Simultaneous detection of cyclopiazonic acid and aflatoxin B1 by HPLC in methanol/water mobile phase

A simple procedure for the simultaneous detection of cyclopiazonic acid (CPA) and aflatoxin B1 from fungal extracts is presented, using a methanol and water mobile phase and fluorescence detection. This methodology has been tested with standard solutions of both mycotoxins CPA and Aflatoxin B1 and with methanolic extracts of Aspergillus section Flavi strains, previously characterized for their mycotoxin production profile. Previously available methodology required the use of two different chromatographic runs for these mycotoxins, with distinct columns and detectors (fluorescence detection with a post-column photochemical derivatization (PHRED) for aflatoxin B1 and UV detection for CPA). The proposed method detects both mycotoxins in a single run. Data from these assays will be presented and discussed.

In Portugal, maize is the cereal that involves more agriculture explorations. Aspergillus spp., among other species, are usually associated with this cereal, during drying and storage, making this commodity susceptible to mycotoxins (such as aflatoxins, ochratoxins, and cyclopiazonic acid). The aim of this study was to evaluate the mycotoxigenic potential of isolated Aspergillus strains from these maize samples and correlate it with the sampling place, the weather conditions, and local practices during drying and storage. The samples were collected between November 2008 and April 2009 in maize association of producers facilities in Coimbra, Santarém and Portalegre. The isolated strains were divided in three distinct groups, Aspergillus section Flavi, Aspergillus section Nigri and others Aspergillus. The preliminary results show that there are differences between the incidence of these groups in the three sampling places, especially in Coimbra, probably due to a lower mean temperatures and higher humidity levels. These data will be presented and discussed.

Mycotoxins are fungal secondary metabolites found in some agricultural commodities which are toxic for humans and animals in small amounts. Mycotoxins are a global problem which can be partially controlled through prevention strategies that can be applied along the food and feed chain production. However, when mycotoxin formation can not be avoided and they come to be present in commodities some remediation strategies can also be used to reduce its levels on products, its bioavailability or its toxic effects. Among these remediation strategies, the biological methods are recently holding a relevant position, being widely studied in the last years. As a result, a great number of microorganisms that can degrade or detoxify several mycotoxins and the application of some of them were reported. Moreover, several enzymes which mediate these biological processes were identified, being by themselves studied in order to develop new biotechnological approaches to control the mycotoxin problem on commodities. The main enzymes known to detoxify ochratoxin A, their action and their present application in order to counteract the referred problem are reviewed and critically assessed.

The occurrence of mycotoxigenic moulds such as Aspergillus, Penicillium and Fusarium in food and feed has an important impact on public health, by the appearance of acute and chronic mycotoxicoses in humans and animals, which is more severe in the developing countries due to lack of food security, poverty and malnutrition. This mould contamination also constitutes a major economic problem due the lost of crop production. A great variety of filamentous fungi is able to produce highly toxic secondary metabolites known as mycotoxins. Most of the mycotoxins are carcinogenic, mutagenic, neurotoxic and immunosuppressive, being ochratoxin A (OTA) one of the most important. OTA is toxic to animals and humans, mainly due to its nephrotoxic properties. Several approaches have been developed for decontamination of mycotoxins in foods, such as, prevention of contamination, biodegradation of mycotoxins-containing food and feed with microorganisms or enzymes and inhibition or absorption of mycotoxin content of consumed food into the digestive tract. Some group of Gram-positive bacteria named lactic acid bacteria (LAB) are able to release some molecules that can influence the mould growth, improving the shelf life of many fermented products and reducing health risks due to exposure to mycotoxins. Some LAB are capable of mycotoxin detoxification. Recently our group was the first to describe the ability of LAB strains to biodegrade OTA, more specifically, Pediococcus parvulus strains isolated from Douro wines. The pathway of this biodegradation was identified previously in other microorganisms. OTA can be degraded through the hydrolysis of the amide bond that links the L-β-phenylalanine molecule to the ochratoxin alpha (OTα) a non toxic compound. It is known that some peptidases from different origins can mediate the hydrolysis reaction like, carboxypeptidase A an enzyme from the bovine pancreas, a commercial lipase and several commercial proteases. So, we wanted to have a better understanding of this OTA degradation process when LAB are involved and identify which molecules where present in this process. For achieving our aim we used some bioinformatics tools (BLAST, CLUSTALX2, CLC Sequence Viewer 7, Finch TV). We also designed specific primers and realized gene specific PCR. The template DNA used came from LAB strains samples of our previous work, and other DNA LAB strains isolated from elderberry fruit, silage, milk and sausages. Through the employment of bioinformatics tools it was possible to identify several proteins belonging to the carboxypeptidase family that participate in the process of OTA degradation, such as serine type D-Ala-D-Ala carboxypeptidase and membrane carboxypeptidase. In conclusions, this work has identified carboxypeptidase proteins being one of the molecules present in the OTA degradation process when LAB are involved.

Lactic acid bacteria (LAB) play a key role in the biopreservation of a wide range of fermented food products, such as yogurt, cheese, fermented milks, meat, fish, vegetables (sauerkraut, olives and pickles), certain beer brands, wines and silage, allowing their safe consumption, which gave to these bacteria a GRAS (Generally Recognised as Safe) status. Besides that, the use of LAB in food and feed is a promising strategy to reduce the exposure to dietary mycotoxins, improving their shelf life and reducing health risks, given the unique mycotoxin decontaminating characteristic of some LAB. Mycotoxins present carcinogenic, mutagenic, teratogenic, neurotoxic and immunosuppressive effects over animals and Humans, being the most important ochratoxin A (OTA), aflatoxins (AFB1), trichothecenes, zearalenone (ZEA), fumonisin (FUM) and patulin. In a previous work of our group it was observed OTA biodegradation by some strains of Pediococcus parvulus isolated from Douro wines. So, the aim of this study was to enlarge the screening of the biodetoxification over more mycotoxins besides OTA, including AFB1, and ZEA. This ability was checked in a collection of LAB isolated from vegetable (wine, olives, fruits and silage) and animal (milk and dairy products, sausages) sources. All LAB strains were characterized phenotypically (Gram, catalase) and genotypically. Molecular characterisation of all LAB strains was performed using genomic fingerprinting by MSP- PCR with (GTG)5 and csM13 primers. The identification of the isolates was confirmed by 16S rDNA sequencing. To study the ability of LAB strains to degrade OTA, AFB1 and ZEA, a MRS broth medium was supplemented with 2.0 g/mL of each mycotoxin. For each strain, 2 mL of MRS supplemented with the mycotoxins was inoculated in triplicate with 109 CFU/mL. The culture media and bacterial cells were extracted by the addition of an equal volume of acetonitrile/methanol/acetic acid (78:20:2 v/v/v) to the culture tubes. A 2 mL sample was then collected and filtered into a clean 2 mL vial using PP filters with 0.45 m pores. The samples were preserved at 4 °C until HPLC analysis. Among LAB tested, 10 strains isolated from milk were able to eliminate AFB1, belonging to Lactobacillus casei (7), Lb. paracasei (1), Lb. plantarum (1) and 1 to Leuconostoc mesenteroides. Two strains of Enterococcus faecium and one of Ec. faecalis from sausage eliminated ZEA. Concerning to strains of vegetal origin, one Lb. plantarum isolated from elderberry fruit, one Lb. buchnerii and one Lb. parafarraginis both isolated from silage eliminated ZEA. Other 2 strains of Lb. plantarum from silage were able to degrade both ZEA and OTA, and 1 Lb. buchnerii showed activity over AFB1. These enzymatic activities were also verified genotypically through specific gene PCR and posteriorly confirmed by sequencing analysis. In conclusion, due the ability of some strains of LAB isolated from different sources to eliminate OTA, AFB1 and ZEA one can recognize their potential biotechnological application to reduce the health hazards associated with these mycotoxins. They may be suitable as silage inoculants or as feed additives or even in food industry.

A ocorrência de bolores micotoxigénicos pertencentes aos géneros Aspergillus, Penicillium e Fusarium em alimentos para consumo Humano e animal, tem um impacto importante sobre a saúde pública e constitui também um importante problema económico. Isto é devido à síntese por este tipo de fungos filamentosos de metabolitos altamente tóxicos conhecidos como micotoxinas. A maioria das micotoxinas são substâncias cancerígenas, mutagénicas, neurotóxicas e imunossupressoras, sendo a ocratoxina A (OTA) uma das mais importantes. A OTA é uma micotoxina, tóxica para os animais e Humanos principalmente devido às suas propriedades nefrotóxicas. Alguns grupos de bactérias gram positivas nomeadamente as bactérias do ácido láctico (BAL) são capazes de controlar o crescimento de fungos, melhorando e aumentando a vida útil de muitos produtos fermentados e, assim, reduzir os riscos para a saúde provocados pela exposição às micotoxinas. Algumas BAL são, também, capazes de destoxificar certas micotoxinas. Em trabalhos anteriores do nosso grupo foi observada a biodegradação da OTA por estirpes de Pediococcus parvulus isoladas de vinhos do Douro. Assim, com este trabalho, pretendeu-se compreender com maior detalhe o processo de biodegradação da OTA pelas referidas estirpes e identificar quais as enzimas que estão associadas à sua biodegradação. Para atingir este objetivo utilizaram-se algumas ferramentas ioinformáticas (BLAST, CLUSTALX2, CLC Sequence Viewer 7, Finch TV), desenharam-se primers específicos e realizaram-se PCR específicos para os genes envolvidos. Através da utilização de ferramentas de bioinformática, foi possível identificar várias proteínas que pertencem à família das carboxipeptidases e que podem eventualmente participar no processo da degradação da OTA, tais como D-Ala-D-Ala carboxipeptidase serínica e carboxipeptidase membranar. Estas BAL podem desempenhar um papel importante na destoxificação da OTA, sendo as carboxipeptidases uma das enzimas envolvidas na sua biodegradação.

OBJECTIVE: To assess the effect of a new feed soy product fermented by Enterococcus faecium and Lactobacillus jugurti on the serum lipid levels of rabbits with induced hypercholesterolemia. METHODS: Thirty-two rabbits were divided into 4 groups as follows: 1) control (C); 2) hypercholesterolemic (H); 3) hypercholesterolemic + fermented product (HPF); and 4) control + fermented product (CPF). The H and HPF groups were fed with a diet with 0.15% (p/p) cholesterol in the first 15 days. C and CPF groups received regular food preparation. The HPF and CPF groups received 10 mL daily of the fermented 30 days. Blood samples were drawn at the beginning of the study and at the 15th and 30th days. Concentrations of total cholesterol, HDL-cholesterol, and triglycerides were analyzed. RESULTS: After 15 days, the HPF group showed a total cholesterol concentration lower (18.4%) than that of the H group (p=0.05), but this difference disappeared after 30 days. No change was observed in total cholesterol levels of C and CPF groups. After 15 days, the HDL-cholesterol was higher (17.8%) in the HPF group, but the triglyceride levels remained unchanged in all groups during the same period of time. CONCLUSION: The soy fermented product caused an 18.4% reduction in total cholesterol and a 17.8% increase in the HDL-fraction. It may, therefore, be a possible coadjutor in the treatment of hypercholesterolemia.

The influence of the feed composition upon the actual degrees of separation attained at the top and bottom sections of a thermogravitational column is discussed using the classical phenomenological theory of Furry, Jones, and Onsager. It is shown that, except for a feed composition of C 0 = 0.5 (mass fraction), the separation profile is nonsymmetric, i.e., the separations in the top and bottom sections of the column are nonsymmetric with respect to the feed composition, the asymmetry increasing as the feed composition moves away from C 0 = 0.5. An equation for the determination of the optimum feed location as a function of the feed composition is derived.
Operation modes for the electric vehicle in smart grids and smart homes : present and proposed modes

This paper presents the main operation modes for an electric vehicle (EV) battery charger framed in smart grids and smart homes, i.e., are discussed the present-day and are proposed new operation modes that can represent an asset towards EV adoption. Besides the well-known grid to vehicle (G2V) and vehicle to grid (V2G), this paper proposes two new operation modes: Home-to-vehicle (H2V), where the EV battery charger current is controlled according to the current consumption of the electrical appliances of the home (this operation mode is combined with the G2V and V2G); Vehicle-for-grid (V4G), where the EV battery charger is used for compensating current harmonics or reactive power, simultaneously with the G2V and V2G operation modes. The vehicle-to-home (V2H) operation mode, where the EV can operate as a power source in isolated systems or as an off-line uninterruptible power supply to feed priority appliances of the home during power outages of the electrical grid is presented in this paper framed with the other operation modes. These five operation modes were validated through experimental results using a developed 3.6 kW bidirectional EV battery charger prototype, which was specially designed for these operation modes. The paper describes the developed EV battery charger prototype, detailing the power theory and the voltage and current control strategies used in the control system. The paper presents experimental results for the various operation modes, both in steady-state and during transients.

Los materiales lignocelulósicos residuales de las actividades agroindustriales pueden ser aprovechados como fuente de lignina, hemicelulosa y celulosa. El tratamiento químico del material lignocelulósico se debe enfrentar al hecho de que dicho material es bastante recalcitrante a tal ataque, fundamentalmente debido a la presencia del polímero lignina. Esto se puede lograr también utilizando hongos de la podredumbre blanca de la madera. Estos producen enzimas lignolíticas extracelulares fundamentalmente Lacasa, que oxida la lignina a CO2. Tambien oxida un amplio rango de sustratos ( fenoles, polifenoles, anilinas, aril-diaminas, fenoles metoxi-sustituídos, y otros), lo cual es una buena razón de su atracción para aplicaciones biotecnológicas. La enzima tiene potencial aplicación en procesos tales como en la delignificación de materiales lignocelulósicos y en el bioblanqueado de pulpas para papel, en el tratamiento de aguas residuales de plantas industriales, en la modificación de fibras y decoloración en industrias textiles y de colorantes, en el mejoramiento de alimentos para animales, en la detoxificación de polutantes y en bioremediación de suelos contaminados. También se la ha utilizado en Q.Orgánica para la oxidación de grupos funcionales, en la formación de enlaces carbono- nitrógeno y en la síntesis de productos naturales complejos. HIPOTESIS: Los hongos de podredumbre blanca, y en condiciones óptimas de cultivo producen distintos tipos de enzimas oxidasas, siendo las lacasas las más adecuadas para explorarlas como catalizadores en los siguientes procesos: Delignificación de residuos de la industria forestal con el fin de aprovechar tales desechos en la alimentación animal. Decontaminación/remediación de suelos y/o efluentes industriales. Se realizarán los estudios para el diseño de bio-reactores que permitan responder a las dos cuestiones planteadas en la hipótesis. Para el proceso de delignificación de material lignocelulósico se proponen dos estrategias: 1- tratar el material con el micelio del hongo adecuando la provisión de nutrientes para un desarrollo sostenido y favorecer la liberación de la enzima. 2- Utilizar la enzima lacasa parcialmente purificada acoplada a un sistema mediador para oxidar los compuestos polifenólicos. Para el proceso de decontaminación/remediación de suelos y/o efluentes industriales se trabajará también en dos frentes: 3) por un lado, se ha descripto que existe una correlación positiva entre la actividad de algunas enzimas presentes en el suelo y la fertilidad. En este sentido se conoce que un sistema enzimático, tentativamente identificado como una lacasa de origen microbiano es responsable de la transformación de compuestos orgánicos en el suelo. La enzima protege al suelo de la acumulación de compuestos orgánicos peligrosos catalizando reacciones que involucran degradación, polimerización e incorporación a complejos del ácido húmico. Se utilizarán suelos incorporados con distintos polutantes(por ej. policlorofenoles ó cloroanilinas.) 4) Se trabajará con efluentes industriales contaminantes (alpechínes y/o el efluente líquido del proceso de desamargado de las aceitunas). The lignocellulosic raw materials of the agroindustrial activities can be taken advantage as source of lignin, hemicellulose and cellulose. The chemical treatment of this material is not easy because the above mentioned material is recalcitrant enough to such an assault, due to the presence of the lignin. This can be achieved also using the white-rot fungi of the wood. It produces extracellular ligninolitic enzymes, fundamentally Laccase, which oxidizes the lignin to CO2. The enzyme has application in such processes as in the delignification of lignocellulosic materials and in the biobleaching of fibers for paper industry, in the treatment of waste water of industrial plants, in the discoloration in textile industries, in the improvement of food for ruminants, in the detoxification of polutants and in bioremediation of contaminated soils. HYPOTHESIS: The white-rot fungi produce different types of enzymes, being the laccases the most adapted to explore them as catalysts in the following processes: Delignification of residues of the forest industry in order to take advantage of such waste in the animal feed. Decontamination of soils and / or waste waters. The studies will be conducted for the design of bio reactors that allow to answer to both questions raised in the hypothesis. For the delignification process of lignocellulosic material they propose two strategies: 1- to treat the material with the fungi 2-to use the partially purified enzyme to oxidize the polyphenolic compounds. For the soil and/or waste water decontamination process, we have: 3- Is know that the enzyme protects to the soil of the accumulation of organic dangerous compounds catalyzing reactions that involve degradation, polymerization and incorporation to complexes of the humic acid. There will be use soils incorporated into different pollutants. 4- We will work with waste waters (alpechins or the green olive debittering effluents.

La introducción de nuevos recursos genéticos es esencial en la evolución de la agricultura destinada a la producción de materia prima de alta calidad. La evaluación de nuevo germoplasma, con el objetivo de su difusión comercial o incorporación a programas de mejoramiento, se realiza a través de ensayos comparativos de rendimiento en diferentes ambientes. Las forrajeras anuales de invierno son casi insustituibles para mantener la cadena forrajera en la región pampeana subhúmeda seca y semiárida. En la actualidad tanto triticales como tricepiros son alternativas muy promisorias para su utilización como doble propósito, así como para explorar la posibilidad de producir harinas diferenciadas para uso en la industria de la alimentación humana. En el país, los triticales se han difundido para pastoreo y se requiere de nuevos cultivares. Los tricepiros se trabajan exclusivamente en la Argentina y es necesario desarrollar germoplasma. Se planea identificar germoplasma superior mediante selección en introducciones y cruzamientos propios, ensayos comparativos de pasto y grano, estudiar la estabilidad citológica mediante análisis cromosómicos y genómicos y caracterizar líneas avanzadas por descriptores morfológicos.The introduction of new genetic resources is essential in the agriculture evolution, especially when high quality production is the main objective. New germoplasm must be tested several years in order to be part of breeding programs or to be commercialized due to environmental differences among years. In dry subhumid and semiarid pampeana region, annual winter feed crops are necessary in order to have forage production during winter. Nowadays Triticales and Tricepiros are promising alternatives to be used as double purpose for animal feed and fodder, as well as to obtain differenced flour to human food. In Argentina, Triticale has spread for forage uses and new cultivars are required. Tricepiros are sown only in this country; therefore there is a need to develop Tricepiro germoplasm. The outline includes: identify superior germoplasm in introductions and own crosses through selection; establish comparative forage and grain trials; cytological estability study using chromosomal and genomic analysis and characterize advanced lines through morphological features.

La quinoa (Chenopodium quinoa Willd), es un pseudocereal originario de la región Andina. Fue utilizada como alimento básico por los pueblos nativos. La quinoa, la papa y el maíz constituyeron el trinomio base de la alimentación indígena de este continente. La colonización española fue desplazando su cultivo a favor del trigo europeo y otros cereales, quedando reducida a las zonas altas de la región andina. La Quínoa ha adquirido una considerable atención en los últimos tiempos, principalmente por la calidad de sus proteínas y la ausencia de gluten en ella. Su empleo está ampliamente difundido en los países andinos, especialmente Bolivia y Perú, con un notable crecimiento de la superficie sembrada. En nuestro país la explotación de este cultivo se ubica principalmente en las provincias norteñas de Salta y Jujuy. En estos últimos años se ha reivindicado su cultivo y los granos privados de saponinas son considerados como un excelente alimento, reconocido por la OMS, la FAO y la NASA. Además de la calidad de sus lípidos y vitaminas, y al elevado contenido en almidón, la quinoa posee una proteína de excelente calidad nutricional y libre de gluten, lo que hace a este grano especialmente indicado para la alimentación de personas que sufren de la enfermedad celíaca o del síndrome de intestino irritado. El presente proyecto está orientado al aprovechamiento integral del grano de quinoa. Es nuestra intensión aquí, demostrar que dicho grano, cultivado en la provincia de Córdoba, permitirá elaborar productos alimenticios asi como también derivados de su industrialización. Para este objetivo se cuenta con las instalaciones de la Planta Piloto del Instituto de Ciencia y Tecnología de los Alimentos (ICTA), de la UNC, así como de intrumental moderno y acorde, como HPLC, GC, Espectrofotómetro UV-Vis, rotavapores de laboratorio e industrial, cámara fría, balanzas analíticas y de precisión, muflas, estufas, molinos y tamices, así como también, contamos con profesionales, algunos de ellos realizando su tesis doctoral en este tema. En cuanto a los objetivos que se persiguen, se espera obtener productos tales como sopas, papillas, productos para panadería y galletería y salsas. En el plano industrial, se pretende elaborar concentrados proteicos, almidón y saponinas. Como se dijo más arriba, a nivel internacional la quinoa ha comenzado a extender sus fronteras, y es así que hoy el principal productor mundial de este grano, Bolivia, destina un porcentaje importante de su producción a la exportación. La creciente demanda mundial de quinoa a hecho que se constituya en un cultivo estratégico y de alto valor, con precios internacionales que rondan los U$S 1200 la tonelada. Si a esto unimos que la planta presenta una gran resistencia a la sequía, que se adapta bien a terrenos salitrosos, arenosos y pobres, podemos comprender la importancia que adquiere para nuestra provincia, toda vez que en la misma existen zonas geográficas potencialmente aptas para su cultivo. Quinoa (Chenopodium quinoa Willd) is a pseudocereal originating in the Andean region. It was used as a staple food by native peoples. Quinoa, potatoes and corn were the tree most important indigenous staple food to this part of South America. Spanish colonization was marginalized cultivation in favor of European wheat and other grains, displacing it to the highlands of the Andean region. Quinoa has recently gained considerable attention, mainly by its protein quality and lack of gluten. Its use is widespread in the Andean countries, especially Bolivia and Peru, with a notable increase in plantings. In our country, the exploitation of this crop is located mainly in the northern provinces of Salta and Jujuy. In recent years its cultivation has been promoted, and the grains once free of saponins are considered an excellent food, recognized by WHO, FAO and NASA. In addition to its lipid and vitamins, and high starch contain, quinoa protein has an excellent nutritional value and it is free of gluten, making it particularly suitable for this grain to feed people with celiac disease or irritable bowel syndrome. This project aims at an integral development of quinoa grain. It is our intention here to demonstrate that this grain grown in the province of Córdoba, can produce food products resulting from local industrialization. This team has access to the facilities of the Pilot Plant of the Institute of Science and Food Technology (ICTA) of the UNC, and the modern equipments in it, as HPLC, GC, UV-Vis spectrophotometer, laboratory and industrial rotary evaporators, cold storage, analytical and precision balances, flasks, ovens, grinders and screens. Also, we have an important professional staff, some of them doing their thesis on this subject. With regard to the objectives pursued, we expect to obtain products such as soups, baby food, bakery products and biscuits and sauces. At the industrial level, it aims at producing protein concentrates, starch and saponins.

Surgical procedures such as osteotomy and hip replacement involve the cutting of bone with the aid of various manual and powered cutting instruments including manual and powered bone saws. The basic mechanics of bone sawing processes are consistent with most other material sawing processes such as for wood or metal. Frictional rubbing between the blade of the saw and the bone results in the generation of localised heating of the cut bone. Research studies have been carried out which consider the design of the bone saw which deals with specifics of the saw teeth geometry and research which examines the effect of drilling operations on heating of the bone has shown that elevated temperatures will occur from frictional overheating. This overheating in localised areas is known to have an impact on the rate of healing of the bone post operation and the sharpness life of the blade. The purpose of this study was to measure the temperature at three zones at fixed intervals of 3mm, 6mm, and 9mm away from the cutting zone. It should be noted that it was the first time that this measurement technique was used to measure the temperature gradient through the bone specimen thereby establishing the extent to which clinicians are experiencing thermal injury during sawing of bone while using a reciprocating saw. The effect of various cutting feed rate on temperature elevation was also investigated in this research. The results showed that there will be a region of bone at least 9mm either side of the cutting blade experiencing thermal injury as temperatures in this region exceeded the threshold temperature of 44°C for necrosis (cell death).
