An analysis of the dietary content of haematophagous insects can provide important information about the transmission networks of certain zoonoses. The present study evaluated the potential of polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) analysis of the mitochondrial cytochrome B (cytb)gene to differentiate between vertebrate species that were identified as possible sources of sandfly meals. The complete cytb gene sequences of 11 vertebrate species available in the National Center for Biotechnology Information database were digested with Aci I, Alu I, Hae III and Rsa I restriction enzymes in silico using Restriction Mapper software. The cytb gene fragment (358 bp) was amplified from tissue samples of vertebrate species and the dietary contents of sandflies and digested with restriction enzymes. Vertebrate species presented a restriction fragment profile that differed from that of other species, with the exception of Canis familiaris and Cerdocyon thous. The 358 bp fragment was identified in 76 sandflies. Of these, 10 were evaluated using the restriction enzymes and the food sources were predicted for four: Homo sapiens (1), Bos taurus (1) and Equus caballus (2). Thus, the PCR-RFLP technique could be a potential method for identifying the food sources of arthropods. However, some points must be clarified regarding the applicability of the method, such as the extent of DNA degradation through intestinal digestion, the potential for multiple sources of blood meals and the need for greater knowledge regarding intraspecific variations in mtDNA.

Avaliou-se o comprometimento funcional de pacientes com Charcot-Marie-Tooth provenientes da duplicação 17p11.2-p12 (CMT1A), utilizando o SF-36, que é um questionário para medir a qualidade de vida. Vinte e cinco pacientes de ambos os sexos com idades ≥10 anos e diagnóstico molecular de CMT1A foram selecionados. Idade, sexo, condições sociodemográficas e profissionais foram pareados com o Grupo Controle (sem histórico familiar de neuropatia). Os resultados mostraram que o maior impacto da CMT1A na qualidade de vida ocorreu nos domínios social e emocional dos pacientes avaliados. A capacidade funcional também tende a ser significativamente afetada, enquanto outros indicadores de deficiência física foram preservados. Por fim, os aspectos sociais e emocionais dos pacientes acometidos por CMT1A costumam ser negligenciados na assistência médica prestada aos pacientes brasileiros, e devem ser melhor compreendidos a fim de oferecer uma assistência global à saúde, resultando em adequada qualidade de vida.

Species from the Solenopsis saevissima (Smith) (Hymenoptera: Formicidae) species group are native to South America and have a cosmopolitan distribution because they have been accidentally introduced in many countries around the world. In Brazil, they have a wide distribution, including urban areas. The present study was conducted to investigate the characterization of Solenopsis genus populations associated with urban/human interference sites in Brazil by analyzing the mitochondrial gene cytochrome oxidase I and estimating the degree of relatedness of these populations to make inferences about their phylogeny and also observe the patterns of mitochondrial haplotype (mitotype) distribution across their range. The results revealed complete geographical coherence and polyphyly for the Solenopsis invicta Buren and Solenopsis saevissima species groups, which confirms the diversity of the genera. It also suggests the possibility that reproductively-isolated populations occur, resulting in the evolutionary process of speciation. No predominant haplotype was found in the populations analyzed, but some were more prevalent.

This study aims to characterize and to compare the students writing difficulties from 1st to 4th grades of public education. The study included 80 students divided into GI (20 students from 1st grade), GII (20 students from 2nd grade), GIII (20 students from 3rd grade) and GIV (20 students from 4th grade). The procedure applied was the Assessment of Learning Difficulties in Writing. The results revealed a median learning difficulty in students of GI (greater average of errors in writing). The increasing of schooling determine a decrease of the average of these errors from GI to GIV. The study revealed that the students are still in a process of construction and appropriate of the writing system, since the average of writing errors decreased as the school level increased.

A doença de Charcot-Marie-Tooth (CMT) é a mais freqüente das neuropatias hereditárias, mas a sua história natural é pouco conhecida. Apesar da maioria dos pacientes apresentarem uma evolução com características mais “benignas”, é associado à doença com um comprometimento funcional, o que pode ser acompanhado de limitações e incapacitações. O presente estudo teve como objetivo avaliar a qualidade de vida de pacientes afetados pela CMT1A. Considerando que a qualidade de vida pode sofrer influência de variáveis sócio-culturais, um estudo neste contexto mostra-se oportuno. Nenhum estudo de nosso conhecimento realizou tal avaliação no contexto brasileiro. Fizeram parte deste estudo pacientes com CMT decorrente da duplicação 17p11.2-p12 (CMT1A) que foram avaliados quanto ao comprometimento funcional por meio da aplicação do SF-36. Esse questionário é utilizado internacionalmente para aferir a qualidade de vida dos indivíduos. Os resultados obtidos foram submetidos à análise estatística. Como resultados não se observaram associações entre o gênero e os domínios avaliados para a qualidade de vida. Os dados obtidos revelaram que a qualidade de vida dos pacientes com CMT1A se mostrou comprometida principalmente quanto aos seus Aspectos Sociais (p = 0,02) e Aspectos emocionais (p = 0,04). Adicionalmente, houve uma tendência ao comprometimento da Capacidade Funcional (p = 0,08), sendo que os outros índices de avaliação de comprometimento físico se mostraram preservados. A presença ou ausência de ocupação parece influenciar os Aspectos Sociais apresentando uma tendência de diferença significativa (p = 0,07), possuindo o grupo com ocupação valores superiores. Apesar da CMT1A apresentar piora clínica ao longo dos anos, a idade mais avançada não implicou em uma pior percepção da qualidade de vida no grupo de pacientes estudados no presente trabalho... This paper presents methodology for an integrated treatment of three variable phenomena in Brazilian portuguese: (i) encoding of first-person plural into the forms nós (we) and a gente (the people), (ii) verbal agreement with the pronoun nós and (iii) verbal agreement with the pronominal form a gente. Based on the theoretical framework provided by Labovian sociolinguistics (LABOV, 1966, 1972), the methodology is applied to a sample of Brazilian portuguese spoken in the countryside of São Paulo State (GONÇALVES, 2007). The results indicate that distinct factors predominate in the choice of the alternative forms of each phenomenon: in the verbal agreement with a gente, linguistic factors are the most prominent; in the verbal agreement with nós, social factors are the most salient; and in the use of nós/a gente both linguistic and social factors prevail.

The use of molecular markers may auxiliary the buffalo breeding. The oxytocin (OXT) and the adrenergic receptor alpha(1A) (ADRA1A) may be involved in milk ejection in ruminants. The aim of this study was to verify the existence of polymorphisms in the OXT and ADRA1A genes and their associations with milk production traits. A total of 220 buffaloes were genotyped using PCR-RFLP for both genes. The SNP identified in the ADRA1A gene was associated with protein percentage in dairy buffaloes. This is the first report of such association in the literature, which has not been studied in other species.

The interaction of cytochrome c (cyt c) with cardiolipin (CL) induces protein conformational changes that favor peroxidase activity. This process has been correlated with CL oxidation and the induction of cell death. Here we report evidence demonstrating the generation of singlet molecular oxygen [O-2((1)Delta(g))] by a cyt c-CL complex in a model membrane containing CL. The formation of singlet oxygen was directly evidenced by luminescence measurements at 1270 nm and by chemical trapping experiments. Singlet oxygen generation required cyt c-CL binding and occurred at pH values higher than 6, consistent with lipid-protein interactions involving fully deprotonated CL species and positively charged residues in the protein. Moreover, singlet oxygen formation was specifically observed for tetralinoleoyl CL species and was not observed with monounsaturated and saturated CL species. Our results show that there are at least two mechanisms leading to singlet oxygen formation: one with fast kinetics involving the generation of singlet oxygen directly from CL hydroperoxide decomposition and the other involving CL oxidation. The contribution of the first mechanism was clearly evidenced by the detection of labeled singlet oxygen [O-18(2)((1)Delta(g))] from liposomes supplemented with 18-oxygen-labeled CL hydroperoxides. However quantitative analysis showed that singlet oxygen yield from CL hydroperoxides was minor (<5%) and that most of the singlet oxygen is formed from the second mechanism. Based on these data and previous findings we propose a mechanism of singlet oxygen generation through reactions involving peroxyl radicals (Russell mechanism) and excited triplet carbonyl intermediates (energy transfer mechanism).
