La importancia de los sistemas de recomendación ha experimentado un crecimiento exponencial como consecuencia del auge de las redes sociales. En esta tesis doctoral presentaré una amplia visión sobre el estado del arte de los sistemas de recomendación. Incialmente, estos estaba basados en fitrado demográfico, basado en contendio o colaborativo. En la actualidad, estos sistemas incorporan alguna información social al proceso de recomendación. En el futuro utilizarán información implicita, local y personal proveniente del Internet de las cosas. Los sistemas de recomendación basados en filtrado colaborativo se pueden modificar con el fin de realizar recomendaciones a grupos de usuarios. Existen trabajos previos que han incluido estas modificaciones en diferentes etapas del algoritmo de filtrado colaborativo: búsqueda de los vecinos, predicción de las votaciones y elección de las recomendaciones. En esta tesis doctoral proporcionaré un nuevo método que realizar el proceso de unficación (pasar de varios usuarios a un grupo) en el primer paso del algoritmo de filtrado colaborativo: cálculo de la métrica de similaridad. Proporcionaré una formalización completa del método propuesto. Explicaré cómo obtener el conjunto de k vecinos del grupo de usuarios y mostraré cómo obtener recomendaciones usando dichos vecinos. Asimismo, incluiré un ejemplo detallando cada paso del método propuesto en un sistema de recomendación compuesto por 8 usuarios y 10 items. Las principales características del método propuesto son: (a) es más rápido (más eficiente) que las alternativas proporcionadas por otros autores, y (b) es al menos tan exacto y preciso como otras soluciones estudiadas. Para contrastar esta hipótesis realizaré varios experimentos que miden la precisión, la exactitud y el rendimiento del método. Los resultados obtenidos se compararán con los resultados de otras alternativas utilizadas en la recomendación de grupos. Los experimentos se realizarán con las bases de datos de MovieLens y Netflix. ABSTRACT The importance of recommender systems has grown exponentially with the advent of social networks. In this PhD thesis I will provide a wide vision about the state of the art of recommender systems. They were initially based on demographic, contentbased and collaborative filtering. Currently, these systems incorporate some social information to the recommendation process. In the future, they will use implicit, local and personal information from the Internet of Things. As we will see here, recommender systems based on collaborative filtering can be used to perform recommendations to group of users. Previous works have made this modification in different stages of the collaborative filtering algorithm: establishing the neighborhood, prediction phase and determination of recommended items. In this PhD thesis I will provide a new method that carry out the unification process (many users to one group) in the first stage of the collaborative filtering algorithm: similarity metric computation. I will provide a full formalization of the proposed method. I will explain how to obtain the k nearest neighbors of the group of users and I will show how to get recommendations using those users. I will also include a running example of a recommender system with 8 users and 10 items detailing all the steps of the method I will present. The main highlights of the proposed method are: (a) it will be faster (more efficient) that the alternatives provided by other authors, and (b) it will be at least as precise and accurate as other studied solutions. To check this hypothesis I will conduct several experiments measuring the accuracy, the precision and the performance of my method. I will compare these results with the results generated by other methods of group recommendation. The experiments will be carried out using MovieLens and Netflix datasets.

The present paper describes the preliminary stages of the development of a new, comprehensive model conceived to simulate the evacuation of transport airplanes in certification studies. Two previous steps were devoted to implementing an efficient procedure to define the whole geometry of the cabin, and setting up an algorithm for assigning seats to available exits. Now, to clarify the role of the cabin arrangement in the evacuation process, the paper addresses the influence of several restrictions on the seat-to-exit assignment algorithm, maintaining a purely geometrical approach for consistency. Four situations are considered: first, an assignment method without limitations to search the minimum for the total distance run by all passengers along their escaping paths; second, a protocol that restricts the number of evacuees through each exit according to updated FAR 25 capacity; third, a procedure which tends to the best proportional sharing among exits but obliges to each passenger to egress through the nearest fore or rear exits; and fourth, a scenario which includes both restrictions. The four assignment strategies are applied to turboprops, and narrow body and wide body jets. Seat to exit distance and number of evacuees per exit are the main output variables. The results show the influence of airplane size and the impact of non-symmetries and inappropriate matching between size and longitudinal location of exits.

The colony shape of four yeast species growing on agar medium wasmeasured for 116 days by image analysis. Initially, all the colonies are circular, with regular edges. The loss of circularity can be quantitatively estimated by the eccentricity index, Ei, calculated as the ratio between their orthogonal vertical and horizontal diameters. Ei can increase from 1 (complete circularity) to a maximum of 1.17–1.30, depending on the species. One colony inhibits its neighbour only when it has reached a threshold area. Then, Ei of the inhibited colony increases proportionally to the area of the inhibitory colony. The initial distance between colonies affects those threshold values but not the proportionality, Ei/area; this inhibition affects the shape but not the total surface of the colony. The appearance of irregularities in the edges is associated, in all the species, not with age but with nutrient exhaustion. The edge irregularity can be quantified by the Fourier index, Fi, calculated by the minimum number of Fourier coefficients that are needed to describe the colony contour with 99% fitness. An ad hoc function has been developed in Matlab v. 7.0 to automate the computation of the Fourier coefficients. In young colonies, Fi has a value between 2 (circumference) and 3 (ellipse). These values are maintained in mature colonies of Debaryomyces, but can reach values up to 14 in Saccharomyces.All the species studied showed the inhibition of growth in facing colony edges, but only three species showed edge irregularities associated with substrate exhaustion. Two-phase plant communities with an engineer conforming conspicuous patches and affecting the performance and patterns of coexisting species are the norm under stressful conditions. To unveil the mechanisms governing coexistence in these communities at multiple spatial scales, we have developed a new point-raster approach of spatial pattern analysis, which was applied to a Mediterranean high mountain grassland to show how Festuca curvifolia patches affect the local distribution of coexisting species. We recorded 22 111 individuals of 17 plant perennial species. Most coexisting species were negatively associated with F. curvifolia clumps. Nevertheless, bivariate nearest-neighbor analyses revealed that the majority of coexisting species were confined at relatively short distances from F. curvifolia borders (between 0-2 cm and up to 8 cm in some cases). Our study suggests the existence of a fine-scale effect of F. curvifolia for most species promoting coexistence through a mechanism we call 'facilitation in the halo'. Most coexisting species are displaced to an interphase area between patches, where two opposite forces reach equilibrium: attenuated severe conditions by proximity to the F. curvifolia canopy (nutrient-rich islands) and competitive exclusion mitigated by avoiding direct contact with F. curvifolia.

One of the main problems relief teams face after a natural or man-made disaster is how to plan rural road repair work tasks to take maximum advantage of the limited available financial and human resources. Previous research focused on speeding up repair work or on selecting the location of health centers to minimize transport times for injured citizens. In spite of the good results, this research does not take into account another key factor: survivor accessibility to resources. In this paper we account for the accessibility issue, that is, we maximize the number of survivors that reach the nearest regional center (cities where economic and social activity is concentrated) in a minimum time by planning which rural roads should be repaired given the available financial and human resources. This is a combinatorial problem since the number of connections between cities and regional centers grows exponentially with the problem size, and exact methods are no good for achieving an optimum solution. In order to solve the problem we propose using an Ant Colony System adaptation, which is based on ants? foraging behavior. Ants stochastically build minimal paths to regional centers and decide if damaged roads are repaired on the basis of pheromone levels, accessibility heuristic information and the available budget. The proposed algorithm is illustrated by means of an example regarding the 2010 Haiti earthquake, and its performance is compared with another metaheuristic, GRASP.

A pesar de los avances en materia de predicción, los desastres naturales siguen teniendo consecuencias devastadoras. Entre los principales problemas a los que se enfrentan los equipos de ayuda y rescate después de un desastre natural o provocado por el hombre se encuentra la planificación de las tareas de reparación de carreteras para conseguir la máxima ventaja de los limitados recursos económicos y humanos. En la presente Tesis Fin de Máster se intenta dar solución al problema de la accesibilidad, es decir, maximizar el número de supervivientes que consiguen alcanzar el centro regional más cercano en un tiempo mínimo mediante la planificación de qué carreteras rurales deberían ser reparadas dados unos recursos económicos y humanos limitados. Como se puede observar, es un problema combinatorio ya que el número de planes de reparación y conexiones entre las ciudades y los centros regionales crece de forma exponencial con el tamaño del problema. Para la resolución del problema se comienza analizando una adaptación básica de los sistemas de colonias de hormigas propuesta por otro autor y se proponen múltiples mejoras sobre la misma. Posteriormente, se propone una nueva adaptación más avanzada de los sistemas de colonias de hormiga al problema, el ACS con doble hormiga. Este sistema hace uso de dos tipos distintos de hormigas, la exploradora y la trabajadora, para resolver simultáneamente el problema de encontrar los caminos más rápidos desde cada ciudad a su centro regional más cercano (exploradora), y el de obtener el plan óptimo de reparación que maximice la accesibilidad de la red (trabajadora). El algoritmo propuesto se ilustra por medio de un ejemplo de gran tamaño que simula el desastre natural ocurrido en Haití, y su rendimiento es comparado con la combinación de dos metaheurísticas, GRASP y VNS.---ABSTRACT---In spite of the advances in forecasting, natural disaster continue to ocasionate devastating consequences. One of the main problems relief teams face after a natural or man-made disaster is how to plan rural road repair work to take maximum advantage of the limited available financial and human resources. In this Master´s Final Project we account for the accesability issue, that is, to maximize the number of survivors that reach the nearest regional center in a minimum time by planning whic rural roads should be repaired given the limited financial and human resources. This is a combinatorial problem since the number of possible repairing solutions and connections between cities and regional centers grows exponentially with the size of the problem. In order to solve the problem, we analyze the basic ant colony system adaptation proposed by another author and point out multiple improvements on it. Then, we propose a novel and more advance adaptation of the ant colony systems to the problem, the double- ant ACS. This system makes use of two diferent type of ants, the explorer and the worker, to simultaneously solve the problem of finding the shorthest paths from each city to their nearest regional center (explorer), and the problem of identifying the optimal repairing plan that maximize the network accesability (worker). The proposed algorithm is illustrated by means of a big size example that simulates the natural disaster occurred in Haiti, and its performance is compared with a combination of two metaheuristics, GRASP and VNS.

The initial step in most facial age estimation systems consists of accurately aligning a model to the output of a face detector (e.g. an Active Appearance Model). This fitting process is very expensive in terms of computational resources and prone to get stuck in local minima. This makes it impractical for analysing faces in resource limited computing devices. In this paper we build a face age regressor that is able to work directly on faces cropped using a state-of-the-art face detector. Our procedure uses K nearest neighbours (K-NN) regression with a metric based on a properly tuned Fisher Linear Discriminant Analysis (LDA) projection matrix. On FG-NET we achieve a state-of-the-art Mean Absolute Error (MAE) of 5.72 years with manually aligned faces. Using face images cropped by a face detector we get a MAE of 6.87 years in the same database. Moreover, most of the algorithms presented in the literature have been evaluated on single database experiments and therefore, they report optimistically biased results. In our cross-database experiments we get a MAE of roughly 12 years, which would be the expected performance in a real world application.

Existe un amplio catálogo de posibles soluciones para resolver la problemática de las zapatas de medianería así como, por extensión, las zapatas de esquina como caso particular de las anteriores. De ellas, las más habitualmente empleadas en estructuras de edificación son, por un lado, la utilización de una viga centradora que conecta la zapata de medianería con la zapata del pilar interior más próximo y, por otro, la colaboración de la viga de la primera planta trabajando como tirante. En la primera solución planteada, el equilibrio de la zapata de medianería y el centrado de la respuesta del terreno se consigue gracias a la colaboración del pilar interior con su cimentación y al trabajo a flexión de la viga centradora. La modelización clásica considera que se logra un centrado total de la reacción del terreno, con distribución uniforme de las tensiones de contacto bajo ambas zapatas. Este planteamiento presupone, por tanto, que la viga centradora logra evitar cualquier giro de la zapata de medianería y que el pilar puede, por ello, considerarse perfectamente empotrado en la cimentación. En este primer modelo, el protagonismo fundamental recae en la viga centradora, cuyo trabajo a flexión conduce frecuentemente a unas escuadrías y a unas cuantías de armado considerables. La segunda solución, plantea la colaboración de la viga de la primera planta, trabajando como tirante. De nuevo, los métodos convencionales suponen un éxito total en el mecanismo estabilizador del tirante, que logra evitar cualquier giro de la zapata de medianería, dando lugar a una distribución de tensiones también uniforme. Los modelos convencionales existentes para el cálculo de este tipo de cimentaciones presentan, por tanto, una serie de simplificaciones que permiten el cálculo de las mismas, por medios manuales, en un tiempo razonable, pero presentan el inconveniente de su posible alejamiento del comportamiento real de la cimentación, con las consecuencias negativas que ello puede suponer en el dimensionamiento de estos elementos estructurales. La presente tesis doctoral desarrolla un contraste de los modelos convencionales de cálculo de cimentaciones de medianería y esquina, mediante un análisis alternativo con modelos de elementos finitos, con el objetivo de poner de manifiesto las diferencias entre los resultados obtenidos con ambos tipos de modelización, analizar cuáles son las variables que más influyen en el comportamiento real de este tipo de cimentaciones y proponer un nuevo modelo de cálculo, de tipo convencional, más ajustado a la realidad. El proceso de investigación se desarrolla mediante una etapa experimental virtual que utiliza como modelo un pórtico tipo de edificación, ortogonal, de hormigón armado, con dos vanos y número variable de plantas. Tras identificar el posible giro de la cimentación como elemento clave en el comportamiento de las zapatas de medianería y de esquina, se adoptan como variables de estudio aquellas que mayor influencia puedan tener sobre el citado giro de las zapatas y sobre la rigidez del conjunto del elemento estructural. Así, se han estudiado luces de 3 m a 7 m, diferente número de plantas desde baja+1 hasta baja+4, resistencias del terreno desde 100 kN/m2 hasta 300 kN/m2, relaciones de forma de la zapata de medianería de 1,5 : 1 y 2 : 1, aumento y reducción de la cuantía de armado de la viga centradora y variación del canto de la viga centradora desde el mínimo canto compatible con el anclaje de la armadura de los pilares hasta un incremento del 75% respecto del citado canto mínimo. El conjunto de pórticos generados al aplicar las variables indicadas, se ha calculado tanto por métodos convencionales como por el método de los elementos finitos. Los resultados obtenidos ponen de manifiesto importantes discrepancias entre ambos métodos que conducen a importantes diferencias en el dimensionamiento de este tipo de cimentaciones. El empleo de los métodos tradicionales da lugar, por un lado, a un sobredimensionamiento de la armadura de la viga centradora y, por otro, a un infradimensionamiento, tanto del canto de la viga centradora, como del tamaño de la zapata de medianería y del armado de la viga de la primera planta. Finalizado el análisis y discusión de resultados, la tesis propone un nuevo método alternativo, de carácter convencional y, por tanto, aplicable a un cálculo manual en un tiempo razonable, que permite obtener los parámetros clave que regulan el comportamiento de las zapatas de medianería y esquina, conduciendo a un dimensionamiento más ajustado a las necesidades reales de este tipo de cimentación. There is a wide catalogue of possible solutions to solve the problem of party shoes and, by extension, corner shoes as a special case of the above. From all of them, the most commonly used in building structures are, on one hand, the use of a centering beam that connects the party shoe with the shoe of the nearest interior pillar and, on the other hand, the collaboration of the beam of the first floor working as a tie rod. In the first proposed solution, the balance of the party shoe and the centering of the ground response is achieved thanks to the collaboration of the interior pillar with his foundation along with the bending work of the centering beam. Classical modeling considers that a whole centering of the ground reaction is achieved, with uniform contact stress distribution under both shoes. This approach to the issue presupposes that the centering beam manages to avoid any rotation of the party shoe, so the pillar can be considered perfectly embedded in the foundation. In this first model, the leading role lies in the centering beam, whose bending work usually leads to important section sizes and high amounts of reinforced. The second solution, consideres the collaboration of the beam of the first floor, working as tie rod. Again, conventional methods involve a total success in the stabilizing mechanism of the tie rod, that manages to avoid any rotation of the party shoe, resulting in a stress distribution also uniform. Existing conventional models for calculating such foundations show, therefore, a series of simplifications which allow calculation of the same, by manual means, in a reasonable time, but have the disadvantage of the possible distance from the real behavior of the foundation, with the negative consequences this could bring in the dimensioning of these structural elements. The present thesis develops a contrast of conventional models of calculation of party and corner foundations by an alternative analysis with finite element models with the aim of bring to light the differences between the results obtained with both types of modeling, analysis which are the variables that influence the real behavior of this type of foundations and propose a new calculation model, conventional type, more adjusted to reality. The research process is developed through a virtual experimental stage using as a model a typical building frame, orthogonal, made of reinforced concrete, with two openings and variable number of floors. After identifying the possible spin of the foundation as the key element in the behavior of the party and corner shoes, it has been adopted as study variables, those that may have greater influence on the spin of the shoes and on the rigidity of the whole structural element. So, it have been studied lights from 3 m to 7 m, different number of floors from lower floor + 1 to lower floor + 4, máximum ground stresses from 100 kN/m2 300 kN/m2, shape relationships of party shoe 1,5:1 and 2:1, increase and decrease of the amount of reinforced of the centering beam and variation of the height of the centering beam from the minimum compatible with the anchoring of the reinforcement of pillars to an increase of 75% from the minimum quoted height. The set of frames generated by applying the indicated variables, is calculated both by conventional methods such as by the finite element method. The results show significant discrepancies between the two methods that lead to significant differences in the dimensioning of this type of foundation. The use of traditional methods results, on one hand, to an overdimensioning of the reinforced of the centering beam and, on the other hand, to an underdimensioning, both the height of the centering beam, such as the size of the party shoe and the reinforced of the beam of the first floor. After the analysis and discussion of results, the thesis proposes a new alternative method, conventional type and, therefore, applicable to a manual calculation in a reasonable time, that allows to obtain the key parameters that govern the behavior of party and corner shoes, leading to a dimensioning more adjusted to the real needings of this type of foundation.

El análisis de los factores que determinan el establecimiento y supervivencia de orquídeas epífitas, incluyen: a) las condiciones microambientales de los bosques que las mantienen, b) preferencias por las características de los hospederos donde crecen, c) limitación en la dispersión de semillas, d) interacciones planta-planta, y e) asociaciones micorrízicas para la germinación y resultan esenciales para el desarrollo de estrategias para la conservación y manejo de este grupo de plantas. Este trabajo ha evaluado la importancia de estos factores en Epidendrum rhopalostele, orquídea epífita del bosque de niebla montano, a través de los análisis de los patrones espaciales de los árboles que la portan y de la propia orquídea, a escala de población, estudios de asociación y métodos moleculares. Estos últimos han consistido en el uso de marcadores AFLP para el análisis de la estructura genética de la orquídea y en la secuenciación-clonación de la región ITS para la identificación de los hongos micorrízicos asociados. El objetivo de esta tesis es, por tanto, una mejor comprensión de los factores que condicionan la presencia de orquídeas epífitas en los remanentes de bosque de niebla montano y una evaluación de las implicaciones para la conservación y mantenimiento de sus hábitats y la permanencia de sus poblaciones. El estudio fue realizado en un fragmento de bosque de niebla montano de sucesión secundaria situado al este de la Cordillera Real, en los Andes del sur de Ecuador, a 2250 m.s.n.m y caracterizado por una pendiente marcada, temperatura media anual de 20.8°C y precipitación anual de 2193 mm. En este fragmento se mapearon, identificaron y caracterizaron todos los árboles presentes con DBH > 1 cm y todos los individuos de Epidendrum rhopalostele. Así mismo se tomaron muestras de hoja para obtener ADN de todas las orquídeas registradas y muestras de raíces de individuos con flor de E. rhopalostele, uno por cada forófito, para el análisis filogenético de micorrizas. Análisis espaciales de patrones de puntos basados en la K de Ripley y la distancia al vecino más cercano fueron usados para los árboles, forófitos y la población de E. rhopalostele. Se observó que la distribución espacial de árboles y forófitos de E. rhopalostele no es aleatoria, ya que se ajusta a un proceso agregado de Poisson. De ahí se infiere una limitación en la dispersión de las semillas en el fragmento estudiado y en el establecimiento de la orquídea. El patrón de distribución de la población de E. rhopalostele en el fragmento muestra un agrupamiento a pequeña escala sugiriendo una preferencia por micro-sitios para el establecimiento de la orquídea con un kernel de dispersión de las semillas estimado de 0.4 m. Las características preferentes del micro-sitio como tipos de árboles (Clusia alata y árboles muertos), tolerancia a la sombra, corteza rugosa, distribución en los dos primeros metros sugieren una tendencia a distribuirse en el sotobosque. La existencia de una segregación espacial entre adultos y juveniles sugiere una competencia por recursos limitados condicionada por la preferencia de micro-sitio. La estructura genética de la población de E. rhopalostele analizada a través de Structure y PCoA evidencia la presencia de dos grupos genéticos coexistiendo en el fragmento y en los mismos forófitos, posiblemente por eventos de hibridización entre especies de Epidendrum simpátricas. Los resultados del análisis de autocorrelación espacial efectuados en GenAlex confirman una estructura genético-espacial a pequeña escala que es compatible con un mecanismo de dispersión de semillas a corta distancia ocasionada por gravedad o pequeñas escorrentías, frente a la dispersión a larga distancia promovida por el viento generalmente atribuida a las orquídeas. Para la identificación de los micobiontes se amplificó la región ITS1-5.8S-ITS2, y 47 secuencias fueron usadas para el análisis filogenético basado en neighborjoining, análisis bayesiano y máximum-likelihood que determinó que Epidendrum rhopalostele establece asociaciones micorrízicas con al menos dos especies diferentes de Tulasnella. Se registraron plantas que estaban asociadas con los dos clados de hongos encontrados, sugiriendo ausencia de limitación en la distribución del hongo. Con relación a las implicaciones para la conservación in situ resultado de este trabajo se recomienda la preservación de todo el fragmento de bosque así como de las interacciones existentes (polinizadores, micorrizas) a fin de conservar la diversidad genética de esta orquídea epífita. Si fuere necesaria una reintroducción se deben contemplar distancias entre los individuos en cada forófito dentro de un rango de 0.4 m. Para promover el reclutamiento y regeneración de E. rhopalostele, se recomienda que los forófitos correspondan preferentemente a árboles muertos o caídos y a especies, como Clusia alata, que posean además corteza rugosa, sean tolerantes a la sombra, y en el área del sotobosque con menor luminosidad. Además es conveniente que las orquídeas en su distribución vertical estén ubicadas en los primeros metros. En conclusión, la limitación en la dispersión, las características del micro-sitio, las interacciones intraespecíficas y con especies congenéricas simpátricas y las preferencias micorrízicas condicionan la presencia de esta orquídea epífita en este tipo de bosque. ABSTRACT The analysis of factors that determine the establishment and survival of epiphytic depends on factors such as a) microenvironmental conditions of forest, b) preference for host characteristics where orchids grow, c) seed dispersal limitation, d) plant-plant interaction, e) priority mycorrhizal associations for germination, are essential for the development of strategies for management and conservation. This work evaluated the importance of these factors in Epidendrum rhopalostele, an epiphytic orchid of montane cloud forest through the analysis of spatial patterns of host trees and the orchid, in a more specific scale, with association studies and molecular methods, including AFLPs for orchid population genetic structure and the sequencing of the ITS region for associated mycorrhizal fungi. The aim of this thesis is to understand the factors that condition the presence of epiphytic orchids in the remnants of montane cloud forest and to assess the implications for the conservation and preservation of their habitats and the persistence of the orchid populations. The study was carried out in a fragment of montane cloud forest of secondary succession on the eastern slope of Cordillera Real in the Andes of southern Ecuador, located at 2250 m a.s.l. characterized by a steep slope, mean annual temperature of 20.8°C and annual precipitation of 2193 mm. All trees with DBH > 1 cm were mapped, characterized and identified. All E. rhopalostele individuals present were counted, marked, characterized and mapped. Leaf samples of all orchid individuals were collected for DNA analysis. Root samples of flowering E. rhopalostele individuals were collected for phylogenetic analysis of mycorrhizae, one per phorophyte. Spatial point pattern analysis based on Ripley`s K function and nearest neighbor function was used for trees, phorophytes and orchid population. We observed that spatial distribution of trees and phorophytes is not random, as it adjusts to a Poisson cluster process. This suggests a limitation for seed dispersal in the study fragment that is affecting orchid establishment. Furthermore, the small-scale spatial pattern of E. rhopalostele evidences a clustering that suggests a microsite preference for orchid establishment with a dispersal kernel of 0.4 m. Microsite features such as types of trees (dead trees or Clusia alata), shade tolerance trees, rough bark, distribution in the first meters suggest a tendency to prefer the understory for their establishment. Regarding plant-plant interaction a spatial segregation between adults and juveniles was present suggesting competition for limited resources conditioned for a microsite preference. Analysis of genetic structure of E. rhopalostele population through Structure and PCoA shows two genetic groups coexisting in this fragment and in the same phorophyte, possibly as a result of hybridization between sympatric species of Epidendrum. Our results of spatial autocorrelation analysis develop in GenAlex confirm a small-scale spatial-genetic structure within the genetic groups that is compatible with a short-distance dispersal mechanism caused by gravity or water run-off, instead of the long-distance seed dispersal promoted by wind generally attributed to orchids. For mycobionts identification ITS1-5.8S-ITS2 rDNA region was amplified. Phylogenetic analysis was performed with neighborjoining, Bayesian likelihood and maximum-likelihood for 47 sequences yielded two Tulasnella clades. This orchid establishes mycorrhizal associations with at least two different Tulasnella species. In some cases both fungi clades were present in same root, suggesting no limitation in fungal distribution. Concerning the implications for in situ conservation resulting from this work, the preservation of all forest fragment and their interactions (pollinators, mycorrhiza) is recommended to conserve the genetic diversity of this species. If a reintroduction were necessary, distances between individuals in each phorophyte within a range of 0.4 m, are recommended. To promote recruitment and regeneration of E. rhopalostele it is recommended that phorophytes correspond to dead or fallen trees or species, such as Clusia alata. Trees that have rough bark and are shade tolerant are also recommended. Furthermore, regarding vertical distribution, it is also convenient that orchids are located in the first meter (in understory, area with less light). In conclusion, limitation on seed dispersal, microsite characteristics, plant-plant interactions or interaction with cogeneric sympatric species and mycorrhizal preferences conditioned the presence of this epiphytic orchid in this fragment forest.

El presente trabajo tiene como objetivo evaluar el aprendizaje de los deportes colectivos en el alumnado de segundo ciclo de Educación Secundaria Obligatoria (ESO) utilizando una herramienta denominada Game Performance Assessment Instrument (GPAI). Para ello se ha realizado una revisión de dos modelos de enseñanza existentes: el modelo técnico y el modelo comprensivo. Así mismo hemos revisado dos tipos de procesos de evaluación; por un lado la evaluación sumativa, más relacionada con el modelo técnico; por otro lado la evaluación formativa, más próxima al modelo comprensivo. A continuación se ha llevado a cabo una intervención didáctica aplicando el modelo de enseñanza comprensivo (TGfU) y el instrumento mencionado anteriormente (GPAI). Los resultados obtenidos han sido utilizados para proponer un sistema de evaluación basado en el modelo comprensivo con la utilización del GPAI y a través de la coevaluación como forma de asentar el aprendizaje en el alumnado. Se concluye que el modelo de enseñanza comprensivo (TGfU) con el GPAI y una evaluación entre iguales es bastante óptimo para favorecer el proceso de enseñanzaaprendizaje de los deportes colectivos en el alumnado de segundo ciclo de Educación Secundaria Obligatoria (ESO). ABSTRACT The present work has the aim to assess the learning of collective sports in Secondary Education students using a tool called Game Performance Assessment Instrument (GPAI). For that we have used to revision of two teaching models: the technical model and Teaching Games for Understanding (TGfU). At the same time, we have reviewed two kinds of assessment process; on one hand summative assessment which is related to technical model; on the other hand formative assessment nearest to Teaching Games for Understanding (TGfU). Next it has been done a didactic intervention using the Teaching Games for Understanding (TGfU) and that instrument mentioned beforehand. The results obtained have been used to suggest an assessment system based on Teaching Games for Understanding (TGfU) using the GPAI through the peer assessment as a way of teaching.

Sequence analysis based on multiple isolates representing essentially all genera and species of the classic family Volvocaeae has clarified their phylogenetic relationships. Cloned internal transcribed spacer sequences (ITS-1 and ITS-2, flanking the 5.8S gene of the nuclear ribosomal gene cistrons) were aligned, guided by ITS transcript secondary structural features, and subjected to parsimony and neighbor joining distance analysis. Results confirm the notion of a single common ancestor, and Chlamydomonas reinharditii alone among all sequenced green unicells is most similar. Interbreeding isolates were nearest neighbors on the evolutionary tree in all cases. Some taxa, at whatever level, prove to be clades by sequence comparisons, but others provide striking exceptions. The morphological species Pandorina morum, known to be widespread and diverse in mating pairs, was found to encompass all of the isolates of the four species of Volvulina. Platydorina appears to have originated early and not to fall within the genus Eudorina, with which it can sometimes be confused by morphology. The four species of Pleodorina appear variously associated with Eudorina examples. Although the species of Volvox are each clades, the genus Volvox is not. The conclusions confirm and extend prior, more limited, studies on nuclear SSU and LSU rDNA genes and plastid-encoded rbcL and atpB. The phylogenetic tree suggests which classical taxonomic characters are most misleading and provides a framework for molecular studies of the cell cycle-related and other alterations that have engendered diversity in both vegetative and sexual colony patterns in this classical family.

The transcription factors nuclear factor of activated T cells (NFAT) and activator protein 1 (AP-1) coordinately regulate cytokine gene expression in activated T-cells by binding to closely juxtaposed sites in cytokine promoters. The structural basis for cooperative binding of NFAT and AP-1 to these sites, and indeed for the cooperative binding of transcription factors to composite regulatory elements in general, is not well understood. Mutagenesis studies have identified a segment of AP-1, which lies at the junction of its DNA-binding and dimerization domains (basic region and leucine zipper, respectively), as being essential for protein–protein interactions with NFAT in the ternary NFAT/AP-1/DNA complex. In a model of the ternary complex, the segment of NFAT nearest AP-1 is the Rel insert region (RIR), a feature that is notable for its hypervariability in size and in sequence amongst members of the Rel transcription factor family. Here we have used mutational analysis to study the role of the NFAT RIR in binding to DNA and AP-1. Parallel yeast one-hybrid screening assays in combination with alanine-scanning mutagenesis led to the identification of four amino acid residues in the RIR of NFAT2 (also known as NFATC1 or NFATc) that are essential for cooperativity with AP-1 (Ile-544, Glu-545, Thr-551, and Ile-553), and three residues that are involved in interactions with DNA (Lys-538, Arg-540, and Asn-541). These results were confirmed and extended through in vitro binding assays. We thus conclude that the NFAT RIR plays an essential dual role in DNA recognition and cooperative binding to AP-1 family transcription factors.

DNA exhibits a surprising multiplicity of structures when it is packed into dense aggregates. It undergoes various polymorphous transitions (e.g., from the B to A form) and mesomorphous transformations (from hexagonal to orthorhombic or monoclinic packing, changes in the mutual alignment of nearest neighbors, etc). In this report we show that such phenomena may have their origin in the specific helical symmetry of the charge distribution on DNA surface. Electrostatic interaction between neighboring DNA molecules exhibits strong dependence on the patterns of molecular surface groups and adsorbed counter-ions. As a result, it is affected by such structural parameters as the helical pitch, groove width, the number of base pairs per helical turn, etc. We derive expressions which relate the energy of electrostatic interaction with these parameters and with the packing variables characterizing the axial and azimuthal alignment between neighboring macromolecules. We show, in particular, that the structural changes upon the B-to-A transition reduce the electrostatic energy by ≈kcal/mol per base pair, at a random adsorption of counter ions. Ion binding into the narrow groove weakens or inverts this effect, stabilizing B-DNA, as it is presumably the case in Li+-DNA assemblies. The packing symmetry and molecular alignment in DNA aggregates are shown to be affected by the patterns of ion binding.

Yeast and vertebrate nuclear pores display significant morphological similarity by electron microscopy, but sequence similarity between the respective proteins has been more difficult to observe. Herein we have identified a vertebrate nucleoporin, Nup93, in both human and Xenopus that has proved to be an evolutionarily related homologue of the yeast nucleoporin Nic96p. Polyclonal antiserum to human Nup93 detects corresponding proteins in human, rat, and Xenopus cells. Immunofluorescence and immunoelectron microscopy localize vertebrate Nup93 at the nuclear basket and at or near the nuclear entry to the gated channel of the pore. Immunoprecipitation from both mammalian and Xenopus cell extracts indicates that a small fraction of Nup93 physically interacts with the nucleoporin p62, just as yeast Nic96p interacts with the yeast p62 homologue. However, a large fraction of vertebrate Nup93 is extracted from pores and is also present in Xenopus egg extracts in complex with a newly discovered 205-kDa protein. Mass spectrometric sequencing of the human 205-kDa protein reveals that this protein is encoded by an open reading frame, KIAAO225, present in the human database. The putative human nucleoporin of 205 kDa has related sequence homologues in Caenorhabditis elegans and Saccharomyces cerevisiae. To analyze the role of the Nup93 complex in the pore, nuclei were assembled that lack the Nup93 complex after immunodepletion of a Xenopus nuclear reconstitution extract. The Nup93-complex–depleted nuclei are clearly defective for correct nuclear pore assembly. From these experiments, we conclude that the vertebrate and yeast pore have significant homology in their functionally important cores and that, with the identification of Nup93 and the 205-kDa protein, we have extended the knowledge of the nearest-neighbor interactions of this core in both yeast and vertebrates.

Current evidence indicates that methylation of cytosine in mammalian DNA is restricted to both strands of the symmetrical sequence CpG, although there have been sporadic reports that sequences other than CpG may also be methylated. We have used a dual-labeling nearest neighbor technique and bisulphite genomic sequencing methods to investigate the nearest neighbors of 5-methylcytosine residues in mammalian DNA. We find that embryonic stem cells, but not somatic tissues, have significant cytosine-5 methylation at CpA and, to a lesser extent, at CpT. As the expression of the de novo methyltransferase Dnmt3a correlates well with the presence of non-CpG methylation, we asked whether Dnmt3a might be responsible for this modification. Analysis of genomic methylation in transgenic Drosophila expressing Dnmt3a reveals that Dnmt3a is predominantly a CpG methylase but also is able to induce methylation at CpA and at CpT.
