibronectina (FN) no estroma medular ósseo in vivo, principalmente na região subendosteal (local de fixação da célula tronco progenitora hemopoética). Já in vitro, no estroma medular ósseo, observou-se tanto o aumento quanto a diminuição de FN e a presença de suas isoformas. Essas alterações de FN parecem estar envolvidas com a hipoplasia da medula óssea (MO) em camundongos desnutridos. As modificações quantitativas de FN podem ser devidas: (i) à ação das metaloproteinases de matriz (MMP) responsáveis pela degradação das proteínas da MEC; (ii) aos inibidores de metaloproteinases (TIMP) que regulam a degradação da MEC; (iii) às alterações transcricionais, reguladas pela via de AKT/mTOR, que controla os splicing alternativos na FN, resultando em isoformas dessa proteína; (iv) a processos pós-transcricionais modulados por LC3, que aumenta a tradução do RNAm de FN. Assim, o objetivo deste estudo foi elucidar os mecanismos que alteram o turnover de FN no estroma medular ósseo em modelo murino de DP. Utilizamos camundongos, C57BL/6J machos, adultos, separados em dois grupos: controle e desnutrido, alimentados, ad libitum, com ração contendo 12% e 2% de proteína, respectivamente. Após cinco semanas de indução à desnutrição os camundongos foram eutanasiados, e coletado o material biológico. Avaliamos: o estado nutricional, o hematológico, a histologia da MO femoral bem como a determinação imunohistoquímica da FN, MMP-2 e MMP-9, determinação da expressão de FN e suas isoformas em células totais da MO, o estabelecimento do estroma medular ósseo in vitro, por 28 e 35 dias de cultivo. A partir das culturas foram avaliadas a expressão de RNAm de FN e suas isoformas, MMP-2, MMP-9, TIMP-1, TIMP-2, AKT, mTOR e LC3α e β, quantificação de MMP-2, MMP-9, TIMP-1, TIMP-2,TNFα, TGFβ e IL-1β e determinação de LC3β e proteínas da via de AKT/mTOR. Não observamos alterações na expressão do RNAm de FN e suas isoformas ex vivo e in vitro, mas um aumento da deposição de FN na MO.Também não observamos modificações na imunolocalização de MMP-2 e MMP-9 na MO e na atividade dessas proteínas no sobrenadante de culturas de células estromais in vitro, mas houve aumento da expressão do RNAm de MMP-9 em 28 dias de cultivo. Não detectamos alterações na expressão de RNAm e na concentração de TIMP-1 e TIMP-2 no sobrenadante das culturas. Houve redução significativa de TNFα e TGFβ no sobrenadante das culturas de 28 dias. Observamos aumento da expressão do RNAm de mTOR em culturas de 28 dias e LC3α e LC3β em 35 dias de células estromais. Encontramos menor fosforilação de PI3K, AKT, PTEN, mTOR e mTOR total e aumento de LC3β em culturas de 28 dias, mas redução de LC3β em 35 dias. Em função dos dados inferimos que a DP conduz a alterações da FN que não estão relacionadas à ação de MMPs e TIMPs e sim a modificações de LC3β e da via de AKT/mTOR.

This dissertation uses a political ecology approach to examine the relationship between tourism development and groundwater in southwest Nicaragua. Tourism in Nicaragua is a booming industry bolstered by ‘unspoiled’ natural beauty, low crime rates, and government incentives. This growth has led to increased infrastructure, revenue, and employment opportunities for many local communities along the Pacific coast. Not surprisingly, it has also brought concomitant issues of deeper poverty, widening gaps between rich and poor, and competition over natural resources. Adequate provisions of freshwater are necessary to sustain the production and reproduction of tourism; however, it remains uncertain if groundwater supplies can keep pace with demand. The objective of this research is to assess water supply availability amidst tourism development in the Playa Gigante area. It addresses the questions: 1) are local groundwater supplies sufficient to sustain the demand for freshwater imposed by increased tourism development? and 2) is there a power relationship between tourism development and control over local freshwater that would prove inequitable to local populations? Integrating the findings of groundwater monitoring, geological mapping, and ethnographic and survey research from a representative stretch of Pacific coastline, this dissertation shows that diminishing recharge and increased groundwater consumption is creating conflict between stakeholders with various levels of knowledge, power, and access. Although national laws are structured to protect the environment and ensure equitable access to groundwater, the current scramble to secure water has powerful implications on social relations and power structures associated with tourism development. This dissertation concludes that marginalization due to environmental degradation is attributable to the nexus of a political promotion of tourism, poorly enforced state water policies, insufficient water research, and climate change. Greater technical attention to hydrological dynamics and collaboration amongst stakeholders are necessary for equitable access to groundwater, environmental sustainability, and profitability of tourism.

Attempts to address the ever increasing achievement gap among students have failed to explain how and why educational traditions and teaching practices perpetuate the devaluing of some and the overvaluing of others. This predicament, which plagues our educational system, has been of increased concern, given the growing racial diversity among college students and the saturation of White faculty in the academy. White faculty make up the majority, 79%, of all faculty in the academy. White faculty, whether consciously or unconsciously, are less likely to interrogate how race and racism both privilege them within the academy and influence their faculty behaviors. The result of this cyclical, highly cemented process suggests that there is a relationship between racial consciousness and White faculty members' ability to employ behaviors in their classroom that promote equitable educational outcomes for racially minoritized students. An investigation of the literature revealed that racial consciousness and the behaviors of White faculty in the classroom appeared to be inextricably linked. A conceptual framework, Racial Consciousness and Its Influence on the Behaviors of White Faculty in the Classroom was developed by the author and tested in this study. Constructivist grounded theory was used to explore the role White faculty believe they play in the dismantling of the white supremacy embedded in their classrooms through their faculty behaviors. A substantive theory subsequently emerged. Findings indicate that White faculty with a higher level of racial consciousness employ behaviors in their classroom reflective of a more expansive view of equality in their pursuit of social justice, which they consider synonymous with excellence in teaching. This research bears great significance to higher education research and practice, as it is the first of its kind to utilize critical legal scholar Kimberlé Crenshaw's (1988) restrictive and expansive views of equality framework to empirically measure and describe excellence in college teaching. Implications for faculty preparation and continued education are also discussed.

Hiring minority coaches is a serious concern in the National Football League (NFL). The NFL's hiring practices are inconsistent, specifically for minority candidates. The author investigates why NFL teams underutilize minority coaches with research from Central Florida University and the University of Pennsylvania. Research findings suggest that minority hires positively affect the NFL, yet the hiring process remains weak. Case study examples show the poor decision-making trends of NFL personnel, implying that although minority coaches' success is better than non-minorities, the negative perception of minorities, as aspiring head coaches and leaders, is a barrier. As a result, the NFL has a unique opportunity to improve its hiring process by aligning its approach to hiring within the guidelines of federal law.

A conditioning procedure is proposed allowing to install into the concrete specimens any selected value of water saturation degree with homogeneous moisture distribution. This is achieved within the least time and the minimum alteration of the concrete specimens. The protocol has the following steps: obtaining basic drying data at 50 °C (water absorption capacity and drying curves); unidirectional drying of the specimens at 50 °C until reaching the target saturation degree values; redistribution phase in closed containers at 50 °C (with measurement of the quasi-equilibrium relative humidities); storage into controlled environment chambers until and during mass transport tests, if necessary. A water transport model is used to derive transport parameters of the tested materials from the drying data, i.e., relative permeabilities and apparent water diffusion coefficients. The model also allows calculating moisture profiles during isothermal drying and redistribution phases, thus allowing optimization of the redistribution times for obtaining homogeneous moisture distributions.

Mercury intrusion porosimetry (MIP) has been widely used to evaluate the quality of concrete through the pore size distribution parameters. Two of these parameters are the critical pore diameter (Dcrit) and the percentage of the most interconnected net of pores compared to the total volume of pores. Some researchers consider Dcrit as the diameter obtained from the inflexion point of the cumulative mercury intrusion curve while others consider Dcrit as the diameter obtained from the point of abrupt variation in the same curve. This study aims to analyze two groups of concretes of varying w/c ratios, one cast with pozzolanic cement and another with high initial strength cement, in order to determine which of these diameters feature a better correlation with the quality parameters of the concretes. The concrete quality parameters used for the evaluations were (1) the w/c ratios and (2) chloride diffusion coefficients measured at approximately 90 days. MIP cumulative distributions of the same concretes were also measured at about 90 days, and Dcrit values were determined (1) from the point of abrupt variation and alternatively, (2) from the inflexion point of each of these plots. It was found that Dcrit values measured from the point of abrupt variation were useful indicators of the quality of the concrete, but the Dcrit values based on the inflexion points were not. Hence, it is recommended that Dcrit and the percentage of the most interconnected volume of pores should be obtained considering the point of abrupt variation of the cumulative curve of pore size distribution.

The annual Young Academics network of the Association of European School of Planning (AESOP YA) conference, entitled Differences and Connections, was held for the first time in a Southern Italian city, Palermo, Italy, during 23–26 March 2015. The call for papers attracted a wide range of authors within the field of planning and other related fields. Forty-five contributions by young academic scholars, representing 19 countries, were selected by the conference scientific committee to critically explore the themes of the conference.

El trabajo explora la emergencia de temas y la constitución de problemas públicos que integran la agenda de la seguridad en Argentina en la etapa democrática a partir de procesos vinculados a la problematización pública de muertes violentas constituidas como casos conmocionantes. Con estos fines, el análisis integra la reconstrucción en el registro de la acción pública de casos conmocionantes con una revisión de trabajos que desde las ciencias sociales abordan dimensiones de la muerte violenta y problemas vinculados a la agenda nacional de la seguridad. Como punto de llegada del análisis destacamos la capacidad que en la actualidad manifiesta la inseguridad para absorber e integrar dimensiones en tanto problema que se constituye como un guion cultural privilegiado para la comprensión de la violencia, que sintetiza sentidos sociales circulantes sobre el riesgo y que sirve de plataforma para la demanda política de amplios sectores de la ciudadanía.

The German-Russian project CARBOPERM - Carbon in Permafrost, origin, quality, quantity, and degradation and microbial turnover - is devoted to studying soil organic matter history, degradation and turnover in coastal lowlands of Northern Siberia. The multidisciplinary project combines research from various German and Russian institutions and runs from 2013 to 2016. The project aims assessing the recent and the ancient trace gas budget over tundra soils in northern Siberia. Studied field sites are placed in the permafrost of the Lena Delta and on Bol'shoy Lyakhovsky, the southernmost island of the New Siberian Archipelago in the eastern Laptev Sea. Field campaigns to Bol'shoy Lyakhovsky in 2014 (chapter 2) were motivated by research on palaeoenvironmental and palaeoclimate reconstruction, sediment dating, near surface geophysics and microbiological research. In particular the field campaigns focussed on: - coring Quaternary strata with a ages back to ~200.000 years ago as found along the southern coast; they allow tracing microbial communities and organic tracers (i.e. lipids and biomarkers, sedimentary DNA) in the deposits across two climatic cycles (chapter 3), - instrumenting a borehole with a thermistor chain for measuring permafrost temperatures (chapter 3), - sampling Quaternary strata for dating permafrost formation periods based on the optical stimulated luminescence (OSL) technique (chapter 4), - sampling soil and geologic formations for carbon content in order to highlight potential release of CO2 and methane based on incubation experiments (chapter 5), - profiling near surface permafrost using ground-penetrating radar and geoelectrics for defining the spatial depositional context, where the cores are located (chapters 6 + 7).

Trotz der steigenden Bedeutung von familienexternen Nachfolgeregelungen wie MBO und MBI bestehen hinsichtlich dieser Nachfolgeoptionen noch große Forschungslücken. Obwohl die Informationsökonomie einen viel versprechenden Ansatz darstellt, um ein fundierteres Verständnis zu erlangen, ist sie in diesem Kontext noch zu wenig angewandt worden. Dies wäre jedoch vor allem in Bezug auf einen MBI sinnvoll, da dort die deutlichsten Informationsasymmetrien zwischen Übergeber und Nachfolger auftreten können. In diesem Beitrag bedienen wir uns daher der Informationsökonomie und analysieren die verschiedenen Informationsasymmetrien bei einem MBI im Detail. Außerdem zeigen wir verschiedene Möglichkeiten auf, wie die entsprechenden Asymmetrien überwunden werden können. Damit leisten wir einen wertvollen Beitrag zu Wissenschaft und Praxis.
