This study analyzes an accident in which two maintenance workers suffered severe burns while replacing a circuit breaker panel in a steel mill, following model of analysis and prevention of accidents (MAPA) developed with the objective of enlarging the perimeter of interventions and contributing to deconstruction of blame attribution practices. The study was based on materials produced by a health service team in an in-depth analysis of the accident. The analysis shows that decisions related to system modernization were taken without considering their implications in maintenance scheduling and creating conflicts of priorities and of interests between production and safety; and also reveals that the lack of a systemic perspective in safety management was its principal failure. To explain the accident as merely non-fulfillment of idealized formal safety rules feeds practices of blame attribution supported by alibi norms and inhibits possible prevention. In contrast, accident analyses undertaken in worker health surveillance services show potential to reveal origins of these events incubated in the history of the system ignored in practices guided by the traditional paradigm.

Áreas agrícolas trocam enormes fluxos de CO2, oferecendo uma oportunidade para mitigar o efeito estufa. Neste trabalho, estudou-se o potencial de sequestro de carbono em razão da conversão no manejo das principais atividades agrícolas do Brasil. Dados de vários estudos têm indicado que no sistema soja/milho e nas respectivas rotações, ocorre um sequestro de carbono no solo significativo ao longo dos anos de conversão do plantio convencional para o plantio direto, com uma média de 0,41 Mg C ha-1 ano-1. O mesmo efeito tem sido observado nos canaviais, porém há maiores acúmulos de carbono no solo quando as áreas de cana-de-açúcar são convertidas da colheita baseada na queima para a mecanizada, em que grandes quantidades de palha são deixadas na superfície do solo (1,8 Mg C ha-1 ano-1). Esse maior potencial de acúmulo de carbono no solo nos canaviais, comparado com outras culturas, está diretamente relacionado com a maior produção primária dessa cultura. Apesar disso, muito desse potencial de sequestro é perdido, uma vez que os canaviais são reformados, sob preparo intensivo do solo. As áreas de pasto mostram uma depleção nos estoques de carbono, quando convertidas de áreas naturais; porém, a integração dessas áreas com agricultura pode promover o aumento nos estoques de carbono do solo. Os trabalhos têm mostrado que as principais atividades agrícolas do Brasil possuem um grande potencial de mitigação, especialmente na forma de acúmulo de carbono no solo, sendo uma oportunidade para estratégias futuras.

Information about orthodontic movement of teeth with hypercementosis is scarce. As cementum deposition continues to occur, cementum is expected to change the shape of the root and apex over time, but this has not yet been demonstrated. Nor has it ever been established whether it increases or decreases the prevalence of root resorption during orthodontic treatment. The unique biological function of the interconnected network of cementocytes may play a role in orthodontic movement and its associated root resorptions, but no research has ever been conducted on the topic. Unlike cementum thickness and hypercementosis, root and apex shape has not yet been related to patient age. A study of the precise difference between increased cementum thickness and hypercementosis is warranted. Hypercementosis refers to excessive cementum formation above and beyond the extent necessary to fulfill its normal functions, resulting in abnormal thickening with macroscopic changes in the tooth root, which may require the delivery of forces that are different from conventional mechanics in their intensity, direction and distribution. What are the unique features and specificities involved in moving teeth that present with hypercementosis? Bodily movements would be expected to occur, since inclination might prove difficult to achieve, but would the root resorption index be higher or lower?

Este trabalho teve como objetivo avaliar o efeito do estresse salino sobre a eficiência fotoquímica do fotossistema II (PSII) nas cultivares de manga 'Haden', 'Palmer', 'Tommy Atkins' e 'Ubá' enxertadas sobre o porta-enxerto 'Imbu'. Foi utilizada solução nutritiva de Hoagland modificada contendo 0; 15; 30e 45 mmol L-1 NaCl. Aos 97 dias após a exposição ao estresse salino, foram avaliados os parâmetros da fluorescência da clorofila (F0, Fm, Fv, F0/Fm, Fv/Fm, Fv'/Fm', ΦPSII = [(Fm'-Fs)/(Fm')], D = (1- Fv'/Fm') e ETR = (ΦPSII×PPF×0,84×0,5). Aos 100 dias, foram avaliados a emissão foliar, a área média de folhas (cm²), o índice de toxidez nas folhas e o índice de abscisão foliar. em todas as cultivares, em graus diferenciados, ocorreram decréscimo na eficiência fotoquímica do fotossistema II, na emissão de folhas, e aumento nos índices de toxidez e abscisão foliar, intensificados nas concentrações a partir de 15 mmol L-1 NaCl. As plantas cultivadas em 45 mmol L-1 NaCl apresentaram decréscimos na razão Fv/Fm de 27,9; 18,7; 20,5 e 27,4%, incremento no índice de toxidez foliar de 33,0; 67,5; 41,6 e 80,8% e no índice de abscisão foliar de 71,8; 29,2; 32,5 e 67,9% para as cultivares 'Haden', 'Palmer', 'Tommy Atkins' e 'Uba', respectivamente. Os decréscimos na razão Fv/Fm foram acompanhados de redução na emissão de folhas e aumento no índice de toxidez foliar, mostrando, portanto, o potencial da fluorescência da clorofila na detecção precoce de estresse salino em mangueira.

Here we compute the static potential in scalar QED(3) at leading order in 1/Nf. We show that the addition of a non-minimal coupling of Pauli-type (is an element of(mu nu alpha)j(mu)partial derivative(nu)A(alpha)), although it breaks parity, it does not change the analytic structure of the photon propagator and consequently the static potential remains logarithmic ( confining) at large distances. The non-minimal coupling modifies the potential, however, at small charge separations giving rise to a repulsive force of short range between opposite sign charges, which is relevant for the existence of bound states. This effect is in agreement with a previous calculation based on Moller scattering, but differently from such calculation we show here that the repulsion appears independently of the presence of a tree level Chern-Simons term which rather affects the large distance behaviour of the potential turning it into a constant.

The coumarin antibiotics are potent inhibitors of DNA replication whose target is the enzyme DNA gyrase, an ATP-dependent bacterial type II topoisomerase. The coumarin drugs inhibit gyrase action by competitive binding to the ATP-binding site of DNA gyrase B protein. The production of new biologically active products has stimulated additional studies on coumarin-gyrase interactions. In this regard, a 4.2 kDa peptide mimic of DNA gyrase B protein from Escherichia coli has been designed and synthesized. The peptide sequence includes the natural fragment 131-146 (coumarin resistance-determining region) and a segment containing the gyrase-DNA interaction region (positions 753-770). The peptide mimic binds to novobiocin (K-a = 1.4 +/- 0.3 x 10(5) m(-1)), plasmid (K-a = 1.6 +/- 0.5 x 10(6) m(-1)) and ATP (K-a = 1.9 f 0.4 x 10(3) m(-1)), results previously found with the intact B protein. on the other hand, the binding to novobiocin was reduced when a mutation of Arg-136 to Leu-136 was introduced, a change previously found in the DNA gyrase B protein from several coumarin-resistant clinical isolates of Escherichia coLi. In contrast, the binding to plasmid and to ATP was not altered. These results suggest that synthetic peptides designed in a similar way to that described here could be used as mimics of DNA gyrase in studies which seek a better understanding of the ATP, as well as coumarin, binding to the gyrase and also the mechanism of action of this class of antibacterial drugs. Monocrotaline is a pyrrolizidine alkaloid present in plants of the Crotalaria species, which causes cytotoxicity and genotoxicity, including hepatotoxicity in animals and humans. It is metabolized by cytochrome P-450 in the liver to the alkylating agent dehydromonocrotaline. We evaluated the effects of monocrotaline and its metabolite on respiration, membrane potential and ATP levels in isolated rat liver mitochondria, and on respiratory chain complex I NADH oxidase activity in submitochondrial particles. Dehydromonocrotaline, but not the parent compound, showed a concentration-dependent inhibition of glutamate/malate-supported state 3 respiration (respiratory chain complex 1), but did not affect succinate-supported respiration (complex II). Only dehydromonocrotaline dissipated mitochondrial membrane potential, depleted ATP, and inhibited complex I NADH oxidase activity (IC50 = 62.06 mu M) through a non-competitive type of inhibition (K-I = 8.1 mu M). Therefore, dehydromonocrotaline is an inhibitor of the activity of respiratory chain complex I NADH oxidase, an action potentially accounting for the well-documented monocrotaline's hepatotoxicity to animals and humans. The mechanism probably involves change of the complex I conformation resulting from modification of cysteine thiol groups by the metabolite. (c) 2007 Elsevier Ltd. All rights reserved.

Quinolones constitute a family of compounds with a potent antibiotic activity. The enzyme DNA gyrase, responsible for the replication and transcription processes in DNA of bacteria, is involved in the mechanism of action of these drugs. In this sense, it is believed that quinolones stabilize the so-called 'cleavable complex' formed by DNA and gyrase, but the whole process is still far from being understood at the molecular level. This information is crucial in order to design new biological active products. As an approach to the problem, we have designed and synthesized low molecular weight peptide mimics of DNA gyrase. These peptides correspond to sequences of the subunit A of the enzyme from Escherichia coli, that include the quinolone resistance-determining region (positions 75-92) and a segment containing the catalytic Tyr-122 (positions 116-130). The peptide mimic of the non-mutated enzyme binds to ciprofloxin (CFX) only when DNA and Mg2+ were present (Kd = 1.6 × 10 -6 m), a result previously found with DNA gyrase. On the other hand, binding was reduced when mutations of Ser-83 to Leu-83 and Asp-87 to Asn-87 were introduced, a double change previously found in the subunit A of DNA gyrase from several CFX-resistant clinical isolates of E. coli. These results suggest that synthetic peptides designed in a similar way to that described here can be used as mimics of gyrases (topoisomerases) in order to study the binding of the quinolone to the enzyme-DNA complex as well as the mechanism of action of these antibiotics. Chagas' disease is a serious health problem for Latin America. The situation is worsened by the lack of efficient chemotherapy. The two available commercial drugs, benznidazole and nifurtimox, are more effective in the acute phase of the disease. Nitrofurazone is active against Trypanosoma cruzi, however its high toxicity precludes its current use in parasitosis. Hydroxymethylnitrofurazone is a prodrug of nitrofurazone. It is more active against Trypanosoma cruzi than nitrofurazone, besides being less toxic. This work shows the voltammetric behavior of nitrofurazone and a comparison with those of metronidazole and chloramphenicol using cyclic, linear sweep and differential pulse voltammetries. For these drugs also the prediction of the diffusion coefficients using Wilke-Chang equation was performed. The reduction of nitrofurazone is pH-dependent and in acidic medium the hydroxylamine derivative, involving four electrons, is the principal product formed. In aqueous-alkaline medium and with a glassy carbon electrode pre-treatment the reduction of nitrofurazone occurs in two steps, the first involving one electron to form the nitro-radical anion and the second corresponding to the hydroxylamine derivative formation. Hydroxymethylnitrofurazone presented the same voltammetric behavior and electroactivity, indicating that the molecular modification performed in nitrofurazone did not change its capacity to be reduced. A brief discussion regarding the differences in biological activity between the two compounds is also presented. ©2005 Sociedade Brasileira de Química.

Objetives: The aim of this study was to verify the anticariogenic effect of acidulate solutions with low NaF concentration, using pH-cycling model and bovine enamel. Material and methods: Enamel blocks were submitted to the surface microhardness (SMH) test and randomly divided in 12 experimental and one placebo groups. The blocks were submitted to pH cycling for 7 days, with daily applications once/day of 0.05% NaF and 0.1% NaF and twice/day of 0.02% NaF solutions. Four different pH: 4.0. 5.0, 6.0 and 7.0 were used. Next, SMH test was again used to determine the surface microhardness percentage change (%SMH). Data obtained for %SMH were homogeneous and passed through variance analyses and Tukey's test (5%) as far as fluoride concentrations and pH. Resulls:The results showed that pH influenced %SMH in 0.02% NaF and 0.05% NaF solutions with pH 4.0, which had less mineral loss compared to pH 7.0 (p̃0.05). The 0.02% NaF - pH 4.0, and 0.05% NaF- pH 7.0 groups showed similar results (p>0.05). A dose-response relationship was observed among the tested solutions, with better anticariogenic effect for the 0.1% NaF solution. Conclusion: The results suggest that the addition of citric acid to acidulate mouth rinses reduce mineral loss.
