Poly(ß,L-malic acid) (PMLA) was made to interact with the cationic anticancer drug Doxorubicin (DOX) in aqueous solution to form ionic complexes with different compositions and an efficiency near to 100%. The PMLA/DOX complexes were characterized by spectroscopy, thermal analysis, and scanning electron microscopy. According to their composition, the PMLA/DOX complexes spontaneously self-assembled into spherical micro or nanoparticles with negative surface charge. Hydrolytic degradation of PMLA/DOX complexes took place by cleavage of the main chain ester bond and simultaneous release of the drug. In vitro drug release studies revealed that DOX delivery from the complexes was favored by acidic pH and high ionic strength

Nutritional factors, especially the protein and fat content of the diet, may change pancreatic morphology after ethanol induced injury. This study was performed to delineate the combined effects of a low fat diet and longterm ethanol ingestion on the rat pancreas. Male Sprague-Dawley rats were maintained with five different diets for 12 weeks and the pancreas removed on the day they were killed. Rats fed a very low fat diet without ethanol (5% of total calories as lipid) developed malnutrition, pancreatic steatosis, and reduction in zymogen granules content. Animals fed a 35% lipid diet with ethanol also developed pancreatic steatosis but changes in zymogen granules content were not detected. Both malnutrition and longterm ethanol consumption increased pancreatic cholesterol ester content, and their effects were additive. Pancreatic steatosis was accompanied with hypercholesterolaemia. Amylase, lipase, and cholesterol esterase content were reduced in malnourished rats; but longterm ethanol ingestion, regardless of the nutritional state, increased lipase content and decreased amylase. It is suggested that high serum cholesterol concentrations and increased pancreatic lipase activity could cause accumulation of cholesterol esters in acinar cells. Fat accumulation in the pancreas has been reported as the earliest histopathological feature in alcoholic patients and may be responsible for cytotoxic effects on the acinar cells at the level of the cell membrane. Although it is difficult to extrapolate results in this animal study to the human situation, the results presented in this work might explain the higher incidence of pancreatitis is malnourished populations as well as in alcoholic subjects that is reported in dietary surveys.

O paclobutrazol é utilizado para o manejo da produção de manga na maioria dos pomares, com aplicação no solo, o que contribui para a maior persistência no ambiente. Assim, este trabalho teve como objetivo avaliar o efeito de reguladores de crescimento vegetal aplicado via foliar, prohexadione-Ca, etil-trinexapac e cloreto de chlormequat, e via solo, paclobutrazol, no controle da brotação vegetativa e na floração de mangueiras 'Kent'. O experimento foi conduzido na região do Vale do São Francisco, em Petrolina-PE, no período de dezembro de 2007 a julho de 2008, em mangueiras cultivadas no espaçamento de 8 m nas entrelinhas e 5 m entre plantas. Foram avaliados os efeitos dos tratamentos com reguladores vegetais aplicados via foliar e via solo, com relação ao vigor (número, comprimento e número de folhas dos novos fluxos vegetativos) e à floração (percentagem e comprimento das panículas). Verificou-se que o tratamento com paclobutrazol, combinado com o prohexadione-Ca, foi mais eficiente no controle do crescimento vegetativo dos ramos da mangueira 'Kent', mas não com relação à floração, em que não foram observadas diferenças siginificativas entre os tratamentos.

Työssä tutkittiin liimauksen reversiota ja häviämistä erilaisilla hydrofobiliimoilla alkaalisessa paperinvalmistuksessa käytettävien täyteaineiden kanssa. Liimauksen reversiota ja häviämistä arvioitiin arkkimuotilla valmistetuista koearkeista mittaamalla hydrofobiliimautuneisuutta kuvaavia ominaisuuksia kuten raakareunan absorptio ja kontaktikulma. Liimauksen reversiota kiihdytettiin lämpökäsittelyllä ja UV-säteilytyksellä. Liimauksen häviämistä tutkittiin muuttamalla viiraveden alkaalisuutta ja käyttämällä kalsiumkarbonaattia täyteaineena. Verrattaessa käytettyjä hydrofobiliimoja ASA1+AKD -liimayhdistelmällä ja AKD -liimauksella saavutettiin paperille paras liimautuneisuus. ASA2 -liimauksella saavutettiin paperille parempi liimautuneisuus kuin ASA1 -liimalla. Kalsiumkarbonaatilla täytetyillä arkeilla mitattiin korkeampi liimautuneisuus kuin kaoliinilla täytetyillä arkeilla. Kokeellisessa osassa tutkittiin myös 4 tuntia kestävän 105°C lämpökäsittelyn ja UV-säteilytyksen sekä viiraveden alkaalisuuden vaikutusta hydrofobiliimauksen reversioon ja häviämiseen. Liimauksen reversiota aikaansai UV-säteily ja lämpökäsittely. UV-säteily ja lämpökäsittely näyttävät aikaansaavan kovalenttisen esterisidoksen katkeamisen tai hydrolysoitumisen liimamolekyylin ja selluloosan karboksyyliryhmän välillä. Liimauksen reversiota havaittiin jokaisella hydrofobiliimalla. Liimauksen häviämistä aikaansai korkea viiraveden alkaalisuus (520 ppm CaCO3). Liimauksen häviämisen aiheuttamaa alhaisempaa liimautuneisuutta havaittiin myös viiraveden alkaalisuuden ollessa normaalia tasoa (250 ppm CaCO3), kun täyteaineena käytettiin saostettua kalsiumkarbonaattia.

Triheptanoin-enriched diets have been successfully used in the experimental treatment of various metabolic disorders. Maximal therapeutic effect is achieved in the context of a ketogenic diet where triheptanoin oil provides 30-40% of the daily caloric intake. However, pre-clinical studies using triheptanoin-rich diets are hindered by the difficulty of administering to laboratory animals as a solid foodstuff. In the present study, we successfully synthesized triheptanoin to the highest standards of purity from glycerol and heptanoic acid, using sulfonated charcoal as a catalyst. Triheptanoin oil was then formulated as a solid, stable and palatable preparation using a ketogenic base and a combination of four commercially available formulation agents: hydrophilic fumed silica, hydrophobic fumed silica, microcrystalline cellulose, and talc. Diet compliance and safety was tested on C57Bl/6 mice over a 15-week period, comparing overall status and body weight change. Practical applications: This work provides a complete description of (i) an efficient and cost-effective synthesis of triheptanoin and (ii) its formulation as a solid, stable, and palatable ketogenic diet (triheptanoin-rich; 39% of the caloric intake) for rodents. Triheptanoin-rich diets will be helpful on pre-clinical experiments testing the therapeutic efficacy of triheptanoin in different rodent models of human diseases. In addition, using the same solidification procedure, other oils could be incorporated into rodent ketogenic diet to study their dosage and long-term effects on mammal health and development. This approach could be extremely valuable as ketogenic diet is widely used clinically for epilepsy treatment.

Este trabalho teve por objetivo a caracterização molecular de 13 acessos de Psidium spp. (Myrtaceae) identificados previamente quanto à reação ao nematoide da goiabeira. A extração do DNA das amostras foi executada conforme o protocolo de Shillito e Saul (1988). Os marcadores moleculares do tipo fAFLP, foram obtidos utilizando-se do 'fAFLP Regular Plant Genomes Fingerprinting Kit' (Applied Biosystems do Brasil Ltda.) onde foram testadas 24 combinações seletivas de primers, das quais 18 apresentaram amplificação que gerou 272 marcadores polimórficos. Para a análise dos marcadores, foram utilizados os softwares GeneScan (ABI Prism versão 1.0) e Genotyper (ABI Prism versão 1.03), e os dados coletados foram transformados em matriz binária que foi analisada no software PAUP (Phylogenetic Analysis Using Parcimony - versão 3.01). Foram também calculados índices de distância genética intra e interespecífica entre os materiais. Verificou-se que os marcadores AFLP foram eficientes na discriminação dos acessos entre si, bem como apontou similaridade genética entre os acessos identificados como resistentes ao nematoide Meloidogyne enterolobii, característica esta passível de exploração no futuro.

O rambutan é uma frutífera exótica que apresenta alto potencial de mercado, e suas mudas podem ser obtidas por sementes ou vegetativamente. A produção de mudas via sementes é rotineiramente feita no Estado de São Paulo, tendo-se alta variabilidade no pomar, além de demorar mais tempo para entrar em produção. Embora caracteres morfológicos sejam amplamente usados na diferenciação de variedades, as técnicas moleculares permitem a comparação e a identificação genética dos materiais. Diante disso, o presente trabalho foi realizado, comparando progênies e plantas-matrizes de rambutan, por fAFLP. As análises foram realizadas no Laboratório de Bioquímica de Microrganismos e Plantas, do Departamento de Tecnologia - Faculdade de Ciências Agrárias e Veterinárias - UNESP - Câmpus de Jaboticabal-SP, utilizando 06 plantas de rambutan, denominadas: A; B; C; D; E e F. Foram coletadas folhas de 15 plântulas oriundas de cada planta-matriz e realizou-se a extração de DNA, sendo as amostras quantificadas em biofotômetro, e os marcadores fAFLP, obtidos de acordo com o protocolo AFLP Plant Mapping Protocol (Applied Biosystems), utilizando as combinações de pares de primers: ACG/CAC; ACT/CAT; ACA/CTT e ACC/CTT. Pode ser concluído que o uso de marcadores moleculares é eficiente na distinção de materiais e na obtenção de distância genética; não é recomendada a obtenção de mudas via sementes quando a finalidade é a de instalação de pomar comercial.

Diplomityön tavoitteena oli kehittää prosessi, jolla rypsiöljystä voidaan valmistaa dieselöljyn korvaavaa biodieseliä. Työssä tutustuttiin raaka-aineiden sekä sivu- ja lopputuotteiden aine- ja laatuominaisuuksiin, biodieselin käytettävyyteen ja käytössä oleviin valmistusprosesseihin. Työn aikana kehitettiin uusi tekniikka biodieselin tuottamiseen. Diplomityön tuloksena syntyi pilot-laitteen suunnitelma, jonka mukaan laite rakennettiin. Pilot-laitteen koeajot varmistivat tekniikan toimivuuden ja lopputuote täyttää biodieselille asetetut vaatimukset. Menetelmä tarvitsee vielä jatkokehittelyä tuotantokustannuksien alentamiseksi.

Many strategies for treating diseases require the delivery of drugs into the cell cytoplasm following internalization within endosomal vesicles. Thus, compounds triggered by low pH to disrupt membranes and release endosomal contents into the cytosol are of particular interest. Here, we report novel cationic lysine-based surfactants (hydrochloride salts of Nε- and Nα-acyl lysine methyl ester) that differ in the position of the positive charge and the length of the alkyl chain. Amino acid-based surfactants could be promising novel biomaterials in drug delivery systems, given their biocompatible properties and low cytotoxic potential. We examined their ability to disrupt the cell membrane in a range of pH values, concentrations and incubation times, using a standard hemolysis assay as a model of endosomal membranes. Furthermore, we addressed the mechanism of surfactant-mediated membrane destabilization, including the effects of each surfactant on erythrocyte morphology as a function of pH. We found that only surfactants with the positive charge on the α-amino group of lysine showed pH-sensitive hemolytic activity and improved kinetics within the endosomal pH range, indicating that the positive charge position is critical for pH-responsive behavior. Moreover, our results showed that an increase in the alkyl chain length from 14 to 16 carbon atoms was associated with a lower ability to disrupt cell membranes. Knowledge on modulating surfactant-lipid bilayer interactions may help us to develop more efficient biocompatible amino acid-based drug delivery devices.

Introducción: La incidencia de la presentación fetal podálica en España es del 3,8%. La moxibustión es parte integral de la milenaria medicina tradicional china. La técnica consiste en la colocación de un palillo incandescente de Artemisia vulgaris a unos pocos centímetros del punto de acupuntura Zhiyin (punto 67 del meridiano de vejiga, ubicado en la base externa de la uña del quinto dedo del pie). Los objetivos son: 1. Conocer el porcentaje de fetos que han rotado a cefálica mediante la moxibustión en gestantes que presenten una malposición fetal a partir de las 32 semanas de embarazo; 2. Identificar las complicaciones materno-fetales en la aplicación de la técnica. Sujetos: Se estudiaron 18 gestantes de más de 32 semanas de embarazo. Material y métodos: Mediante un estudio descriptivo de intervención, analizamos el porcentaje de fetos que rotaron a presentación cefálica. Resultados: Quince gestantes (83,9%) realizaron el tratamiento adecuadamente y las 3 restantes (16,7%) lo realizaron de manera ocasional y terminaron con una cesárea electiva por nalgas. Conclusiones: Parece razonable concluir que el cumplimiento del tratamiento influye en el tipo de parto (χ2= 12,600; gl= 1; p= 0,000), aunque no modifica el Apgar del recién nacido. Esta técnica se presenta como una alternativa económica, segura, sencilla y práctica para la versión fetal de la presentación podálica.
