The aim of this study was to determine if DNA polymorphism within runt-related gene 2 (RUNX2)/core binding factor A1 (CBFA1) is related to bone mineral density (BMD). RUNX2 contains a glutamine-alanine repeat where mutations causing cleidocranial dysplasia (CCD) have been observed. Two common variants were detected within the alanine repeat: an 18-bp deletion and a synonymous alanine codon polymorphism with alleles GCA and GCG (noted as A and G alleles, respectively). In addition, rare mutations that may be related to low BMD were observed within the glutamine repeat. In 495 randomly selected women of the Geelong Osteoporosis Study (GOS), the A allele was associated with higher BMD at all sites tested. The effect was maximal at the ultradistal (UD) radius (p = 0.001). In a separate fracture study, the A allele was significantly protective against Colles' fracture in elderly women but not spine and hip fracture. The A allele was associated with increased BMD and was protective against a common form of osteoporotic fracture, suggesting that RUNX2 variants may be related to genetic effects on BMD and osteoporosis.

Much of northern Australia’s tropical savannas are subject to annual intense and extensive late dry season wildfires, much of this occurring on Aboriginal land. Based on the successful West Arnhem Land Fire Abatement (WALFA) model, which has resulted in significantly reduced greenhouse gas emissions, fire abatement programmes are planned for other significant regions of northern Australia. This study offers an introduction to the ideas behind a proposed environmental and social benchmarking project that aims to evaluate the potential benefits of expanding the fire abatement program in northern Australia, under the leadership of NAILSMA and its partners. Gaining a better understanding of the biodiversity, social and cultural outcomes of these fire abatement activities is an important component of demonstrating multiple benefits of these programmes. We emphasize the role of both biodiversity and cultural mapping to establish benchmarks and baseline states, with the involvement of Indigenous communities being a key element to optimize social and biodiversity benefits. Consultation with Traditional Owners and ranger groups to establish an agreed set of targets, indicators and sampling protocols and methodologies are critical component of this process. Examples of preliminary work to date are provided.

We previously used Gene Expression Signature technology to identify methazolamide (MTZ) and related compounds with insulin sensitizing activity in vitro. The effects of these compounds were investigated in diabetic db/db mice, insulin-resistant diet-induced obese (DIO) mice, and rats with streptozotocin (STZ)-induced diabetes. MTZ reduced fasting blood glucose and HbA1c levels in db/db mice, improved glucose tolerance in DIO mice, and enhanced the glucose-lowering effects of exogenous insulin administration in rats with STZ-induced diabetes. Hyperinsulinemic-euglycemic clamps in DIO mice revealed that MTZ increased glucose infusion rate and suppressed endogenous glucose production. Whole-body or cellular oxygen consumption rate was not altered, suggesting MTZ may inhibit glucose production by different mechanism(s) to metformin. In support of this, MTZ enhanced the glucose-lowering effects of metformin in db/db mice. MTZ is known to be a carbonic anhydrase inhibitor (CAI); however, CAIs acetazolamide, ethoxyzolamide, dichlorphenamide, chlorthalidone, and furosemide were not effective in vivo. Our results demonstrate that MTZ acts as an insulin sensitizer that suppresses hepatic glucose production in vivo. The antidiabetic effect of MTZ does not appear to be a function of its known activity as a CAI. The additive glucose-lowering effect of MTZ together with metformin highlights the potential utility for the management of type 2 diabetes.

RUNX2 is an essential transcription factor required for skeletal development and cartilage formation. Haploinsufficiency of RUNX2 leads to cleidocranial displaysia (CCD) a skeletal disorder characterised by gross dysgenesis of bones particularly those derived from intramembranous bone formation. A notable feature of the RUNX2 protein is the polyglutamine and polyalanine (23Q/17A) domain coded by a repeat sequence. Since none of the known mutations causing CCD characterised to date map in the glutamine repeat region, we hypothesised that Q-repeat mutations may be related to a more subtle bone phenotype. We screened subjects derived from four normal populations for Q-repeat variants. A total of 22 subjects were identified who were heterozygous for a wild type allele and a Q-repeat variant allele: (15Q, 16Q, 18Q and 30Q). Although not every subject had data for all measures, Q-repeat variants had a significant deficit in BMD with an average decrease of 0.7SD measured over 12 BMD-related parameters (p = 0.005). Femoral neck BMD was measured in all subjects (−0.6SD, p = 0.0007). The transactivation function of RUNX2 was determined for 16Q and 30Q alleles using a reporter gene assay. 16Q and 30Q alleles displayed significantly lower transactivation function compared to wild type (23Q). Our analysis has identified novel Q-repeat mutations that occur at a collective frequency of about 0.4%. These mutations significantly alter BMD and display impaired transactivation function, introducing a new class of functionally relevant RUNX2 mutants.

Business analytics (BA) systems are an important strategic investment for many organisations and can potentially contribute significantly to firm performance. Establishing strong BA capabilities is currently one of the major concerns of chief information officers. This research project aims to develop a BA capability maturity model (BACMM). The BACMM will help organisations to scope and evaluate their BA initiatives. This research-in-progress paper describes the current BACMM, relates it to existing capability maturity models and explains its theoretical base. It also discusses the design science research approach being used to develop the BACMM and provides details of further work within the research project. Finally, the paper concludes with a discussion of how the BACMM might be used in practice.

Universities in Australia, as with universities in the US and UK, are required more and more to adjust to student cohorts which represent a range of national, cultural and linguistic diversity. Many are concerned about the readiness of these students for study in an English-speaking medium and about their subsequent experiences and success. This presentation reports on the outcomes of “The English Language Growth” project, conducted across five Australian universities. This study, funded by the Australian Learning and Teaching Council (ALTC), represents the largest of its type in Australia. The project investigated the strategies that international students from non-English speaking backgrounds use to continue to improve their English once entering a university in Australia.
