La vid silvestre se considera como el ancestro autóctono de las vides cultivadas y una enorme reserva genética en peligro de extinción. La prospección llevada a cabo entre 2003 y 2004 permitió catalogar 51 localizaciones de vides silvestres españolas, la mayoría de ellas ubicadas en riberas de ríos. Estos ejemplares se incluyeron en el Banco de Germoplasma de la Finca "El Encín" (BGVCAM - Alcalá de Henares, Madrid, España). En primer lugar, se caracterizó la cantidad y la distribución de su diversidad genética utilizando 25 loci empleando microsatélites nucleares (SSR). Hemos analizado también la posible coexistencia en el hábitat natural de vides silvestres con vides cultivadas naturalizadas y portainjertos. De este modo, los análisis fenotípicos y genéticos identificaron el 19% de las muestras recogidas como derivadas de genotipos cultivados, siendo, o bien vides cultivadas naturalizadas o genotipos híbridos derivados de cruces espontáneos entre vides silvestres y cultivadas. La diversidad genética de las poblaciones de vides silvestres fue similar a la observada en el grupo de las cultivadas. El análisis molecular mostró que el germoplasma de cultivadas y silvestres es genéticamente divergente con bajo nivel de introgresión. Se ha identificado cuatro grupos genéticos, con dos de ellos fundamentalmente representados por los genotipos de vides cultivadas y dos por las accesiones silvestres. El análisis de los vínculos genéticos entre las vides silvestres y cultivadas podría sugerir una contribución genética de las accesiones silvestres españolas a las actuales variedades occidentales. En segundo lugar, se realizó un profundo estudio morfológico "ex situ " y se contrastaron con los resultados de la caracterización realizada en 182 variedades comerciales españolas de la misma colección. Todos los individuos silvestres mostraron diferencias morfológicas con Vitis vinifera L subsp. vinifera, pero no se encontraron diferencias significativas dentro Vitis vinifera L. subsp. sylvestris, ni por localización geográfica ni por sexo. Los resultados de este estudio describen las principales características morfológicas de las vides silvestres españolas y sus rasgos diferenciales con su pariente cultivada. Por último, se analizó la composición antociánica presente en 21 accesiones de vides silvestres de la Península Ibérica conservadas en el BGVCAM de la Finca "El Encín" y seleccionadas basándose en diferencias ampelográficas y caracterización molecular. La concentración de antocianinas es similar la encontrad en vides cultivadas con destino a la vinificación. Las accesiones estudiadas mostraron una variabilidad considerable en su perfil antociánico y fue posible distinguir varios grupos. Sin embargo, la presencia de material silvestre con perfiles antociánicos poco comunes o inexistentes en variedades españolas, sugiere que la variabilidad genética relacionada con antocianinas en poblaciones españolas de vides silvestres podría ser más alta que la de variedades cultivadas comúnmente consideradas de origen español. ABSTRACT The wild grapevine is considered an autochthonous relative of cultivated vines and a huge gene pool endangered in Europe. Prospecting carried out between 2003 and 2004 enabled to inventory 51 Spanish sites with wild grapevines, most of them located near rivers. These individuals were grafted in the collection of "El Encín" (BGVCAM - Alcalá de Henares, Madrid, Spain). Firstly, werw characterized the amount and distribution of their genetic diversity using 25 nuclear SSR loci. We have also analysed the possible coexistence in the natural habitat of wild grapevines with naturalized grapevine cultivars and rootstocks. In this way, phenotypic and genetic analyses identified 19% of the collected samples as derived from cultivated genotypes, being either naturalized cultivars or hybrid genotypes derived from spontaneous crosses between wild and cultivated grapevines. The genetic diversity of wild grapevine populations was similar than that observed in the cultivated group. The molecular analysis showed that cultivated germplasm and wild germplasm are genetically divergent with low level of introgression. We identified four genetic groups, with two of them fundamentally represented among cultivated genotypes and two among wild accessions. The analyses of genetic relationships between wild and cultivated grapevines could suggest a genetic contribution of wild accessions from Spain to current Western cultivars. Secondly, a morphological study was done "ex situ" and were compared with data from 182 Spanish commercial cultivars grown in the same collection. All wild individuals showed morphological differences with Vitis vinifera L. ssp. vinifera but no significant differences were found within Vitis vinifera L subsp. sylvestris neither by geographic origin nor by sex. A pattern with the main characteristics of Spanish wild grapevines is suggested. Ultimately, were investigated the anthocyanin composition of 21 mostly Spanish wild grapevine accessions preserved at BGVCAM "El Encín" and selected in consideration of observed ampelographic differences and molecular characterization. Total anthocyanin concentration was similar to that found in winegrape cultivars. The accessions studied showed considerable variability in their anthocyanin fingerprints and it was possible to distinguish several groups, similar to previous reports on the anthocyanin fingerprint of winegrapes. The anthocyanin composition of wild grapevine accessions was similar to that of cultivated grapes. Nevertheless, the presence of wild accessions with anthocyanin fingerprints uncommon or nonexistent in Spanish cultivated varieties suggests that the genetic variability related to anthocyanins in Spanish wild grapevine populations may be higher than that of cultivated varieties commonly considered of Spanish origin.

Experiments were performed to confirm that the aldimine bond formation is a spontaneous reaction, because attempts to find an enzyme catalyzing the last decisive step in betaxanthin biosynthesis, the aldimine formation, failed. Feeding different amino acids to betalain-forming hairy root cultures of yellow beet (Beta vulgaris L. subsp. vulgaris “Golden Beet”) showed that all amino acids (S- and R-forms) led to the corresponding betaxanthins. We observed neither an amino acid specificity nor a stereoselectivity in this process. In addition, increasing the endogenous phenylalanine (Phe) level by feeding the Phe ammonia-lyase inhibitor 2-aminoindan 2-phosphonic acid yielded the Phe-derived betaxanthin. Feeding amino acids or 2-aminoindan 2-phosphonic acid to hypocotyls of fodder beet (B. vulgaris L. subsp. vulgaris “Altamo”) plants led to the same results. Furthermore, feeding cyclo-3-(3,4-dihydroxyphenyl)-alanine (cyclo-Dopa) to these hypocotyls resulted in betanidin formation, indicating that the decisive step in betacyanin formation proceeds spontaneously. Finally, feeding betalamic acid to broad bean (Vicia faba L.) seedlings, which are known to accumulate high levels of Dopa but do not synthesize betaxanthins, resulted in the formation of dopaxanthin. These results indicate that the condensation of betalamic acid with amino acids (possibly including cyclo-Dopa or amines) in planta is a spontaneous, not an enzyme-catalyzed reaction.

NADP-dependent isocitrate dehydrogenase (NADP-ICDH) activity is increased in roots of Eucalyptus globulus subsp. bicostata ex Maiden Kirkp. during colonization by the ectomycorrhizal fungus Pisolithus tinctorius Coker and Couch. To investigate the regulation of the enzyme expression, a cDNA (EgIcdh) encoding the NADP-ICDH was isolated from a cDNA library of E. globulus-P. tinctorius ectomycorrhizae. The putative polypeptide sequence of EgIcdh showed a high amino acid similarity with plant NADP-ICDHs. Because the deduced EgICDH protein lacks an amino-terminal targeting sequence and shows highest similarity to plant cytosolic ICDHs, it probably represents a cytoplasmic isoform. RNA analysis showed that the steady-state level of EgIcdh transcripts was enhanced nearly 2-fold in ectomycorrhizal roots compared with nonmycorrhizal roots. Increased accumulation of NADP-ICDH transcripts occurred as early as 2 d after contact and likely led to the observed increased enzyme activity. Indirect immunofluorescence microscopy indicated that NADP-ICDH was preferentially accumulated in the epidermis and stele parenchyma of nonmycorrhizal and ectomycorrhizal lateral roots. The putative role of cytosolic NADP-ICDH in ectomycorrhizae is discussed.

The wild ancestor of cultivated barley, Hordeum vulgare subsp. spontaneum (K. Koch) A. & Gr. (H. spontaneum), is a source of wide genetic diversity, including traits that are important for malting quality. A high β-amylase trait was previously identified in H. spontaneum strains from Israel, and transferred into the backcross progeny of a cross with the domesticated barley cv Adorra. We have used Southern-blot analysis and β-amy1 gene characterization to demonstrate that the high β-amylase trait in the backcross line is co-inherited with the β-amy1 gene from the H. spontaneum parent. We have analyzed the β-amy1 gene organization in various domesticated and wild-type barley strains and identified three distinct β-amy1 alleles. Two of these β-amy1 alleles were present in modern barley, one of which was specifically found in good malting barley cultivars. The third allele, linked with high grain β-amylase activity, was found only in a H. spontaneum strain from the Judean foothills in Israel. The sequences of three isolated β-amy1 alleles are compared. The involvement of specific intron III sequences, in particular a 126-bp palindromic insertion, in the allele-dependent expression of β-amylase activity in barley grain is proposed.

Grafting experiments between phytochrome B antisense and wild-type potato (Solanum tuberosum L. subsp. andigena [line 7540]) plants provide evidence that phytochrome B is involved in the production of a graft-transmissible inhibitor of tuberization, the level of which is reduced in the antisense plants, allowing them to tuberize in noninducing photoperiods.

As metalo-β-lactamases (MBL) são capazes de hidrolisar os carbapenêmicos, a classe de antimicrobianos com maior potência para o tratamento de infecções graves e de maior uso clinico. Dentre as MBL, o grupo mais recentemente descrito e que apresentou rápida disseminação em todo o mundo é o da New-Delhi-Metalo- β-lactamases (NDM). Nas enterobactérias, os genes que codificam essas enzimas estão mais frequentemente localizados em plasmídeos. O estudo da estabilidade de plasmídeos que albergam o gene blaNDM-1 é importante para entender a predominância de espécies que carregam esses plasmídeos, desvendar mecanismos moleculares envolvidos na sua persistência e para desenvolver novas drogas que possam diminuir a sua persistência. Estudos recentes sobre estabilidade plasmidial evidenciaram que a maprotilina é capaz de induzir perda plasmidial de até 90% em E. coli K12. Neste trabalho, foi estudado o efeito da maprotilina na indução de cura de plasmídeos, que albergam o gene blaNDM-1, em diferentes espécies da família Enterobacteriaceae. Nove isolados pertencentes a diferentes espécies foram incluídas no estudo. Os plasmídeos foram caracterizados quanto ao seu tamanho por eletroforese e por sequenciamento de DNA no sistema Illumina. A persistência plasmidial foi determinada pelo método de contagem em placa em LB ágar com e sem tratamento com maprotilina em concentrações sub-inibitórias (50mg/L). O experimento foi conduzido por 10 dias, representando aproximadamente 100 gerações. Neste estudo evidenciou-se que o grupo das enterobactérias estão envolvidas na disseminação de plasmídeos com blaNDM-1, sendo que plasmídeos do grupo IncF estão mais relacionados a essa dispersão. A maprotilina teve efeito de cura plasmidial em todos os isolados exceto em E. hormaechei \"subsp. oharae\" e C. freundii. O isolado P. rettgeri apresentou maior taxa de perda plasmidial e a análise comparativa da sequência nucleotídica do plasmídeo indicou que a presença da IS5 pode estar relacionada com a diminuição da persistência plasmidial. Diferenças na persistência plasmidial, quando tratados com maprotilina, entre E. hormaechei \"subsp. steigerwaltii\" e E. hormaechei \"subsp. oharae\" sugerem que E. hormaechei \"subsp. oharae\" pode ser um possível disseminador de plasmídeos albergando blaNDM-1, devido a processos de adaptação co-evolutivos.

Seven wild and cultivated Salvia species and two Phlomis species, used traditionally in Valencian medicine to treat a variety of external and internal ailments, were studied. New ethnobotanical data are provided, obtained from semistructured interviews with 34 people in the Valencian area. A seasonal characterization of the essential oil of a wild sage, Salvia blancoana Webb & Heldr. subsp. mariolensis Figuerola, by GC-FID and GC-MS was carried out as a means to ensure quality control of endemic traditional species such as this one, which has been commercialized by local industries. A comparison with the essential oil of Salvia lavandulifolia Vahl subsp.lavandulifolia allowed inclusion of the wild sage within the commercial 'Spanish sage' oil.
