Es realitza una breu introducció a la Química Supramolecular, la qual estudia les estructures i funcions de les associacions que resulten de la unió d'espècies moleculars a través d'enllaços intermoleculars, no covalents. El reconeixement molecular, reactivitat i transport són les funcions bàsiques de les espècies supramoleculars. El present treball tracta de la química supramolecular d'anions i del potential efecte catalític de complexos de Zinc amb lligands hexaazamacrocilics en reaccions de hidròlisis d'esters de fosfat. Els lligands descrits en aquest treball són lligands macrocíclics hexadentats formats per amines secundàries o terciàries enllaçades a cadenes alquíliques de dos o tres àtoms de carbonis formant dos braços, que estan unides per un espaiador, El conjunt de lligands que s'estudien permeten estudiar els diferents factors, geomètrics, electrònics i estèrics que controlen els fenòmens de reconeixement molecular. El treball descriu en detall els procediments de síntesi i la caracterització d'aquest conjunt de lligands. S'ha realitzat un estudi sistemàtic dels diferents factors que afecten als fenòmens de reconeixement molecular entre lligands macrocíclics (L), descrits anteriorment i substrats aniònics (S): fosfats, polifosfats i dicarboxilats. Els estudis s'han realitzat tant en dissolució aquosa, a partir de mesures potenciomètriques, com en estat sòlid per difracció de Raig-X a partir de la obtenció de les estructures cristal·lines dels complexos. La força de l'enllaç en els complexos ternaris H:L:S es racionalitza en termes d'enllaç per pont d'hidrogen, interaccions electrostàtiques i interaccions per -Stacking. Així mateix es racionalitza la importància de les dimensions i forma de la cavitat en el grau d'interacció. Actualment hi ha un gran interès en l'estudi del paper que juguen els ions metàl·lics en el centre actiu dels metal·loenzims hidrolítics tal com la carboxipeptidasa, que catalitza la hidròlisi d'aminoàcids C-terminals de substrats polipeptídics, l'anhidrasa carbònica, que catalitza la reacció d'hidratació del CO2 per donar hidrogencarbonat, la fosfatasa alcalina, que actua catalitzant la hidròlisi no específica de monoesters de fosfats a pH alcalí. El Zn(II) és un metall que es troba sovint formant part dels centres actius dels metal·loenzims i és el segon element de transició més abundant en els organismes vius. Per aquest motiu són útils els models sintètics que puguin actuar com a mimetitzadors de reaccions bioinorgàniques en les que els metal·loenzims juguen un paper com a catalitzadors. Els models sintètics són complexos de metalls de transició formats per un lligand o lligands en el seu entorn de coordinació més immediat que tinguin efectes electrònics i estèrics els més similar possible als que es produeixen en el centre actiu dels enzims. Donat la importància dels complexos de Zn(II) com a models biomimètics de metal·loenzims hidrolítics, s'han sintetitzat i caracteritzat complexos dinuclears de Zn(II) amb lligands hexaazamacrocíclics descrits anteriorment. S'ha realitzat un estudi de la seva activitat catalítica en la hidròlisi d'un ester activat, l'acetat de p-nitrofenil, un substrat característic que s'utilitza per analitzar l'activitat catalítica dels models biomimètics.

Chitosan is a biocompatible and biodegradable amino polysaccharide, which is soluble in aqueous solutions at pH < 6.5. It has been widely used for developing drug delivery systems because of its excellent mucoadhesive properties. Although many studies report on chitosan being mucoadhesive, the nature of interactions between chitosan and mucin remains poorly defined. Here, we have examined the role of primary amino groups and the role of electrostatic attraction, hydrogen bonding, and hydrophobic effects on aggregation of gastric mucin in the presence of chitosan. Reducing the number of amino groups through their half acetylation results in expansion of chitosan’s pH-solubility window up to pH 7.4 but also reduces its capacity to aggregate mucin. We demonstrated that electrostatic attraction forces between chitosan and gastric mucin can be suppressed in the presence of 0.2 mol/L sodium chloride; however, this does not prevent the aggregation of mucin particles in the presence of this biopolymer. The presence of 8 mol/L urea or 10% v/v ethanol in solutions also affects mucin aggregation in the presence of chitosan, demonstrating the role of hydrogen bonding and hydrophobic effects, respectively, in mucoadhesion.

The interaction between four flavonoids (catechin, epicatechin, rutin and quercetin) and bovine serum albumin (BSA) was investigated using tryptophan fluorescence quenching. Quenching constants were determined using the Stern-Volmer equation to provide a measure of the binding affinity between the flavonoids and BSA. The binding affinity was found to be strongest for quercetin, and ranked in the order quercetin>rutin>epicatechin=catechin. The pH in the range of 5 to 7.4 does not affect significantly (p<0.05) the association of rutin, epicatechin and catechin with BSA, but quercetin exhibited a stronger affinity at pH 7.4 than at lower pH (p<0.05). Quercetin has a total quenching effect on BSA tryptophan fluorescence at a molar ratio of 10:1 and rutin at approximately 25:1. However, epicatechin and catechin did not fully quench tryptophan fluorescence over the concentration range studied. Furthermore, the data suggested that the association between flavonoids and BSA did not change molecular conformation of BSA and that hydrogen bonding, ionic and hydrophobic interaction are equally important driving forces for protein-flavonoid association.

The ability to generate very stable assemblies via non-covalent interactions has enabled materials to be constructed that were not feasible via traditional covalent bond formation processes. A series of low molecular mass bisurethane and bisurea polymers have been developed that form stable self-assembled networks through hydrogen bonding interactions. Thermo-responsive polymers were generated by end-capping poly(ethylene-co-butylene) or polybutadiene chains with the bisurethane or bisurea motif. Microphase separation is observed via TEM and small-angle X-ray scattering (SAXS) for the modified pseudo polymers and significant differences in the temperature dependence of microphase separation are analysed via SAXS. The importance of the polarity of the end groups is manifested in distinct temperature-dependent microphase separation behaviour. Statistical approaches have been applied to examine amino acid pairing preferences within parallel beta-sheets. The main chain hydrogen bonding pattern in parallel beta-sheets means that, for each residue pair, only one of the residues is involved in main chain hydrogen bonding with the strand containing the partner residue. We call this the hydrogen bonded (HB) residue and the partner residue the non-hydrogen bonded (nHB) residue, and differentiate between the favorability of a pair and that of its reverse pair, e.g. Asn(HB)-Thr(nHB)versus Thr(HB)-Asn(nHB). Significantly (p < or = 0.000001) favoured pairings were rationalised using stereochemical arguments. For instance, Asn(HB)-Thr(nHB) and Arg(HB)-Thr(nHB) were favoured pairs, where the residues adopted favoured chi1 rotamer positions that allowed side-chain interactions to occur. In contrast, Thr(HB)-Asn(nHB) and Thr(HB)-Arg(nHB) were not significantly favoured, and could only form side-chain interactions if the residues involved adopted less favourable chi1 conformations. The favourability of hydrophobic pairs e.g. Ile(HB)-Ile(nHB), Val(HB)-Val(nHB) and Leu(HB)-Ile(nHB) was explained by the residues adopting their most preferred chi1 and chi2 conformations, which enabled them to form nested arrangements. Cysteine-cysteine pairs are significantly favoured, although these do not form intrasheet disulphide bridges. Interactions between positively and negatively charged residues were asymmetrically preferred: those with the negatively charged residue at the HB position were more favoured. This trend was accounted for by the presence of general electrostatic interactions, which, based on analysis of distances between charged atoms, were likely to be stronger when the negatively charged residue is the HB partner. The Arg(HB)-Asp(nHB) interaction was an exception to this trend and its favorability was rationalised by the formation of specific side-chain interactions. This research provides rules that could be applied to protein structure prediction, comparative modelling and protein engineering and design. Glycogen phosphorylase (GP) is currently exploited as a target for inhibition of hepatic glycogenolysis under high glucose conditions. Spirohydantoin of glucopyranose and N-acetyl-beta-D-glucopyranosylamine have been identified as the most potent inhibitors of GP that bind at the catalytic site. Four spirohydantoin and three beta-D-glucopyranosylamine analogs have been designed, synthesized and tested for inhibition of GP in kinetic experiments. Depending on the functional group introduced, the K(i) values varied from 16.5 microM to 1200 microM. In order to rationalize the kinetic results, we determined the crystal structures of the analogs in complex with GP. All the inhibitors bound at the catalytic site of the enzyme, by making direct and water-mediated hydrogen bonds with the protein and by inducing minor movements of the side chains of Asp283 and Asn284, of the 280s loop that blocks access of the substrate glycogen to the catalytic site, and changes in the water structure in the vicinity of the site. The differences observed in the Ki values of the analogs can be interpreted in terms of variations in hydrogen bonding and van der Waals interactions, desolvation effects, ligand conformational entropy, and displacement of water molecules on ligand binding to the catalytic site.

The structures of benzoic acid (C6H5COOH) and 2-hydroxybenzoic acid (C6H4OHCOOH) have been determined in the gas phase by electron diffraction using results from quantum chemical calculations to inform restraints used on the structural parameters. Theoretical methods (HF and MP2/6-311+G(d, p)) predict two conformers for benzoic acid, one which is 25.0 kJ mol(-1) (MP2) lower in energy than the other. In the low-energy form, the carboxyl group is coplanar with the phenyl ring and the O-H group eclipses the C=O bond. Theoretical calculations (HF and MP2/6-311+ G(d, p)) carried out for 2-hydroxybenzoic acid gave evidence for seven stable conformers but one low-energy form (11.7 kJ mol-1 lower in energy (MP2)) which again has the carboxyl group coplanar with the phenyl ring, the O-H of the carboxyl group eclipsing the C=O bond and the C=O of the carboxyl group oriented toward the O-H group of the phenyl ring. The effects of internal hydrogen bonding in 2-hydroxybenzoic acid can be clearly observed by comparison of pertinent structural parameters between the two compounds. These differences for 2-hydroxybenzoic acid include a shorter exocyclic C-C bond, a lengthening of the ring C-C bond between the substituents, and a shortening of the carboxylic single C-O bond.

Three new supramolecular assemblies of co-crystallized metal complexes and aliphatic dicarboxylic acids, {[Cu(pic)(2)(H2O)(2)](H(2)mal)}(n) (1), {[Cu(pic)(2)(H2O)(2)](H(2)mal)(2)(H2O)(2)}(n) (2) and {[Cu(pic)(2)(MeOH)](H(2)succ)}(n) (3) {Hpic = 2-picolinic acid, H(2)mal = malonic acid and H(2)succ = succinic acid} have been synthesized and characterized by X-ray single-crystal structure determination. The crystal packings of the complexes reveal that supramolecular associations of the monomeric complex units lead to the formation of layers through hydrogen bonding. In all the complexes, the dicarboxylic acid units connect the 2-D layers to act as pillars. The interaction between water molecules and the dicarboxylic acid plays an important role in the overall supramolecular assembly. (C) 2008 Elsevier Ltd. All rights reserved.

The solvent-induced transition between self-assembled structures formed by the peptide AAKLVFF is studied via electron microscopy, light scattering, and spectroscopic techniques. The peptide is based on a core fragment of the amyloid beta-peptide, KLVFF, extended by two alanine residues. AAKLVFF exhibits distinct structures of twisted fibrils in water or nanotubes in methanol. For intermediate water/methanol compositions, these structures are disrupted and replaced by wide filamentous tapes that appear to be lateral aggregates of thin protofilaments. The orientation of the beta-strands in the twisted tapes or nanotubes can be deduced from X-ray diffraction on aligned stalks, as well as FT-IR experiments in transmission compared to attenuated total reflection. Strands are aligned perpendicular to the axis of the twisted fibrils or the nanotubes. The results are interpreted in light of recent results on the effect of competitive hydrogen bonding upon self-assembly in soft materials in water/methanol mixtures.

The title compound, [Cu(C4H8N3O2)(2)]center dot 2C(5)H(9)NO, consists of a neutral copper complex, in which the Cu II centre coordinates to two bis(methoxycarbimido) aminate ligands, solvated by two molecules of 1-methylpyrrolidin-2-one. The complex is planar and centrosymmetric, with the Cu II centre occupying a crystallographic inversion centre and adopting approximately square-planar geometry. N-H center dot center dot center dot O hydrogen-bonding interactions exist between the amine NH groups of the ligands and the O atoms of the 1-methylpyrrolidin-2-one molecules. The associated units pack to form sheets.

A new layered ammonium manganese(II) diphosphate, (NH4)(2)[Mn-3(P2O7)(2)(H2O)(2)], has been synthesised under solvothermal conditions at 433 K in ethylene glycol and the structure determined at 293 K using single-crystal X-ray diffraction data (M-r = 584.82, monoclinic, space group P2(1)/a, a = 9.4610( 8), b = 8.3565( 7), c = 9.477(1) Angstrom, beta = 99.908(9) degrees, V = 738.07 Angstrom(3), Z = 2, R = 0.0351 and R-w = 0.0411 for 1262 observed data (I > 3(sigma(I))). The structure consists of chains of cis- and trans-edge sharing MnO6 octahedra linked via P2O7 units to form layers of formula [Mn3P4O14(H2O)(2)](2-) in the ab plane. Ammonium ions lie between the manganese-diphosphate layers. A network of interlayer and ammonium-layer based hydrogen bonding holds the structure together. Magnetic measurements indicate Curie - Weiss behaviour above 30 K with mu(eff) = 5.74(1) mu(B) and theta = -23(1) K, consistent with the presence of high-spin Mn2+ ions and antiferromagnetic interactions. However, the magnetic data reveal a spontaneous magnetisation at 5 K, indicating a canting of Mn2+ moments in the antiferromagnetic ground state. On heating (NH4)(2)[Mn-3(P2O7)(2)(H2O)(2)] in water at 433 K under hydrothermal conditions, Mn-5(HPO4)(2)(PO4)(2).4H(2)O, synthetic hureaulite, is formed.

The title compound, poly[[mu-cyanoureato-tri-mu-hydroxidodicopper(II)] dihydrate], {[Cu-2(C2H2N3O)(OH)(3)]center dot 2H(2)O}(n), is a new layered copper(II) hydroxide salt (LHS) with cyanoureate ions and water molecules in the interlayer space. The three distinct copper(II) ions have distorted octahedral geometry: one Cu (symmetry (1) over bar) is coordinated to six hydroxide groups (4OH + 2OH), whilst the other two Cu atoms (symmetries (1) over bar and 1) are coordinated to four hydroxides and two N atoms from nitrile groups of the cyanoureate ions (4OH + 2N). The structure is held together by hydrogen-bonding interactions between the terminal-NH2 groups and the central cyanamide N atoms of organic anions associated with neighbouring layers.

The title compound, potassium nickel(II) digallium tris-( phosphate) dihydrate, K[NiGa2(PO4)(3)(H2O)(2)], was synthesized hydrothermally. The structure is constructed from distorted trans-NiO4(H2O)2 octahedra linked through vertices and edges to GaO5 trigonal bipyramids and PO4 tetrahedra, forming a three-dimensional framework of formula [NiGa2(PO4)(3)(H2O)(2)](-). The K, Ni and one P atom lie on special positions (Wyckoff position 4e, site symmetry 2). There are two sets of channels within the framework, one running parallel to the [10 (1) over bar] direction and the other parallel to [001]. These intersect, forming a three-dimensional pore network in which the water molecules coordinated to the Ni atoms and the K+ ions required to charge balance the framework reside. The K+ ions lie in a highly distorted environment surrounded by ten O atoms, six of which are closer than 3.1 angstrom. The coordinated water molecules are within hydrogen-bonding distance to O atoms of bridging Ga-O-P groups.

Specific monomer sequences in aromatic copolyimides are recognized through their -stacking and hydrogen-bonding interactions with a sterically and electronically complementary molecular tweezer. These interactions enable the tweezer molecule to read monomer sequences comprising up to 27 aromatic rings by multiple adjacent binding to neighboring sites on the polymer chain.
